Protein glycosylation is a type of protein post-translational modification. One specific example is the modification of proteins with O-linked β-N-acetylglucosamine (O-GlcNAc) and O-linked α-N-acetylgalactosamine (O-GalNAc). Enhanced levels of both O-GalNAc and O-GlcNAc in bladder cancer (BlCa) have been reported previously. However, the interplay between O-GalNAc and O-GlcNAc has yet to be explored. Herein, we find that the expression level of core1 β-1,3-galactosyltransferase (C1GalT1), which is responsible for extending and maturing mucin-type O-glycans, is increased in BlCa. This increase is accompanied by O-GlcNAc modification of C1GalT1. This modification stabilizes C1GalT1 expression and strengthens its interaction with its chaperone Cosmc. Mutation at Thr229 or Thr233 attenuates C1GalT1 stability and facilitates its degradation via the proteasome pathway. Furthermore, a decrease in C1GalT1 inhibits the pro-tumorigenic effect on bladder cancer cells by suppressing glycolysis.

Objective: This study aimed to enrich the functional lactic acid bacteria resource bank and to search for excellent starting strains of lactic acid bacteria with the antagonistic ability of Vibrio parahaemolyticus. Methods: Lactobacillus strains with broad-spectrum bacteriostatic potential were screened by Oxford cup bacteriostatic test, and their biological characteristics were discussed by growth and metabolic performance, gastrointestinal fluid tolerance, salt tolerance, antibiotic sensitivity and bacteriostatic spectrum. Results: Six strains of lactic acid bacteria were screened by using V. parahaemolyticus as indicator bacteria. They were classified into L. paracasei, L. fermentus and L. plantarum by morphological, physiological and biochemical identification and molecular biological identification. Among them, the bacteriostatic activity of L. paracasei A1 was the best, the total number of colonies within 24 hours exceeded 1×109 CFU/mL, and the pH value of fermentation broth was stable at about 4.1. After artificial simulated gastric juice treatment for 2 h, the survival rate was 54.61%. After artificial simulated intestinal juice treatment for 8 h, the survival rate still reached 45.46%. After 10% NaCl stress treatment for 24 h, the total number of viable bacteria was ＞ 1×105 CFU/mL. Moreover, the crude bacteriocin extract of L. paracasei A1 showed good antibacterial activity against 13 pathogenic bacteria, had broad-spectrum antibacterial potential, and no resistance to 8 common antibiotics was found. Conclusion: L. paracasei A1, which can inhibit V. parahaemolyticus and has excellent biological characteristics, was obtained.

After the completion of the three steps of "circling back", the lunar exploration has changed from the short-term single point exploration to the long-term exploration of lunar scientific research stations with the con-struction of lunar surface infrastructure, the higher requirements for the planning and demonstration, overall de-sign, system development and in-orbit exploration of lunar exploration missions are put forward. The thought of model based system engineering (MBSE) is adopted, a model-based forward process for the system analysis of the lunar scientific research station proposal is proposed, and the system model is taken as a carrier to analyze the mis-sion overall, mission requirements and task application scenario. By carrying out the model-based mission analysis of the lunar scientific research station, the process of the mission of the lunar scientific research station has been pre-liminarily realized.

An accurate intrinsic structural model is essential to describing the high-temperature deformation behavior of metal materials. Support Vector Regression (SVR) has strong regression analysis capabilities, but its application research in constructing constitutive models of 25CrMo4 steel still needs to be improved. In this study, we use grid search, particle swarm optimization, improved genetic algorithm, and improved gray wolf optimization to optimize SVR parameters. A constitutive relationship model for 25CrMo4 steel under high-temperature compression based on SVR was established through training using experimental data models. The predicted data of SVR constitutive models with different optimization algorithms were compared with experimental data. Statistical values, such as average absolute percentage error (AAPE), mean absolute percentage error (MAPE), and correlation coefficient (R2), were introduced to evaluate the accuracy of each model. The particle swarm optimization-SVR model achieved the best performance, with an AAPE of 0.455 38, MAPE of 0.489 09%, and R2 of 0.999 74. Furthermore, compared to other models, it requires the least time. This model has a higher accuracy than other commonly used instantaneous models. These findings can provide a basis for selecting appropriate deformation parameters and preventing hot working defects of 25CrMo4 steel, thus helping to improve the manufacturing process and material properties.

Introduction: Issuance pricing is an important part of the operation of the securities market. Its pricing is directly related to the interests of issuers, investors and underwriters, as well as the regulatory and resource allocation functions of the securities issuance market. When the industry expectations vary greatly, the company has greater TMT heterogeneity. When the market expectation gap expands, the possibility of risk conversion increases. IPO underpricing occurs in stocks of developed countries, developing countries and emerging countries. Because the issuer and the underwriter investors have different information, for example, the information advantage of the underwriter will bring risks to investors when purchasing IPO shares. IPO underpricing is a compensation for the risks brought by information asymmetry.Methods: At present, little attention has been paid to how the value of listed companies affects the underpricing of intellectual property. This paper will fill in this gap and empirically study the impact of management ability of senior management team on IPO underpricing. Therefore, starting from the Chinese stock market, this paper studies the evidence of vertical parallel executives, heterogeneity of senior management team and IPO underpricing.Results: The average values of NCSKEW (C1) and DUVOL (C2) were −0.301 and −0.203, respectively, which were close to the descriptive statistics of the study.Discussion: The dependent variable data in this paper has certain reliability. The minimum value of NCSKEW is −0.361 and the maximum value is 0.392, indicating that the fragmentation risk of different stocks is relatively high.

Background: Jiedu Sangen Decoction (JSD), a traditional Chinese medicine formula, has been widely applied in the treatment of gastrointestinal cancer, especially in colorectal cancer. Our study mainly aimed to assess the combined efficacy of Jiedu Sangen aqueous extract (JSAE) and a PD-L1 inhibitor (PI) in colon cancer cells migration and invasion, along with epithelial-mesenchymal transition, and then provide deep insights into the potential mechanism. Methods: We explored the inhibitory effects on invasion and metastasis and the reverse effect on EMT process in CT-26 colon cancer cell via Transwell migration assay, Matrigel invasion assay and confocal laser scanning microscopy. Furthermore, regulation in expression of EMT-related proteins and molecular biomarkers and underlying signal pathway proteins were detected through Western blotting and IHC. Results: The combination of JSD and PD-L1 inhibitor could inhibit migration, invasive ability and EMT of CT-26 cells in a concentration-dependent manner. Meanwhile, JSD combined with PD-L1 inhibitor could also remarkably reverse EMT and metastasis in vivo. In addition, the protein expression of N-cadherin, Slug, Snail, Vimentin was down-regulated along with E-cadherin s up-regulation with the combination of JSD and PD-L1 inhibitor, while that of PI3K/AKT was notably down-regulated. Conclusions: These findings indicated that JSAE and a PD-L1 inhibitor could drastically inhibit the migration and invasion of colorectal cancer by reversing EMT through the PI3K/AKT signaling pathway.

The aims of this study were to investigate the status quo of self-management behaviors in stroke patients at the recovery stage and to explore its influencing factors.

Lichong Shengsui Yin (LCSSY) is an effective and classic compound prescription of Traditional Chinese Medicines (TCMs) used for the treatment of ovarian cancer. To investigate its pharmacodynamic basis for treating ovarian cancer, the multi-dimensional spectrum-effect relationship was determined. Four compositions (I to IV) were obtained by extracting LCSSY successively with supercritical CO2 fluid extraction, 75% ethanol reflux extraction, and the water extraction-ethanol precipitation method. Nine samples for pharmacological evaluation and fingerprint analysis were prepared by changing the content of the four compositions. The specific proportions of the four compositions were designed according to a four-factor, three-level L9(34) orthogonal test. The pharmacological evaluation included in vitro tumor inhibition experiments and the survival extension rate in tumor-bearing nude mice. The fingerprint analyzed by chromatographic condition I (high-performance liquid chromatography-photodiode array detec tor，HPLC-PDA) identified 19 common peaks. High-performance liquid chromatography-photodiode array detector-Evaporative Light-scattering Detector (HPLC-PDA-ELSD )hyphenated techniques were used to compensate for the use of a single detector, and the fingerprint analyzed by chromatographic condition II identified 28 common peaks in PDA and 23 common peaks in ELSD. Furthermore, multiple statistical analyses were utilized to calculate the relationships between the peaks and the pharmacological results. The union of the regression and the correlation analysis results were the peaks of X5, X9, X11, X12, X16, X18, Y5, Y8, Y12, Y14, Y20, Z4, Z5, Z6, and Z8. The intersection of the regression and the correlation analysis results were the peaks of X11, X12, X16, X18, Y5, Y12, and Z5. The correlated peaks were assigned by comparing the fingerprints with the negative control samples and reference standard samples, and identifying the structure using high-performance liquid chromatography-mass spectrometry detector(HPLC-MS). The results suggested that the pharmacodynamic basis of LCSSY on anti-ovarian cancer activities were germacrone, furandiene, β-elemene, calycosin-7-glucoside, ononin, epimedin B, icariin, ginsenoside Rc, astragaloside, ginsenoside Rd, astragaloside II, and some unknown components.
