To study the quality discrepancy of jujube vinegar brewed from different jujube varieties, six types of jujube vinegar made from different varieties of jujube were evaluated by the analysis of basic physicochemical components (such as pH, total acid, total phenol and total flavone), color, in vitro functional activity (antioxidant capacity and lipid-lowering ability), organic acids, amino acids, and volatile aroma components, combined with electronic nose and sensory evaluation. The results showed that there were significant differences in the quality of jujube vinegar brewed from different jujubes. In terms of physical and chemical analysis, the total acid and total flavonoids of Xinjiang jujube vinegar were the highest, which were 54.87 g/L and 1168.84 mg RE/L, respectively. By contrast, the total phenolic content of Xinjiang Jun-jujube jujube vinegar was the highest, which was 1815.63 mg GAE/L. Henan Lingbao jujube vinegar and Fuping jujube vinegar had better color values. Xinjiang Jujube vinegar showed the best antioxidant and lipid lowering activities. Fuping jujube vinegar had the lowest total organic acid content of 47.07 g/L. Xinjiang gray jujube vinegar and Xinjiang Jun-jujube vinegar had higher essential amino acid and total amino acid content, while Hebei Fuping jujube vinegar had lower amino acid content. A total of 41 volatile aromas were detected in 6 kinds of jujube vinegar, and there were certain differences in the types and contents of volatile aromas contained in different kinds of jujube vinegar. The sensory evaluation showed that all jujubes were suitable for brewing jujube vinegar, among which Xinjiang Hui-jujube vinegar had the best sensory acceptance. In summary, there were certain differences in the quality of 6 kinds of jujube vinegar. The high functional activity of Xinjiang Jun-jujube vinegar made it more suitable as a functional fruit vinegar. Based on the high sensory acceptance of Xinjiang Hui-jujube vinegar, the Xinjiang Hui-jujube would be suitable for the vinegar processing too.

Abstract Observational and modeling studies have elucidated the influential role played by the southern and northern extratropical Pacific (SEP and NEP) forcing in shaping dynamics of tropical Pacific climate variability. However, the relative importance of the NEP and SEP and the timescale on which they impact the tropics remain unclear. Using a linear inverse model (LIM) that selectively incorporates or excludes tropical‐extratropical coupling, we find a reduction in tropical interannual variability (∼40%) and low‐frequency (sub‐decadal to decadal) variability in the southeastern tropical Pacific region (∼70%) in the absence of SEP. Conversely, the absence of NEP yields no significant impact on tropical interannual variability but markedly diminishes low‐frequency variability in the central tropical Pacific region (∼70%). LIM and statistic diagnostics on CMIP6 models show the low‐frequency to total variability ratio in the tropical Pacific depending on their NEP and SEP representation. Models with more (less) low‐frequency power tend to show stronger NEP (SEP) dynamics.

Aluminum thermal welding joints are widely used in the maintenance welding of heavy-haul railways due to their easy handling and high efficiency. However, due to their inherent welding characteristics, welding results in certain differences in the material’s physical properties at the welding zone compared to adjacent base materials, leading to the occurrence of short-wave irregularity under long-term wheel–rail interactive forces. In order to explore the evolution characteristics of weld irregularity, dynamic characteristics, and plastic deformation under long-term wheel–rail impact, a detailed tracking test was conducted on a normal aluminum weld, and the process from being put on the track to being damaged and replaced was evaluated. At the same time, a rigid–flexible coupling model was established for subsequent analysis, and plastic damage was analyzed using the finite element model. The results show that the service life of the weld can be divided into three different stages: the initial stage, the intermediate stage, and the damage stage. In the damage stage, a temporary separation occurred between the wheel and rail, leading to a sudden change in the wheel–rail interaction. The weight of 250 MT at the weld reached the repairment control limit. The concentration effect of equivalent plastic deformation was most serious at 2~5 mm below the rail head.

Abstract The ocean circulation is modulated by meandering currents and eddies. Forecasting their evolution is a key target of operational models, but their forecast skill remains limited. We propose a machine learning approach that improves the output of an ocean circulation model by learning and predicting its systematic biases. This method can be applied a priori to any region, and is tested in the Gulf of Mexico, where the Loop Current (LC) and the large anticyclonic eddies that detach from it are major forecasting targets. The LC dynamics are recurrent and lie on a low‐dimensional dynamical attractor. Building upon the information gained analyzing this low dimensional attractor, we improve the representation of sea surface anomalies in model outputs through information from satellite altimeter data using a Sequence‐to‐Sequence model, which is a special class of Recurrent Neural Network. Building upon the HYCOM‐NCODA analysis system, we deliver a correction to the forecast at the observation resolution. For at least 15 days the proposed method learns to forecast the systematic bias in the HYCOM‐NCODA, outperforming persistence, and improving the forecast. This data‐driven approach is fast and can be implemented as an added step to any dynamical hindcasting or forecasting model. It offers an interesting avenue for further developing hybrid modeling tools. In these tools, fundamental physical conservations are preserved through the integration of partial differential equations which obey them. In addition, the method highlights specific deficiencies of the hindcast system that deserve further investigation in the future.

Background: Upper blepharoplasty is one of the most commonly performed esthetic surgeries, but injection pain during local anesthesia often disturbs patients. The objective of this study was to identify the pain levels associated with anesthetizing the upper eyelids in blepharoplasty and determine whether injection with thin needles is associated with less pain and bleeding than injection with thick needles. Methods: The study included 50 patients who underwent bilateral upper blepharoplasty. The eyelids were anesthetized using 2% lidocaine with 1:100 ​000 epinephrine. One upper eyelid was randomly injected with a thin needle (26G) and the other with a thick needle (22G). The pain level was scored by patients immediately after the injection of each eyelid using a visual analog scale (VAS) ranging from 0 to 10. The incidence of eyelid hematomas caused by the anesthetic injection was also recorded. Results: The VAS scores in eyelids injected with thin needles were not significantly lower than those in eyelids injected with thick needles (4.0 vs. 4.0, P=0.393). The formation of eyelid bruises caused by thin-needle injection was less frequent than that caused by thick-needle injection (16% vs. 26%), but there was no significant difference between the two groups (P=0.326). Conclusion: Using thin needles for local anesthesia during upper blepharoplasty could not significantly decrease injection pain or hematoma occurrence compared with that associated with using thick needles.

The Gulf of Mexico is a very productive and economically important system where riverine runoff acts as a linkage between the continental shelf and the open ocean, providing nutrients in addition to freshwater. This work investigates the three-dimensional transport and pathway structure of this river runoff offshore the continental shelf using ensembles of numerical simulations with different configurations regarding grid resolution (mesoscale resolving and submesoscale permitting) and river setup using suites of 5-months long integrations covering nearly 3 years. The riverine forcing is applied only at the surface over an area around the river mouth, a strategy often adopted in numerical studies, or as a meridional flux with a vertical extension. The simulated flow captures the southward offshore transport of river runoff driven by its interaction with the largest mesoscale circulations in the basin, the Loop Current and Loop Current eddies. This pathway is strong and well-document during summer but also active and relevant in winter, despite a less obvious surface signature. The most intense transport occurs primarily at the peripheries of the Loop Current and the detached eddies, and the freshwater is subducted as deep as 600 m around the mesoscale anticyclonic eddies. Submesoscale motions strengthen slightly the spread of freshwater plumes in summer but their contribution is negligible, if not negative, in winter. Differences in the freshwater distribution and transport volume among runs are small and generally less than 10% among ensembles, with overall slightly higher volume of freshwater transported off-shore and at depth in submesoscale permitting runs that include a velocity flux in their riverine input representation.

Fine-scale larval dispersal and connectivity processes are key to species survival, growth, recovery and adaptation under rapidly changing disturbances. Quantifying both are required to develop any effective management strategy. In the present work, we examine the dispersal pattern and potential connectivity of a common deep-water coral, Paramuricea biscaya, found in the northern Gulf of Mexico by evaluating predictions of physical models with estimates of genetic connectivity. While genetic approaches provide estimates of realized connectivity, they do not provide information on the dispersal process. Physical circulation models can now achieve kilometer-scale resolution sufficient to provide detailed insight into the pathways and scales of larval dispersal. A high-resolution regional ocean circulation model is integrated for 2015 and its advective pathways are compared with the outcome of the genetic connectivity estimates of corals collected at six locations over the continental slope at depths comprised between 1,000 and 3,000 m. Furthermore, the likely interannual variability is extrapolated using ocean hindcasts available for this basin. The general connectivity pattern exhibits a dispersal trend from east to west following 1,000 to 2,000-m isobaths, corresponding to the overall westward near-bottom circulation. The connectivity networks predicted by our model were mostly congruent with the estimated genetic connectivity patterns. Our results show that although dispersal distances of 100 km or less are common, depth differences between tens to a few hundred meters can effectively limit larval dispersal. A probabilistic graphic model suggests that stepping-stone dispersal mediated by intermediate sites provides a likely mechanism for long-distance connectivity between the populations separated by distances of 300 km or greater, such as those found in the DeSoto and Keathley canyons.

The degree of connectivity among populations influences their ability to respond to natural and anthropogenic stressors. In marine systems, determining the scale, rate, and directionality of larval dispersal is therefore, central to understanding how coral metapopulations are interconnected and the degree of resiliency in the event of a localized disturbance. Understanding these source-sink dynamics is essential to guide restoration efforts and for the study of ecology and evolution in the ocean. The patterns and mechanisms of connectivity in the deep-sea (>200 m deep) are largely understudied. In this study, we investigated the spatial diversity patterns and metapopulation connectivity of the octocoral Paramuricea biscaya throughout the northern Gulf of Mexico (GoM). Paramuricea biscaya is one of the most abundant corals on the lower continental slope (between 1,200 and 2,500 m) in the GoM. The 2010 Deepwater Horizon oil spill (DWH) directly impacted populations of this species and thus are considered primary targets for restoration. We used a combination of seascape genomic analyses, high-resolution ocean circulation modeling, and larval dispersal simulations to quantify the degree of population structuring and connectivity among P. biscaya populations. Evidence supports the hypotheses that the genetic diversity of P. biscaya is structured by depth, and that larval dispersal among connected populations is asymmetric due to dominant ocean circulation patterns. Our results suggest that there are intermediate unsampled populations in the central GoM that serve as stepping stones for dispersal. The data suggest that the DeSoto Canyon area, and possibly the West Florida Escarpment, critically act as sources of larvae for areas impacted by the DWH oil spill in the Mississippi Canyon. This work illustrates that the management of deep-sea marine protected areas should incorporate knowledge of connectivity networks and depth-dependent processes throughout the water column.

Submesoscale circulations influence momentum, buoyancy and transport of biological tracers and pollutants within the upper turbulent layer. How much and how far into the water column this influence extends remain open questions in most of the global ocean. This work evaluates the behavior of neutrally buoyant particles advected in simulations of the northern Gulf of Mexico by analyzing the trajectories of Lagrangian particles released multiple times at the ocean surface and below the mixed layer. The relative role of meso- and submesoscale dynamics is quantified by comparing results in submesoscale permitting and mesoscale resolving simulations. Submesoscale circulations are responsible for greater vertical transport across fixed depth ranges and also across the mixed layer, both into it and away from it, in all seasons. The significance of the submesoscale-induced transport, however, is far greater in winter. In this season, a kernel density estimation and a detailed vertical mixing analysis are performed. It is found that in the large mesoscale Loop Current eddy, upwelling into the mixed layer is the major contributor to the vertical fluxes, despite its clockwise circulation. This is opposite to the behavior simulated in the mesoscale resolving case. In the “submesoscale soup,” away from the large mesoscale structures such as the Loop Current and its detached eddies, upwelling into the mixed layer is distributed more uniformly than downwelling motions from the surface across the base of the mixed layer. Maps of vertical diffusivity indicate that there is an order of magnitude difference among simulations. In the submesoscale permitting case values are distributed around 10–3 m2 s–1 in the upper water column in winter, in agreement with recent indirect estimates off the Chilean coast. Diffusivities are greater in the eastern portion of the Gulf, where the submesoscale circulations are more intense due to sustained density gradients supplied by the warmer and saltier Loop Current.

Pectin methylesterase (PME) is a methyl ester group hydrolytic enzyme of either plant or microbial origin. Importantly, endogenous PMEs in fruits can catalyze the demethoxylation of pectin with a bulk release of methanol, largely impacting the fruit juice and wine industries. Here, we demonstrated radio frequency (RF) heating for inactivation of endogenous PMEs and investigated the relevant mechanisms underpinning enzymatic inactivation. The RF heating curve indicated that the optimal heating rate was achieved at an electrode gap of 90 mm (compared to 100 mm and 110 mm) and that the inactivation rate of the enzyme increases with heating time. RF heating exhibited better effects on enzymatic inactivation than traditional water heating, mainly by changing the secondary structures of PMEs, including α-helix, β-sheet, β-turn, and random coil. Moreover, fluorescence spectroscopy indicated changes in the tertiary structure with a significant increase in fluorescence intensity. Significantly, application of RF heating for inactivation of PMEs resulted in a 1.5-fold decrease in methanol during the fermentation of jujube wine. Collectively, our findings demonstrated an effective approach for inactivating endogenous PMEs during the bioprocesses of fruits.

High-speed railways adopt continuous welded rail to maintain the smoothness and continuity of the rail surface. However, the welded joint became one of the weakest parts. In order to clear the characteristics and mechanical properties of the new reinforced device, a dynamic three-dimensional vehicle-reinforced device-track coupling model is established. The mechanical characteristics of the track structure under high-speed train load were simulated and analyzed. After installing the new reinforced device, the dynamic response and service life of the track structure are obviously improved compared with the unreinforced rail. When the train speed is 300 km/h, the dynamic bending stress at the bottom of rail is reduced by 26.90%, the vertical and lateral acceleration of the rail are reduced by 42.78% and 21.56%, the vertical and lateral displacement of the rail are reduced by 6.36% and 8.67%, and the theoretical service life of the rail is greatly extended.

The 2010 Macondo oil well blowout consisted in a localized, intense infusion of petroleum hydrocarbons to the deep waters of the Gulf of Mexico. A substantial amount of these hydrocarbons did not reach the ocean surface but remained confined at depth within subsurface plumes, the largest and deepest of which was found at ∼ 1000–1200 m of depth, along the continental slope (the deep plume). This review outlines the challenges the science community overcame since 2010, the discoveries and the remaining open questions in interpreting and predicting the distribution, fate and impact of the Macondo oil entrained in the deep plume. In the past 10 years, the scientific community supported by the Gulf of Mexico Research Initiative (GoMRI) and others, has achieved key milestones in observing, conceptualizing and understanding the physical oceanography of the Gulf of Mexico along its northern continental shelf and slope. Major progress has been made in modeling the transport, evolution and degradation of hydrocarbons. Here we review this new knowledge and modeling tools, how our understanding of the deep plume formation and evolution has evolved, and how research in the past decade may help preparing the scientific community in the event of a future spill in the Gulf or elsewhere. We also summarize briefly current knowledge of the plume fate – in terms of microbial degradation and geochemistry – and impacts on fish, deep corals and mammals. Finally, we discuss observational, theoretical, and modeling limitations that constrain our ability to predict the three-dimensional movement of waters in this basin and the fate and impacts of the hydrocarbons they may carry, and we discuss research priorities to overcome them.

Mesoscale eddies and fronts in the ocean greatly impact lateral transport and in turn the trajectories of sinking particles. Such influence was explored for April and October 2012 in the Gulf of Mexico using numerical simulations performed with a regional model at 1-km horizontal resolution. Results are compared qualitatively to field samples from two sediment traps located at GC600 (27°22.5 N, 90°30.7 W) and AT357 (27°31.5 N, 89°42.6 W), 81 km apart. In April the traps collected a comparable amount of material, while in October the flux at GC600 greatly exceeded that at AT357. Through inverse calculations, several thousand particle trajectories were reconstructed multiple times from the ocean surface to the depth of the traps (approximately 1,000 m) using a range of sinking velocities, 20–100 m d–1. Taken together, model results and trap data indicate that cross-shore transport of riverine input induced by mesoscale eddies, and convergence and divergence processes at the scale of a few kilometers, significantly impact the trajectory of sinking particles. The large majority of modeled particles reach the bottom faster than would be expected by their sinking speeds alone. This finding is associated with submesoscale-induced horizontal convergence in the mixed layer that aggregates particles preferentially in downwelling regions, accelerating their descent. Furthermore, this study confirms that the cone of influence of vertical fluxes is highly variable in both space and time in the presence of an energetic eddy field, especially for particles with sinking velocity of 50 m d–1 or less. It also demonstrates that the variability of vertical fluxes in the Gulf of Mexico is highly complex and can be understood only by considering the mesoscale circulation and seasonal cycle of primary productivity, which in turn are linked to riverine inputs, wind forcing and the seasonal cycle of the mixed-layer depth.

The existence of multipotent adipose-derived stem cells isolated from human orbital fat (OF) tissue has shown great therapeutic potential in tissue engineering and regenerative medicine. But the use of stem cells for therapeutic applications is influenced by their proliferative and differentiation potentials, which may be affected by the age of the donor. So far there is little knowledge about the effects of donor age on the biological properties of human orbital adipose-derived stem cells (OASCs). The intraorbital fat protrusion in the lower eyelids occurs as an aging process, and the protruded fat is routinely removed during aesthetic surgeries. Based on the ease of OF harvest and the availability of OASCs, we investigated in this study the relationship between age and the differentiation and proliferation potentials of human OASCs. Human orbital adipose samples were harvested from young (with normal lower eyelid appearance) and old donors (having protruded fat pads in the lower eyelids). The morphological properties of orbital adipocytes were assessed and the fat cell size displayed a decreasing trend with advancing age. OASCs were isolated from the fat samples, expanded in vitro and cultured under appropriate inducive conditions. Compared to the young cells, although no difference was found in the cell yield and phenotype expression, aged OASCs showed fewer progenitor cell numbers, reduced proliferative rates, increased senescent features and decreased differentiation potentials towards adipogenic, osteogenic and chondrogenic lineages. Our data suggested that using autologous OASCs from elderly patients for potential therapeutic purposes might be restricted.

microRNAs (miRNAs), small noncoding RNAs of 19-25 nt, play an important roles in the pathological processes of tumorigenesis. The object of this study was to study the expression and function of miR-203 and to found its target gene in osteosarcoma. In our study, we found the expression level of miR-203 was significantly downregulated in osteosarcoma cell lines and tissues. In addition, overexpression of miR-203 inhibited the osteosarcoma cell proliferation and migration and inhibited Mesenchymal-to-Epithelial reversion Transition (MErT). Moreover, we identified RAB22A as a direct target of miR-203 and RAB22A overexpression blocks the roles of miR-203 in osteosarcoma cell. Furthermore, we demonstrated that RAB22A expression was upregulated in human osteosarcoma cell lines and tissues. Take together, our results demonstrated that miR-203 act as a tumor suppressor miRNA through regulating RAB22A expression and suggested its involvement in osteosarcoma progression and carcinogenesis.

[Objectives] To explore the effects of different sterilization conditions on nutrition and flavor of apple vinegar. [Methods] Five kinds of high temperature short time (HTST) sterilization conditions were selected to treat apple vinegar, and the volatile aroma components and the content of active components in apple vinegar before and after sterilization were analyzed. [Results] Compared with the control, the contents of total acid and malic acid in the samples after sterilization changed little, but the contents of citric acid increased significantly ( P < 0. 01), and the contents of total phenols, ascorbic acid and total flavonoids decreased. Ethyl acetate, isopentyl acetate, ethyl caprylate, phenethyl acetate, 1-pentanol, phenylethyl alcohol, acetic acid, and sec-butyl ether were the characteristic aroma components which contributed to the flavor of apple vinegar. As sterilization temperature increased, the content of esters decreased, while the content of acids, alcohols and aldehydes increased. The contents of nutrition, active components and volatile aroma components in apple vinegar under 100 °C and 30 s sterilization conditions had little change compared with other sterilization conditions, so 100 °C and 30 s were the optimal sterilization conditions. [Conclusions] Under different sterilization conditions, the content of flavor components in apple vinegar will change greatly, which will affect the quality of apple vinegar. [ABSTRACT FROM AUTHOR]

[Objectives] The paper was to study the effect of magnetic field technology on physicochemical properties of fresh-cut apple during storage. [Methods] Using fresh-cut apples as materials, magnetic field assisted refrigeration and conventional refrigeration control groups were set up, and the preservation effects of magnetic field assisted refrigeration on fresh-cut apples were tested by measuring the changes of microorganisms and physicochemical indicators. In this experiment, the conditions of magnetic field assisted refrigeration group was set as follows: temperature 4 °C, magnetic field intensity 5 mT, static magnetic field, and storage for 6 d. [Results] At 6 d post storage, the weight loss rate, total number of colonies and total number of fungi of fresh-cut apples increased; the i" value and hardness showed a downward trend; the soluble solid content first increased and then decreased; and the total acid decreased first and then increased. The above indicators in magnetic field assisted refrigeration group changed more gently than that in conventional refrigeration group. Compared with conventional assisted refrigeration, the weight loss rate of fresh-cut apples treated by magnetic field combined with 4 9C refrigeration was reduced by 68.00%; the hardness was increased by 12.23%; the total number of colonies and total number of fungi were reduced by 64.15% and 37.50%; and the L" value did not change much. High content of soluble solids and total acid was maintained. [Conclusions] Magnetic field assisted refrigeration delays quality deterioration of fresh-cut apples during storage, prolongs shelflife, and further improves their edible quality and commodity value. Due to its unique advantages, molecular marker technology is widely applied in the research of forest tree species. This paper reviewed the application of molecular marker technology in tree species resource diversity, germplasm identification, genetic map construction, gene mapping and marker-assisted selection (MAS) breeding. Objectives: To compare functional properties of mulberry black tea fermented by Phellinus igniarius and Eurotium cristatum. Methods: The functional components of mulbery black tea fermented by P. igniarus and E. cristatum, the deep processing and health care effects of two probiotic fungi were explored. Results: The polysaccharide content in the tea fermented by P. igniarius was the highest at 0.410 mg/mL, and the hydroxyl radical scavenging rate was the strongest, reaching 60. 31% ; the content of flavonoids in the tea fermented by E. cristatum was the highest, reaching 60. 65 pg/mL, and the content of polysaccharides was 83. 17% higher than that of mulbery black tea, reaching 0.279 mg/mL. Conclusions: The mulbery black tea fermented by P. igniarius and E. cristatum has improved in different func-tional components, and has its own advantages in aroma and mouthfeel, so it ha certain research value and market prospect.
