An environmental enrichment protocol is essential for testing experimental models because it upholds animal welfare, aligns with ethical principles in animal experimentation, and reduces the number of animals needed. Calomys callosus , a South American rodent from the Cricetidae family, is bred in rodent animal facilities for its ease of handling, longevity, prolificacy, and effectively mimicking diseases like Toxoplasmosis, Leishmaniasis, Chagas, and Schistosomiasis. There are no reports on environmental enrichments for this species or their impact on reproductive parameters. This study aimed to analyze the influence of the Environmental Enrichment Program (EEP) on the reproductive and zootechnical performance of C. callosus kept in the Rodents Animal Facilities Complex of Universidade Federal de Uberlândia (UFU). Two experimental groups were established: with environmental enrichment EE+ and without environmental enrichment EE-. The materials used in the experimental design were changed weekly and alternated between dietary, occupational, physical/cognitive, and non-enrichment items. After the inclusion of the EEP, an improvement in the reproductive indices of C. callosus was identified in the EE+ group. These improvements included increased female precocity, a decreased interbirth interval, and a higher number of pairs producing more offspring. The postpartum zootechnical indices were also better, such as the number of animals born alive, improved weaning rates, and a reduced average number of deaths from birth to weaning. After the inclusion of the EEP, the general health status of C. callosus improved, reducing cases of non-infectious lumbar alopecia. Currently, there is a wide application in the literature of the use of the Fourier Transform Infrared Spectroscopy (FTIR) technique. This basic tool has also proven to be efficient for detecting molecules associated with hosts and pathogens in infections, as well as other molecules present in humans and animals' biological samples. However, there is a crisis in science data reproducibility. This crisis can also be observed in data from experimental animal models (EAMs). When it comes to rodents, a major challenge is to carry out sanitary monitoring, which is currently expensive and requires a large volume of biological samples, generating ethical, legal, and psychological conflicts for professionals and researchers. We carried out a survey of data from the relevant literature on the use of this technique in different diagnostic protocols and combined the data with the aim of presenting the technique as a promising tool for use in EAM. Since FTIR can detect molecules associated with different diseases and has advantages such as the low volume of samples required, low cost, sustainability, and provides diagnostic tests with high specificity and sensitivity, we believe that the technique is highly promising for the sanitary and stress and the detection of molecules of interest of infectious or non-infectious origin.

The aim of the present research was to add to the growing literature on dopamine and gambling disorder (GD) by assessing whether GD is associated with dopamine transporter (DAT) density in the ventral striatum compared to healthy controls and whether DAT density was associated with key characteristics of GD (e.g., abstinence, craving). In a cross-sectional investigation using single-photon emission computed tomography with a technetium- 99 m-labeled tropane derivative as a radiotracer with SPECT imaging, fifteen participants with GD and 15 controls (non-gambling individuals, matched for age, gender, handedness, and smoking status) were measured. The GD group completed self-reported questionnaires regarding gambling. Striatal DAT density did not differ between the two groups. Conversely, striatal DAT density correlated significantly with various measures of recent gambling, but not with measures of chronic gambling. Multivariate analysis, adjusted for age and smoking status, showed that DAT density in the left striatum correlated positively with time spent gambling and gambling craving in the last month, whereas DAT density in the right striatum correlated negatively with abstinence self-efficacy. Fertilization in flowering plants requires a complex series of coordinated events involving interaction between the male and female gametophyte. We report here molecular data on one of the key events underpinning this process - the death of the receptive synergid cell and the coincident bursting of the pollen tube inside the ovule to release the sperm. We show that two REM transcription factors, VALKYRIE (VAL) and VERDANDI (VDD), both targets of the ovule identity MADS-box complex SEEDSTICK-SEPALLATA3, interact to control the death of the receptive synergid cell. In vdd-1/+ mutants and VAL_RNAi lines, we find that GAMETOPHYTIC FACTOR 2 (GFA2), which is required for synergid degeneration, is downregulated, whereas expression of FERONIA (FER) and MYB98, which are necessary for pollen tube attraction and perception, remain unaffected. We also demonstrate that the vdd-1/+ phenotype can be rescued by expressing VDD or GFA2 in the synergid cells. Taken together, our findings reveal that the death of the receptive synergid cell is essential for maintenance of the following generations, and that a complex comprising VDD and VAL regulates this event., (© 2016. Synchronous colorectal cancers (syCRCs) are physically separated tumours that develop simultaneously. To understand how the genetic and environmental background influences the development of multiple tumours, here we conduct a comparative analysis of 20 syCRCs from 10 patients. We show that syCRCs have independent genetic origins, acquire dissimilar somatic alterations, and have different clone composition. This inter- and intratumour heterogeneity must be considered in the selection of therapy and in the monitoring of resistance. SyCRC patients show a higher occurrence of inherited damaging mutations in immune-related genes compared to patients with solitary colorectal cancer and to healthy individuals from the 1,000 Genomes Project. Moreover, they have a different composition of immune cell populations in tumour and normal mucosa, and transcriptional differences in immune-related biological processes. This suggests an environmental field effect that promotes multiple tumours likely in the background of inflammation.

Chitin is a natural biopolymer and the second most abundant after cellulose. This polysaccharide can be found in the biomass in different polymorphic forms. Chitosan is one of the most important derivatives obtained from the deacetylation of chitin. In this work, Molecular Dynamics simulations of chitin and chitosan nanoparticles enabled us to evaluate their different conformation and solubility properties. The role of secondary metabolites in the determination of cell identity has been an area of particular interest over recent years, and studies strongly indicate a connection between cell fate and the regulation of enzymes involved in secondary metabolism. In Arabidopsis thaliana, the maternally derived seed coat plays pivotal roles in both the protection of the developing embryo and the first steps of germination. In this regard, a characteristic feature of seed coat development is the accumulation of proanthocyanidins (PAs - a class of phenylpropanoid metabolites) in the innermost layer of the seed coat. Our genome-wide transcriptomic analysis suggests that the ovule identity factor SEEDSTICK (STK) is involved in the regulation of several metabolic processes, providing a strong basis for a connection between cell fate determination, development and metabolism. Using phenotypic, genetic, biochemical and transcriptomic approaches, we have focused specifically on the role of STK in PA biosynthesis. Our results indicate that STK exerts its effect by direct regulation of the gene encoding BANYULS/ANTHOCYANIDIN REDUCTASE (BAN/ANR), which converts anthocyanidins into their corresponding 2,3-cis-flavan-3-ols. Our study also demonstrates that the levels of H3K9ac chromatin modification directly correlate with the active state of BAN in an STK-dependent way. This is consistent with the idea that MADS-domain proteins control the expression of their target genes through the modification of chromatin states. STK might thus recruit or regulate histone modifying factors to control their activity. In addition, we show that STK is able to regulate other BAN regulators. Our study demonstrates for the first time how a floral homeotic gene controls tissue identity through the regulation of a wide range of processes including the accumulation of secondary metabolites.

Cancer genetic heterogeneity offers a wide repertoire of molecular determinants to be screened as therapeutic targets. Here, we identify potential anticancer targets by exploiting negative genetic interactions between genes with driver loss-of-function mutations (recessive cancer genes) and their functionally redundant paralogs. We identify recessive genes with additional copies and experimentally test our predictions on three paralogous pairs. We confirm digenic negative interactions between two cancer genes (SMARCA4 and CDH1) and their corresponding paralogs (SMARCA2 and CDH3). Furthermore, we identify a trigenic negative interaction between the cancer gene DNMT3A, its functional paralog DNMT3B, and a third gene, DNMT1, which encodes the only other human DNA-methylase domain. Although our study does not exclude other causes of synthetic lethality, it suggests that functionally redundant paralogs of cancer genes could be targets in anticancer therapy., (Copyright © 2013 The Authors. MADS domain transcription factors are key regulators of eukaryotic development. In plants, the homeotic MIKC MADS factors that regulate floral organ identity have been studied in great detail. Based on genetic and protein-protein interaction studies, a floral quartet model was proposed that describes how these MADS domain proteins assemble into higher order complexes to regulate their target genes. However, despite the attractiveness of this model and its general acceptance in the literature, solid in vivo proof has never been provided. To gain deeper insight into the mechanisms of transcriptional regulation by MADS domain factors, we studied how SEEDSTICK (STK) and SEPALLATA3 (SEP3) directly regulate the expression of the reproductive meristem gene family transcription factor-encoding gene VERDANDI (VDD). Our data show that STK-SEP3 dimers can induce loop formation in the VDD promoter by binding to two nearby CC(A/T)6GG (CArG) boxes and that this is essential for promoter activity. Our in vivo data show that the size and position of this loop, determined by the choice of CArG element usage, is essential for correct expression. Our studies provide solid in vivo evidence for the floral quartet model.

Background: MADS-domain transcription factors play important roles during plant development. The Arabidopsis MADS-box gene SHORT VEGETATIVE PHASE (SVP) is a key regulator of two developmental phases. It functions as a repressor of the floral transition during the vegetative phase and later it contributes to the specification of floral meristems. How these distinct activities are conferred by a single transcription factor is unclear, but interactions with other MADS domain proteins which specify binding to different genomic regions is likely one mechanism., Results: To compare the genome-wide DNA binding profile of SVP during vegetative and reproductive development we performed ChIP-seq analyses. These ChIP-seq data were combined with tiling array expression analysis, induction experiments and qRT-PCR to identify biologically relevant binding sites. In addition, we compared genome-wide target genes of SVP with those published for the MADS domain transcription factors FLC and AP1, which interact with SVP during the vegetative and reproductive phases, respectively., Conclusions: Our analyses resulted in the identification of pathways that are regulated by SVP including those controlling meristem development during vegetative growth and flower development whereas floral transition pathways and hormonal signaling were regulated predominantly during the vegetative phase. Thus, SVP regulates many developmental pathways, some of which are common to both of its developmental roles whereas others are specific to only one of them.

The nuclear factor-Y (NF-Y), a trimeric, CCAAT-binding transcriptional activator with histone-like subunits, was until recently considered a prototypical promoter transcription factor. However, recent in vivo chromatin immunoprecipitation assays associated with microarray methodologies (chromatin immunoprecipitation on chip experiments) have indicated that a large portion of target sites (40%-50%) are located outside of core promoters. We applied the tethered particle motion technique to the major histocompatibility complex class II enhancer-promoter region to characterize i), the progressive compaction of DNA due to increasing concentrations of NF-Y, ii), the role of specific subunits and domains of NF-Y in the process, and iii), the interplay between NF-Y and the regulatory factor-X, which cooperatively binds to the X-box adjacent to the CCAAT box. Our study shows that NF-Y has histone-like activity, since it binds DNA nonspecifically with high affinity to compact it. This activity, which depends on the presence of all trimer subunits and of their glutamine-rich domains, seems to be attenuated by the transcriptional cofactor regulatory factor-X. Most importantly NF-Y-induced DNA compaction may facilitate promoter-enhancer interactions, which are known to be critical for expression regulation.

The behavior of male and female Mongolian gerbils was continuously video-recorded from 24 h before the parturition to the end of postpartum estrus. We noted that the mean interval between the delivery of the last pup and the first mounting was 13 h and 32 min. Over the whole duration of the postpartum estrus (7 h and 41+/-57 min), females spent significantly more time in crouching over, pup licking, nest building and "digging" activities, and were more recipient of allogrooming than males. Maternal pup retrievals were not very frequent (14.0+/-5.6 episodes), but males never retrieved pups nor exhibited a full sequence of nest building activity. Males spent a longer mean time in bodily interactions with females, as compared to the time they spent with pups, and engaged in intense copulatory activity (592.8+/-40.5 mounts or 1 episode per 46.7s); even during the delivery process males attempted to force copulation, but females rejected mounting in all cases. We conclude that females exhibited higher levels of parental care than males, and our findings suggest that males compete with pups in attracting the female's attention, since they actively disrupt maternal care or persistently persecute the female in order to copulate.

We investigated the effects of travel costs on transporting nest material and nest-building activity in golden hamsters. Nest-deprived animals were submitted to run alleys 30, 90 and 180 cm long to access a source containing paper strips as nest material (Experiment 1) or were submitted to the same travel costs in 24-h experimental sessions (Experiment 2). We noted that increased travel costs were related to a decreased number of trips to the source, larger amounts (cm(2)) of nest material transported per trip (although total loads also decreased in longer alleys), longer intervals between trips, and increased time spent at the source and in nest building activity. Foraging efficiency (i.e. size of load divided by the time spent at the source) decreased as a function of travel costs, and animals transported their loads in two fundamental ways: in 30-cm alleys, they simply used their mouth to pull the paper strips, but in 90- or 180-cm alleys they transported the loads in their cheek pouches. The animals were faster when returning to the home-cage and their running speed (cm/s) increased as a function of the length of the alley, showing that animals are under different environmental pressures when searching for resources and subsequently running back with the load to the nest. Both male and female subjects were sensitive to travel costs, but males engaged in nest building activity more promptly and exhibited higher mean performances in most measures. We conclude that nest material is a good reinforcer, and our major results are in accordance with the predictions of microeconomic and optimal foraging theories.

The effects of litter size on maternal care, body weight and infant development of golden hamsters were investigated from a longitudinal perspective. Litters were culled to 1,3,6 and 9 pups, and the behavior and body weight of mothers and pups were recorded from the 5th to the 25th postpartum day. We noted that the time spent by mothers in bodily interactions with pups decreased as a function of litter size; maternal pup retrievals reached their maximum around the 13-15th day, which coincided with the increased locomotor activity of pups at this time; the total number of pup retrievals by the mother increased as a function of the litter size, but mothers of larger litters were more 'efficient' (i.e. they failed less frequently in exhibiting a full sequence of retrievals) and exhibited a low litter-size proportional mean number of retrievals. All mothers gradually lost body mass throughout lactation, and decrease in body weight was significantly related to litter size. The mean body weight gain (%) by pups decreased as a function of litter size, but we also noted that single and larger litter pups exhibited a decreased body mass (grams) by the 15th day, suggesting that infant development may be impaired at both extremes of experimental conditions. We concluded that the behavior of mothers and pups was affected by the litter size, and it appeared that the litter had an optimal size-not so large as to overlap the mother's physical capacity, and not so small as to fail to compensate for the parental investment.

The effects of social deprivation schedules and days of testing on the play-fighting of juvenile golden hamsters were investigated. In Experiment 1, subjects totally (individually housed) or partially isolated (individually housed, but separated by a wire mesh 2 cm from another animal) for a period of 48 h spent significantly increased peer-peer bodily contact, play fighting, and play-fighting relative time (% of bodily contact time spent in play fighting), but exhibited decreased locomotor activity on the 1st day of testing. In Experiment 2, subjects reared since the 10th postnatal day with their mothers alone (chronically deprived of peer-peer social interactions) exhibited similar performances, except that locomotor activity was not affected. All dyads exhibited increased pinning turbulence (number of pinnings divided by time spent in play fighting) on the 1st day of testing, and the interactions between variables (deprivation schedules and days of testing) were statistically significant in most cases, suggesting that unfamiliarity between interactors also affect the performances of animals. We conclude that golden hamsters are sensitive to social deprivation, mothers do not substitute peer companions, animals reared in single-infant litters basically behave like socially-isolated animals, and the effects of induced social deprivation are reversible and disappear on days following testing.

Cross-fostering technique can be defined as adoption of infants by adults of other species. This phenomenon is poorly investigated because very young animals have few opportunities to interact peacefully with non-conspecific adults, either in captivity or in natural conditions. This study describes the induction of cross-fostering in captivity between white tufted-ear (Callithrix jacchus) and black tufted-ear marmosets (Callithrix penicillata). We conclude that this technique can be very useful for preserving the life of rejected by parents or orphan infants, mainly in the case of species with low reproduction rate in captivity or those threatened by extinction, as well for investigating the environmental effects on the typical behavior of species (courtship, food preference, vocalization patterns, e.g.).

Hand preference in two adult C. jacchus (one male and one female) and five C. penicillata (three adult females and two 70-day-old male and female infants) was examined in this study. Animals were housed in large wire mesh cages and daily tested twice during 40 consecutive days in 15-minute experimental sessions. Tests were carried out with dog food pellets and were as follows: 1) food taking from a glass cylinder (12 cm long x 5 cm diameter), 2) food taking from an opaque plastic flask (11 cm long x 9.5 cm diameter) behind wire mesh, 3) food taking under the cage floor and 4) spontaneous activity (food manipulation, self-grooming and scratching behavior, and grasping wire mesh). It was observed that 1) subjects exhibit a high manual ability in all testing conditions, 2) hand biases occurred, but the subjects either reversed hand preference or the preference was ambiguous as the performances are examined in a longitudinal perspective, 3) hand biases were clearly more visible in test 1, which may be due to the complexity of this task itself, 4) hand biases were not similar in all testing conditions--i.e., subjects were left--in one test and right-handed in another-, and 5) the main results were very similar in both C. jacchus and C. penicillata and seem to be not dependent on either sex or age.

Play fighting of homosexual and heterosexual dyads in golden hamsters was studied. The behaviors of male-male (MM), female-female (FF), and female-male (FM) dyads were recorded from the 25th to the 31st day of age. Play fighting activity of heterosexual dyads was depressed since MF and FM dyads exhibited a significantly high physical contact time and engaged in less play fighting, with a significantly lower time spent on such activity. Alternation ratio in the pinning posture was very high and subjects did not differ in the mean number of active (top) and passive pinning (under) in each dyad, showing an inexistence of sex dominance. In most cases, differences in the performances of MM and FF dyads were not significant. We have concluded that play fighting in golden hamsters may be predominantly modulated by environmental stimuli (sex of the partner) rather than by endogenous stimuli (androgen level).

The effects of bilateral optic enucleation on play fighting in golden hamster infants were studied. The behaviors of blind-blind (BB), blind-intact (BI) and intact-intact (II) dyads were recorded from the 30th to the 36th day of age. It has been noted that the differences in physical contact, play fighting and grooming time, as well as in the percentage of time spent in play fighting, the amount of locomotion and other activities of BB, BI and II were small and statistically insignificant. The mean number of active and passive pinning in all groups was equivalent and the alternation of active and passive pinning, or vice versa, was around 50%, showing that posture during play fighting is not an indicator of dominance. It has been concluded that vision is not important for either the initiation or the modulation of play fighting in golden hamsters.
