Nutrient import into eutrophic estuarine-coastal zones is modulated by river discharge and anthropogenic activities (agriculture, industry, sewage, etc.). Comparing anthropogenic vs. hydrological influences on nutrient loading is important for ecosystem managers but has been a long-standing challenge. As an attempt to fill this gap, a retrospective study was conducted using multiyear (2012–2022) data in one of the most eutrophic coastal seas in China, the Jiangsu coastal waters. Of the two main pollutants on the Jiangsu coast, dissolved inorganic nitrogen (DIN) is extremely overloaded relative to phosphate (PO4). Their spatial distribution shows clear terrestrial sources as concentrations decrease offshore. Although in each survey DIN and PO4 are enriched in low-salinity waters, their spatially averaged concentrations are out of phase with river discharge on the seasonal and interannual scales. A box model was applied to decompose contributions of river discharge and nutrient import by anthropogenic activities. Results show that the human impacts on the interannual nutrient variability exceed those of hydrological factors on the Jiangsu coast. Anthropogenic nutrient loading decreases during 2012–2020, paralleled with the reduced fertilizer use and other nonpoint sources. Sporadic nutrient fluxes from point sources are likely responsible for the aseasonal nutrient-rich patches. Our study demonstrates that water quality in the Jiangsu coastal waters is highly affected by anthropogenic activities. Findings in this study have broad implications for local monitoring strategies and restoration efforts and provide insight into assessing human stress from field data in global eutrophic ecosystems.

Based on satellite remote sensing dataset and survey data during upwelling season of 2015, the spatial structures of phytoplankton biomass and community for the first time in the eastern Hainan upwelling (EHU) and its adjacent area, the eastern Leizhou Peninsula upwelling (ELPU) were illustrated. It is found that a significant cold tongue with high salinity and low temperature along the eastern Hainan coast driven by upwelling-favorable summer monsoon. The ELPU was relative weaker than the EHU because of its wide and gentle continental slope. Due to mixing by tides and waves, DO concentration with high value (>6.0 mg/L) were almost homogenous from surface to 30 m depth at the EHU. Beneath that, low DO water (

Vertical variability in the bio-optical properties of seawater in the northern South China Sea (NSCS) including inherent optical properties (IOPs) and chlorophyll a concentration (Chl) were studied on the basis of in situ data collected in summer 2008 using an absorption/attenuation spectrophotometer. An empirical model was developed to estimate Chl profiles based on the absorption line height at long wavelengths, with a relative root mean square error of 37.03%. Bio-optical properties exhibited large horizontal and vertical spatial variability. As influenced by coastal upwelling and the Zhujiang River (Pearl River) discharge, both IOPs and Chl exhibited high values in the surface waters of the inner shelf, which tended to decrease with distance offshore. Subsurface maximum layers of IOPs and Chl were observed in the middle and outer shelf regions, along with significantly higher values of attenuation coefficients beneath this layer that rapidly increased towards the bottom. In the open ocean, both IOPs and Chl exhibited consistent variability, with the subsurface maximum layer typically located at 34–84 m. Phytoplankton were found to be one of the major components in determining the vertical variability of bio-optical properties, with their vertical dynamics influenced by both physical forcing and light attenuation effects. The depth of the subsurface maximum layer was found to be closely related to the fluctuation of the oceanic thermocline and the depth of the euphotic zone, which also affected the total integrated biomass of the upper ocean. Typically high values of attenuation coefficients observed in the bottom waters of the continental shelf reflected the transport of particulate matter over the bottom boundary layer. Our results reveal large spatial differences in bio-optical profiles in response to complex marine ecodynamics in the NSCS. From the perspective of marine research, high-resolution optical measurements are clearly advantageous over conventional bottle sampling.

The aim of this study was to identify the symptom burden of perioperative oral cancer patients, its trajectory, and the factors influencing it.A longitudinal, repeated measures design with consecutively identified sampling was used to recruit oral cancer patients scheduled for surgical treatment. Data collected included sociodemographic and clinical information, nutritional risk by the Nutritional Risk Screening 2002, and symptom burden by M. D. Anderson Symptom Inventory-Head and Neck Module (MDASI-HN) at preoperation, 7 days postsurgery, and 1 month postsurgery.Perioperative patients with oral cancer had multiple symptoms. Pain, difficulty swallowing/chewing, and mouth/throat sores (61.9%-76.1%) were the most prevalent symptoms before surgery. The symptom burden was the highest at 7 days after surgery, with the most prevalent symptoms, including difficulty swallowing/chewing, difficulty with voice/speech, and problems with mucus (87.8%-95.4%). At 1 month postsurgery, the 3 main symptoms were numbness or tingling, difficulty swallowing/chewing, and difficulty with voice/speech (all 87.8%). Treatment stage, job, comorbidity, cancer stage, adjuvant therapy, and Nutritional Risk Screening 2002 score were correlated with symptom burdens.Our study illustrates that perioperative oral cancer patients have multiple symptoms and high symptom burdens, especially at 7 days postsurgery, with prominent symptoms and symptom burdens varying with the treatment stage.

This paper briefly introduces the concept, classification, and mechanism of action of positive thinking therapy; reviews the application and research progress of positive thinking therapy in perioperative care of surgical patients at home and abroad; presents the shortcomings and defects in the development; and aims to provide intervention, reference, and basis for the development of positive thinking therapy in perioperative care of surgical patients. One hundred and eight patients are undergoing PCI surgery in our cardiology department; 50 patients undergoing percutaneous coronary intervention were selected as the control group, and 58 patients undergoing percutaneous coronary intervention were selected as the observation group. Traditional health education was employed in the control group, while empowerment education based on timing theory was used in the observation group. The two groups were observed and compared in terms of self-care competence, anxiety and depression, medication adherence score, and exercise adherence. Conclusion. Empowerment education based on timing theory can improve self-care ability of PCI patients, reduce patients’ anxiety and depression, and improve patients’ medication and exercise compliance.

The standing stock of phytoplankton carbon is a basic and essential property for understanding oceanic ecosystems, biogeochemical cycles, and regional climates. However, current related algorithms mainly focus on remote-sensed application, which cannot describe the vertical profile of phytoplankton carbon throughout the whole euphotic zone. In this study, we modified a previous absorption-based bio-optical algorithm to acquire vertical variabilities of the total and size-partitioned phytoplankton carbon based on field data from the South China Sea (SCS). The mean absolute errors and the biases between estimated and field picophytoplankton carbon were SCS basin. The picophytoplankton carbon was always the fundamental component of the total phytoplankton carbon within the whole euphotic zone. The dominant picophytoplankton species changed from Synechococcus-like cyanobacteria at the sea surface to pico-sized haptophytes at the phytoplankton carbon maximum layer. The strong covariation between total phytoplankton carbon and chlorophyll-a concentration suggested that they can be converted into each other through an accurate carbon-to-chlorophyll ratio in the open SCS. These results provide essential information that can be used to decipher the three-dimensional structure of total and size-partitioned phytoplankton carbon in the open SCS.

Phosphorus in the soil accessible to plants can easily be combined with calcium ion, the content of which is high in karst rocky desertification (KRD) regions, thereby resulting in a low utilization efficiency of phosphorus. The application of phosphate-solubilizing bacteria (PSB) from the KRD region would facilitate enhanced phosphate availability in the soil. In the present study, the strains belonging to Acinetobacter, Paraburkholderia, and Pseudomonas with efficient phosphate-solubilizing ability were isolated from fruit tree rhizosphere soils in KRD regions. Particularly, Acinetobacter sp. Ac-14 had a sustained and stable phosphate-solubilizing ability (439–448 mg/L, 48–120 h). Calcium carbonate decreased the phosphate-solubilizing ability in liquid medium; however, it did not affect the solubilization index in agar-solidified medium. When cocultivated with Arabidopsis thaliana seedling, Ac-14 increased the number of lateral roots, fresh weight, and chlorophyll content of the seedlings. Metabolomics analysis revealed that Ac-14 could produce 23 types of organic acids, majorly including gluconic acid and D-(-)-quinic acid. Expression of Ac-14 glucose dehydrogenase gene (gcd) conferred Pseudomonas sp. Ps-12 with a sustained and stable phosphate-solubilizing ability, suggesting that the production of gluconic acid is an important mechanism that confers phosphate solubilization in bacteria. Moreover, Ac-14 could also produce indole acetic acid and ammonia. Collectively, the isolated Ac-14 from KRD regions possess an efficient phosphate-solubilizing ability and plant growth-promoting effect which could be exploited for enhancing phosphorus availability in KRD regions. This study holds significance for the improvement of soil fertility and agricultural sustainable development in phosphorus-deficient KRD regions.

Increasing evidence has supported the crucial role of CARD14 in the pathogenesis of psoriasis, whereas the precise cellular signaling involved in skin physiopathology remains poorly understood. In this article, we show that neither genetic ablation of Il17a nor elimination of T cells was sufficient to restrain the skin inflammation in a CARD14-E138A-mutation-induced psoriasis-like mouse model, whereas depletion of Il23, which extremely blocked the IL-23/T17 axis, was more effective. Targeting CBM complex by conditional deletion of MALT1 or BCL10 in keratinocytes abrogated both the cutaneous and systemic inflammation of heterozygous Card14E138A/+ mice. Selective inactivation of keratinocyte-specific MALT1 proteolytic activity strongly ameliorated the Card14E138A/+- and Card14ΔQ136/+-induced skin disease, which was reproduced by using the imiquimod-induced mouse model. Together, our results suggest a sequence of events under CARD14-mutation-induced psoriasis condition that keratinocyte-intrinsic activation of CBM complex initiates the skin inflammation depending on the IL-23/T17 axis. Targeting keratinocytes by inactivation of MALT1 paracaspase activity might be a promising therapeutic target for early psoriasis treatment.

The light scattering and backscattering coefficients of particles (bp( λ ) and bbp( λ )) and their association with particle concentration, size distribution, and composition off the eastern Hainan coast in the South China Sea (SCS) were analyzed between August and September 2013. Variations in bp(532) and bbp(532) were correlated to changes in the LISST-based total cross-sectional area and volume concentration of particles (AC and VC). Two significantly different covariations between bbp( λ ) and bp( λ ) and chlorophyll a concentration (Chla) were observed, and all samples were clearly separated into two groups (Group A and Group B) using the threshold relationship between bbp and Chla. Samples in Group B, most of which were collected in the nepheloid layer, showed higher bp/AC and bbp/AC than those in Group A. This indicates the presence of inorganic particles with higher np in Group B than in Group A. The relationship between bbp and Chla for Group A could be well fitted by a published model involving a constant backscattering background independent of Chla. A significant relationship (R2 = 0.85) was observed between bbp and Chla in the Chla maximum layer, which was different from that in other layers. This finding might imply that both an increase in the micro-phytoplankton community and a decrease in the contribution of background backscattering to bbp at the Chla maximum layer. Understanding this variability of beam-scattering properties off the eastern Hainan coast can improve our understanding of marine biogeochemical processes at a regional scale, and can also promote the application of these regional bio-optical relationships for ocean color.

Typhoons can affect various hydrodynamic processes, such as upwelling, vertical mixing, and entrainment, which in turn influence the growth and redistribution of phytoplankton. In this study, responses of phytoplankton to consecutive typhoons Barijat and Mangkhut in summer 2018 in the northern South China Sea (NSCS) were investigated through combining remote sensing, numerical simulation, and Argo profile data. Variations of physical and biological properties induced by Barijat and Mangkhut over the study region were observed. Regional chlorophyll-a (Chla) algorithms over the NSCS were assessed in order to accurately quantify typhoon-induced variations of phytoplankton. Chla concentration decreased slightly over the region of interest after the passage of Barijiat while increased obviously after the passage of Mangkhut. Satellite-derived Chla increased by ~63% on average after the consecutive typhoons. Responses of phytoplankton size structure (PSS) were further investigated over the continental shelf and the deep ocean in the NSCS. For the continental shelf, the increment of micro- and nano-phytoplankton was about 1.5-fold as high as that of pico-phytoplankton, compared with a 0.5-fold increment for the deep ocean. This study attested the discrepant responses of different phytoplankton size classes to typhoons. It was of great significance to expand our understanding of ocean physical and ecological responses to extreme phenomena. Our finding underscores the potential of remote sensing to investigate typhoon-induced variation of PSS and could be the key to accurately assess typhoon-induced carbon fluxes, primary production, and potential fishing ground in the future.

The phytoplankton size class (PSC) plays an important role in biogeochemical processes in the ocean. In this study, a regional model of PSCs is proposed to retrieve vertical PSCs from the total minus water absorption coefficient (at-w(λ)) and Chlorophyll a concentration (Chla). The PSC model is developed by first reconstructing phytoplankton absorption and Chla from at-w(λ), and then extracting PSC from them using the support vector machine (SVM). In situ bio-optical data collected in the South China Sea from 2006 to 2013 were used to train the SVM. The proposed PSC model was subsequently validated using an independent PSC dataset from the Northeast South China Sea Cruise in 2015. The results indicate that the PSC model performed better than the three components model, with a value of r2 between 0.35 and 0.66, and the absolute percentage difference between 56% and 181%. On the whole, our PSC model shows a remarkable utility in terms of inferring vertical PSCs from the South China Sea.

The diurnal variation of the diffuse attenuation coefficient for downwelling irradiance at 490 nm (Kd(490)) has complex characteristics in the coastal regions. However, owing to the scarcity of in situ data, our knowledge on the diurnal variation is inadequate. In this study, an optical-buoy dataset was used to investigate the diurnal variation of Kd(490) in the coastal East China Sea, and to evaluate the Kd(490) L2 products of geostationary ocean color imager (GOCI), as well as the performance of six empirical algorithms for Kd(490) estimation in the Case-2 water. The results of validation show that there was high uncertainty in GOCI L2 Kd(490), with mean absolute percentage errors (MAPEs) of 69.57% and 68.86% and root mean square errors (RMSEs) of 0.70 and 0.71 m−1 compared to buoy-measured Kd12(490) and Kd13(490), respectively. Meanwhile, with the coefficient of determination (R2) of 0.71, as well as the lowest MAPE of 27.31% and RMSE of 0.29 m−1, the new dual ratio algorithm (NDRA) performed the best in estimating Kd(490) in the target area, among the six algorithms. Further, four main types of Kd(490) diurnal variation were found from buoy data, showing different variabilities compared to the area closer to the shore. One typical diurnal variation pattern showed that Kd(490) decreased at flood tide and increased at ebb tide, which was confirmed by GOCI images through the use of NDRA. Hydrometeorological factors influencing the diurnal variations of Kd(490) were also studied. In addition to verifying the predominant impact of tide, we found that the dominant effect of tide and wind on the water column is intensifying sediment resuspension, and the change of sediment transport produced by them are secondary to it.

Quantitative estimates of particulate matter [ PM) concentration in sea ice using remote sensing data is helpful for studies of sediment transport and atmospheric dust deposition flux. In this study, the difference between the measured dirty and estimated clean albedo of sea ice was calculated and a relationship between the albedo difference and PM concentration was found using field and laboratory measurements. A semianalytical algorithm for estimating PM concentration in sea ice was established. The algorithm was then applied to MODIS data over the Bohai Sea, China. Comparisons between MODIS derived and in situ measured PM concentration showed good agreement, with a mean absolute percentage difference of 31.2%. From 2005 to 2010, the MODIS-derived annual average PM concentration was approximately 0.025 g/L at the beginning of January. After a month of atmospheric dust deposition, it increased to 0.038 g/L. Atmospheric dust deposition flux was estimated to be 2.50 t/km(2)/month, similar to 2.20 t/km(2)/month reported in a previous study. The result was compared with on-site measurements at a nearby ground station. The ground station was close to industrial and residential areas, where larger dust depositions occurred than in the sea, but although there were discrepancies between the absolute magnitudes of the two data sets, they demonstrated similar trends.

Medium Resolution Imaging Spectrometer MERIS, Moderate Resolution Imaging Spectroradiometer MODIS, and Sea-viewing Wide Field-of-view Sensor SeaWiFS particulate organic carbon POC concentration products for the South China Sea SCS were compared with in situ data collected from October 2007 to December 2013. Spectral remote-sensing reflectance Rrs,λ was also measured to help understand POC algorithm performance. A strict comparison of the satellite-derived POC and in situ measurements showed that MERIS, MODIS, and SeaWiFS underestimated in situ values by 29.1, 11.7, and 31.5%, respectively. Similar results were obtained with a relaxed matching criterion. Through analysis of the causes of product uncertainty, the results suggested that satellite retrieval of Rrs,λ and the global POC algorithm both have an impact on inversion accuracy. However, the formulation of the POC algorithm seems to be more critical. When a regional algorithm was developed to obtain satellite-derived POC, both the strict and relaxed comparison results showed significant improvement, but for coastal waters, both algorithms had larger errors. Other factors affecting the comparison are also discussed.

The spatiotemporal variability of phytoplankton biomass has been widely studied because of its importance in biogeochemical cycles. Chlorophyll a (Chl-a)—an essential pigment present in photoautotrophic organisms—is widely used as an indicator for oceanic phytoplankton biomass because it could be easily measured with calibrated optical sensors. However, the intracellular Chl-a content varies with light, nutrient levels, and temperature and could misrepresent phytoplankton biomass. In this study, we estimated the concentration of phytoplankton carbon—a more suitable indicator for phytoplankton biomass—using a regionally adjusted bio-optical algorithm with satellite data in the South China Sea (SCS). Phytoplankton carbon and the carbon-to-Chl-a ratio (θ) exhibited considerable variability spatially and seasonally. Generally, phytoplankton carbon in the northern SCS was higher than that in the western and central parts. The regional monthly mean phytoplankton carbon in the northern SCS showed a prominent peak during December and January. A similar pattern was shown in the central part of SCS, but its peak was weaker. Besides the winter peak, the western part of SCS had a secondary maximum of phytoplankton carbon during summer. θ exhibited significant seasonal variability in the northern SCS, but a relatively weak seasonal change in the western and central parts. θ had a peak in September and a trough in January in the northern and central parts of SCS, whereas in the western SCS the minimum and maximum θ was found in August and during October–April of the following year, respectively. Overall, θ ranged from 26.06 to 123.99 in the SCS, which implies that the carbon content could vary up to four times given a specific Chl-a value. The variations in θ were found to be related to changing phytoplankton community composition, as well as dynamic phytoplankton physiological activities in response to environmental influences; which also exhibit much spatial differences in the SCS. Our results imply that the spatiotemporal variability of θ should be considered, rather than simply used a single value when converting Chl-a to phytoplankton carbon biomass in the SCS, especially, when verifying the simulation results of biogeochemical models.

The energetic mesoscale and submesoscale processes in the Southwest Indian Ocean (SWIO) have a significant effect on phytoplankton growth revealed by satellite observations and numerical simulations. Based on observations from two Bio-Argo floats deployed in the SWIO, a prominent phenomenon of subsurface Chl-a maximum (SCM) is observed in this region during spring, summer and autumn. After comparing the Chl-a profiles with and without mixed layer restratification (ML restratification), we found that the ML restratification favors to increase the upper ocean Chl-a concentration in spring, autumn and winter, while decrease Chl-a concentration in summer, which could be explained by nutrient-light colimitation on growth rates. Further analysis imply ML restratification is likely caused by the surface heating and up-front wind-driven Ekman transport.

Using large amounts of bio-optical data collected in the South China Sea (SCS) from 2003 to 2016, this study checks the consistency between well-known semi-analytical algorithms (SAAs)—the quasi-analytical algorithm (QAA) and the default generalized inherent optical property (GIOP-DC)—in retrieving the non-water absorption coefficient (anw(λ)), phytoplankton absorption coefficient (aph(λ)) and particulate backscattering coefficient (bbp(λ)) from remote-sensing reflectance (Rrs(λ)) data at 412, 443, 490, 531, and 555 nm. The samples from the SCS are further separated into oligotrophic and mesotrophic water types for the comparison of the SAAs. Several findings are made: First, the values of anw(λ) derived from the two SAAs deliver similar performance, with R2 values ranging from 0.74 to 0.85 and 0.74 to 0.87, implying absolute percent error differences (APDs) from 37.93% to 74.88% and from 32.32% to 71.75% for the QAA and GIOP-DC, respectively. The QAA shows a value of R2 between 0.64 and 0.91 and APDs between 43.57% to 83.53%, while the GIOP-DC yields R2 between 0.76 to 0.89 and APDs between 44.65% to 79.46% when estimating aph(λ). The values of bbp(λ) derived from the QAA are closer to the in-situ bbp(λ) values, as indicated by the low values of the normalized centered root-mean-square deviation and normalized standard deviation, which are close to one. Second, a regionally tuned estimation of aph(λ) is proposed and recommended for the SCS. This consistency check of inherent optical properties products from SAAs can serve as reference for algorithm selection for further applications, including primary production, carbon, and biogeochemical models of the SCS, and can provide guidance for improving aph(λ) estimation.

An empirical algorithm is proposed to estimate suspended particulate matter (SPM) ranging from 0.675 to 25.7 mg L-1 in the turbid Pearl River estuary (PRE). Comparisons between model predicted and in situ measured SPM resulted in R2s of 0.97 and 0.88 and mean absolute percentage errors (MAPEs) of 23.96% and 29.69% by using the calibration and validation data sets, respectively. The developed algorithm demonstrated the highest accuracy when compared with existing ones for turbid coastal waters. The diurnal dynamics of SPM was revealed by applying the proposed algorithm to reflectance data collected by a moored buoy in the PRE. The established algorithm was implemented to Hyperspectral Imager for the Coastal Ocean (HICO) data and the distribution pattern of SPM in the PRE was elucidated. Validation of HICO-derived reflectance data by using concurrent MODIS/Aqua data as a benchmark indicated their reliability. Factors influencing variability of SPM in the PRE were analyzed, which implicated the combined effects of wind, tide, rainfall, and circulation as the cause.

The presence of foam influences the accuracy of satellite-derived water-leaving radiance. A model has been developed to estimate the augmented reflectance ratio (A(λ,U)) due to differences in the fraction of whitecap coverage (w) on the ocean surface. A(λ,U) can be calculated from the product of w and ρ(λ,U), where ρ(λ,U) is the augmented ratio of the reflectance of background water (Rb(λ)) caused by the presence of whitecaps. Our results showed that the average A(400~700,U) in the visible region was approximately 1.3% at U = 9 m∙s−1, 2.2% at U = 10 m∙s−1, 4.4% at U = 12 m∙s−1, 7.4% at U = 14 m∙s−1, 19% at U = 19 m∙s−1 and 37.9% at U = 24 m∙s−1, making it is necessary to consider the augmented reflectance ratio for remote sensing applications. By estimating remote sensing augmented reflectance using A(λ,U), it was found that the result was in good agreement with previous studies conducted in other areas with U from 9 to 12 m∙s−1. Since Rb(λ) is temporally and spatially variable, our model considered the variation of Rb(λ), whereas existing models have assumed that Rb(λ) is constant. Therefore, the proposed model is more suitable for estimating the augmented reflectance ratio due to whitecaps.

We examined regional empirical equations for estimating the surface concentration of particulate organic carbon (POC) in the South China Sea. These algorithms are based on the direct relationships between POC and the blue-to-green band ratios of spectral remotely sensed reflectance, Rrs (λB)/Rrs(555). The best error statistics among the considered formulas were produced using the power function POC (mg/m3)=262.173 [Rrs(443)/Rrs(555)]−0.940. This formula resulted in a small mean bias of approximately −2.52%, a normalized root mean square error of 31.1%, and a determination coefficient of 0.91. This regional empirical equation is different to the results of similar studies in other oceanic regions. Our validation results suggest that our regional empirical formula performs better than the global algorithm, in the South China Sea. The feasibility of this band ratio algorithm is primarily due to the relationship between POC and the green-toblue ratio of the particle absorption coefficient. Colored dissolved organic matter can be an important source of noise in the band ratio formula. Finally, we applied the empirical algorithm to investigate POC changes in the southwest of Luzon Strait.

Ubiquitination of proliferating cell nuclear antigen (PCNA) plays an important role in DNA damage response. Ectopic expression of PCNA fused at either terminus with ubiquitin (Ub) lacking two C-terminal glycine residues induces translesion DNA synthesis which resembles synthesis mediated by PCNA monoubiquitination. PCNA fused with Ub containing the C-terminal Gly residues at the C-terminus can be further polyubiquitinated in a Gly-dependent manner, which inhibits cell proliferation and induces ATR-dependent replication checkpoint. In this study, we surprisingly found that PCNA fused to a head-to-tail linear Ub chain induces apoptosis in a Ub chain length-dependent manner. Further investigation revealed that the apoptotic effect is actually induced by the linear Ub chain independently from PCNA, as the Ub chain fused to GFP or an epitope tag still efficiently induces apoptosis. It is revealed that the artificial linear Ub chain differs from endogenously encoded linear Ub chains in that its Ubs contain a Ub-G76S substitution, making the Ub chain resistant to cleavage by deubiquitination enzymes. We demonstrated in this study that ectopic expression of the artificial Ub chain alone in cultured human cancer cells is sufficient to inhibit tumor growth in a xenograft mouse model, making the linear Ub chain a putative anti-cancer agent.

The canonical Wnt/β-Catenin signaling pathway is widely involved in regulating diverse biological processes. Dysregulation of the pathway results in severe consequences, such as developmental defects and malignant cancers. Here, we identified Ube2s as a novel activator of the Wnt/β-Catenin signaling pathway. It modified β-Catenin at K19 via K11-linked polyubiquitin chain. This modification resulted in an antagonistic effect against the destruction complex/β-TrCP cascade-orchestrated β-Catenin degradation. As a result, the stability of β-Catenin was enhanced, thus promoting its cellular accumulation. Importantly, Ube2s-promoted β-Catenin accumulation partially released the dependence on exogenous molecules for the process of embryonic stem (ES) cell differentiation into mesoendoderm lineages. Moreover, we demonstrated that UBE2S plays a critical role in determining the malignancy properties of human colorectal cancer (CRC) cells in vitro and in vivo. The findings in this study extend our mechanistic understanding of the mesoendodermal cell fate commitment, and provide UBE2S as a putative target for human CRC therapy.

Recent advances in hydrologic optics provide a potential tool for extracting maps of optical properties of optically significant constituents. During summer experiments in the northern South China Sea, a procedure for inverting optical properties in optically complex coastal upwelling waters was performed. We tested an optical inversion model (OIM) that provides estimates of the absorption attributable to optically significant constituents (including phytoplankton, non-algal particles (NAP), colored dissolved organic matter (CDOM)), as well as the chlorophyll a concentration and the phytoplankton size class, from the total spectral absorption. The OIM provided reasonable estimates of these bio-optical products, and attempts to invert the bio-optical properties in the coastal upwelling were successful. Several bio-optical parameters displayed robust relationships to the hydrologic characteristics. The biomass was strongly enhanced in the upwelling, where the phytoplankton population was dominated by the microphytoplankton. The inherent optical properties in the surface waters delivered distinct responses to the coastal upwelling. The distribution of backscattering ratios and particulate size distribution slopes indicated that the offshore surface waters were mainly dominated by small organic NAP, while the upwelling surface water was characterized by larger particles. We propose a bio-optical inversion model that retrieves the absorption contributions of phytoplankton and colored detrital matter (CDM), as well as the phytoplankton size classes (PSCs), from total minus water absorption spectra. The model is based on three-component separation of phytoplankton size structure and a genetic algorithm. The model performance was tested on two independent datasets (the NASA bio-Optical Marine Algorithm Dataset (NOMAD) and the northern South China Sea (NSCS) dataset). The relationships between the estimated and measured values were strongly linear, especially for aCDM (412), and the Root Mean Square Error (RMSE) of the CDM exponential slope (SCDM) was relatively low. Next, the inversion model was directly applied to in-situ total minus water absorption spectra determined by an underwater meter during a cruise in September 2008, to retrieve the phytoplankton size structure in the seawater. By comparing the measured and retrieved chlorophyll a concentrations, we demonstrated that total and size-specific chlorophyll a concentrations could be retrieved by the model with relatively high accuracy. Finally, we applied the bio-optical inversion model to investigate changes in phytoplankton size structure induced by an anti-cyclonic eddy in the NSCS.

A previously developed model was modified to derive three phytoplankton size classes (micro-, nano-, and pico-phytoplankton) from the overall chlorophyll-a concentration, assuming that each class has a specific absorption coefficient. The modified model performed well using in-situ data from the northern South China Sea, and the results were reliable and accurate. The relative errors of the size-fractioned chlorophyll-a concentration for each size class were: micro-: 21%, nano-: 41%, pico-: 26%, and nano+pico: 23%. The model was then applied on ocean color remote sensing data to examine the distribution and variation of phytoplankton size classes in northern South China Sea on a large scale.

In eukaryotic cells, Rev7 interacts with Rev3 and functions as a regulatory subunit of Polζ, a translesion DNA synthesis (TLS) polymerase. In addition to its role in TLS, mammalian Rev7, also known as Mad2B/Mad2L2, participates in multiple cellular activities including cell cycle progression and double-strand break repair through its interaction with several proteins. Here we show that in mammalian cells, Rev7 undergoes ubiquitin/proteasome-mediated degradation upon UV irradiation in a time-dependent manner. We identified the Rev7 N-terminal destruction box as the degron and Cul4A/B as putative E3 ligases in this process. We also show that the nucleotide excision repair (NER) pathway protein HR23B physically interacts and colocalizes with Rev7 in the nuclear foci after UV irradiation and protects Rev7 from accelerated degradation. Furthermore, a similar Rev7 degradation profile was observed in cells treated with the UV-mimetic agent 4-nitroquinoline 1-oxide but not with cisplatin or camptothecin, suggesting a role of the NER pathway protein(s) in UV-induced Rev7 degradation. These data and the observation that cells deficient in Rev7 are sensitized to UV irradiation while excessive Rev7 protects cells from UV-induced DNA damage provide a new insight into the potential interplay between TLS and NER.

Seagrass in optically shallow water can generate optical signals that can be tracked remotely. Unfortunately the signals from the bottom are relatively weak and can be affected by the water column when concentrations of suspended particles, chlorophyll and coloured dissolved organic matter are high. An optical model simulating the propagation of light for retrieving the bottom reflectance was developed. Implementation of the method was found to be effective for improving the accuracy of coastal habitat maps, and essential for deriving empirical relationships between remotely sensed data and interesting features in the marine environment. The appropriate wavebands for seagrass mapping, which generally lay between 500 and 630 nm and 680 and 710 nm, were obtained by means of full visual inspection and analysis of the correct spectra. Additionally, a strong relationship between the reflectance value at 715 nm and Leaf Area Index was found, with a correlation coefficient of 0.99.

Internal waves can bring nutrients to the upper level of water bodies and facilitate phytoplankton photosynthesis. Internal waves occur frequently in the northern portion of the South China Sea and inflict an important effect on chlorophyll a distribution. In this study, in-situ observation and satellite remote sensing data were used to study the effects of internal waves on chlorophyll a distribution. Based on the in-situ observations, lower chlorophyll a concentrations were present in the middle and bottom level in areas in which internal waves occur frequently, while the surface chlorophyll a distribution increased irregularly, and a small area with relatively higher chlorophyll a concentrations was observed in the area around the Dongsha Island. Satellite remote sensing showed that the chlorophyll a concentration increased in the area near Dongsha Island, where internal waves frequently occurred. The results of the increased chlorophyll a concentration in the surface water near Dongsha Island in the northern portion of the South China Sea indicated that internal waves could uplift phytoplankton and facilitate phytoplankton growth.

From 15 to 28 August in 2007, a Chaetoceros socialis bloom was detected in the Pearl River Estuary water with chlorophyll a concentration (Chl a) up to 30 mg m(-3) and cell density up to 10(6) cells L-1. Time series of bio-optical measurements was obtained at a single site (114.29 degrees E, 22.06 degrees N) with the mooring of marine optical buoy. Light absorption properties of seawater experienced large variability throughout the algal bloom. Absorption by colored dissolved organic matter (CDOM) was one of the dominant optical components of the light absorption (30-70%) especially for pre- and post-bloom waters, and it tended to decrease with Chl a during the algal bloom. Absorption by phytoplankton was another dominant optical component (18-50%) and increased rapidly with Chl a. Phytoplankton and accompanying material played dominant roles in light absorption as indicated by the relationship between absorption coefficient and Chl a. At high pigment concentrations, water samples showed significantly lower specific phytoplankton absorption, compared with pre- and post-bloom conditions, with the specific phytoplankton concentration at 443 nm varied between 0.011 and 0.022 m(2) mg(-1) and that at 676 nm between 0.007 and 0.018 m(2) mg(-1); small values of blue-to-red ratio of phytoplankton were also observed. These lower values were associated with variations in phytoplankton size structure. Spectral variability of phytoplankton absorption and total absorption (not including the fixed background absorption by pure water itself) could be expressed as simple linear functions linking absorption at one wavelength to the absorption at the other wavelengths, with the slope of the relationship changing with wavelength. The absorption coefficients by non-algal particles and CDOM follow the general exponential functions with remarkably limited variability in the exponent with means of 0.0105 and 0.0166 nm(-1), respectively. These spectral dependencies of absorption coefficients provide useful information for retrieving inherent optical properties from reflectance data in a remote-sensing context. (C) 2010 Elsevier Ltd. Ocean colour remote sensing is used as a tool to detect phytoplankton size classes (PSCs). In this study, the Medium Resolution Imaging Spectrometer (MERIS), Moderate Resolution Imaging Spectroradiometer (MODIS), and Sea-viewing Wide Field-of-view Sensor (SeaWiFS) phytoplankton size classes (PSCs) products were compared with in-situ High Performance Liquid Chromatography (HPLC) data for the South China Sea (SCS), collected from August 2006 to September 2011. Four algorithms were evaluated to determine their ability to detect three phytoplankton size classes. Chlorophyll-a (Chl-a) and absorption spectra of phytoplankton (aph(λ)) were also measured to help understand PSC’s algorithm performance. Results show that the three abundance-based approaches performed better than the inherent optical property (IOP)-based approach in the SCS. The size detection of microplankton and picoplankton was generally better than that of nanoplankton. A three-component model was recommended to produce maps of surface PSCs in the SCS. For the IOP-based approach, satellite retrievals of inherent optical properties and the PSCs algorithm both have impacts on inversion accuracy. However, for abundance-based approaches, the selection of the PSCs algorithm seems to be more critical, owing to low uncertainty in satellite Chl-a input data

For studies in bio-optical oceanography, visible light properties by classes of dissolved organic matter should be characterized. The regional adjustment of model parameters is one approach which is being widely used to refine bio-optical models. In the present study, buoy and laboratory data were acquired, and during the 15-day observation period an algal bloom event occurred. The absorption coefficient of CDOM at 443 nm, a(g)(443), changed in the range of 0.09-0.35 m(-1) and 0.1-0.34 m(-1) for two depths, 0.32 m and 2.3 m., respectively, throughout the entire period. CDOM absorption was larger for bloom conditions than for non-bloom conditions. In addition, the fraction of CDOM in total absorption was higher during the bloom event than that of non-bloom conditions. The spectral slope of CDOM absorption, S., regressed over 400-500 nm, ranged from 0.015 to 0.0185 nm(-1) with an average of 0.0166 nm(-1). CDOM fluorescence intensity (Fcdom) was obtained using an internal Raman standard and varied over the range of 467.44-1538.23 in relative units. Variations in Fcdom showed a similar pattern to that of CDOM absorption. A robust non-linear relationship between Fcdom and CDOM absorption was found, with a correlation coefficient of 0.893, throughout the whole observation. An attempt to describe S-g with absorption showed a promising consequence that can be described with a negative correlation during the bloom, however, without any distinguishable tendency for non-bloom time span. A two-band ratio algorithm was also carried out for retrieving CDOM absorption. The variations in optical properties of CDOM may be related to the complicated environment in the estuarine waters, which may result from different sources of CDOM. (C) 2009 Elsevier Ltd.
