A restrição dietética de sódio reduz a pressão arterial e morbimortalidade cardiovascular. No entanto, a redução intensa no consumo de sódio aumenta a atividade do sistema-renina-angiotensina-alosterona, reduz a sensibilidade periférica à insulina e eleva os lípides plasmáticos, fatores de risco para doenças metabólicas e cardiovasculares. Por outro lado, o treinamento físico aeróbio (TFA) é uma ferramenta para prevenção e tratamento das disfunções metabólicas, especialmente devido à sua ação positiva sobre a sensibilidade periférica à insulina. O objetivo do presente estudo foi avaliar em camundongos dislipidêmicos, knockout para o receptor de LDL (LDLr-KO), os efeitos da restrição de sódio dietético e TFA sobre a resistência periférica à insulina, conteúdo de lípides e expressão de genes relacionados ao metabolismo de lípides e resistência à insulina no fígado. Para isso, camundongos LDLr KO machos com 12 semanas de idade, alimentados com dieta nomossódica (NS) ou hipossódica (HS) foram mantidos sedentários (S) ou treinados (T) por 90 dias. Foram determinados massa corporal, lípides plasmáticos, resistência periférica à insulina, conteúdo hepático de triglicérides (TG), atividade da enzima citrato sintase e a expressão de genes associados ao metabolismo de lípides, bem como a resistência periférica à insulina. Os resultados foram comparados por ANOVA de 2 fatores e pós-teste de Tukey. Comparado ao grupo NS, o grupo HS apresentou maior massa corporal, menor sensibilidade periférica à insulina e conteúdo aumentado de TG plasmático e hepático. Tanto a menor sensibilidade periférica à insulina quanto a concentração de TG hepático, induzidos pela dieta HS, foram prevenidos pelo TFA. No fígado, o grupo HS, comparado ao grupo NS, apresentou aumento na expressão de genes associados à resistência à insulina (Apoc3, G6pc, Pck1) e redução de genes relacionados à capacidade oxidativa (Prkaa1, Prkaa2, Ppara, Lipe) e montagem de lipoproteínas (Mttp). O grupo HS-T apresentou maior a atividade hepática da enzima citrato sintase, quando comparado aos demais grupos. Em conclusão, o TFA preveniu o acúmulo de TG hepático, induzido pela dieta HS, devido ao seu papel protetor sobre sensibilidade periférica à insulina e estímulo à expressão de genes envolvidos no metabolismo oxidativo hepático Low-sodium (LS) diet reduces blood pressure and cardiovascular morbidity and mortality. On the other hand, aerobic exercise training (AET) is an effective tool for the prevention and treatment of metabolic disorders, especially due to its positive action on peripheral insulin sensitivity. The aim of the present study was to evaluate the effects of LS diet and AET in peripheral insulin resistance, and hepatic TG content and expression of genes related to lipid metabolism and peripheral insulin resistance. Twelve-week-old male dyslipidemic mice - LDL receptor knockout mice (LDLr-KO) - fed normal sodium (NS) or LS diet were kept sedentary (S) or trained (T) for 90 days. Body mass, plasma lipids, peripheral insulin resistance, hepatic triglycerides (TG) content, activity of citrate synthase and expression of genes associated with lipid metabolism and peripheral insulin resistance were determined. Results were compared by 2-way ANOVA and Tukey\'s post-test. Compared to NS, LS group presented higher body weight, reduced peripheral insulin sensitivity and, elevated plasma and hepatic TG concentration. Both the impairment in peripheral insulin sensitivity and the increase in hepatic TG were prevented by the AET. In the liver, compared to NS group, LS group increased the expression of genes related to insulin resistance (Apoc3, G6pc, Pck1) and reduced those associated with oxidative capacity (Prkaa1, Prkaa2, Ppara, Lipe) and assembly of lipoproteins (Mttp). The LS-T group showed higher activity of citrate synthase in the liver, compared to other groups. In conclusion, AET prevented the accumulation of hepatic TG induced by the LS diet, due to its protective role on peripheral insulin sensitivity and stimulation of the expression of genes involved in hepatic oxidative metabolism

Objectives Because the plasma campesterol/cholesterol ratio does not differ between groups that absorb different amounts of cholesterol, the authors investigated whether the plasma Phytosterols (PS) relate to the body's cholesterol synthesis rate measured as non-cholesterol sterol precursors (lathosterol). Method The authors studied 38 non-obese volunteers (58±12 years; Low-Density Lipoprotein Cholesterol ‒ LDL-C ≥ 130 mg/dL) randomly assigned to consume 400 mL/day of soy milk (Control phase) or soy milk + PS (1.6 g/day) for four weeks in a double-blind, cross-over study. PS and lathosterol were measured in plasma by gas chromatography coupled to mass spectrophotometry. Results PS treatment reduced plasma total cholesterol concentration (-5.5%, p < 0.001), LDL-C (-7.6%, p < 0.001), triglycerides (-13.6%, p < 0.0085), and apolipoprotein B (apo B) (-6.3%, p < 0.008), without changing high density lipoprotein cholesterol (HDL-C concentration), but plasma lathosterol, campesterol and sitosterol expressed per plasma cholesterol increased. Conclusions The lathosterol-to-cholesterol plasma ratio predicted the plasma cholesterol response to PS feeding. The highest plasma lathosterol concentration during the control phase was associated with a lack of response of plasma cholesterol during the PS treatment period. Consequently, cholesterol synthesis in non-responders to dietary PS being elevated in the control phase indicates these cases resist to further synthesis rise, whereas responders to dietary PS, having in the control phase synthesis values lower than non-responders, expand synthesis on alimentary PS. Responders absorb more PS than non-responders, likely resulting from responders delivering into the intestinal lumen less endogenous cholesterol than non-responders do, thus facilitating greater intestinal absorption of PS shown as increased plasma PS concentration.

Non-cholesterol sterols are transported in plasma lipoproteins and are consequently important in cholesterol metabolism. We investigated the distribution of non-cholesterol sterol precursors of cholesterol synthesis (NCSPCS), oxysterols, and phytosterols in lipoproteins of healthy subjects differing according to HDL-Cholesterol (HDL-C) plasma levels. Elevated NCSPCS (desmosterol, lathosterol) in the High HDL group suggests that HDL exports these sterols from cells, but not the cholesterol metabolite 24-OHC which was higher in the Low HDL group than in the High HDL group. 27-hydroxycholesterol (27OH-C) plasma levels did not differ between groups. Percentage of NCSPCS and phytosterols predominates in LDL, but did not differ between groups. Thirty percent of desmosterol and lathosterol are present in HDL, with the High HDL group carrying higher percentage of these sterols. A high percentage of campesterol and sitosterol in HDL suggests that phytosterols are absorbed by enterocytes, and that HDL could be a marker of the ABCA1/ApoA1 intestinal activity.

Background: A low-sodium (LS) diet reduces blood pressure, contributing to the prevention of cardiovascular diseases. However, intense dietary sodium restriction impairs insulin sensitivity and worsens lipid profile. Considering the benefits of aerobic exercise training (AET), the effect of LS diet and AET in hepatic lipid content and gene expression was investigated in LDL receptor knockout (LDLr-KO) mice. Methods: Twelve-week-old male LDLr-KO mice fed a normal sodium (NS) or LS diet were kept sedentary (S) or trained (T) for 90 days. Body mass, plasma lipids, insulin tolerance testing, hepatic triglyceride (TG) content, gene expression, and citrate synthase (CS) activity were determined. Results were compared by 2-way ANOVA and Tukey’s post-test. Results: Compared to NS, LS increased body mass and plasma TG, and impaired insulin sensitivity, which was prevented by AET. The LS-S group, but not the LS-T group, presented greater hepatic TG than the NS-S group. The LS diet increased the expression of genes related to insulin resistance (ApocIII, G6pc, Pck1) and reduced those involved in oxidative capacity (Prkaa1, Prkaa2, Ppara, Lipe) and lipoprotein assembly (Mttp). Conclusion: AET prevented the LS-diet-induced TG accumulation in the liver by improving insulin sensitivity and the expression of insulin-regulated genes and oxidative capacity.

Experimental and clinical studies have demonstrated the effect of phytosterols (PS) on reducing plasma levels of cholesterol and LDL-c, but the effects of plant sterols beyond cholesterol-lowering are still questionable. Since inflammation and endothelial dysfunction are involved in the pathogenesis of atherosclerosis, this study aims to evaluate the effect of PS on biomarkers involved in atherosclerosis progression and whether these effects are independent of alterations in plasma LDL-c levels. Thirty-eight moderately hypercholesterolemic volunteers (58 &plusmn, 12 years, LDL-c &ge, 130 mg/dL) were randomly assigned to consume 400 mL/day of soy milk or soy milk + PS (1.6 g/day) for 4 weeks in a double-blind, placebo-controlled, cross-over study. Blood samples were collected and lipid profiles and biomarkers for inflammation and endothelial dysfunction determined. The results showed that PS treatment reduced endothelin-1 plasma concentration by 11% (p = 0.02) independently of variations in plasma levels of LDL-c. No alterations were observed regarding fibrinogen, IL-6, hs-CRP, SAA, TNF&alpha, or VCAM-1 between placebo and PS-treated groups. Furthermore, PS reduced total plasma cholesterol concentration (&minus, 5,5%, p &lt, 0.001), LDL-c (&minus, 6.4%, p &lt, 0.05), triglycerides (&minus, 8.3%, p &lt, 0.05), and apo B (&minus, 5.3%, p &lt, 0.05), without changing HDL-c concentration (p &gt, 0.05). Therefore, PS supplementation effectively lowers endothelin-1 independently of the reductions in plasma levels of LDL-c, contributing to the comprehension of the effect of plant sterols on endothelial function and prevention of cardiovascular diseases.

A low-sodium (LS) diet has been shown to reduce blood pressure (BP) and the incidence of cardiovascular diseases. However, severe dietary sodium restriction promotes insulin resistance (IR) and dyslipidemia in animal models and humans. Thus, further clarification of the long-term consequences of LS is needed. Here, we investigated the effects of chronic LS on gastrocnemius gene and protein expression and lipidomics and its association with IR and plasma lipids in LDL receptor knockout mice. Three-month-old male mice were fed a normal sodium diet (NS; 0.5% Na; n = 12–19) or LS (0.06% Na; n = 14–20) over 90 days. Body mass (BM), BP, plasma total cholesterol, triacylglycerol (TG), glucose, hematocrit, and IR were evaluated. LS increased BM (9%), plasma TG (51%), blood glucose (19%), and IR (46%) when compared with the NS. RT-qPCR analysis revealed that genes involved in lipid uptake and oxidation were increased by the LS: Fabp3 (106%), Prkaa1 (46%), and Cpt1 (74%). Genes and proteins (assessed by Western blotting) involved in insulin signaling were not changed by the LS. Similarly, lipid species classically involved in muscle IR, such as diacylglycerols and ceramides detected by ultra-high-performance liquid chromatography coupled to mass spectrometry, were also unchanged by LS. Species of phosphatidylcholines (68%), phosphatidylinositol (90%), and free fatty acids (59%) increased while cardiolipins (41%) and acylcarnitines (9%) decreased in gastrocnemius in response to LS and were associated with glucose disposal rate. Together these results suggest that chronic LS alters glycerophospholipid and fatty acids species in gastrocnemius that may contribute to glucose and lipid homeostasis derangements in mice.

Os óxidos de colesterol modulam o desenvolvimento da aterosclerose por mediarem a síntese, captação e exportação de colesterol, além de inflamação e citotoxicidade na parede arterial. O exercício físico regular previne e regride a lesão aterosclerótica, por melhorar o perfil lipídico, transporte reverso de colesterol e defesas antioxidantes. A proteína cinase ativada por AMP (AMPK) é um importante mediador dos efeitos metabólicos do exercício físico. Em macrófagos, sua ativação vincula-se ao aumento no efluxo de colesterol e diminuição na captação de LDL. Entretanto, não está claro se o treinamento físico modula as concentrações de óxidos de colesterol, refletindo seu benefício sobre a prevenção da aterosclerose, e se esses efeitos podem ser mediados pela AMPK. O objetivo do presente estudo foi avaliar, em camundongos dislipidêmicos, o papel de 6 semanas de treinamento físico aeróbio sobre: o infiltrado de colesterol arterial e a distribuição de óxidos de colesterol no arco aórtico e no plasma; a expressão gênica de proteínas envolvidas na metabolização de óxidos de colesterol na parede arterial; e o efeito da ativação da AMPK em macrófagos, in vitro, sobre a concentração dos óxidos de colesterol e expressão de genes envolvidos na metabolização de óxidos de colesterol. Para tanto, camundongos machos knockout para apolipoproteína E, com 16 semanas de idade, alimentados com dieta padrão, foram incluídos no estudo. O treinamento físico foi realizado em esteira, 15 m/min, por 60 min, 5 dias/semana, durante 6 semanas. Lípides plasmáticos e glicose foram determinados por ensaio enzimático e glicosímetro, respectivamente, antes e após o treinamento físico. Colesterol arterial e óxidos de colesterol foram avaliados por cromatografia gasosa acoplada à espectrometria de massa. A expressão de genes envolvidos no metabolismo de lípides foi avaliada RT-qPCR. Os resultados foram comparados por ANOVA de um fator com pós-teste de Newman-Keuls ou teste t de Student. Peso corporal, colesterol total, TG, HDL-c, glicose e óxidos de colesterol no plasma foram semelhantes entre os grupos. O treinamento físico aumentou a concentração de 7alfa-OH C (70%) e reduziu a de colesterol (32%) na aorta. Além disso, o exercício físico aumentou a expressão gênica da Cyp27a1 (54%), Cd36 (75%), Cat (70%), Prkaa1 (AMPKalfa1) (40%) e Prkaa2 (AMPKalfa2) (51%) e reduziu Abcg1 (31%), Olr1 (LOX-1) (65%), Cyp7b1 (35%) e Ch25h (48%). Nenhuma alteração foi observada na expressão de Abca1, Nr1h3 (LXRalfa) e Nr1h2 (LXRbeta). Nos macrófagos, a ativação da AMPK por AICAR, reduziu o conteúdo de 7alfa-OH C após estimulo com HDL2. O tratamento com AICAR aumentou a expressão gênica de Abca1 (52%) e Cd36 (220%) e diminuiu Prkaa1 (19%) e Cyp27a1 (47%), e não alterou Abcg1, Nr1h3 e Nr1h2. Em conclusão, em camundongos dislipidêmicos, o treinamento físico aeróbio, por 6 semanas, aumentou a concentração de 7 beta -OH C, o que se vincula à maior expressão de Cd36 no arco aórtico. A rápida difusão de óxidos de colesterol, como via complementar ao transporte reverso de colesterol, pode também ser favorecida pelo aumento e redução, respectivamente, na expressão de Cyp27a1 e Cyp7b1, favorecendo maior liberação de 27-OH C das células. Juntamente com suas ações diretas que beneficiam o transporte reverso de colesterol, previamente descritas o treinamento físico diminui a concentração de colesterol na parede arterial, prevenindo a aterosclerose. Regular physical exercise prevents and regresses atherosclerosis by improving lipid metabolism, reverse cholesterol transport and antioxidant defenses. AMP-activated protein kinase (AMPK) plays an important role in the beneficial metabolic adaptations of physical exercise. In macrophages, its activation is related to the enhancement in cholesterol efflux and reduction in LDL uptake. However, it is not clear whether exercise training benefits in atherosclerosis is mediated by its action in oxysterols concentrations, and whether this can be modulated by AMPK. The aim of this study was to evaluate the role of a 6-week aerobic exercise training program in dyslipidemic mice in the arterial and plasma accumulation of cholesterol and oxysterols subspecies; expression of genes related to oxysterols metabolisms in the aortic arch, and the effect of AMPK activation in macrophage on the concentration of oxysterols and expression of genes linked to oxysterols metabolism. Sixteen-week-old male apoE knockout mice fed a chow diet were included in the protocol. Animals were trained in a treadmill running, 15 m/min, 60 min, 5 days/week, during 6 weeks. Plasma lipids and glucose were determined by enzymatic techniques and glycosometer, respectively. Cholesterol in aortic arch and oxysterols were measured by gas chromatography/mass spectrometer. The expression of genes involved in lipid metabolism was determined by RT-qPCR. Results (mean ± SD) were compared by one-way ANOVA with Newman-Keuls posttest or Student\'s t-test. Body weight and plasma total cholesterol, TG, HDL-c, glucose, and oxysterols were similar among groups. The exercise training enhanced 7beta-hydroxycholesterol (70%) and reduced cholesterol (32%) in the aortic arch. In addition, exercise increased Cyp27a1 (54%), Cd36 (75%), cat (70%), Prkaa1 (AMPKalpha1) (40%) and Prkaa2 (AMPKalpha2) (51%) mRNA. No changes were observed in the expression of Abca1, Nr1h3 (LXRalpha) and Nr1h2 (LXRbeta). In macrophages, the activation of AMPK by AICAR, reduced 7beta-hydroxycholesterol level after stimulation by HDL2. Treatment with AICAR increased Abca1 (52%) and Cd36 (220%), decreased Prkaa1 (19%) e Cyp27a1 (47%), and did not change Abcg1, Nr1h3 e Nr1h2. In conclusion, in dyslipidemic mice aerobic exercise training increases the nonenzymatic-driven oxysterol, 7beta-hydroxycholesterol, which is related to the enhanced expression of Cd36. The rapid diffusion of oxysterols, as a complementary pathway for the reverse cholesterol transport, may also be favored by the increase and reduction of Cyp27a1 and Cyp7b1 expressions, respectively, which in turns favors 27-OH C desorption from cells. Together with its direct role in improving reverse cholesterol transport as previously reported, aerobic exercise training diminishes cholesterol accumulation in the arterial wall preventing atherosclerosis. Based on in vitro assays, the AMPK activation does not seem to contribute to the effect of exercise in increasing oxysterols

Resumo Objetivo: Avaliar em qual fase da síndrome de Vogt-Koyanagi-Harada (SVKH) os pacientes recebem o primeiro atendimento em serviço especializado. Métodos: Foram analisados prontuários de 14 pacientes atendidos no Setor de Uveítes do Hospital Universitário Clementino Fraga Filho da UFRJ no período de janeiro de 2014 a março de 2017. Nesta análise, foram observados o sexo, a idade, a fase da doença e a acuidade visual destes pacientes com ao menos doença provável da SVKH. Resultados: Observamos que 35,4% dos pacientes apresentavam a doença ainda na fase uveítica e que 78,5% destes pacientes apresentava acuidade visual igual ou pior que 0,05. Destes pacientes, 78,5% eram do sexo feminino e 21,5% do sexo masculino e a mediana de idades foi de 34 anos. Conclusão: Os pacientes analisados obtiveram dificuldade em ter acesso precoce a um setor especializado, afetando assim, diretamente o tratamento e prognóstico visual. Abstract Objective: To evaluate in which phase of Vogt-Koyanagi-Harada (VKH) syndrome the patients receive the first attendance in specialized service. Methods: A retrospective study was conduted to evaluate medical records of 14 patients with VKH in the Clementino Fraga Filho University Hospital of the Federal University of Rio de Janeiro from January 2014 to March 2017. In this analysis, gender, age, stage of disease and visual acuity of these patients with at least probable VKH were recorded. Results: Of these patients, 78.5% were female and 21.5% male and the median age was 34 years. We observed that 35.4% of the patients had the disease still in the uveitic phase and that 78.5% of these patients had visual acuity equal to or worse than 0.05. Conclusion: There is a delay in the admission of these patients to a specialized sector, thus affecting directly the treatment and visual prognosis.

Background: Oxysterols are bioactive lipids that control cellular cholesterol synthesis, uptake, and exportation besides mediating inflammation and cytotoxicity that modulate the development of atherosclerosis. Aerobic exercise training (AET) prevents and regresses atherosclerosis by the improvement of lipid metabolism, reverse cholesterol transport (RCT) and antioxidant defenses in the arterial wall. We investigated in dyslipidemic mice the role of a 6-week AET program in the content of plasma and aortic arch cholesterol and oxysterols, the expression of genes related to cholesterol flux and the effect of the exercise-mimetic AICAR, an AMPK activator, in macrophage oxysterols concentration. Methods: Sixteen-week old male apo E KO mice fed a chow diet were included in the protocol. Animals were trained in a treadmill running, 15 m/min, 5 days/week, for 60 min (T; n = 29). A control group was kept sedentary (S; n = 32). Plasma lipids and glucose were determined by enzymatic techniques and glucometer, respectively. Cholesterol and oxysterols in aortic arch and macrophages were measured by gas chromatography/mass spectrometry. The expression of genes involved in lipid metabolism was determined by RT-qPCR. The effect of AMPK in oxysterols metabolism was determined in J774 macrophages treated with 0.25 mM AICAR. Results: Body weight and plasma TC, TG, HDL-c, glucose, and oxysterols were similar between groups. As compared to S group, AET enhanced 7β-hydroxycholesterol (70%) and reduced cholesterol (32%) in aorta. In addition, exercise increased Cyp27a1 (54%), Cd36 (75%), Cat (70%), Prkaa1 (40%), and Prkaa2 (51%) mRNA. In macrophages, the activation of AMPK followed by incubation with HDL2 increased Abca1 (52%) and Cd36 (220%) and decrease Prkaa1 (19%), Cyp27a1 (47%) and 7α-hydroxycholesterol level. Conclusion: AET increases 7β-hydroxycholesterol in the aortic arch of dyslipidemic mice, which is related to the enhanced expression of Cd36. In addition, the increase and reduction of Cyp27a1 and Cyp7b1 in trained mice may contribute to enhance levels of 27-OH C. Both oxysterols may act as an alternative pathway for the RCT contributing to the reduction of cholesterol in the aortic arch preventing atherogenesis.

Objective: Children of parents with bipolar disorder (BD) are at heightened risk for developing mood and other psychiatric disorders. We proposed to evaluate the environment of families with at least one parent with BD type I (BDF) with affected offspring (aBDF) and unaffected offspring (uBDF) compared with control families without a history of DSM-IV Axis I disorder (CF). Method: We used the Family Environment Scale (FES) to evaluate 47 BDF (aBDF + uBDF) and 30 CF. Parents were assessed through the Structured Clinical Interview for DSM-IV Axis I Disorders (SCID-I). Diagnosis of the offspring was determined through the Schedule for Affective Disorders and Schizophrenia for School-Age Children/Present and Lifetime Version (K-SADS-PL) interview. Results: There were statistically significant differences between aBDF, uBDF and CF in cohesion ( p = 0.003), intellectual-cultural orientation ( p = 0.01), active-recreational orientation ( p = 0.007), conflict ( p = 0.001), control ( p = 0.01), moral-religious emphasis ( p = 0.01) and organization ( p = 0.001). The aBDF showed higher levels of control ( p = 0.02) when compared to the uBDF. Conclusions: Families with a BD parent presented more dysfunctional interactions among members. Moreover, the presence of BD or other psychiatric disorders in the offspring of parents with BD is associated with higher levels of control. These results highlight the relevance of psychosocial interventions to improve resilience and family interactions.

Cholesterol is an essential component in the structure and function of cell membranes and has been associated with the major pathological signatures of Alzheimer’s disease (AD). To maintain brain cholesterol homeostasis, it is converted into 24(S)-hydroxycholesterol (24OHC) which can be driven through the blood–brain barrier. Several studies have already described a decrease in 24OHC and an increase of 27(S)-hydroxycholesterol (27OHC) in AD, as a reflection of disease burden, the loss of metabolically active neurons and the degree of structural atrophy. It is also well known that peripheral cholesterol is altered in AD patients. However, there are no data regarding effects of AD treatment in this cholesterol pathway. Since a study from our group indicated a significant increase in membrane phospholipid metabolism by donepezil, the aim of this study was to evaluate the effect of short- and long-term donepezil treatment on cholesterol and metabolites 24OHC and 27OHC in plasma of AD patients and in healthy volunteers. At baseline, we found a decrease of 24OHC (p = 0.003) in AD patients. Cholesterol levels increased with donepezil treatment (p = 0.04) but no differences were observed regarding 24OHC and 27OHC. Background Regular exercise prevents and regresses atherosclerosis by improving lipid metabolism and antioxidant defenses. Exercise ameliorates the reverse cholesterol transport (RCT), an antiatherogenic system that drives cholesterol from arterial macrophages to the liver for excretion into bile and feces. In this study we analyzed the role of aerobic exercise on the in vivo RCT and expression of genes and proteins involved in lipid flux and inflammation in peritoneal macrophages, aortic arch and liver from wild type mice. Methods Twelve-week-old male mice were divided into sedentary and trained groups. Exercise training was performed in a treadmill (15 m/min, 30 min/day, 5 days/week). Plasma lipids were determined by enzymatic methods and lipoprotein profile by fast protein liquid chromatography. After intraperitoneal injection of J774-macrophages the RCT was assessed by measuring the recovery of 3H-cholesterol in plasma, feces and liver. The expression of liver receptors was determined by immunoblot, macrophages and aortic mRNAs by qRT-PCR. 14C-cholesterol efflux mediated by apo A-I and HDL2 and the uptake of 3H-cholesteryl oleoyl ether (3H-COE)-acetylated-LDL were determined in macrophages isolated from sedentary and trained animals 48 h after the last exercise session. Results Body weight, plasma lipids, lipoprotein profile, glucose and blood pressure were not modified by exercise training. A greater amount of 3H-cholesterol was recovered in plasma (24 h and 48 h) and liver (48 h) from trained animals in comparison to sedentary. No difference was found in 3H-cholesterol excreted in feces between trained and sedentary mice. The hepatic expression of scavenger receptor class B type I (SR-BI) and LDL receptor (B-E) was enhanced by exercise. We observed 2.8 and 1.7 fold rise, respectively, in LXR and Cyp7a mRNA in the liver of trained as compared to sedentary mice. Macrophage and aortic expression of genes involved in lipid efflux was not systematically changed by physical exercise. In agreement, 14C-cholestrol efflux and uptake of 3H-COE-acetylated-LDL by macrophages was similar between sedentary and trained animals. Conclusion Aerobic exercise in vivo accelerates the traffic of cholesterol from macrophages to the liver contributing to prevention and regression of atherosclerosis, independently of changes in macrophage and aorta gene expression.
