Considering that COVID-19 is a respiratory disease transmitted by contact and places of incarceration are an environment that imposes additional risks for viral spread, this study sought to identify cases of SARS-CoV-2 infection in socio-educational units. We evaluated the seroprevalence of IgM and IgG for SARS-CoV-2 in workers and adolescents confined in five socio-educational units. Abstract

Objectives: The treatment of wounds accounts for a considerable fraction of health expenses as well as serious socioeconomic problems. The use of natural substances stands out as a source of new therapeutic discoveries for the wound healing. Thus, this review compiled scientific findings on the applicability of carvacrol and thymol, or essential oils containing at least one of these compounds, for the treatment of wounds. Methods: This review was performed at PubMed, SCOPUS, Web of Science databases using keywords as wound healing, thymol/carvacrol and essential oils. Thirteen studies were selected for discussion. Key findings: Thymol/carvacrol was able to act in the three phases of wound healing. In the first phase, they showed modulatory effect of the inflammatory cytokines, oxidative stress and antimicrobial power. In the second phase, they promoted re‐epithelialization, angiogenesis and development of granulation tissue. Finally, in the third phase, they improve the collagen deposition and modulated the growth of fibroblasts and keratinocytes. Conclusions: These compounds present a high potential for the development of new therapeutic for wound repair. However, dose, efficacy and safety of these compounds for the treatment of wounds, as well as the mechanisms by which those effects can be observed, are challenges for future studies. Abstract

Throughout the history of the development of treatments proposed for pain, medicinal plants and their secondary metabolites have played a key role. However, even more effective therapeutic options for the treatment of painful conditions are necessary. In this sense, the present study aimed to evaluate the effect of the inclusion complex of β-cyclodextrin (β-CD) containing carvacrol, a phenolic monoterpene present in the essential oil of medicinal plants of the genus Origanum, Thymus (Lamiaceae), on the oncology nociception (induced by tumor cell sarcoma 180, S180) in experimental animals. This thesis is formed by three chapters, consisting of a review of patents and two regular papers. Initially, the review Terpenes and derivatives as a new perspective for pain treatment: a patent review was elaborated, which describes patents on the analgesic properties of terpenes and prospects created from these records. After conducting the experimental protocols, the article Evidence for the Involvement of descending pain- inhibitory mechanisms in the attenuation of cancer pain induced by carvacrol , a monoterpene of Origanum oil was constructed, in which it has been shown that carvacrol (CARV) reduces nociception induced by tumor cells (S180), decreases the number of Fos positive neurons in lamina I of the Lumbar spinal cord and activates the brain nuclei involved in the descending inhibitory pain pathway, such as the periaqueductal gray, nucleus raphe magnus and locus coeruleus. Subsequently, the inclusion complex consisting of carvacrol and β-cyclodextrin (CARV/β-CD) was developed and characterized physicochemically through the method of co-evaporation. CARV complexation was able to increase the time of the antinociceptive effects of this compound in animals with S180 in 15 hours and improved the pharmacological response at lower doses, in addition to promoting a clear increase in solubility and stability. These data formed the patent submitted and approved by the Center for Innovation and Technology Transfer (CINTEC UFS) and forms the scientific basis of the article Encapsulation of β-cyclodextrin in carvacrol improves the pharmacological response on cancer pain . From these findings, we conclude that the analgesic effect of carvacrol on cancer pain can be enhanced by employing inclusion complexes containing β-cyclodextrin and may represent an interesting object of further translational research and clinical trials in order clinical evidence to be obtained, which is necessary for the development of new drugs for cancer-associated pain management. Ao longo da história do desenvolvimento de propostas terapêuticas para o tratamento da dor, as plantas medicinais e seus metabólitos secundários tem exercido um papel fundamental. No entanto, ainda são necessárias opções terapêuticas mais eficazes para o tratamento de condições dolorosas. Neste sentido, o presente trabalho buscou avaliar o efeito do complexo de inclusão de β-ciclodextrina (β-CD) contendo carvacrol, um monoterpeno fenólico presente no óleo essencial de plantas medicinais do gênero Origanum e Thymus (Lamiaceae), sobre a nocicepção oncológica (induzida por células tumorais sarcoma 180, S180) em animais de experimentação. A tese é composta por três capítulos, constituídos de uma revisão de patentes e dois artigos científicos. Inicialmente foi elaborada a revisão Terpenes and derivatives as a new perspective for pain treatment: a patent review , a qual descreve as patentes sobre as propriedades analgésicas dos terpenos e as perspectivas criadas a partir destes registros. Após a realização dos protocolos experimentais foi elaborado o artigo Evidence for the involvement of descending pain-inhibitory mechanisms in the attenuation of cancer pain induced by carvacrol, a monoterpene of Origanum oil , no qual foi demonstrado que o carvacrol (CARV) reduz a nocicepção induzida por células tumorais (S180), diminui o número de neurônios Fos positivos na lâmina I da medula lombar e ativa núcleos cerebrais envolvidos na via descendente inibitória da dor, como a substância cinzenta periaquedutal, núcleo magno da rafe e locus ceruleus. Posteriormente, foi desenvolvido e caracterizado físico-quimicamente o complexo de inclusão constituído de β-ciclodextrina e carvacrol (CARV/β-CD) através do método de co-evaporação. A complexação do CARV foi capaz de aumentar o tempo do efeito antinociceptivo deste composto nos animais com o S180 em 15 horas e melhorar a resposta farmacológica em doses menores, além de promover nítido aumento de solubilidade e estabilidade. Estes dados compuseram a patente submetida e aprovada pelo Centro de Inovação e Transferência de Tecnologia (CINTEC UFS) e constitui a base científica do artigo Encapsulation of carvacrol in β-cyclodextrin improve the pharmacological response on cancer pain . A partir destes achados é possível concluir que o efeito analgésico do carvacrol sobre a dor oncológica pode ser aprimorado pelo emprego de complexos de inclusão contendo β-ciclodextrina e pode representar um interessante objeto de novas pesquisas translacionais e ensaios clínicos a fim de que sejam obtidas as evidências clínicas necessárias para o desenvolvimento de novos medicamentos destinados ao manejo da dor associada ao câncer.

Abstract

Morinda citrifolia Linn (popularly known as ‘‘Noni’’) has been used in folk medicine by Polynesians for over 2,000 years. It is reported to have a broad range of therapeutic effects, including effects against headache, fever, arthritis, gingivitis, respiratory disorders, infections, tuberculosis, and diabetes. The aim of this study was to investigate the antioxidant, anti-inflammatory, antinociceptive, and antibacterial properties of the aqueous extract from M. citrifolia leaves (AEMC). Antioxidant activity was observed against lipid peroxidation, nitric oxide, and hydroxyl radicals. The antinociceptive effect of AEMC was observed in the acetic acid–induced writhing test at the higher dose. Moreover, AEMC significantly reduced the leukocyte migration in doses of 200 and 400 mg/kg and showed mild antibacterial activity. Together, the results suggest that properties of M. citrifolia leaf extract should be explored further in order to achieve newer tools for managing painful and inflammation conditions, including those related to oxidant states.

Abstract

Cymbopogon winterianus Jowitt (‘Java citronella’) is an important essential oil yielding aromatic grass cultivated in India and Brazil and its volatile essential oils extracted from its leaves are used in perfumery, cosmetics, pharmaceuticals and flavoring industries. However, there is no report on any psychopharmacological study of C. winterianus leaf essential oil (LEO) available to date. In this study, the pharmacological effects of the LEO were investigated in animal models and its phytochemical analyses. GC-MS analysis showed a mixture of monoterpenes, as citronellal (36.19%), geraniol (32.82%) and citronellol (11.37%). LEO exhibited an inhibitory effect on the locomotor activity of mice, an antinociceptive effect by increasing the reaction time in the writhing and capsaicin tests. All doses induced a significant increase in the sleeping time of animals not having modified however, the latency. The LEO did not alter the remaining time of the animals on the rota-rod apparatus. Abstract

Amburana cearensis, popularly known as "umburana-de-cheiro" or "cumaru" is a tree widely distributed in northeastern of Brazil, its wood is used in furniture making and has considerable therapeutic properties in folk medicine. Several compounds have been isolated and identified from A. cearensis, including protocatechuic acid, coumarins, flavonoids (isokaempferide, kaempferol, afrormosin and 4’-methoxyfisetin) and the phenol glucosides (amburosides A and B) among others. This species has antiinflammatory, analgesic, antispasmodic and bronchodilator. Despite its economic and pharmacology importance few studies are found in the literature about that species. Therefore, this study attempts to review the chemistry and pharmacology of this plant species Amburana cearensis, conhecida popularmente como “umburana-de-cheiro” ou “cumaru”, é uma planta arbórea amplamente distribuída no Nordeste brasileiro, sua madeira é utilizada na movelaria e tem apreciáveis propriedades terapêuticas na medicina popular. Abstract

Carvacrol, a monoterpenic phenol present in essential oils of the Labiatae family, has been used through the ages as a source of flavor in food and for medicinal purposes. Borneol is a monoterpene found in several species of Artemisia and Dipterocarpaceae, used for anxiety, pain and anesthesia in traditional Chinese. Citral, a mixture of two geometrical isomers (neral and geranial), is one of the most important compounds in some citrus oils and has central nervous system (CNS) properties. The anticonvulsant effect of carvacrol (CARV), (−)-borneol (BOR) and citral (CIT) was investigated in two animal models of epilepsy. Mice were pretreated with CARV, BOR or CIT (50, 100, and 200 mg/kg, i.p.) 30 min before pentylenetetrazole (PTZ) or maximal electroshock (MES) tests, the two most important animal epilepsy tests. The latency for development of convulsions and protection percentage was recorded. In order to investigate the involvement of GABAergic system, flumazenil was utilized. These monoterpenes, CARV in a higher, but not in a lower dose (p < 0.001), BOR and CIT in all doses (p < 0.05 or p < 0.001), were capable of promoting an increase of latency for the development of convulsions induced by PTZ. Additionally, these compounds were efficient in preventing the tonic convulsions (p < 0.05) induced by MES. However, the GABAergic neurotransmitter system might be involved, at least in BOR effects. Abstract

AIM: In the present study we verified the anticonvulsant properties of the new tryptamine analogue, N-salicyloyltryptamine (NST), in rodents. METHODS AND RESULTS: In the evaluation of the anticonvulsant activity, NST protected the animals from the incidence of seizures induced by pentylenetetrazole (PTZ) and picrotoxin (PIC), in doses of 100 and 200 mg/kg. NST (100 and 200 mg/kg, i.p.) significantly eliminated the extensor reflex of maximal electric-induced seizure tests in 40% of the experimental animals. However, in the PTZ model FLU (10 mg/kg, i.p.), an antagonist of the benzodiazepine (BZD) site in the GABAA-BZD receptor complex, inhibited the prolongation of seizure latency induced by NST. Abstract

ABSTRACTIntroduction: Depression is a serious mood disorder and is one of the most common mental illnesses. Despite the availability of several classes of antidepressants, a substantial percentage of patients are unresponsive to these drugs, which have a slow onset of action in addition to producing undesirable side effects. Some scientific evidence suggests that cyclodextrins (CDs) can improve the physicochemical and pharmacological profile of antidepressant drugs (ADDs). The purpose of this paper is to disclose current data technology prospects involving antidepressant drugs and cyclodextrins.Areas covered: We conducted a patent review to evaluate the antidepressive activity of the compounds complexed in CDs, and we analyzed whether these complexes improved their physicochemical properties and pharmacological action. The present review used 8 specialized patent databases for patent research, using the term ‘cyclodextrin’ combined with ‘antidepressive agents’ and its related terms. We found 608 patents. In the end, considering the inclusion criteria, 27 patents reporting the benefits of complexation of ADDs with CDs were included.Expert opinion: The use of CDs can be considered an important tool for the optimization of physicochemical and pharmacological properties of ADDs, such as stability, solubility and bioavailability.

Abstract

Context: Pain corresponds to the most frequent reason for visits to physicians, and its control by conventional drugs is accompanied by several side effects, making treatment difficult. For this reason, new chemical entities derived from natural products still hold great promise for the future of drug discovery to pain treatment. Objective: The objective of this study was to evaluate the antinociceptive and anti-inflammatory profiles ofp-cymene (PC), a monocyclic monoterpene, and its possible mechanisms of action. Materials and methods: Mice treated acutely with PC (25, 50, or 100 mg/kg, i.p.) were screened for carrageenan-induced hyperalgesia and the inflammatory components of its cascade (30–180 min), carrageenan-induced pleurisy (4 h), and tail-flick test (1–8 h). Also, we observed the PC effect on the generation of nitric oxide by macrophages and the activation of neurons in the periaqueductal gray (PAG) by immunofluorescence. Results: PC reduced (p < 0.001) the hyperalgesia induced by carrageenan, TNF-α, dopamine, and PGE2. PC decrease total leukocyte migration (100 mg/kg:p < 0.01), neutrophils (50 and 100 mg/kg:p < 0.05 and 0.001), and TNF-α (25, 50, and 100 mg/kg:p < 0.01, 0.05, and 0.001, respectively), besides reducing NO production (p < 0.05)in vitro. PC produced antinociceptive effect in tail-flick test (p < 0.05), which was antagonized by naloxone, naltrindole, nor-BNI, and CTOP, and increased (p < 0.001) the number of c-Fos-immunoreactive neurons in PAG. Discussion and conclusion: These results provide information about the anti-hyperalgesic and anti-inflammatory properties of PC suggesting a possible involvement of the opioid system and modulating some pro-inflammatory cytokines. Abstract

Aristolochia trilobataL. is an aromatic plant, popularly known as “mil-homens”, and its essential oil (EO) is generally used to treat colic, diarrhea and dysentery disorders. We evaluated the antinociceptive effect of A. trilobatastem EO and of its major compound, the (R)-(-)-6-methyl-5-hepten-2-yl acetate (sulcatyl acetate: SA), using acetic acid (0.85%)-induced writhing response and formalin-induced (20 μL of 1%) nociceptive behavior in mice. We also evaluated the EO and SA effect on motor coordination, using the rota-rod apparatus. EO (25, 50 and 100 mg/kg) or SA (25 and 50 mg/kg) reduced nociceptive behavior in the writhing test (p<0.001). EO (100 mg/kg) and SA (25 and 50 mg/kg) decreased the nociception on the first phase of the formalin test (p<0.05). On the second phase, EO (25: p<0.01; 50: p<0.05 and 100 mg/kg: p<0.001) and SA (25 and 50 mg/kg; p<0.001) reduced the nociceptive response induced by formalin. EO and SA were not able to cause changes in the motor coordination of animals. Together, our results suggest that EO has an analgesic profile and SA seems to be one of the active compounds in this effect.

Abstract

Background and Aim: Although reverse zoonotic transmission events from humans to domestic cats have been described, there is currently little evidence of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) circulation in stray cats. Due to the evidence of natural and experimental infections in cats and the capacity to disseminate the virus among them, this study aimed to identify the SARS-CoV-2 antigen in stray cats from the Federal University of Sergipe in Brazil. Materials and Methods: One hundred twenty six stray cats from the university were screened for SARS-CoV-2 antigens by random sampling. Throat swab samples were tested for the virus using rapid antigen detection tests. Results: Of the 126 animals tested, 30 (23.60%) were positive for SARS-CoV-2 antigens. To our knowledge, for the first time, this study detected the SARS-CoV-2 antigen in stray cats and confirmed the presence of SARS-CoV-2 infections in Brazil's stray cat population. Conclusion: The detection of SARS-CoV-2 in stray cats poses a risk for infected and healthy animals and possibly for humans who attend the university daily. As a limitation of the study, the small sample size necessitates caution when interpreting the results. This underscores the need for further research in this area to help control diseases in stray animals during potential pandemics.
