Abstract

Avian genomes are characterized as being more compact than other amniotes, with less diversity and density of transposable elements (TEs). In addition, birds usually show bimodal karyotypes, exhibiting a great variation in diploid numbers. Some species present unusually large sex chromosomes, possibly due to the accumulation of repetitive sequences. Avian retrotransposon-like element (AviRTE) is a long interspersed nuclear element (LINE) recently discovered in the genomes of birds and nematodes, and it is still poorly characterized in terms of chromosomal mapping and phylogenetic relationships. In this study, we mapped AviRTE isolated from the Trogon surrucura genome into the T. surrucura (TSU) karyotype. Furthermore, we analyzed the phylogenetic relationships of this LINE in birds and other vertebrates. Our results showed that the distribution pattern of AviRTE is not restricted to heterochromatic regions, with accumulation on the W chromosome of TSU, yet another species with an atypical sex chromosome and TE hybridization. The phylogenetic analysis of AviRTE sequences in birds agreed with the proposed phylogeny of species in most clades, and allowed the detection of this sequence in other species, expanding the distribution of the element. Abstract

The genome organization of woodpeckers has several distinctive features e.g., an uncommon accumulation of repetitive sequences, enlarged Z chromosomes, and atypical diploid numbers. Despite the large diversity of species, there is a paucity of detailed cytogenomic studies for this group and we thus aimed to rectify this. Genome organization patterns and hence evolutionary change in the microchromosome formation of four species (Colaptes campestris, Veniliornis spilogaster, Melanerpes candidus, and Picumnus nebulosus) was established through fluorescence in situ hybridization using bacterial artificial chromosomes originally derived from Gallus gallus and Taeniopygia guttata. Findings suggest that P. nebulosus(2n = 110), which was described for the first time, had the most basal karyotype among species of Picidae studied here, and probably arose as a result of fissions of avian ancestral macrochromosomes. We defined a new chromosomal number for V. spilogaster (2n = 88) and demonstrated microchromosomal rearrangements involving C. campestris plus a single, unique hitherto undescribed rearrangement in V. spilogaster. This comprised an inversion after a fusion involving the ancestral microchromosome 12 (homologous to chicken microchromosome 12). We also determined that the low diploid number of M. candidus is related to microchromosome fusions. Woodpeckers thus exhibit significantly rearranged karyotypes compared to the putative ancestral karyotype.

Abstract

The Cuculiformes are a family of over 150 species that live in a range of habitats, such as forests, savannas, and deserts. Here, bacterial artificial chromosome (BAC) probes (75 from chicken and 14 from zebra finch macrochromosomes 1–10 +ZW and for microchromosomes 11–28 (except 16)) were used to investigate chromosome homologies between chicken and the squirrel cuckoo (Piaya cayana). In addition, repetitive DNA probes were applied to characterize the chromosome organization and to explore the role of these sequences in the karyotype evolution of P. cayana. We also applied BAC probes for chicken chromosome 17 and Z to the guira cuckoo (Guira guira) to test whether this species has an unusual Robertsonian translocation between a microchromosome and the Z chromosome, recently described in the smooth-billed ani (Crotophaga ani). Our results revealed extensive chromosome reorganization with inter- and intrachromosomal rearrangements in P. cayana, including a conspicuous chromosome size and heterochromatin polymorphism on chromosome pair 20. Furthermore, we confirmed that the Z-autosome Robertsonian translocation found in C. ani is also found in G. guira, not P. cayana. Abstract

Passeriformes birds are widely recognized for their remarkable diversity, with over 5700 species described so far. Like most bird species, they possess a karyotype characteristic of modern birds, which includes a bimodal karyotype consisting of a few pairs of macrochromosomes and many pairs of microchromosomes. Although the karyotype is typically 2n = 80, the diploid number can atypically vary greatly, ranging from 56 to approximately 100 chromosomes. In this study, we aimed to understand the extent of conservation of the karyotype’s organizational structure within four species of this group using Bacterial Artificial Chromosomes via Fluorescence In Situ Hybridization (BAC-FISH) with microchromosome probes from Chicken (Gallus gallus) or Zebra Finch (Taeniopygia guttata) per microchromosomes (GGA10-28, except GGA16). By examining the chromosome complement of four passerine species—the Streaked Flycatcher (Myiodynastes maculatus), Shiny Cowbird (Molothrus bonariensis), Southern House Wren (Troglodytes aedon), and Double-collared Seedeater (Sporophila caerulescens)—we discovered a new chromosome number for Southern House Wren. Through FISH experiments, we were able to observe the same pattern of microchromosome organization as in the common ancestor of birds. As a result, we propose a new diploid number for Southern House Wren and confirm the conservation status of microchromosome organization, which may confer evolutionary advantages to this group.

Abstract

Microchromosomes, once considered unimportant elements of the genome, represent fundamental building blocks of bird karyotypes. Shorebirds (Charadriiformes) comprise a wide variety of approximately 390 species and are considered a valuable model group for biological studies. Despite this variety, cytogenetic analysis is still very scarce in this bird order. Thus, the aim of this study was to provide insight into the Charadriiformes karyotype, with emphasis on microchromosome evolution in three species of shorebirds—Calidris canutus, Jacana jacana, and Vanellus chilensis—combining classical and molecular approaches. Cross-species FISH mapping applied two BAC probes for each microchromosome, GGA10–28 (except GGA16). The experiments revealed different patterns of microchromosome organization in the species investigated. Hence, while in C. canutus, we found two microchromosomes involved in chromosome fusions, they were present as single pairs in V. chilensis. We also described a new chromosome number for C. canutus (2n = 92). Hence, this study contributed to the understanding of genome organization and evolution of three shorebird species.

Abstract

Interchromosomal rearrangements involving microchromosomes are rare events in birds. To date, they have been found mostly in Psittaciformes, Falconiformes, and Cuculiformes, although only a few orders have been analyzed. Hence, cytogenomic studies focusing on microchromosomes in species belonging to different bird orders are essential to shed more light on the avian chromosome and karyotype evolution. Based on this, we performed a comparative chromosome mapping for chicken microchromosomes 10 to 28 using interspecies BAC-based FISH hybridization in five species, representing four Neoaves orders (Caprimulgiformes, Piciformes, Suliformes, and Trogoniformes). Our results suggest that the ancestral microchromosomal syntenies are conserved in Pteroglossus inscriptus (Piciformes), Ramphastos tucanus tucanus (Piciformes), and Trogon surrucura surrucura (Trogoniformes). On the other hand, chromosome reorganization in Phalacrocorax brasilianus (Suliformes) and Hydropsalis torquata (Caprimulgiformes) included fusions involving both macro- and microchromosomes. Fissions in macrochromosomes were observed in P. brasilianus and H. torquata. Relevant hypothetical Neognathae and Neoaves ancestral karyotypes were reconstructed to trace these rearrangements. We found no interchromosomal rearrangement involving microchromosomes to be shared between avian orders where rearrangements were detected. Our findings suggest that convergent evolution involving microchromosomal change is a rare event in birds and may be appropriate in cytotaxonomic inferences in orders where these rearrangements occurred.

Abstract

The Passeriformes is the most diverse and cytogenetically well-known clade of birds, comprising approximately 5,000 species. The sooty-fronted spinetail (Synallaxis frontalis Aves: Furnariidae) species, which belongs to the order Passeriformes, is typically found in South America, where it is widely distributed. Polymorphisms provide genetic variability, important for several evolutionary processes, including speciation and adaptation to the environment. The aim of this work was to analyze the possible cytotypes and systemic events involved in the species polymorphism. Of the sampled 19 individuals, two thirds were polymorphic, an event supposedly linked to mutations resulting from genomic evolution that can be transmitted hereditarily. A chromosomal polymorphism was detected between the 1st and 3rdpairs of autosomal macrochromosomes. This type of polymorphism is related to a pericentric inversion in regions involving chromosomal rearrangements. Differently from other polymorphism studies that report a link between polymorphic chromosomes and phenotypic changes, S. frontalis did not present any morphological variation in the sampled individuals.

Abstract

The order Passeriformes is the largest group of species karyotyped among birds, however little is known about thecytogenetic of the Mimidae family, registering only karyology basic data (giemsa staining). The aim of this studywas to analyze the chromosomal complement from the species Mimus saturninus by conventional staining anddifferential chromosome banding. Diploid number and chromosome morphology were determined, as well as thedistribution pattern of constitutive heterochromatin (CBG-banding), GTG-banding andAgNOR staining (NORs).The Chalk-browed Mockingbird has 2n=80. The first and fourth pairs are submetacentric and the second, third andfifth are acrocentric. The remaining chromosomes pairs of the complement have telocentric morphology. The Zchromosome is submetacentric and the W is metacentric. CBG-banding showed positive staining in thepericentromeric region of most macrochromosomes and microchromosomes and also at Z chromosome, differentlyfrom W chromosome which appeared totally heterochromatic. The GTG-banding was similar to Gallus gallus and inother species which have already been GTG-banded. The NORs were identified in a pair of microchromosomescharacterized by presenting a remarkable secondary constriction. This can be considered as a plesiomorphiccharacteristic for M. saturninus once baseline groups (Paleognathae) also showed a pair of microchromosomesbearing NORs., A ordem Passeriformes é o maior grupo de espécies cariotipadas entre as aves, porém pouco se sabe sobre a citogenética da família Mimidae, registrando apenas dados básicos de cariologia (coloração giemsa). O objetivo deste estudo foi analisar o complemento cromossômico da espécie Mimus saturninus por coloração convencional e bandas diferenciais de cromossomos. O número diplóide e a morfologia cromossômica foram determinados, bem como o padrão de distribuição da heterocromatina constitutiva (banda CBG), banda GTG e coloração AgNOR (NORs). O Mockingbird sob giz possui 2n = 80. O primeiro e o quarto pares são submetacêntricos e o segundo, terceiro e quinto são acrocêntricos. Os pares cromossômicos restantes do complemento têm morfologia telocêntrica. O cromossomo Z é submetacêntrico e o W é metacêntrico. A banda CBG mostrou coloração positiva na região pericentromérica da maioria dos macrocromossomos e microcromossomos e também no cromossomo Z, diferentemente do cromossomo W, que parecia totalmente heterocromático. A banda GTG foi semelhante à Gallus gallus e em outras espécies que já foram banda GTG. As NORs foram identificadas em um par de microcromossomos caracterizados por apresentar uma constrição secundária notável. Isso pode ser considerado uma característica plesiomórfica para M. saturninus, uma vez que os grupos de linha de base (Paleognathae) também mostraram um par de microcromossomos contendo NORs.

Abstract

The domestic buffaloes were divided in two groups: the river buffalo, which has chromosome number 2n=50 and the swamp buffalo, 2n=48. The crosses between these animals result in F1 specimens with 2n=49 which are viable and fertile. In Brazil the Baio type and the Carabao breed buffalo are found in small number and maintained in genetic conservation programs that aim to keep these groups separated, this way conserving their genetic heritage in situ as well as ex situ. The objective of this study was to evaluate cytogenetically buffaloes from Carabao breed, Baio type and its F1, F2, F3 and F4 crossbreeds kept in a genetic conservation program. The metaphases were obtained from blood culture of 50 animals Carabao breed, 45 Baio type and 10 animals belonging to the crossbreed progeny. The Baio type presented 2n=50 and the Carabao breed 2n=48. In the progeny were observed specimens with chromosome number 2n=48 or 2n=49 with morphological variations in the first pair of chromosomes. The diploid number 2n=49 confirms crossbreeding of the animals from the conservation program. Thus, the exclusion of these animals from the original herds is highly recommended in order to keep separate genotypes for each genetic group., Os búfalos domésticos são divididos em dois grupos: os búfalos de rio, com número diploide 2n= 50 e os búfalos de pântano, 2n= 48. O cruzamento F1 entre estes resultam em animais com 2n= 49, que são viáveis e férteis. No Brasil os búfalos do tipo Baio e da raça Carabao são mantidos em um programa de conservação genética que visa manter esses grupos separados, conservando seu patrimônio genético in situ e ex situ. O objetivo deste estudo foi avaliar citogeneticamente búfalos da raça Carabao, tipo Baio e suas crusas F1, F2, F3 e F4 mantidos em um programa de conservação genética. As metáfases foram obtidas a partir de cultura de linfócitos de 50 animais da raça Carabao, 45 do tipo Baio e 10 descendentes de uma fêmea mestiça. Os animais do tipo Baio apresentaram 2n= 50, os da raça Carabao 2n= 48. Para os descendentes foram observados 2n= 48 e 2n= 49, com variações morfológicas no primeiro par. O número diplóides 2n= 49 confirma a presença de animais cruzados no programa de conservação. Assim, a exclusão destes animais e seus descendentes é altamente recomendada dos rebanhos originais a fim de manter os genótipos separados para cada grupo genético.
