Chinese Kunming dog (CKD) is a working breed widely used by army, customs and police across the country. To better evaluate a breeding population's potential working performance and implement selective breeding for future offspring, a temperament test with five subsets, including following to trainer (FO), reaction to stranger (RS), tease by towel (TT), chase and fetch (CF) and tug of war (TW), was annually operated by the Kunming Police Dog Base since 2019. To fully utilize this behavioral data, 92 breeding animals of CKDs were genotyped by a 50K SNP chip, and a GWAS method supporting repeated measurement was adopted to estimate heritabilities and explore genetic variants associated with the five subsets. The heritabilities estimated for the five subsets ranged from 0,03 to 0.14, which showed a possibility for genetic improvement. Meanwhile, in total seven novel SNPs were identified associated with subsets FO, TT, CF and TW. In addition to that five of the seven loci are separately located within ACOT11, PRDM16, SAMD12, CPA1 and ENSCAFG00000023173 genes. Nevertheless, genetic effects of these novel loci on dog temperament should be further validated by a larger dataset in the future.

Summary: The canine transmissible venereal tumor (CTVT) is a clonal cell-mediated cancer with a long evolutionary history and extensive karyotype rearrangements in its genome. However, little is known about its genetic similarity to human tumors. Here, using multi-omics data we identified 11 germline gene fusions (GGFs) in CTVT, which showed higher genetic susceptibility than others. Additionally, we illustrate a mechanism of a complex gene fusion of three gene segments (HSD17B4-DMXL1-TNFAIP8) that we refer to “greedy fusion”. Our findings also provided evidence that expressions of GGFs are downregulated during the tumor regressive phase, which is associated with DNA methylation level. This study presents a comprehensive landscape of gene fusions (GFs) in CTVT, which offers a valuable genetic resource for exploring potential genetic mechanisms underlying the development of cancers in both dogs and humans.

Abstract Background The international Dog10K project aims to sequence and analyze several thousand canine genomes. Incorporating 20 × data from 1987 individuals, including 1611 dogs (321 breeds), 309 village dogs, 63 wolves, and four coyotes, we identify genomic variation across the canid family, setting the stage for detailed studies of domestication, behavior, morphology, disease susceptibility, and genome architecture and function. Results We report the analysis of > 48 M single-nucleotide, indel, and structural variants spanning the autosomes, X chromosome, and mitochondria. We discover more than 75% of variation for 239 sampled breeds. Allele sharing analysis indicates that 94.9% of breeds form monophyletic clusters and 25 major clades. German Shepherd Dogs and related breeds show the highest allele sharing with independent breeds from multiple clades. On average, each breed dog differs from the UU_Cfam_GSD_1.0 reference at 26,960 deletions and 14,034 insertions greater than 50 bp, with wolves having 14% more variants. Discovered variants include retrogene insertions from 926 parent genes. To aid functional prioritization, single-nucleotide variants were annotated with SnpEff and Zoonomia phyloP constraint scores. Constrained positions were negatively correlated with allele frequency. Finally, the utility of the Dog10K data as an imputation reference panel is assessed, generating high-confidence calls across varied genotyping platform densities including for breeds not included in the Dog10K collection. Conclusions We have developed a dense dataset of 1987 sequenced canids that reveals patterns of allele sharing, identifies likely functional variants, informs breed structure, and enables accurate imputation. AIM: To investigate the expression changes of MMP-12 during the long-term axon regeneration induced by the lens injury after the optic nerve clamp trauma in sprague-dawley(SD)rats.METHODS: The optic nerve injury model and lens injury model of SD rats were established, and the 24 experimental animals were divided into control group; lens injury group; optic nerve injury group; lens injury combined with optic nerve injury group, with 6 rats in each group. Reference transcriptome sequencing was used to analyze the expression changes of differentially expressed genes in the injured optic nerve region, and relevant differentially expressed genes with high expression were screened. Quantitative real-time polymerase chain reaction(qRT-PCR)and enzyme-linked immunosorbent assay(ELISA)were used to quantify the expression changes of matrix metalloproteinase-12(MMP-12)in the injured optic nerve region.RESULTS: The Principal Component Analysis of transcriptome sequencing indicated that lens injury combined with optic nerve injury was the principal component of gene expression change. Analysis of gene expression differences showed that the expression of MMP-12 gene was up-regulated in the lens injury combined with optic nerve injury group. Objective The percutaneous IS screws and the minimally invasive percutaneous plate are the most popular internal methods for Zone II unstable sacral fractures. However, the choice of fixation remains controversial for orthopaedic surgeons. The purpose of study was to evaluate and compare the clinical results of percutaneous iliosacral (IS) screw fixation under three‐dimensional (3D) navigation and minimally invasive percutaneous plate fixation in the treatment of Zone II unstable sacral fractures. Methods A retrospective study was performed, including 64 patients with Zone II unstable sacral fractures who underwent percutaneous IS screw fixation under 3D navigation (navigation group) and minimally invasive percutaneous plate fixation (plate group) from January 2011 and March 2021 in our department. The age, gender, fracture type, mechanism of injury, injury severity score (ISS), time from admission to operation, operative time, intraoperative blood loss, hospital stay, incision length, follow‐up time, time to clinical healing, and complications were recorded and analyzed. Matta standard was used to assess fracture reduction outcomes. The Majeed function system assessed functional outcomes at the last follow‐up. Results The average follow‐up time was (14.42 ± 1.57) months in the navigation group and (14.79 ± 1.37) months in the plate group. No statistical difference between the two groups in age, gender, fracture type, mechanism of injury, ISS, time from admission to operation, and time to clinical healing. Social isolation (SI) exerts diverse adverse effects on brain structure and function in humans. To gain an insight into the mechanisms underlying these effects, we conducted a systematic analysis of multiple brain regions from socially isolated and group-housed dogs, whose brain and behavior are similar to humans. Our transcriptomic analysis revealed reduced expression of myelin-related genes specifically in the white matter of prefrontal cortex (PFC) after SI during the juvenile stage. Despite these gene expression changes, myelin fiber organization in PFC remained unchanged. Surprisingly, we observed more mature oligodendrocytes and thicker myelin bundles in the somatosensory parietal cortex in socially isolated dogs, which may be linked to an increased expression of ADORA2A, a gene known to promote oligodendrocyte maturation. Additionally, we found a reduced expression of blood-brain barrier (BBB) structural components Aquaporin-4, Occludin, and Claudin1 in both PFC and parietal cortices, indicating BBB disruption after SI. In agreement with BBB disruption, myelin-related sphingolipids were increased in cerebrospinal fluid in the socially isolated group. These unexpected findings show that SI induces distinct alterations in oligodendrocyte development and shared disruption in BBB integrity in different cortices, demonstrating the value of dogs as a complementary animal model to uncover molecular mechanisms underlying SI-induced brain dysfunction.

In general, there is a steep decrease in cryogenic (−196 °C) impact absorbed energy of high Mn austenitic steels after aging at 600–800 °C, which is also the temperature range of aging precipitation. Thus, the precipitation hardening is hardly used in hot-rolled high Mn austenitic steel for cryogenic application and its effect on cryogenic impact toughness is also not understood up to now. In the present work, A high Mn austenitic steel alloyed with Al and V was designed, and the effect of aging temperature on the evolution of microstructure and mechanical properties was investigated by means of electron backscattered diffraction, transmission electron microscopy and X-ray diffraction. The grain size is nearly not affected by aging temperature, exhibiting similar grain size of 6.7–8.1 μm, but the volume fraction and size of precipitated particles greatly depend on aging temperature. The average particle size slightly increases from 5.6 to 6.7 nm with increasing aging temperature from 550 to 650 °C, whereas the precipitated particles are greatly coarsened at 700 °C and the average particle size reaches 16.3 nm. These fine particles can provide a sufficient precipitation hardening of about 200 MPa, leading to a high yield strength of 587 MPa. Meanwhile, this strong precipitation hardening only slight lowers cryogenic impact absorbed energy from 132 to 97 J, also implying that the above-mentioned aging brittle effect should be suppressed or very weak.

China is rich in abundant lithium resources characterized by considerable reserves and a concentrated distribution of metallogenic zones or belts, with proven reserves of 4046.8×103 t (calculated based on Li2O) by 2021. China is also a big consumer of lithium. By 2019, China ’s lithium consumption in the battery sector alone had reached 99×103 t, with an average annual growth rate of nearly 26%. China has become the world ’s largest importer of lithium resources, showing a severely unbalanced relationship between supply and demand for lithium resources. Therefore, there is an urgent need for the prospecting, exploitation, and study of lithium resources in China. This study collected, organized, and summarized the data on the major lithium deposits in China, analyzed and compared the spatial-temporal distribution patterns, geological characteristics, and metallogenic regularity of these lithium deposits, and summarized the prospecting and research achievements over the last decade. The major lithium deposits in China are distributed in provinces and regions such as Qinghai, Jiangxi, Sichuan, Tibet, and Xinjiang. These deposits are mostly small in scale. According to different genetic types, this study divided lithium deposits into granitic pegmatite type, granite type, saline lake brine type, underground brine type, and sedimentary type, as well as new types including hot spring type and magmatic-hydrothermal type, and summarized the characteristics and key metallogenic factors of these different types of deposits. Sixteen metallogenic prospect areas of lithium deposits were delineated according to the deposit types and the distribution patterns of metallogenic belts. The paper introduced the research progress in major metallogenic models and lithium extraction techniques made over the past decade. Based on the comprehensive analysis of the prospecting potential of lithium deposits, the authors concluded that the future prospecting of lithium resources in China should focus on lithium metallogenic belts, the deep and peripheral areas of currently determined large-scale pegmatite-type lithium deposits, geophysical-geochemical anomalous areas with mineralization clues, and areas with developed large-scale low-grade associated granite-type and sedimentary lithium resources. The study aims to serve as a guide for the future prospecting of lithium deposits in China.©2022 China Geology Editorial Office.

A novel processing route involving twin-roll casting and two-stage cold rolling was proposed to manufacture Inconel 718 alloy. Due to the sub-rapid solidification rate (412.5 °C/s) of as-cast strip, the element segregation can be alleviated and the growth of Laves phase was effectively suppressed. It was also observed that the dissolution of Laves phases during subsequent homogenization was significantly accelerated because of their refined size. The corresponding homogenization time can be shortened from dozens of hours to 1 h. Additionally, the non-uniform microstructure of cold-rolled and annealed samples caused by coarse grains from homogenization treatment can be eliminated by two-stage cold rolling method. Furthermore, a large amount of granular δ phases precipitated during intermediate annealing. After double aging treatment, the final products exhibited an excellent combination of strength and elongation. The ultimate tensile strength when deformed at room temperature and at 650 °C were about 1475 and 1120 MPa, and the corresponding total elongation were ∼ 15% and ∼ 22%. These results indicated that the twin-roll casting was a promising route to produce Inconel 718 alloy.

Introduction: Thoracolumbar kyphosis (TLK) is a common feature in patients with spinal deformities. However, due to limited studies, the impacts of TLK on gait have not been reported. The objective of the study was to quantify and evaluate the impacts of gait biomechanics of patients with TLK secondary to Scheuermann’s disease.Methods: Twenty cases of Scheuermann’s disease patients with TLK and twenty cases of asymptomatic participants were recruited into this study. And the gait motion analysis was conducted.Results: The stride length was shorter in the TLK group compared to control group (1.24 ± 0.11 m vs. 1.36 ± 0.21 m, p = 0.04). Compared to control group, the stride time and step time were more prolonged in the TLK group (1.18 ± 0.11s vs. 1.11 ± 0.08 s, p = 0.03; 0.59 ± 0.06 s vs. 0.56 ± 0.04 s, p = 0.04). The gait speed of the TLK group was significantly slower than it of control group (1.05 ± 0.12 m/s vs. 1.17 ± 0.14 m/s, p = 0.01); In the sagittal plane, the range of motion (ROM) of the hip in the TLK group was significantly smaller than that of the control group (37.71 ± 4.35° vs. 40.05 ± 3.71°, p = 0.00). In the transverse plane, the adduction/abduction ROMs of the knee and ankle, as well as the internal and external rotation of the knee, were smaller in TLK group than ROMs in the control group (4.66 ± 2.21° vs. 5.61 ± 1.82°, p = 0.00; 11.48 ± 3.97° vs. 13.16 ± 5.6°, p = 0.02; 9.00 ± 5.14° vs. 12.95 ± 5.78°, p = 0.00).Discussion: The main finding of this study was that measurements of gait patterns and joint movement of the TLK group were significantly lower than those of the control group. And these impacts have the potential to exacerbate degenerative progress of joints in the lower extremities. These abnormal features of gait can also serve as a guideline for physicians to focus on TLK in these patients.

Abstract Background Posterior pilon fracture is a relatively common clinical fracture involving the posterior articular surface of the distal tibia. Currently, this form of fracture is receiving increasing attention. The surgical approach and technique for the treatment of posterior pilon fractures are still controversial. The purpose of this retrospective study was to compare the clinical and imaging outcomes of pilon fractures after treatment with the open fibula fracture line (OFFL) surgical technique versus the traditional posterolateral approach (TPL). Methods A retrospective analysis of patients with posterior pilon fractures treated using the open fibula fracture line technique and the traditional posterolateral approach between January 2015 and March 2020. Thirty-one cases were included in the open fibula fracture line technique group and twenty-eight cases were included in the traditional posterolateral approach group. We used the Burwell-Charnley scale to assess the effectiveness of surgical repositioning. The clinical outcomes were evaluated using American Orthopaedic Foot & Ankle Society ankle-hind foot score (AOFAS) and visual analog score (VAS). Results The overall anatomic reduction rate was slightly better in the open fibula fracture line group than in the conventional posterolateral group (81% vs. 71%, p = 0.406), but there was no statistically significant difference between the two groups. There were no statistically significant differences between the two groups in terms of fracture healing time and time to full weight bearing (p > 0.05). At the final follow-up, the AOFAS functional score of the open fibula fracture line group was statistically superior to that of the conventional posterolateral group (p 0.05). Conclusion The trans-fibular fracture approach provides a better surgical option for specific types of posterior pilon fractures with a high rate of anatomic repositioning and a good near-term outcome. In this paper, we aim to investigate the (2+1)-dimensional Heisenberg ferromagnetic spin chain equation that is used to describe the nonlinear dynamics of magnets. Two recent effective technologies, namely, the variational method and subequation method, are employed to construct the abundant soliton solutions. By these two methods, diverse solutions such as the bright soliton, dark soliton, bright-dark soliton, perfect periodic soliton, and singular periodic soliton are successfully extracted. The numerical results are illustrated in the form of 3-D plots and 2-D curves by choosing proper parametric values to interpret the dynamics of wave profiles. Finally, the physical interpretation of the acquired results is elaborated in detail. The results obtained in this study are helpful to explain some physical meanings of some nonlinear physical models in electromagnetic waves.

During acute reperfusion, the expression profiles of long noncoding RNAs in adult rats with focal cerebral ischemia undergo broad changes. However, whether long noncoding RNAs are involved in neuroprotective effects following focal ischemic stroke in rats remains unclear. In this study, RNA isolation and library preparation was performed for long noncoding RNA sequencing, followed by determining the coding potential of identified long noncoding RNAs and target gene prediction. Differential expression analysis, long noncoding RNA functional enrichment analysis, and co-expression network analysis were performed comparing ischemic rats with and without ischemic postconditioning rats. Rats were subjected to ischemic postconditioning via the brief and repeated occlusion of the middle cerebral artery or femoral artery. Quantitative real-time reverse transcription-polymerase chain reaction was used to detect the expression levels of differentially expressed long noncoding RNAs after ischemic postconditioning in a rat model of ischemic stroke. The results showed that ischemic postconditioning greatly affected the expression profile of long noncoding RNAs and mRNAs in the brains of rats that underwent ischemic stroke. The predicted target genes of some of the identified long noncoding RNAs (cis targets) were related to the cellular response to ischemia and stress, cytokine signal transduction, inflammation, and apoptosis signal transduction pathways. In addition, 15 significantly differentially expressed long noncoding RNAs were identified in the brains of rats subjected to ischemic postconditioning. Nine candidate long noncoding RNAs that may be related to ischemic postconditioning were identified by a long noncoding RNA expression profile and long noncoding RNA-mRNA co-expression network analysis. Expression levels were verified by quantitative real-time reverse transcription-polymerase chain reaction. These results suggested that the identified long noncoding RNAs may be involved in the neuroprotective effects associated with ischemic postconditioning following ischemic stroke. The experimental animal procedures were approved by the Animal Experiment Ethics Committee of Kunming Medical University (approval No. KMMU2018018) in January 2018.

Abstract Background Up to now in the surgical treatment of Kümmell’s disease combined with thoracolumbar kyphosis, little research has focused on the evaluation of the imaging and clinical outcomes of restoring the normal alignment and sagittal balance of the spine. This study aimed to evaluate the short to mid-term radiographic and clinical outcomes in the treatment of Kümmell’s disease with thoracolumbar kyphosis. Methods From February 2016 to May 2018, 30 cases of Kümmell’s disease with thoracolumbar kyphosis were divided into group A and B according to whether the kyphosis was combined with neurological deficits. All of the cases underwent surgical treatment to regain the normal spinal alignment and sagittal balance. The radiographic outcomes and clinical outcomes of the cases were retrospectively evaluated. The sagittal imaging parameters including sagittal vertebral axis (SVA),thoracic kyphosis (TK),thoracolumbar kyphosis (TLK),lumbar lordosis (LL),pelvic incidence (PI),pelvic tilt (PT),and sacral slope (SS) before operation,immediately after operation,and the last follow-up of each case were measured and evaluated. The clinical results included the Oswestry Disability Index (ODI) and the Numerical Rating Scale (NRS) of the two groups. Statistical software SPSS21.0 was used to analyze the data. Results In group A: Mean SVA before operation was 75 mm and 26.7 mm at the final postoperative evaluation (P = 0.000); Mean TLK before operation was 39°, and 7.1° at the final postoperative evaluation (P = 0.000); Mean NRS before operation was 4.7, compared with 0.9 at the final postoperative evaluation (P = 0.000). In group B: Mean preoperative SVA was 62.5 mm and decreases to 30.7 mm at the final postoperative evaluation (P = 0.000); Mean TLK before operation was 33°, and 9.7° 2 years post-operation (P = 0.000); Mean NRS prior to surgery was 4.0, and 0.8 at the last follow-up evaluation (P = 0.000). The improvement of the NRS scores of groups A and B was related to the improvement of the cobb angle (P = 0.020); (P = 0.009) respectively. Conclusion In the treatment of Kümmell’s disease with thoracolumbar kyphosis,to restore the normal alignment and sagittal balance can obtain a satisfactory radiographic and clinical short and medium-term effects.

This paper gives a study to the system of ion sound and Langmuir waves (SEISLWs). Seven sets of the traveling wave solutions (TWSs) like the bright solitary (Type-1, Type-2, Type-3, Type-4), kinky bright solitary, dark solitary and periodic wave solutions are developed through three effective methods, which are simplified extended tanh-function method, He’s frequency formulation and the energy balance theory. The dynamic behaviors of the absolute, real and imaginary parts of the solutions are also plotted via the 3-D contours. The results in this work fully prove that the proposed methods are powerful tools to construct abundant TWS for the PDEs arising in physics.

In this article, the gamma function method, for the first time ever, is used to solve the nonlinear cubic-quintic Duffing oscillators. The nonlinear cubic-quintic Duffing oscillators with and without the damped and quadratic terms are considered respectively. By the gamma function method, it only needs one-step to get the approximate solution. The comparisons with the existing solutions reveal that the proposed method is simple but effective in solving the small amplitude oscillation.

This article mainly studies the vibration of the carbon nanotubes embedded in elastic medium. A new novel method called the Hamiltonian-based method is applied to determine the frequency property of the nonlinear vibration. Finally, the effectiveness and reliability of the proposed method is verified through the numerical results. The obtained results in this work are expected to be helpful for the study of the nonlinear vibration.

Two high manganese austenitic steels with different grain size were prepared to comprehensively investigate size effect on twinning and resultant cryogenic impact toughness at −196 °C. The microstructure was characterized by means of electron back-scattered diffraction, transmission electron microscopy and X-ray diffraction. It is found that the Charpy impact absorbed energy can be enhanced by 36% via increasing grain size from ~11 to ~47 μm, exhibiting an inverse size effect of cryogenic impact toughness. The major difference in deformed microstructure is that the number fraction of type III grains (intense twinning in grain interior) increases from ~16% to ~56% as the grain size increases from ~11 to ~47 μm. Moreover, it has been demonstrated that the larger grain size, inducing better Charpy impact toughness at −196 °C, is due to the following aspects: (1) stronger dynamic grain refinement; (2) higher dislocation density and wider extended dislocations contributing to stronger strain hardening and better plasticity as well as (3) relatively homogeneous plastic deformation.

This paper provides an investigation on nonlinear dynamic behaviors of the (3+1)-dimensional B-type Kadomtsev—Petviashvili equation, which is used to model the propagation of weakly dispersive waves in a fluid. With the help of the Cole—Hopf transform, the Hirota bilinear equation is established, then the symbolic computation with the ansatz function schemes is employed to search for the diverse exact solutions. Some new results such as the multi-wave complexiton, multi-wave, and periodic lump solutions are found. Furthermore, the abundant traveling wave solutions such as the dark wave, bright-dark wave, and singular periodic wave solutions are also constructed by applying the sub-equation method. Finally, the nonlinear dynamic behaviors of the solutions are presented through the 3-D plots, 2-D contours, and 2-D curves and their corresponding physical characteristics are also elaborated. To our knowledge, the obtained solutions in this work are all new, which are not reported elsewhere. The methods applied in this study can be used to investigate the other PDEs arising in physics.

Tea is an important beverage crop with a large and heterozygous genome. Here, the authors assemble the genome of the cultivar Longjing 43 and conduct a population genetics study to reveal divergent selection for disease resistance and flavor between the two variety groups.

Abstract Objective The data provided herein represent the whole-genome resequencing data related to three wolves and three Iranian local dogs. The understanding of genome evolution during animal domestication is an interesting subject in genome biology. Dog is an excellent model for understanding of domestication due to its considerable variety of behavioral and physical traits. The Zagros area of current day Iran has been identified as one of the initial centers of animal domestication. The availability of the complete genome sequences of Iranian local canids can be a valuable resource for researchers to address questions and testing hypotheses on the dog domestication process. Data description We collected blood samples from six Iranian local canids including two hunting dogs (Saluki breed), a mastiff dog (Qahderijani ecotype) and three wolves. We extracted genomic DNA from blood samples. Sequence data were produced using the Illumina HiSeq 2500 system. All sequence data are available in the National Genomics Data Center (NGDC), Genome Sequence Archive (GSA) database under the accession of CRA001324 and the National Center for Biotechnology Information (NCBI) under the accession of PRJNA639312. The short-read sequences with the mean depth of 16X were aligned to the dog reference genome (CanFam3.1) and achieved 99% coverage of the reference assembly. The obtained information from this experiment will be useful in evolutionary biology.

Abstract Background Advances in genome technology have simplified a new comprehension of the genetic and historical processes crucial to rapid phenotypic evolution under domestication. To get new insight into the genetic basis of the dog domestication process, we conducted whole-genome sequence analysis of three wolves and three dogs from Iran which covers the eastern part of the Fertile Crescent located in Southwest Asia where the independent domestication of most of the plants and animals has been documented and also high haplotype sharing between wolves and dog breeds has been reported. Results Higher diversity was found within the wolf genome compared with the dog genome. A total number of 12.45 million SNPs were detected in all individuals (10.45 and 7.82 million SNPs were identified for all the studied wolves and dogs, respectively) and a total number of 3.49 million small Indels were detected in all individuals (3.11 and 2.24 million small Indels were identified for all the studied wolves and dogs, respectively). A total of 10,571 copy number variation regions (CNVRs) were detected across the 6 individual genomes, covering 154.65 Mb, or 6.41%, of the reference genome (canFam3.1). Further analysis showed that the distribution of deleterious variants in the dog genome is higher than the wolf genome. Also, genomic annotation results from intron and intergenic regions showed that the proportion of variations in the wolf genome is higher than that in the dog genome, while the proportion of the coding sequences and 3′-UTR in the dog genome is higher than that in the wolf genome. The genes related to the olfactory and immune systems were enriched in the set of the structural variants (SVs) identified in this work. Conclusions Our results showed more deleterious mutations and coding sequence variants in the domestic dog genome than those in wolf genome. By providing the first Iranian dog and wolf variome map, our findings contribute to understanding the genetic architecture of the dog domestication.

Summary: Although gray wolves (Canis lupus) are one of the most widely distributed terrestrial mammals, their origins in China are not well understood. We sequenced six specimens from wolf skins, showing that gray wolves from Southern China (SC) derive from a single lineage, distinct from gray wolves from the Tibetan Plateau and Northern China, suggesting that SC gray wolves may form a distinct subpopulation. Of SC gray wolves, one wolf from Zhejiang carries a genetic component from a canid and had gene flow from a population related to or further diverged from wolves than the dhole. This may indicate that interspecific gene flow likely played an important role in shaping the speciation patterns and population structure in the genus Canis. Burdock roots are rich in inulin-type burdock fructooligosaccharide (BFO). However, the antidiabetic effect of BFO remains unknown. In this study, BFO was purified using DEAE-52 and G-75. The purity of BFO was 99.4%, the peak-average molecular weight (Mp) was 2958 Da. BFO inhibited α-glucosidase activity with an IC50 of 0.4996 mg/mL. Streptozotocin (STZ) and high-fat diet (HFD) induced diabetic mice were used to test the anti diabetic effect of BFO. BFO reduced fasting blood glucose (FBG) levels, relieved rapid weight loss, and improved glucose tolerance after six weeks of treatment. In addition, BFO reduced serum triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), reactive oxygen species (ROS), malondialdehyde (MDA), and nitric oxide synthetase (NOS) levels, while increasing superoxide dismutase (SOD) and high-density lipoprotein cholesterol (HDL-C) levels. BFO also reduced islet, liver, and kidney damage. Based on these findings, BFO possesses potent antidiabetic effects on diabetic mice.

A 6063 aluminum alloy thin slab with a cross-section of 260 × 40 mm2 was prepared by a vertical bending continuous casting (VC) process. The effects of homogenization, hot rolling and subsequent heat treatments on the microstructure and properties of the as-cast slabs and as-rolled sheets were investigated by scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), field emission electron probe (EPMA), macro- and micro-structure observation and tensile tests. The results show that the as-cast structure of AA 6063 was dominated by fine and uniform equiaxed grains. After homogenization, all Mg2Si phases dissolved back, and the Fe-containing intermetallic phases changed from acicular β-Al5FeSi phase to spherical α-Al8Fe2Si phase. Homogenizing heat treatment before hot rolling can improve the mechanical properties of the alloy. However, in the case of direct rolling without homogenization, the alloy still has good mechanical properties; the strength and plasticity are comparable to that obtained through traditional direct chill (DC) casting, homogenizing and extruding processes, indicating that the VC process has the potential to realize continuous casting and rolling.

In this paper, a novel and simple approach called the variational direct method (VDM) is applied to construct the exact traveling wave solutions of the system of ion sound and Langmuir waves (SEISLWs), which is under the influence of the ponderomotive force caused by a nonlinear force experienced by a charged particle in an inhomogeneous oscillating electromagnetic field due to high-frequency field. These exact solutions include the bright soliton, bright-like soliton, kinky-bright soliton, bright-dark soliton and periodic wave solutions, which are all presented through the numerical results in the form of 3-D plots. Obtained results show that the VDM is simple, straightforward and powerful, which is expected to open new perspectives for the traveling wave theory.

The soliton solutions of the (2 + 1)-dimensional nonlinear electrical transmission line equation are discussed by Md. Abdul Kayum, et al. (Results in Physics, Volume18, 2020, 103269). In this paper, we obtain its periodic solution by the variational method, which is expected to shed a light on the study of the solitary wave theory.

Aim: This study aims to evaluate the safety and efficacy of fluorodeoxyglucose positron emission tomography/computed tomography ( 18 F-FDG-PET/CT) guided intensity-modulated radiation therapy (IMRT) for patients with peritoneal metastases. Patients and Methods: A total of 55 patients with peritoneal metastases were treated with 18F-FDG-PET/CT-guided IMRT (BIMRT) from January 2012 to January 2019. They were prescribed with a fraction of the median dose of 2 Gy to a total dose of 50.4 Gy. The multivariate analysis was used the Cox proportional hazard model and the Kaplan-Meier plot was used to perform local control rate (LCR), progression-free survival (PFS), and overall survival (OS) analysis. Results: The 1-year, 2-year, and 3-year LCR were 72.7%, 36.4%, and 9.1%, respectively; the 1-year, 2-year, and 3-year PFS were 69.1%, 30.9%, and 7.3%, respectively, and the median PFS time was 18 months. The 1-year, 3-year and 5-year OS were 70.9%, 28.7%, and 4.2%, respectively. Based on the multivariate analysis using the Cox proportional hazard model, the Karnofsky performance status (KPS) score and radiotherapy joint chemotherapy (RJC) method were independent prognostic-related indicators (P < 0.0001). Conclusion: BIMRT may be a safe and effective treatment for patients with peritoneal metastases, especially for patients who cannot undergo surgery. In addition, the results indicated that the patient’s KPS score and RJC method were independent prognostic-related indicators for patients survival time.

Context: Garcinia mangostana Linn. (Guttiferae) pericarp is used as a traditional medicine in South Asia to treat inflammatory diseases. Objective: This study investigates therapeutic effects of G. mangostana pericarp ethanol extract (MAN) on collagen-induced arthritis (CIA) and interactions with methotrexate in vivo. Materials and methods: Male Sprague-Dawley rats with CIA were treated with MAN (0.5 g/kg/day), methotrexate (0.5 mg/kg, bw) or combination of both for 36 days, respectively (n = 8/group). Another eight healthy and CIA rats served as normal and model control, respectively. Therapeutic effects were evaluated based on paw edema and arthritis score during the experiment and serological markers at the end of the study period. Histological and radiological examinations were used to assess joint destructions. The immune status was investigated by immunohistochemistry and flow cytometry. The domestic dog has evolved to be an important biomedical model for studies regarding the genetic basis of disease, morphology and behavior. Genetic studies in the dog have relied on a draft reference genome of a purebred female boxer dog named “Tasha” initially published in 2005. Derived from a Sanger whole genome shotgun sequencing approach coupled with limited clone-based sequencing, the initial assembly and subsequent updates have served as the predominant resource for canine genetics for 15 years. While the initial assembly produced a good-quality draft, as with all assemblies produced at the time, it contained gaps, assembly errors and missing sequences, particularly in GC-rich regions, which are found at many promoters and in the first exons of protein-coding genes. Here, we present Dog10K_Boxer_Tasha_1.0, an improved chromosome-level highly contiguous genome assembly of Tasha created with long-read technologies that increases sequence contiguity >100-fold, closes >23,000 gaps of the CanFam3.1 reference assembly and improves gene annotation by identifying >1200 new protein-coding transcripts. The assembly and annotation are available at NCBI under the accession GCF_000002285.5.

In this paper, two low carbon microalloyed steels, named as steel A and steel B, were fabricated by ultra fast cooling (UFC). In both steels, the microstructures containing quasi polygonal ferrite (QF), acicular ferrite (AF) and granular bainite (GB) can be obtained by UFC process. The amount of AF in steel B is more than that in steel A. The size and distribution of precipitates (Nb/Ti carbonitrides) in steel B are finer and more dispersed than those of in steel A due to relatively low finish cooling temperature. The mechanical properties of both steels are effectively enhanced by UFC process. UFC process produces low-temperature transformation microstructures containing a significant amount of AF. The mechanical properties of steel B were more satisfactory than those of steel A due to the finer average grain size, the greater amount of the volume fractions and smaller size of secondary phases.

Heavy gauge pipeline steels experience a low qualification in drop-weight-tear test properties because of the low cooling capability of conventional thermomechanical controlled processing. To solve this problem, a new-generation thermomechanical-controlled processing technology based on ultrafast cooling was applied to prepare heavy gauge pipeline steels. The microstructure, strengthening and toughening mechanisms of 25.4 mm X70 and 22 mm X80 pipeline steels that were processed by ultrafast cooling were studied. The microstructures of the 25.4 mm X70 and 22 mm X80 pipeline steels consisted of bainitic ferrite, M-A island and acicular ferrite with a large fraction above 85%. The grain size and high-angle grain boundary fraction of X70 pipeline steel were 2.7 μm and 43%, respectively, whereas those of the X80 pipeline steel were 2.4 μm and 45%, respectively. The strengthening and toughening mechanisms were studied for the ultrafast cooling method. The main strengthening mechanism for 25.4 mm X70 pipeline steel was solution and grain-refining strengthening and precipitation strengthening with contributions of ~456 MPa and ~90.5 MPa, respectively. In the 22 mm X80 pipeline steel, the main strengthening mechanism was the solution and grain-refining strengthening, and dislocation strengthening with contributions of ~475 MPa and ~109.8 MPa, respectively.

Spicy foods elicit a pungent or hot and painful sensation that repels almost all mammals. Here, we observe that the tree shrew (Tupaia belangeri chinensis), which possesses a close relationship with primates and can directly and actively consume spicy plants. Our genomic and functional analyses reveal that a single point mutation in the tree shrew's transient receptor potential vanilloid type-1 (TRPV1) ion channel (tsV1) lowers its sensitivity to capsaicinoids, which enables the unique feeding behavior of tree shrews with regards to pungent plants. We show that strong selection for this residue in tsV1 might be driven by Piper boehmeriaefolium, a spicy plant that geographically overlaps with the tree shrew and produces Cap2, a capsaicin analog, in abundance. We propose that the mutation in tsV1 is a part of evolutionary adaptation that enables the tree shrew to tolerate pungency, thus widening the range of its diet for better survival.

Ctenolophon englerianus Mildbr. is endemic to West Africa. The wood of this species is very strong, and is widely used as building material in local regions. In this study, we determined its complete plastome sequence. This is the first reported complete plastome sequence in the family Ctenolophonaceae. The plastome of C. englerianus was found to possess a total length of 161,553 bp containing two inverted repeats (IRs) of 27,469 bp, a large single copy (LSC) region of 89,386 bp, and a small single copy (SSC) region of 17,229 bp. The plastome contains 110 unique genes, consisting of 76 protein-coding genes, 30 tRNA genes and 4 rRNA genes. The rpl32, rps16 and infA genes were lost. To validate the phylogenetic relationships of C. englerianus in Malpighiales, we have selected seven representative families from three major clades of Malpighiales to construct phylogenetic tree. According to the phylogenetic topologies, C. englerianus has a close relationship with Erythroxylum novogranatence.

Advanced glycation end products (AGEs), the direct modulators of β-cells, have been shown to cause insulin-producing β-cell dysfunction and apoptosis through increase of intracellular reactive oxygen species (ROS) production. Sesamin has been demonstrated to possess antioxidative activity. This study was designed to investigate whether sesamin protects against AGEs-evoked β-cell damage via its antioxidant property. The effects of sesamin were examined in C57BL/6J mice and MIN6 cell line. In in vivo studies, mice were intraperitoneally injected with AGEs (120 mg/kg) and orally treated with sesamin (160 mg/kg) for four weeks. Intraperitoneal glucose tolerance and insulin releasing tests were performed. Insulin content, ROS generation and β-cell apoptosis in pancreatic islets were also measured. In in vitro studies, MIN6 cells were pretreated with sesamin (50 or 100 μM) and then exposed to AGEs (200 mg/L) for 24 h. Insulin secretion, β-cell death, ROS production as well as expression and activity of NADPH oxidase were determined. Sesamin treatment obviously ameliorated AGE-induced β-cell dysfunction and apoptosis both in vivo and in vitro. These effects were associated with decreased ROS production, down-regulated expression of p67phox and p22phox, and reduced NADPH oxidase activity. These results suggest that sesamin protects β-cells from damage caused by AGEs through suppressing NADPH oxidase-mediated oxidative stress.

Background/Aims: Tendon injuries are common, difficult to cure and usually healed with fibrosis and scar tissue. The aim of this study was to evaluate tendon derived stem cells (TDSCs) and platelet rich plasma (PRP) in the treatment of collagenase induced Achilles tendinopathy in rat. Methods: Four and 8 weeks (n=18) after TDSCs, PRP, PRP with TDSC or PBS (control) injection into collagenase or saline (sham) injected rat Achilles tendon, tendon tissue was harvested and tendon quality was evaluated by histology and biomechanical testing. TDSCs were cultured and treated by 10% PRP, and the FAK/ERK1/2 signaling pathway and tenocyte-related genes were detected by western blot analysis. Results: Compared to the control, PRP treatment resulted in better healing of injured tendons with improved histological outcomes and biomechanical functions. The addition of TDSCs to PRP treatment significantly enhanced the effects of PRP treatment alone. TDSC injection alone had little effect on tendon healing. PRP and PRP with TDSC treatments of collagenase induced tendon injuries also increased the mRNA and protein expression of tenocyte-related genes (type I collagen, SCX, Tenascin C) and activated the focal adhesion kinase (FAK) and extracellular-regulated kinase (ERK) 1/2 signaling pathways. Treatment of TDSCs in vitro with 10% PRP significantly increased the phosphorylation levels of FAK and ERK1/2 and the protein levels of tenocyte-related genes (Col I, SCX and Tenascin C). Inhibition of the FAK and ERK1/2 signaling pathways abolished the effect of PRP. Conclusion: This study concludes that PRP combined with TDSCs is potentially effective for the treatment of tendinopathy. The PRP induced, FAK and ERK1/2 dependent activation of tenocyte related genes in TDSCs in vitro suggests that the beneficial healing effect of the PRP with TDSC combination might occur by means of an improved TDSC differentiation toward the tenocyte lineage. Thus, a PRP with TDSC combination therapy may be clinically useful.

Canine transmissible venereal tumour (CTVT) is a clonally transmissible cancer that originated approximately 11,000 years ago and affects dogs worldwide. Despite the clonal origin of the CTVT nuclear genome, CTVT mitochondrial genomes (mtDNAs) have been acquired by periodic capture from transient hosts. We sequenced 449 complete mtDNAs from a global population of CTVTs, and show that mtDNA horizontal transfer has occurred at least five times, delineating five tumour clades whose distributions track two millennia of dog global migration. Negative selection has operated to prevent accumulation of deleterious mutations in captured mtDNA, and recombination has caused occasional mtDNA re-assortment. These findings implicate functional mtDNA as a driver of CTVT global metastatic spread, further highlighting the important role of mtDNA in cancer evolution.

The Belgian Malinois (BM) is an excellent working dog that typically shows a circling behavior when placed in a confined space. Moreover, individuals showing moderate running in circles (one kind of obsessive compulsive behavior) in confined spaces typically show better work performance compared to those without the circling behavior or to those with a serious circling behavior (which can be defined as an obsessive compulsive disorder (OCD)). To determine whether the candidate gene CDH2, Cadherin 2, which is associated with OCD in the Doberman pinscher breed of dogs and in humans, was linked with this behavioral character in the BM, population genetic analyses were performed on a BM population and a natural population of the Chinese indigenous dog (CID). Many genetic signals of balancing selection were detected for one specific region of the CDH2 gene, which suggests that a genomic block, which is included in the CDH2 gene, experienced balancing selection in the BM, and that the CDH2 gene might be associated with the behavioral characteristics of the BM dog (a balance between circling behavior and work performance). Moreover one specific variant, G63913941A, which creates a predicted transcription factor-binding site, may be the key mutation in the CDH2 gene affecting the behavior of BMs by allowing the binding of a transcription factor and increasing CDH2 expression.

Coat color in dog breeds is an excellent character for revealing the power of artificial selection, as it is extremely diverse and likely the result of recent domestication. Coat color is generated by melanocytes, which synthesize pheomelanin (a red or yellow pigment) or eumelanin (a black or brown pigment) through the pigment type-switching pathway, and is regulated by three genes in dogs: MC1R (melanocortin receptor 1), CBD103 (β-defensin 103), and ASIP (agouti-signaling protein precursor). The genotypes of these three gene loci in dog breeds are associated with coat color pattern. Here, we resequenced these three gene loci in two Kunming dog populations and analyzed these sequences using population genetic approaches to identify evolutionary patterns that have occurred at these loci during the recent domestication and breeding of the Kunming dog. The analysis showed that MC1R undergoes balancing selection in both Kunming dog populations, and that the Fst value for MC1R indicates significant genetic differentiation across the two populations. In contrast, similar results were not observed for CBD103 or ASIP. These results suggest that high heterozygosity and allelic differences at the MC1R locus may explain both the mixed color coat, of yellow and black, and the difference in coat colors in both Kunming dog populations.

The majority of renal cell carcinomas (RCCs) are characterized by loss of function of the tumor suppressor gene von Hippel Lindau (VHL), which acts as ubiquitin ligase for hypoxia-inducible factor-1α (HIF-1α). In the absence of VHL, HIF-1α protein becomes stabilized and contributes to tumorigenesis. Recent data demonstrate the antitumor efficacy of VHL promoter in RCC cells. This study demonstrates that N-Myc downstream-regulated gene 2 (NDRG2) is a potential regulator of VHL. NDRG2 is involved in proliferation and invasion of cancer cell, furthermore it is frequently down-regulated in renal cell carcinoma. Herein we evaluated the effect of NDRG2 overexpression on proliferation and invasion in human renal cancer cells. The human renal cancer cell line 786-O and A498 were infected with Ad-NDRG2 or Ad-LacZ. Overexpression of NDRG2 not only inhibited the growth of the cells, but also suppressed the invasion. Further study showed that the tumor suppressor gene VHL were up-regulated, whereas transcription factor HIF-1a and vascular endothelial growth factor (VEGF) were down-regulated in 786-O cells infected by Ad-NDRG2. Finally, in a nude mouse model, intratumoral injections of Ad-NDRG2 every 3 days for a total of seven times significantly inhibited the growth and angiogenesis of xenografted 786-O tumors. In conclusion, these data indicate that NDRG2 may be involved in proliferation and invasion by impacting the expression of VHL and HIF-1α. NDRG2 may be an attractive therapeutic target for renal cell carcinoma.

BACKGROUND AND OBJECTIVE:The investigation concerning the B7-H1 expression in colorectal cancer cells is at an early stage. It is unclear whether B7-H1 expression may have diagnostic or prognostic value in colorectal carcinoma. Additionally, how B7-H1 is associated with the clinical features of colorectal carcinoma is not known. In order to investigate the relationship between B7-H1 and colorectal cancer, we analyzed B7-H1 expression and its effect in clinical specimens and HCT116 cells. METHODS:Paraffin-embedded specimens from 143 eligible patients were used to investigate the expression of CD274 by immunohistochemistry. We also examined whether B7-H1 itself may be related to cell proliferation, apoptosis, migration and invasion in colon cancer HCT116 cells. RESULTS:Our results show that B7-H1 was highly expressed in colorectal carcinoma and was significantly associated with cell differentiation status and TNM (Tumor Node Metastasis) stage. Patients with positive B7-H1 expression showed a trend of shorter survival time. Using multivariate analysis, we demonstrate that positive B7-H1 expression is an independent predictor of colorectal carcinoma prognosis. Our results indicate that B7-H1 silencing with siRNA inhibits cell proliferation, migration and invasion. Furthermore, cell apoptosis was also increased by B7-H1 inhibition. CONCLUSIONS:Positive B7-H1 expression is an independent predictor for colorectal carcinoma prognosis. Moreover, knockdown of B7-H1 can inhibit cell proliferation, migration and invasion.

The synchronization between fractional-order hyperchaotic systems and integer-order hyperchaotic systems via sliding mode controller is investigated. By designing an active sliding mode controller and choosing proper control parameters, the drive and response systems are synchronized. Synchronization between the fractional-order Chen chaotic system and the integer-order Chen chaotic system and between integer-order hyperchaotic Chen system and fractional-order hyperchaotic Rössler system is used to illustrate the effectiveness of the proposed synchronization approach. Numerical simulations coincide with the theoretical analysis.

In the crystal structure of the title compound, C6H6N3+·C6H2N3O7−, anions and cations are linked into chains along [010] by intermolecular N—H...O hydrogen bonds. These chains are further stabilized by weak C—H...O hydrogen bonds and π–π stacking interactions with a centroid–centroid distance of 3.908 (1) Å.

BACKGROUND: A major reduction of genetic diversity in mtDNA occurred during the domestication of East Asian pigs. However, the extent to which genetic diversity has been lost in the nuclear genome is uncertain. To reveal levels and patterns of nucleotide diversity and to elucidate the genetic relationships and demographic history of domestic pigs and their ancestors, wild boars, we investigated 14 nuclear markers (including 8 functional genes, 2 pseudogenes and 4 intergenic regions) from 11 different chromosomes in East Asia-wide samples and pooled them with previously obtained mtDNA data for a combined analysis. PRINCIPAL FINDINGS: The results indicated that domestic pigs and wild boars possess comparable levels of nucleotide diversity across the nuclear genome, which is inconsistent with patterns that have been found in mitochondrial genome. CONCLUSIONS: This incongruence between the mtDNA and nuclear genomes is suggestive of a large-scale backcross between male wild boars and female domestic pigs in East Asia. Our data reveal the impacts of founder effects and backcross on the pig genome and help us better understand the complex demographic histories of East Asian pigs, which will be useful for future work on artificial selection.
