Plant height (PH) is associated with lodging resistance and planting density, which is regulated by a complicated gene network. In this study, we identified a spontaneous dwarfing mutation in maize, m30, with decreased internode number and length but increased internode diameter. A candidate gene, ZmCYP90D1, which encodes a member of the cytochrome P450 family, was isolated by map-based cloning. ZmCYP90D1 was constitutively expressed and showed highest expression in basal internodes, and its protein was targeted to the nucleus. A G-to-A substitution was identified to be the causal mutation, which resulted in a truncated protein in m30. Loss of function of ZmCYP90D1 changed expression of hormone-responsive genes, in particular brassinosteroid (BR)-responsive genes which is mainly involved in cell cycle regulation and cell wall extension and modification in plants. The concentration of typhasterol (TY), a downstream intermediate of ZmCYP90D1 in the BR pathway, was reduced. A haplotype conferring dwarfing without reducing yield was identified. ZmCYP90D1 was inferred to influence plant height and stalk diameter via hormone-mediated cell division and cell growth via the BR pathway.

Abstract Background Branching is a plastic character that affects plant architecture and spatial structure. The trait is controlled by a variety of plant hormones through coordination with environmental signals. Plant AT-rich sequence and zinc-binding protein (PLATZ) is a transcription factor that plays an important role in plant growth and development. However, systematic research on the role of the PLATZ family in apple branching has not been conducted previously. Results In this study, a total of 17 PLATZ genes were identified and characterized from the apple genome. The 83 PLATZ proteins from apple, tomato, Arabidopsis, rice, and maize were classified into three groups based on the topological structure of the phylogenetic tree. The phylogenetic relationships, conserved motifs, gene structure, regulatory cis-acting elements, and microRNAs of the MdPLATZ family members were predicted. Expression analysis revealed that MdPLATZ genes exhibited distinct expression patterns in different tissues. The expression patterns of the MdPLATZ genes were systematically investigated in response to treatments that impact apple branching [thidazuron (TDZ) and decapitation]. The expression of MdPLATZ1, 6, 7, 8, 9, 15, and 16 was regulated during axillary bud outgrowth based on RNA-sequencing data obtained from apple axillary buds treated by decapitation or exogenous TDZ application. Quantitative real-time PCR analysis showed that MdPLATZ6 was strongly downregulated in response to the TDZ and decapitation treatments, however, MdPLATZ15 was significantly upregulated in response to TDZ, but exhibited little response to decapitation. Furthermore, the co-expression network showed that PLATZ might be involved in shoot branching by regulating branching-related genes or mediating cytokinin or auxin pathway. Conclusion The results provide valuable information for further functional investigation of MdPLATZ genes in the control of axillary bud outgrowth in apple.

Hydrological simulation in karst areas is of great importance and challenge. It is a practical way to enhance the performance of existing hydrological models in karst areas by coupling karst modules that represent hydrological processes in these areas. The near-surface critical zone structure affects runoff generation in karst areas significantly and its complex hydrological processes could be simplified with threshold behaviors. This study proposed a three-thresholds-based karst runoff generation module (3T-KRGM), which used three reservoirs to represent water storage in the soil zone, soil–epikarst interface, and epikarst zone. The 3T-KRGM is coupled with the Xinanjiang (XAJ) model to extend the applicability of the model to karst areas. Both the improved XAJ model and the original XAJ model were used in the Shibantang watershed, which is a typical karst watershed located in southwest China. The results indicate that the performance of daily discharge simulations was obviously improved by introducing the 3T-KRGM. In addition, both the parameter sensitivity analysis and baseflow simulation demonstrate that the 3T-KRGM is rational in structure. The 3T-KRGM could also be easily coupled into other hydrological models, thus benefiting the hydrological simulation in karst areas. HIGHLIGHTS We proposed a three-thresholds-based karst runoff generation module to improve model performance in karst areas.; The theoretical basis of the module is a modification of a three-thresholds mechanism revealed by hillslope experiments.; We coupled the module into the Xinanjiang model to extend its applicability in karst areas.; The advantages of the module for hydrological simulation were verified from various aspects.;

Temporal modeling is the essential to achieve video super-resolution. Most models use alignment or recurrent methods to directly exploit the temporal information of consecutive frames. However, the feature information extracted directly from the input frames is coarse, which affects the performance and generalization ability of the model. Thus, in this paper, we propose to investigate the role of recurrent feature updating networks. The feature update module is proposed to update and optimize the input information to improve the accuracy of features. We design the model as a multi-stage form to improve performance and generalization. Moreover, to further improve the performance of the model, we propose the difference supplement module. It allows the model to exploit the temporal differences between groups to complement the missing information in the output of each stage. Experiments demonstrate that our proposed model achieves 27.78 dB, 30.44 dB, and 39.38 dB on Vid4, SPMCS, and UDM10, respectively, which indicates that our proposed model achieves state-of-the-art performance on video super-resolution. The precision instruments and equipment are often utilized in low-frequency and micro-amplitude vibration systems, in which many vibration isolators of rubber materials are widely used. Ignoring the low-frequency amplitude will result in errors in the fatigue life design of the vibration isolators and predicting the dynamic response of each frequency band accurately becomes necessary. However, integer-order models cannot describe the frequency dependence of rubber materials, while the fractional-order models can describe it instead. On the other hand, the elastic restoring force is strongly nonlinear under large deformation, and the vibration of the nonlinear system contains multiple harmonic components. In order to solve those issues, the fractional nonlinear Nishimura model is used to characterize the constitutive relation of vibration isolators such as air springs, which are made of carbon black filled natural rubber. The high-order harmonic balance method is used to obtain the steady-state response of the vibration system, while the fourth-order Runge–Kutta method is applied to simulate the dynamic response of the system in the low-frequency region, and the Lyapunov exponent is used to determine the stability of the system. Furthermore, the influence of parameters on the amplitude–frequency characteristics of different frequency bands is also studied, and a method to solve the optimal damping coefficient is proposed based on the primary resonance amplitude–frequency curves. The results show that there is a diversity of periodic motions in the process of adjacent super-harmonic resonance transition. Numerical simulations also demonstrate that multi-periodic motions coexist in the system. The motion transition law between the polymorphic coexistence region and its adjacent regions is summarized.

Abstract Ammopiptanthus mongolicus is the only evergreen broad-leaved shrub present in arid areas of Northwest China and plays an important role in maintaining the stability of the local desert ecosystem. It can survive under extreme temperatures (e.g., extreme low temperature: − 24.8 °C and extreme high temperature: 37.7 °C). To understand the gene expression-physiological regulation network of A. mongolicus in extreme temperature environments, we monitored the changes in gene expression and photosynthetic traits of the leaves. The results showed that at low temperatures, the net photosynthetic rates (A), Fv'/Fm' and electron transport rate (ETR) decreased, the Fv/Fm ratio was only 0.32, and the proportion of nonregulatory heat dissipation Y(NO) increased. Based on a KEGG analysis of the differentially expressed genes, 15 significantly enriched KEGG pathways were identified, which were mainly related to circadian rhythm, photosynthesis, lipid metabolism, carbohydrate metabolism, plant hormones and other life activities. At high temperatures, the A value increased, and the proportion of regulatory energy dissipation Y(NPQ) increased. The KEGG analysis identified 24 significantly enriched KEGG pathways, which are mainly related to circadian rhythm, carbon sequestration of photosynthesis, carotenoid biosynthesis, secondary metabolites, cofactors and vitamin metabolism. In general, at the expense of photosynthesis, A. mongolicus can ensure the survival of leaves by increasing Y(NO) levels, regulating the circadian rhythm, increasing the synthesis of unsaturated fatty acids and changing the role of plant hormones. Under high-temperature stress, a high photosynthetic capacity was maintained by adjusting the stomatal conductance (gsw), increasing Y(NPQ), consuming excess light energy, continuously assembling and maintaining PSII, and changing the production of antioxidants.

Effective aggregation of temporal information of consecutive frames is the core of achieving video super-resolution. Many scholars have utilized structures such as sliding windows and recurrences to gather the spatio-temporal information of frames. However, although the performances of constructed video super-resolution models are improving, the sizes of the models are also increasing, exacerbating the demand on the equipment. Thus, to reduce the stress on the device, we propose a novel lightweight recurrent grouping attention network. The parameters of this model are only 0.878 M, which is much lower than the current mainstream model for studying video super-resolution. We have designed a forward feature extraction module and a backward feature extraction module to collect temporal information between consecutive frames from two directions. Moreover, a new grouping mechanism is proposed to efficiently collect spatio-temporal information of the reference frame and its neighboring frames. The attention supplementation module is presented to further enhance the information gathering range of the model. The feature reconstruction module aims to aggregate information from different directions to reconstruct high-resolution features. Experiments demonstrate that our model achieves state-of-the-art performance on multiple datasets.

Apple scar skin viroid (ASSVd) can infect apple trees and cause scar skin symptoms. However, the associated physiological mechanisms are unclear in young saplings. In this study, ASSVd-infected and control ‘Odysso’ and ‘Tonami’ apple saplings were examined to clarify the effects of ASSVd on apple tree growth and physiological characteristics as well as the leaf metabolome. The results indicated that leaf ASSVd contents increased significantly after grafting and remained high in the second year. Leaf size, tree height, stem diameter, branch length, and leaf photosynthetic efficiency decreased significantly in viroid-infected saplings. In response to the ASSVd infection, the chlorophyll a and b contents decreased significantly in ‘Odysso’, but were unchanged in ‘Tonami’. Moreover, the N, P, K, Fe, Mn, and Ca contents decreased significantly in the leaves of viroid-infected ‘Odysso’ or ‘Tonami’. Similarly, the CAT and POD contents decreased significantly in the viroid-infected saplings, but the SOD content increased in the viroid-infected ‘Tonami’ saplings. A total of 15 and 40 differentially abundant metabolites were respectively identified in the metabolome analyses of ‘Odysso’ and ‘Tonami’ leaves. Specifically, in the viroid-infected ‘Odysso’ and ‘Tonami’ samples, the L-2-aminobutyric acid, 6″-O-malonyldaidzin, and D-xylose contents increased, while the coumarin content decreased. These metabolites are related to the biosynthesis of isoflavonoids and phenylpropanoids as well as the metabolism of carbohydrates and amino acids. These results imply that ASSVd affects apple sapling growth by affecting physiological characteristics and metabolism of apple leaves. The study data may be useful for future investigations on the physiological mechanisms underlying apple tree responses to ASSVd.

This paper uses “prolepsis,” a process of reaching into the past to inform present and future practices, to understand 12 English-as-a-second language (ESL) teachers’ practices of supporting English language learners (ELLs) through remote teaching during the COVID-19 pandemic from 2020-2021 in British Columbia and to envision some different current and future post-pandemic classroom literacies for diverse learners. Accounts of these ESL teachers’ synthetical moments of teaching and supporting ELLs during the pandemic suggest that they had to navigate “new” areas of teaching, including attending to students’ social-emotional learning (SEL), connecting with ELL parents, teaching and engaging students via technology-supported instruction, and co-teaching with mainstream teachers, on the basis of limited or no pre-pandemic experience. These insights suggest a need to widen the focus on ESL teachers’ knowledge and expertise in applied linguistics and instructional strategies to include classroom literacies in integrating SEL into ESL instruction, adopting interactive, student-driven instructional designs and practices afforded by multimodal technologies, maintaining multiple channels of communication with parents and students, and team-teaching with classroom teachers to provide tailored language support for ELLs.

Floral fragrance is an important trait that contributes to the ornamental properties and pollination of crabapple. However, research on the physiological and molecular biology of the floral volatile compounds of crabapple is rarely reported. In this study, metabolomic and transcriptomic analyses of the floral volatile compounds of standard Malus robusta flowers (Mr), and progeny with strongly and weakly fragrant flowers (SF and WF, respectively), were conducted. Fifty-six floral volatile compounds were detected in the plant materials, mainly comprising phenylpropane/benzene ring-type compounds, fatty acid derivatives, and terpene compounds. The volatile contents were significantly increased before the early flowering stage (ES), and the contents of SF flowers were twice those of WF and Mr flowers. Odor activity values were determined for known fragrant volatiles and 10–11 key fragrant volatiles were identified at the ES. The predominant fragrant volatiles were methyl benzoate, linalool, leaf acetate, and methyl anthranilate. In the petals, stamens, pistil, and calyx of SF flowers, 26 volatiles were detected at the ES, among which phenylpropane/benzene ring-type compounds were the main components accounting for more than 75% of the total volatile content. Functional analysis of transcriptome data revealed that the phenylpropanoid biosynthesis pathway was significantly enriched in SF flowers. By conducting combined analyses between volatiles and differentially expressed genes, transcripts of six floral scent-related genes were identified and were associated with the contents of the key fragrant volatiles, and other 23 genes were potentially correlated with the key volatile compounds. The results reveal possible mechanisms for the emission of strong fragrance by SF flowers, and provide a foundation for improvement of the floral fragrance and development of new crabapple cultivars.

This study was designed to examine the role of early bilingual home literacy experiences (HLE) (including parent–child shared reading, parents’ direct teaching in Chinese and English, the availability of books in both languages, and children’s access to digital devices for bilingual learning) in the biliteracy development of 66 Chinese–Canadian first graders during the COVID-19 pandemic. Descriptive analyses reveal that overall, parents report higher engagement in English than in Chinese across the four HLE measures. Parent’s engagement in bilingual HLE differs by gender, SES, and immigration status. Pearson correlational analyses of English reading, decoding, and bilingual oral receptive vocabulary reveal that the four dimensions of HLE are not strongly related to English early literacy skills but are positively related to Chinese receptive vocabulary. Finally, hierarchical regression analyses indicate that the availability of books in Chinese and parent–child shared reading in Chinese are key factors associated with Chinese receptive vocabulary score variance; the amount of time using digital devices is found to be significantly related to English reading comprehension, but not Chinese vocabulary; and parents’ direct teaching is not significant with either English early literacy skills or Chinese receptive vocabulary. These findings have important implications for parental engagement in early bilingual home literacy activities and early literacy instruction in school.

The purpose of this study is to examine preservice Chinese language teachers’ cognition in teaching intercultural communicative competence. In the study we collected data through in-depth interviews with seven preservice teachers in a Master of Education program (Teaching Chinese as a Second Language, TCSL) at a university in Hong Kong SAR, China. The findings indicated that the participants had a relatively positive attitude and inclination toward the development of students’ intercultural communicative competence, while their conceptualizations of culture tended to be static and ambiguous. In addition, the participants’ objectives in teaching intercultural communicative competence were found to be more attitude-than knowledge- or skill-oriented. The study offers valuable insights that preservice language teachers’ cognition plays a crucial role in their future professional development and calls for curricular innovations with intercultural aims in teacher education programs.

Every year, a number of Chinese undergraduates from urban teacher universities are selected as volunteer pre-service teachers to teach in schools located in underdeveloped rural areas. In this qualitative case study, the researchers explore four pre-service teachers’ 1-year experience as volunteer educators in rural schools, their communities of practice in the south and west of China, and present their reflections on the challenges, including how their responses (re)shaped their teacher identity. It is found that these pre-service teachers have built their social capital through rural teaching experience, and they have begun to construct their professional teacher identity within that transitional community of teaching practice. The paper contributes to discussions of pre-service teacher education and pre-service teacher identity construction in the context of secondary education in rural China and in other parts of the world.

Drawing on research about young children’s literacy development, this review article discusses a recent paradigmatic turn for understanding the child and childhood from human-centerism to posthumanism. Building on the new materialist tradition (e.g., Barad, 2007) and the assemblage theory of Deleuze and Guattari (1987, 1997), the posthuman lens enables researchers and educators to see children as parts of entangled networks of relationships who continuously intra-act with their peers, teachers, materials, and the other nonhuman entities and activities produced constantly by the child-material entanglements. As such, the posthumanist perspective expands the current research on early literacy by offering new possibilities for re-conceptualizing the child, the materials or resources for early literacy, and the meaning of childhood and children’s play. These new ways of seeing the child, the materials, and childhood have also generated new pedagogical practices that are material-oriented, intra-active, and flexible. The review concludes by providing directions for conducting research from a posthuman perspective in the field of early literacy education.

In this study, an investigation on the free vibration of the beam with material properties and cross section varying arbitrarily along the axis direction is studied based on the so-called Spectro-Geometric Method. The cross-section area and second moment of area of the beam are both expanded into Fourier cosine series, which are mathematically capable of representing any variable cross section. The Young’s modulus, the mass density and the shear modulus varying along the lengthwise direction of the beam, are also expanded into Fourier cosine series. The translational displacement and rotation of cross section are expressed into the Fourier series by adding some polynomial functions which are used to handle the elastic boundary conditions with more accuracy and high convergence rate. According to Hamilton’s principle, the eigenvalues and the coefficients of the Fourier series can be obtained. Some examples are presented to validate the accuracy of this method and study the influence of the parameters on the vibration of the beam. The results show that the first four natural frequencies gradually decrease as the coefficient of the radius β increases, and decreases as the gradient parameter n increases under clamped–clamped end supports. The stiffness of the functionally Timoshenko beam with arbitrary cross sections is variable compared with the uniform beam, which makes the vibration amplitude of the beam have different changes.

Global climate change has greatly influenced the ecosystems in the Tibetan Plateau. Many studies focused on the direct effects of climate warming on the headwater regions by mean temperature, while less investigating its implication for the eco-environment. To address this, the study discussed the spatial-temporal variations of the bio-related climate indicators ≥0 °C annual accumulated temperature AAT0 and its lasting days LDT0, and corresponding ≥5 °C indicators AAT5 and LDT5 on the source region of the Yellow River (SRYR). The stationarity of indicators during 1979–2018 were tested by Pettitt test, and trends checked by linear regression analysis and Mann-Kendall test. Normalized difference vegetation index NDVI (2001–2016) was adopted to detect the correlation between vegetation activities and indicators. Results show that the AAT and LDT0 exhibited significant increasing trend over the SRYR, while the LDT5 significantly increased mainly under 4000 m. Most LDT extended due to the combined efforts of the early onset and late termination of the given temperature. 1997 was detected in the abrupt change analysis of AAT0 both on the basin scale and most area, and was adopted to divide the period into two stages. The regional mean AAT0 linearly grew at a rate of 96 °C decade−1 during the entire period, and 104 °C decade−1 during the second stage. Except for a drastic jump in the areal mean values, there was a distinct upward-shift of isoline in elevation between stages. NDVI showed strong correlativity with ≥0 °C indicators on the basin scale, according to the Pearson, Spearman and Kendall correlation coefficients, ranging from 0.5 to 0.7. Spatially, the overlap area between Pearson’s γ ≥ 0.5 and linearly rising AAT0 reached 50%, which was fully covered with significantly increasing AAT0 during the recent stage. Thus the rapid growth of ≥0 °C indicators would effectively accelerate NDVI over this major alpine grasslands, especially around the eastern low regions, where indicators are higher and grow faster.

The rapid increase of Asian immigrant students in Canadian classrooms demands more systematic and increased language support to ensure all English language learners (ELLs) achieve success in school. However, research has found mixed results on the usefulness of current English as a Second Language (ESL) support programs and a growing dissatisfaction among students and parents about ESL, suggesting further investigation is needed to improve the provision of ESL in the schools. This paper details how one school and one ESL teacher responded to the needs of newly arrived Asian (i.e., Chinese) ELL students by documenting the school’s and teacher’s journey in revamping the pull-out ESL program into a culturally responsive English for Academic Purposes (EAP) program with a focus on immersion, community engagement, and a pedagogy of cultural reciprocity. The case has important implications for redesigning current ESL programs in the context of changing immigration.

Based on a conversational analysis (CA) of eight videoed EFL lessons from technology-enhanced primary classrooms in a rural suburb of a major city in China, this study examines the nature of technology-assisted practices and their influence on teacher-student interaction in the target language. The analysis revealed that the technology-assisted practices mainly served as an alternative presentation tool to meet a range of traditional pedagogical goals and facilitated minimal spontaneous language use among the students. The findings call for research to explore better pedagogical use of technology to promote students’ active language production to truly achieve educational equalization for rural students.

Half-size adenosine triphosphate-binding cassette transporter subgroup G (ABCG) genes play crucial roles in regulating the movements of a variety of substrates and have been well studied in several plants. However, half-size ABCGs have not been characterized in detail in apple (Malus × domestica Borkh.). Here, we performed a genome-wide identification and expression analysis of the half-size ABCG gene family in apple. A total of 46 apple half-size ABCGs were identified and divided into six clusters according to the phylogenetic analysis. A gene structural analysis showed that most half-size ABCGs in the same cluster shared a similar exon–intron organization. A gene duplication analysis showed that segmental, tandem and whole-genome duplications could account for the expansion of half-size ABCG transporters in M. domestica. Moreover, a promoter scan, digital expression analysis and RNA-seq revealed that MdABCG21 may be involved in root's cytokinin transport and that ABCG17 may be involved in the lateral bud development of M. spectabilis ‘Bly114’ by mediating cytokinin transport. The data presented here lay the foundation for further investigations into the biological and physiological processes and functions of half-size ABCG genes in apple. The western Pacific subtropical high (WPSH) is one of the key systems affecting the summer rainfall over the Yangtze River Valley in China. In this study, the forecasting capacity of the WPSH for summer rainfall and streamflow is evaluated based on the WPSH index (WPSHI) derived from the NCEP/NCAR reanalysis dataset. It has been found that WPSHI can identify extreme flood years with a higher skill than normal wet years. Specifically, exceedance probability forecasting based on WPSHI has higher skills for higher thresholds of rainfall. For streamflow, adding WPSHI as a predictor only enhances the skill for higher thresholds of streamflow relative to models based on antecedent streamflow. Under the same framework, performances of two postprocessing approaches for dynamical forecasts, i.e., the model output statistics (MOS) approach and the reanalysis-based (RAN) approach are compared. Hindcasts from Climate Forecast System version 2 from the National Center for Environmental Prediction (CFSv2) are used to calculate WPSHI, which is used as the predictor for rainfall and streamflow. The result shows that the RAN approach performs better than the MOS approach. This study emphasizes the fact that the forecasting skill of exceedance probability would largely depend on the selected threshold of the predictand, and this fact should be noticed in future studies in the long-term forecasting field.

Persistent educational inequity for immigrant and refugee students and their families calls for instructional practices centering on access, quality, and social justice. Drawing on two qualitative case studies, this article examines how three U.S. urban school teachers attended to the systemic inequalities and unique challenges confronting immigrant and refugee students both inside their classrooms and outside the school. Our analyses show that the teachers strategically enacted various critical instructional practices, including linguistically responsive pedagogy, translanguaging, and sociopolitically responsive pedagogy. The teachers' agentic practices have important implications for teacher education and professional development for immigrant and refugee learners in urban settings.

Shoot branching is regulated by phytohormones, including cytokinin (CK), strigolactone (SL), and auxin in axillary buds. The correlative importance of these phytohormones in the outgrowth of apple axillary buds remains unclear. In this study, the outgrowth dynamics of axillary buds of a more-branching mutant (MB) and its wild-type (WT) of Malus spectabilis were assessed using exogenous chemical treatments, transcriptome analysis, paraffin section, and reverse transcription-quantitative PCR analysis (RT-qPCR). High contents of CK and abscisic acid coincided in MB axillary buds. Exogenous CK promoted axillary bud outgrowth in the WT but not in MB, whereas exogenous gibberellic had no significant effect on bud outgrowth in the WT. Functional analysis of transcriptome data and RT-qPCR analysis of gene transcripts revealed that MB branching were associated with CK signaling, auxin transport, and SL signaling. Transcription of the SL-related genes MsMAX1, MsD14, and MsMAX2 in the axillary buds of MB was generally upregulated during bud outgrowth, whereas MsBRC1/2 were generally downregulated both in WT and MB. Exogenous SL inhibited outgrowth of axillary buds in the WT and the apple varieties T337, M26, and Nagafu 2, whereas axillary buds of the MB were insensitive to SL treatment. Treatment with N-1-naphthylphalamic acid (NPA; an auxin transport inhibitor) inhibited bud outgrowth in plants of the WT and MB. The transcript abundance of MsPIN1 was generally decreased in response to NPA and SL treatments, and increased in CK and decapitation treatments, whereas no consistent pattern was observed for MsD14 and MsMAX2. Collectively, the present results suggest that in apple auxin transport from the axillary bud to the stem may be essential for the outgrowth of axillary buds, and at least, is involved in the process of bud outgrowth.

In our study, the effects of water stress on photosynthesis and photosynthetic electron transport chain (PETC) were studied in several ways, including monitoring the change of gas exchange parameters, modulated chlorophyll fluorescence, rapid fluorescence induction kinetics, reactive oxygen species (ROS), antioxidant enzyme activities and D1 protein levels in apple leaves. Our results show that when leaf water potential (ψw) is above –1.5 MPa, the stomatal limitation should be the main reason for a drop of photosynthesis. In this period, photosynthetic rate (PN), stomatal conductance (Gs), transpiration rate (E) and intercellular CO2 concentration (Ci) all showed a strong positive correlation with ψw. Modulated chlorophyll fluorescence parameters related to photosynthetic biochemistry activity including maximum photochemical efficiency (Fv/Fm), actual photochemical efficiency of PSII (ΦPSII), photochemical quenching coefficient (qP) and coefficient of photochemical fluorescence quenching assuming interconnected PSII antennae (qL) also showed a strong positive correlation as ψw gradually decreased. On the other hand, in this period, Stern-Volmer type non-photochemical quenching coefficient (NPQ) and quantum yield of light-induced non-photochemical fluorescence quenching [Y(NPQ)] kept going up, which shows an attempt to dissipate excess energy to avoid damage to plants. When ψw was below –1.5 MPa, PN continued to decrease linearly, while Ci increased and a ‘V’ model presents the correlation between Ci and ψw by polynomial regression. This implies that, in this period, the drop in photosynthesis activity might be caused by non-stomatal limitation. Fv/Fm, ΦPSII, qP and qL in apple leaves treated with water stress were much lower than in control, while NPQ and Y(NPQ) started to go down. This demonstrates that excess energy might exceed the tolerance ability of apple leaves. Consistent with changes of these parameters, excess energy led to an increase in the production of ROS including H2O2 and O2•−. Although the activities of antioxidant enzymes like catalase (CAT), superoxide dismutase (SOD) and peroxidase (POD) increased dramatically and ascorbate peroxidase (APX) decreased in apple leaves with drought stress, it was still not sufficient to scavenge ROS. Consequently, the accumulation of ROS triggered a reduction of net D1 protein content, a core protein in the PSII reaction center. As D1 is responsible for the photosynthetic electron transport from plastoquinone A (QA) to plastoquinone B (QB), the capacity of PETC between QA and QB was considerably downregulated. The decline of photosynthesis and activity of PETC may result in the shortage of adenosine triphosphate (ATP) and limitation the regeneration of RuBP (Jmax), a key enzyme in CO2 assimilation. These are all non-stomatal factors and together contributed to decreased CO2 assimilation under severe water stress.

The headwater regions in the Tibetan Plateau play an essential role in the hydrological cycle, however the variation characteristics in the long-term precipitation and throughout-the-year apportionment remain ambiguous. To investigate the spatio-temporal variability of precipitation in the source region of the Yellow River (SRYR), different time scale data during 1979–2015 were studied based on Shannon entropy theory. Long-term marginal disorder index (LMDI) was defined to evaluate the inter-annual hydrologic budget for annual (AP) and monthly precipitation (MP), and annual marginal disorder index (AMDI) to measure intra-annual moisture supply disorderliness for daily precipitation (DP). Results reveal that the AP over the SRYR exhibits remarkable variation, with an inclination rate of 2.7 mm/year, and a significant increasing trend. The climatic trend reversed from warm–dry to warm–wet around the turn of this century. The start of the wet season has advanced from May instead of June, supported by the proportion of MP in AP and the LMDI for May are both comparable with the values during June–September. May contributes the main changes in AP, as it is the only month in the wet season which shows a significant increasing trend during 1979–2015, and has a value in the LMDI that divides the basin in half spatially, the same as AP, with a high value in the northwest and low in the southeast. The AMDI roughly rises with latitude in spatial distribution, with wetlands and glaciers disturbing the continuity of the pattern for a relatively perennial moisture supply. AP has increased on northwest high-altitude areas first and then the southern corner since the beginning of this century. Wetting is mainly attributed to the enhanced southwest monsoon and the warming-induced freeze-thaw process. Meanwhile, AMDI variation concentrated on the Zoige Plateau Wetland, the headwater corner, the summit and part of the North Slope in the Bayan Har Mountain, as a result of a single or combined effect of global climate change and human protection.

β-galactosidases are cell wall hydrolases that play an important role in fruit softening. However, PpBGALs mechanism impacting on ethylene-dependent peach fruit softening was still unclear. In this study, we found that PpBGAL4, -6, -8, -10, -16, and -17 may be required for ethylene-dependent peach softening and PpBGAL10, -16 may make a main contribution to it among 17 PpBGALs. Utilization of virus-induced gene silencing (VIGS) showed that fruits were firmer than those of the control at 4 and 6 days after harvest (DAH) when PpBGAL10 and PpBGAL16 expression was down-regulated. Suppression of PpBGAL10 and PpBGAL16 expression also reduced PpPG21 and PpPME3 transcription, and polygalacturonase (PG) and pectinmethylesterases (PME) activity. Overall, total cell wall material and protopectin slowly declined, water-soluble pectin slowly increased, and cellulose and hemicellulose was altered significantly at 4 DAH, relative to control fruit. In addition, PpACO1 expression and ethylene production were also suppressed at 4 DAH because of inhibiting PpBGAL10 and PpBGAL16 expression. These results suggested that down-regulation of PpBGAL10 and PpBGAL16 expression delays peach fruit softening by decreasing PG and PME activity, which inhibits cell wall degradation and ethylene production.

Under the impact of climate change and human activities, the stationarity of hydrometeorological extreme value series has been losing in many regions, which makes occurrence rules of hydrometeorological extreme events more complicated. In this study, the efficiencies of trend test methods such as Spearman rank correlation test and Mann-Kendall test, as well as the efficiencies of change-point test methods such as moving T test, moving rank sum test, Pettitt test, and sequential Mann-Kendall test were analyzed quantitatively through Monte Carlo simulation. Five representative level stations in the Yangtze River estuary were selected, and the methods listed above were used in the trend and change-point detection of the annual maximum tidal level records in the period of 1950–2008. It was found that obvious rising tendency existed in the annual maximum tidal level series for all these 5 stations, and year 1980 (for 3 stations) and year 1979 (for 2 stations) were statistically significant change-points. Two subseries were divided with the change-point as the dividing point for all these actual series in the stations. Frequency analyses were carried out, respectively, for all of the subseries, and the impact of nonstationary changes in annual maximum tidal levels on probability distribution was evaluated quantitatively.

BBX proteins play important roles in regulating plant growth and development including photomorphogenesis, photoperiodic regulation of flowering, and responses to biotic and abiotic stresses. At present, the genomes of seven Rosaceae fruit species have been fully sequenced. However, little is known about the BBX gene family and their evolutionary history in these Rosaceae species. Therefore, in this study total, 212 BBX genes were investigated from seven Rosaceae species (67 from Malus × domestica, 40 from Pyruscommunis, 22 from Rosa Chinesis, 20 from Prunuspersica, 21 from Fragariavesca, 22 from Prunusavium, and 20 from Rubusoccidentalis). The chemical properties, gene structures, and evolutionary relationships of the BBX genes were also studied. All the BBX genes were grouped into six subfamilies on the basis of their phylogenetic relationships and structural features. Analysis of gene structure, segmental and tandem duplication, gene phylogeny, and tissue-specific expression with the ArrayExpress database showed their diversification in function, quantity, and structure. The expression profiles of 19 MdBBX genes in different tissues were evaluated through qRT-PCR. These genes showed distinct transcription level among the tested tissues (bud, flower, fruit, stem, and leaf). Moreover, expression patterns of 19 MdBBX genes were examined during flowering induction time under flowering-related hormones and treatments (GA3, 6-BA, and sucrose). The expressions of the candidates BBX genes were affected and showed diverse expression profile. Furthermore, changes in response to these flowering-related hormones and treatment specifying their potential involvement in flowering induction. Based on these findings, BBX genes could be used as potential genetic markers for the growth and development of plants particularly in the area of functional analysis, and their involvement in flower induction in fruit plants.

Design annual runoff is a classical issue in traditional univariate hydrological frequency analysis (HFA). We developed a multivariate HFA approach for designs for a study region covering the confluence of two streams. HFA relies on the assumption that probability distribution is consistent in the past, present, and future; however, it has been asserted as incorrect under an uncertain and changing environment. A change-point was detected in our study and adopted to divide runoff into two periods, with no significant trends in all subseries. The post-change design annual runoff witnessed dramatic mean value decline by about half at four frequencies (50%, 75%, 90% and 95%), which were selected in the bivariate analysis. Probability distribution models were constructed with univariate p-III distributions through Clayton, Frank, and Gumbel copulas and independence. Frank copula showed a generally better match with observations than others. The traditional approach, adding up the same-frequency results from both tributaries independently, was disproved by the systematically smaller design values in independence model than copulas and the 40% asynchronous encounter probability. The 25.6% worst synchronous dry-dry encounter situation may be a concern for water resource managers. Consequently, multivariate HFA should prevail as design approach in terms of water resources security.

This mixed-methods study was designed to examine how different home literacy environments shape gender differences in Chinese-Canadian first graders' (N = 76) bilingual lexical attainment and literacy engagement. Quantitative analyses indicated that girls outperform boys in both Chinese (L1) and English (L2) vocabulary. Gender differences were not associated with the quantity of language exposure at home in either language but rather with the quality of home language input. Home Language Quality was found to be differentially associated with gender, with boys receiving quality input in both languages at home. Results reveal that parents engaged in more quality home literacy practices with girls than boys. Parental expectations and beliefs in gender differences, particularly girls' superiority in language learning and self-regulation, shaped gendered home literacy engagement.

Mitochondrial protein translation that is essential for aerobic energy production includes four essential steps of the mitochondrial ribosome cycle, namely, initiation, elongation, termination of the polypeptide, and ribosome recycling. Translation termination initiates when a stop codon enters the A site of the mitochondrial ribosome where it is recognized by a dedicated peptide release factor (RF). However, RFs and mechanisms involved in translation in plant mitochondria, especially in monocotyledons, remain largely unknown. Here, we identified a crumpled kernel (crk5 allele) mutant, with significantly decreased kernel size, 100-kernel weight, and an embryo-lethal phenotype. The Crk5 allele was isolated using map-based cloning and found to encode a mitochondrial localization RF2a. As it is an ortholog of Arabidopsis mitochondrial RF2a, we named the gene ZmmtRF2a. ZmmtRF2a is missing the 5th-7th exons in the crk5 resulting in deletion of domains containing motifs GGQ and SPF that are essential for release activity of RF, mitochondrial ribosome binding, and stop codon recognition. Western blot and qRT-PCR analyses indicate that the crk5 mutation results in abnormal mitochondrion structure and function. Intriguingly, we observed a feedback loop in the crk5 with up-regulated transcript levels detected for several mitochondrial ribosome and mitochondrial-related components, in particular mitochondrial complexes CI, CIV, and a ribosome assembly related PPR. Together, our data support a crucial role for ZmmtRF2a in regulation of mitochondrial structure and function in maize. [ABSTRACT FROM AUTHOR]

Abstract Salmonella, the prevailing zoonotic pathogen within the Enterobacteriaceae family, holds the foremost position in global bacterial poisoning incidents, thereby signifying its paramount importance in public health. Consequently, the imperative for expeditious and uncomplicated detection techniques for Salmonella in food is underscored. After more than two decades of development, loop-mediated isothermal amplification (LAMP) has emerged as a potent adjunct to the polymerase chain reaction, demonstrating significant advantages in the realm of isothermal amplification. Its growing prominence is evident in the increasing number of reports on its application in the rapid detection of Salmonella. This paper provides a systematic exposition of the technical principles and characteristics of LAMP, along with an overview of the research progress made in the rapid detection of Salmonella using LAMP and its derivatives. Additionally, the target genes reported in various levels, including Salmonella genus, species, serogroup, and serotype, are summarized, aiming to offer a valuable reference for the advancement of LAMP application in Salmonella detection. Finally, we look forward to the development direction of LAMP and expect more competitive methods to provide strong support for food safety applications.
