Este proyecto consistió en plantear un sistema de vermicompostaje para el aprovechamiento de los residuos orgánicos generados en el establecimiento penitenciario de mediana seguridad carcelaria Villahermosa, ubicada en la ciudad Santiago de Cali, incluyendo la implementación de una huerta urbana y actividades complementarias con las personas privadas de la libertad, con el propósito de crear un espacio para su resocialización. Por esta razón, se crearon grupos focales coordinados por los estudiantes de pasantía para el reconocimiento y profundización de la información técnica durante cada actividad. Según lo anterior, se desarrolló el primer objetivo donde se llevaron a cabo las visitas al establecimiento penitenciario para el planteamiento del sistema integrado de aprovechamiento de biorresiduos en las camas de vermicompostaje, posteriormente se observó el área del terreno, se hizo un estudio detallado de cuales biorresiduos generó el establecimiento y encontramos que los residuos generados eran aptos para el funcionamiento de las camas de vermicompostaje, se hizo la recolección y se procedió a pesar los biorresiduos para que de esta forma se pudiera cuantificar la cantidad de biorresiduos y saber cuántas camas se pueden construir y cual cantidad de humus se puede generar. Teniendo en cuenta lo anterior, se procedió a desarrollar el segundo objetivo, donde se estableció el modelo de huerta urbana que permitió el aprovechamiento del humus generado en el vermicompostaje producido por los biorresiduos. Para conocer el estado del suelo donde se ubicó la huerta urbana, se realizó un análisis de suelo en el laboratorio Centro Internacional de Agricultura Tropical (CIAT), donde se determinaron las condiciones del suelo y se obtuvo como resultado un suelo rico en materia orgánica para asimilar de manera fácil y rápida el crecimiento de las plántulas que se sembraron en esta huerta urbana con la ayuda de las personas privadas de la libertad. También, se realizaron encuestas dirigidas a las personas privadas de la libertad, en la cual se les brindó asesorías en cada fase del proyecto generando empatía y buen manejo del escenario para el proceso de resocialización, estas personas hicieron acompañamiento durante el proceso brindando ayuda al momento de la implementación, de igual manera se realizaron grupos focales en los cuales se repartieron tareas semanalmente y se les hizo acompañamiento con el fin de que con su buena conducta y buen empeño al momento de realizar las actividades, les quedara un aprendizaje más y mediante el proyecto ellos tuvieran el beneficio de ingerir los alimentos, cultivados por ellos mismos. Finalmente, se puede evidenciar que se logró en su totalidad la ejecución de los objetivos, cumpliendo con el planteamiento del sistema integrado de aprovechamiento de los biorresiduos por vermicompostaje, posteriormente, el modelo de huerta urbana cumplió con los estándares técnicos establecidos cumpliendo con el cierre de ciclos planteado para que a posteriori pueda ser utilizado el aprovechamiento del humus cuando se implementen las camas de vermicompostaje en el establecimiento, por último, se cumplió con el proceso de 16 resocialización con las actividades y grupos focales, para el cumplimiento de los objetivos se estimó la evaluación de costos, que permitió calcular el beneficio en ahorro para el establecimiento penitenciario, al ser un proyecto que aplica los principios de economía circular ya no sería necesario invertir en cierta cantidad de alimentos porque la huerta urbana los brinda por otro lado, la inversión en costos de materiales para el sistema de vermicompostaje fue de $ 10,406,729 y la huerta urbana de $ 6,879,510 los costos de inversión incluyen un ingreso económico como incentivo para las personas privadas de la libertad. Taking into account the above, we proceeded to develop the second objective, where we established the urban garden model that allowed the use of the humus generated in the vermicomposting produced by the biowaste. In order to know the state of the soil where the urban garden was located, a soil analysis was carried out at the International Center for Tropical Agriculture (CIAT) laboratory, where the soil conditions were determined and the result was a soil rich in organic matter to easily and quickly assimilate the growth of the seedlings that were planted in this urban garden with the help of the people deprived of their freedom. We also conducted surveys directed to the persons deprived of liberty, in which we provided them with advice in each phase of the project, generating empathy and good management of the scenario for the process of resocialization, these people accompanied us during the process providing help at the time of implementation, Similarly, focus groups were held in which weekly tasks were distributed and they were accompanied so that with their good behavior and good effort at the time of performing the activities, they would learn more and through the project they would have the benefit of eating the food grown by themselves. The cone photoreceptor-enriched cultures derived from embryonic chick retinas have become an indispensable tool for researchers around the world studying the biology of retinal neurons, particularly photoreceptors. The applications of this system go beyond basic research, as they can easily be adapted to high throughput technologies for drug development. However, genetic manipulation of retinal photoreceptors in these cultures has proven to be very challenging, posing an important limitation to the usefulness of the system. We have recently developed and validated an ex ovo plasmid electroporation technique that increases the rate of transfection of retinal cells in these cultures by five-fold compared to other currently available protocols(1). In this method embryonic chick eyes are enucleated at stage 27, the RPE is removed, and the retinal cup is placed in a plasmid-containing solution and electroporated using easily constructed custom-made electrodes. The retinas are then dissociated and cultured using standard procedures. This technique can be applied to overexpression studies as well as to the downregulation of gene expression, for example via the use of plasmid-driven RNAi technology, commonly achieving transgene expression in 25% of the photoreceptor population. The video format of the present publication will make this technology easily accessible to researchers in the field, enabling the study of gene function in primary retinal cultures. We have also included detailed explanations of the critical steps of this procedure for a successful outcome and reproducibility.
