1. Trapping and release of deuterium in helium-induced tungsten fuzz at low temperature

Abstract

The release of deuterium (D) atoms from a nanostructure ‘fuzz’ tungsten (W) after a sequential plasma irradiation of helium (He) and D was explored. Thermal desorption spectroscopy revealed that more than 60% of total D amount was already released at >423 K. A sophisticated measurement with an aberration-corrected scanning TEM combined with electron energy-loss spectrometer (STEM-EELS) directly showed the trapping of D atoms into He bubbles after the plasma irradiation and the release of D from the He bubbles at elevated temperature at 423 K. The He bubbles containing D were characterized by (i) relatively large volume and (ii) small He energy-loss shift below 2 eV. It is suggested that the shifts in the energy-loss peaks of He and D could be used as indicators of D absorption within He bubbles.

2. Conducting head-mounted eye-tracking research with young children with autism and children with increased likelihood of later autism diagnosis

Abstract

Abstract Background Over the past years, researchers have been using head-mounted eye-tracking systems to study young children’s gaze behaviors in everyday activities through which children learn about the world. This method has great potential to further our understanding of how millisecond-level gaze behaviors create multisensory experiences and fluctuate around social environments. While this line of work can yield insight into early perceptual experiences and potential learning mechanisms, the majority of the work is exclusively conducted with typically-developing children. Sensory sensitivities, social-communication difficulties, and challenging behaviors (e.g., disruption, elopement) are common among children with developmental disorders, and they may represent potential methodological challenges for collecting high-quality data. Results In this paper, we describe our research practices of using head-mounted eye trackers with 41 autistic children and 17 children with increased likelihood of later autism diagnosis without auditory or visual impairments, including those who are minimally or nonspeaking and/or have intellectual disabilities. The success rate in gathering data among children with autism was 92.68%. 3 of 41 children failed to complete the play-session, resulting in an 86.36% success rate among 1–4-year-olds and a 100.00% success rate among 5–8-year-olds. 1 of 17 children with increased likelihood of later autism diagnosis failed to complete the play-session, resulting in a success rate of 94.11%. There were numerous “challenging” behaviors relevant to the method. The most common challenging behaviors included taking the eye-tracking device off, elopement, and becoming distressed. Overall, among children with autism, 88.8% of 1–4-year-olds and 29.4% of 5–8-year-olds exhibited at least one challenging behavior. Conclusions Research capitalizing on this methodology has the potential to reveal early, socially-mediated gaze behaviors that are relevant for autism screening, diagnosis, and intervention purposes. We hope that our efforts in documenting our study methodology will help researchers and clinicians effectively study early naturally-occuring gaze behaviors of children during non-experimental contexts across the spectrum and other developmental disabilities using head-mounted eye-tracking. Ultimately, such applications may increase the generalizability of results, better reflect the diversity of individual characteristics, and offer new ways in which this method can contribute to the field.

3. Lithological variation of asteroid Ryugu samples returned by the Hayabusa 2 spacecraft: Assessment from the 18 particles distributed to the initial analysis “Stone' team

Abstract

JAXA’s Hayabusa2 spacecraft successfully returned~5.4g of C-type asteroid Ryugu materials on Dec. 2020 [e.g., 1] and the recovered samples were extensively analyzed by initial analysis teams and Phase2 curation teams. The reported results show that Ryugu samples are similar to CI chondritesin chemistry and mineralogy, showing evidence for aqueous alteration in the parent body [e.g., 2,3]. As a “stone” team of the initial analysis, we received 18coarse particles (>1 mm) of Ryugu samples to characterize their mineralogy and petrology. We found that the samples were breccias of mm-to-sub mm size clasts. All of the clasts are mainly composed of Mg-Fe phyllosilicates, but the alteration degree appears slightly different from one clast to another. Here we report a lithological variation of Ryugu samples to propose their reasonable lithological classification based upon different mineral assemblages and to discuss the formation and evolution of the Ryugu parent body.

Background: The Global Anticoagulant Registry in the FIELD–Atrial Fibrillation (GARFIELD-AF) is an ongoing prospective noninterventional registry, which is providing important information on the baseline characteristics, treatment patterns, and 1-year outcomes in patients with newly diagnosed non-valvular atrial fibrillation (NVAF). This report describes data from Indian patients recruited in this registry. Abstract

Debris avalanches caused by volcano sector collapse often form characteristic depositional landforms such as hummocks. Sedimentological and geomorphological analyses of debris avalanche deposits (DADs) are crucial to clarify the size, mechanisms, and emplacement of debris avalanches. We describe the morphology of hummocks on the northeastern flank of Mt. Erciyes in Kayseri, central Turkey, likely formed in the late Pleistocene. Using a remotely piloted aircraft system (RPAS) and the structure-from-motion and multi-view stereo (SfM–MVS) photogrammetry, we obtained high-definition digital elevation model (DEM) and orthorectified images of the hummocks to investigate their geometric features. We estimated the source volume of the DAD by reconstructing the topography of the volcano edifice using a satellite-based DEM. We examined the topographic cross sections based on the slopes around the scar regarded as remnant topography. Spatial distribution of hummocks is anomalously concentrated at a certain distance from the source, unlike those that follow the distance–size relationship. The high-definition land surface data by RPAS and SfM revealed that many of the hummocks are aligned toward the flow direction of the debris avalanche, suggesting that the extensional regime of the debris avalanche was dominant. However, some displaced hummocks were also found, indicating that the compressional regime of the flow contributed to the formation of hummocks. These indicate that the flow and emplacement of the avalanche were constrained by the topography. The existing caldera wall forced the initial eastward flow to move northward, and the north-side caldera wall forced the flow into the narrow and steepened outlet valley where the sliding debris underwent a compressional regime, and out into the unconfined terrain where the debris was most likely emplaced on an extensional regime. Also, the estimated volume of 12–15 × 108 m3 gives a mean thickness of 60–75 m, which is much deeper than the reported cases of other DADs. This suggests that the debris avalanche must have flowed further downstream and beyond the current DAD extent. Assessments of the DAD incorporating the topographic constraints can provide further insights into the risk and mitigation of potential disasters in the study area.

Abstract

A method that delivers a high yield and excellent quality of essential oil, which retains most of its value-added compounds, and undergoes least change after the extraction process, is greatly sought after. Although chemical free methods are acceptable, they call for an extensive processing time, while the yield and quality from these methods are often disappointing. This work utilizes subcritical water technology to address these issues. In this undertaking, essential oil was extracted from Aquilaria malaccensis wood by way of subcritical conditions, and characterized through gas chromatography/mass spectroscopy (GC/MS). Optimization through response surface methodology revealed temperature to be the most critical factor for the extraction process, while the optimum conditions for temperature, sample-to-solvent ratio, and time for subcritical water extraction was revealed as 225 °C, 0.2 gr/mL, and 17 min, respectively. The subcritical water extraction technique involves two simultaneous processes, which are based on good fitting to the two-site kinetic and second order model. In comparison to the hydrodistillation method, GC/MS results indicated that the quality of A. malaccensis’ wood oils, derived through the subcritical water technique, are of significantly better quality, while containing many constructive value-added compounds, such as furfural and guaiacol, which are useful for the production of pesticides and medicines. Pore size, functional groups, and morphology analysis revealed the occurrence of substantial damage to the samples, which facilitated an improved extraction of bio-products. In comparison to conventional methods, the use of the subcritical method not only involves a shorter processing time, but also delivers a higher oil yield and quality.

13. Anisotropic magnetic excitations from single-chirality antiferromagnetic state in Ca-kapellasite

Abstract

We present a ^{35}Cl NMR study for the spin S=1/2 perfect kagome antiferromagnet Ca-kapellasite [CaCu_{3}(OH)_{6}Cl_{2}·0.6H_{2}O] with a magnetic transition at T^{*}=7.2K. The static magnetic structure in the ground state has been determined to be a chirality-ordered Q=0 state, which is selected by a finite Dzyaloshinskii-Moriya interaction. The low-energy magnetic excitations in the ordered state are investigated by a nuclear spin-lattice relaxation rate measurement. We detect a weakly temperature-dependent contribution in the magnetic fluctuations perpendicular to the kagome plane in addition to the dispersive spin-wave contribution in the kagome plane. The low-energy magnetic excitations from the coplanar spin structure are attributed to the zero mode originating from the flat band in this kagome antiferromagnet.

Abstract

Background: In the PAOLA-1/ENGOT-ov25 primary analysis, maintenance olaparib plus bevacizumab demonstrated a significant progression-free survival (PFS) benefit in newly diagnosed advanced ovarian cancer patients in clinical response after first-line platinum-based chemotherapy plus bevacizumab, irrespective of surgical status. Prespecified, exploratory analyses by molecular biomarker status showed substantial benefit in patients with a BRCA1/BRCA2 mutation (BRCAm) or homologous recombination deficiency (HRD; BRCAm and/or genomic instability). We report the prespecified final overall survival (OS) analysis, including analyses by HRD status. Patients and methods: Patients were randomized 2: 1 to olaparib (300 mg twice daily; up to 24 months) plus bevacizumab (15 mg/kg every 3 weeks; 15 months total) or placebo plus bevacizumab. Analysis of OS, a key secondary endpoint in hierarchical testing, was planned for ∼60% maturity or 3 years after the primary analysis. Results: After median follow-up of 61.7 and 61.9 months in the olaparib and placebo arms, respectively, median OS was 56.5 versus 51.6 months in the intention-to-treat population [hazard ratio (HR) 0.92, 95% confidence interval (CI) 0.76-1.12; P = 0.4118]. Subsequent poly(ADP-ribose) polymerase inhibitor therapy was received by 105 (19.6%) olaparib patients versus 123 (45.7%) placebo patients. In the HRD-positive population, OS was longer with olaparib plus bevacizumab (HR 0.62, 95% CI 0.45-0.85; 5-year OS rate, 65.5% versus 48.4%); at 5 years, updated PFS also showed a higher proportion of olaparib plus bevacizumab patients without relapse (HR 0.41, 95% CI 0.32-0.54; 5-year PFS rate, 46.1% versus 19.2%). Myelodysplastic syndrome, acute myeloid leukemia, aplastic anemia, and new primary malignancy incidence remained low and balanced between arms. Conclusions: Olaparib plus bevacizumab provided clinically meaningful OS improvement for first-line patients with HRD-positive ovarian cancer. These prespecified exploratory analyses demonstrated improvement despite a high proportion of patients in the placebo arm receiving poly(ADP-ribose) polymerase inhibitors after progression, confirming the combination as one of the standards of care in this setting with the potential to enhance cure. in press

Abstract

Samples of the carbonaceous asteroid Ryugu were brought to Earth by the Hayabusa2 spacecraft. We analyzed 17 Ryugu samples measuring 1 to 8 millimeters. Carbon dioxide–bearing water inclusions are present within a pyrrhotite crystal, indicating that Ryugu’s parent asteroid formed in the outer Solar System. The samples contain low abundances of materials that formed at high temperatures, such as chondrules and calcium- and aluminum-rich inclusions. Abstract

Snakebite is a neglected disease and serious health problem in Brazil, with most bites being caused by snakes of the genus Bothrops. Although serum therapy is the primary treatment for systemic envenomation, it is generally ineffective in neutralizing the local effects of these venoms. In this work, we examined the ability of 7,8,3'-trihydroxy-4'-methoxyisoflavone (TM), an isoflavone from Dipteryx alata, to neutralize the neurotoxicity (in mouse phrenic nerve-diaphragm preparations) and myotoxicity (assessed by light microscopy) of Bothrops jararacussu snake venom in vitro. The toxicity of TM was assessed using the Salmonella microsome assay (Ames test). Incubation with TM alone (200 μg/mL) did not alter the muscle twitch tension whereas incubation with venom (40 μg/mL) caused irreversible paralysis. Preincubation of TM (200 μg/mL) with venom attenuated the venom-induced neuromuscular blockade by 84% ± 5% (mean ± SEM; n = 4). The neuromuscular blockade caused by bothropstoxin-I (BthTX-I), the major myotoxic PLA2 of this venom, was also attenuated by TM. Histological analysis of diaphragm muscle incubated with TM showed that most fibers were preserved (only 9.2% ± 1.7% were damaged; n = 4) compared to venom alone (50.3% ± 5.4% of fibers damaged; n = 3), and preincubation of TM with venom significantly attenuated the venom-induced damage (only 17% ± 3.4% of fibers damaged; n = 3; p < 0.05 compared to venom alone). TM showed no mutagenicity in the Ames test using Salmonella strains TA98 and TA97a with (+S9) and without (−S9) metabolic activation. These findings indicate that TM is a potentially useful compound for antagonizing the neuromuscular effects (neurotoxicity and myotoxicity) of B. jararacussu venom.

Abstract

As the decommissioning of Fukushima Daiichi nuclear power plant (NPP) progresses, the issue of how to deal with tritiated water has been attracting attention, both domestically and internationally. This article summarises the live discussion at the International Symposium on Tritiated Water, which was held by the Japan Health Physics Society (JHPS) in June 2020. Two issues – the scientific safety of tritiated water and social consensus building – were covered in the live discussion. The importance of further disclosure and dissemination of information based on steady monitoring was highlighted. It was also pointed out that scientific knowledge and scientific research data are merely the bottom line to achieve social consensus. Through the discussions, it was recognised that the role of JHPS is not only to look at the technical issues of safety, but also to look at social issues from the point of view of radiation protection, and to support the solution of these issues.

Abstract

In recent years, heavy rain which frequently occurred in various places in Japan have been caused severe damage. It is important to identify the damaged area for disaster recovery and reconstruction. In this study, we focus on the optical satellite images that are easy to process and interpret, and extract the damaged area by combining a land cover classification method using machine learning and an additive color mixture method. As the results, it is possible to visually express the land cover changes before and after the disasters in a specific category and to extract the damaged area from the optical satellite image.

Abstract

Study question Does the result of endometrial microbiome metagenomic analysis (EMMA), a next generation sequencing (NGS)-based test of the intrauterine microbiome, have an impact on pregnancy rate after the test? Summary answer After recommend treatment with antimicrobial and probiotic therapy, the group diagnosed as dysbiosis by EMMA achieved pregnancy significantly earlier than the group with Normal result. What is known already Using NGS technology, EMMA testing can determine the composition of the endometrial microbiome by analysing bacterial 16S ribosomal RNA with a focus on the lactobacillus population. Endometrial flora in patients undergoing in vitro fertilization (IVF) is often composed of pathogenic microorganisms which decrease implantation rates, such as the Enterobacteriaceae family, Staphylococcus spp., Escherichia coli and Gram-negative bacteria. Other studies also indicate that Lactobacillus spp. is a major microorganism in the endometrium. Especially, lactobacillus-dominated microbiota (LDM, defined as > 90% Lactobacillus spp.) in the endometrium has been reported to lead to better pregnancy outcomes than non-LDM ( Study design, size, duration This study was a prospective, multicenter cohort study of 527 patients (under 42 years old) with recurrent implantation failure (RIF) and recurrent pregnancy loss (RPL) at 14 IVF treatment facilities in Japan from June 2019 to August 2021. Defining RIF as three or more failed implantation attempts and RPL as two or more miscarriages, we examined the prognosis of two additional embryo transfers (ETs) after the EMMA test in patients who underwent the test. Participants/materials, setting, methods Endometrial tissue was obtained by aspiration from patients in day 15-25 of their menstrual cycles, and sample tissues were analyzed by NGS for EMMA. Participant centers treated patients according to the therapies specified in the reports, including antibiotic treatments, probiotic treatments, re-analysis, and embryo transfer. Multivariate analysis was performed using a generalized linear model with the endpoint of ongoing pregnancy. For the time-to-event analyses, we used Kaplan-Meier survival analysis to compare time to ongoing pregnancy. Main results and the role of chance The results of the first EMMA were as follows: 229 patients (43.4%) were normal with Lactobacillus spp. >90% (Normal group), 110 patients (20.9%) were abnormal with less than 90% Lactobacillus spp. and predominantly pathogenic bacteria (Abnormal group), and 188 patients (35.7%) were mild with low absolute amounts of bacteria and ultralow biomass indicating almost sterile (Mild + Ultralow group). There were no significant differences in background factors such as age, duration of infertility, number of previous ETs, or history of deliveries among the three groups. Gardnerella was the most pathogenic bacteria detected in patients with Abnormal EMMA results. All patients in the Abnormal group were treated with antimicrobials and probiotics, and those in Mild + Ultralow group were treated with probiotics. Odds ratio for ongoing pregnancy rate was 1.10(95%CI 0.67-1.82, p = 0.699) in Abnormal group and 1.23(95%CI 0.80-1.89, p = 0.342) in Mild + Ultralow group, respectively. After the intervention, ongoing pregnancies were comparable to those in Normal group.Analysis of time to pregnancy using Kaplan-Meier survival curves showed that Abnormal group had a significantly higher rate of ongoing pregnancies during the observation period than the other groups (p = 0.031). Limitations, reasons for caution Since this study was not necessarily limited to euploid embryos transferred after testing, an aging bias cannot be excluded. Since this study was conducted with all patients receiving EMMA, the effectiveness of the test needs to be further validated by comparison to patients without EMMA testing. Wider implications of the findings This study is the first multicenter study to demonstrate that the intervention based on EMMA reports improve pregnancy outcome in the patients with RIF and RPL. We suggest that the EMMA procedure, which aims at establishing an appropriate uterine microbiome, may be important for implantation and pregnancy continuation. 26. Investigation of the existence of self compacting properties in high performance concrete through experimental tests

Abstract

The self compacting concrete is characterized by its capacity to flow inside the formwork filling it exclusively by the force of the gravity with adequate cohesion and viscosity in such a way that segregation does not occur. One of its characteristic is the presence of fines which provide the necessary cohesion,and grains with maximum diameter of 20 mm. This work presents some procedures and experimental methods that make it possible to evaluate self compacting properties of high performance concrete. First, a bibliographical review on the subject was carried out, and later, the equipment used for the accomplishment of the assays were manufactured, in order to verify the properties related to the self compacting concrete: cohesion, viscosity and segregation. As for the work, two concretes were produced with Portland ARI Cement, thick sand, stone powder, sand 0, superplasticizer made of ether-carboxilate chains that differentiate from each other for the presence of active silica in one of them and fly ash in the other. Based on the results, it was verified whether the high performance concrete had self compacting characteristics. In this case, both were considered positive. It was also analyzed the behavior of these concretes in their hardened state by means of the compressive strength test. The Self Compacting Concrete has many advantages such as: reduction in the number of employees, shorter construction period, the non-use of the vibrator and the filling of formworks with high density of… or of complex geometry.

Abstract

IL-27 is an immunoregulatory cytokine whose essential function is to limit immune responses. We found that the gene encoding cholesterol 25-hydroxylase (Ch25h) was induced in CD4(+) T cells by IL-27, enhanced by TGF-β, and antagonized by T-bet. Ch25h catalyzes cholesterol to generate 25-hydroxycholesterol (25OHC), which was subsequently released to the cellular milieu, functioning as a modulator of T cell response. Extracellular 25OHC suppressed cholesterol biosynthesis in T cells, inhibited cell growth, and induced nutrient-deprivation cell death without releasing high-mobility group box-1. This growth inhibitory effect was specific to actively proliferating cells with high cholesterol demand and was reversed when extracellular cholesterol was replenished. Interestingly, Ch25h expressing CD4(+) T cells that received IL-27 and TGF-β signals became refractory to 25OHC-mediated growth inhibition in vitro. Nonetheless, IL-27 treated T cells negatively affected viability of bystander cells in a paracrine manner, but only if the bystander cells were in the early phases of activation. In mouse models of skin inflammation due to autoreactive T cells or chemically induced hypersensitivity, genetic deletion of Ch25h or Il27ra led to worsened outcomes. Thus, Ch25h is an immunoregulatory metabolic switch induced by IL-27 and dampens excess bystander T effector expansion in tissues through its metabolite derivative, 25OHC. This study reveals regulation of cholesterol metabolism as a modality for controlling tissue inflammation and thus represents a mechanism underlying T cell immunoregulatory functions.

Abstract

We demonstrated atomic diffusion bonding (ADB) of wafers using AlN films at room temperature. Results showed that crystal lattice rearrangement occurred at the bonded interface and integrated AlN film with c-axis perpendicular to the film plane formed.

Abstract

Objective : To explore the medical literature on techniques of tissue and sperm handling after surgical retrieval for intracytoplasmic sperm injection (ICSI). Methods : A search was performed in PubMed and Google Scholar databases, according to a modified Preferred Reporting Items for Systemic Reviews and Meta-Analyses (PRISMA) guideline, considering the studies investigating tissue handling and sperm selection techniques for ICSI. Results : Overall, 42 articles were included in this study, investigating sample handling, methods for sperm selection, and the use of chemical compounds to improve sperm motility and fertilisation rates. Conclusion : The ideal sperm handling method should provide a high sperm count, high vitality and appropriate sperm function, without side-effects. In this review the most common and useful techniques are described and the best combination strategies discussed in clinical scenarios.

Abstract

The Optimized Energy Degrading Optics (OEDO) beamline was constructed at RIBF to produce slowed-down, well-focused radioactive-isotope beams at 10–50 MeV/u, and began operation in June 2017. We examined the performance of the OEDO system by decelerating high-energy medium-mass radioactive ions. The main components of the OEDO beamline are two superconducting quadrupole triplet magnets and one radio-frequency deflector. This paper introduces the unique design of the ion optics of the beamline and describes the achieved performance of the beamline in the commissioning and following physics experiments. We will discuss the properties of energy-degraded beams produced by the OEDO beamline by comparison of simulations and the achieved performance.

Abstract

A fast ionization chamber with a high counting rate was commissioned at the 8 MV tandem accelerator complex of Kyushu University in Japan. Heavy ion beams consisting of 12C and 16O with Ebeam = 36 MeV were used to investigate the detector performance. By using various beam intensities ranging from 103 to 106 particles per second (pps), the maximum counting rate of the ion counter was measured to be about 8 × 105 pps. The detector performance under different gases was also investigated using two common gases in the ionization chamber: isobutane and P-10 gases. The results show that the negative effects due to pileups, which are often observed at high counting rates, can be greatly reduced when P-10 gas is used.

Abstract

This paper suggests the use of two optical elements for laser transfer in Thomson scattering diagnostics, which are widely used for other laser applications. YAG lasers amplified by flash lamps (repetition frequency of 50–100 Hz having a few Joules output) can sometimes have adverse effects such as peaked beam profiles and wavefront distortion. We used a beam homogenizer, which is a refractive optical element, to change the beam profile from peaked to non-peaked at a designated position. Using a super-Gaussian output profile when amplifying lasers facilitated longer—by a factor of 1.5 (=300 mm/200 mm) compared with top-hat beam profile—sustained distances, i.e. the distance along the laser path having a non-peaked profile. In terms of beam quality, wavefront distortion broke the beam profile in the far field. We used a deformable mirror to correct and flatten the wavefront of the beam. A YAG laser having a wavefront distortion of 0.46λ peak-to-valley was used to test the application of the deformable mirror, the results of which improved wavefront distortion to a wavelength of 0.1λ peak-to-valley.

37. Cloning of rat lysosomal acid lipase cDNA and identification of the mutation in the rat model of Wolman's disease.

Abstract

Lysosomal acid lipase (LAL) is a hydrolase essential for the intracellular degradation of cholesteryl esters and triglycerides. We previously reported a rat model of Wolman's disease (Wolman rat) that is deficient for LAL activity. In this study, we cloned rat LAL (RLAL) cDNA and investigated abnormal LAL gene expression in the Wolman rat. We cloned the RLAL gene from a cDNA library made from normal rat liver mRNA using the human LAL cDNA as a probe, subcloned the RLAL cDNA into pBlueScript vector, and sequenced it. Next, we constructed a cDNA library from a Wolman rat liver, and used the RLAL cDNA as a probe to isolate the Wolman RLAL cDNA for comparison. The normal RLAL cDNA contains 3150 bp including an 1194 bp open reading frame and three poly A signals at the 3' end. The deduced amino acid sequence contained 397 amino acids, showed 79.9% homology with human LAL, and had the same functional domains at the same sites as human LAL. Northern blot analysis revealed that the RLAL mRNA from normal rat was 3.2 kb in length, while the RLAL mRNA from Wolman rat was only 1.4 kb. Nucleotide sequence analysis showed that Wolman rat LAL cDNA had the same sequence as a RLAL cDNA from the 5'-untranslated region to nt 1101, followed by a 60 bp replacement from nt 1102 to nt 1161 with poly A signal and a 3' 1.8 kb deletion. The deduced amino acid sequence demonstrated the substitution of 367Ile to Asn, 368Pro to stop codon, and deletion of the C-terminal 29 amino acids. Genomic Southern blot analysis disclosed a large deletion at the 3' end of the gene. These results identify the molecular defect in the Wolman RLAL, and suggest that the C-terminus of RLAL is essential for the activity and/or stability of the enzyme.

Abstract

Background Blood pressure (BP) variability has reportedly been a risk factor for various clinical events. To clarify the influence of BP visit‐to‐visit variability on adverse events in patients with nonvalvular atrial fibrillation, a post hoc analysis of the J‐RHYTHM Registry was performed. Methods and Results Of 7406 outpatients with nonvalvular atrial fibrillation from 158 institutions, 7226 (age, 69.7±9.9 years; men, 70.7%), in whom BP was measured 4 times or more (14.6±5.0 times) during the 2‐year follow‐up period or until occurrence of an event, constituted the study group. SD and coefficient of variation of BP values were calculated as BP variability. Thromboembolism, major hemorrhage, and all‐cause death occurred in 110 (1.5%), 121 (1.7%), and 168 (2.3%) patients, respectively. When patients were divided into quartiles of systolic BP‐SD (P =0.015 for thromboembolism; HR, 2.60, 95% CI, 1.36–4.97, P =0.004 for major hemorrhage; and HR, 1.85, 95% CI, 1.11–3.07, P =0.018 for all‐cause death) after adjusting for components of the CHA 2 DS 2 ‐VASc score, warfarin and antiplatelet use, atrial fibrillation type, BP measurement times, and others. These findings were consistent when BP‐coefficient of variation was used instead of BP‐SD. Conclusions Systolic BP visit‐to‐visit variability was significantly associated with all adverse events in patients with nonvalvular atrial fibrillation. Further studies are needed to clarify the causality between BP variability and adverse outcomes in patients with nonvalvular atrial fibrillation. Abstract

The purpose of this research is to develop a semi-rigid column base of reinforced concrete structure to control stress distribution. In this paper, we report on the structural experiments carried out to grasp more detailed structural performance of semi-rigid columns with tapered part. Structural experiments were carried out changing conditions of axial force, taper angle and concrete strength. As a result of experiments, all the specimens showed that the strength decreased rapidly with the occurrence of burst fracture at the cross-section switching part. In addition, it was confirmed that the yield shear reinforcements and the tensile fracture occurred at the reduced part. In this paper, we clarified the detailed structural performance of semi-rigid columns with tapered part. Especially, it was confirmed that different characteristics of fracture properties and strain distribution occur depending on the taper angle under high axial force.

Abstract

Objective To present clinical and laboratory data of a Brazilian social program for cancer fertility preservation. Methods We carried out a descriptive observational study between July 2011 and December 2018. 246 patients were included from a social program in a private assisted reproduction clinic in Santo Andre/Brazil for oocyte cryopreservation before starting oncological treatment. Results 246 cancer patients resorted to fertility preservation before initiating cancer treatment. These were diagnosed with 27 different types of cancer, and the breast type is the most prevalent. 2528 MII oocytes (mean of 10.3 oocytes per patient) were vitrified. Four patients thawed their oocytes to submit in vitro fertilization, three had embryos transferred and one achieved pregnancy. Conclusion Preservation of fertility offers patients, especially at reproductive age, a viable way to perform their cancer treatment without compromising future gestation. It is important that professionals duly counsel oncological patients so, if they wish, they can have the possibility to guarantee her fertility preserved.

Abstract

Objective To assess the recovery of thawed blastocysts submitted to quarter laser assisted hatching and examine potential correlations between the procedure and pregnancy rates. Methods This cross-sectional study included only single-blastocyst transfers performed from July 2017 to December 2018. A total of 765 blastocysts were thawed and immediately submitted to quarter laser assisted hatching in the zona pellucida; they were subsequently incubated for three hours until transfer time, at which time they were examined for collapse or expansion; expanded blastocysts were further evaluated for herniation. The Chi-square test was used in statistical analysis. Results 627 blastocysts expanded (81.9%) and yielded a pregnancy rate of 40% (251/627). 138 blastocysts collapsed after thawing (18.0%) and yielded a pregnancy rate of 25.4% (35/138) (p=0.001). Additional analysis of the subgroup of expanded blastocysts revealed that the 385 herniated blastocysts (61.4%) yielded a pregnancy rate of 43.9% (169/385). The remaining 242 non-herniated blastocysts (38.6%) yielded a pregnancy rate of 33.9% (82/242) (p=0.013). Statistical significance was attributed to events with a p Conclusion Quarter laser assisted hatching is a safe, valid, and relatively easy-to-use procedure for thawed blastocysts. Blastocysts that expanded and herniated after quarter laser assisted hatching presented statistically superior results.

Abstract

We demonstrate an increase in the interparticle distance of a colloidal dipolar chain in a nematic liquid crystal (NLC). Applying an in-plane electric field perpendicular to the rubbing direction induces a splay-bend wall defect in the middle of the electrode gap, which traps a dipolar chain. Above the Freedericksz threshold electric field, the interparticle distance increases with increasing applied electric field, owing to the reorientation of the NLC molecules. The maximum increase is 32% of the particle diameter.

Abstract

Pneumococcal serotype replacement is an important issue after the introduction of pneumococcal conjugate vaccine (PCV) in children. After the introduction of 13-valent PCV, the incidence of invasive pneumococcal diseases (IPD) caused by Streptococcus pneumoniae serotype 12F (Sp12F) have increased in some countries; however, an outbreak of Sp12F has not reported in the post-13-valent PCV era. We experienced a local outbreak of Sp12F during March through May 2016 in Tsuruoka city, Japan after the introduction of 13-valent PCV in 2013. The IPD patients were two children and seven adults, three of whom died with a rapid disease progress. Although the clear transmission route was not determined, eight of the nine patients (89%) had close contact with children, which suggests that transmitted colonisation of Sp12F among children and adults might be the source of transmission. Continuous monitoring of IPDs, along with the determination of pneumococcal serotypes, is warranted in the post–13-valent PCV era. New IPD control strategies may be needed if this fatal outbreak continues to occur.

50. Identification, purification, and immunochemical characterization of a tocopherol-binding protein in rat liver cytosol.

Abstract

Tocopherol binding activity accompanying a rat liver cytosolic protein with molecular weight of 30-36 kDa has been demonstrated previously, although the isolation of the protein has not been reported. We now report the purification of an alpha-tocopherol-binding protein (TBP) from rat liver cytosol utilizing three chromatographic procedures: gel filtration, Affi-Gel Blue affinity chromatography, and chromatofocusing. Three peaks of specific alpha-tocopherol-binding activity were resolved on Affi-Gel Blue, referred to as AFB-1A, 1B, and 2. A 32-kDa homogeneous form was obtained after chromatofocusing of AFB-1B. D-alpha-[3H]tocopherol was displaced from homogeneous TBP in the presence of 500-fold excess of nonlabeled alpha-tocopherol, indicating the specificity of the binding. Anti-TBP rabbit antisera identified only one protein in rat hepatic cytosol on Western blotting. TBP immunoreactivity was found in the cytosol of rat liver and the lysate of fractionated hepatocytes, but not in the cytosol of other organs (including the heart, spleen, testes, and lung) nor in the lysate of fractioned Ito cells, endothelial cells, or Kupffer cells isolated from rat liver. Semi-quantitative ELISA demonstrated that rat liver cytosol contained approximately 2 mg TBP/g of cytosol protein. This immunoreactivity was associated with only the 30-36 kDa gel filtration fractions of rat liver cytosol and with both AFB-1A and -1B but not with AFB-2.
