HIV-1-spezifische T-Zellen spielen eine wichtige Rolle bei der Kontrolle der HIV-1-Infektion. Basierend auf Vorarbeiten der Arbeitsgruppe untersuchte ich verschiedene CTL-Epitope hinsichtlich ihrer Kreuzreaktivität zwischen HIV-1 und anderen Mikroorganismen. Bei einem dieser Epitope handelte es sich um das immundominante RI9-Epitop (RQDILDLWI) aus dem HIV-1-Nef-Protein, das mit einer guten Kontrolle von HIV-1 assoziiert ist und eine ähnliche Sequenz zeigte sich im PBP2-Protein von Chlamydophila pneumoniae (CP-YI9). Es konnte eine Kreuzreaktion zwischen Nef-RI9 und CP-YI9 gezeigt werden. Die Erkennung von Chlamydien-infizierten Zelllinien oder PBP2-transfizierten Zelllinien konnte jedoch nicht nachgewiesen werden, was darauf hindeutet, dass das optimale CP-YI9-Peptid nicht aus dem PBP2-Protein prozessiert wird. Ein weiteres Epitop ist das IMP-GL9-Epitop (GILGFVFTL) aus dem Matrix-Protein des Influenza-A-Virus, bei dem eine Kreuzreaktion zum HIV-1-SL9-Epitop (SLYNTVATL) schon gezeigt wurde. In einer Analyse einer Kohorte mit 174 HIV-1-infizierten Patienten konnte ich zeigen, dass IMP-GL9/SL9-kreuzreaktiven CTL häufig (43,1%) bei HIV-1-infizierten Patienten gefunden werden. Ich fand zwar klare Hinweise, dass die Erkennung von SL9 an sich, mit einem besseren Ansprechen auf die antiretrovirale Therapie assoziiert ist, jedoch fand ich keine Hinweise, dass IMP-GL9/SL9-kreuzreaktive CTL einen günstigen Effekt auf die Kontrolle von HIV-1 aufweisen. Es wurden nur selten IMP-GL9-spezifische CTL bei HIV-1-negativen Spendern gefunden und es wurden auch keine IMP-GL9/SL9-kreuzreaktive T-Zellen beobachtet. Dies weist darauf hin, dass die in den HIV-1-infizierten Patienten detektierten SL9/IMP-GL9-kreuzreaktiven CTL nicht durch Influenza, sondern durch HIV-1 selbst induziert wurden, was damit auch erklären könnte, dass diese kreuzreaktiven CTL keinen günstigen Effekt auf die HIV-1 Kontrolle hatten. Ein weiteres interessantes Epitop ist das EBV-GL9 (GLCTLVAML) von EBV, das kreuzreaktiv gegenüber dem IMP-GL9-Epitop ist. In meiner Arbeit konnte ich zeigen, dass auch SL9/EBV-GL9-spezifische kreuzreaktive T-Zellen unter 157 getesteten HIV-1-infizierten Patienten gefunden werden. Diese SL9/EBV-GL9-kreuzreaktiven T-Zellen werden deutlich seltener detektiert (16 %) als IMP-GL9/SL9-kreuzreaktive CTL. Ich konnte weiterhin einen TCR mit IMP-GL9-Erkennung und zwei TCR mit SL9-Erkennung klonieren und funktionell charakterisieren. Diese TCR zeigten eine eindeutige Peptid-Spezifität für das Peptid, mit dem die jeweilige ursprüngliche CTL-Linie generiert wurde, jedoch keine Kreuzreaktion. Weitere potenzielle kreuzreaktive Epitope sind in der Sequenz der RT von HIV-1 zu finden, die im katalytischen Zentrum eine Sequenzhomologie mit humanen endogenen Retroviren (HERV) aufweist. Ich analysierte daher in mehr als 150 Patienten die CTL-Antwort gegen HIV-1 und verschiedene HERVs und konnte eine Reihe von CTL-Linien generieren, die eine Spezifität gegen die RT von HIV-1 bzw. HERV aufweisen und einige zeigten auch Kreuzreaktivität zwischen HIV-1 und HERV. Ich konnte einen HIV-1-RT-YV9-spezifischen TCR klonieren und nach Transfektion TCR-kodierter mRNA in PBMC funktionell charakterisieren. Da dieser TCR keine Kreuzreaktion gegenüber den vorhandenen HERV-spezifischen Peptiden zeigte, wäre er sehr geeignet für die Immuntherapie der HIV-1-Infektion. Von unserer Arbeitsgruppe konnte auch ein kreuzreaktives Antikörper-Epitop aus dem HIV-1 Gag p24-Protein mit Sequenzübereinstimmung in verschiedenen HERV identifiziert werden. In einer groß angelegten Analyse von 336 Seren aus Patientengruppen mit verschiedenen Erkrankungen konnten bei einigen Patienten eine Reaktion gegenüber diesem Peptid im ELISA nachgewiesen werden. Dies deutet auf das Vorhandensein von kreuzreaktiven Antikörpern hin, sodass vermutet werden könnte, dass HERV oder andere Retroviren an der Pathogenese von Autoimmunkrankheiten beteiligt sein könnten. Auch HIV-1-exponierte seronegative Patienten wurden auf das Vorhandensein von HIV-1-spezifischen bzw. kreuzreaktiven CTL-Antworten untersucht. In zwei Patienten konnten HIV-1-spezifische CTL-Linien generiert werden. Somit konnten in der Promotionsarbeit CTL und Antikörper nachgewiesen werden, welche mit unterschiedlichen Epitopen aus Influenza, EBV und HERV kreuzreagieren. Die Klonierung und funktionelle Expression von TCR gegen konservierte CTL-Epitope in HIV-1 und Influenza könnte neue therapeutische Optionen durch eine genetische Immunisierung eröffnen. HIV-1-specific CTLs play an important role in controlling HIV-1-infections. I investigated different CTL epitopes concerning their cross-reactivity between HIV-1 and other pathogens based on preliminary studies of the work group. One of these epitopes was the immunodominant RI9 (RQDILDLWI) of HIV-1 Nef-protein, which is associated with a better control of HIV-1 and a similar sequence is in the PBP2-protein of Chlamydophila pneumoniae (CP-YI9). A cross-reaction between Nef-RI9 and CP-YI9 was shown in different patients. The recognition of cell-lines infected with C. pneumoniae or PBP2 transfected cell-lines was not established. This is probably due to the non-processing of optimal CP-YI9 epitope from the PBP2-protein. Another epitope is the IMP-GL9 epitope (GILGFVFTL) of the matrix protein of influenza A virus, in which a cross-reaction to HIV-1-SL9-epitope (SLYNTVATL) was previously shown. In my analysis of a cohort of 174 HIV-1-infected patients, it was shown, that such an IMP-GL9/SL9-cross-reactive CTL-line was frequently detected (43,1 %). I found clear evidence for a better response onto the antiretroviral therapy associated with the recognition of SL9, but there is no evidence of a positive effect of such IMP-GL9/SL9-cross-reactive CTL on the course of the HIV-1-infection. HIV-1-negative blood donors rarely generated IMP-GL9-specific CTL and IMP-GL9/SL9 cross-reactive T-cells weren´t observed. This indicated that the detected SL9/IMP-GL9-cross-reactive CTL in HIV-1-infected patients weren´t induced by Influenza, but rather by HIV-1 itself, which could also elucidate that this cross-reactive CTL had no favorable effect on the HIV-1 control. A further interesting epitope is the EBV-GL9 (GLCTLVAML) from EBV, which is cross-reactive to IMP-GL9. In my work, I showed that SL9/EBV-GL9-specific cross-reactive T-cells were found among 157 tested HIV-1-infected patients. These SL9/EBV-GL9 cross-reactive T-cells were more rarely found (16 %) as IMP-GL9/SL9 cross-reactive T-cells. Furthermore, I could generate and characterize one TCR with recognition of the IMP-GL9 epitope and two TCR with recognition of the SL9 epitope. The TCRs showed clear peptide specificity for the peptide with which the original T-cell line was stimulated, but without cross-reaction. More potential cross-reactive epitope are in the sequence of the RT of HIV-1 found. The catalytic center exhibit sequence homology with human endogenous retroviruses (HERV). I analyzed in more than 150 patients the CTL-answer against HIV-1 and different HERVS and I generated different CTL-lines with a specificity for HIV-1 and HERV, respectively. Some of these CTL-lines showed a cross-reactivity between HIV-1 and HERV. Additionally, an RT-YV9-specific TCR was generated, and after transfection of TCR-encoded mRNA in PBMC, functionally characterized. Since this TCR showed no cross-reaction to available HERV-specific peptides, this is a potential agent for immunotherapy of the HIV-1-infection. Our work group also identified a cross-reactive antibody epitope in HIV-1 Gag p24 protein. Moreover, there is compliance with different HERV sequences. In a large analysis of 336 sera from patient groups with different disease patterns, a reaction with this peptide was detected in some of the patients in the ELISA. This pointed to the existence of cross-reactive antibodies, so that it was assumed that HERV or other retroviruses could play a role in the pathogenesis of autoimmune diseases. Furthermore HIV-1-exposed seronegative persons were analyzed for the existence of HIV-1-specific and cross-reactive CTL-responses, respectively. In two patients, HIV-specific CTL-lines to different HIV-specific epitopes were generated. In this PhD-thesis CTL and antibody which cross-react with different epitopes from influenza, EBV and HERV were established. The generation and functional expression of TCR against conserved CTL-epitopes in HIV-1 and influenza could open new therapeutic options though genetic immunization.

NONEXPRESSOR OF PATHOGENESIS-RELATED PROTEINS1 (NPR1; also known as NIM1) is a master regulator of systemic acquired resistance (SAR). SAR is induced by salicylic acid (SA), leading to the expression of PATHOGENESIS-RELATED (PR) genes. Current evidence suggests that NPR1 is part of a transcription complex tethered to activation sequence-1 (as-1)-like cis-acting elements in PR-1 gene promoters through TGA transcription factors, and that SA-dependent PR-1 gene expression is regulated by NIM1-INTERACTING (NIMIN) proteins. In Arabidopsis, NPR1 is active only after SA induction. Regulation of Arabidopsis NPR1 activity has been proposed to comprise cysteine-156 (Cys-156), mediating SA-induced cytoplasmic oligomer-nuclear monomer exchange, and Cys-521 and Cys-529, mediating SA-dependent transcriptional activation. Tobacco NPR1 does not harbour these residues. To understand the function of tobacco NPR1, we analysed its biochemical capabilities in a heterologous system: yeast. Tobacco NPR1 differs from Arabidopsis NPR1 in its subcellular localization and its transactivation potential. Yet, both tobacco and Arabidopsis NPR1, as well as tobacco NIM1-like1, alter some of their biochemical activities in response to SA. Whereas the addition of SA to yeast growth medium induces transcriptional activity in tobacco NPR1, its interaction with NIMIN2-type proteins is suppressed. The effects of SA are specific, sensitive and occur coordinately. They are abolished completely by mutation of the arginine residue within the invariable penta-amino acid motif LENRV, as present in the nonfunctional Arabidopsis nim1-4 allele. Furthermore, NPR1 proteins with the LENRV domain coincidently harbour a broad and strongly conserved NIMIN1/NIMIN2 binding site. Our data suggest that NPR1 and some NPR1-like proteins are sensitive to the plant hormone SA, altering some of their biochemical capabilities to enable stimulus-dependent gene expression. The sensitivity of NPR1 proteins to SA, together with their differential interaction with diverse NIMIN proteins, seems a plausible molecular basis for the timely and coordinated activation of PR genes during SAR.

NONEXPRESSOR OF PATHOGENESIS-RELATED PROTEINS1 (NPR1; also known as NIM1) is a master regulator of systemic acquired resistance (SAR). SAR is induced by salicylic acid (SA), leading to the expression of PATHOGENESIS-RELATED (PR) genes. Current evidence suggests that NPR1 is part of a transcription complex tethered to activation sequence-1 ( as-1)-like cis-acting elements in PR-1 gene promoters through TGA transcription factors, and that SA-dependent PR-1 gene expression is regulated by NIM1-INTERACTING (NIMIN) proteins. In Arabidopsis, NPR1 is active only after SA induction. Regulation of Arabidopsis NPR1 activity has been proposed to comprise cysteine-156 (Cys-156), mediating SA-induced cytoplasmic oligomer-nuclear monomer exchange, and Cys-521 and Cys-529, mediating SA-dependent transcriptional activation. Tobacco NPR1 does not harbour these residues. To understand the function of tobacco NPR1, we analysed its biochemical capabilities in a heterologous system: yeast. Tobacco NPR1 differs from Arabidopsis NPR1 in its subcellular localization and its transactivation potential. Yet, both tobacco and Arabidopsis NPR1, as well as tobacco NIM1-like1, alter some of their biochemical activities in response to SA. Whereas the addition of SA to yeast growth medium induces transcriptional activity in tobacco NPR1, its interaction with NIMIN2-type proteins is suppressed. The effects of SA are specific, sensitive and occur coordinately. They are abolished completely by mutation of the arginine residue within the invariable penta-amino acid motif LENRV, as present in the nonfunctional Arabidopsis nim1-4 allele. Furthermore, NPR1 proteins with the LENRV domain coincidently harbour a broad and strongly conserved NIMIN1/NIMIN2 binding site. Our data suggest that NPR1 and some NPR1-like proteins are sensitive to the plant hormone SA, altering some of their biochemical capabilities to enable stimulus-dependent gene expression. The sensitivity of NPR1 proteins to SA, together with their differential interaction with diverse NIMIN proteins, seems a plausible molecular basis for the timely and coordinated activation of PR genes during SAR. Introduction:Post-transplant morbidity and mortality are majorly determined by GvL effect counter-balanced by GvHD. Treatment with systemic steroids represents the first-line therapy for GvHD, but is associated with increased incidence of infection and relapse. ECP can reduce the extent of GvHD while preserving anti-virus/-tumor activity. However, ECP, a time-consuming and relatively expensive therapy, requires specialized equipment and staff expertise as well as the mechanism of action remains unclear. Therefore, this study was performed to address these unsolved questions.

Introduction:Genetically modified T cells expressing chimeric antigen receptors (CARTs) are currently among the most encouraging treatment approaches in cancer immunotherapy. Efficacy and persistence of adoptively transferred CARTs are linked to the proportion of naïve-like T cells (TN) and stem cell memory-like (TSCM) T cells defined by stem cell-like properties such as a high proliferative and self-renewal capacity. Optimization of CART production is important to shift the balance towards these subsets. The PI3K/mTOR/AKT pathway is one of the main pathways involved in T cell differentiation. In the current study we evaluated the inhibition of PI3K by the clinically approved small molecule idelalisib during culturing of CARTs in order to increase less differentiated T cell subpopulations in the final cell product.

- Abstract
Human JC and BK polyomaviruses (JCV/BKV) can establish a latent infection without any clinical symptoms in healthy individuals. In immunocompromised hosts infection or reactivation of JCV and BKV can cause lethal progressive multifocal leukoencephalopathy (PML) and hemorrhagic cystitis, respectively. Vaccination with JCV/BKV derived antigen epitope peptides or adoptive transfer of virus-specific T cells would constitute an elegant approach to clear virus-infected cells. Furthermore, donor leukocyte infusion (DLI) is another therapeutic approach which could be helpful for patients with JCV/BKV infections.So far, only few immunodominant T cell epitopes of JCV and BKV have been described and therefore is a fervent need for the definition of novel epitopes. In this study, we identified novel T cell epitopes by screening libraries of overlapping peptides derived from the major capsid protein VP1 of JCV. Virus like particles (VLPs) were used to confirm naturally processing. Two human leucocyte antigen (HLA)-A*02-restricted epitopes were characterized by fine mapping with overlapping peptides and nonamer peptide sequences were identified. Cytokine release profile of the epitope-specific T cells was analyzed by enzyme-linked immunospot (ELISPOT) assays and by flow cytometry. We demonstrated that T cell responses were of polyfunctional nature with the potential of epitope-specific killing and cross-reactivity between JCV and BKV. These novel epitopes might constitute a new potential tool to design effective diagnostic and therapeutic approaches against both polyomaviruses.

- Abstract
Background: It has been reported that HIV-1-specific cytotoxic T cells (CTL) recognizing the HLA-A2-restricted p17 epitope SLYNTVATL (SL9) can cross-react with the HLA-A2-restricted influenza matrix epitope GILGFVFTL (GL9). So far, the prevalence of GL9-cross-reacting HIV-1-specific CTL in larger cohorts of HIV-1-infected patients is unknown, and there are no data yet on whether SL9/GL9-cross-reactive CTL may influence the course of HIV-1 infection., Methods: We analyzed the presence of SL9/GL9-cross-reacting CTL in a cohort of 175 HLA-A2-positive HIV-1-infected patients. Peripheral blood mononuclear cells were stimulated in vitro with SL9 and GL9 peptides, and outgrowing cell lines regarding cross-reactivity and recognition of viral variants in γ-interferon enzyme-linked immunospot assays were analyzed., Results: SL9- and GL9-specific CTL could be generated in 52.6% and 53.7% of 175 patients, respectively. Both SL9- and GL9-specific CTL were more frequently observed in patients on antiretroviral therapy (ART). Of the 92 SL9-specific CTL and the 94 GL9-specific CTL, 65.2% and 66%, respectively, showed at least partial SL9/GL9 cross-reactivity. SL9/GL9-cross-reactive CTL could be detected in 42.9% of the 175 patients. Recognition of SL9 was associated with lower viral loads and higher CD4 cell counts in patients on ART. Patients with GL9/SL9 cross-reactivity displayed similar CD4 cell counts than patients without GL9/SL9-cross-reactive cells. GL9/SL9-cross-reactive cells were associated with higher viral loads in patients on ART., Conclusions: Partially SL9/GL9-cross-reactive CTL are frequently observed in HIV-1-infected patients. So far, we could not detect a significant influence of the presence of SL9/GL9-cross-reacting CTL on the course of HIV-1 infection.
