At work, at school, at the gym club, or even at home, consumers often face challenging situations in which they are motivated to perform their best. This research demonstrates that activating performance goals, whether in cognitive or physical domains, leads to an increase in the consumption of high-calorie foods at the expense of good nutrition. This effect derives from beliefs that the function of food is to provide energy for the body (food as fuel) coupled with poor nutrition literacy, leading consumers to overgeneralize the instrumental role of calories for performance. Indeed, nutrition experts choose very different foods (lower in calorie, higher in nutritional value) than lay consumers in response to performance goals. Also, performance goals no longer increase calorie intake when emphasizing the hedonic function of food (food for pleasure). Hence, while consumer research often interprets the overconsumption of pleasurable and unhealthy high-calorie foods as a consequence of hedonic goals and self-control failures, our research suggests that this overconsumption may also be explained by a maladaptive motivation to manage energy intake. Oxidative stress due to obesity plays a detrimental role in the testicular microenvironment and sperm parameters. We explored the impact of a hypercaloric diet in male BALB/c mice as a condition to trigger damage to the spermatogenic process and the antioxidant effect of Aspalathus linearis as well. We used a hypercaloric diet in animals divided into 3 groups: Control, Hypercaloric diet control (HC) and Hypercaloric diet and Rooibos infusion (HCR). Morphometric parameters, enzyme dosages, cell viability, and tubular histopathology were evaluated. Body weight increased in HCR animals at weeks 3, 4, and 8. We found a reduction in seminiferous epithelium height, with an increase in the tubular diameter of the HCR group. Catalase levels were lower in HC and HCR, while carbonyl protein was decreased in HC. We estimate that it induces oxidative stress (OS) capable of affecting the seminiferous epithelium and that the infusion of A. linearis does not demonstrate a potential benefit in cell preservation. Dens invaginatus in molar teeth is extremely rare. This report presents a case in a right mandibular second molar. The pulp was necrotic and there was a buccal sinus tract associated with the tooth. The dens invaginatus was revealed following a periapical radiograph that also showed a periradicular lesion. Conventional root canal treatment was carried out; a calcium hydroxide paste was used as an intracanal dressing. The root canal space was filled with gutta-percha and sealer using a thermomechanical compaction technique. At follow-up, after 1 year, the tooth was symptom-free and there was radiographic evidence of healing. As a result, endodontic surgery was not required. [ABSTRACT FROM AUTHOR]

The aim was to report the diagnosis and successful clinical management of a patient with pathological anatomic variation in a mandibular first molar, caused by internal and external resorption and to evaluate radiographically the long term clinical outcome of the root canal treatment. A case is presented in which a previous caries removal procedure with pulp exposure on a mandibular molar developed into internal resorption of the pulp chamber floor and the distal canal. A preoperative periapical radiograph showed apical external resorption of the molar roots and condensing osteitis associated with the mesial root. Root canal treatment was completed in two visits, with calcium hydroxide used as an interappointment dressing for 1 month. The purpose of this study was to test the effect of acute dairy calcium intake on exercise energy metabolism and endurance performance. Trained female runners completed two trials. Each trial consisted of a 90-min glycogen depletion run followed by a self-paced 10K time trial, conducted one hour after consumption of a high dairy (500 mg Ca+2) or low dairy (80 mg Ca+2) meal. During the 90-min run, blood samples and respiratory gases were collected. No treatment main effects of acute dairy intake were found for respiratory exchange ratio (RER), calculated fat oxidation, lactate, glycerol, or 10K time. Intrahousehold resource allocations have important implications for policies targeted at household resources and thus household welfare. Recent literature has shown, for single headed households, that the Supplemental Nutrition Assistance Program benefits may be inadequate to reach a nutritious diet due to a "time deficit" gap in food production. This paper first develops a simple theoretically based time adjustment multiplier to address this benefit inadequacy for single and dual headed households. A method is then developed for estimating spousal time in food production with limited data. The estimated time deficit gap and time adjustment multiplier are smaller for dual headed households than single headed households. For single headed households, the time adjusted benefits are about $107 per household per week higher than the unadjusted benefits. For dual headed households, ignoring the spousal time contribution, the time adjusted benefits are about $73 per household per week higher than the unadjusted benefits. However, by including the spousal time contribution, this benefit shortfall is reduced by about $50. These findings demonstrate the importance of taking into account intrahousehold time allocation in evaluating the adequacy of the Supplemental Nutrition Assistance Program benefits. Spontaneous activity of vascular smooth muscle is present in small arteries and some venous tissues like the hepatic portal vein. Whereas the ability to generate rhythmic membrane potential changes is expressed in a high number of primary oscillators, the generation of physiological tone and phasic activity requires synchronization of specialized pacemaker activity (Interstitial Cajal-like cells) by intercellular propagation and regeneration of excitation or a strong coupling mechanism of smooth muscle cells. The aim of this study was to deduce oscillator coupling by analyzing the spatiotemporal homogeneity of calcium oscillations within a native tissue preparation. Portal vein tissue was loaded with a calcium-sensitive dye (Fluo-3). By combining confocal microscopy and computation of spatial auto- and cross-correlation of the calcium signals, temporal and spatial coupling between cells was characterized. Spontaneous oscillations of calcium signals were measured at different predefined regions of interest. Cross-correlation analysis of these signals revealed that their damping was very similar in all directions of the investigated z-plane. In single experiments, improved cell-to-cell coupling was seen when noradrenaline (1–10 μM) was added to the bath-solution. With the chosen parameters of frame refresh, the velocity of signal propagation was faster than the maximum detectable velocity, but it could be estimated to exceed 0.1 mm/s. Correlative Network Analysis is a new and very useful tool to determine the functional coupling parameters of quasi-homogenous biological networks and their temporal changes. Aim: This study described outcomes following treatment for lactose intolerance, which is common in children.Methods: The medical records of children aged 6-18 years who underwent lactose hydrogen breath testing at Dana-Dwek Children's Hospital, Tel Aviv, Israel, from August 2012 to August 2014 were analysed. We compared 154 children with gastrointestinal symptoms and positive lactose hydrogen breath tests to 49 children with negative test results.Results: Of the 154 children in the study group, 89 (57.8%) were advised to follow a lactose-restricted diet, 32 (20.8%) were advised to avoid lactose completely, 18 (11.7%) were instructed to use substitute enzymes, and 15 (9.7%) did not receive specific recommendations. Only 11 patients (7.1%) received recommendations to add calcium-rich foods or calcium supplements to their diet. Lactose reintroduction was attempted in 119 of 154 patients (77.3%), and 65 of 154 (42.2%) experienced clinical relapses. At the final follow-up of 3.3 years, 62.3% of the study children were still observing a restricted diet. Older children and those who were symptomatic during lactose hydrogen breath testing were more likely to be on a prolonged restricted diet.Conclusion: Our long-term follow-up of lactose-intolerant children showed that only a third were able to achieve a regular diet. Abstract: Within the last decade, calcium decline has emerged as a stressor for many soft water lakes. A legacy of long‐term acid deposition and logging, calcium decline has been implicated in the loss of large herbivores, such as Daphnia, and changes in community structure as taxa with presumed low calcium demand also increase. Although lake surveys, paleolimnological studies, and laboratory experiments have provided considerable evidence that declining Daphnia abundances are associated with low calcium concentration, causal relationships for zooplankton response to calcium decline have not been assessed using field experiments with natural zooplankton communities. We conducted a six‐week field experiment under low food conditions typical of Canadian Shield lakes, using four calcium concentrations (0.6 mg/L, 1.0 mg/L, 1.4 mg/L, and 2.4 mg/L) and zooplankton communities originating from eight lakes along a gradient from 1.78 mg Ca/L to 24.8 mg Ca/L, to examine the effect of calcium on growth rates and reproduction of individual and groups of taxa. Population growth rates for daphniids, Bosmina spp., and three cyclopoids declined as calcium concentration decreased. At low calcium, growth rates of Daphnia pulex, Daphnia catawba, and Mesocyclops edax were negative indicating declining populations. There was no effect of calcium on reproduction of the most common cladocerans. Although we detected some variation in growth rates amongst source lakes, zooplankton communities did not differ in their response to low calcium. As calcium concentrations continue to decline in soft water lakes, reduced zooplankton growth rates may result in shifts in zooplankton community structure and overall declines in total zooplankton production. Alterations in intracellular-calcium (Ca 2+ ) i homeostasis is critical to Aeromonas hydrophila -induced headkidney macrophages (HKM) apoptosis of Clarias gariepinus , though the implications are poorly understood. Here, we describe the role of intermediate molecules of Ca 2+− signaling pathway that are involved in HKM apoptosis. We observed phosphoinositide-3-kinase/phospholipase C is critical for (Ca 2+ ) i release in infected HKM. Heightened protein kinase-C (PKC) activity and phosphorylation of MEK1/2-ERK1/2 was noted which declined in presence of 2-APB, Go6976 and PD98059, inhibitors to IP3-receptor, conventional PKC isoforms (cPKC) and MEK1/2 respectively implicating Ca 2+ /cPKC/MEK-ERK1/2 axis imperative in A. hydrophila- induced HKM apoptosis. Significant tumor necrosis factor-α (TNFα) production and its subsequent reduction in presence of MEK-ERK1/2 inhibitor U0126 suggested TNFα production downstream to cPKC-mediated signaling via MEK1/2-ERK1/2 pathway. RNAi and inhibitor studies established the role of TNFα in inducing caspase-8-mediated apoptosis of infected HKM. Inactivation of L-type calcium channel (Cav1.2) is an important determinant of the length of the cardiac action potential. Here, we report a key role of the voltage-sensing segment IS4 in Cav1.2 inactivation. Neutralization of IS4 charges gradually shifted the steady-state inactivation curve on the voltages axis from 5.1 ± 3.7 mV in single point mutant IS4(K1Q) to −26.7 ± 1.3 mV in quadruple mutant IS4(K1Q/R2Q/R3Q/R4Q) compared to wild-type (WT) and accelerated inactivation. The slope factor of the Boltzmann curve of inactivation was decreased from 17.4 ± 3.5 mV (IS4(K1Q)) to 6.2 ± 0.7 mV (IS4(K1Q/R2Q/R3Q/R4Q)). Neutralizations of single or multiple charges in IIS4 and IIIS4 did not significantly affect the time course of inactivation. Neutralization of individual IVS4 charges shifted the inactivation curve between 17.4 ± 1.7 mV (IVS4(R2Q)) and −4.6 ± 1.4 mV (IVS4(R4Q)) on the voltage axis and affected the slope of the inactivation curves (IVS4(R2Q): 10.2 ± 1.2 mV, IVS4(R4Q): 9.7 ± 0.7 mV and IVS4(K5Q): 8.1 ± 0.7 mV vs WT: 14.1 ± 0.8 mV). IS4(K1Q) attenuated while IS4(K1Q/R2Q/R3Q) and IS4(K1Q/R2Q/R4Q/R3Q) enhanced the development of inactivation. Shifts in the voltage dependence of inactivation curves induced by IS4 neutralizations significantly correlated with shifts of the voltage dependence of channel activation ( r = 0.95, p < 0.01) indicating that IS4 movement is not only rate limiting for activation but also initiates inactivation. The paradoxical decrease of the slope factor of the steady-state inactivation and acceleration of inactivation kinetics upon charge neutralization in segment IS4 may reflect the loss of stabilizing interactions of arginines and lysine with surrounding residues. High inclusion concentrations of dietary calcium (Ca) from limestone may have detrimental effects on broiler performance, such as increased gastric pH and a reduction in phosphorus (P) and amino acid digestibility. To assess the potential for feeding diets with lower total Ca concentrations, the effect of a novel highly soluble source of calcium (HSC) on bird performance, nutrient digestibility and skeletal integrity of 1120 male Cobb-500 broilers was compared with that of limestone. Eight dietary treatments in a 2 × 2 × 2 factorial design consisted of two sources of Ca (limestone or HSC), two concentrations of Ca (6.0 or 7.7 g/kg in the starter and 4.0 or 5.7 g/kg in the grower), two phytase levels (0 or 500 FTU/kg). The effect of dietary treatments on growth performance and feed intake were measured from Day 1 to Day 40 and foot ash and leg health on Day 40. At the end of the starter period, in the absence of phytase, changing Ca source from limestone to HSC at 6.0 g/kg total Ca decreased both bodyweight gain and feed intake (P < 0.05). The addition of phytase to diets containing 6.0 g/kg total Ca provided by HSC increased bodyweight gain (P < 0.05), as well as increasing feed intake (P < 0.05). At the end of the grower period, diets supplemented with phytase increased bodyweight gain (P < 0.001), birds fed the higher Ca concentration were also observed to have an increased bodyweight gain (P < 0.001). Apparent ileal Ca digestibility significantly increased with the addition of phytase to diets containing 4.0 g/kg total Ca provided by HSC (P < 0.05), this was also observed for P digestibility. In a 12-week duration parallel group randomised clinical trial, we evaluated the effect of increasing calcium (Ca) intake on body composition and insulin resistance in patients with type 2 diabetes (T2DM). Thirty-six subjects with low habitual Ca intake (<600 mg d −1 ), consumed low-Ca diet (CD group, 800 mg d −1 ) or high-Ca fat-free milk diet (MD group, 1500 mg d −1 ). MD group final anthropometric measures (body weight, BMI, waist circumference, waist-hip ratio, and fat mass) decreased compared with baseline. MD group showed greater decrease in waist circumference compared with CD group. Final fasting glucose decreased in CD group compared with baseline. Both groups reduced glycated haemoglobin. Consumption of high-Ca diet from dairy for 12 weeks was effective in reducing abdominal adiposity, but provided no additional effect on glycaemic control in overweight patients with T2DM. The importance of adequate calcium intakes for healthy growth and bone development has long been recognised. Recent evidence suggests that calcium supplementation may have sex-specific effects on bone growth in childhood. The aim was to describe the long-term effects of calcium supplementation in pregnant Gambian women with a low calcium intake (ISCRTN96502494) on offspring height, weight, bone and body composition in childhood, and whether the effects differ by sex. Children of mothers who participated in the original calcium supplementation trial were measured at age 8–12 years using dual-energy X-ray absorptiometry and peripheral quantitative computed tomography. Linear models tested for sex*supplement interactions before and after adjusting for current age and size in early life. 447 children, aged 9.2(SD 0.9) years, were measured. Significant sex*supplement interactions ( p < 0.05) were observed for many of the anthropometric and bone outcomes, Females whose mothers received calcium (F-Ca) were shorter, lighter with smaller bones and less bone mineral than those whose mothers received placebo (F-P), differences (SE) ranged from height = − 1.0 (0.5)% to hip BMC − 5.5 (2.3)%. Males from mothers in the calcium group (M-Ca) had greater mid-upper arm circumference (MUAC) (+ 2.0 (1.0)%, p = 0.05) and fat mass (+ 11.6 (5.1)%, p = 0.02) and tended towards greater BMC and size than those whose mothers were in the placebo group (M-P). The differences in anthropometry and body composition were robust to adjustment for current height and weight, whereas all bone differences became non-significant. F-P were taller with more BMC than M-P, whereas F-Ca had similar sized bones and mineral content to M-Ca. Calcium supplementation of pregnant women with low calcium intakes altered the childhood trajectories of growth and bone and body composition development of their offspring in a sex-specific manner, resulting in slower growth among females compared to placebo and accelerated growth among males by age 8–12 years. The suprachiasmatic nucleus (SCN) times the daily rhythms of behavioral processes including feeding. Beyond the SCN, the hypothalamic arcuate nucleus (ARC), involved in feeding regulation and metabolism, and the epithalamic lateral habenula (LHb), implicated in reward processing, show circadian rhythmic activity. These brain oscillators are functionally coupled to coordinate the daily rhythm of food intake. In rats, a free choice high-fat high-sugar (fcHFHS) diet leads to a rapid increase of calorie intake and body weight gain. Interestingly, under a fcHFHS condition, rats ingest a similar amount of sugar during day time (rest phase) as during night time (active phase), but keep the rhythmic intake of regular chow-food. The out of phase between feeding patterns of regular (chow) and highly rewarding food (sugar) may involve alterations of brain circadian oscillators regulating feeding. Here, we report that the fcHFHS diet is a successful model to induce calorie intake, body weight gain and fat tissue accumulation in mice, extending its effectiveness as previously reported in rats. Moreover, we observed that whereas in the SCN the day-night difference in the PER2 clock protein expression was similar between chow-fed and fcHFHS-fed animals, in the LHb, this day-night difference was altered in fcHFHS-exposed animals compared to control chow mice. These findings confirm previous observations in rats showing disrupted daily patterns of feeding behavior under a fcHFHS diet exposure, and extend our insights on the effects of the diet on circadian gene expression in brain clocks. Layer 5 (L5) is a major neocortical output layer containing L5A slender-tufted (L5A-st) and L5B thick-tufted (L5B-tt) pyramidal neurons. These neuron types differ in their in vivo firing patterns, connectivity and dendritic morphology amongst other features, reflecting their specific functional role within the neocortical circuits. Here, we asked whether the active properties of the basal dendrites that receive the great majority of synaptic inputs within L5 differ between these two pyramidal neuron classes. To quantify their active properties, we measured the efficacy with which action potential (AP) firing patterns backpropagate along the basal dendrites by measuring the accompanying calcium transients using two-photon laser scanning microscopy in rat somatosensory cortex slices. For these measurements we used both "artificial" three-AP patterns and more complex physiological AP patterns that were previously recorded in anesthetized rats in L5A-st and L5B-tt neurons in response to whisker stimulation. We show that AP patterns with relatively few APs (3APs) evoke a calcium response in L5B-tt, but not L5A-st, that is dependent on the temporal pattern of the three APs. With more complex in vivo recorded AP patterns, the average calcium response was similar in the proximal dendrites but with a decay along dendrites (measured up to 100 µm) of L5B-tt but not L5A-st neurons. Interestingly however, the whisker evoked AP patterns--although very different for the two cell types--evoke similar calcium responses. In conclusion, although the effectiveness with which different AP patterns evoke calcium transients vary between L5A-st and L5B-tt cell, the calcium influx appears to be tuned such that whisker-evoked calcium transients are within the same dynamic range for both cell types. Objective: An experiment was conducted to investigate the effect of dietary Ca concentrations in low non-phytate phosphorus (NPP) diets containing phytase on growth performance, bone mineralization, litter quality, and footpad dermatitis (FPD) incidence in growing broiler chickens. Methods: A total of 1,800 21-day-old Ross 308 growing broiler chickens were allotted to 1 of 6 dietary treatments with 6 replicated cages. Six diets were formulated to provide increasing Ca concentrations of 4.0, 5.0, 6.0, 7.0, 8.0, or 9.0 g/kg in diets. The concentrations of NPP in all diets were maintained at 3.0 g/kg, and phytase was supplemented to all diets at the level of 1,000 fytase units (FTU)/kg. At the end of the 14-d feeding trial, birds were euthanized for tibia sampling, and litter samples were collected from 3 areas in the cage. The FPD incidence was measured based on a 6-point scoring system. Results: Dietary Ca concentrations had no effect on growth performance of growing broiler chickens. However, a tendency (linear, p = 0.05) for decreased feed efficiency was observed as dietary Ca concentrations were increased. The concentrations of Ca and P in the tibia of broiler chickens increased (linear and quadratic, p<0.01) with increasing Ca concentrations in low NPP diets containing phytase. Litter pH, moisture, and N contents were not affected by increasing Ca concentrations in low NPP diets containing phytase. However, a tendency (quadratic, p = 0.10) for increased FPD incidence with increasing dietary Ca concentrations was observed. Conclusion: Dietary Ca concentrations from 4.0 to 9.0 g/kg in low NPP diets containing phytase have little effects on growth performance of growing broiler chickens. However, Ca and P concentrations in the tibia are decreased if dietary Ca concentrations are less than 5.0 g/kg. Maintaining a healthy diet has important implications for physical and mental health. One factor that may influence diet and food consumption is inhibitory control-the ability to withhold a dominant response in order to correctly respond to environmental demands. We examined how N2 amplitude, an ERP that reflects inhibitory control processes, differed toward high- and low-calorie food stimuli and related to food intake. A total of 159 participants (81 female; M age = 23.5 years; SD = 7.6) completed two food-based go/no-go tasks (one with high-calorie and one with low-calorie food pictures as no-go stimuli) while N2 amplitude was recorded. Participants recorded food intake using the Automated Self-Administered 24-hour Dietary Recall system. Inhibiting responses toward high-calorie stimuli elicited a larger (i.e., more negative) no-go N2 amplitude; inhibiting responses toward low-calorie stimuli elicited a smaller no-go N2 amplitude. Participants were more accurate during the high-calorie than low-calorie task, but took longer to respond on go trials toward high-calorie rather than low-calorie stimuli. When controlling for age, gender, and BMI, larger high-calorie N2 difference amplitude predicted lower caloric intake (β = 0.17); low-calorie N2 difference amplitude was not related to caloric intake (β = −0.03). Exploratory analyses revealed larger high-calorie N2 difference amplitude predicted carbohydrate intake (β = 0.22), but not protein (β = 0.08) or fat (β = 0.11) intake. Results suggest that withholding responses from high-calorie foods requires increased recruitment of inhibitory control processes, which may be necessary to regulate food consumption, particularly for foods high in calories and carbohydrates. The influence of temperature on electrode potential was studied & it can be used in the determination of thermodynamic function like ΔG, ΔH & ΔS. The electrochemical impedance spectroscopy (EIS) technique is also employed to study the electrochemical & surface reactions. Three kinds of novel diatomite/ground calcium carbonate composite humidity control materials were prepared with different calcination temperatures using diatomite and ground calcium carbonate (GCC) as raw materials. The microstructure and morphology properties of samples were studied by nitrogen gas adsorption, mercury intrusion porosimetry (MIP) and fractal dimensions on the basis of gas adsorption isotherms with FHH methods. Fourier transform infrared spectroscope (FTIR), scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were used to characterize the mineral composition and surface properties. Furthermore, X-ray diffraction (XRD), thermo-gravimetry and differential scanning calorimetry (TG-DSC) were used to analyze the formation mechanism of composite material. It was indicated that DG750 showed the better moisture adsorption performance. The 72 h moisture adsorbed amount of DG750 reached 11.66%, 8.81% and 8.00% at 98%RH, 85%RH and 75%RH, respectively, which improved about 0.46, 0.54 and 0.53-fold as those of diatomite. The hydrophilic calcium silicate, calcium oxide and calcium hydroxide were formed in the DG750 during the calcinations process. As compared with the raw materials, the content of mesoporous component increases in the DG750, which is in favor of capillary condensation and improving moisture adsorption ability. Duchenne muscular dystrophy (DMD) is caused by defects in the DMD gene and results in progressive wasting of skeletal and cardiac muscle due to an absence of functional dystrophin. Cardiomyopathy is prominent in DMD patients and contributes significantly to mortality. This is particularly true following respiratory interventions that reduce death rate and increase ambulation and consequently cardiac load. Cardiomyopathy shows an increasing prevalence with age and disease progression and over 95% of patients exhibit dilated cardiomyopathy by the time they reach adulthood. Development of the myopathy is complex and elevations in intracellular calcium, functional muscle ischemia and mitochondrial dysfunction characterise the pathophysiology. Current therapies are limited to treating symptoms of the disease and there is therefore an urgent need to treat the underlying genetic defect. Our objective was to evaluate the effect of oral calcium administration on clinical cure, survival, subsequent presentation of peripartal health disorders, and reproductive performance of Holstein cows diagnosed with puerperal metritis (PM) under certified organic management. A second objective was to evaluate the metabolic status at calving and at the time of PM diagnosis (d 0) in affected and matched healthy cows. Cows diagnosed with PM (n = 200) were assigned randomly to receive 1 of 2 treatments: (1) control received 3.75 mL of Optimum UterFlush [Van Beek Natural Science, Orange City, IA, containing yucca extract, cinnamaldehyde, thymol, and a proprietary blend of carvacrol (4-isopropyl-2-methylphenol, at 0.47 g/mL)] diluted in 117 mL of distilled water by intrauterine infusion, administered every other day for a total of 3 treatments (n = 100); (2) calcium-supplemented (CA) received the same intrauterine treatment plus 6 oral capsules providing calcium ('O' Cal-D Cap, Bio-Vet Inc., Barneveld, WI; 7.5-9.0 g of Ca/capsule) once per day, for 3 consecutive days after diagnosis of PM. All cows received hypertonic saline solution (500 mL of 7.2% solution i.v. once), dextrose (500 mL of 50% solution i.v. once), and oral aspirin (5 boluses/d for 3 d). Outcome variables included fever, presence of fetid vaginal discharge, and uterine score at d 6 and 14 after diagnosis, survival at 30 and 100 d in milk, reproductive performance, and incidence of other health disorders after PM. A group of 200 control healthy cows (CH) was matched with PM cows at d 0, and calcium and fatty acid serum concentrations were determined at calving and at the day of diagnosis of PM (d 0). Calcium status was also assessed in PM cows at d 1, 2, 3, and 6 after diagnosis. Treatment effects were tested by logistic regression, repeated measures analysis, and ANOVA. Average calcium serum concentrations at d 0 were lower in PM cows (1.57 mmol/L) compared with CH cows (2.10 mmol/L). In PM cows, calcium concentrations at d 1, 2, 3, and 6 after diagnosis were significantly higher in the CA group. Fatty acid serum concentrations at calving and at d 0 were higher in PM cows compared with CH cows (0.48 vs. 0.37 mmol/L and 0.49 vs. 0.35 mmol/L, for calving and d 0). No effect was observed for calcium administration on health and survival outcomes. However, the proportion of cows inseminated by 150 d in milk was greater for CA compared with control cows (66 vs. 55%). In conclusion, supplementing oral calcium at the time of diagnosing PM had no effect on health. High fatty acid concentrations at calving were significant risk factors for occurrence of PM. Astrocytic excitability relies on cytosolic calcium increases as a key mechanism, whereby astrocytes contribute to synaptic transmission and hence learning and memory. While it is a cornerstone of neurosciences that experiences are remembered, because transmitters activate gene expression in neurons, long-term adaptive astrocyte plasticity has not been described. Here, we investigated whether the transcription factor CREB mediates adaptive plasticity-like phenomena in astrocytes. We found that activation of CREB-dependent transcription reduced the calcium responses induced by ATP, noradrenaline, or endothelin-1. As to the mechanism, expression of VP16-CREB, a constitutively active CREB mutant, had no effect on basal cytosolic calcium levels, extracellular calcium entry, or calcium mobilization from lysosomal-related acidic stores. Rather, VP16-CREB upregulated sigma-1 receptor expression thereby increasing the release of calcium from the endoplasmic reticulum and its uptake by mitochondria. Sigma-1 receptor was also upregulated in vivo upon VP16-CREB expression in astrocytes. We conclude that CREB decreases astrocyte responsiveness by increasing calcium signalling at the endoplasmic reticulum-mitochondria interface, which might be an astrocyte-based form of long-term depression. The effect of Ca2+ions on the cytotoxic ability of boron heterocyclic compound dipotassium-trioxohydroxytetrafluorotriborate (K2[B3O3F4OH]), onin vitrotumor cells (mammary adenocarcinoma 4T1, melanoma B16F10 and squamous cell carcinoma SCCVII) and non-tumoral fibroblast cells (mouse dermal L929 and hamster lung V79) was examined. At small concentrations of Ca2+ions (0.42 mM), K2[B3O3F4OH] (3.85 mM) has a very strong cytotoxic effect on all cancer cells tested (89.1, 85.6 and 84.6%) and significantly less effect on normal cells (19.5 and 24.2%), respectively. Applying larger concentrations of Ca2+ions (9.42–72.42 mM), at the same concentration of K2[B3O3F4OH], no significant cytotoxic effect was detected on cancer cells and normal cells investigated. The selective ability of K2[B3O3F4OH], in the medium with a low concentration of Ca2+ions has a strong cytotoxic effect on cancer cells and very weak effect in normal cells, opens up the possibility of its application in antitumor therapy. Rats overfed during lactation show higher visceral adipose tissue (VAT) mass and metabolic dysfunctions at adulthood. As both vitamin D and glucocorticoids change adipogenesis, parameters related to metabolism and action of these hormones in the adipocyte can be altered in rats raised in small litters (SL). We also studied the antiobesity effects of high calcium diet since it decreases visceral fat in obesity models. On postnatal day (PN) 3, litter size was adjusted to 3 pups/dam (SL) to induce overfeeding. Control litters (NL) remained with 10 pups/dam until weaning. From PN120 to PN180, half of the SL rats were fed standard chow (SL) and the other half was fed a calcium-supplemented chow (SL-Ca, 10 g CaCO3/kg). Both SL groups were heavier and hyperphagic when compared with the NL group; however, SL-Ca rats ate less than SL. SL-Ca rats had decreased VAT mass and adipocyte size, associated with lower hypothalamic NPY content, VAT fat acid synthase content and leptinemia. At PN120, SL rats had increased plasma 25(OH)D3, Cyp27b1 mRNA and glucocorticoid receptor (GR-α) in the VAT, but lower vitamin D receptor (Vdr) mRNA. At PN180, Cyp27b1 and GR-α remained higher, while Vdr normalized in SL rats. SL-Ca rats had normal VAT Cyp27b1 and GR-α, but lower Vdr. Thus, higher body mass and glucocorticoid receptors in the VAT of SL rats are normalized by calcium-enriched diet, and Vdr expression in this tissue is reduced, suggesting a possible role of glucocorticoids and vitamin D in calcium action in the adipocyte. In neuronal axons, the ratio of motile-to-stationary mitochondria is tightly regulated by neuronal activation, thereby meeting the need for local calcium buffering and maintaining the ATP supply. However, the molecular players and detailed regulatory mechanisms behind neuronal mitochondrial movement are not completely understood. Here, we found that neuronal activation-induced mitochondrial anchoring is regulated by Disrupted-in-schizophrenia 1 (DISC1), which is accomplished by functional association with Syntaphilin (SNPH). DISC1 deficiency resulted in reduced axonal mitochondrial movement, which was partially reversed by concomitant SNPH depletion. In addition, a SNPH deletion mutant lacking the sequence for interaction with DISC1 exhibited an enhanced mitochondrial anchoring effect than wild-type SNPH. Moreover, upon neuronal activation, mitochondrial movement was preserved by DISC1 overexpression, not showing immobilized response of mitochondria. Taken together, we propose that DISC1 in association with SNPH is a component of a modulatory complex that determines mitochondrial anchoring in response to neuronal activation. The present investigation evaluated the ability of an experimental di-calcium phosphate (DCP) desensitising agent used alone or combined with phytosphingosine (PHS) to occlude dentine tubules and resist a citric acid (CA) or artificial saliva (AS) challenge. Three groups of human dentine specimens (DS) were treated with the following: (1) PHS alone, (2) DCP or (3) a combination of PHS and DCP. Dentine hydraulic conductance of DS was evaluated using a digital flow sensor at 6.9 kPa. The average fluid volume for each of the treated DS was used to calculate the total dentine permeability reduction (%P) prior to and following CA immersion for 1 min or AS immersion for 4 weeks. The treated DS were subjected to both scanning electron microscopy (SEM) and Fourier transform infrared (FTIR) spectroscopy analysis. Statistically significant differences (%P) were identified between the groups by ANOVA and Fisher's multiple comparison test (p < 0.05), respectively. Interestingly, both PHS and DCP appeared to work synergistically. DS treated with DCP or PHS/DCP demonstrated a significant reduction (%P) prior to and following CA or AS challenge (p < 0.05). Both the SEM and FTIR analyses showed consistent brushite crystals occluding the dentine tubules. Conversely, the application of PHS alone failed to demonstrate any significant reduction of dentine permeability (p > 0.05) or show any evidence of occlusion of the dentine tubules. DCP can be used alone or combined with PHS to decrease the dentine permeability as well as to resist a CA and AS challenge. Obesity is related to diabetes, higher oxidative stress and nonalcoholic fatty liver disease, and dietetic therapies, for instance calcium-rich diet, can improve these dysfunctions. Rats raised in small litters (SL) had increased fat depots and insulin resistance at adulthood associated with higher liver oxidative stress and microsteatosis. Thus, we evaluated if dietary calcium can improve these changes. In PN3, litter size was adjusted to 3 pups (SL group) to induce overfeeding, while controls had 10 pups until weaning. At PN120, SL group was randomly divided into: rats fed with standard chow or fed with calcium supplementation (SL–Ca group, 10 g/kg chow) for 60 days. At PN180, dietary calcium normalized food consumption, visceral fat, plasma aspartate aminotransferase (AST) and glycaemia. Concerning oxidative balance, calcium restored both higher hepatic lipid peroxidation and protein carbonylation as well as higher plasma lipid peroxidation. Higher fatty acid synthase (FAS) content, steatosis and lower protein kinase B (Akt) in SL group were normalized by dietary calcium and SL–Ca rats had lower hepatic cholesterol. Thus, calcium supplementation improved the insulin sensitivity, redox balance and steatosis in the liver. Purpose: To investigate the reliability of ungated, high-pitch dual-source CT for coronary artery calcium (CAC) screening.Materials and Methods: One hundred and eighty-five smokers underwent a dual-source CT examination with acquisition of two sets of images during the same session: (a) ungated, high-pitch and high-temporal resolution acquisition over the entire thorax (i.e., chest CT); (b) prospectively ECG-triggered acquisition over the cardiac cavities (i.e., cardiac CT).Results: Sensitivity and specificity of chest CT for detecting positive CAC scores were 96.4 % and 100 %, respectively. There was excellent inter-technique agreement for determining the quantitative CAC score (ICC = 0.986). The mean difference between the two techniques was 11.27, representing 1.81 % of the average of the two techniques. The inter-technique agreement for categorizing patients into the four ranks of severity was excellent (weighted kappa = 0.95; 95 % CI 0.93-0.98). Normocalcaemic hyperparathyroidism is a common biochemical finding, usually identified during an assessment of bone or renal health. Hypercalcaemia must be considered by calculation of adjusted calcium, and a careful history taken to assess dietary calcium intake and for the possibility of a malabsorption syndrome. 25-hydroxyvitamin D (25 OHD) should be measured and replaced if indicated. The management plan for the patient is influenced by the context in which calcium and PTH were measured. Field experiments were conducted in 2006–2007 and 2007–2008 to evaluate the effects of three calcium carbide (CaC2) based formulations (slow release sources of acetylene and ethylene) - matrix-1 [(M-1)(21% CaC2, 58% polyethylene and 21% plaster of paris)], matrix-2 [(M-2)(42% CaC2, 48% polyethylene and 10% plaster of paris)] and paint coated [(PC)(35% CaC2, 44% paint and 21% plaster of paris)] - on nitrification inhibition; growth, yield and nitrogen (N)-uptake of wheat. All the three CaC2formulations (CCF,s) inhibited nitrification, reduced plant height, and improved N-uptake, spike bearing tillers, 1000-grains weight and yield of Inqulab-91 and Bhakhar-02 wheat cultivars. Increase in grain yield was found maximum (22% over the control) with M-1 applied at 30 kg CaC2ha−1; followed by M-2 and PC. Furthermore, CaC2significantly reduced lodging in Bhakhar-02 plots. Introduction: Probiotics are live microorganisms which when administered in adequate amounts confer a health benefit on the host. Sufficient calcium intake has been reported to support bone growth and prevent bone loss during the ageing process. Aim: To determine the effect of Lactobacillus helveticus MTCC 5463 probiotic dietary intervention on serum calcium & haematological parameters in geriatric population. Materials and Methods: Healthy volunteers with age ranging from 64-74 years were recruited from the nearby residential areas in and around Anand, Gujarat. Study duration was from 2012 to 2015. Of the 112 subjects initially enrolled in the trial, 36 withdrew before the intervention because of not matching with criterias. Of the 76 participants, 5 subjects (4%) under test group and 12 subjects (11%) under placebo left the study. We had 59 subjects who successfully completed a double blind cross over trial. Probiotic fermented milk products (in form of "Lassi") was prepared by supplementing toned milk with honey and fermenting with probiotic Lactobacillus helveticus MTCC 5463 and Streptococcus thermophilus MTCC 5460. The final product had at least 108 CFU/ml of viable Lactobacillus helveticus MTCC 5463 at the time of feeding. During feeding period, 200 ml of fermented product containing the test strain to one group and a similar product but without the test strain as placebo were fed regularly at the time of breakfast in morning for 4 weeks. Subjects of each group were given a washout period of 4 weeks before they were crossed over and included to the other group. The study was approved by institutional ethics committee. Results: The socio-demographic and clinical profiles were similar at baseline. The mean (SD) calcium level improved significantly in test {9.36 (0.45) vs 8.45 (0.61), p<0.001}. No significant effect was observed with respect to haemoglobin & haematological parameters. Conclusion: The well-documented probiotic Lactobacillus helveticus MTCC 5463 confirmed increase in serum calcium level but no effect on haematological parameters when administered to geriatrics. Objectives: To investigate whether dietary calcium intake will modify the relationship between serum 25-hydroxyvitamin D3 (25OHD) with intact serum parathyroid hormone (PTH) concentrations in apparently healthy Indian adolescents.Study Design: Cross-sectional study.Setting and Participants: Apparently healthy adolescents aged 10-14 years (n=181), from Gujarat, western India. Study conducted from January 2012 to March 2014.Methods: Serum 25OHD concentrations and intact serum PTH concentrations (both using chemiluminescent microparticle immunoassay) were measured. Diet was recorded through 24 h diet recall and calcium intake was computed (C-diet V.2.1). To assess relationship between 25OHD and PTH, data were dichotomised according to median calcium intakes (520 mg/day) and relationship between serum 25OHD and PTH in the two subgroups was plotted.Results: Subjects with calcium intakes above median (>520 mg/day) had lower intact serum PTH values for given serum 25OHD concentration while those with calcium intakes below median (<520 mg/day) had higher intact serum PTH values for given serum 25OHD concentration. Serum 25OHD concentration was negatively correlated with intact serum PTH concentration at lower as well as higher calcium intakes (r=- 0.606 and -0.483, respectively, p<0.01 for both). Using a regression analysis, predicted values for intact serum PTH concentration for the given serum 25OHD concentrations were plotted. The plot revealed a negative shift with increasing calcium intake.Conclusions: Dietary calcium intake modifies the relationship between serum 25OHD concentrations and intact serum PTH concentrations. The endoplasmic reticulum (ER) is at the epicenter of astrocyte Ca2+ signaling. We sought to identify the molecular mechanism underlying store-operated calcium entry that replenishes ER stores in mouse Muller cells. Store depletion, induced through blockade of sequestration transporters in Ca2+-free saline, induced synergistic activation of canonical transient receptor potential 1 (TRPC1) and Orai channels. Store-operated TRPC1 channels were identified by their electrophysiological properties, pharmacological blockers, and ablation of the Trpcl gene. Ca2+ release-activated currents (/CRAC) were identified by ion permeability, voltage dependence, and sensitivity to selective Orai antagonists Synta66 and GSK7975A. Depletion-evoked calcium influx was initiated at the Müller end-foot and apical process, triggering centrifugal propagation of Ca2+ waves into the cell body. EM analysis of the end-foot compartment showed high-density ER cisternae that shadow retinal ganglion cell (RGC) somata and axons, protoplasmic astrocytes, vascular endothelial cells, and ER-mitochondrial contacts at the vitreal surface of the end-foot. The mouse retina expresses transcripts encoding both Stim and all known Orai genes; Müller glia predominantly express stromal interacting molecule 1 (STIM1), whereas STIM2 is mainly confined to the outer plexiform and RGC layers. Elimination of TRPC 1 facilitated Müller gliosis induced by the elevation of intraocular pressure, suggesting that TRPC channels might play a neuroprotective role during mechanical stress. By characterizing the properties of storeoperated signaling pathways in Müller cells, these studies expand the current knowledge about the functional roles these cells play in retinal physiology and pathology while also providing further evidence for the complexity of calcium signaling mechanisms in CNS astroglia. Calcium imaging has been used as a promising technique to monitor the dynamic activity of neuronal populations. However, the calcium trace is temporally smeared which restricts the extraction of quantities of interest such as spike trains of individual neurons. To address this issue, spike reconstruction algorithms have been introduced. One limitation of such reconstructions is that the underlying models are not informed about the biophysics of spike and burst generations. Such existing prior knowledge might be useful for constraining the possible solutions of spikes. Here we describe, in a novel Bayesian approach, how principled knowledge about neuronal dynamics can be employed to infer biophysical variables and parameters from fluorescence traces. By using both synthetic and in vitro recorded fluorescence traces, we demonstrate that the new approach is able to reconstruct different repetitive spiking and/or bursting patterns with accurate single spike resolution. Furthermore, we show that the high inference precision of the new approach is preserved even if the fluorescence trace is rather noisy or if the fluorescence transients show slow rise kinetics lasting several hundred milliseconds, and inhomogeneous rise and decay times. In addition, we discuss the use of the new approach for inferring parameter changes, e.g. due to a pharmacological intervention, as well as for inferring complex characteristics of immature neuronal circuits. Calcium (Ca2+) mobilization is a central theme in various plant signal transduction pathways. We demonstrate that Arabidopsis thaliana cyclic nucleotide-gated channel 2 (AtCNGC2) is involved in jasmonic acid (JA)-induced apoplastic Ca2+ influx in Arabidopsis epidermal cells. Ca2+ imaging results showed that JA can induce an elevation in the cytosolic cAMP concentration ([cAMP]cyt), reaching a maximum within 3 min. Dibutyryl cAMP (db-cAMP), a cell membrane-permeable analogue of cAMP, induced an increase in the cytosolic Ca2+ concentration ([Ca2+]cyt), with a peak at 4 min. This [Ca2+]cyt increase was triggered by the JA-induced increase in [cAMP]cyt. W-7[N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide], an antagonist of calmodulin, positively modulated the JA-induced increase in [Ca2+]cyt, while W-5[N-(6-aminohexyl)-1-naphthalenesulfonamide], an inactive antagonist of calmodulin, had no apparent effect. db-cAMP and JA positively induced the expression of primary (i.e. JAZ1 and MYC2) and secondary (i.e. VSP1) response genes in the JA signalling pathway in wild-type Arabidopsis thaliana, whereas they had no significant effect in the AtCNGC2 mutant 'defense, no death (dnd1) plants. These data provide evidence that JA first induces the elevation of cAMP, and cAMP, as an activating ligand, activates the AtCNGC2 channel, resulting in apoplastic Ca2+ influx through AtCNGC2. Increasing calcium (Ca) intake is important for female athletes with a risk of weak bone caused by inadequate food intake. The aim of the present study was to examine the preventive effect of Ca supplementation on low bone strength in young female athletes with inadequate food intake, using the rats as an experimental model. Seven-week-old female Sprague-Dawley rats were divided into four groups: the sedentary and ad libitum feeding group (SED), voluntary running exercise and ad libitum feeding group (EX), voluntary running exercise and 30% food restriction group (EX-FR), and a voluntary running exercise, 30% food-restricted and high-Ca diet group (EX-FR+Ca). To Ca supplementation, we used 1.2% Ca diet as "high-Ca diet" that contains two-fold Ca of normal Ca diet. The experiment lasted for 12 weeks. As a result, the energy availability, internal organ weight, bone strength, bone mineral density, and Ca absorption in the EX-FR group were significantly lower than those in the EX group. The bone strength and Ca absorption in the EX-FR+Ca group were significantly higher than those in the EX-FR group. However, the bone strength in the EX-FR+Ca group did not reach that in the EX group. These results suggested that Ca supplementation had a positive effect on bone strength, but the effect was not sufficient to prevent lower bone strength caused by food restriction in young female athletes. The present work deals with a novel sonochemical synthesis of calcium carbonate (CaCO 3 ) by reverse miniemulsion technique using ultrasound and compares the results with conventional route of synthesis without the ultrasound. The effect of power density on the obtained particle size, crystallinity, crystallite size and morphology of the prepared CaCO 3 were investigated and optimized. XRD, SEM, TEM, FTIR were used for the characterization of prepared CaCO 3 particles. The results evinced that the size of the obtained calcium carbonate particles accomplished by acoustic cavitation method was about 20 nm, exhibited uniform morphology and gave a maximum of 98% conversion. It was clearly observed that conventionally synthesized CaCO 3 particles had wide particle size distribution (10–150 nm) and gave lower yields than sonochemical synthesis method. This work also established that sonochemical synthesis method saves more than 80% of energy as compared to the conventional method of synthesis. Finally it is evident that ultrasonication technique could be a very efficient and faster synthesis tool in controlling particle size and morphology resulting in the overall reduction in reaction time and energy. The objective of this study was to investigate whether other calcium forms have the similar effect, in particular, after postharvest dip in calcium solution. However, current postharvest dip/spinning/drying method profoundly reduces the shelf life due to mechanical damages. Background/Aims: Although osteosarcoma (OS) is the most common primary malignancy of bone, its molecular pathogenesis remains to be fully understood. We previously found the calcium-binding protein S100A6 was expressed in ~80% of the analyzed OS primary and/or metastatic tumor samples. Here, we investigate the role of S100A6 in OS growth and progression. Methods: S100A6 expression was assessed by qPCR and Western blotting. Overexpression or knockdown of S100A6 was carried out to determine S100A6's effect on proliferation, cell cycle, apoptosis, tumor growth, and osteogenic differentiation. Results: S100A6 expression was readily detected in human OS cell lines. Exogenous S100A6 expression promoted cell proliferation in vitro and tumor growth in an orthotopic xenograft model of human OS. S100A6 overexpression reduced the numbers of OS cells in G1 phase and increased viable cells under serum starvation condition. Conversely, silencing S100A6 expression induced the production of cleaved caspase 3, and increased early stage apoptosis. S100A6 knockdown increased osteogenic differentiation activity of mesenchymal stem cells, while S100A6 overexpression inhibited osteogenic differentiation. BMP9-induced bone formation was augmented by S100A6 knockdown. Conclusion: Our findings strongly suggest that S100A6 may promote OS cell proliferation and OS tumor growth at least in part by facilitating cell cycle progression, preventing apoptosis, and inhibiting osteogenic differentiation. Fresh-cut ‘Braeburn’ apple slices were dipped into cold water (4 °C for 2 min) or hot water (HWT, 48 °C or 55 °C for 2 min) followed by dips into 0 or 6% w/v aqueous calcium ascorbate (CaAsc, 2 min, 0 °C) and stored in air up to 28 d at 4 °C. Microbial counts, changes in browning and sensory acceptance were determined to indicate changes in quality. Changes in antioxidant levels were measured using free radical scavenging activity (DPPH), reducing activity (FRAP), ascorbic acid content (AA) and polyphenolic content (by HPLC). CaAsc dips had a strong impact reducing the browning through increasing the flesh luminosity and hue angle. 6% CaAsc in fresh-cut apples extended the overall acceptability from less than 7 d to 14 d. Immediately after CaAsc treatment, AA content was 5 fold higher (0.25–1.25 g kg −1 ) than those not dipped into CaAsc. However, the combination of HWT treatments and CaAsc dips led to seven fold increased levels of AA inside the apple tissue (0.25–1.85 g kg −1 ) and consequently increased the antioxidant activity. HWT did not increase the AA content when not combined with CaAsc dips. The HWT CaAsc dip extended the overall acceptability to 21 d compared to 14 d for samples not heated but dipped into CaAsc. Shelf life was ultimately limited by sensory quality. At day 28, total plate counts were reduced from 5.3 log cfu/g (untreated slices) to 4.6 log cfu/g in the 6% CaAsc dips and further to 3.9 log cfu/g with the combination of HWT and CaAsc dip. Changes in the content of phenolic compounds with time, HWT and CaAsc dip were generally not significant except for slightly increased quercetin and phloridzin levels and decreased p -coumaric and procyanidins over time. A novel and potentially cost effective filtration scheme for removal of nitrate from groundwater, characterized by production of low salinity waste brine that can be easily discharged to sewerage systems and high product-water recovery, is proposed. The inherent preference of particular NF membranes for rejecting chloride and sodium over nitrate ions is utilized in a preliminary NF stage to remove Na + , Cl + , Ca 2+ and Mg 2+ to a side stream. In a second stage, RO is applied to remove NO 3 − and the RO permeate is mixed with the side stream of the NF stage to create product water low in nitrate, yet with a balanced composition consisting all the required species and minerals. The number of NF stages depends mainly on the rejection efficiency of the NF membrane. Based on Israeli regulations for both drinking water and required composition of brines discharged to the sewage, a treatment scheme composed of a single and double NF stages followed by RO is shown to reach water recoveries of 91.6% and 94.3%, respectively. Each NF stage raises the energy cost by approximately 0.5 cent/m 3 product water. However, this cost is easily paid back by the inherent additional advantages of the combined scheme, i.e., less water treated by the RO, significant increase in total recovery ratio, no need in re-mineralization of the product water and minimization of calcium carbonate precipitation potential on the RO membrane. The principles for process design are described, making the specific treatment scheme proposed here easily adjustable to other regulatory requirements and other water characteristics. Store operated calcium entry (SOCE) is thought to primarily regulate calcium homeostasis in neurons. Subsequent to identification of Orai as the SOCE channel in nonexcitable cells, investigation of Orai function in neurons demonstrated a requirement for SOCE in Drosophila flight. Here, by analysis of an Orai mutant and by controlled expression of a dominant-negative Drosophila Orai transgene, we show that Orai-mediated SOCE is required in dopaminergic interneurons of the flight circuit during pupal development. Expression of dominant-negative Orai in dopaminergic neurons of pupae abolished flight. The loss of Orai-mediated SOCE alters transcriptional regulation of dopaminergic neurons, leading to downregulation of the enzyme tyrosine hydroxylase, which is essential for dopamine synthesis, and the dopamine transporter, which is required for dopamine uptake after synaptic release. This review assesses (1) the potential role of calcium supplements in the prevention and treatment of osteoporosis and osteoporotic fractures, and (2) the safety of calcium supplements with respect to cardiovascular health as well. With regard to (1), a total calcium intake of < 800 mg/day is associated with increased loss of bone mineral density in peri- and postmenopausal women with an increase in fracture risk. Hereby, the effect of calcium supplements on fracture prevention is dependent primary on baseline calcium intake. The strongest protective effect has been reported in individuals with a calcium intake < 700 mg/day and in high-risk groups. A calcium intake of about 1000–1200 mg/day seems to be sufficient for general fracture prevention. With regard to (2), an analysis of the data based on the Hill criteria does not demonstrate convincing evidence that calcium supplements increase cardiovascular risk. In the long term, total calcium intake of 2500 mg/day (from food and supplements) continues to be classified as safe. This value should not be exceeded for an extended period of time. Background: The risk stratification of patients with diabetes mellitus (DM) is a major objective for the clinicians, and it can be achieved by coronary flow velocity reserve (CFVR) or with coronary artery calcium score (CS). CS evaluates underlying coronary atherosclerotic plaque burden and CFVR estimates both presence of coronary artery stenosis and microvascular function. Consequently, CFVR may provide unique risk information beyond the extent of coronary atherosclerosis. Aim: Our aim is to assess joint prognostic value of CFVR and CS in asymptomatic DM patients. Materials and methods: We prospectively included 200 asymptomatic patients (45,5 % male, mean age 57,35 ± 11,25), out of which, there were 101 asymptomatic patients with DM and 99 asymptomatic patients without DM, but with one or more conventionally risk factors for coronary artery disease. We analyzed clinical, biochemical, metabolic, inflammatory parameters, CS by Agatston method, transthoracic Doppler echocardiography CFVR of left anterior descending artery and echocardiographic parameters. Results: Total CS and CS LAD were significantly higher, while mean CFVR was lower in diabetics compared to the nondiabetics. During 1 year follow-up, 24 patients experienced cardio-vascular events (one cardiovascular death, two strokes, three myocardial infarctions, nine new onsets of unstable angina and nine myocardial revascularizations): 19 patients with DM and five non DM patients, (p = 0,003). Overall event free survival was significantly higher in non DM group, compared to the DM group (94,9 % vs. 81,2 %, p = 0,002 respectively), while the patients with CS ≥200 and CFVR <2 had the worst outcome during 1 year follow up in the whole study population as well as in the DM group. At multivariable analysis CFVR on LAD (HR 12.918, 95 % CI 3.865-43.177, p < 0.001) and total CS (HR 13.393, 95 % CI 1.675-107.119, p = 0.014) were independent prognostic predictors of adverse events in DM group of patients. Conclusion: Both CS and CFVR provide independent and complementary prognostic information in asymptomatic DM patients. When two parameters are analyzed together, the risk stratification ability improves, even when DM patients are analyzed together with non DM patients. As a result, DM patients with CS ≥200 and CFVR <2 had the worst outcome. Purpose: Myocardial perfusion imaging (MPI) has limitations in the presence of balanced multivessel disease (MVD) and left main (LM) coronary artery disease, occasionally resulting in false-normal results despite the high cardiovascular risk associated with this condition. The purpose of this study was to assess the incidence of severe coronary artery disease (CAD) in the presence of a very high Agatston coronary artery calcium (CAC) score (>1,000) in stable symptomatic patients without known CAD but with normal MPI results. Methods: A total of 2,659 prospectively acquired consecutive patients were referred for MPI and evaluation of CAC score by CT. Of this patient population, 8 % (222/2,659) had ischemia without myocardial infarction (MI) on MPIand 11 % (298/2,659) had abnormal MPI (MI and/or ischemia). On presentation 1 % of the patients (26/2,659) were symptomatic, had a CAC score >1,000 and normal MPI results. The definition of normal MPI was strict and included a normal hemodynamic response without ischemic ECG changes and normal imaging, particularly absence of transient ischemic dilation. All of these 26 patients with a CAC score >1,000 and normal MPI findings underwent cardiac catheterization. Results: Of these 26 patients, 58 % (15/26) had severe disease (≥70 % stenosis) leading to revascularization. Of this group, 47 % (7/15) underwent percutaneous intervention, and 53 % (8/15) underwent coronary artery bypass grafting. All of these 15 patients had either MVD (14/15) or LM coronary artery disease (1/15), and represented 0.6 % (15/2,659) of all referred patients (95 % CI 0.3 - 0.9 %). The majority, 90 % (8/9), had severe CAD with typical chest pain. Conclusion: A very high CAC score (>1,000) with normal MPI in a small subset of symptomatically stable patients was associated with a moderate incidence of severe CAD (95 % CI 37 - 77 %). Larger studies and/or a meta-analysis of small studies are needed to more precisely estimate the incidence of CAD in this population. Background: The role of calcium hydroxylapatite injection pharyngoplasty after failed sphincter pharyngoplasty surgery has not previously been described. We report our technique of injecting calcium hydroxylapatite into the pharyngoplasty site to add bulk to specific areas of the velopharyngeal port. Methods: A retrospective review of children undergoing calcium hydroxylapatite injection pharyngoplasty for persistent velopharyngeal insufficiency after sphincter pharyngoplasty was performed. Our surgical technique is described. Outcome measures included postoperative perceptual assessment of resonance, improvement in phoneme-specific nasometry scores, and rate of revision surgery. Results: Fourteen patients were included in the study. Improvement in hypernasality on perceptual assessment was noted in the majority of children (85.7%). The rate of achievement of normal resonance on perceptual analysis was 35.7%. A significant decrease in nasometry scores was noted for the following phonemes: /pa/, /sa/, /sha/, and /pi/. There were no intraoperative complications or immediate postoperative complications associated with calcium hydroxylapatite injection. Conclusions: Our early experience suggests that calcium hydroxylapatite injection after failed sphincter pharyngoplasty is a safe, minimally invasive technique with favorable short-term results. © 2015 S. A high-fat diet not only leads to obesity, but also leads to a predisposition towards insulin resistance (IR), which is characterized by hyperinsulinemia and reduced glucose tolerance. However, the etiology of IR remains to be fully elucidated. The present study investigated whether calcium-sensing receptor (CaSR) is involved in the development of IR in rats fed a high-fat diet. IR was induced in the rats by feeding with a fat emulsion via gavage for 2, 4, 6 or 8 weeks. Reverse transcription-quantitative polymerase chain reaction (RT-q-PCR) and western blot analysis were performed to investigate whether CaSR-associated proteins were affected. The gavage of fat emulsion for 8 weeks induced a notable decline in the insulin sensitivity index (ISI) between -4.98 and -5.60. With 6 weeks of gavage, a significant difference in the ISI was observed between the IR and control groups. The results of the RT-qPCR and western blot analysis demonstrated that phosphatidylinositol 3-kinase/Akt pathway, which is a pathway closely associated with the CaSR signaling pathway, was significantly inhibited in the rats with IR. The results of the present study provided evidence that CaSR is associated with the development of IR in rats fed a high-fat diet and suggested that CaSR may be important in the pathogenesis of diabetes.
