Die vorliegende Arbeit untersucht die Wirkungen von CpG-DNA auf die Verteilung von Thrombozyten und Leukozyten im Blut und in den lymphatischen Organen sowie mögliche Mechanismen, die dabei eine Rolle spielen. CpG-DNA führt bereits nach 3 Stunden zu einer transienten Leukopenie im Blut, die v. a. die Lymphozyten betrifft und etwas verzögert auch zu einer Granulozytopenie führt. In dieser Phase kommt es zu markanten Veränderungen im Differentialblutbild mit einem starken prozentualen Anstieg der Granulozyten. Nach 4 bis 5 Tagen erreichten die untersuchten Parameter wieder die Ausgangswerte. Gleichzeitig mit dem Zellabfall im Blut kommt es zu einer deutlichen Vergrößerung von Milz und Lymphknoten durch eine Erhöhung der Zellzahl in diesen Organen. Dies lässt vermuten, dass Umverteilungsprozesse von Zellen aus dem Blut in lymphatische Organ hierbei eine wichtige Rolle spielen. Eine Beteiligung neu synthetisierter Chemokine ELC und SDF nach Gabe der CpG-DNA bei der frühen Phase der Umverteilung der Zellen konnte nicht nachgewiesen werden. Thrombozyten fallen nach CpG-DNA-Gabe ebenfalls stark ab. Bei TNFR(p55)-/--Mäusen ist dieser Effekt jedoch nicht zu beobachten. Damit konnte gezeigt werden, dass TNFa in der Vermittlung der Thrombozytopenie eine wichtige Rolle spielt., The dissertation describes the effects of CpG-DNA on the distribution of thrombocytes and leucocytes in the blood and lymphatic organs as well as possible mechanisms which are involved in this process. Simultaneously with the decrease of cell numbers in the blood there is a clear augmentation of the spleen and the lymphnodes due to an increase of cell numbers in these organs. This leads to the conclusion that redistributionprocesses of cells from the blood into lymphatic organs play a crucial role. In this early phase there was no evidence of any effect of newly synthesized chemokines ELC and SDF on the redistribution of the mentioned cells After the application of CpG-DNA there is an explicit drop of thrombocytes, too. In TNFR(p55)-/- mice this effect could not be detected. This proofs that TNFa is indispensable for the observed thrombopenia

In dieser Arbeit wurde unter Verwendung von Modellplasmiden, die für das Protein Ovalbumin bzw. das Peptid SIINFEKL kodieren, untersucht, ob Plasmid-DNA zur Immunstimulation verschiedener muriner Zellpopulationen (CTL, B-Zellen, dendritische Zellen) in der Lage ist und inwiefern diese einer TLR9-Abhängigkeit unterliegt. Es konnte mittels in vitro Experimenten gezeigt werden, dass pDNA B-Zellen, dendritische Zellen und murine Makrophagen zu Zytokinproduktion und vermehrter Oberflächenexpression kostimulatorischer Moleküle induzieren kann. Die Proliferation von B-Zellen ist hingegen nicht durch Plasmid-DNA induzierbar. Plasmid-DNA ist weiterhin an der DC-Aktivierung beteiligt. Es konnte nachgewiesen werden, dass sowohl die Zytokinsynthese, als auch die Expression kostimulatorischer Oberflächenmarker durch pDNA induziert wird. Unter Verwendung von dendritischen Zellen aus TLR9-/- Mäusen konnte darüberhinaus gezeigt werden, dass die Induktion von Zytokinproduktion, wie auch die Oberflächenexpression kostimulatorischer Moleküle TLR9-vermittelt ist. Bei der in vivo-Vakzinierung von Mäusen war die Induktion peptidspezifischer zytotoxischer Lyse überraschenderweise nicht TLR9-vermittelt, da keine Unterschiede zwischen den wt- und ko-Milzzellen festzustellen waren. Eine Umgehung des TLR-9 Signalwegs via anderer TLRs konnte durch MyD88-/- Milzzellen ausgeschlossen werden. Diese Ergebnisse widersprechen den bisherigen Annahmen, dass CpG-DNA/TLR9-Wechselwirkungen eine wichtige Rolle bei Langzeit-Immunisierungsprotokollen spielen., We analyzed whether the immunobiology of vaccinating plasmid DNA containing a transcription unit for OVA (SIINFEKL) is influenced by immunostimulatory CpG motifs in the plasmid backbone and functions via Toll-like receptor 9 (TLR9). Indeed, plasmid DNA activated in vitro b-cells, macrophages and dendritic cells (DCs) provided they expressed the CpG-DNA receptor TLR9. Activation resulted in cytokine production and up-regulated expression of costimulatory molecules CD40 and CD86. In vivo, however, even upon repeated vaccination with plasmid DNA, priming of OVA-specific CTL and clonal expansion of SIINFEKL-specific CD8 T cells were equal in TLR9-positive and TLR9- or MyD88-negative mice. Overall, these results negate a dominant role of CpG-DNA/TLR9 interactions in long-term vaccination protocols.

Chlamydia pneumoniae ist ein obligat intrazelluläres Bakterium, das insbesondere Atemwegsinfektionen hervorruft. In dieser Arbeit wird gezeigt, dass mit Chlamydia pneumoniae infizierte HeLa 229-Zellen gegen experimentell (mit Staurosporin oder anti-CD95) induzierte Apoptose geschützt sind. Apoptose ist eine physiologische, für höherstehende Lebewesen wichtige Form des Zelltodes. Sie kann durch verschiedene Stimuli ausgelöst werden und ist Folge spezialisierter Signaltransduktion. In diesem Signaltransduktionsweg ist die Aktivierung der Caspasen-3, -8 und -9 und die Freisetzung von Cytochrom c aus den Mitochondrien durch Infektion mit Chlamydia pneumoniae blockiert. Hierzu wird die bakterielle Proteinsynthese zu einem frühen Zeitpunkt des Infektionszyklus benötigt. Wie weiter gezeigt, spielen zelluläre Einflüsse durch das NF-kB-System, HSP-70- oder Bcl-2-Überexpression keine Rolle., Chlamydia pneumoniae is an obligate intracellular bacterium which frequently causes airway infection. In this signal transduction pathway the activation of caspases-3, -8 and -9 and the release of cytochrome c from mitochondria is blocked by infection with Chlamydia pneumoniae. Bacterial protein synthesis is needed for this at an early stage of the infection cycle. As further showed cellular influences of the NF-kB-system, HSP-70- or Bcl-2-overexpression do not play any role.

Der aktivierende Rezeptor NKG2D erkennt eine Reihe unterschiedlicher Liganden, die zumeist von gestressten Geweben und Tumoren überexprimiert werden. Bislang sind für alle Liganden gleichermaßen immunstimulatorische Effekte auf NK-Zellen und CD8+ T-Zellen beschrieben worden. Das NKG2D Rezeptor-/Liganden-System wurde deswegen als ein Sensorsystem zum Erkennen entstehender Tumoren und zur Aktivierung von Immunantworten interpretiert. In dieser Arbeit sollte die NKG2D-Liganden-Interaktion auf antigen-spezifische CD8+ T-Zellen charakterisiert werden. Dabei konnte erstmalig ein Unterschied zwischen einzelnen Liganden beschrieben werden. So führen die NKG2D-Liganden H60 und MICA im Gegensatz zu den ebenfalls untersuchten Liganden Rae1 und MULT1 zu einem stark suppressiven Effekt auf die T-Zell-Proliferation. Der von H60 und MICA ausgelöste Effekt benötigt IL-10 und wird über einen von NKG2D verschiedenen Rezeptor vermittelt. Diese Befunde könnten erklären, warum in vivo trotz hoher Expression von NKG2D-Liganden z.B. auf Tumoren gelegentlich keine effektiven Immunantworten ausgelöst werden können und stellt dabei einen Zusammenhang mit einer bestimmten Gruppe von NKG2D-Liganden her., The activating receptor NKG2D recognizes a wide range of different ligands, some of which are primarily expressed in "stressed" tissues or on tumor cells. Until now, similar stimulatory effects on natural killer and CD8+ T cells have been described for all NKG2D ligands, and the NKG2D receptor/ligand system has therefore been interpreted as a sensor system involved in tumor immune surveillance and activation of immune responses. The aim of this work was the characterization of the NKG2D receptor/ligand interactions on antigen-specific CD8+ T cells. For the first time differences between distinct ligands could be shown. The NKG2D ligands H60 and MIC class 1 chain-related protein A (MICA) in contrast to other ligands like Rae1 and MULT1 can also mediate strong suppressive effects on T cell proliferation. Responsiveness to H60- and MICA-mediated suppression requires IL-10 and involves a receptor other than NKG2D. These findings might provide explanations for the observation that strong in vivo NKG2D ligand expression, such as that on tumor cells, sometimes fails to support effective immune responses and links this observation to a distinct subgroup of NKG2D ligands.
