While demyelination is the most common etiology of longitudinally extensive myelopathy, other causes are important to recognize. In this study, we present the case of a longitudinal cervical lesion with a very rare cause. We discuss the approach to the differential diagnosis and workup for longitudinal myelopathy. This clinical reasoning case also illustrates the anatomical relationship between symptomatic spinal cord lesions and nonsymptomatic intracranial etiologies.

Background This study analyzed the motor development and suspected developmental coordination disorder of very and moderately preterm (+0 gestational age), late preterm (34+0–36+6 gestational week), and early-term (37+0–38+6 gestational week) children compared to their full-term peers with a national population-based sample in China. Methods A total of 1673 children (799 girls, 874 boys) aged 3–10 years old were individually assessed with the Movement Assessment Battery for Children-second edition (MABC-2). The association between gestational age and motor performance of children was analyzed using a multilevel regression model. Results The global motor performance [β = – 5.111, 95% confidence interval (CI) = – 9.200 to – 1.022; P = 0.015] and balance (β = – 5.182, 95% CI = – 5.055 to – 1.158; P = 0.003) for very and moderately preterm children aged 3–6 years old were significantly lower than their full-term peers when adjusting for confounders. Late preterm and early-term children showed no difference. Moreover, very and moderately preterm children aged 3–6 years had a higher risk of suspected developmental coordination disorder (DCD) (≤ 5 percentile of MABC-2 score) when adjusting for potential confounders [odds ratio (OR) = 2.931, 95% CI = 1.067–8.054; P = 0.038]. Late preterm and early-term children showed no difference in motor performance from their full-term peers (each P > 0.05). Conclusions Our findings have important implications for understanding motor impairment in children born at different gestational ages. Very and moderately preterm preschoolers have an increased risk of DCD, and long-term follow-up should be provided for early detection and intervention.

Based on Ansteel’s new No. 5 blast furnace (BF), the distribution rule of hot blast on the circumference of blast furnace hearth under different blast volume was studied. The results show that, the blast volume distribution rule is similar under different blast volumes, that is, there are four regions with large blast volumes at the 0°, 90°, 180° and 270° positions of the bustle pipe. Under different blast volumes, the difference of blast volume near 90° and 270° is close to 0, which has a minor effect on the uneven distribution of gas flow and circumferential asymmetry of packed bed in BF. However, the blast volume of the tuyere near the 180° is always larger than that of the tuyere near 0°, and with the increase of blast volume from 4600 to 5000 nm3/min, this difference keeps increasing, 0.69 to 0.95 nm3/min. This phenomenon will lead to an increase in the coke consumption on the 180° side, and cause a higher descending velocity of coke than that on the 0° side, this difference increases from 0.39 to 0.54 m, which could further result in the unevenness of blast volume distribution and the circumferential asymmetry of packed bed in BF. Comparing with the actual production in Ansteel, the results obtained in this work are in good agreement with the phenomenon in practical production.

A high-resolution synchrotron X-ray diffraction study of a single-crystal YCrO3 compound was employed to obtain its crystallographic information, such as lattice parameters, atomic positions, bond lengths and angles, and local crystalline distortion size and mode. The measurements were taken at 120 K (below the antiferromagnetic transition temperature T N ≃ 141.5 K), 300 K (between T N and the ferroelectric transition temperature T C ≃ 473 K) and 500 K (above T C). Using the high intensity of synchrotron X-rays, it was possible to refine collected patterns with the previously proposed noncentrosymmetric monoclinic structural model (P1211, No. 4) and determine detailed structural parameters. Meanwhile, for a controlled study, the data were refined with the centrosymmetric orthorhombic space group (Pmnb, No. 62). The lattice constants a, b and c and the unit-cell volume increased nearly linearly upon heating. With the P1211 space group, the distributions of bond lengths and angles, as well as local distortion strengths, were observed to be more dispersed. This implies that (i) the local distortion mode of Cr2O6 at 120 K correlates with the formation of canted antiferromagnetic order by Cr1–Cr2 spin interactions, primarily via intermediate O3 and O4 ions; and (ii) the previously reported dielectric anomaly may have a microscopic origin in the strain-balanced Cr1—O3(O4) and Cr2—O5(O6) bonds as well as the local distortion modes of Cr1O6 and Cr2O6 octahedra at 300 K. Local crystalline distortion is shown to be an important factor in the formation of ferroelectric order. The comprehensive set of crystallographic information reported here allows for a complete understanding of the unique magnetic and ferroelectric properties of YCrO3.

This study aims to establish an optimization procedure to define the cut-offs of quantitative assays for acetylcholine receptor antibody (AChRAb), evaluate their diagnostic performance in myasthenia gravis (MG), and explore the association with clinical features.Samples from a representative cohort of 77 MG patients, 80 healthy controls (HC) and 80 other autoimmune diseases (OAD) patients were tested using competitive inhibition ELISA and RIA. Raw values (OD and cpm) and processed values (inhibition rate, binding rate and concentration) were used to define the cut-offs with statistical methods, a rough method, and receiver operating characteristic (ROC) curve. Optimal cut-offs were selected by comparing false positive rates in HC and OAD individuals. The diagnostic performance was evaluated in whole MG cohort and subgroups. Agreement between ELISA and RIA for AChRAb positivity were examined with Kappa test and McNemar test. Clinical association with AChRAb was explored by comparison among subgroups and with Spearman rank correlation.The optimal cut-offs for AChRAb positivity were determined as OD ≤ 1.79 for ELISA and cpm ≥ 1234.12 for RIA, which derived from statistical method and performed better than those derived from ROC curves. The sensitivity and specificity were 74.03%, 100% for ELISA, and 74.03%, 99.37% for RIA. There was good agreement between ELISA and RIA for AChRAb positivity in whole cohort and subgroups (weighted к ≥ 0.71, p 0.01; McNemar test, p 0.05). Levels of AChRAb were different in MG subgroups (p 0.01). Correlation between Quantitative Myasthenia Gravis scores and AChRAb levels was moderate for ELISA and RIA (rThe raw testing values of ELISA and RIA were found as optimal quantitative measures of AChRAb levels. There are good agreements on diagnostic performance between two assays. Quantitative values are more informative than positivity in association with clinical features.

A taxonomic identification using polyphasic approach was performed on strain TH16-21T, which was isolated from the interfacial sediment of Taihu Lake, PR China. Strain TH16-21T was Gram-stain-negative, aerobic, rod-shaped and catalase-positive. Phylogenetic analysis based on the 16S rRNA gene and genomic sequences indicated that strain TH16-21T was classified within the genus of Flavobacterium . The 16S rRNA gene sequence of strain TH16-21T showed the highest similarity to Flavobacterium cheniae NJ-26T (98.9 %). The average nucleotide identity and digital DNA–DNA hybridization values between strain TH16-21T and F. cheniae NJ-26T were 91.2 and 45.9 %, respectively. The respiratory quinone was menaquinone 6. The major cellular fatty acids (>10 %) comprised iso-C15 : 0, iso-C16 : 0, iso-C15 : 1 G and iso-C16 : 0 3-OH. The genomic DNA G+C content was 32.2 mol%. Phosphatidylethanolamine, six amino lipids and three phospholipids were the main polar lipids. Based on the phenotypic features and phylogenetic position, a novel species with the name Flavobacterium lacisediminis sp. nov. is proposed. The type strain is TH16-21T (=MCCC 1K04592T=KACC 22896T).

Background Pulmonary fibrosis (PF) is a severe chronic interstitial lung disease with high mortality, and there is currently a lack of definite and effective treatment methods. Studies have found that gentiopicroside, a secoiridoid glycoside compound derived from plants of the Gentiana genus, can significantly improve pulmonary inflammation and fibrosis lesions in mice with pulmonary fibrosis. However, the mechanism of its anti-fibrotic effect is unclear. Therefore, in this study, we used the virtual computer technology of network pharmacology to theoretically explore the mechanism of Gentiana rigescens 's anti-pulmonary fibrosis effect, providing theoretical reference and direction for subsequent experiments.Results A total of 10 active compounds and 920 drug-target proteins were identified from the TCMSP database. The compound-target-pathway-disease network showed that G. rigescens could potentially treat PF by regulating the MDM2, ERBB2 and VEGFA, signaling pathways through its key targets, including AKT1, TNF, and MAPK1. The protein‒protein interaction network revealed that these targets had strong interactions with each other, indicating a potential synergistic effect of G. rigescens in treating PF. The GO and KEGG enrichment analyses further supported the potential anti-pulmonary fibrosis mechanisms of Gentiana rigescens, including regulating the inflammatory response, ECM-receptor interaction, and TGF-beta signaling pathways.Conclusion Our study provides a systematic analysis of the potential anti-pulmonary fibrosis mechanisms of G. rigescens based on network pharmacology. These findings could contribute to the development of novel treatments for PF and provide a basis for further experimental studies.

We investigated the safety and explore potential efficacy of batoclimab administered subcutaneously in Chinese patients with generalized myasthenia gravis (gMG).A randomized, double-blinded, placebo-controlled, parallel phase II study was conducted. First, in the double-blinded treatment period, eligible patients received batoclimab (680 mg), batoclimab (340 mg), or placebo on days 1, 8, 15, 22, 29, and 36. In the open-label treatment period, patients received batoclimab (340 mg) on days 50, 64, and 78. In the follow-up period, patients were examined on days 92, 106, and 120. The primary endpoint was Myasthenia Gravis Activities of Daily Living (MG-ADL) score change on day 43 from baseline.In total, 30 eligible patients were enrolled, with 11, 10, and 9 patients in the batoclimab 680 mg, batoclimab 340 mg, and placebo groups, respectively. MG-ADL score changes from baseline to day 43 were -2.2 ± 0.9, -4.7 ± 0.6, and -4.4 ± 1.0 in the placebo, batoclimab 340 mg, and 680 mg groups, respectively. Similar changes were observed in Quantitative Myasthenia Gravis, Myasthenia Gravis Composite, and 15-item Myasthenia Gravis Quality of Life scores in the placebo, batoclimab 340 mg, and 680 mg groups, respectively. The proportion of patients with clinically significant improvement on day 43 was higher in the batoclimab groups. On day 120, all four scales in the placebo group had more significant improvement compared with the batoclimab groups, with total serum IgG levels reaching a plateau. No death or treatment-emergent adverse events (TEAEs) led to study discontinuation.Batoclimab is effective and safe in Chinese patients with gMG.This study was registered at ClinicalTrials.gov (NCT04346888) on 15 April 2020, with the first patient enrolled on 23 July 2020.

BACKGROUND Complex tibial plateau fractures can seriously affect quality of life and physical and mental health of patients. The anatomical relationship between the proximal tibial bone and soft tissue is complex, resulting in different types of tibial plateau fractures. Violent trauma can lead to displaced fracture, serious soft tissue injury, and potentially, dislocation of the knee joint. Therefore, tibial plateau fractures are extremely unstable. AIM To assess the use of locking compression plate (LCP) + T-type steel plate for postoperative weight bearing and functional recovery of complex tibial plateau fractures. METHODS Ninety-seven patients with complex tibial plateau fractures who underwent surgery at our hospital were selected for retrospective study. Forty-nine patients had been treated with LCP + T-type steel plate limited internal fixation (study group), and 48 patients with bilateral ordinary steel plate support (control group). The operation process index, postoperative rehabilitation related index, Rasmussen score of the knee joint, tibial plateau varus angle (TPA), tibial plateau retroversion angle (PA), and surgical complications of the two groups were compared. RESULTS The operation time and intraoperative bone graft volume in the study group were lower than those in the control group (P < 0.05). There were no significant differences in surgical bleeding, anterior external incision length, postoperative drainage, hospital stay duration, and fracture healing time between the groups (P > 0.05). There was no significant difference in the TPA and PA angle between the groups immediately and 18 mo after surgery (P > 0.05). At 12 mo after surgery, the Rasmussen scale score was higher in the study group than in the control group (P < 0.05). There was no significant difference in the Rasmussen scale score at 18 mo after surgery, and the radiology score at 12 and 18 mo after surgery, between the two groups (P > 0.05). The postoperative complication rate in the study group (3.77%) was lower than that in the control group (15.09%; P < 0.05). CONCLUSION LCP + T-type steel plate internal fixation has advantages in terms of minimizing trauma and enabling early postoperative functional exercise, promoting functional recovery and lower limb weight-bearing, and reducing postoperative complications.

Transparent glass-ceramic is a highly attractive class of materials for photonic application. Transition metal ions are promising candidates for luminescence centers thanks to the sensitivity to the ligand ions of d-d electron transition. Here, the broadband second near-infrared (NIR-II) photoluminescence was achieved via precipitation of ZnGa2O4:Ni2+ nanocrystals within a zinc gallium-silicate glass matrix by the crystallization process. Excited by 260 nm UV photon, ZnGa2O4:Ni2+ GC displays NIR-II emission peaking at 1240 nm and has a full width at half maximum wider than 288 nm, attributing to 3T2g(3F)→3A2g(3F) electronic transition of octahedral coordinated Ni2+ ions. Moreover, broad persistent luminescence of Ni2+ ions can be effectively obtained after irradiation in the UV light. Based on the electron paramagnetic resonance spectra and thermoluminescence method, a physical model was constructed to demonstrate the mechanism of persistent luminescence. This developed afterglow luminescent glass-ceramic exhibits a potential application in biological imaging.

As typical solid-state electrolytes (SSEs), {Na}$_{1+x}${Zr}$_2${Si}$_{x}${P}$_{3-x}${O}$_{12}$ NASICONs provide an ideal platform for solid-state batteries (SSBs) that display higher safety and accommodate higher energy densities. The critical points for achieving SSBs with higher efficiencies are to improve essentially the ionic conductivity and to reduce largely the interfacial resistance between SSEs and cathode materials, which would necessitate extremely high level of craftsmanship and high-pressure equipment. An alternative to higher-performance and lower-cost SSBs is additive manufacturing. Here, we report on an effective additive, MgF$_2$, which was used in synthesizing NASICONs, resulting in SSEs with fewer defects and higher performance. With an addition of mere 1 wt$\%$ MgF$_2$ additive, the total room-temperature ionic conductivity of the NASICON electrolyte reaches up to 2.03 mS cm$^{-1}$, improved up to $\sim$ 181.3$\%$, with an activation energy of 0.277 eV. Meanwhile, the stability of the Na plating/stripping behavior in symmetric cells increases from 236 to 654 h. We tried to reveal the microscopic origins of the higher ionic conductivity of MgF$_2$-doped NASICONs by comprehensive in-house characterizations. Our study discovers a novel MgF$_2$ additive and provides an efficient way to prepare higher-performance SSEs, making it possible to fabricate lower-cost SSBs in industries., Comment: 16 pages, 7 figures

The myelin regulatory factor (MYRF; MIM# 608329) gene was first identified as a critical transcription factor involved in oligodendrocyte differentiation and central nervous system myelination. With the recent development of exome sequencing, pathogenic variants of MYRF had been considered as the cause of cardiac-urogenital syndrome (CUGS), 46,XY and 46,XX disorders/differences of sex development (DSDs), and nanophthalmos. Herein, we described a 4-year-7-month-old “girl” with ventricular septal defect, atrial septal defect, patent ductus arteriosus, severe pulmonary hypertension, moderate-to-severe tricuspid regurgitation, enlarged coronary sinus, left superior vena cava, and right lung hypoplasia at birth. Later, the patient developed short stature and amblyopia. Further examination revealed a karyotype 46,XY and visible uterus, whereas the presence of gonads were not explored. Laparoscopy revealed dysplasia of testicular gonad. Whole-exome sequencing (WES) was performed and a de novo heterozygous mutation in MYRF was identified, known as c.2817G > A/p. W939* (NM_001127392.3). Therefore, this case report presented multiple clinical manifestations with syndromic symptoms of CUGS, 46,XY DSD, and ocular symptoms. These new data expanded the phenotype of the MYRF variant and may benefit to characterize the phenotypes caused by the variants of this gene.

RAV transcription factors (TFs) are unique to higher plants and contain both B3 and APETALA2 (AP2) DNA binding domains. Although sets of RAV genes have been identified from several species, little is known about this family in wheat. In this study, 26 RAV genes were identified in the wheat genome. These wheat RAV TFs were phylogenetically clustered into three classes based on their amino acid sequences. A TaRAV gene located on chromosome 1D was cloned and named TaRAV1. TaRAV1 was expressed in roots, stems, leaves, and inflorescences, and its expression was up-regulated by heat while down-regulated by salt, ABA, and GA. Subcellular localization analysis revealed that the TaRAV1 protein was localized in the nucleus. The TaRAV1 protein showed DNA binding activity in the EMSA assay and transcriptional activation activity in yeast cells. Overexpressing TaRAV1 enhanced the salt tolerance of Arabidopsis and upregulated the expression of SOS genes and other stress response genes. Collectively, our data suggest that TaRAV1 functions as a transcription factor and is involved in the salt stress response by regulating gene expression in the SOS pathway.

Epoxy resin (EP), as a kind of dielectric polymer, exhibits the advantages of low-curing shrinkage, high-insulating properties, and good thermal/chemical stability, which is widely used in electronic and electrical industry. However, the complicated preparation process of EP has limited their practical applications for energy storage. In this manuscript, bisphenol F epoxy resin (EPF) was successfully fabricated into polymer films with a thickness of 10~15 μm by a facile hot−pressing method. It was found that the curing degree of EPF was significantly affected by changing the ratio of EP monomer/curing agent, which led to the improvement in breakdown strength and energy storage performance. In particular, a high discharged energy density (Ud) of 6.5 J·cm−3 and efficiency (η) of 86% under an electric field of 600 MV·m−1 were obtained for the EPF film with an EP monomer/curing agent ratio of 1:1.5 by hot pressing at 130 °C, which indicates that the hot−pressing method could be facilely employed to produce high−quality EP films with excellent energy storage performance for pulse power capacitors.

With the increase in pet keeping among families in China, there has been a corresponding increase in the incidence of allergic diseases such as bronchial asthma and allergic rhinoconjunctivitis. Pet allergies pose important social and health problems, making their management in children crucial. There is an increasing trend of sensitization to cat and dog dander among Chinese children. The process underlying allergic sensitization is complex and is associated with an IgE-mediated type I hypersensitivity reaction that involves the upregulation of type 2 T-helper (Th2) cells, resulting in Th2 immunity. Compared with traditional serum-specific IgE (sIgE) and skin tests, molecular diagnosis may be better at predicting the progression of diseases. The association between pet keeping and allergic diseases in children remains controversial. Although the hygiene hypothesis is widely accepted in Chinese children, exposure to pets in early life remains a risk factor for allergies. Pet avoidance and allergy immunotherapy (AIT) are recommended for children with pet allergies. However, pet avoidance is difficult to maintain, and AIT is not widely available in China. There is an imbalance between the increase in allergic diseases and the shortage of prevention and treatment options. We hope that advancements in medicine, diagnosis, and healthcare policies will help resolve this issue.

Metastasis is the major cause of death in patients with pancreatic ductal adenocarcinoma (PDAC), and circulating tumor cells (CTCs) play an important role in the development of metastasis. However, few studies have uncovered the metastasis mechanism of PDAC based on CTCs. In this study, the existing bulk RNA-sequencing (bulk RNA-seq) and single-cell sequencing (scRNA-seq) data for CTCs in pancreatic cancer were obtained from the Gene Expression Omnibus (GEO) database. Analysis of tumor-infiltrating immune cells (TIICs) by CIBERSORT showed that the CTCs enriched from the peripheral blood of metastatic PDAC were found to contain a high proportion of T cell regulators (Tregs) and macrophages, while the proportion of dendritic cells (DCs) was lower than that enriched from localized PDAC. Through weighted gene co-expression network analysis (WGCNA) and the result of scRNA-seq, we identified the hub module (265 genes) and 87 marker genes, respectively, which were highly associated with metastasis. The results of functional enrichment analysis indicated that the two gene sets mentioned above are mainly involved in cell adhesion and cytoskeleton and epithelial–mesenchymal transition (EMT). Finally, we found that HMGB3 was the hub gene according to the Venn diagram. The expression of HMGB3 in PDAC was significantly higher than that in normal tissues (protein and mRNA levels). HMGB3 expression was significantly positively correlated with both EMT-related molecules and CTC cluster–related markers. Furthermore, it was also found that HMGB3 mutations were favorably related to tumor-associated immune cells through the TIMER2.0 online tool. We further demonstrated that PDAC patients with higher HMGB3 expression had significantly worse overall survival (OS) in multiple datasets. In summary, our study suggests that HMGB3 is a hub gene associated with EMT in CTCs, the formation of CTC clusters, and infiltration patterns of immune cells favorable for tumor progression and metastasis to distant organs.

BackgroundThe absolute change in the severity score between the baseline and pre-specified time frame (absolute criterion) was recommended as a criterion for myasthenia gravis (MG) treatment response. But heterogeneity of disease severity might dilute major changes in individual patients. The rationality of relative criterion (improvement percentage) had not been evaluated in treatment response in patients with MG.ObjectivesTo investigate the consistency between an absolute criterion and a relative criterion in the evaluation of treatment response in patients with MG.MethodsWe retrospectively analyzed the treatment response to a 3-month standardized treatment protocol with only glucocorticoid in 257 MG patients native to immunological treatments. With the commonly used absolute criterion, cut-offs of relative criteria were generated with the receiver operating characteristic (ROC) curve in the whole cohort and in patients with different degrees of baseline severity stratified by pre-treatment quantitative myasthenia gravis score (QMGS). The consistency between absolute and relative criteria was examined with Cohen's Kappa test and Venn diagrams.ResultsThe absolute and relative criteria had an overall substantial consistency (Kappa value, 0.639, p < 0.001) in the cohort. The Kappa values were substantial to almost perfect in mild and moderate groups and moderate in severe groups between the absolute and relative criteria (all p ≤ 0.001). More patients were classified as responsive with an absolute criterion while as unresponsive with a relative criterion in the moderate and severe groups.ConclusionsThe overall consistency between absolute and relative criteria was substantial in the whole cohort. The inconsistency between the two criteria was mainly from the moderate or severe patients at the baseline.

GaFeO3-type oxides are promising multiferroic materials due to the coexistence of spontaneous magnetization and ferroelectric polarization properties at room temperature. As these ferroic properties feature a large anisotropy, single crystals are required. However, the magnetization of GaFeO3-type single crystals remains low at room temperature. In this study, we largely enhanced the magnetization at room temperature of GaFeO3-type single crystals by increasing the Fe content and co-doping Sc3+ and Al3+. Single crystals of Al xSc0.1− xGa0.6Fe1.3O3 ( x = 0.01–0.04) were prepared using the optical floating-zone method. The single crystals were rod-shaped, with a diameter and length of ∼6 mm and 7 cm, respectively. X-ray diffraction measurements confirmed the ferroelectric polarization of the crystals. In addition, they exhibited room-temperature ferrimagnetism, with Curie temperature in the range of 326–338 K; the crystals exhibit magnetic anisotropy along the a-axis. The magnetization of the single crystal at 300 K and 0.3 kOe was 13 emu g−1, which is over ten times larger than those of previously reported single crystals with GaFeO3-type crystal structure.

By introducing halogens to the A/Ga/Q (A = Na, K; Q = S, Se) system, one new chalcohalide namely Na2GaS2Cl was successfully obtained. It crystallizes in the orthorhombic space group Cmcm (63). Na2GaS2Cl has a layered structure consisting of two dimensional [GaS2]∞ layers which are stacked in “face to face” and “back to back” arrays and separated by Na+ and Cl− ions. Interestingly, supertetrahedral building units [Ga4S10] (T2) which are rarely found in metal chalcogenides and metal chalcohalides are formed in this structure. Moreover, the distances of two adjacent layers are around four times larger than the ionic radius of the Na+ ion, which is very likely to provide a perfect environment for the storage and migration of Na+ ions. In particular, the volume concentration of the Na+ cations in this compound is as high as 1.54 × 1022 cm−3. The UV-vis-NIR spectroscopy measurement reveals that the optical band gap of this title compound is 3.06 eV. The electronic structural calculations on Na2GaS2Cl show that the band gap is mainly determined by the [GaS4] groups and Na–Cl ionic bonding.

Background/aim Curcumin is a polyphenol that exerts a variety of pharmacological activities and plays an anti-cancer role in many cancer cells. It was recently reported that gasdermin E (GSDME) is involved in the progression of pyroptosis. Materials and methods HepG2 cells were treated with various concentrations of curcumin and cell viability was examined using MTT assay, apoptosis was analysed using flow cytometry, reactive oxygen species (ROS) levels using dihydroethidium, LDH release using an LDH cytotoxicity assay, and protein expression using western blot. Results Curcumin increased the expression of the GSDME N-terminus and proteins involved in pyrolysis, promoted HspG2 cell pyrolysis and increased intracellular ROS levels. Moreover, inhibition of the production of intracellular ROS with n-acetylcysteine (NAC) improved the degree of apoptosis and pyrolysis induced by curcumin. Conclusion Curcumin induces HspG2 cell death by increasing apoptosis and pyroptosis, and ROS play a key role in this process. This study improves our understanding of the potential anti-cancer properties of curcumin in liver cancer.

Many large and super-large copper deposits have been discovered and explored in the Tibet Plateau, which makes it the most important copper resource reserve and development base in China. Based on the work of the research team, the paper summarizes the geological characteristics of the main copper deposits in Tibet and puts forward a further prospecting direction. A series of large accumulated metal deposits or ore districts from subduction of Tethys oceanic crust to India-Asia collisionhave been discovered, such as Duolong Cu (Au) ore district and Jiama copper polymetallic deposit. The ore deposits in the Duolong ore district are located in the lowstand domain, the top of lowstand domain, and the highstand domain of the same magmatic-hydrothermal metallogenic system, and their relative positions are the indicators for related deposits in the Bangong Co-Nujiang metallogenic belt. The polycentric metallogenic model of the Jiama copper polymetallic deposit is an important inspiration for the exploration of the porphyry mineralization related to collision orogeny. Further mineral exploration in the Tibet Plateau should be focused on the continental volcanic rocks related to porphyry-epithermal deposits, orogenic gold deposits, hydrothermal Pb-Zn deposits related to nappe structures, skarn Cu (Au) and polymetallic deposits, and the Miocene W-Sn polymetallic deposits. ©2021 China Geology Editorial Office.

Background As the third most abundant element, aluminum is widespread in the environment. Previous studies have shown that aluminum has a neurotoxic effect and its exposure can impair neuronal development and cognitive function. Aim To study the effects of aluminum on epigenetic modification in neural stem cells and neurons. Methods Neural stem cells were isolated from the forebrain of adult mice. Neurons were isolated from the hippocampi tissues of embryonic day 16-18 mice. AlCl3 at 100 and 200 μmol/L was applied to stem cells and neurons. Results Aluminum altered the differentiation of adult neural stem cells and caused apoptosis of newborn neurons while having no significant effects on the proliferation of neural stem cells. Aluminum application also significantly inhibited the dendritic development of hippocampal neurons. Mechanistically, aluminum exposure significantly affected the levels of DNA 5-hydroxy-methylcytosine, 5-methylcytosine, and N6-methyladenine in stem cells and neurons. Conclusion Our findings indicate that aluminum may regulate neuronal development by modulating DNA modifications.

Nasopharyngeal carcinoma (NPC) is a unique head and neck cancer with highly aggressive and metastatic potential in which distant metastasis is the main reason for treatment failure. Till present, the underlying molecular mechanisms of NPC metastasis remains poorly understood. Here, we identified S100 calcium-binding protein A14 (S100A14) as a functional regulator suppressing NPC metastasis by inhibiting the NF-kB signaling pathway and reversing the epithelial-mesenchymal transition (EMT). S100A14 was found to be downregulated in highly metastatic NPC cells and tissues. Immunohistochemical staining of 202 NPC samples revealed that lower S100A14 expression was significantly correlated with shorter patient overall survival (OS) and distant metastasis-free survival (DMFS). S100A14 was also found as an independent prognostic factor for favorable survival. Gain- and loss-of-function studies confirmed that S100A14 suppressed the in vitro and in vivo motility of NPC cells. Mechanistically, S100A14 promoted the ubiquitin-proteasome-mediated degradation of interleukin-1 receptor-associated kinase 1 (IRAK1) to suppress NPC cellular migration. Moreover, S100A14 and IRAK1 established a feedback loop that could be disrupted by the IRAK1 inhibitor T2457. Overall, our findings showed that the S100A14-IRAK1 feedback loop could be a promising therapeutic target for NPC metastasis.

The variation of water level is the main environmental factor controlling the growth of aquatic vegetation. It is of significance to understand its influences on aquatic vegetation coverage in sub-lakes under different hydrolo-gical control modes. Taking the free connected sub-lake Bang Lake and locally controlled sub-lake Dahuchi Lake of Poyang Lake as a case and based on remote sensing cloud computing platform of the Google Earth Engine (GEE), we used the pixel binary model to estimate aquatic vegetation coverage from 2000 to 2019, and analyzed the temporal and spatial differentiation characteristics, and the variation trend was simulated by combining the method of Sen+M-K. We analyzed the water level change characteristics during the study period and the relationship between the hydrological parameters and the aquatic vegetation coverage area of sub-lakes with different hydrological connectivity was explored by setting up the water level fluctuation parameters. The results showed that the aquatic vegetation coverage of Bang Lake was more susceptible to water level changes, while Dahuchi Lake was more stable. The aquatic vegetation was patchily and sporadically distributed in the years with low vegetation coverage. In the years with high vegetation coverage, it was distributed in a ring-like pattern, spreading from the center of the lake to the shore. The aquatic vegetation coverage of Bang Lake was more likely influenced by water level fluctuation rate, while the aquatic vegetation coverage of Dahuchi Lake was more likely influenced by the flooding duration of 17 m characteristic water level. The flooding duration of 19 m characteristic water level had a strong negative correlation with the aquatic vegetation coverage of Bang Lake and Dahuchi Lake. The trend of aquatic vegetation in Bang Lake was dominated by stabilization and slight improvement, while that in Dahuchi Lake was dominated by stabilization and significant degradation. Our results could help to further understand the dynamics of water hydrological ecosystem with different hydrological connectivity and provide a reference for lake management and conservation.水位变化是影响水生植被生长的主控环境因子,研究其对不同水文管控模式子湖的水生植被覆盖度的影响具有重要的现实意义。本研究基于谷歌地球引擎(GEE)遥感云计算平台,以鄱阳湖自由连通子湖蚌湖、局部控制子湖大湖池为研究对象,采用像元二分法估算2000—2019年间水生植被生长期的植被覆盖度并分析其时空分异特征,运用Sen+M-K相结合的方法对其变化趋势进行模拟分析,构建水位波动参数集,分析研究期间水位变化特征,探讨不同水文连通性子湖各水文参数与水生植被覆盖面积的关系。结果表明: 蚌湖的水生植被覆盖度更易受水位变化的影响,大湖池更稳定。在植被覆盖度较低的年份,水生植被呈斑状零星分布,较高的年份则呈环带状分布,且植被覆盖度由湖心至湖岸逐渐升高。蚌湖的水生植被覆盖度更易受水位波动率的影响,而大湖池的水生植被覆盖度更易受17 m特征水位水淹时长的影响。19 m特征水位水淹时长与蚌湖和大湖池的水生植被覆盖度均有很强的负相关性。蚌湖的水生植被变化趋势以稳定不变或轻微改善为主,而大湖池的水生植被变化趋势以稳定不变或显著退化为主。本研究有助于进一步了解不同水文连通性湖泊的水文生态系统动力学的变化及其对生态系统结构和功能的影响,可为湖泊的管理和保护提供参考。.
