Twinning plays a prominent role in the plastic deformation of low-symmetry crystals such as magnesium alloys. However, the capture of twin nucleation has been challenging since the process is rapid and difficult to predict, making it impossible to observe directly. Because of this, the understanding of the fundamental twinning mechanism through experiments and simulation techniques incorporating twinning has progressed slowly. In this study, a quantitative analysis integrating an in-situ three-point bending experiment and crystal plasticity modeling was conducted on the twin nucleation in AZ31 magnesium alloys with different grain sizes. The threshold for triggering twin nucleation has been quantified. The prediction ability of three widely used criteria for twin nucleation, i.e., geometrically necessary dislocation (GND) density, von Mises stress, and stored energy density, is evaluated. The results suggest that only the stored energy density is able to capture the twin nucleation location accurately in all the considered grain sizes. The critical value exhibits an exponential reduction with the increase in grain size. Based on the quantitative analysis, the mechanism of the grain size effect on twin nucleation is proposed.

Reconstructing critical-sized craniofacial bone defects is a global healthcare challenge. Current methods, like autologous bone transplantation, face limitations. Bone tissue engineering offers an alternative to autologous bone, with traditional approaches focusing on stimulating osteogenesis via the intramembranous ossification (IMO) pathway. However, IMO falls short in addressing larger defects, particularly in clinical scenarios where there is insufficient vascularisation. This review explores redirecting bone regeneration through endochondral ossification (ECO), a process observed in long bone healing stimulated by hypoxic conditions. Despite its promise, gaps exist in applying ECO to bone tissue engineering experiments, requiring the elucidation of key aspects such as cell sources, biomaterials and priming protocols. This review discusses various scaffold biomaterials and cellular sources for chondrogenesis and hypertrophic chondrocyte priming, mirroring the ECO pathway. The review highlights challenges in current endochondral priming and proposes alternative approaches. Emphasis is on segmental mandibular defect repair, offering insights for future research and clinical application. This concise review aims to advance bone tissue engineering by addressing critical gaps in ECO strategies.

The deformation behavior of hot-roll-bonded Mg/Al laminated metal composites (LMCs) is investigated, focusing on the crack formation around the interface. The material demonstrates a positive strain rate sensitivity mainly arising from the Mg matrix. A diffusion layer was formed at the Mg/Al interface with a serrated morphology during the two-pass rolling process, which consists of the Al3Mg2 and Mg17Al12 phases. The mechanical responses of the LMCs are evaluated using uniaxial tension, interfacial debonding tests (in normal and shear modes), and microstructure characterization techniques, which clarify the interfacial heterogeneity. The fracture constants of the interface were calibrated and incorporated in the finite element models to predict the failure modes of heterogeneous Mg/Al interface under three-point bending. With the increased thickness ratio of the Mg layer, the damage accumulation rate decreases at the interface and increases in the Mg matrix. As a result, the failure mode transferred from interface failure to matrix fracture. The findings of the present work provide new insight into the failure mechanism in laminated metal composites under bending conditions and can provide theoretical guidance for the plastic forming of such materials.

Abstract Aim Autophagy plays essential roles in abdominal aortic aneurysm (AAA) development and progression. The objective of this study was to verify the autophagy-related genes (ARGs) underlying AAA empirically and using bioinformatics analysis. Methods Two gene expression profile datasets GSE98278 and GSE57691 were downloaded from the Gene Expression Omnibus (GEO) database, and principal component analysis was performed. Following, the R software (version 4.0.0) was employed to analyze potentially differentially expressed genes related with AAA and autophagy. Subsequently, the candidate genes were screened using protein–protein interaction (PPI), gene ontology (GO) enrichment analysis, and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. Finally, quantitative real-time polymerase chain reaction (RT-qPCR) was performed to detect the RNA expression levels of the top five selected abnormal ARGs in clinical samples obtained from the normal and AAA patients. Results According to the information contained (97 AAA patients and 10 healthy controls) in the two datasets, a total of 44 differentially expressed autophagy-related genes (6 up-regulated genes and 38 down-regulated genes) were screened. GO enrichment analysis of differentially expressed autophagy-related genes (DEARGs) demonstrated that some enrichment items were associated with inflammation, and PPI analysis indicated interaction between these genes. RT-qPCR results presented that the expression levels of IL6, PPARG, SOD1, and MAP1LC3B were in accordance with the bioinformatics prediction results acquired from the mRNA chip. Conclusion Bioinformatics analysis identified 44 potential autophagy-related differentially expressed genes in AAA. Further verification by RT- qPCR presented that IL6, PPARG, SOD1, and MAP1LC3B may affect the development of AAA by regulating autophagy. These findings might help explain the pathogenesis of AAA and be helpful in its diagnosis and treatment.

Autologous bone replacement remains the preferred treatment for segmental defects of the mandible; however, it cannot replicate complex facial geometry and causes donor site morbidity. Bone tissue engineering has the potential to overcome these limitations. Various commercially available calcium phosphate-based bone substitutes (Novabone®, BioOss®, and Zengro®) are commonly used in dentistry for small bone defects around teeth and implants. However, their role in ectopic bone formation, which can later be applied as vascularized graft in a bone defect, is yet to be explored. Here, we compare the above-mentioned bone substitutes with autologous bone with the aim of selecting one for future studies of segmental mandibular repair. Six female sheep, aged 7–8 years, were implanted with 40 mm long four-chambered polyether ether ketone (PEEK) bioreactors prepared using additive manufacturing followed by plasma immersion ion implantation (PIII) to improve hydrophilicity and bioactivity. Each bioreactor was wrapped with vascularized scapular periosteum and the chambers were filled with autologous bone graft, Novabone®, BioOss®, and Zengro®, respectively. The bioreactors were implanted within a subscapular muscle pocket for either 8 weeks (two sheep), 10 weeks (two sheep), or 12 weeks (two sheep), after which they were removed and assessed by microCT and routine histology. Moderate bone formation was observed in autologous bone grafts, while low bone formation was observed in the BioOss® and Zengro® chambers. No bone formation was observed in the Novabone® chambers. Although the BioOss® and Zengro® chambers contained relatively small amounts of bone, endochondral ossification and retained hydroxyapatite suggest their potential in new bone formation in an ectopic site if a consistent supply of progenitor cells and/or growth factors can be ensured over a longer duration.

The periosteum is a thin layer of connective tissue covering bone. It is an essential component for bone development and fracture healing. There has been considerable research exploring the application of the periosteum in bone regeneration since the 19th century. An increasing number of studies are focusing on periosteal progenitor cells found within the periosteum and the use of hydrogels as scaffold materials for periosteum engineering and guided bone development. Here, we provide an overview of the research investigating the use of the periosteum for bone repair, with consideration given to the anatomy and function of the periosteum, the importance of the cambium layer, the culture of periosteal progenitor cells, periosteum-induced ossification, periosteal perfusion, periosteum engineering, scaffold vascularization, and hydrogel-based synthetic periostea.

Periosteum is a highly vascularized membrane lining the surface of bones. It plays essential roles in bone repair following injury and reconstruction following invasive surgeries. To broaden the use of periosteum, including for augmenting in vitro bone engineering and/or in vivo bone repair, we have developed an ex vivo perfusion bioreactor system to maintain the cellular viability and metabolism of surgically resected periosteal flaps. Each specimen was placed in a 3D printed bioreactor connected to a peristaltic pump designed for the optimal flow rates of tissue perfusate. Nutrients and oxygen were perfused via the periosteal arteries to mimic physiological conditions. Biochemical assays and histological staining indicate component cell viability after perfusion for almost 4 weeks. Our work provides the proof-of-concept of ex vivo periosteum perfusion for long-term tissue preservation, paving the way for innovative bone engineering approaches that use autotransplanted periosteum to enhance in vivo bone repair.

The initial texture effect of strain rate sensitivity response in magnesium alloys was studied using the quasi in situ electron backscatter diffraction technique. The strain accumulations, under uniaxial loading of both tensile and compression along the rolling direction, showed strain rate dependency. For the AZ31 magnesium sheets with strong basal texture (max intensity to 11.2), twinning first occurs at the onset of plastic deformation, resulting in macroscopic strain rate insensitivity. With further loading, the strain rate sensitivity significantly increases (with the strain rate sensitivity coefficient m increased from 0.0025 to 0.012), arising from the initiation of slip. For the WE43 alloy sheets with weak basal texture (max intensity to 2.6), the overall deformation is sensitive to strain rate under both tensile and compression. The twinning growth rate and twinned volume fraction have been used to identify whether the process is twin-dominated or slip-dominated during the plastic deformation of a magnesium alloy.

Abdominal aortic aneurysm (AAA) is a common and serious disease with a high mortality rate, but its genetic determinants have not been fully identified. In this feasibility study, we aimed to elucidate the transcriptome profile of AAA and further reveal its molecular mechanisms through the Oxford Nanopore Technologies (ONT) MinION platform. Overall, 9574 novel transcripts and 781 genes were identified by comparing and analysing the redundant-removed transcripts of all samples with known reference genome annotations. We characterized the alternative splicing, alternative polyadenylation events and simple sequence repeat (SSR) loci information based on full-length transcriptome data, which would help us further understand the genome annotation and gene structure of AAA. Moreover, we proved that ONT methods were suitable for the identification of lncRNAs via identifying the comprehensive expression profile of lncRNAs in AAA. The results of differentially expressed transcript (DET) analysis showed that a total of 7044 transcripts were differentially expressed, of which 4278 were upregulated and 2766 were downregulated among two groups. In the KEGG analysis, 4071 annotated DETs were involved in human diseases, organismal systems and environmental information processing. These pilot findings might provide novel insights into the pathogenesis of AAA and provide new ideas for the optimization of personalized treatment of AAA, which is worthy of further study in subsequent studies.

Free flap surgery is currently the only successful method used by surgeons to reconstruct critical-sized defects of the jaw, and is commonly used in patients who have had bony lesions excised due to oral cancer, trauma, infection or necrosis. However, donor site morbidity remains a significant flaw of this strategy. Various biomaterials have been under investigation in search of a suitable alternative for segmental mandibular defect reconstruction. Hydrogels are group of biomaterials that have shown their potential in various tissue engineering applications, including bone regeneration, both through in vitro and in vivo pre-clinical animal trials. This review discusses different types of hydrogels, their fabrication techniques, 3D printing, their potential for bone regeneration, outcomes, and the limitations of various hydrogels in preclinical models for bone tissue engineering. This review also proposes a modified technique utilizing the potential of hydrogels combined with scaffolds and cells for efficient reconstruction of mandibular segmental defects.

This brief review attempts to summarize research advances in the mechanical toughness and structures of double-network (DN) hydrogels. The focus is to provide a critical and concise discussion on the toughening mechanisms, damage recoverability, stress relaxation, and biomedical applications of tough DN hydrogel systems. Both conventional DN hydrogel with two covalently cross-linked networks and novel DN systems consisting of physical and reversible cross-links are discussed and compared. Covalently cross-linked hydrogels are tough but damage-irreversible. Physically cross-linked hydrogels are damage-recoverable but exhibit mechanical instability, as reflected by stress relaxation tests. This remains one significant challenge to be addressed by future research studies to realize the load-sustaining applications proposed for tough hydrogels. With their special structure and superior mechanical properties, DN hydrogels have great potential for biomedical applications, and many DN systems are now fabricated with 3D printing techniques.

Breast cancer is the most common and biggest health threat for women. There is an urgent need to develop novel breast cancer therapies to overcome the shortcomings of conventional surgery and chemotherapy, which include poor drug efficiency, damage to normal tissues, and increased side effects. Drug delivery systems based on injectable hydrogels have recently gained remarkable attention, as they offer encouraging solutions for localized, targeted, and controlled drug release to the tumor site. Such systems have great potential for improving drug efficiency and reducing the side effects caused by long-term exposure to chemotherapy. The present review aims to provide a critical analysis of the latest developments in the application of drug delivery systems using stimuli-responsive injectable hydrogels for breast cancer treatment. The focus is on discussing how such hydrogel systems enhance treatment efficacy and incorporate multiple breast cancer therapies into one system, in response to multiple stimuli, including temperature, pH, photo-, magnetic field, and glutathione. The present work also features a brief outline of the recent progress in the use of tough hydrogels. As the breast undergoes significant physical stress and movement during sporting and daily activities, it is important for drug delivery hydrogels to have sufficient mechanical toughness to maintain structural integrity for a desired period of time.

Background: Persistent sciatic artery is a rare embryological vascular anomaly, with an incidence of 0.01%–0.05%. Up to 60% of persistent sciatic artery patients will develop aneurysms that can subsequently lead to distal embolization and a high risk of limb loss. Method: Here we report a case with acute limb ischemia caused by thrombus in a right persistent sciatic artery aneurysm. The patient underwent endovascular treatment by deploying a 10 × 150 mm stent graft (Viabahn) in the persistent sciatic artery and two self-expanding bare stents (10 × 40 mm, 10 × 60 mm, SMART) in the stent graft to reinforce the radial resistive force. In addition, we conducted a literature review of articles published in PubMed from 2001 to 2018 regarding stent graft repair of complete persistent sciatic artery aneurysms. A total of 13 articles reported 13 patients with complete persistent sciatic artery aneurysms who underwent endovascular stent graft repair. Result: A favorable result was obtained for this patient, and computed tomographic angiography at 6 months of follow-up revealed patent stent graft. Most articles reported favorable results. Conclusion: Favorable results can be achieved with endovascular stent graft repair and anticoagulation therapy for complete persistent sciatic artery aneurysms.

Coral bleaching events have been predicted to occur more frequently in the coming decades with global warming. The susceptibility of corals to bleaching during thermal stress episodes is dependent on many factors and an understanding of these underlying drivers is crucial for conservation management. In 2013, a mild bleaching episode ensued in response to elevated sea temperature on the sediment-burdened reefs in Singapore. Surveys of seven sites highlighted variable bleaching susceptibility among coral genera-Pachyseris and Podabacia were the most impacted (31% of colonies of both genera bleached). The most susceptible genera such as Acropora and Pocillopora, which were expected to bleach, did not. Susceptibility varied between less than 6% and more than 11% of the corals bleached, at four and three sites respectively. Analysis of four of the most bleached genera revealed that a statistical model that included a combination of the factors (genus, colony size and site) provided a better explanation of the observed bleaching patterns than any single factor alone. This underscored the complexity in predicting the coral susceptibility to future thermal stress events and the importance of monitoring coral bleaching episodes to facilitate more effective management of coral reefs under climate change.

For example, with millions of datayou will need million set of keys. To manage and keep track of these keys,complexity and operational inefficiency becomes an issue. How to manage keysbecomes a challenging task. Adequate understanding of these new challenges isessential to effectively devise new key management policies and mechanisms toguard against them. We discuss many of these methods in this new context tofill this gap
