Abstract During the filling process of LNG tank trucks, due to the long-term operation of filling equipment in low temperature and high-pressure conditions, the sealing parts in the equipment are prone to failure, leading to leaks. The reasons for the leakage of LNG filling equipment were analyzed, and the diffusion of LNG after different equivalent leakage hole diameters and different wind speeds were numerically analyzed. A gas leak monitoring system suitable for LNG filling stations was established based on TDLAS technology. The analysis results show that when the equivalent leakage hole diameter is greater than 4 mm, the height of natural gas diffusion will exceed 5 m within 2 s without wind; the concentration of natural gas above the leakage point is inversely proportional to the height, and the decrease in concentration at a height of 5 m is between 50% and 60% compared with a height of 0.5 m; the wind will bend the natural gas diffusion shape downwind, and the degree of bending decreases as the equivalent leakage hole diameter increases; the optimal laser incidence angle of the laser gas sensor is between 30° and 50°, and the theoretical effective monitoring area width of the sensor when the installation height is 5 m is 4.46 m within the optimal laser incidence angle range.

ObjectiveTo analyze the relationship between sleep and myopia in primary and secondary school students with different durations of outdoor activities in Beijing, aiming to provide an evidence for myopia prevention and control in the students. MethodsFrom September to December 2021, 24 326 students recruited using stratified random cluster sampling from 16 districts in Beijing municipality underwent visual acuity testing and an on-site anonymous online questionnaire survey. A multivariate logistic regression model was used to analyze the relationship between sleep duration and myopia in students with different durations of outdoor activities. ResultsOf the 22 957 valid participants, 71.6% were identified as having myopia, 61.0% reported insufficient time spent outdoors, and 70.0% reported insufficient sleep. The prevalence of myopia was higher in participants with insufficient sleep than in those with sufficient sleep (73.6% vs. 67.2%, χ2 = 96.970; P < 0.001). Multivariate logistic regression analysis showed that after adjustment for gender, urban/rural residence, education level, parental myopia, daily after-school study time, daily TV viewing time, daily screen viewing time, and whether they turn off the light when viewing screens after dark, among participants who reported insufficient time spent outdoors, those with insufficient sleep had an increased risk of myopia (odds ratio = 1.10, 95% confidence interval: 1.03 – 1.18; P < 0.05) compared to those with sufficient sleep; subsequent gender stratification analysis showed that the risk of myopia was increased in male and all participants, but not in female participants. ConclusionAmong students with inadequate outdoor activity, the prevalence of myopia was higher among those with inadequate sleep, suggesting that parents and teachers should encourage students to participate in more outdoor activity and ensure adequate sleep to reduce the risk of myopia.

Abstract Background Wu et al. introduced a modified radiographic system that allows classification of all forms of CTD with excellent interobserver and intraobserver reliability. No study to date has evaluated the radiographic characteristics of Wu et al. type C3 CTD with osseous attachment at the level of the metacarpal. Objective This study aimed to evaluate the radiographic features of type C3 CTD according to the system of Wu et al., to describe the different anatomical subtypes of the duplication, and to propose a categorization approach to distinguish diverse surgical strategies based on the radiographic anatomy of this specific subtype of duplication. Methods We performed a retrospective analysis of 215 patients (221 thumbs) diagnosed with Wu et al. type C3 CTD at our Institution between 2015 and 2021. We evaluated all CTDs by examining the alignment of the interphalangeal (IP) and metacarpophalangeal (MP) joints and by assessing the presence of abnormal hypertrophic epiphysis of the primary thumb on posteroanterior (PA) radiographs. The proposed classification system has four types: Type I with good alignment of both MP and IP joints, Type II with ulnar deviation of the MP joint, Type III with radial deviation in the MP joint and Type IV with abnormal hypertrophic epiphysis of the distal phalanx of the main thumb with ulnar deviation of the IP joint with or without ulnar deviation of the MP joint. Results There were 140 male and 75 female patients with CTD (221 thumbs). There were 65 left, 144 right and 6 bilateral forms. The right-to-left, male-to-female and unilateral-to-bilateral ratios were 2.2:1, 1.9:1 and 35.8:1 respectively. The mean age at surgery was 22.3 ± 11.8 months (range, 8–80). The proposed classification system allowed the classification of all CTDs (n = 221). Specifically, 53 fingers were classified as Type I (24%), 136 as Type II (61.5%), 21 as Type III (9.5%), and 11 as Type IV (5%). Conclusion The proposed system is based on radiographic pathoanatomy and complements that of Wu et al. by identifying four distinct subtypes of deformity. It has the potential to improve inter-professional communication and guide surgery in patients with Wu et al. type C3 CTD. However, our results are preliminary and further research is needed to validate them. Level of evidence III.

Abstract Background This work aims to explore circ_ROBO1’s function in nasopharyngeal carcinoma (NPC). Methods circ_ROBO1 expression in NPC tissues and cell lines was measured. The regulation of circ_ROBO1 and/or miR-324-3p on the proliferation, migration, invasion, and apoptosis of NPC cells was investigated by functional experiments. The interplay between circ_ROBO1, miR-324-3p, and NME1 was explored. Tumor growth and metastasis were studied in mice. Results circ_ROBO1 was overexpressed in NPC. Knockdown of circ_ROBO1 repressed proliferation, migration, and invasion and induced apoptosis of NPC cells. Loss of circ_ROBO1 reduced tumor growth and metastasis in mice. circ_ROBO1 competed with miR-324-3p to upregulate NME1. Lowering miR-324-3p expression impaired the effect of knockdown of circ_ROBO1on NPC cells. Conclusion Overexpressed circ_ROBO1 promotes NPC development by modifying the miR-324-3p/NME1 axis.

IntroductionThe increasing global demand for starch has created an urgent need to identify more efficient and sustainable production methods. However, traditional starch sources, such as crop-based options, experience significant bottlenecks due to limitations in land use, water consumption, and the impacts of climate change. Therefore, there is a pressing need to explore and develop new sources of starch.MethodsWe develop a novel duckweed cultivation technology that combines nutrients limitation and CO2 supplementation to achieve very high starch content. In this study, we integrated whole-genome sequencing, epigenomics, transcriptomics, enzyme activity, and composition variation to elucidate the mechanisms of efficient starch accumulation in duckweed in terms of starch accumulation and carbon partitioning, regulation of the expression of genes in the starch metabolic pathway, and sucrose biosynthesis and transportation.Results and discussionAlthough Landoltia punctata exhibits dramatic gene family contraction, its starch content and productivity reached 72.2% (dry basis) and 10.4 g m-2 d-1, respectively, in 10 days, equivalent to a yield of 38.0 t ha-1 y-1, under nutrient limitation treatment with elevated CO2 levels. We also examined the mechanism of high starch accumulation in duckweed. This phenomenon is associated with the regulation of DNA methylation and transcription factors as well as the significantly upregulated transcription levels and the increased activities of key enzymes involved in starch biosynthesis. Moreover, while nitrogen redistribution was increased, sucrose biosynthesis and transportation and lignocellulose biosynthesis were reduced. These alterations led to a reduction in lignocellulose and protein contents and ultimately an increase in the accumulation of starch in the chloroplasts.ConclusionThis work demonstrates the potential of duckweed as a highly efficient starch producer.

Background Intracranial atherosclerotic stenosis is a leading cause of ischemic stroke and recurrent events due to plaque instability. High‐resolution magnetic resonance imaging identifies plaque enhancement as a key marker of instability. This study evaluated the efficacy of combined high‐intensity statins and proprotein convertase subtilisin/kexin type 9 inhibitors in plaque stabilization. Methods In this prospective, single‐arm study, patients with acute stroke and intracranial atherosclerotic stroke in the M1 segment of the middle cerebral artery or basilar artery were enrolled. After 24 weeks of intensive statin and evolocumab therapy, high‐resolution magnetic resonance imaging assessments of intracranial vessels were conducted at baseline and follow‐up. The primary end point was the change in stenosis degree; secondary end points included changes in plaque enhancement (contrast ratio) and plaque burden. ObjectiveTo investigate the current status of family structure and its relationship with health-related behaviors among primary and secondary school students in Beijing, and to provide a basis for promoting health-related behaviors among these students. MethodsUsing a stratified cluster random sampling method, a questionnaire survey was conducted among 26 403 primary school, junior high school, senior high school, and vocational high school students from 16 districts in Beijing from September 2022 to February 2023. Chi-square test and multivariate logistic regression analysis were used to analyze the correlation between health-related behaviors and cohabitants among primary and secondary school students. ResultsThe rate of consuming sugar-sweetened beverages ≥ 1 time per day was 7.5% (1 955/26 403) among primary and secondary school students in Beijing; the rate of consuming ≥ 3 kinds of vegetables per day was 44.2% (11 614/26 403); the rate of outdoor activities ≥ 2 hours during the day was 35.0% (8 838/26 403); and the rate of using mobile electronic devices ≥ 2 hours per day was 42.0% (8 547/26 403). The rate of depressive tendency among middle school students was 17.9% (3 130/17 486). Living with parents was a protective factor for consuming sugar-sweetened beverages ≥ 1 time per day (OR = 0.80), while living with grandparents was a risk factor (OR = 1.26). Living with parents was a protective factor for consuming < 3 kinds of vegetables per day (OR = 0.76) and depressive tendency (OR = 0.70). Living with grandparents (OR = 0.93) and siblings (OR = 0.89) were protective factors for < 2 hours of outdoor activity time per day. Living with parents was a protective factor for the rate of using mobile electronic products ≥ 2 hours per day (OR = 0.72), while living with grandparents (OR = 1.16) and siblings (OR = 1.12) were risk factors. ConclusionLiving with parents is the most important factor affecting health behaviors, and it is associated with most health-related behaviors and has a positive effect on living with parents. Living with grandparents and siblings may increase the risk of using electronic products, but it also increases outdoor activity time. In health education, interventions should be focused according to the different characteristics of cohabitants.

The administration of T cells that have been modified to carry chimeric antigen receptors (CARs) aimed at B cells has been an effective strategy in treating B cell malignancies. This breakthrough has spurred the creation of CAR T cells intended to specifically reduce or alter the faulty immune responses associated with autoimmune disorders. Early positive outcomes from clinical trials involving CAR T cells that target the B cell protein CD19 in patients suffering from autoimmune diseases driven by B cells have been reported. Additional strategies are being developed to broaden the use of CAR T cell therapy and enhance its safety in autoimmune conditions. These include employing chimeric autoantireceptors (CAAR) to specifically eliminate B cells that are reactive to autoantigens, and using regulatory T cells (Tregs) engineered to carry antigen-specific CARs for precise immune modulation. This discussion emphasizes key factors such as choosing the right target cell groups, designing CAR constructs, defining tolerable side effects, and achieving a lasting immune modification, all of which are critical for safely integrating CAR T cell therapy in treating autoimmune diseases.

Abstract Copper plays vital roles in numerous cellular processes and its imbalance can lead to oxidative stress and dysfunction. Recent research has unveiled a unique form of copper-induced cell death, termed cuproptosis, which differs from known cell death mechanisms. This process involves the interaction of copper with lipoylated tricarboxylic acid cycle enzymes, causing protein aggregation and cell death. Recently, a growing number of studies have explored the link between cuproptosis and cancer development. This review comprehensively examines the systemic and cellular metabolism of copper, including tumor-related signaling pathways influenced by copper. It delves into the discovery and mechanisms of cuproptosis and its connection to various cancers. Additionally, the review suggests potential cancer treatments using copper ionophores that induce cuproptosis, in combination with small molecule drugs, for precision therapy in specific cancer types.

Abstract Many plants are facultatively asexual, balancing short-term benefits with long-term costs of asexuality. During range expansion, natural selection likely influences the genetic controls of asexuality in these organisms. However, evidence of natural selection driving asexuality is limited, and the evolutionary consequences of asexuality on the genomic and epigenomic diversity remain controversial. We analyzed population genomes and epigenomes of Spirodela polyrhiza, (L.) Schleid., a facultatively asexual plant that flowers rarely, revealing remarkably low genomic diversity and DNA methylation levels. Within species, demographic history and the frequency of asexual reproduction jointly determined intra-specific variations of genomic diversity and DNA methylation levels. Genome-wide scans revealed that genes associated with stress adaptations, flowering and embryogenesis were under positive selection. These data are consistent with the hypothesize that natural selection can shape the evolution of asexuality during habitat expansions, which alters genomic and epigenomic diversity levels.

Objectives: Growing evidence suggests a link between organophosphate insecticides and depression disorder. These chemicals are metabolized and subsequently expelled through the urinary tract. The present study aims to investigate whether dialkyl phosphate metabolites associate with depression score and severity among the general population. Methods: This cross-sectional study used data from the National Health and Nutrition Examination Survey (NHANES). Depression was evaluated by the Patient Health Questionnaire-9 (PHQ-9). All urinary dialkyl phosphate metabolites were quantitatively analyzed. The survey’s complex design parameters and sampling weights were considered. Results: 3035 eligible individuals were included. The estimated prevalence of mild and major depression was 18.3% (95% confidence interval [CI]: 16.9–19.7%) and 9.9% (95% CI: 8.7–11.0%). For each incremental unit in the level of urinary dimethyl phosphate (DMP), individuals were found to have a higher depression score of 0.77 and a significantly increased odds ratio (OR) of 1.13 (95% CI: 1.12–1.13) for mild depression and 2.75 (95% CI: 2.74–2.76) for major depression. Conclusions: Our findings indicate positive and independent associations between urinary dialkyl phosphate metabolites and an elevated risk of depression among the general population.

In today’s digital age, multi-energy systems (MES) have become an indispensable part of the social infrastructure, providing people with diversified energy support such as electricity, gas, water and so on. However, with the increasing popularity and networking of MES, the network security threats they face are becoming more and more serious, especially the threat of network attacks. This makes it essential to detect attacks on MES and precisely classify attack types in order to establish effective defense strategies. In this paper, a Dual-Head output network attack detection and classification method based on parallel CNN-BiLSTM network is proposed. The method adopts a parallel structure and can process different aspects of information at the same time, speeding up the training and inference process of the whole network, making the system respond more quickly to potential network attacks, and improving real-time and efficiency. The multi-model fusion structure can give full play to the advantages of CNN and BiLSTM in processing different types of data, so that the system can capture attack characteristics more comprehensively in many aspects, and improve the overall detection and classification performance. The dual-head output not only improves the system’s ability to accurately detect attacks, but also can effectively classify different types of attacks in detail, which helps to formulate more targeted defense strategies. In addition, in order to effectively evaluate our proposed method, the network traffic data required for the experiment were collected in an environment very similar to the actual operating environment of a multi-energy system. Finally, the experiment verifies that our method can not only realize effective detection of network attacks, but also accurately classify different types of attacks.

Summary: Recent advances in cancer research have unveiled a significant yet previously underappreciated aspect of oncology: the presence and role of intratumoral microbiota. These microbial residents, encompassing bacteria, fungi, and viruses within tumor tissues, have been found to exert considerable influence on tumor development, progression, and the efficacy of therapeutic interventions. This review aims to synthesize these groundbreaking discoveries, providing an integrated overview of the identification, characterization, and functional roles of intratumoral microbiota in cancer biology. We focus on elucidating the complex interactions between these microorganisms and the tumor microenvironment, highlighting their potential as novel biomarkers and therapeutic targets. The purpose of this review is to offer a comprehensive understanding of the microbial dimension in cancer, paving the way for innovative approaches in cancer diagnosis and treatment.

Abstract Background Nosocomial infections caused by Serratia marcescens mostly occurred in pediatrics and it was very rarely reported after adult surgery. Here, an intracranial abscess caused by Serratia marcescens was reported. Case summary We report a rare case of a postoperative intracranial abscess caused by Serratia marcescens in a 63-year-old male patient with a left parietal mass. The patient underwent resection of the mass on June 1, 2022, and the postoperative pathology revealed an angiomatous meningioma, WHO I. He then experienced recurrent worsening of right limb movements, and repeated cranial CT scans showed oozing blood and obvious low-density shadows around the operation area. Delayed wound healing was considered. Subsequently, a large amount of pus was extracted from the wound. The etiological test showed that Serratia marcescens infection occurred before the removal of the artificial titanium mesh. Antibiotics were initiated based on the results of drug susceptibility tests. At present, the patient is recovering well and is still closely monitored during follow-up. Conclusion It is rare for Serratia marcescens to cause brain abscesses without any obvious signs of infection. This report provided in detail our experience of a warning postoperative asymptomatic brain abscess caused by an uncommon pathogen.

Traditional fermented foods could produce a variety of functional enzymes and plenty of flavor compounds through microbial fermentation. The production process of fermented foods would be constantly enriched with abundant enzymes, and it is the current hotspot to study these functional enzymes systematically and scientifically. In this study, the traditional isolation and culture of functional enzymes from fermented food are analyzed, and it can identify the enzyme activity of key functional enzymes. Furthermore, this work summarizes the recent research on functional enzymes in fermented foods by high-throughput technologies, such as PICRUSt analysis based on 16S rRNA and ITS sequencing, metagenome, metatranscriptome and metaproteome. It is found that, compared to analysis by traditional isolation and culture, single high-throughput technology can obtain more functional enzymes information, analyze complex metabolic pathways and identify active functional enzymes. The research on functional enzymes by combinations of high-throughput technologies is also reviewed, and it is found that combination of different methods can obtain more information to analyze the fermentation process of traditional food comprehensively and deeply. It is also found that, based on analysis of high-throughput technology, some researchers have analyzed the specific functional enzymes in traditional foods by heterologous expression. This study comprehensively summarizes results and methods of functional enzymes in various traditional fermented foods, aiming to provide technical foundation for the research and application of functional enzymes in traditional fermented foods.

CD24 is a glycosylphosphatidylinositol-anchored protein that is expressed in a wide range of tissues and cell types. It is involved in a variety of physiological and pathological processes, including cell adhesion, migration, differentiation, and apoptosis. Additionally, CD24 has been studied extensively in the context of cancer, where it has been found to play a role in tumor growth, invasion, and metastasis. In recent years, there has been growing interest in CD24 as a potential therapeutic target for cancer treatment. This review summarizes the current knowledge of CD24, including its structure, function, and its role in cancer. Finally, we provide insights into potential clinical application of CD24 and discuss possible approaches for the development of targeted cancer therapies.

Promoters are one of the most critical elements in regulating gene expression. They are considered essential biotechnological tools for heterologous protein production. The one most widely used in plants is the 35S promoter from cauliflower mosaic virus. However, our study for the first time discovered the 35S promoter reduced the expression of exogenous proteins under increased antibiotic stress. We discovered an endogenous strong promoter from duckweed named LpSUT2 that keeps higher initiation activity under antibiotic stress. Stable transformation in duckweed showed that the gene expression of eGFP in the LpSUT2:eGFP was 1.76 times that of the 35S:eGFP at 100 mg.L-1 G418 and 6.18 times at 500 mg.L-1 G418. Notably, with the increase of G418 concentration, the gene expression and the fluorescence signal of eGFP in the 35S:eGFP were weakened, while the LpSUT2:eGFP only changed slightly. This is because, under high antibiotic stress, the 35S promoter was methylated, leading to the gene silencing of the eGFP gene. Meanwhile, the LpSUT2 promoter was not methylated and maintained high activity. This is a previously unknown mechanism that provides us with new insights into screening more stable promoters that are less affected by environmental stress. These outcomes suggest that the LpSUT2 promoter has a high capacity to initiate the expression of exogenous proteins. In conclusion, our study provides a promoter tool with potential application for plant genetic engineering and also provides new insights into screening promoters.

Abstract Triple‐positive breast cancer (TPBC) poorly responds to current standard neoadjuvant therapy (trastuzumab plus pertuzumab and chemotherapy). Our previous MUKDEN 01 study showed a promising total pathological complete response (tpCR) rate of 30.4% with neoadjuvant pyrotinib (pan‐human epidermal growth factor receptor tyrosine kinase inhibitor) plus dalpiciclib (cyclin‐dependent kinase 4/6 inhibitor) and letrozole, but the efficacy remains suboptimal. This pilot study (NCT05228951) explored adding trastuzumab to this triplet neoadjuvant regimen in patients with stage II–III TPBC. The primary endpoint was tpCR (ypT0/is, ypN0) rate. Between February 2022 and June 2022, 12 patients were enrolled, and seven (58%; 95% confidence interval [CI], 27.7%–84.8%) patients achieved tpCR. The rate of residual cancer burden (RCB) 0–I was 75% (95% CI, 46.8%–91.1%). The objective response rate (ORR) was 92% (95% CI, 64.6%–98.5%). Mean Ki‐67 level was significantly reduced from 45.0% (95% CI, 19.5%–70.5%) at baseline to 17.2% (95% CI, 0.7%–33.7%) after neoadjuvant therapy (p = 0.03). The most common grade 3 adverse events were diarrhea (four [33%]) and decreased neutrophil count (three [25%]). No grade 4 adverse events or treatment‐related deaths occurred. This four‐drug neoadjuvant regimen shows promising pathological response with an acceptable safety profile in patients with TPBC. A randomized controlled trial (NCT05638594) of this regimen is being conducted.

Abstract Background Chinese Nong-favor daqu, the presentative liquor starter of Baijiu, has been enriched with huge amounts of enzymes in degrading various biological macromolecules by openly man-made process for thousand years. According to previous metatranscriptomics analysis, plenty of α-glucosidases were identified to be active in NF daqu and played the key role in degrading starch under solid-state fermentation. However, none of α-glucosidases was characterized from NF daqu, and their actual functions in NF daqu were still unknown. Results An α-glucosidase (NFAg31A, GH31-1 subfamily), the second highest expressed α-glucosidases in starch degradation of NF daqu, was directly obtained by heterologous expression in Escherichia coli BL21 (DE3). NFAg31A exhibited the highest sequence identities of 65.8% with α-glucosidase II from Chaetomium thermophilum, indicating its origin of fungal species, and it showed some similar features with homologous α-glucosidase IIs, i.e., optimal activity at pH ~ 7.0 and litter higher temperature of 45 ℃, well stability at 41.3 ℃ and a broad pH range of pH 6.0 to pH 10.0, and preference on hydrolyzing Glc-α1,3-Glc. Besides this preference, NFAg31A showed comparable activities on Glc-α1,2-Glc and Glc-α1,4-Glc, and low activity on Glc-α1,6-Glc, indicating its broad specificities on α-glycosidic substrates. Additionally, its activity was not stimulated by any of those detected metal ions and chemicals, and could be largely inhibited by glucose under solid-state fermentation. Most importantly, it exhibited competent and synergistic effects with two characterized α-amylases of NF daqu on hydrolyzing starch, i.e., all of them could efficiently degrade starch and malto-saccharides, two α-amylases showed advantage in degrading starch and long-chain malto-saccharides, and NFAg31A played the competent role with α-amylases in degrading short-chain malto-saccharides and the irreplaceable contribution in hydrolyzing maltose into glucose, thus alleviating the product inhibitions of α-amylases. Conclusions This study provides not only a suitable α-glucosidase in strengthening the quality of daqu, but also an efficient way to reveal roles of the complicated enzyme system in traditional solid-state fermentation. This study would further stimulate more enzyme mining from NF daqu, and promote their actual applications in solid-state fermentation of NF liquor brewing, as well as in other solid-state fermentation of starchy industry in the future.

Abstract Purpose The specific neurovascular compression (NVC) event responsible for the symptomatic manifestation of hemifacial spasm (HFS) remains difficult to assess accurately using magnetic resonance imaging (MRI). We aim to evaluate the MRI characteristics of HFS. Method We retrospectively included patients with HFS and divided them into a test group (n = 186) and a validation group (n = 28). The presence, severity, and offending vessel type of NVC in each portion, and the orientation of the offending vessel around the facial nerve, were recorded. Conditional logistic regression analyses were performed to evaluate correlations using test group. The validation group was used to verify whether our findings improved diagnostic performance. Results Deformity in the proximal cisternal segment was significantly correlated with HFS occurrence (odds ratio [OR]: 256.58, p = .002), whereas contact was not (p = .233). ObjectiveThis study aimed (i) to evaluate the radiographic characteristics of patients with congenital thumb duplication (CTD) type C2 according to the classification of Wu et al., (ii) to describe the various subtypes of type C2 CTD, and (iii) to propose a classification system that allows the identification of different surgical strategies based on the radiographic anatomy of this specific subtype of duplication.MethodsWe retrospectively reviewed 92 patients (92 thumbs) with type C2 CTD according to the Wu et al. classification in our institution between August 2015 and April 2021. All CTDs were classified according to the interphalangeal joint alignment of the main thumb at the posteroanterior radiograph of the thumb before operation: type I (no deviation), type II (ulnar deviation), and type III (radial deviation).ResultsAll CTDs (n = 92) could be classified according to the proposed classification system: 76 (82.6%) were type I, 10 (10.9%) were type II, and six were type III (6.5%). According to the Kim system of subtype classification, there were 55 (59.8%) type 1, 24 (26.1%) type 2, and 13 (14.1%) type 3 cases.ConclusionsThe suggested classification completes the Wu et al. system and has the potential to guide surgical treatment in children with type C2 CTD.Level of evidenceIII.

This review encapsulates the expanding spectrum of CD24 functions, delving into its involvement in immune regulation, cancer immune microenvironment, and its potential as a therapeutic target in autoimmune diseases and beyond. The ‘magic’ of CD24, once solely attributed to cancer, now inspires a new paradigm in understanding its multifunctionality in human health and disease, offering exciting prospects for medical advancements.

Glioma, as the most frequently occurring primary malignancy in the central nervous system, significantly impacts patients’ quality of life and cognitive abilities. Ferroptosis, a newly discovered form of cell death, is characterized by significant iron accumulation and lipid peroxidation. This process is fundamentally dependent on iron. Various factors inducing ferroptosis can either directly or indirectly influence glutathione peroxidase, leading to reduced antioxidant capabilities and an increase in lipid reactive oxygen species (ROS) within cells, culminating in oxidative cell death. Recent research indicates a strong connection between ferroptosis and a range of pathophysiological conditions, including tumors, neurological disorders, ischemia-reperfusion injuries, kidney damage, and hematological diseases. The regulation of ferroptosis to intervene in the progression of these diseases has emerged as a major area of interest in etiological research and therapy. However, the exact functional alterations and molecular mechanisms underlying ferroptosis remain to be extensively studied. The review firstly explores the intricate relationship between ferroptosis and glioma, highlighting how ferroptosis contributes to glioma pathogenesis and how glioma cells may resist this form of cell death. Then, we discuss recent studies that have identified potential ferroptosis inducers and inhibitors, which could serve as novel therapeutic strategies for glioma. We also examine the current challenges in targeting ferroptosis in glioma treatment, including the complexity of its regulation and the need for precise delivery methods. This review aims to provide a comprehensive overview of the current state of research on ferroptosis in glioma, offering insights into future therapeutic strategies and the broader implications of this novel cell death pathway in cancer biology.

Glioblastoma (GBM), one of the most aggressive forms of brain cancer, has limited treatment options. Recent years have witnessed the remarkable success of checkpoint inhibitor immunotherapy across various cancer types. Against this backdrop, several clinical trials investigating checkpoint inhibitors for GBM are underway in multiple countries. Furthermore, the integration of immunotherapy with traditional treatment approaches is now emerging as a highly promising strategy. This review summarizes the latest advancements in checkpoint inhibitor immunotherapy for GBM treatment. We provide a concise yet comprehensive overview of current GBM immunotherapy options. Additionally, this review underscores combination strategies and potential biomarkers for predicting response and resistance in GBM immunotherapies.

Brain-inspired navigation technologies combine environmental perception, spatial cognition, and target navigation to create a comprehensive navigation research system. Researchers have used various sensors to gather environmental data and enhance environmental perception using multimodal information fusion. In spatial cognition, a neural network model is used to simulate the navigation mechanism of the animal brain and to construct an environmental cognition map. However, existing models face challenges in achieving high navigation success rate and efficiency. In addition, the limited incorporation of navigation mechanisms borrowed from animal brains necessitates further exploration. On the basis of the brain-inspired navigation process, this paper launched a systematic study on brain-inspired environment perception, brain-inspired spatial cognition, and goal-based navigation in brain-inspired navigation, which provides a new classification of brain-inspired cognition and navigation techniques and a theoretical basis for subsequent experimental studies. In the future, brain-inspired navigation technology should learn from more perfect brain-inspired mechanisms to improve its generalization ability and be simultaneously applied to large-scale distributed intelligent body cluster navigation. The multidisciplinary nature of brain-inspired navigation technology presents challenges, and multidisciplinary scholars must cooperate to promote the development of this technology.

CD47 is a 50 kDa five-spanning membrane receptor that plays a crucial role in multiple cellular processes, including myeloid cell activation, neutrophils transmigration, vascular remodeling, leukocyte adhesion and trans-endothelial migration. Recent studies have revealed that CD47 is a highly expressed anti-phagocytic signal in several types of cancer, and therefore, blocking of CD47 has shown an effective therapeutic potential in cancer immunotherapy. In addition, CD47 has been found to be involved in a complex interplay with microglia and other types of cells, and increasing evidence indicates that CD47 can be targeted as part of immune modulatory strategies for non-neoplastic diseases as well. In this review, we focus on CD47 and its role in non-neoplastic diseases, including neurological disorders, atherosclerosis and autoimmune diseases. In addition, we discuss the major challenges and potential remedies associated with CD47-SIRPα-based immunotherapies.

For the calculation of transient fluid–solid coupling temperature field of transformer, the traditional Galerkin finite element full order model has the defect that the calculation time is up to hours-level, which is difficult to meet the timeliness requirements of the digital twin model. Firstly, the implementation architecture of transformer digital twin based on IoT is established, and the necessity of multiphysical field simulation and model reduction technology for building the digital twin model is introduced. Then, based on the method of transformer digital twin implementation,it is proposed to combine the proper orthogonal decomposition (POD) with the finite element (FE) to establish the Galerkin FE full order model for the governing equation of the transient temperature field, the reduced order mode composed of the first several bases is intercepted to build the reduced order model of transient fluid–solid coupling temperature field. Finally, the POD reduced order model is applied to calculate the temperature rise of a transformer. According to the actual transformer temperature rise test, the temperature values of different measuring points are selected to compare the calculation error and time of the reduced order model and the full order model. The results show that the error between the third order model and full order model meets the criterion of POD truncation error, and the calculation time is shortened by 192 times, which can meet the seconds-level calculation requirements.

For many years, the methods of cancer treatment are usually surgery, chemotherapy and radiation therapy. Although these methods help to improve the condition, most tumors still have a poor prognosis. In recent years, immunotherapy has great potential in tumor treatment. Chimeric antigen receptor T-cell immunotherapy (CAR-T) uses the patient's own T cells to express chimeric antigen receptors. Chimeric antigen receptor (CAR) recognizes tumor-associated antigens and kills tumor cells. CAR-T has achieved good results in the treatment of hematological tumors. In 2017, the FDA approved the first CAR-T for the treatment of B-cell acute lymphoblastic leukemia (ALL). In October of the same year, the FDA approved CAR-T to treat B-cell lymphoma. In order to improve and enhance the therapeutic effect, CAR-T has become a research focus in recent years. The structure of CAR, the targets of CAR-T treatment, adverse reactions and improvement measures during the treatment process are summarized. This review is an attempt to highlight recent and possibly forgotten findings of advances in chimeric antigen receptor T cell for treatment of hematological tumors.

Abstract Cytogenetically normal acute myeloid leukemia (CN-AML) is a heterogeneous disease with different prognoses. Researches on prognostic biomarkers and therapy targets of CN-AML are still ongoing. Instead of protein-coding genes, more and more researches were focused on the non-coding RNAs especially long non-coding RNAs (lncRNAs) which may play an important role in the development of AML. Although a large number of lncRNAs have been found, our knowledge of their functions and pathological process is still in its infancy. The purpose of this research is to identify the key lncRNAs and explore their functions in CN-AML by reconstructing the lncRNA–miRNA–mRNA network based on the competitive endogenous RNA (ceRNA) theory. We reconstructed a global triple network based on the ceRNA theory using the data from National Center for Biotechnology Information Gene Expression Omnibus and published literature. According to the topological algorithm, we identified the key lncRNAs which had both the higher node degrees and the higher numbers of lncRNA–miRNA pairs and total pairs in the ceRNA network. Meanwhile, Gene Ontology (GO) and pathway analysis were performed using databases such as DAVID, KOBAS and Cytoscape plug-in ClueGO respectively. The lncRNA–miRNA–mRNA network was composed of 90 lncRNAs,33mRNAs,26 miRNAs and 259 edges in the lncRNA upregulated group, and 18 lncRNAs,11 mRNAs,6 miRNAs and 45 edges in the lncRNA downregulated group. The functional assay showed that 53 pathways and 108 GO terms were enriched. Three lncRNAs (XIST, TUG1, GABPB1-AS1) could possibly be selected as key lncRNAs which may play an important role in the development of CN-AML. Particularly, GABPB1-AS1 was highly expressed in CN-AML by both bioinformatic analysis and experimental verification in AML cell line (THP-1) with quantitative real‐time polymerase chain reaction. In addition, GABPB1-AS1 was also negatively correlated with overall survival of AML patients. The lncRNA–miRNA–mRNA network revealed key lncRNAs and their functions in CN-AML. Particularly, lncRNA GABPB1-AS1 was firstly proposed in AML. We believe that GABPB1-AS1 is expected to become a candidate prognostic biomarker or a potential therapeutic target.

The generation of music lyrics by artificial intelligence (AI) is frequently modeled as a language-targeted sequence-to-sequence generation task. Formally, if we convert the melody into a word sequence, we can consider the lyrics generation task to be a machine translation task. Traditional machine translation tasks involve translating between cross-lingual word sequences, whereas music lyrics generation tasks involve translating between music and natural language word sequences. The theme or key words of the generated lyrics are usually limited to meet the needs of the users when they are generated. This requirement can be thought of as a restricted translation problem. In this paper, we propose a fuzzy training framework that allows a model to simultaneously support both unrestricted and restricted translation by adopting an additional auxiliary training process without constraining the decoding process. This maintains the benefits of restricted translation but greatly reduces the extra time overhead of constrained decoding, thus improving its practicality. The experimental results show that our framework is well suited to the Chinese lyrics generation and restricted machine translation tasks, and that it can also generate language sequence under the condition of given restricted words without training multiple models, thereby achieving the goal of green AI.

This study introduces the application of deep-learning technologies in automatically generating guidance for independent reading. The study explores and demonstrates how to incorporate the latest advances in deep-learning-based natural language processing technologies in the three reading stages, namely, the pre-reading stage, the while-reading stage, and the post-reading stage. As a result, the novel design and implementation of a prototype system based on deep learning technologies are presented. This system includes connections to prior knowledge with knowledge graphs and summary-based question generation, the breakdown of complex sentences with text simplification, and the auto-grading of readers' writing regarding their comprehension of the reading materials. Experiments on word sense disambiguation, named entity recognition and question generation with real-world materials in the prototype system show that the selected deep learning models on these tasks obtain favorable results, but there are still errors to be overcome before their direct usage in real-world applications. Based on the experiment results and the reported performance of the deep learning models on reading-related tasks, the study reveals the challenges and limitations of deep learning technologies, such as inadequate performance, domain transfer issues, and low explain ability, for future improvement.

Semantic role labeling (SRL) is dedicated to recognizing the semantic predicate-argument structure of a sentence. Previous studies in terms of traditional models have shown syntactic information can make remarkable contributions to SRL performance; however, the necessity of syntactic information was challenged by a few recent neural SRL studies that demonstrate impressive performance without syntactic backbones and suggest that syntax information becomes much less important for neural semantic role labeling, especially when paired with recent deep neural network and large-scale pre-trained language models. Despite this notion, the neural SRL field still lacks a systematic and full investigation on the relevance of syntactic information in SRL, for both dependency and both monolingual and multilingual settings. This paper intends to quantify the importance of syntactic information for neural SRL in the deep learning framework. We introduce three typical SRL frameworks (baselines), sequence-based, tree-based, and graph-based, which are accompanied by two categories of exploiting syntactic information: syntax pruning-based and syntax feature-based. Experiments are conducted on the CoNLL-2005, -2009, and -2012 benchmarks for all languages available, and results show that neural SRL models can still benefit from syntactic information under certain conditions. Furthermore, we show the quantitative significance of syntax to neural SRL models together with a thorough empirical survey using existing models.

The local endocytosis of membrane proteins is critical for many physiological processes in plants, including the regulation of growth, development, nutrient absorption, and osmotic stress response. Much of our knowledge on the local endocytosis of plasma membrane (PM) protein only focuses on the polar growth of pollen tubes in plants and neuronal axon in animals. However, the role of local endocytosis of PM proteins in guard cells has not yet been researched. Here, we first cloned duckweed SUT2 (sucrose transporter 2) protein and then conducted subcellular and histological localization of the protein. Our results indicated that LpSUT2 (Landoltia punctata 0202 SUT2) is a PM protein highly expressed on guard cells. In vitro experiments on WT (wild type) lines treated with high sucrose concentration showed that the content of ROS (reactive oxygen species) in guard cells increased and stomatal conductance decreased. We observed the same results in the lines after overexpression of the LpSUT2 gene with newfound local endocytosis of LpSUT2. The local endocytosis mainly showed that LpSUT2 was uniformly distributed on the PM of guard cells in the early stage of development, and was only distributed in the endomembrane of guard cells in the mature stage. Therefore, we found the phenomenon of guard cell LpSUT2 local endocytosis through the changes of duckweed stomata and concluded that LpSUT2 local endocytosis might be dependent on ROS accumulation in the development of duckweed guard cells. This paper might provide future references for the genetic improvement and water-use efficiency in other crops.

Novel selective enzymatic refining of sweet potato processing residues requires judicious enzyme selection and enzyme discovery. We prepared a pectinaceous cell wall polysaccharide fraction from sweet potato using an enzymatic a treatment to preserve the natural linkages and substitutions. Polysaccharide composition and linkage analysis data confirmed the pectinaceous polysaccharide fraction to be a rhamnogalacturonan I-rich fraction with a high content of arabinogalactan Type I. We hypothesized that the post-harvest tuber pathogenic fungus Penicillium sclerotigenum would harbor novel enzymes targeting selective sweet potato pectin modification. As part of the study, we also report the first genome sequence of P. sclerotigenum. We incubated the sweet potato pectinaceous fraction with P. sclerotigenum. Using proteomics accompanied by CUPP-bioinformatics analysis, we observed induced expression of 23 pectin-associated degradative enzymes. We also identified six abundantly secreted, induced proteins that do not correspond to known CAZymes, but which we suggest as novel enzymes involved in pectin degradation. For validation, the predicted CUPP grouping of putative CAZymes and the exo-proteome data obtained for P. sclerotigenum during growth on sweet potato pectin were compared with proteomics and transcriptomics data reported previously for pectin-associated CAZymes from Aspergillus niger strain NRRL3. The data infer that P. sclerotigenum has the capacity to express several novel enzymes that may provide novel opportunities for sweet potato pectin modification and valorization of sweet potato starch processing residues. In addition, the methodological approach employed represents an integrative systematic strategy for enzyme discovery.

Abstract Background Daqu is the most important fermentation starter for Chinese liquor, with large number of microbes and enzymes being openly enriched in the Daqu system over thousands of years. However, only a few enzymes have been analyzed with crude protein for total liquefying power and saccharifying power of Daqu. Therefore, the complex enzymatic system present in Daqu has not been completely characterized. Moreover, their pivotal and complicated functions in Daqu are completely unknown. Results In this study, a novel α-amylase NFAmy13B, from GH13_5 subfamily (according to the Carbohydrate-Active enZYmes Database, CAZy) was successfully heterologous expressed by Escherichia coli from Chinese Nong-flavor (NF) Daqu. It exhibited high stability ranging from pH 5.5 to 12.5, and higher specific activity, compared to other GH13_5 fungal α-amylases. Moreover, NFAmy13B did not show activity loss and retained 96% residual activity after pre-incubation at pH 11 for 21 h and pH 12 for 10 h, respectively. Additionally, 1.25 mM Ca2+ significantly improved its thermostability. NFAmy13B showed a synergistic effect on degrading wheat starch with NFAmy13A (GH13_1), another α-amylase from Daqu. Both enzymes could cleave maltotetraose and maltopentaose in same degradation pattern, and only NFAmy13A could efficiently degrade maltotriose. Moreover, NFAmy13B showed higher catalytic efficiency on long-chain starch, while NFAmy13A had higher catalytic efficiency on short-chain maltooligosaccharides. Their different catalytic efficiencies on starch and maltooligosaccharides may be caused by their discrepant substrate-binding region. Conclusions This study mined a novel GH13_5 fungal α-amylase (NFAmy13B) with outstanding alkali resistance from Nong-flavor (NF) Daqu. Furthermore, its synergistic effect with NFAmy13A (GH13_1) on hydrolyzing wheat starch was confirmed, and their possible contribution in NF Daqu was also speculated. Thus, we not only provide a candidate α-amylase for industry, but also a useful strategy for further studying the interactions in the complex enzyme system of Daqu.

CD47 is ubiquitously expressed on the surface of cells and plays a critical role in self-recognition. By interacting with SIRPα, TSP-1 and integrins, CD47 modulates cellular phagocytosis by macrophages, determines life span of individual erythrocytes, regulates activation of immune cells, and manipulates synaptic pruning during neuronal development. As such, CD47 has recently be regarded as one of novel innate checkpoint receptor targets for cancer immunotherapy. In this review, we will discuss increasing awareness about the diverse functions of CD47 and its role in immune system homeostasis. Then, we will discuss its potential therapeutic roles against cancer and outlines, the possible future research directions of CD47- based therapeutics against cancer.

The purpose is to guarantee the security of fifth generation mobile communication technology (5G) network in power Internet of Things environment and improve the ability of wireless network communication to resist attacks. First, in terms of attack prevention, the 5G network security structure is proposed to replace the plaintext information commonly used in the original system with Ciphertext based on software defined network (SDN), thereby alleviating the security risks of the data dimension. Second, concerning attack detection, the signal is identified by using the imperfections and differences of equipment manufacturing based on the above security structure, preventing the attacker from further harming the sensitive data leaked. Researchers found that the SDN-based 5G network attack prevention scheme avoids the centralized exposure of sensitive data, improves security, reduces computational overhead, and simplifies encryption logic. Without affecting the bandwidth, the existing 5G network system is greatly prevented from attacks. The detection mechanism is not limited by the low-dimensional feature space, and it demonstrates strong robustness and stability. It can effectively detect the attacks on 5G network system in power Internet of Things (IoT). This study provides an important reference for the protection of 5G communication network attacks under IoT.

Simplified Molecular Input Line Entry System (SMILES) provides a text-based encoding method to describe the structure of chemical species and formulize general chemical reactions. Considering that chemical reactions have been represented in a language form, we present a symbol only model to generally predict the yield of organic synthesis reaction without considering complex quantum physical modeling or chemistry knowledge. Our model is the first deep neural network application that treats chemical reaction text segments as embedding representation to the most recent deep natural language processing. Experimental results show our model can effectively predict chemical reactions, which achieves a high accuracy of 99.76% on practicality judgment and the Root Mean Square Error (RMSE) is around 0.2 for yield prediction. Our work shows the great potential for automatic yield prediction for organic reactions under general conditions and further applications in synthesis path prediction with the least modeling cost.

Downstream of the sluice gate or weir, wave-type flows inevitably occur in stilling basins with no tailwater. This paper aims to investigate the hydraulic performance of wave-type flows at a sill-controlled stilling basin through experimental research. The flow pattern, bottom pressure profiles along the stilling basin, and the air concentrations on the bottom and the sidewall were examined in five sill-controlled stilling basins by altering the sill position and the height. The results show that wave-type flow patterns contain submerged and non-submerged jumps, which are relevant to ambient pressure head and air entrainment. The bottom pressure profiles are related to larger pressure fluctuations at large unit discharges and two peak pressure values in the vicinity of the sill. The air concentrations on the bottom and the sidewall decrease with the increasing unit discharge. The flow zone in the vicinity of the sill should be focused upon concerning protection against cavitation damage because of the slight air entrainment and significant pressure fluctuations. These findings advance our understanding of wave-type flows, and their ambient pressure heads and air entrainment are useful for designing the sill-controlled stilling basin in hydraulic engineering.

In order to stabilize the output fluctuation of wind power generation, this paper applies control strategy to control the action of the battery energy storage system according to the stability target, and quickly makes up for the system demand. In this paper, the optimal scheduling of new energy power system with energy storage is studied. This model predictive control is introduced into a certain system and optimized by model predictive control strategy so that the combination can match the predetermined expectations. Then, the model predictive control strategy is used to improve the schedulability of the wind farm. The objective function is to minimize the amount of action, and the composite output can track the scheduling instructions. Considering the cost, the way of storage and operation cost is optimized, and the penalty cost of wind abandonment is also reduced. Through rolling optimization and feedback correction, the model predictive control is optimized and solved, and finally, the action instructions of the storage system are obtained so that the storage output can effectively calm the wind. Power makes the combination schedulable. The optimized wind power/energy storage output is equivalent to the benign and dispatchable power supply of the power grid.

Phosphate-solubilizing bacteria (PSB) can alleviate available phosphorus (AP)-deficiency without causing environmental pollution like chemical phosphate fertilizers. However, the research and application of PSB on the barren rocky soil is very rare. We screened six PSB from sweetpotato rhizosphere rocky soil. Among them, Ochrobactrum haematophilum FP12 showed the highest P-solubilizing ability of 1,085.00 mg/L at 7 days, which was higher than that of the most reported PSB. The assembled genome of PSB FP12 was 4.92 Mb with P-solubilizing and plant growth-promoting genes. In an AP-deficient environment, according to transcriptome and metabolomics analysis, PSB FP12 upregulated genes involved in gluconic acid synthesis and the tricarboxylic acid cycle, and increased the concentration of gluconic acid and malic acid, which would result in the enhanced P-solubilizing ability. Moreover, a series of experiments in the laboratory and field confirmed the efficient role of the screened PSB on significantly increasing AP in the barren rocky soil and promoting sweetpotato yield. So, in this study, we screened highly efficient PSB, especially suitable for the barren rocky soil, and explored the P-solubilizing mechanism. The research will reduce the demand for chemical phosphate fertilizers and promote the environment-friendly agricultural development.

With the rapid development of railways towards high speed and larger carrying capacity, the problem of wear and fatigue damage between wheel/rail is gradually becoming serious. However, traditional pearlite wheel/rail has reached the limit, which leads to more attention to developing a novel wheel/rail material. This study aims to report a novel carbide-free bainite wheel steel. The wear-resistance of novel steel was tested by a rolling-sliding wear experiment under heavy-haul condition and investigated the impacts of the running speeds on the damage mechanism of wear and fatigue. The results show that the yield strength of the bainite wheel was as high as 950 MPa and the hardness was 415 HV, which was superior to most of the reported typical wheel steel. During the process of wear, the surface damage of the wheel was mainly adhesive wear and fatigue damage, and the gradient strain layer (GS layer) was formed on the wheel surface. As the running speed increased, fatigue damage gradually became more serious than adhesive wear, and the shear stress and strain of the GS layer were enhanced. The higher thickness and hardening were produced on the GS layer, which is the main reason for the higher wear-resistance of the bainitic wheel under higher running speeds. In addition, the wear-resistance of the novel wheel steel was better than that of the reported wheel steel. This novel bainitic wheel is a promising wheel for heavy-haul condition applications, which could provide a guide in choosing bainitic wheel steel for the railway.

Molecular farming utilizes plants as a platform for producing recombinant biopharmaceuticals. Duckweed, the smallest and fastest growing aquatic plant, is a promising candidate for molecular farming. However, the efficiency of current transformation methods is generally not high in duckweed. Here, we developed a fast and efficient transformation procedure in Lemna minor ZH0403, requiring 7–8 weeks from screening calluses to transgenic plants with a stable transformation efficiency of 88% at the DNA level and 86% at the protein level. We then used this transformation system to produce chicken interleukin-17B (chIL-17B). The plant-produced chIL-17B activated the NF-κB pathway, JAK-STAT pathway, and their downstream cytokines in DF-1 cells. Furthermore, we administrated chIL-17B transgenic duckweed orally as an immunoadjuvant with mucosal vaccine against infectious bronchitis virus (IBV) in chickens. Both IBV-specific antibody titer and the concentration of secretory immunoglobulin A (sIgA) were significantly higher in the group fed with chIL-17B transgenic plant. This indicates that the duckweed-produced chIL-17B enhanced the humoral and mucosal immune responses. Moreover, chickens fed with chIL-17B transgenic plant demonstrated the lowest viral loads in different tissues among all groups. Our work suggests that cytokines are a promising adjuvant for mucosal vaccination through the oral route. Our work also demonstrates the potential of duckweed in molecular farming.

Frequency stability is an important factor for the safety and stability of the power system operation. In a traditional power system, the operation stability is ensured by the inertia response, primary frequency modulation, and secondary frequency modulation. In recent years, in order to achieve the goal of carbon neutralization and carbon peaking, China has made great efforts in new energy development. With large-scale new energy connected to the power grid, the proportion of traditional conventional synchronous units has gradually declined. At the same time, a large number of power electronic devices have been used in the power grid, which led to the capability decline of the inertia response and primary frequency modulation. For example, the East China Power Grid has experienced a sharp frequency drop in such an environment. In order to solve the above problems, the operation principle and control mode of various new energy resources are analyzed in this paper. Moreover, the process and principle of power grid frequency response are studied and the evaluation index of frequency response capability is proposed. The research results can quantitatively evaluate the system inertia response and primary frequency modulation level and provides a judgment tool for dispatching operators and system planners.

Iron is an essential trace mineral element in almost all living cells and organisms. However, cellular iron metabolism pathways are disturbed in most cancer cell types. Cancer cells have a high demand of iron. To maintain rapid growth and proliferation, cancer cells absorb large amounts of iron by altering expression of iron metabolism related proteins. However, iron can catalyze the production of reactive oxygen species (ROS) through Fenton reaction. Nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is an important player in the resistance to oxidative damage by inducing the transcription of antioxidant genes. Aberrant activation of Nrf2 is observed in most cancer cell types. It has been revealed that the over-activation of Nrf2 promotes cell proliferation, suppresses cell apoptosis, enhances the self-renewal capability of cancer stem cells, and even increases the chemoresistance and radioresistance of cancer cells. Recently, several genes involving cellular iron homeostasis are identified under the control of Nrf2. Since cancer cells require amounts of iron and Nrf2 plays pivotal roles in oxidative defense and iron metabolism, it is highly probable that Nrf2 is a potential modulator orchestrating iron homeostasis and redox balance in cancer cells. In this hypothesis, we summarize the recent findings of the role of iron and Nrf2 in cancer cells and demonstrate how Nrf2 balances the oxidative stress induced by iron through regulating antioxidant enzymes and iron metabolism. This hypothesis provides new insights into the role of Nrf2 in cancer progression. Since ferroptosis is dependent on lipid peroxide and iron accumulation, Nrf2 inhibition may dramatically increase sensitivity to ferroptosis. The combination of Nrf2 inhibitors with ferroptosis inducers may exert greater efficacy on cancer therapy.

Coded caching is a promising method for solving caching problems in content-centric wireless networks. To enhance the security of coded caching for practical purposes, this paper investigates a secure coded caching scheme for defending against an eavesdropper who may possess prior knowledge before eavesdropping on content delivery. A novel key-based solution is proposed to minimize the leakage of meaningful information to the eavesdropper during the delivery phase. In the proposed solution, the central server observes the strength of the eavesdropper and introduces several keys to dynamically encrypt the broadcast signals within a weak security constraint. The amount of cache memory occupied by the keys is carefully designed to achieve the highest possible cache gain in terms of the rate of content delivery. The tradeoff between the cache memory usage and the transmission rate with varying eavesdropper strength is characterized. For a given size of the total cache memory, the tradeoff between the amounts of memory used for data and key storage is also presented. We report simulation results to support our analytical findings and show that no keys are needed to achieve weak security against a low-strength eavesdropper. Even for an eavesdropper of high strength, weak security can be introduced at a negligible cost given a large cache size.

Accurate malignancy level classification of lung nodules can reduce lung cancer mortality rate effectively. In this study, we present a fast and efficient CAD system to improve the performance of nodules malignancy level classification. Firstly, to reduce false positives (FPs), we propose a novel vessel segmentation method which measures vessel likelihood by tubular-like structures discriminating from multiple views. The method can recognize irregular vascular structures robustly and sensitively, and achieve fast vessel segmentation. In addition, a mathematical description for 3D pulmonary entities using neighbor centroids clustering is provided as a fundamental condition for spatial feature extraction. To optimize features extraction, we formulate a gray values cumulative function and a patches selection function based on the mathematical description, to generate axial spatial outline and spatial density distribution samples of the entities, respectively. Then, we use Edge Orientation Histogram (EOH) to extract edge features from the spatial outline and propose a multi-scale path LBP (MSPLBP) to extract the texture feature of the density distribution samples. Finally, the fused EOH and MSPLBP are classified into 6 malignancy levels by three state-of-the-art classifiers. The experimental results show that the vessel segmentation method achieves an average F1_Score of 78.14% and AUC value under PR curves of 0.8149. Moreover, our system reaches an average accuracy of 95.88% and consumes average 176.26 seconds for evaluating a CT set on malignancy level classification. These results indicate that the system can segment vessels exactly, and classify the malignancy level of nodules efficiently. Our system is the potential to be a powerful tool for early diagnosis of lung cancer.

Background: In industrial yeasts, selection and breeding for resistance to multiple stresses is a focus of current research. The objective of this study was to investigate the tolerance to multiple stresses of Saccharomyces cerevisiae obtained through an adaptive laboratory evolution strategy involving a repeated liquid nitrogen freeze–thaw process coupled with multi-stress shock selection. We also assessed the related resistance mechanisms and very high-gravity (VHG) bioethanol production of this strain. Results: Elite S. cerevisiae strain YF10-5, exhibiting improved VHG fermentation capacity and stress resistance to osmotic pressure and ethanol, was isolated following ten consecutive rounds of liquid nitrogen freeze–thaw treatment followed by plate screening under osmotic and ethanol stress. The ethanol yield of YF10-5 was 16% higher than that of the parent strain during 35% (w/v) glucose fermentation. Furthermore, there was upregulation of three genes (HSP26, HSP30, and HSP104) encoding heat-shock proteins involved in the stress response, one gene (TPS1) involved in the synthesis of trehalose, and three genes (ADH1, HXK1, and PFK1) involved in ethanol metabolism and intracellular trehalose accumulation in YF10-5 yeast cells, indicating increased stress tolerance and fermentative capacity. YF10-5 also showed excellent fermentation performance during the simultaneous saccharification and fermentation of VHG sweet potato mash, producing 13.40% (w/v) ethanol, which corresponded to 93.95% of the theoretical ethanol yield. Conclusions: A multiple-stress-tolerant yeast clone was obtained using adaptive evolution by a freeze–thaw method coupled with stress shock selection.
