Coloring agents in foods and drinks have been popular for centuries. This study aims to analyze the presence of ten synthetic colors (namely, (allura red (E129), amaranth (E123), sunset yellow (E110), tetrazine (E102), fast green (E143), ponceau 4R (New Coccine) (E124), erythrosin B (E127), brilliant blue FCF (E133), brilliant black (E151) and carmoisine (E122))) in food and drink samples using ultra-high-performance liquid chromatography diode array detection (UHPLC-DAD). The present analytical method was carried out using Agilent Poroshell 120 HPH-C18 column, 3 × 100 mm, 2.7 µm, and a mobile phase consisting of 10 mM Na2HPO4, pH 7, mixed with methanol as a time-increment gradient solution until the time was 20 min, then decreased with time until the time was 26 min. The pH was set by orthophosphoric acid at 7 and 5 μL injection volume, 0.50 mL flow rate, and the elution systems were monitored at 428 nm for E102, 518 nm for E124, E110, E129, E122, 530 nm for E151, E127, 622 nm for E143, and E133, respectively. The limit of detection and quantification for all colors ranged from 0.017 to 0.025 and 0.057 and 0.082 mg L−1, respectively. The correlation coefficient values ranged between 0.9991 and 1.0. The selectivity of the assay revealed no interference from other components in the analyzed samples. The percent recovery and precision (intra- and inter-day) of the spiked samples were within the acceptable limits of the ICH guidelines. Five analytical parameters were employed, and the results showed a new, novel, and robust method according to ICH guidelines for analyzing these colors. While most of the investigated food and drinks fell within the accepted range, some fell outside. The current sample preparation and analytical methods are comprehensive and universal for extracting and measuring synthetic colors in various food and drink samples. [ABSTRACT FROM AUTHOR]

This study outlines the development of a cost-effective, extended-release febuxostat (FEB) tablet using activated charcoal as an adsorbent to enhance drug release. FEB, a BCS Class II drug, presents formulation challenges due to low solubility and high lipophilicity. We evaluated eight formulations with varying FEB-to-charcoal ratios using FTIR and DSC for physical interactions and followed USP standards for overall assessment. The optimal 1:0.25 FEB-to-charcoal ratio demonstrated a consistent 12 h zero-order release pattern. In vivo studies indicated a significantly extended plasma profile compared to immediate-release tablets. The optimal tablets demonstrated acceptable hardness and disintegration times. This innovative approach enhances patient compliance, improves bioavailability, and reduces production costs, offering a promising solution for controlled FEB delivery. [ABSTRACT FROM AUTHOR]

Cancer nanomedicines, characterized by submicrometer-sized formulations, aim to optimize the biodistribution of anticancer drugs by minimizing off-target effects, reducing toxicity, enhancing target site accumulation, and improving overall efficacy. Numerous nanomedicines have been developed to improve the effectiveness and safety of traditional anticancer treatments. These include formulations with carbon nanotubes, nanodiamonds, enzyme-responsive nanoparticles for controlled drug release, dendrimers as nanoparticle drug carriers, quantum dot nanocarrier systems for precise drug delivery, solid lipid nanoparticles, and polymeric nanoparticles designed for targeted drug delivery. Additionally, nanotechnology has been explored in cancer treatment through gene therapy. Despite these advances, the complex nature of carrier materials and functional integration presents challenges in preparing these candidates for clinical translation. Nanotechnology, with its unique features at the nanoscale, offers novel possibilities for developing cancer therapies while increasing efficacy and safety. Although only a few nanotherapeutics have obtained clinical approval, exciting uses for nanotechnology are on the horizon. Nanoparticles possess unique transport, biological, optical, magnetic, electrical, and thermal capabilities due to their small size within the light wavelength spectrum. This results in high surface area-to-volume ratios, allowing for the incorporation of various supporting components in addition to active medicinal substances. These properties aid in solubilization, degradation protection, delayed release, immune response evasion, tissue penetration, imaging, targeted distribution, and triggered activation. In summary, the future of nanomedicine holds promise for introducing innovative platforms in cancer treatment. N-nitrosamine pollutants are probable carcinogens. Regulatory agencies declared their presence in the drugs unsafe for human consumption and demanded their recall. Using ultra-performance liquid chromatography-atmospheric pressure chemical ionization-tandem mass spectrometry (UPLC-APCI-MS/MS) in tablet dosage form based on International Conference on Harmonization (ICH) tripartite guideline criteria, we aim to develop and test a new approach for identifying and validating nitrosamine-contaminants, N -nitrosodimethylamine (NDMA) and N -nitrosodiethylamine (NDEA) in irbesartan, olmesartan and metformin. The column was Phenomenex Luna-C18, 100 × 3.0 mm and 3.0 μm. A mobile gradient phase of formic acid in either water or methanol separated the impurities. NDMA and NDEA had retention times of 0.85 and 2.55 min, respectively. The detector's linearity was established at concentrations ranging from 0.6 to 100 ng/mL. R2 for NDMA and NDEA were 0.9996 and 0.9998, respectively, with a linear response function established at 0.6–100 ng/mL. Limit of detection and limit of quantification for NDMA and NDEA were 0.35, 0.29 and 0.55, 0.37 ng/mL, respectively. On average, recovery rates for NDMA and NDEA ranged from 96.0 to 98.4 and 96.2 to 98.0%, respectively. The relative standard deviation for NDMA and NDEA was 3.46 and 2.69, respectively. According to the ICH guidelines, the developed method was quick, sensitive and valid. Type 2 diabetes mellitus is a multifactorial disorder whose primary manifestation usually initiates with elevated blood sugar levels. Several antidiabetic agents are used to manage type 2 diabetes mellitus, of which empagliflozin is an oral sodium-glucose co-transporter (SGLT-2) inhibitor in the kidney. This research aims to develop and validate a simple analytical method for determining empagliflozin levels in biological fluid and to further evaluate grapefruit juice's impact on empagliflozin pharmacokinetics in rats. High-Performance Liquid Chromatography (HPLC) was used to establish a simple, rapid, and accurate method for determining empagliflozin levels in rat plasma, in the presence of grapefruit juice. Four groups of rats (n = 10 rats in each) were used in the preclinical study. Group A (healthy rats) received empagliflozin alone; Group B (healthy rats) received empagliflozin with grapefruit; Group C (diabetic rats) received empagliflozin with grapefruit; and Group D (healthy, negative control) received no medication. The rats (n = 10) were given grapefruit juice instead of water for seven days before receiving the empagliflozin dose (0.16 mg/kg). Some pharmacokinetic parameters for each group were determined. The maximum plasma concentration (Cmax) and area under the curve (AUC) of empagliflozin in Group A without grapefruit intake were 730 ng/mL and 9264.6 ng × h/mL, respectively, with Tmax (2 h). In Group B, Cmax was 1907 ng/mL and AUC was 10,290.75 ng × h/mL in the presence of grapefruit, with Tmax (1 h); whereas, in Group C, the Cmax was 2936 ng/mL and AUC was 18657 ng × h/mL, with Tmax (2 h). In conclusion, our results showed that the co-administration of grapefruit with empagliflozin should be cautiously monitored and avoided, in which grapefruit elevates the plasma level of empagliflozin. This study aimed to develop and verify a simple HPLC-based quantitative approach to simultaneously determine the phosphodiesterase-5 inhibitors (PDE5Is) sildenafil, vardenafil, udenafil, avanafil, and tadalafil in a tablet dosage form mixed with honey obtained form Jordanian market in rat plasma. PDE5Is block phosphodiesterase-5 (PDE-5). This blockage, in turn, triggers vasodilation by phosphorylating downstream effector molecules. Chromatographic separation was performed on a HypersilTM C18 column (150 mm × 4.6 mm, 5 µm, Thermo Fisher Inc., Waltham, MA, USA). An acetonitrile:10% Triethylamine solution (57:43) at pH 5.5 (adjusted with orthophosphoric acid), 20 µL injection volume, 1 mL/min flow rate, 25 °C temperature, and eluent monitoring at 250 nm was used to execute the current approach. Linearity was observed in the 9.6–14.4 µg/mL concentration ranges for sildenafil, udenafil, avanafil, and tadalafil, and 2.4–3.6 µg/mL for vardenafil. Each dosage form was recovered within acceptable limits at three distinct concentrations, and the assay selectivity indicated no interference from the inactive substances in the formulation. Sildenafil, vardenafil, udenafil, avanafil, and tadalafil had retention times of 3.5, 4.3, 6.2, 9.7, and 12.8 min, respectively, and tadalafil was 12.8 min. The present analytical method is comprehensive and universal for measuring the five drugs. Such an analytical method can be routinely used to detect the combination of these drugs. Objective: The message delivered to the public regarding vitamin D is unclear, and contradictory; particularly regarding sun exposure; previous studies in Jordan have revealed high prevalence of low vitamin D among university students. The aim of this study was to investigate university students’ knowledge, attitudes and practice regarding vitamin D. Design: A web-based cross-sectional survey Setting: The survey was completed by students at a Jordanian university in 2019. Method: The survey collected sample characteristics data and information about university students’ knowledge, attitudes, and practice toward vitamin D. The χ² test was used to assess the associations between the sample characteristics and their vitamin D knowledge and practices and to evaluate the association between participants’ knowledge and practices. Binary logistic regression analysis was used to predict supplement use. Results: 496 students completed the questionnaire. The mean knowledge score was 31.3 (± 11.3) out of 100. Women had significantly higher beliefs that vitamin D deficiency is an escalating health issue (p<0.01). Negative practices regarding sun exposure were significantly higher in women than men (p<0.01). The majority of participants recognized that insufficient sun exposure was a cause of vitamin D deficiency, but 50.7 % avoid sun exposure, and 67.6% expose only their face and hands. The consumption of fortified foods (OR 3.59; p<0.001) was the only studied variable associated with vitamin D consumption. Conclusion: There is a gap between knowledge, attitude, and practice regarding vitamin D which can be bridged by promoting Vitamin D related awareness. [ABSTRACT FROM AUTHOR]

Background and Aim: Grapefruit is known in health care to interact with several medications. Interactions on the kinetic level are attributed to any change in pharmacokinetic parameters of the basic kinetic processes, Absorption, and Disposition when taking the drug concurrently with grapefruit juice. This study aims to develop an analytical method to measure Sitagliptin (an antidiabetic agent) in rat plasma alone and, in the presence of grapefruit juice, and evaluate grapefruit juice's impact on the fundamental pharmacokinetic parameters of Sitagliptin. Methods:A simpleHPLC method was developed using water-acetonitrile (pH 6.8) mobile phase and U.V. detector at 267 nm. According to the International Council for Harmonization of Technical Requirements for Pharmaceuticals for Human Use (ICH), linearity, accuracy, and precision were evaluated. Sitagliptin solution (5.75 mg/Kg) was assigned to two groups of rats, a control group and a fed grapefruit juice group. The plasmalevel vs. time profile was constructed for both groups for up to 24 hours. The pharmacokineticparameters were calculated for Sitagliptin with and without grapefruit juice using compartmental and non-compartmental analysis and statistically compared using t-test. Results: The analysis methodsuccessfully measuredSitagliptin in rat plasma showing high linearity and accepted accuracy and precision levelsaccording to the ICH guidelines. Results also showed an increase insitagliptinthat reached systemic circulation when given with grapefruit juice. There was no observed change in elimination parameters or the drug's total residence time in the body, suggesting an interaction on absorption level. Conclusion: Drug-food interaction is observed between sitagliptin and grapefruit, and hence, grapefruit juice consumption should be avoided with the concurrent use of sitagliptin. The aim of this study is assessing Healthcare Professionals (HCPs) knowledge on Chronic Kidney Disease (CKD) and inspecting their attitude regarding referral and perceptions towards statins use in non-dialysis CKD patients. A cross-sectional design was employed using a self-administered questionnaire that was constructed and validated before the study. The questionnaire was distributed to HCPs at two accredited hospitals. A total of 187 individuals including, 48.1% were pharmacists, 40.6% were physicians, and 11.2% were medical students. Female respondents slightly exceeded males, 56.7% vs. 43.3% respectively. Thirty-nine percent of study participants chose medical journals as their fundamental source for updated CKD information. More than 87% of respondents reported that the available CKD Continuing Medical Education (CME) programs are not sufficient. Almost 93% of participants appreciated the benefit of early referral of CKD patients to a nephrologist and 84.5% believed that non-dialysis CKD patients might benefit from using statins. The overall knowledge of participants was average with no significant differences in overall knowledge scores based on their age, gender, profession, experience or monthly salary. In conclusion, HCPs hold a positive attitude towards the early referral of CKD patients to nephrologists and appreciate the considerable value of statins use in improving CKD patients' outcomes.
