Abstract Solute transport family 7A member 7 (SLC7A7) mutations contribute to lysinuric protein intolerance (LPI), which is the mechanism of action that has been extensively studied. In colorectal cancer (CRC), SLC7A7 appears to play a role, but the features and mechanisms are not yet well understood. Survival was analyzed using the Kaplan–Meier analysis. Enrichment analysis was performed to characterize, immune infiltration, methylation, genetic instability, and crucial pathways of SLC7A7. Afterward, functional experiments were conducted in vitro to investigate how SLC7A7 affects tumor metastasis. Mechanistically, quantitative real-time PCR (qRT-PCR), western blot (WB), and methylated RNA immunoprecipitation (me-RIP) were carried out to confirm the methylation modification of SLC7A7 and related functions. High levels of expression of SLC7A7 are predictive of a worse prognosis for CRC patients. Enrichment analysis showed that SLC7A7 was significantly enriched during EMT and could be enriched in the Wnt/β-catenin signaling pathway, immune infiltration analysis of pan-cancer showed that SLC7A7 was significantly enriched in macrophages, and methylation analysis showed that SLC7A7 methylation modification affected the prognosis of specific cancers. SLC7A7 was indicated to promote the migration and invasion of CRC cells in in vitro functional experiments. Mechanistically, SLC7A7 was observed to potentially interact with the Wnt/β-catenin signaling pathway, possibly by influencing adenomatous polyposis coli (APC) expression. Furthermore, we identified that SLC7A7 undergoes N7-methylguanosine (m7G) modification, which may regulate SLC7A7 mRNA stability, with Quaking (QKI) potentially playing a role in this process by recognizing the m7G modification. Our results indicate that SLC7A7 may promote CRC metastasis through the SLC7A7/APC/Wnt/β-catenin signaling pathway. Moreover, m7G modification might be involved in regulating SLC7A7 mRNA stability, highlighting a novel layer of regulation.

Abstract Background Deciphering the in vivo processes of traditional Chinese medicine (TCM) is crucial for identifying new pharmacodynamic substances and new drugs. Due to the complexity and diversity of components, investigating the exposure, metabolism, and disposition remains a major challenge in TCM research. In recent years, a number of non-targeted smart mass-spectrometry (MS) techniques, such as precise-and-thorough background-subtraction (PATBS) and metabolomics, have realized the intelligent identification of in vivo components of TCM. However, the metabolites characterization still largely relies on manual identification in combination with online databases. Results We developed a scoring approach based on the structural similarity and minimal mass defect variations between metabolites and prototypes. The current method integrates three dimensions of mass spectral data including m/z, mass defect of MS1 and MS2, and the similarity of MS2 fragments, which was sequentially analyzed by a R-based mass dataset relevance bridging (MDRB) data post-processing technique. The MDRB technology constructed a component relationship network for TCM, significantly improving metabolite identification efficiency and facilitating the mapping of translational metabolic pathways. By combining MDRB with PATBS through this non-targeted identification technology, we developed a comprehensive strategy for identification, characterization and bridging analysis of TCM metabolite in vivo. As a proof of concept, we adopted the proposed strategy to investigate the process of exposure, metabolism, and disposition of Semen Armeniacae Amarum (CKXR) in mice. Significance The currently proposed analytical approach is universally applicable and demonstrates its effectiveness in analyzing complex components of TCMs in vitro and in vivo. Furthermore, it enables the correlation of in vitro and in vivo data, providing insights into the metabolic transformations among components sharing the same parent nucleus structure. Finally, the developed MDRB platform is publicly available for ( https://github.com/933ZhangDD/MDRB ) for accelerating TCM research for the scientific community. Graphical Abstract

Abstract Background The Oxidative Balance Score (OBS) was created to evaluate an individual’s overall antioxidant status. The objective of this study was to examine the association between OBS and abdominal aortic calcification (AAC) among individuals aged ≥ 40 years. Methods This population-based cross-sectional study used data from the National Health and Nutrition Examination Survey in 2013–2014 and included adults aged ≥ 40 years. Survey-weighted multivariable logistic and restricted cubic spline models were used to assess the association between OBS and AAC. Results Among 2520 participants, 744 were diagnosed with AAC (weighted percentage, 28.13%). Survey-weighted multivariable logistic revealed an inverse association between OBS and AAC [0.98 (0.96, 1.00)], and the nonlinear dose-response relationship was observed. Subgroup analysis and interaction tests revealed that this inverse relationship was consistent across different populations (all P for interaction > 0.05). Conclusions OBS was inversely associated with the prevalence of AAC among individuals aged ≥ 40 years. Maintaining a higher OBS may be beneficial in reducing the burden of AAC.

Abstract This study investigates global extreme precipitation events (EPEs) during warm seasons, with a particular focus on EPEs preceded by extreme heat stress (EPE-Hs) and a comparative analysis with those not (EPE-NHs). Using reanalysis product and Earth System Model data, the spatiotemporal characteristics and temperature sensitivities of EPEs are analyzed. Results show that EPE-Hs, while less frequent, have longer duration and greater magnitude compared to EPE-NHs, particularly in high latitude regions. In the future, a significant increase is projected in the characteristics of EPE-Hs, in contrast to the stable duration and magnitude of EPE-NHs. EPE-Hs demonstrate substantially higher temperature sensitivity than EPE-NHs, especially in low latitudes. The precipitation-temperature scaling relationships diverge markedly between EPE-Hs and EPE-NHs, with notable regional variations. These insights are pivotal for crafting region-specific early warning and adaptation strategies to mitigate the risks associated with extreme precipitation under the backdrop of global warming.

Study region: The Baihe River Basin located in the upper Han River, central China. Study focus: Potential evapotranspiration (PET) is commonly utilized as the forcing data for hydrological simulation. The lack of observations for estimating PET usually limits the calculation of evapotranspiration in hydrological models. Fortunately, gridded PET products with high data availability provide a great opportunity for hydrological simulation. This study assessed the accuracy of five gridded PET products against the gauge-based data. The Distributed Time Variant Gain Model (DTVGM) was modified to evaluate the performance of these five PET products in streamflow simulation. Additionally, the effect of different PET inputs on simulated runoff components, evapotranspiration, and soil moisture were investigated. New hydrological insights for the region: The results indicate that there are large discrepancies in spatial-temporal distribution of five PET products. Among selected products, GLEAM-b is most consistent with the gauge-based PET in the study region. For the streamflow simulation, five gridded PET products can achieve good performance with the Nash-Sutcliffe efficiency (NSE) values exceeding 0.75 in the validation period. This finding demonstrates the feasibility of using gridded PET products for streamflow simulation. Moreover, GLEAM-b performs better than other products in streamflow simulation according to the evaluation indicators. For simulated runoff components and water balance components, baseflow and evapotranspiration are more sensitive to different PET inputs.

To enhance the personal immunity to COVID-19, a third booster dose of inactivated COVID-19 vaccines program campaign was implemented in China. Our study endeavored to compare the dynamics of neutralizing antibodies generated by four distinct booster vaccines against three kinds of live SARS-CoV-2 virus (wild-type, Delta AY.23, and Omicron BA5.2). This cohort study involved 320 healthy individuals, who were randomly assigned to four groups, to receive boosters with inactivated vaccine (COVac and BIBP), the adenovirus type-5-vectored vaccine (Convidecia), and the recombinant protein-based vaccine (Zifivax), respectively, all the vaccines studied had the Wuhan variant as their parental variant. Participants were recruited from December 2021 to June 2022, with a follow-up period of 180 days. We evaluated humoral immune responses and their longevity by measuring the geometric mean titers (GMTs) of neutralizing antibodies against the SARS-CoV-2 virus at various time points post-boost. After 180 days of follow-up, 310 participants completed the study. Across all booster groups, neutralizing antibodies against the wild-type virus declined sharply within the first 90 days, accounting for an 81.24 to 92.34% reduction, then slowed down with gradually decreasing decay rates. By day 14 of post-boost, the ability to neutralize the Delta variant slightly diminished compared to the wild-type, whereas neutralizing antibodies against the Omicron variant exhibited a more pronounced decline, ranging from 10.78 to 19.88 times lower than those against the wild-type. Notably, heterologous boosting with the Convidecia vaccine maintained higher GMTs of neutralizing antibodies against both Delta and Omicron variants compared to the other boosters. At 180 days of post-boost, GMTs of neutralizing antibodies against SARS-CoV-2 had substantially decreased, yet individuals who received the Convidecia vaccine still exhibited higher titers than those who received other boosters. In summary, neutralizing antibody levels significantly waned 180 days after the third vaccine dose, with the most pronounced decline occurring within the initial 90 days. Heterologous boosting with Convidecia demonstrated a more robust, durable, and broad humoral immune response compared to boosting with inactivated vaccines or Zifivax, suggesting that adenovirus vector vaccines possess a special advantage in the realm of vaccine development for preventing infectious diseases.

IntroductionApproximately 15 million premature infants are born each year, many of whom face risks of neurological impairments. Accurate assessment of brain maturity is crucial for timely intervention and treatment planning. Electroencephalography (EEG) is a noninvasive method commonly used for this purpose. However, using all channels and features for brain maturity assessment can lead to high computational burden and overfitting, which can decrease the performance of the prediction system.MethodsIn this study, we propose an automatic prediction framework based on EEG to predict functional brain age (FBA) for assessing brain maturity in preterm infants. To optimize channel selection, we combine Binary Particle Swarm Optimization (BPSO) with Forward Addition (FA) and Backward Elimination (BE) methods. For feature selection, we combine the Pearson Correlation Coefficient (PCC), Recursive Feature Elimination (RFE), and Support Vector Regression (SVR) model.ResultsThe proposed framework achieved a prediction accuracy of 76.71% within ±1 week and 94.52% within ±2 weeks. Effective channel and feature selection significantly improved model performance while reducing computational costs.DiscussionThese results demonstrate that optimizing channel and feature selection can enhance the performance of FBA prediction in preterm infants, offering a more efficient and accurate tool for brain maturity assessment.

Abstract Background Hoylesella marshii can be isolated from human oral cavities affected by dental pulp and periodontal infections, as well as from the dental plaque of healthy individuals, making it a common bacterium within the oral microbiota. However, its role in causing pleural infections in humans is rare. Case presentation A case of purulent pleural effusion occurred shortly after discharge in an elderly patient who had undergone surgery for gastric cancer. The infection was identified as being caused by an obligate anaerobe through laboratory culture, and was further identified as Hoylesella marshii causing pleural infection through 16 S rRNA gene sequence analysis. Susceptibility testing guided precise treatment with cefoperazone-sulbactam and metronidazole. The patient’s clinical symptoms improved rapidly, laboratory test indicators gradually returned to normal, and the patient ultimately recovered. Conclusion Hoylesella marshii can cause pleural infections in humans. Clinical microbiology laboratories should pay special attention to the cultivation of obligate anaerobes when routine aerobic cultures do not show bacterial growth but bacteria are visible on smear staining, and when conventional identification methods fail to identify the bacterium, analysis based on the highly conserved 16 S rRNA gene sequence can accurately and specifically identify the bacterium, guiding clinicians in formulating precise anti-infection strategies.

Epithelial ovarian cancer (EOC) is a common estrogen-sensitive tumor that poses a serious threat to women 's health, and the mortality rate of EOC ranks first among malignant tumors in females. Studies have indicated a strong link between estrogen abnormality and EOC progression. We accidently found that 3-phosphoinositide-dependent protein kinase-1 (PDPK1) is highly expressed in EOC tissues. Further, estrogen also up-regulates the expression of PDPK1 in EOC cells. Notably, the expression of PDPK1 is controlled strictly, and its expression can determine the fate of cells. However, to date, the molecular mechanism by which estrogen elicits PDPK1 expression in EOC cells, and the role of PDPK1 in estrogen-driven EOC cells are not well defined. In this research, we found that a high expression of PDPK1 was associated with poor prognosis in patients with ovarian cancer. Further, estrogen stimulated the increase of PDPK1 protein expression through estrogen receptor ESR1. The depletion or overexpression of PDPK1 affected the inhibition or amplification of estrogen-driven EOC cell proliferation, and the knockdown of PDPK1 suppressed the migration of EOC cells by estrogen while promoting cell apoptosis. This suggests a critical functional association between estrogen and PDPK1 in the process of EOC. The expression of messenger RNA for cyclin A1, cyclin-dependent kinase 2 (CDK2), matrix metallopeptidase 2 (MMP2), and bcl-2 associated x protein (Bax) is regulated by PDPK1 under estrogen treatment. Our results indicated that PDPK1 plays a role as an oncogene in the development of EOC; hence, elucidating the mechanism by which estrogen promotes EOC progression by regulating PDPK1 expression.

At present, the role of active ingredients of traditional Chinese medicine in tumor therapy has gradually attracted people's attention, and anthraquinones, which are structurally similar to adriamycin and epirubicin, are one of the hotspots of research. Emodin (1,3,8-trihydroxy-6-methylanthraquinone) is a natural anthraquinone compound isolated from rhubarb, Polygonum cuspidatum, and aloe vera. In recent years, emodin has received widespread attention for its remarkable anti-tumor effects, and its anti-breast cancer effects are manifested as induction of apoptosis, inhibition of tumor cell proliferation, inhibition of invasion and metastasis of tumor cells, and anti-tumor drug resistance. Moreover, emodin can act against multiple types of breast cancer cells by acting on different targets. In this paper, we reviewed the latest research progress on the anti-breast cancer effects of emodin and its anti-tumor mechanism, to provide reference and information for the treatment of breast cancer and the development of anti-tumor drugs.

Abstract Background Ginger is a common aromatic vegetable with a wide range of functional ingredients and considerable medicinal and nutritional properties. Numerous studies have shown that ginger and its active ingredients have suppressive effects on manifold tumours, including ovarian cancer (OC). However, the molecular mechanism by which ginger inhibits OC is not clear. The aim of this study was to investigate the function and mechanism of ginger in OC. Methods The estimation of n6-methyladenosine (m6A) levels was performed using the m6A RNA Methylation Quantification Kit, and RT-qPCR was used to determine the expression of m6A-related genes and proteins. The m6A methylationome was detected by MeRIP-seq, following analysis of the data. Differential methylation of genes was assessed utilizing RT-qPCR and Western Blotting. The effect of ginger on SKOV3 invasion in ovarian cancer cells was investigated using the wound healing assay and transwell assays. Results Ginger significantly reduced the m6A level of OC cells SKOV3. The 3’UTR region is the major site of modification for m6A methylation, and its key molecular activities include Cell Adhesion Molecules, according to meRIP-seq results. Moreover, it was observed that Ginger aids significantly in downregulating the CLDN7, CLDN11 mRNA, and protein expression. The results of wound healing assay and transwell assay showed that ginger significantly inhibited the invasion of OC cells SKOV3. Conclusions Ginger inhibits ovarian cancer cells’ SKOV3 invasion by regulating m6A methylation through CLDN7, CLDN11, and CD274.

This study employed dual-azimuth scanning MAX-DOAS to monitor vertical column densities of NO2 and HCHO in Shanghai during the summer and winter of 2023, and compared the results with Sentinel-5P TROPOMI data. Dual-azimuth scanning revealed a generally consistent trend in gas concentrations (r > 0.95), but concentrations at 90° were higher than those at 0°, especially near the surface. This suggests that averaging multiple azimuth angles is necessary to better represent regional pollution levels. During the observation period, diurnal patterns revealed that NO2 exhibited a “double peak” in the morning and evening, which was more pronounced in the summer, while HCHO peaked between 13:00 and 15:00. Comparisons with the TROPOMI data demonstrated overall good agreement. However, the probability of TROPOMI’s NO2 and HCHO measurements being lower than those of MAX-DOAS was 80% and 62.5%, respectively. Furthermore, TROPOMI tended to overestimate at high concentrations, with overestimation reaching 41.14% for NO2 when exceeding 9.54 × 1015 molecules/cm2 and 25.93% for HCHO when exceeding 1.26 × 1016 molecules/cm2. Sensitivity analysis of the sampling distance (0–40 km) between TROPOMI samples and the ground-based site, and the sampling time (±5 to ±60 min) relative to the TROPOMI overpass, revealed that using a sampling distance of 15–25 km for NO2 and 10–20 km for HCHO, along with appropriately shortening sampling times in the winter and extending them in the summer, can effectively enhance the consistency between satellite and ground-based observations. These findings not only reveal the spatiotemporal distribution characteristics of regional pollutants but optimize the sampling time and distance parameters for satellite–ground observation validation, providing data support for improving and enhancing the accuracy of satellite retrieval algorithms.

Single-channel seismic (SCS) methods play a crucial role in offshore wind farm assessments, offering rapid and continuous imaging of the subsurface. Conventional SCS methods often fall short in resolution and signal completeness, leading to potential misinterpretations of geological structures. In this study, we propose the application of seismic interferometry as a powerful tool to address these challenges by utilizing multiple reflections that are usually considered as noise. First, we demonstrate the feasibility of using seismic interferometry to approximate the primary wavefield. Then, we evaluate a series of seismic interferometry applied in SCS data, including cross-correlation, deconvolution, and cross-coherence, and determine the most appropriate one for our purpose. Finally, by comparing and analyzing the differences in amplitude, continuity, time–frequency properties, etc., between conventional primary wavefield information and reconstructed primary wavefield information by seismic interferometry, it is proved that incorporating multiples as supplementary information through seismic interferometry significantly enhances data reliability and resolution. The introduction of seismic interferometry provides a more detailed and accurate geological assessment crucial for optimal site selection in offshore wind farm development.

In order to investigate the characteristics and driving factors of keystone species change in vegetation communities, we conducted a network analysis to analyze the complexity of grassland plant communities and keystone species. We conducted a study with six plots of degraded grassland, intact grassland, and various ages of artificial grassland. We systematically analyzed the characteristics of vegetation community and soil physical–chemical properties under different treatments. Our findings revealed that the 5-year-old artificial grassland exhibited lower biodiversity, as indicated by a Shannon–Weiner index of 1.70 and a Pielou’s evenness index of 0.72. In comparison to the degraded grassland, soil organic carbon and nitrate nitrogen levels in the 5-year-old artificial grassland increased by 22% and 13%, respectively, while soil density decreased by 9%. However, despite the relatively stable plant community in the 21-year-old artificial grassland, it had not fully recovered to the level of natural grassland due to the complexity of the plant community and the shift in keystone species. Therefore, it is crucial to consider the impact of soil bulk density and nutrient levels on the complexity of the plant community when restoring degraded alpine meadows through the establishment of artificial grassland.

Network dismantling is an important question that has attracted much attention from many different research areas, including the disruption of criminal organizations, the maintenance of stability in sensor networks, and so on. However, almost all current algorithms focus on unsigned networks, and few studies explore the problem of signed network dismantling due to its complexity and lack of data. Importantly, there is a lack of an effective quality function to assess the performance of signed network dismantling, which seriously restricts its deeper applications. To address these questions, in this paper, we design a new objective function and further propose an effective algorithm named as DSEDR, which aims to search for the best dismantling strategy based on evolutionary deep reinforcement learning. Especially, since the evolutionary computation is able to solve global optimization and the deep reinforcement learning can speed up the network computation, we integrate it for the signed network dismantling efficiently. To verify the performance of DSEDR, we apply it to a series of representative artificial and real network data and compare the efficiency with some popular baseline methods. Based on the experimental results, DSEDR has superior performance to all other methods in both efficiency and interpretability.

Understanding the spatiotemporal distribution and transport of atmospheric water vapor in urban areas is crucial for improving mesoscale models and weather and climate predictions. This study employs Multi-Axis Differential Optical Absorption Spectroscopy to monitor the dynamic distribution and transport flux of water vapor in Beijing within the tropospheric layer (0–4 km) from June 2021 to May 2022. The seasonal peaks in precipitable water occur in August, reaching 39.13 mm, with noticeable declines in winter. Water vapor was primarily distributed below 2.0 km and generally decreases with increasing altitude. The largest water vapor transport flux occurs in the southeast–northwest direction, whereas the smallest occurs in the southwest–northeast direction. The maximum flux, observed at about 1.2 km in the southeast–northwest direction during summer, reaches 31.77 g/m2/s (transported towards the southeast). Before continuous rainfall events, water vapor transport, originating primarily from the southeast, concentrates below 1 km. Backward trajectory analysis indicates that during the rainy months, there was a higher proportion of southeasterly winds, especially at lower altitudes, with air masses from the southeast at 500 m accounting for 69.11%. This study shows the capabilities of MAX-DOAS for remote sensing water vapor and offers data support for enhancing weather forecasting and understanding urban climatic dynamics.

BackgroundThe role of drug repositioning in the treatment of ovarian cancer has received increasing attention. Although promising results have been achieved, there are also major controversies.MethodsIn this study, we conducted a drug-target Mendelian randomisation (MR) analysis to systematically investigate the reported effects and relevance of traditional drugs in the treatment of ovarian cancer. The inverse-variance weighted (IVW) method was used in the main analysis to estimate the causal effect. Several MR methods were used simultaneously to test the robustness of the results.ResultsBy screening 31 drugs with 110 targets, FNTA, HSPA5, NEU1, CCND1, CASP1, CASP3 were negatively correlated with ovarian cancer, and HMGCR, PLA2G4A, ITGAL, PTGS1, FNTB were positively correlated with ovarian cancer.ConclusionStatins (HMGCR blockers), lonafarnib (farnesyltransferase inhibitors), the anti-inflammatory drug aspirin, and the anti-malarial drug adiponectin all have potential therapeutic roles in ovarian cancer treatment.

Abstract Background Patients with nasopharyngeal carcinoma (NPC) undergoing radiotherapy experience significant fatigue, which is frequently underestimated due to the lack of objective indicators for its evaluation. This study aimed to explore the longitudinal association between fatigue and nutrition status 1 week in advance. Methods From January 2021 to June 2022, a total of 105 NPC patients who received intensity‐modulated radiation therapy were enrolled in the observational longitudinal study. The significant outcomes, including the Piper Fatigue Scale‐12 (PFS‐12), the Scored Patient‐Generated Subjective Global Assessment (PG‐SGA), four body composition indices, and the Hospital Anxiety and Depression Scale (HADS), were assessed weekly from pre‐treatment until the completion of radiotherapy (T0–T7) to explore their relationship. Results The trajectories of PFS‐12 and all dimensions for 105 participants reached a peak during the fifth week. Sensory fatigue consistently received the highest scores (T0 = 1.60 ± 2.20, T5 = 6.15 ± 1.57), whereas behavior fatigue exhibited the fastest increase over time (T0 = 1.11 ± 1.86, T5 = 5.47 ± 1.70). Higher PG‐SGA scores were found to be weakly explainable for aggravating fatigue (β = 0.02 ~ 0.04). Unlike generalized additive mixed models, marginal structural models (MSM) produced larger effect values (β = 0.12 ~ 0.21). Additionally, body composition indices showed weakly negative relationships with fatigue in MSMs one week in advance. Conclusions The PG‐SGA may be a more accurate predictor of future‐week fatigue than individual body composition indicators, particularly when HADS is controlled for as a time‐dependent confounder.

Background: Jiuwan decoction has been used to treat chronic eczema since the Qing Dynasty. According to clinical experience, Shizhenqing granules (SZQG), derived from the Jiuwan decoction, exert beneficial clinical effects on acute eczema and reduce recurrence. Therefore, we elucidated the underlying mechanisms of SZQG through network pharmacology, molecular docking, and experimental validation. Methods: The main chemical components of SZQG were identified by ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). And the targets of SZQG against eczema were screened out through online databases. Then, the regulatory network map of the “herbal compound–potential target” and the target protein-protein interaction (PPI) network was constructed. The Gene Ontology (GO) analysis and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were conducted using by R language. Additionally, the interaction between the active compounds and the targets was verified by molecular docking technology. Finally, an experiment in vivo was used to verify the effect and mechanism of SZQG on eczema. Results: Using UHPLC-MS/MS, 158 main chemical compounds of SZQG were identified, and 72 compounds were selected according to the criteria for further analysis. All 237 potential targets of SZQG in eczema were explored using multiple online databases. The network with 14 core targets was screened out, including STAT3, RELA, TNF, JUN, MAPK3, IL-6, PIK3CA, STAT1, MAPK14, MAPK1, IL-4, NFKBIA, IL1B, and MYC. KEGG analyses indicated that the therapeutic effects of SZQG on eczema were predominantly associated with cytokine-cytokine receptor interaction, TNF, MAPK, NF-κB, toll-like receptor, T cell receptor, and Th1 and Th2 cell differentiation signaling pathways. Furthermore, the good affinity between the core compounds and core targets was verified by molecular docking technology, particularly for RELA and MAPK. Animal experiments revealed that SZQG downregulated MAPK14, RELA, T-bet, and GATA3 mRNA expression, reduced immunoglobulin E (IgE) and interleukin-4 (IL-4) serum concentrations, and improved eczema-like lesions in model rats. Conclusion: This study identified potential targets and signaling pathways of SZQG in the treatment of eczema, whereby RELA and MAPK14 may constitute the main therapeutic targets of SZQG in cytokine regulation and reduction of inflammatory responses.

Environmental surveillance (ES) is a useful approach for monitoring circulating viruses, including polioviruses (PVs) and non-polio enteroviruses (NPEVs). In this study, the results of nine years of ES from 2013 to 2021 at six sampling sites in three cities in Fujian Province, China, were summarized. It showed that the sewage samples contained abundant viruses, but the positive rate was affected by different sampling sites. From the 520 samples, 431 PVs, 1,713 NPEVs, and 281 human adenoviruses (HAdVs) were isolated. PV isolates had been markedly affected following the adjustment of the immunization strategy. All but one PV isolate were Sabin-like strains without wild PVs. One isolate was vaccine-derived PV type 3 with 10 variation points in the VP1 region. After May 2016, PV type 2 was no longer detected, and PV type 3 became a superior serotype. Of 1,713 NPEVs, 24 serotypes were identified, including echovirus11 (E11), E6, coxsackievirus B3 (CVB3), CVB5, E7, and E3 were the predominant serotypes (37.65%, 20.96%, 11.50%, 8.87%, 8.23%, and 7.06%, respectively). The temporal dynamic of the six common serotypes was inconsistent. E3 was frequently isolated, but the number of isolates was low, with no obvious peaks. E6, E7, and CVB3 exhibited periodic changes with a high peak every three to four years, and E11 only had one high peak lasting four years. Summer-fall peaks of the echoviruses and spring-winter peaks of CVB were observed in the monthly distribution of virus isolation. The infectious isolates of various serotypes of different species identified from the sewage samples showed that ES is an essential part of pathogen surveillance.

ObjectiveTo examine the effect of hepatitis A virus (HAV) vaccine vaccination on serum HAV antibody level 10 years after HAV vaccine being included into the national Expanded Programme on Immunization (EPI) among the residents in Fujian province for providing evidence to prevention and control of viral hepatitis A.MethodsWe collected blood samples among 4 827 residents aged 0 – 60 years recruited with stratified random cluster sampling in 10 municipalities/prefectures across Fujian province from December 2018 through March 2019. Serum HAV immuno-globulin G (HAV-IgG) of the participants were detected with enzyme-linked immunosorbent assay (ELISA) and compared with those among healthy residents of same age groups in 2009.ResultsThe positive rate of HAV-IgG was 96.86% for all the participants, which was significantly higher than that (49.51%) for the healthy residents aged 0 – 60 years in 2009. Unsignificantly increased serum HAV-IgG positive rate was detected among 20 – 50 years-old childbearing women in comparison to that among the childbearing women of same ages in 2019 (96.73% vs. 83.14%); the positive rate was also increased in the participants less than one year old in contrast to the infants of same age in 2009 (96.76% vs. 32.37%). In the participants aged 1 – 15 years old, the standardized HAV vaccine vaccination rate was 86.06% and significantly higher than that (46.47%) in the residents of same ages in 2009.ConclusionThe positive rate of serum HAV-IgG increased significantly among 0 – 60 years old residents and the HAV-IgG content remained at a relatively high level in the infants less than one year old in 2019, ten years after HAV vaccine being included into EPI in Fujian province.

Objective: To explore the impact of seasonal changes on the T-helper 1 (Th1)/T-helper 2 (Th2) immune balance of the body in order to elucidate the internal immunological mechanisms underlying seasonal eczema. Methods: Eighty 4-week-old male Sprague–Dawley rats were divided into 5 season groups kept in corresponding season simulation environments, and subdivided into the model group and the control group. The Eczema Area and Severity Index (EASI) and scratching frequency were evaluated. The serum levels of immunoglobulin E (IgE), interleukin-2 (IL-2), interleukin-12 (IL-12), and interferon-gamma (IFN-γ), interleukin-4 (IL-4), interleukin-25 (IL-25), and interleukin-31 (IL-31), and melatonin (MT), as well as the MT receptor (MTR) levels in the spleen, were detected by enzyme-linked immunosorbent assay analysis. The mRNA expression levels of T-bet and GATA3 were detected by quantitative real-time polymerase chain reaction. Results: EASI scores and the scratching frequency of the model group were higher in the long summer than in the other 4 seasons. Meanwhile, the serum levels of IgE and the Th2 cytokines IL-4, IL-25, and IL-31, as well as GATA3 mRNA expression levels, were high during the long summer in both groups. However, the serum levels of the Th1 cytokines IL-2, IL-12, and IFN-γ, as well as MT, MTR, and T-bet mRNA levels, were lower during the long summer. In all 5 seasonal groups (spring, summer, long summer, autumn, and winter), the levels of all immune factors, especially IL-4 and IL-31, were higher in the model group than those in the control group, while the concentrations of MT and MTR were lower. Conclusion: Under long light, hot, and humid conditions in the long summer, the body is more likely to suffer from Th2-dominated immune imbalance. This is the internal mechanism behind the high incidence and severity of eczema during the long summer. MT and MTR play a key role in the seasonal onset of eczema.

Due to the complex correlation within data collection, it is a challenging task to detect leakage in the water supply network. The Graph Convolutional Network (GCN) has recently gained significant attention in correlation research. However, most existing GCN-based models assumed that the topology of whole network should be derived from expert knowledge, which is always time-consuming and difficult to acquire. To tackle this problem, a data-driven improved graph convolutional network (IGCN) is proposed based on a self-learning fully connected association graph. Compared with traditional GCN, the proposed model can adaptively learn necessary data relationship information without accurate fixed undirected association graph. On this basis, a leakage-detection IGCN (LD-IGCN) is carried out for leakage detection. Finally, two case studies of water supply networks are utilized to demonstrate the efficiency of the proposed leakage detection method in this paper.

Engineering vehicle recognition based on video surveillance is one of the key technologies to assist illegal land use monitoring. At present, the engineering vehicle recognition mainly adopts the traditional deep learning model with a large number of floating-point operations. So, it cannot be achieved in edge devices with limited computing power and storage in real time. In addition, some lightweight models have problems with inaccurate bounding box locating, low recognition rate, and unreasonable selection of positive training samples for the small object. To solve the problems, the article proposes an improved lightweight Yolo-Fastest V2 for engineering vehicle recognition fusing location enhancement and adaptive label assignment. The location-enhanced feature pyramid network structure combines deep and shallow feature maps to accurately localize bounding boxes. The grouping k-means clustering strategy and adaptive label assignment algorithm select an appropriate anchor for each object based on its shape and Intersection over Union. The study was conducted on Raspberry Pi 4B 2018 using two datasets and different models. Experiments show that our method achieves the optimal combination in speed and accuracy. Specifically, the mAP50 is increased by 7.02% with the speed of 11.24 fps under the engineering vehicle data obtained by video surveillance in a rural area of China.

Sepsis is an abnormal immune response to infections and can trigger MODS. Despite the availability of advanced clinical techniques and monitoring methods, the mortality rate of the disease is still high, posing a heavy burden to patients and the whole society. Hence, the research on novel drugs and targets is particularly important. As a natural phyto-flavonoid, apigenin boasts anti-inflammatory, antioxidant, anti-cancer, anti-viral, and anti-bacterial effects. Besides, in-vitro experiments and animal models have also revealed the crucial role of apigenin in the treatment of infectious diseases and sepsis. In this context, this paper reviews the pharmacological activity and underlying mechanisms of action of apigenin in sepsis treatment and organ protection, as well as the potential apigenin-based therapeutic strategies against sepsis. Therefore, this review will shed new light on the scientific research and clinical treatment of sepsis.

Abstract The phytohormone abscisic acid (ABA) plays important roles in plant growth, development and adaptative responses to abiotic stresses. SNF1-related protein kinase 2s (SnRK2) are key components that activate the ABA core signaling pathway. NUCLEAR PORE ANCHOR (NUA) is a component of the nuclear pore complex (NPC) that involves in deSUMOylation through physically interacting with the EARLY IN SHORT DAYS 4 (ESD4) SUMO protease. However, it is not clear how NUA functions with SnRK2 and ESD4 to regulate ABA signaling. In our study, we found that nua loss-of-function mutants exhibited pleiotropic ABA-hypersensitive phenotype. We also found that ABA-responsive genes remarkably up-regulated in nua by exogenous ABA. The nua snrk2.2 snrk2.3 triple mutant and nua abi5 double mutant partially rescued the ABA-hypersensitive phenotype of nua, thereby suggesting that NUA is epistatic to SnRK2s. Additionally, we observed that esd4-3 mutant was also ABA-hypersensitive. NUA and ESD4 were further demonstrated to physically interact with SnRK2s and negatively regulate ABA signaling by reducing SnRK2s stability. Taken together, our findings uncover a new regulatory mechanism that can modulate ABA signaling.

Objective: To retrospectively study the efficacy of bone cement-enhanced pedicle screws combined with interbody fusion reduction in the treatment of severe lumbar spondylolisthesis, its effect on lumbar function and complications. Methods: From January 2019 to June 2021, 82 cases of severe lumbar spondylolisthesis in our hospital were analyzed. According to the different treatment plans, the patients were divided into two groups: A and B, in which group A patients were treated with pedicle screws combined with fusion and reduction, and group B patients were treated with bone cement reinforced pedicle screws combined with fusion and reduction. Perioperative indexes, pain severity (VAS), Oswestry dysfunction index (ODI), and low back and leg pain score of Japan Orthopedic Association (JOA), spondylolisthesis reduction, intervertebral space height, intervertebral foramen height, complications, and screw loosening were compared between the two groups. Results: There was no significant difference in the amount of intraoperative bleeding between group A and group B (P > 0.05). The operation time of group B was longer than that of group A, and the hospitalization time was shorter than that of group A. The vertebral fusion rate of group B was higher than that of group A (P 0.05). Conclusion: Compared with conventional screw treatment, bone cement-reinforced pedicle screws combined with fusion repositioning can further improve the repositioning rate of slipped vertebrae in the treatment of severe LSL, while achieving a better intervertebral fusion rate. Therefore, the treatment of severe LSL with bone cement reinforced pedicle fusion and reduction is a safe and effective method.

Saffron is located in the upper part of the crocus stigma of iridaceae, which has a long history of medicinal use. Crocin (molecular formula C44H64O24) is a natural floral glycoside ester compound extracted from saffron, which is a type carotenoid. Modern pharmacological studies have shown that crocin has multiple therapeutic effects including anti-inflammatory, anti-oxidant, anti-hyperlipidemic and anti-stone effects. In recent years, crocin has been widely noticed due to its considerable anti-tumor effects manifested by the induction of tumor cell apoptosis, inhibition of tumor cell proliferation, inhibition of tumor cell invasion and metastasis, enhancement of chemotherapy sensitivity and improvement of immune status. The anti-tumor effects have been shown in various malignant tumors such as gastric cancer, liver cancer, cervical cancer, breast cancer and colorectal cancer. In this review, we compiled recent studies on the anti-tumor effects of crocin and summarized its anti-tumor mechanism for developing ideas of treating malignancies and exploring anti-tumor drugs.

Background: Based on the effect of seasonal changes on human visceral function, this study investigated the impact of seasonal photoperiod of the pineal body on hypothalamic-pituitary-adrenal axis-hippocampal-receptor in rats, aiming to reveal the mechanism by which pineal gland melatonin regulates the seasonal secretion of hippocampal neurotransmitters. Methods: Vernal equinox, summer solstice, autumn equinox, and winter solstice were selected as four experimental time points, and rats were randomly divided into normal control group, sham operation group, and pinealectomized group. The seasonal changes in corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH), corticosterone, hypothalamic melatonin receptor (MTR), and hippocampal corticosterone receptor (CORTR) were examined by enzyme-linked immunosorbent assay. Abstract Five severe floods occurred in the Yangtze River Basin, China, between July and August 2020, and the Three Gorges Reservoir (TGR) located in the middle Yangtze River experienced the highest inflow since construction. The world’s largest cascade-reservoir group, which counts for 22 cascade reservoirs in the upper Yangtze River, cooperated in real time to control floods. The cooperation prevented evacuation of 600,000 people and extensive inundations of farmlands and aquacultural areas. In addition, no water spillage occurred during the flood control period, resulting in a world-record annual output of the TGR hydropower station. This work describes decision making challenges in the cooperation of super large reservoir groups based on a case-study, controlling the 4th and 5th floods (from Aug-14 to Aug-22), the efforts of technicians, multi-departments, and the state, and reflects on these. To realize the full potential of reservoir operation for the Yangtze River Basin and other basins with large reservoir groups globally, we suggest: (i) improve flood forecast accuracy with a long leading time; (ii) strengthen and further develop ongoing research on reservoir group cooperation; and (iii) improve and implement institutional mechanisms for coordinated operation of large reservoir groups.

Many studies have reported obvious seasonal differences in the intestinal flora of rats, and this stable distribution of the seasonal flora helps in maintaining the normal physiological function of the host. However, the mechanism underlying these seasonal differences in intestinal flora remains unclear. To explore the correlation among seasonal factors and intestinal water metabolism and intestinal flora, 20 Sprague Dawley (SD) rats were divided into spring, summer, autumn, and winter groups. The environment for the four seasons was simulated using the Balanced Temperature and Humidity Control system. The intestinal water metabolism was evaluated by determining the intestinal transmission function, fecal water content, water content of colonic tissue, and the colonic expression levels of AQP3, AQP4, and AQP8. The composition and relative abundance of intestinal microflora in rats in each season were assessed through 16S rDNA amplifier sequencing, and the relationship between the dominant flora and intestinal water metabolism in each season was analyzed using Spearman correlation analysis. The high temperature and humidity season could lead to an increase in intestinal water metabolism and intestinal water content in rats, whereas the low temperature and humidity season could lead to a decrease, which was closely related to the change in microflora. To explore the molecular mechanism of seasonal changes in intestinal water metabolism, the concentration of colonic 5-HT, VIP, cAMP, and PKA associated with intestinal water metabolism in rats were also examined. Seasonal changes could affect the concentration of colonic 5-HT and VIP in rats, and then regulate AQPs through cAMP/PKA pathway to affect the intestinal water metabolism. These results suggest that seasonal factors affect the level of intestinal water metabolism in rats and result in seasonal differences in intestinal flora.

New anti-cancer drugs are constantly being developed, especially targeted drugs. Although these drugs have achieved significant clinical efficacy, they do not play a significant role in ovarian cancer. Moreover, the research cycle and costs of such drugs are often huge. The repositioning of conventional drugs has gradually become a concern. Statins, as traditional lipid-lowering drugs, play a role mainly by inhibiting HMGCR. In recent years, epidemiological studies and in vitro experiments have confirmed its anti-cancer effect, especially the effect of anti-ovarian cancer. The mutation rate of TP53 in ovarian cancer is as high as 95%, while HMGCR is often highly expressed in TP53 mutant tumors. However, the effect of prospective clinical trials is not ideal. This result seems understandable considering that it seems unrealistic for a lipid-lowering drug to completely inhibit tumor growth. Therefore, statins play more synergistic roles in the treatment of ovarian cancer. Because ovarian cancer is a highly heterogeneous tumor, it may be a good choice to deeply understand the mechanism of statins in the treatment of ovarian cancer and achieve precise treatment by combining it with other drugs.

Abdominal aortic aneurysm (AAA) is a life-threatening disease and there is currently a lack of effective treatment to prevent it rupturing. ScRNA-seq studies of AAA are still lacking. In the study, we analyzed the published AAA scRNA-seq datasets from the mouse elastase-induced model, CaCl2 treatment model, Ang II-induced model and human by using bioinformatic approaches and in silico analysis. A total of 26 cell clusters were obtained and 11 cell types were identified from multiple mouse AAA models. Also, the proportion of Mφ/Mo increased in the AAA group and Mφ/Mo was divided into seven subtypes. There were significant differences in transcriptional regulation patterns of Mφ/Mo in different AAA models. The enrichment pathways of upregulated or downregulated genes from Mφ/Mo in the three mouse datasets were different. The actived regulons of Mφ/Mo had strong specificity and the repressed regulons showed high consistency. The co-upregulated genes as well as actived regulons and co-downregulated genes as well as repressed regulons were closely correlated and formed regulatory networks. Mφ/Mo from human AAA dataset was divided into five subtypes. The proportion of three macrophage subpopulations increased but the proportion of two monocyte subpopulations decreased. In the AAA group, the upregulated or downregulated genes of Mφ/Mo were enriched in different pathways. After further analyzing the genes in Mφ/Mo of both mouse and human scRNA-seq datasets, two genes were upregulated in the four datasets, IL-1B and THBS1. In conclusion, in silico analysis of scRNA-seq revealed that Mφ/Mo and their regulatory related genes as well as interaction networks played an important role in the pathogenesis of AAA.

Rock grain size parameter is the key parameter of reservoir rock physics analysis. The study found that the relationship between the NMRT2spectrum and rock grain size distribution curve is directly related to NMRT2distribution and grain size distribution of rock, so you can use T2 to retrieve the size distribution of rock NMR spectral data. Based on the calculation of the rock core experiment results of grain size distribution by NMR T2 distribution using the conversion method of piecewise nonlinear calibration for each core, adjusting the calibration parameters to make the grain size distribution calculated with the core analysis of grain size distribution approximation, the error is minimum to obtain each core scale parameter conversion. In the end, the core parameters are classified according to the parameters of pore and permeability. Finally, according to the actual NMR T2 distribution curve inversion of underground rock granularity, the inversion results contrast with the core analysis results, and the reliability of the method is verified to provide accurate, continuous rock size distribution profile analysis of geological reservoir rock physics.

Stigmasterol is a phytosterol derived from multiple herbaceous plants such as herbs, soybean and tobacco, and it has received much attention for its various pharmacological effects including anti-inflammation, anti-diabetes, anti-oxidization, and lowering blood cholesterol. Multiple studies have revealed that stigmasterol holds promise as a potentially beneficial therapeutic agent for malignant tumors because of its significant anti-tumor bioactivity. It is reported that stigmasterol has anti-tumor effect in a variety of malignancies (e.g., breast, lung, liver and ovarian cancers) by promoting apoptosis, inhibiting proliferation, metastasis and invasion, and inducing autophagy in tumor cells. Mechanistic study shows that stigmasterol triggers apoptosis in tumor cells by regulating the PI3K/Akt signaling pathway and the generation of mitochondrial reactive oxygen species, while its anti-proliferative activity is mainly dependent on its modulatory effect on cyclin proteins and cyclin-dependent kinase (CDK). There have been multiple mechanisms underlying the anti-tumor effect of stigmasterol, which make stigmasterol promising as a new anti-tumor agent and provide insights into research on its anti-tumor role. Presently, stigmasterol has been poorly understood, and there is a paucity of systemic review on the mechanism underlying its anti-tumor effect. The current study attempts to conduct a literature review on stigmasterol for its anti-tumor effect to provide reference for researchers and clinical workers.

Background Since 2008, Fujian province provided measles-rubella (MR) vaccine at 8 months followed by measles-mumps-rubella (MMR) vaccine at 18 months a one-dose mumps-containing-vaccine (MuCV) schedule. Several mumps outbreaks have occurred recently in Fujian. Serological surveillance can assess population immunity to mumps and identify risk factors for mumps. Methods We conducted a cross-sectional serosurvey of mumps IgG antibodies in the general population of Fujian Province in 2018 and compare results with a similar study conducted in 2009, when the routine schedule had no MuCV. We analyzed changes in mumps epidemiology after implementation of a one-dose MuCV vaccination strategy. Results Mumps seroprevalence was 78.6% (95% CI: 77.4–79.8), and the geometric mean concentration (GMC) of mumps antibodies was 245.8 IU/ml (95% CI:237.3–255.1). MuCV vaccination at 18 months resulted in increased seroprevalence and GMCs. Seroprevalence and GMCs varied by age, gender, and number of doses received. Except for children under 18 months, seroprevalence and GMCs were lowest among 10-15-year-olds. Each year after introduction of the one-dose MuCV vaccination policy, the highest incidence of mumps was among 4–6-year-olds and 9–15-year-olds, gradually shifting to older age groups. Conclusion A one-dose mumps-containing vaccine schedule does not provide sustained and stable mumps immunity in Fujian. To reduce the risk of mumps, we recommend supplementary vaccination of children without a history of receiving at least one MuCV dose or who are seronegative at 10–15 years of age.

KIAA1429 is a major m6A methyltransferase, which plays important biological and pharmacological roles in both human cancer or non-cancer diseases. KIAA1429 produce a tumorigenic role in various cancers through regulating DAPK3, ID2, GATA3, SMC1A, CDK1, SIRT1 and other targets, promoting cell proliferation, migration, invasion, metastasis and tumor growth . At the same time, KIAA1429 is also effective in non-tumor diseases, such as reproductive system and cardiovascular system diseases. The potential regulatory mechanism of KIAA1429 dependent on m6A modification is related to mRNA, lncRNA, circRNA and miRNAs. In this review, we summarized the current evidence on KIAA1429 in various human cancers or non-cancer diseases and its potential as a prognostic target.

Abstract Colorectal cancer is the most common gastrointestinal cancer and causes severe damage to human health. PRDX2 is a member of the peroxiredoxin family reported to have a high level of expression in colorectal cancer. However, the mechanisms by which PRDX2 promotes the proliferation of colorectal cancer are still unclear. Here, the results indicated that PRDX2 expression was upregulated in colorectal cancer and closely correlated with poor prognosis. Functionally, PRDX2 promoted the proliferation of colorectal cancer cells. Mechanistically, PRDX2 could bind RPL4, reducing the interaction between RPL4 and MDM2. These findings demonstrate that the oncogenic property of PRDX2 may be attributed to its regulation of the RPL4-MDM2-p53 pathway, leading to p53 ubiquitinated degradation.

Background: Based on the theory of “five Zang-organs corresponding to the seasons” in traditional Chinese medicine (TCM), physiological functions including emotions vary with the seasons. We aimed to investigate the seasonal photoperiod effects of melatonin (MT) released from the pineal gland on the MT receptor (MTR)-Gs/Gi-cyclic adenosine monophosphate (cAMP)-protein kinase A (PKA)-cAMP response element-binding protein (CREB) signaling pathway in the hippocampus. Methods: Rats were divided into three groups: control, operation (surgery with pineal gland removal), and pseudo-operation groups (same surgery as operation group but without removing pineal gland), and fed at specific time across the four seasons. The levels of MTR, adenylate cyclase (AC), cAMP, PKA, and CREB in the hippocampus were analyzed using ELISA. The concentrations of Gs and Gi were analyzed using Western blot. The expression of CREB mRNA was detected by PCR. AbstractA large number of CO2-bearing gas layers are distributed in the deep layers in the Huangliu and Meishan formations of the Dongfang 13 gas field. The presence of CH4, CO2, and formation water in CO2-bearing gas layers complicates the change of logging response characteristics. In particular, the unclear understanding of the additional conductivity change law formed by the coupling of CO2 and formation water results in low accuracy of the water saturation results, and the aforementioned type of gas layer can easily be misidentified as the gas-water mixed layer. By conducting multiple rock electrical experiments under simulated formation temperature and pressure conditions, core flooding experiments were conducted with different contents of CO2 and CH4 mixed gas to determine the lithology coefficient b, saturation coefficient n, and cementation coefficient m of cores under different experimental conditions. Additionally, we also analyzed the relationship between the electrical parameters of rocks, CO2 content, and water saturation to reveal the influence of coupling of CO2 and formation water on the change of electrical parameters of rocks under high temperature and pressure conditions. The experimental results show that when the mixed gas with different CO2 contents is dissolved in the formation water, there is no difference between the measured lithology coefficient a and the cementation coefficient m; as the CO2 content increases, the lithology coefficient b increases, and the saturation coefficient n decreases. Based on the experimental results, we established a prediction model of the change between lithology coefficient b, saturation coefficient n, and CO2 content. This model improves the accuracy of water saturation calculations in the CO2-bearing gas layers along with that of fluid identification.

β-Sitosterol (SIT), a white powdery organic substance with a molecular formula of C29H50O, is one of the most abundant naturally occurring phytosterols in plants. With a chemical composition similar to that of cholesterol, SIT is applied in various fields such as medicine, agriculture, and chemical industries, owing to its unique biological and physicochemical properties. Modern pharmacological studies have elucidated good anti-tumor therapeutic effect activity of SIT, which mainly manifests as pro-apoptotic, anti-proliferative, anti-metastatic, anti-invasive, and chemosensitizing on tumor cells. In addition, SIT exerts an anti-tumor effect on multiple malignant tumors such as breast, gastric, lung, kidney, pancreatic, prostate, and other cancers. Further, SIT derivatives with structural modifications are promising anti-tumor drugs with significant anti-tumor effects. This review article focuses on recent studies relevant to the anti-tumor effects of SIT and summarizes its anti-tumor mechanism to provide a reference for the clinical treatment of malignant tumors and the development of novel anti-tumor drugs.

The molecular mechanism of AAA formation is still poorly understood and has not been fully elucidated. The study was designed to identify the immune-related genes, immune-RAS in AAA using bioinformatics methods. The GSE175683 datasets were downloaded from the GEO database. The DEseq2 software was used to identify differentially expressed genes (DEGs). SUVA pipeline was used to quantify AS events and RAS events. KOBAS 2.0 server was used to identify GO terms and KEGG pathways to sort out functional categories of DEGs. The CIBERSORT algorithm was used with the default parameter for estimating immune cell fractions. Nine samples from GSE175683 were used to construct the co-disturbed network between expression of SFs and splicing ratio of RAS events. PCA analysis was performed by R package factoextra to show the clustering of samples, and the pheatmap package in R was used to perform the clustering based on Euclidean distance. The results showed that there were 3,541 genes significantly differentially expressed, of which 177 immune-related genes were upregulated and 48 immune-related genes were downregulated between the WT and WTA group. Immune-RAS events were mainly alt5P and IR events, and about 60% of it was complex splicing events in AAA. The WT group and the WTA group can be clearly distinguished in the first principal component by using the splicing ratio of immune-RAS events. Two downregulated genes, Nr4a1 and Nr4a2, and eight upregulated genes, Adipor2, Akt2, Bcl3, Dhx58, Pparg, Ptgds, Sytl1, and Vegfa were identified among the immune-related genes with RAS and DEGs. Eighteen differentially expressed SFs were identified and displayed by heatmap. The proportion of different types of cells and ratio of the average ratio of different cells were quite different. Both M1 and M2 types of macrophages and plasma cells were upregulated, while M0 type was downregulated in AAA. The proportion of plasma cells in the WTA group had sharply increased. There is a correlation between SF expression and immune cells/immune-RAS. Sf3b1, a splicing factor with significantly different expression, was selected to bind on a mass of immune-related genes. In conclusion, our results showed that immune-related genes, immune-RAS, and SFs by genome-wide identification were involved in AAA.

The purpose of this study was to investigate how to detect abnormalities in electric submersible pumps (ESPs) in advance and how to classify the faults by monitoring the production data before pumps break down. Additionally, a new method based on the denoising autoencoder (DAE) and support vector machine (SVM) is proposed. Firstly, the ESP production data were processed and fault-related features were screened using the random forest (RF) algorithm. Secondly, input data were randomly damaged by the addition of noise, a DAE network structure was constructed, and the optimal learning rate, noise reduction coefficient, and other parameters were set. Thirdly, the real-time status of the production data of ESP was monitored with reconstruction errors to detect the point when an abnormality occurs signifying a pending fault. Finally, SVM was used to distinguish the type of fault. Compared with existing fault diagnosis methods, our method not only has the advantages of easy extraction of effective data features, higher accuracy, and strong generalization ability but can also detect an abnormal state indicating a coming fault and identify its type, hence enabling the preparation of an appropriate advance solution.

Effective permeability is a key parameter for evaluating reservoirs and their productivity. With the wide application of resistivity logging tools in drilling, the advantages of resistivity logging in response to the dynamic invasion process of drilling fluids have become increasingly prominent. We analyzed the variation law of the measured resistivity data for different permeability formations at different times. In this study, we proposed an effective permeability modeling method based on the time-lapse resistivity logging data. First, based on the resistivity measurement data at different times, the dynamic resistivity profile of the reservoir was obtained through joint inversion; we then obtained the invasion depth and invasion zone resistivity of the drilling fluid at different times, along with the original formation resistivity. Subsequently, combined with parameters such as the soaking time, fluid viscosity, and saturation change of the drilling fluid, we obtained the phase permeability curve of the reservoir and dynamic effective permeability of the fluid near the wellbore. This study provides basic parameters for subsequent formation analyses and productivity prediction and substantially improves the reservoir evaluation technology from static to dynamic.

Modeling the current distribution of and predicting suitable habitats for threatened species support the species conservation and restoration planning process. Therefore, the purpose of this study was to model the actual distribution and predict environmentally suitable habitats for Carex alatauensis S.R.Zhang 2015, a locally threatened native grass species on the Qinghai–Tibet Plateau. To realize this objective, based on the geographical samples within the natural distribution of C. alatauensis, the dominant climatic factors in its potential distribution range were analyzed using the maximum entropy (MaxEnt) model. The results showed that the average values of the area under the receiver operating characteristic curve (AUC) of the training data were 0.833 ± 0.044, which indicated that the accuracy of the MaxEnt model was pretty high for modeling potential distribution regions of C. alatauensis. The combined results from the Jackknife test and the presented contribution of environmental variables revealed that the annual precipitation, the growth season precipitation, and the precipitation of the driest month were the key climatic factors that restricted the distribution of C. alatauensis on the Qinghai–Tibet Plateau. It is predicted that the potential distribution area of C. alatauensis on the Qinghai–Tibet Plateau is 1.96 × 106 km2, and the most suitable area is 3.7 × 105 km2, mainly located in the Qilian Mountains, the Himalayas, and the Qingtanggula Mountains.

Ovarian cancer is one of the most common malignancies and the highest mortality among gynecological malignancy. The standard therapy options for patients with ovarian cancer are cytoreductive surgery and chemotherapy, and although most patients do better with standard treatment, it is easy to relapse and be resistant to chemotherapy. Therefore, it is important to find new therapeutic strategies. More recently, metabolic reprogramming has been recognized as a hallmark of cancer and has become a potential target for tumor therapy. Mutations of metabolic enzymes are closely related to the development of ovarian cancer. The metabolic reprogramming of ovarian cancer not only provides energy to tumor cells, but also participates in various biological processes as signaling molecules. Succinic acid (SA) is an important metabolic intermediate involved in a number of metabolic pathways, such as TCA cycle and glutamine metabolism, and is also widely present in a variety of plants and vegetables. Studies show abnormal SA metabolism in many tumors and affect tumor formation through a variety of mechanisms. But the role of SA in ovarian cancer is less studied. This paper reviews the role of SA and its abnormal metabolic pathway in ovarian cancer.

Tillering ability is a key agronomy trait for wheat (Triticum aestivum L.) production. Studies on a dwarf monoculm wheat mutant (dmc) showed that ARF11 played an important role in tillering of wheat. In this study, a total of 67 ARF family members were identified and clustered to two main classes with four subgroups based on their protein structures. The promoter regions of T. aestivum ARF (TaARF) genes contain a large number of cis-acting elements closely related to plant growth and development, and hormone response. The segmental duplication events occurred commonly and played a major role in the expansion of TaARFs. The gene collinearity degrees of the ARFs between wheat and other grasses, rice and maize, were significantly high. The evolution distances among TaARFs determine their expression profiles, such as homoeologous genes have similar expression profiles, like TaARF4-3A-1, TaARF4-3A-2 and their homoeologous genes. The expression profiles of TaARFs in various tissues or organs indicated TaARF3, TaARF4, TaARF9 and TaARF22 and their homoeologous genes played basic roles during wheat development. TaARF4, TaARF9, TaARF12, TaARF15, TaARF17, TaARF21, TaARF25 and their homoeologous genes probably played basic roles in tiller development. qRT-PCR analyses of 20 representative TaARF genes revealed that the abnormal expressions of TaARF11 and TaARF14 were major causes constraining the tillering of dmc. Indole-3-acetic acid (IAA) contents in dmc were significantly less than that in Guomai 301 at key tillering stages. Exogenous IAA application significantly promoted wheat tillering, and affected the transcriptions of TaARFs. These data suggested that TaARFs as well as IAA signaling were involved in controlling wheat tillering. This study provided valuable clues for functional characterization of ARFs in wheat.

The impact of drought on ecosystems has become increasingly prominent. This study used the MODIS remote-sensing vegetation index and the standardized precipitation index to quantify vegetation coverage status and meteorological drought level. Using the copula method, the joint distribution of the drought index and related vegetation cover variables were simulated for the first time. The conditional distribution of vegetation biomass was deduced, and the multidimensional response between the vegetation biomass and drought was explored to understand the possible vegetation loss under different drought severity conditions. The three-river headwaters (TRH) region is the source place of the Yangtze River, Yellow River, and Lancang River, where the ecosystem is characterized by the innate vulnerability. Using this region as a case study, the results show that the spatiotemporal evolution of drought and vegetation in the TRH region has apparent regional heterogeneity. The vegetation cover in the eastern region is significantly better than that in the northwestern region, while the vegetation growth trend in the northwestern region is stronger than that in the southeastern region. It is feasible to build a vegetation-drought multidimensional response model based on the copula method. In 92.5% of the TRH region, vegetation cover was significantly affected by the severity of the drought. The impact on the growth of vegetation caused by persistent drought events was higher than that of short-term but high-intensity drought. The vegetation in the TRH area has a certain degree of drought resistance. This study provides outstanding theoretical support and reference for the protection of the TRH basin ecosystem.

Array processing is an interdisciplinary topic of both physics and signal processing. Physical basis of array processing is the orderly-setup of sensors in space that induces regular change of the phase of external sources incident upon sensor array. In the current paper the direction-of-arrival (DoA) estimation of an azimuth-only uniform linear array (ULA) is investigated in theory and method from the fused perspective of signal and physical properties of the incident sources. By organically fusing the stationary assumption on source signals and noises with difference operation on the phases, a distinctive system of linear equations satisfied by the incident sources is theoretically derived for the azimuth-only ULA with the Hankel-block-matrix of signal correlations as coefficient matrix and the elementary power-sum symmetric functions of the propagators of incident sources as the unknowns. Based on the derived system of linear equations, signal model of the incident sources is first proved as a degenerate spatial ARMA process subject to the identical autoregressive and moving average parameters and simultaneously obeying the dimensional homogeneity principle (DHP) in physics. The explicit root-finding polynomial is proposed with the unknowns of the system of linear equations as polynomial coefficients and the propagators as the roots. No extraneous roots and conjugate symmetry constraint on polynomial coefficients are involved. The DoAs and noise variances can be separately estimated under the backgrounds of spatially white and colored noises, which are numerically analyzed with the different coherent lengths of noises. A simple sound experiment is designed and performed to verify the proposed DoA estimation method. It is promising to investigate the DoA estimation of the ULA model, particularly of the ULAs of the multi-dimensional array from the fused perspective of physical and signal properties of the incident sources.

River flooding affects more people worldwide than other natural hazards. Thus, analysis of the changes in flood regime caused by global warming and increasing anthropogenic activities will help us make adaptive plans for future flood management. The nonstationary flood behavior in the upper Yangtze River was examined comprehensively in terms of trend, change point, and periodicity with co-usage of different methods. Results show that there are decreasing tendencies in the corresponding series of annual maximum flood peak flow and flood volume in four out of six control stations, except Pingshan and Wulong stations in the Jinsha River and the Wu River, respectively, and the flood peak occurrence time appears earlier mostly. The uniformity of flood process increases in four main tributaries, while it decreases in mainstream of the Yangtze River (Yichang and Pingshan stations). The rates of both rising limb and recession limb of all the typical flood process flowing through the six stations were analyzed. 77.8% of the rates of rising limb decrease, while 61.1% of the rates of recession limb increase, which is almost consistent with the variation reflected by the uniformity. The change points of most evaluation indicators happened in 1970s–1990s. The first main periodicity of evaluation indicators in Yichang is about 45 years, while that of other stations is about 20 years. Invalidity of stationarity in the flood series can be attributed to the intensified construction on major water conservancy projects, changes of underlying surface, and influences of climatic variables. The contributions of both climatic control and the Three Gorges Dam (TGD) to the variation of the annual flood peak in Yichang station were further quantitatively evaluated, which has verified that the construction of the TGD has played a positive role in peak-flood clipping.
