The aim of this study was to obtain base-line morphometric data on the whole brain and brainstem in the adult male and female helmeted guinea fowl (Numida meleagris), and compare the result in the two sexes. Brain samples of seventy adult helmeted guinea fowls, comprising 35 males and 35 females, intended for slaughter, were extracted for the study. The body weights for both male and female helmeted guinea fowls were 1247 ± 5.51 g and 1230 ± 4.85 g, respectively. The mean brain weights were 3.39 ± 0.02 g and 3.47 ± 0.18 g for male and female, respectively, and the difference in the values was not significant (P > 0.05). The lengths of the brain and medulla oblongata, as well as the lengths of the midbrain and mesencephalic tectum, did not differ between the two sexes. Neither the mean weights nor lengths of the pons differed between the two sexes. In conclusion, the results of this study showed that there was no sex dimorphism observed in the morphometry of the midbrain, medulla oblongata and pons in the helmeted guinea fowl.

Type 2 diabetes occurs as a result of insulin resistance and dysfunction in insulin signaling. Controlling hyperglycemia and activation of insulin signaling are important in the management of type 2 diabetes. This study aimed to evaluate the effect of genistein and Momordica charantia L. fruit (MCF) on oxidative stress, markers of inflammation, and their role in proglucagon and insulin receptor messenger RNA (mRNA) expression by real-time PCR in diabetic rats. Thirty-five albino rats were divided into 7 groups ( n  = 5). Group I (non-diabetic) and group II (diabetic control) were treated with distilled water, and groups III and IV received 250 mg/kg and 500 mg/kg lyophilized MCF, respectively. Groups V and VI received 10 mg/kg and 20 mg/kg genistein, respectively, while group VII received 500 mg/kg metformin. The administration lasted for 28 days. MCF and genistein significantly reduced interleukin (IL)-1β and tumor necrosis factor alpha (TNF-α) levels, which were elevated in the serum of diabetic rats. Treatment with MCF and genistein significantly increased the expression of proglucagon mRNA in the small intestine and insulin receptor mRNA in the liver of diabetic rats. In conclusion, MCF and genistein ameliorate type 2 diabetes complications by preventing the loss of insulin-positive cells, inhibiting IL-1β and TNF-α, and upregulating proglucagon and insulin receptor mRNA expression. Novelty: MCF and genistein have an inhibitory effect on diabetic induced IL-1β and TNF-α production. MCF and genistein upregulate proglucagon and insulin receptor mRNA expression.

Purpose: The present study was aimed at evaluating the role of Momordica charantia L. fruit and Genistein on beta cell, insulin resistance/sensitivity and lipid profile in type 2 diabetic rats., Methods: Thirty-five (35) albino rats were divided into seven (7) groups of 5 rats each comprising of five (5) non-diabetic and thirty (30) diabetic rats. Groups 1 and 2 served as the normal control and diabetic control groups respectively and received distill water, groups 3 and 4 received Mormodica charantia L. at 250 mg/kg and 500 mg/kg respectively . Groups 5 and 6 received Genistein at 10 mg/kg and 20 mg/kg respectively while group 7 received Metformin at 500 mg/kg the experiment lasted for four weeks. All the rats were euthanized at the end of the fourth week., Results: Lipid profile, glucose and insulin levels were determined from the analysis of serum parameters and the histology of the pancreas. A significant reduction ( p  < 0.05) in blood glucose levels was noticed in rats that received Momordica charantia L. (MC) and genistein when compared with diabetic control rats. A significant decrease ( p  < 0.05) in cholesterol, triglyceride, low density lipoprotein (LDL) and very low density lipoprotein (VLDL) levels were also noted in rats that received MC and Genistein when compared with the diabetic control rats. MC and Genistein significantly increased ( P  < 0.05) serum insulin level compared to the diabetic control rats. MC and Genistein significantly decreased ( p  < 0.05) homeostatic model assessment-insulin resistance (HOMA-IR) level compared with the diabetic control group. The experiment was conducted to evaluate changes in feed consumption and water intake among broiler chickens subjected to melatonin treatment during the hot-dry season. A total of 300 broiler chicks were selected and assigned into three groups, by simple random sampling, comprising 100 chicks each: group I was exposed to natural photoperiod of about 12-h light and 12-h darkness cycle (12D/12L), without melatonin supplementation; group II was kept under 24-h continuous lighting (CL), without melatonin supplementation; and group III was raised under 24-h CL and administered daily with melatonin orally at 0.5 mg/kg (CL + MEL). Live weight (LW), feed consumption, and water intake for each group were obtained at weekly intervals over a period of 8 weeks. On day 42 of age, the LW of 2420 ± 50 g/bird was obtained in group III administered with melatonin (CL + MEL), while LW values recorded in the 12D/12L and CL groups not administered with melatonin were 1470.00 ± 30.00 and 1907.00 ± 38.00 g/bird, respectively. The mean weight gain in CL + MEL (345.00 ± 21.01 g) was significantly (P < 0.05) higher than those of the 12D/12L (244.99 ± 18.67 g) and CL (307.48 ± 18.14 g) groups. Feed consumptions were significantly (P < 0.05) different in all the groups. Group II, raised on CL without melatonin supplementation, had the highest feed consumption value of 25.14 ± 0.51 g/bird from day 14, and attained the peak value of 206.77 ± 7.82 g/bird at day 56. The highest overall amount of water intake was recorded in the melatonin-treated group. In conclusion, melatonin administration to broiler chickens enhanced water intake but decreased feed consumption with increase in LW during the hot-dry season.

The hindlimb of the grasscutter (Thryonomys swinderianus, Temminck-1827) was studied using 12 adult (≥6 months) rats of either sex with the mean weights of 1.42 ± 0.20 kg and 0.82 ± 0.13 kg for buck (n = 6) and doe (n = 6), respectively (p < .05). Characteristics of the bones were studied by gross observation after their preparation. Measurements of different segments of the hindlimb were also taken, giving a total average length of 31.80 cm. Correlation coefficients between length of bone and weight of rats were positive (p < .05) in bucks for all bone segments, except the pes (p > .05), whereas those in does were all non-significant (p > .05), with significant sexual dimorphism in the relationships. The average total number of bones in the hindlimb was 92, with no apparent sexual dimorphism. The bones of the hindlimb revealed important differences and similarities in morphology with those of other rodents. The Os coxae presented a relatively large obturator foramen, formed by the ramus of the ischium and shaft of the pubis. The femur had a body and two extremities with a prominent head, well-defined neck and trochanter. The fibula, a slender bone with triangular-shaped proximal extremity, ran down the length of tibia with proximal attachment (via cartilage) and distal fusion, leaving an extensive interosseus space. Experiments were conducted with the aim of determining the influence of melatonin administration on vigilance and tonic immobility (TI) responses of Marshall broiler chickens. The broiler chickens were reared on different lighting regimens and subjected to heat stress during the hot-dry season. Simple random sampling was used to assign 300 broiler chicks into three groups, comprising 100 broiler chicks each. Group I (12D:12L cycle) was raised under natural photoperiod of 12-h light and 12-h darkness, without melatonin supplementation. Group II (CL) was kept under 24-h continuous lighting, without melatonin administration. Group III (CL+MEL) was raised under 24-h continuous lighting; with melatonin supplementation at 0.5mg/kg per os, via drinking water using a syringe. Beginning from day-old, broiler chickens in group III were individually administered with melatonin once daily for 8weeks at 17:00h. TI was induced by manual restraint, and vigilance elicited at self-righting graded for three days, two weeks apart, in 15 labeled broiler chickens from each of the three groups; at 06:00h, 13:00h and 18:00h, starting from week 4-8. Each broiler chicken was laid on its back in a U-shaped cradle, covered with cloth. Thermal microenvironment parameters of dry bulb temperature (DBT) and relative humidity (RH) were recorded at the experimental site, concurrently during the vigilance and TI tests. Inside the broiler chickens' house, the weekly temperature-humidity index (THI) was lowest at week 4 of the study, with the value of 48.60±0.08°C. At week 4, the relationship between the THI and TI induction attempts was stronger in 12D:12L cycle (r=0.589, P<0.001) than CL (r=0.264, P>0.05) or CL+MEL (r=0.096, P>0.05) broiler chickens. This indicated that the broiler chickens on 12D:12L cycle were more active compared to their melatonin-treated counterparts, apparently due to adverse effects of high DBT and high RH on the broiler chickens during the hot-dry season. The highest numbers of TI induction trial attempts were recorded at 13:00h in 12D:12L cycle and CL groups (2.13±0.34 and 2.15±0.22, respectively), when the broiler chickens were at week 8. The overall mean values of induction trial attempts differed significantly (P<0.0001) between the groups; with the lowest mean values of 1.22±0.4 recorded in CL+MEL broiler chickens. At day 42, the lowest mean TI duration of 101.87±10.24s in the CL group, recorded at 06:00h rose (P<0.001) to 184.07±23.69s at 13:00h. The overall mean duration of TI differed significantly (P<0.0001) again between the groups; with the highest mean duration of 167.82±8.35s, recorded in CL+MEL broiler chickens administered with melatonin. The overall mean vigilance behavioural ranking values of 1.85+0.07 and 1.70+0.08, obtained in 12D:12L cycle and CL broiler chickens, respectively were higher (P<0.0001) than the value of 1.44+0.05 recorded in melatonin-treated broiler chickens. The results indicated that broiler chickens belonging to both 12D:12L cycle and CL groups were more emotional, fearful or anxious, compared to CL+MEL broiler chickens. It was concluded that melatonin administration elicits boldness and confidence by suppressing freezing behaviour in broiler chickens, and it may improve their welfare and productivity., (Copyright © 2016 Elsevier Inc. Experiments were performed with the aim of determining the effect of melatonin administration on diurnal fluctuations in cloacal temperature (CT) of Marshall broiler chickens during the hot dry season. Birds in group I (12L:12D cycle) were raised under natural photoperiod of 12-h light and 12-h darkness, without melatonin supplementation, while those in group II (LL) were kept under 24-h continuous lighting, without melatonin administration. Broiler chickens in group III (LL + melatonin) were raised under 24-h continuous lighting, with melatonin supplementation at 0.5 mg/kg per os. The cloacal temperatures of 15 labeled broiler chickens from each group were measured at 6:00, 13:00, and 19:00 h, 7 days apart, from days 14-42. Temperature-humidity index was highest at day 14 of the study, with the value of 36.72 ± 0.82 °C but lowest at day 28 with the value of 30.91 ± 0.80 °C (P < 0.0001). The overall mean hourly cloacal temperature value of 41.51 ± 0.03 °C obtained in the 12L:12D cycle birds was significantly higher (P < 0.001) than the value of 41.16 ± 0.03 °C recorded in the melatonin-treated group but lower than that of 41.65 ± 0.03 °C obtained in the LL birds. Mortality due to hyperthermia commenced at day 28 in both 12L:12D cycle and LL broiler chickens but was delayed till day 42 in LL + MEL broiler chickens. In conclusion, melatonin administration alleviated the deleterious effects of heat stress on broiler chickens by maintaining their cloacal temperature at relatively low values.

A gross morphological study of the brain of the African giant pouched rat (Cricetomys gambianus Waterhouse, 1840) was undertaken in order to document its normal features and assess the structure-function paradigm. The study was conducted by direct observation of 29 adult African giant pouched rats' brains. In the telencephalon, the cerebral cortex was devoid of prominent gyri and sulci, but the large olfactory bulb and tract relaying impulses to the olfactory cortex were very prominent. The large size of the olfactory bulb correlated with the established sharp olfactory acuity of the rodent. In the mesencephalic tectum, the caudal colliculi were bigger than the rostral colliculi, indicating a more acute sense of hearing than sight. In the metencephalon, the cerebellar vermis, the flocculus and the paraflocculus were highly coiled and, thus, well developed. The myelencephalon revealed a better organised ventral surface than dorsal surface; the cuneate fascicle, the intermediate sulcus and the lateral sulcus were not evident on the dorsal surface, but there were clearly visible pyramids and olivary prominence on the ventral surface. In conclusion, the highly coiled cerebellar vermis, flocculus and paraflocculus, as well as the conspicuous pyramids and olivary prominence are indicative of a good motor coordination and balance in the African giant pouched rat.

The study was conducted to evaluate the sexual dimorphism in the morphometric parameters of the whole brain and the brainstem in adult captive African giant pouched rats. Twenty-nine brain samples, comprising 15 females and 14 males, were utilized. Following brain extraction by standard procedures, the mean values of the weights, lengths, diameters, and volumes of the intact brains and the brainstem structures were compared in male and female rats using quantitative analytical statistical methods. The absolute brain weight obtained in the male was significantly higher (p < 0.01) than that of the female while the relative brain weight obtained in the female (0.636 +/- 0.049%) was higher than that of the male rats (0.564 +/- 0.032%), although the difference in the values was not statistically significant (p > 0.05). The lengths of the brain and myelencephalon, as well as the weights of the myelencephalon and mesencephalon, did not differ between the two sexes (p > 0.05). The weights and lengths of the pons did not differ between the two sexes (p > 0.05). There was a significant difference (p < 0.05) in the pyramidal length and a highly significant difference (p < 0.01) in the pyramidal width between the two sexes, with the male rats having higher values. This pioneering sexual dimorphic brain morphometry provides information for further research.

The pathogenesis of sickle vaso-occlusive crisis (VOC) in sickle cell disease (SCD) patients involves the accumulation of rigid sickle cells and the stimulation of an ongoing inflammatory response, as well as the stress of infections. The immune response, via cytokine imbalances and deregulated T-cell subsets, also has been proposed to contribute to the development of VOC. In this study, a panel of high-sensitivity cytokine kits was used to investigate cytokines in the sera of SCD patients in VOC. The results were compared primarily with those for stable SCD patients and secondarily with those for normal healthy people who served as controls. The cytokines studied included interleukin-2 (IL-2), IL-4, and IL-10. Lymphocyte subsets of patients with VOC were also studied and were compared with those of both control groups (20 stable patients without crisis [SCD group] and 20 normal healthy controls [NHC]). The VOC group was notable for remarkably elevated levels of IL-4, among the three cytokines tested, compared with those for the SCD and NHC groups. Patients with VOC also differed from stable SCD patients and NHC by having notably lower IL-10 levels, as well as the lowest ratio of CD4(+) to CD8(+) T cells (0.7). The patterns of the proinflammatory cytokine IL-2 did not differ between VOC and stable SCD patients, but NHC had significantly lower IL-2 levels than both the VOC and SCD groups. Our results demonstrate coexisting levels, both high and low, of TH1- and TH2-type cytokines, as well as diminished levels of T-cell subsets in VOC. These results are discussed in an effort to better understand the importance of the immune system profile in the pathogenesis of sickle cell VOC. Since the possibility that a cytokine imbalance is implicated in the pathogenesis of sickle cell crisis has been raised, our results should prompt further investigation of the host immune response in terms of TH1 and TH2 balance in sickle cell crisis.

Bovine fetal lung tissue was examined histochemically, using alpha-naphthyl butyrate (nonspecific method), to detect the presence of esterase-positive pulmonary alveolar macrophages (PAM). Positively stained PAM were first seen in the alveolar septa and lumina of the lungs of 4 of 8 fetuses 240 days old. Macrophages (n = 100) in 240-day-old fetuses were 6.74 +/- 0.07 microm, and in most cells, esterase-positive granules were sparsely distributed in the cytoplasm. In 10 fetuses 250 days old, the alveolar septa and lumina of lungs contained positively stained macrophages, as did those of all older fetal lungs examined. Compared with macrophages of the 240-day-old fetuses, those (n = 50) of 250-day-old fetuses were significantly larger, 7.32 +/- 0.10 micron (P less than 0.0001), and esterase-positive granules were heavily distributed throughout the cytoplasm. In all fetal lungs, T lymphocytes contained a single, small, esterase-positive granule, compared with the numerous diffuse esterase-positive granules in PAM. Type-II pneumonocytes with well-developed lamellar bodies had no evidence of positive nonspecific esterase reaction in lungs.

The breathing patterns of 20 anesthetized chickens were studied during progressive suppression of intrapulmonary chemoreceptors (IPC) by various concentrations of CO2 and following bilateral vagotomy. The vagotomy breathing pattern, characterized by a marked accentuation of expiratory times with prolonged expiratory pauses, was markedly different from that induced by CO2 inhalation. Removal of neural input to the central respiratory centers from IPC does not appear to be solely responsible for the altered breathing pattern following vagotomy in birds.

Seventy-five pairs of ovaries from Yankasa Sheep aged 1 1 2 to 3 years were collected from a Zaria abattoir for 12 months, to study the ovarian activity. It was observed that ovarian activity in Yankasa sheep was year-round with maximum activity occurring during the rainy and pre-rain seasons. These periods of maximum ovarian activity might be caused by the availability of pasture during the rainy and pre-rain seasons, since Nigerian sheep obtain most of their food by grazing available pasture.

Ovarian activity of Sokoto red goats was studied for 12 months using 141 pairs of ovaries obtained from a Zaria abattoir. Ovarian activity occurred throughout the year but higher activity was observed during the rainy (June to September) and dry (January to March) seasons. The higher ovarian activity during the rainy season is perhaps due to the availability of pasture during this time, while the higher activity during the dry season might be related to the goat's ability to convert leaves and dry forage that are mostly available during this season into useful nutrients. Temperature increase, particularly during the dry season, might be a factor for higher ovarian activity during the same period.
