This environmental assessment focuses on the coastal sediments of the Al-Khafji area in the Saudi Arabian Gulf, with an analysis of the human health risks posed by lead (Pb) and cadmium (Cd) contamination. Single and integrated indices were used to detect contamination and evaluate these metals' non-carcinogenic and carcinogenic impacts on adults and children through ingestion, dermal contact, and inhalation pathways. Sediment quality guidelines and contamination indices indicated the absence of significant contamination levels. The moderate contamination observed in scattered samples did not imply adverse biological effects due to the presence of these two metals in Al-Khafji sediments. The average values of the chronic daily intake (CDI) for both Pb and Cd were higher in children than adults across all three pathways, with ratios of 9.4, 4.7, and 4.7 folds, respectively. The hazard index (HI) values for Pb and Cd were below 1, confirming that the sediments of Al-Khafji are considered acceptable and safe in terms of these potentially toxic elements (PTEs). The average lifetime cancer risk (LCR) values for Pb and Cd were higher in children compared to adults, with ratios of 9.3 and 9.4 folds, respectively. However, all detected LCR levels do not represent a potential carcinogenic health hazard. Nevertheless, a regular monitoring program aimed at detecting early signals of environmental health depletion is recommended.

Introduction The use of e-cigarettes (ECs) has reached unprecedented levels, due to a variety of reasons, including the misconception regarding their safety. Thus, there have been efforts to characterize the effects of EC exposure, including in the context of thirdhand EC (THEC) on a host of disorders, such as cardiovascular disease (CVD). Methods To address this issue, we sought to characterize the effects of THEC on platelet function and thrombus formation, using a novel mouse exposure protocol that resembles real life scenarios. To assess these effects, a host of related in vivo (i.e. tail bleeding time, and ferric chloride injury induced thrombosis model) assays and in vitro platelet specific (e.g. aggregation, and dense granule secretion) investigative assays were conducted. Results Our in vivo characterization demonstrated that THEC exposed mice exhibited a prothrombotic phenotype reflected by their shortened tail bleeding (THEC: 37 ± 15 seconds, versus clean air: 183 ± 56 s) and occlusion times (THEC: 188 ± 39 s, versus clean air: 519 ± 70 s), relative to those exposed to clean air. Importantly, we found no difference in the platelet counts between the THEC and clean air mice. As for the underlying mechanism, separate experiments revealed significantly enhanced platelet aggregation, dense and alpha granule secretion, as well as integrin/GPIIb-IIIa activation and phosphatidylserine exposure in response to thrombin and ADP agonist stimulation. Conclusions Taken together, these results provide evidence that THEC does have the capacity to increase the risk of thrombotic disease, which should increase awareness regarding its underappreciated negative health effects.

To assess the heavy metals concentration in the coastal sediments of the southern Kuwait coast, Fe, Mn, Cu, Pb, Ni, Co, Cd and Cr were measured by inductively coupled plasma mass spectroscopy. Whereas, the south of Kuwait coast is characterized by the presence of tourist resorts, and commercial and oil exports harbors. Moreover, environmental indicators were used to help in evaluating the degree and the intensity of pollutants in these sediments. Geoaccumulation index (Igeo) revealed that the sediments of hard all Hamara and Al-Khiran coasts are moderately polluted by Cu, while Ras Al-Zour and Ras Al-Jula'ia coasts are moderately polluted by Cd. Moreover, the enrichment factor (EF) indicated that the sediments of Hadd Al-Hamara coast are severely enriched with Ni, Cr and Pb, while the Al-Khiran coast is moderate severely enriched with the same metals. Ras Al-Zour and Ras Al-Jula'ia coasts are severely enriched with Ni and very severely enriched with Pb. Simultaneously, all studied sites are extremely severely enriched with Cu and Cd. These results were confirmed by the results of the contamination factor (CF) and the soil pollution index (SPI) indicated that Hadd Al-Hamara and Al-Khiran coasts are highly contaminated with Cu and Cd, while Ras Al-Zour and Ras Al-Jula'ia coasts are highly contaminated with Cd. Generally, the pollution load index showed that the sediments of all studied sites are no heavy metal pollution (PLI < 1). Pollutants might be originated from commercial wastes and construction activities.

Abstract The FDA-approved DNA hypomethylating agents (DHAs) like 5-azacytidine (5AC) and decitabine (DAC) demonstrate efficacy in the treatment of hematologic malignancies. Despite previous reports that showed histone acetylation changes upon using these agents, the exact mechanism underpinning these changes is unknown. In this study, we investigated the relative potency of the nucleoside analogs and non-nucleoside analogs DHAs on DNA methylation reversal using DNA pyrosequencing. Additionally, we screened their effect on the enzymatic activity of the histone deacetylase sirtuin family (SIRT1, SIRT2, SIRT3, SIRT5 and SIRT6) using both recombinant enzymes and nuclear lysates from leukemia cells. The nucleoside analogs (DAC, 5AC and zebularine) were the most potent DHAs and increased the enzymatic activity of SIRT6 without showing any significant increase in other sirtuin isoforms. ChIP-Seq analysis of bone marrow cells derived from six acute myeloid leukemia (AML) patients and treated with the nucleoside analog DAC induced genome-wide acetylation changes in H3K9, the physiological substrate for SIRT6. Data pooling from the six patients showed significant acetylation changes in 187 gene loci at different chromosomal regions including promoters, coding exons, introns and distal intergenic regions. Signaling pathway analysis showed that H3K9 acetylation changes are linked to AML-relevant signaling pathways like EGF/EGFR and Wnt/Hedgehog/Notch. To our knowledge, this is the first report to identify the nucleoside analogs DHAs as activators of SIRT6. Our findings provide a rationale against the combination of the nucleoside analogs DHAs with SIRT6 inhibitors or chemotherapeutic agents in AML due to the role of SIRT6 in maintaining genome integrity and DNA repair.

Introduction Thirdhand smoke (THS) is associated with many public health and disease concerns, such as respiratory illness, cancer, lipidemia, and cardiovascular disease (CVD). We have previously shown that a moderate to long-term exposure to THS increases the risk of thrombosis. However, whether short-term exposure to THS would produce any effects remains to be discovered. Therefore, this study investigated the impact of 1-month THS exposure on platelet function, in vivo and in vitro, and on cytokine response, in a sex-dependent manner. Aims and Methods Secondhand smoke or clean air (CA) exposed upholstery materials for 1 week were kept in cages housed with 5–6 mice, and the procedure was repeated for 4 weeks. These THS-exposed mice were evaluated for thrombogenesis and platelet function assays. In addition, cytokines expression was evaluated from pooled serum. Results Compared to the CA group, THS exposure significantly shortened the tail bleeding time and carotid artery thrombus formation. Moreover, the female mice appeared more sensitive to THS exposure than males. Furthermore, platelet aggregation, dense granule secretion, and P-selectin activation markers were significantly elevated due to THS exposure. In addition, high-throughput screening showed at least 30 cytokines differentially modulated by THS in females relative to 26 in male mice. Conclusions Collectively, these results demonstrate that 1 month of THS exposure represents a high health risk, in part, by triggering a prothrombotic phenotype that appears to be more significant in females, who are at a much higher risk for occlusive CVD. Additionally, changes in cytokine levels mediate some of the THS-induced occlusive effects. Implications This study revealed that THS exposure for 1 month is detrimental to the cardiovascular health of both sexes; however, females could be more aggressively affected than males. The present study is an integral part of our ongoing investigation on the snapper fish (Family: Lutjanidae) and their distribution between the Red Sea, Egypt, and the Arabian Gulf, Qatar. In 2022, the blackspot snapper Lutjanus ehrenbergii was collected from the catch landings of the two countries concurrently, and it was identified using a previously employed DNA barcoding technique. A total of 221 and 210 specimens of L. ehrenbergii collected from the catch landings of the Red Sea, Egypt, and the Arabian Gulf in Qatar, respectively, were employed for the purpose of the current study. Fish were dissected to determine their sex after measuring the total length (TL) for each specimen to the nearest 0.1cm and calculating the total weight to the nearest 0.1g. Ages were calculated through the reading of otolith growth rings. For L. ehrenbergii from the Red Sea, the results of the von Bertalanffy growth parameters were L∞ = 35.6cm, K = 0.32y -1, and to = - 1.30 year for L. ehrenbergii from the Red Sea, and L∞ = 34.8cm, K = 0.32y -1, and to = - 1.40 year for L. ehrenbergii from the Arabian Gulf. Otolith growth rings revealed a maximum lifespan of 4 years. Kc values for L. ehrenbergii samples from the Arabian Gulf ranged from 1.30 to 1.47, whereas samples from the Red Sea exhibited values between 1.28 and 1.48. L. ehrenbergii exhibits a negative allometric growth, reflected by the weight of pooled sexes increasing in proportion to less than the cube of their body length (b= 2.926 to 2.942 in fish from the Red Sea and 2.876 to 2.881 in fish from the Arabian Gulf. The current study indicates that geographical or sex-specific variations in growth should be considered upon assessing the significance of growth studies for managing fish stocks. Electrochemical Impedance Spectroscopy (EIS) is a powerful, rapid and accurate non-destructive method for the evaluation of wide range of materials. In this work, the corrosion behavior of some austenitic stainless steels was studied in 3.5%NaCl. The effect of Nb content and cold deformation on the corrosion resistance was investigated. The occurrence of localized corrosion due to chloride ion attack was examined by visual inspection and SEM-EDS. XRD analysis was used to identify the chemical composition of the outer oxide layer. According to EIS, the corrosion resistance increased by increasing the Nb content. Cold deformation (CD) has a critical effect on the corrosion resistance. The specimens prepared at 23% CD showed the best corrosion resistance. The surface resistance increased double compared with that without CD. Increasing the deformation to 40% and 50% affects negatively the corrosion resistance. XRD reveled presence of Cr-rich film in the specimen bearing the highest Nb-content. It is argued that increasing the Nb-content enhancing formation of surface film enriched with Cr and Ni which has a high corrosion resistance.
