4. Online Local Natural Hazards Education for Young Adults: Assessing Program Efficacy and Changes in Risk Perception for Texas Natural Hazards

Abstract

Risk perceptions can influence how people prepare, react, and respond to a natural hazard. For dependent populations with increased vulnerability, such as youth, adolescents, and young adults, hazard education programs can influence their risk perceptions and increase their hazard awareness. However, most school curriculum does not include in-depth formal natural hazards education; Texas curriculum in secondary schools, for example, does not typically cover local natural hazards or their impacts in-depth due to other curriculum priorities. Thus, students may not be receiving potentially life-saving information on their local natural hazards or how to plan for or respond to disasters. In response, this research presents a formal, online, and youth-centric natural hazard educational (NHE) curriculum that examines the curriculum's effects on risk perception and subject matter proficiency using a local pilot study with college students as proxies for high school adolescents in Texas. Results suggest that the curriculum content improved overall natural hazards knowledge in participants (p < 0.01) and that participants with higher post-curriculum scores demonstrated higher risk perception and hazard awareness. These findings demonstrate how exposure to natural hazards educational programs can increase hazard awareness and coping capacity in young adults and adolescents.

Abstract

With its direct correspondence to electronic structure, angle-resolved photoemission spectroscopy (ARPES) is a ubiquitous tool for the study of solids. When extended to the temporal domain, time-resolved (TR)-ARPES offers the potential to move beyond equilibrium properties, exploring both the unoccupied electronic structure as well as its dynamical response under ultrafast perturbation. Historically, ultrafast extreme ultraviolet (XUV) sources employing high-order harmonic generation (HHG) have required compromises that make it challenging to achieve a high energy resolution - which is highly desirable for many TR-ARPES studies - while producing high photon energies and a high photon flux. We address this challenge by performing HHG inside a femtosecond enhancement cavity (fsEC), realizing a practical source for TR-ARPES that achieves a flux of over 10$^{11}$ photons/s delivered to the sample, operates over a range of 8-40 eV with a repetition rate of 60 MHz. This source enables TR-ARPES studies with a temporal and energy resolution of 190 fs and 22 meV, respectively. To characterize the system, we perform ARPES measurements of polycrystalline Au and MoTe$_2$, as well as TR-ARPES studies on graphite., Comment: 11 pages, 5 figures

Abstract

The interplay between spin and orbital angular momentum in the up-conversion process allows us to control the macroscopic wave front of high harmonics by manipulating the microscopic polarizations of the driving field. We demonstrate control of orbital angular momentum in high harmonic generation from both solid and gas phase targets using the selection rules of spin angular momentum. The gas phase harmonics extend the control of angular momentum to extreme-ultraviolet wavelength. We also propose a bi-color scheme to produce spectrally separated extreme-ultraviolet radiation carrying orbital angular momentum.

9. A Gauge Invariant Formulation of Interband and Intraband Currents in Solids

Abstract

Experiments and simulations in solid-state high harmonic generation often make use of the distinction between interband and intraband currents. These two contributions to the total current have been associated with qualitatively different processes, as well as physically measurable signatures, for example in the spectral phase of harmonic emission. However, it was recently argued [P. F\"oldi, Phys. Rev. B 96, 035112 (2017)] that these quantities can depend on the gauge employed in calculations. Since physical quantities are expected to have gauge-independent values, this raises the question of whether the decomposition of the total current into interband and intraband contributions is physically meaningful, or merely a feature of a particular mathematical representation of nature. In this article, we explore this apparent ambiguity. We show that a closely related issue arises when calculating instantaneous band populations. In both the case of inter/intraband currents and in the case of instantaneous band populations, we propose definitions which are gauge-invariant, and thus allow these quantities to be calculated consistently in any gauge., Comment: 10 pages, 6 figures

Abstract

The objective of this study was to compare the effectiveness of a second tumor necrosis factor inhibitor (TNFi) versus a non‐TNFi biologic following discontinuation of a TNFi for patients with polyarticular‐course juvenile idiopathic arthritis (pJIA). Using the Childhood Arthritis and Rheumatology Research Alliance Registry, patients with pJIA who started receiving a second biologic following a first TNFi were identified. Patients were required to have no active uveitis on the index date and a visit six months after the index date. Outcome measures included Clinical Juvenile Arthritis Disease Activity Score with a maximum of 10 active joints (cJADAS10), cJADAS10 inactive disease (ID; ≤2.5) and cJADAS10 minimal disease activity (MiDA; ≤5). Multiple imputation was used to account for missing data. Adjusted odds ratios (aORs) were calculated using propensity score quintiles to compare outcomes at six months following second biologic initiation. There were 216 patients included, 84% initially received etanercept, and most patients stopped receiving it because of its ineffectiveness (74%). A total of 183 (85%) started receiving a second TNFi, and 33 (15%) started receiving a non‐TNFi. Adalimumab was the most common second biologic received (71% overall, 84% of second TNFi), and tocilizumab was the most common non‐TNFi second biologic received (9% overall, 58% of non‐TNFi). There was no difference between receiving TNFi versus non‐TNFi in cJADAS10 ID (29% vs 25%; aOR 1.23, 95% confidence interval [CI] 0.47–3.20) or at least MiDA (43% vs 39%; aOR 1.11, 95% CI 0.47–2.62) at six months. Most patients with pJIA started receiving TNFi rather than non‐TNFi as their second biologic, and there were no differences in disease activity at six months.

Abstract

Pre-release training is becoming a standard practice in conservation breeding and translocation programs for mammals and birds, but is still relatively neglected for herpetofauna, likely stemming from widespread beliefs that amphibians are “hardwired” and are thus predicted to benefit little from experiential learning. However, experience during development can drive both morphological and behavioral modifications that could benefit post-translocation survival. Here, we developed an antipredator training program for the endangered mountain yellow-legged frog Rana muscosa and evaluated its impact on morphometrics, developmental rates, behavior, and post-release outcomes. Using a controlled, balanced factorial experimental design, we exposed individuals at two developmental stages (tadpoles and postmetamorphic) to visual and olfactory cues from one of its principal predators, the two-striped garter snake Thamnophis hammondii. We found that exposure to predators during the tadpole stage impacted tadpole development, morphology, and behavior. However, we found no evidence that post-metamorphic behavior or postrelease outcomes were influenced by tadpole predator exposure, either on its own or in interaction with post-metamorphic training treatments. Training of postmetamorphic frogs, however, yielded evidence of learning, with downstream effects on behavior and apparent survival after release into the wild. Our findings produce novel insights on the value of anti-predator training in amphibian conservation translocation programs, bolster recent evidence re-evaluating the importance of learning and developmental experience for amphibian species, and provide guidance for selecting optimal developmental windows for training. 23. Differences in susceptibility to tumor necrosis factor alpha-induced apoptosis among MCF-7 breast cancer cell variants.

Abstract

Widespread use of MCF-7 human breast carcinoma cells as a model system for breast cancer has led to variations in these cells between different laboratories. Although several reports have addressed these differences in terms of proliferation and estrogenic response, variations in sensitivity to apoptosis have not yet been described. Tumor necrosis factor alpha (TNF-alpha) has been shown to both induce apoptosis and inhibit proliferation in MCF-7 cells. We observed that TNF-alpha inhibited proliferation in MCF-7 cell variants from three different laboratories (designated M, L, and N). MCF-7 M cells were resistant to TNF-alpha-induced apoptosis, whereas MCF-7 L cells were moderately resistant to the effect of TNF-alpha. A third variant, MCF-7 N, underwent apoptosis when exposed to TNF-alpha. Analysis of the p55 TNF-alpha receptor (TNFR) 1 expression revealed the greatest expression in MCF-7 N cells, whereas the MCF-7 L and M cells expressed 89 and 67% of MCF-7 N cell TNFR1 levels, respectively. Ceramide generation occurred in all three variants in response to TNF-alpha treatment, with MCF-7 N cells expressing the greatest increase. Cleavage of the CPP32/caspase 3 substrate poly(ADP-ribose) was observed in MCF-7 N and L cells as early as 3 and 6 h, respectively, but poly(ADP-ribose) cleavage was not observed in MCF-7 M cells. The delayed protease activation in the L variant may represent the mechanism by which these cells display delayed sensitivity to TNF-a-induced apoptosis. Expression of the Bcl-2, Mcl-1, Bcl-X, Bax, and Bak proteins was analyzed to determine whether the differences in MCF-7 cell sensitivity to apoptosis could be correlated to the differential expression of these proteins. Whereas Bak, Bcl-X, and Mcl-1 levels were identical between variants, the levels of Bcl-2 were 3.5-3.8-fold higher and the levels of Bax were 1.5-1.7-fold lower in the resistant variants (M and L) as compared with those of the sensitive variant (N). Taken together, these results suggest that differences in susceptibility to TNF-alpha-induced apoptosis among MCF-7 breast cancer cell variants may be explained by differences in TNFR expression, ceramide generation, differential expression of the Bcl-2 family of proteins, and protease activation.

Abstract

Risk perceptions can influence how people prepare, react, and respond to a natural hazard. For dependent populations with increased vulnerability, such as youth, adolescents, and young adults, hazard education programs can influence their risk perceptions and increase their hazard awareness. However, most school curriculum does not include in-depth formal natural hazards education; Texas curriculum in secondary schools, for example, does not typically cover local natural hazards or their impacts in-depth due to other curriculum priorities. Thus, students may not be receiving potentially life-saving information on their local natural hazards or how to plan for or respond to disasters. In response, this research presents a formal, online, and youth-centric natural hazard educational (NHE) curriculum that examines the curriculum's effects on risk perception and subject matter proficiency using a local pilot study with college students as proxies for high school adolescents in Texas. Results suggest that the curriculum content improved overall natural hazards knowledge in participants (p < 0.01) and that participants with higher post-curriculum scores demonstrated higher risk perception and hazard awareness. These findings demonstrate how exposure to natural hazards educational programs can increase hazard awareness and coping capacity in young adults and adolescents. [ABSTRACT FROM AUTHOR]

Abstract

Research about the Olympic Games has primarily focused on preparing athletes for competition. Less attention has been paid to the post-Olympic-phase (POP) and athlete well-being during this time. This study explored Australian Olympic athletes’ experiences following the conclusion of the 2016 Rio Olympic Games, including the factors that may have contributed to or challenged their well-being during this time. Eighteen athletes participated in semi-structured interviews and thematic analysis revealed that when Olympic performance appraisal met prior expectations, when athletes planned for a return to work or study, and when support from a variety of sources was readily available, this positively influenced athletes’ well-being during the POP. When these factors were not in place, more challenging post-Games experiences were present, and well-being was compromised. The findings contribute to the broader literature on elite athlete well-being and at an applied level, may be used to inform targeted programs that focus on supporting athletes after an Olympic campaign.

46. First-in-human clinical trial to assess the safety, tolerability and pharmacokinetics of P218, a novel candidate for malaria chemoprotection

Abstract

AIMS: This first-in-human clinical trial of P218, a novel dihydrofolate reductase inhibitor antimalarial candidate, assessed safety, tolerability, pharmacokinetics and food effects in healthy subjects. METHODS: The study consisted of two parts. Part A was a double-blind, randomized, placebo-controlled, parallel group, ascending dose study comprising seven fasted cohorts. Eight subjects/cohort were randomized (3:1) to receive either a single oral dose of P218 (10, 30, 100, 250, 500, 750 and 1000 mg) or placebo. Part B was an open-label, cross-over, fed/fasted cohort (eight subjects) that received a 250 mg single dose of P218 in two treatment periods. RESULTS: P218 was generally well tolerated across all doses; 21 treatment-emergent adverse events occurred in 15/64 subjects. Nine adverse events in five subjects, all of mild intensity, were judged drug related. No clinically relevant abnormalities in ECG, vital signs or laboratory tests changes were observed. P218 was rapidly absorbed, with Cmax achieved between 0.5 and 2 hours post dose. Plasma concentrations declined bi-exponentially with half-life values ranging from 3.1 to 6.7 hours (10 and 30 mg), increasing up to 8.9 to 19.6 hours (doses up to 1000 mg). Exposure values increased dose-proportionally between 100 and 1000 mg for P218 (parent) and three primary metabolites (P218 β-acyl glucuronide, P218-OH and P218-OH β-acyl glucuronide). Co-administration of P218 with food reduced Cmax by 35% and delayed absorption by 1 hour, with no significant impact on AUC. CONCLUSION: P218 displayed favourable safety, tolerability and pharmacokinetics. In view of its short half-life, a long-acting formulation will be needed for malaria chemoprotection.

Abstract

A comprehensive analysis of both the molecular genetic and phenotypic responses of any organism to the space flight environment has never been accomplished because of significant technological and logistical hurdles. Moreover, the effects of space flight on microbial pathogenicity and associated infectious disease risks have not been studied. The bacterial pathogen Salmonella typhimurium was grown aboard Space Shuttle mission STS-115 and compared with identical ground control cultures. Global microarray and proteomic analyses revealed that 167 transcripts and 73 proteins changed expression with the conserved RNA-binding protein Hfq identified as a likely global regulator involved in the response to this environment. Hfq involvement was confirmed with a ground-based microgravity culture model. Space flight samples exhibited enhanced virulence in a murine infection model and extracellular matrix accumulation consistent with a biofilm. Abstract

Background: While research into Olympic Athletes’ career transitions and retirement has led to a deeper understanding of important factors for athletes in this context, considerably less is known about the experiences of athletes in the immediate phase following an Olympic Games. Objectives: The purpose of this research was to investigate Australian Olympic athletes’ experiences during the period of time immediately following the conclusion of the 2016 Rio Olympic Games. This involved investigating specific factors that influenced athletes’ post-games realities of success, failure, and return to everyday life. Design: Qualitative-inductive Methods: Eighteen Australian Rio Olympic Games athletes (Female N=9, Male N=9) from a variety of team and individual sports participated in semi-structured interviews that explored their post-Olympic Games experiences. Thematic analysis was used to inductively analyse the data. Results: In the period immediately following the Rio Olympic campaign, many athletes felt a sense of relief before coming to terms with a post-Olympic ‘come down’. While positive and negative transitions back to reality following the Rio Games were influenced by performance expectations, positive transitions generally occurred when athletes had made plans for the post-Games phase and received strong support from family, teammates, and sport governing bodies. Conversely, negative experiences tended to occur where funding ceased, coach-athlete relationships fell apart, or team structures were dissolved following the Olympic event. Conclusions: Overall, athletes had a variety of experiences during the post-Olympic period and as such, it is critical to consider their needs individually. The findings of this project have implications at the micro (athlete, coach) and macro (National Sport Organisation) levels that could be used to better inform the targeted development of post-Olympic programs.
