17. Mixed‐charge cellulose nanocrystal modified cellulose acetate membrane with endotoxin scavenging ability and antibacterial properties.

Abstract

Blending modification is used to improve the hemocompatibility of cellulose acetate (CA) dialysis membranes while endowing the CA membrane with endotoxin scavenging and antibacterial capabilities. First, polydopamine (PDA) and polyethylenimine (PEI) are used as negatively charged and positively charged moieties, respectively, together with cellulose nanocrystals (CNCs) to construct mixed charge CNCs (PEI‐PDA@CNCs). Then PEI‐PDA@CNCs is blended with the CA casting solution, and the modified CA dialysis membrane is prepared through a phase inversion process. Through biocompatibility, adsorption, antibacterial, and filtration experiments, various properties of PEI‐PDA@CNCs modified CA membrane (PDCA) are systematically tested. The results show that the PDCA membrane exhibited good blood compatibility and noncytotoxicity. At the same time, the PDCA membrane has antibacterial properties and excellent endotoxin adsorption capacity. Compared with the CA membrane, the clearance rates of PDCA membrane for Staphylococcus aureus and Escherichia coli reach 54.0% ± 1.5% and 52.9% ± 5.9% respectively. In the dynamic adsorption experiment, the adsorption capacity of PDCA membrane for endotoxin increases to 2322.1 ± 45.9 EU/g. The research results indicate that PEI‐PDA@CNC‐modified multifunctional CA membrane with endotoxin adsorption and antibacterial properties has potential application prospects in the fields of hemodialysis and blood purification. 24. Photopolymerizable and Antibacterial Hydrogels Loaded with Metabolites from Lacticaseibacillus rhamnosus GGfor Infected Wound Healing

Abstract

In response to increasing antibiotic resistance and the pressing demand for safer infected wound care, probiotics have emerged as promising bioactive agents. To address the challenges associated with the safe and efficient application of probiotics, this study successfully loaded metabolites from Lacticaseibacillus rhamnosus GG(LGG) into a gelatin cross-linked macromolecular network by an in situ blending and photopolymerization method. The obtained LM-GelMA possesses injectability and autonomous healing capabilities. Importantly, the incorporation of LGG metabolites endows LM-GelMA with excellent antibacterial properties against Staphylococcus aureusand Escherichia coli, while maintaining good biocompatibility. In vivo assessments revealed that LM-GelMA can accelerate wound healing by mitigating infections induced by pathogenic bacteria. This is accompanied by a reduction in the expression of key proinflammatory cytokines such as TNF-α, IL-6, VEGFR2, and TGF-β, leading to increased re-epithelialization and collagen formation. Moreover, microbiological analysis confirmed that LM-GelMA can modulate the abundance of beneficial wound microbiota at family and genus levels. This study provides a facile strategy and insights into the functional design of hydrogels from the perspective of wound microenvironment regulation.

28. Data-driven discovery of the dimensions characterizing emotion experience

Abstract

The current study will probe the quantitative structure of emotions using large-scale online testing. We will present various short narratives and short video clips designed to evoke a wide range of real-world emotions and collect ratings on a rich set of affective scales to quantitatively characterize individual emotion experiences and discover low-dimensional space of emotions in a data-driven manner. Further, utilizing comprehensive psychological measures collected from participants, we will explore individual differences in emotion experiences.

Abstract

This project is conducted by the COVID-DYNAMIC Team (http://conte.caltech.edu/content/covid-dynamic-investigator-team) The COVID-19 pandemic is confronting people with significant changes in affectively-laden media exposure as well as in social behavior. To quantify longitudinal effects that could inform public health policy, we will collect anonymized data over the internet on subject's implicit and explicit emotional biases, and characterize how these might relate to social, economic, moral and political judgments and decision-making. We will sample approximately 1000 English-speaking adult participants in the US over the internet anonymously, recruited through Prolific.co. We will also sample an extant registry of approximately 100 community-dwelling adults in Los Angeles that are part of the Caltech Conte Center on the Neurobiology of Social Decision-Making (http://conte.caltech.edu/content/covid-dynamic). We plan to test participants longitudinally for approximately one hour, up to one year. We expect that the frequency of testing will vary depending on the development of the pandemic, from weekly to monthly. Subject recruitment and testing began on April 5, 2020, with additional waves re-tested in the subsequent two weeks, and data collection ongoing. At this point, we have only analyzed the data from these three waves for quality control and instrument design purposes, but have not analyzed any data for any hypothesis testing. This preregistration includes methods of participant recruitment, sample size, inclusion and exclusion criteria, and general study procedures. Subsequent preregistrations will provide updates and cover specific studies that we will conduct using the data collected with the procedures described here.

38. Emotion Experience from Stories, Videos and Everyday Life: Structure and Individual Differences

Abstract

Most studies of emotion have as their subject matter the emotion experiences that people can describe and rate. By contrast to this approach from psychology, studies in animals, and some biological studies in humans, focus on behavior and its adaptive function. These two literatures typically use very different corresponding features by which to characterize emotion: categories or dimensions describing feelings for which we have convenient words, for the former (e.g., happiness, pleasantness), and functional properties for the latter (e.g., persistence, generalizability, approachabil- ity). In this thesis I use both sets of ratings, and I ask whether the latter, biologically inspired features could also be used to characterize people’s emotion experiences, and might reveal novel dimensions of variability. They also typically use different sets of stimuli to induce the emotions: lexical stimuli in which participants are asked to imagine something hypothetical are common in human studies; ecologically valid stimuli that at least the subjects cannot distinguish from the real world are common in animal studies. Here I used three domains of stimuli: stories, videos, and real-life experiences, in the same set of participants, permitting a unique comparison. I took advantage of a sample of approximately 1000 Americans who were surveyed longitudinally over the internet during the COVID-19 pandemic. I collected ratings of emotion experiences evoked by three classes of stimuli: a validated set of short stories, a validated set of short videos, and actual experiences in real life across multiple waves. I found that all three types of emotion experiences could be characterized by low dimensional spaces, with the first two factors that accounted for most of the variance in people’s ratings corresponding to the dimensions of valence and arousal, in line with prior work.
