Abstract Background The gut microbiota significantly influences the health and growth of red-spotted grouper (Epinephelus akaara), a well-known commercial marine fish from Fujian Province in southern China. However, variations in survival strategies and seasons can impact the stability of gut microbiota data, rendering it inaccurate in reflecting the state of gut microbiota. Which impedes the effective enhancement of aquaculture health through a nuanced understanding of gut microbiota. Inspired by this, we conducted a comprehensive analysis of the gut microbiota of wild and captive E. akaara in four seasons. Results Seventy-two E. akaara samples were collected from wild and captive populations in Dongshan city, during four different seasons. Four sections of the gut were collected to obtain comprehensive information on the gut microbial composition and sequenced using 16S rRNA next-generation Illumina MiSeq. We observed the highest gut microbial diversity in both captive and wild E. akaara during the winter season, and identified strong correlations with water temperature using Mantel analysis. Compared to wild E. akaara, we found a more complex microbial network in captive E. akaara, as evidenced by increased abundance of Bacillaceae, Moraxellaceae and Enterobacteriaceae. In contrast, Vibrionaceae, Clostridiaceae, Flavobacteriaceae and Rhodobacteraceae were found to be more active in wild E. akaara. However, some core microorganisms, such as Firmicutes and Photobacterium, showed similar distribution patterns in both wild and captive groups. Moreover, we found the common community composition and distribution characteristics of top 10 core microbes from foregut to hindgut in E. akaara. Conclusions Collectively, the study provides relatively more comprehensive description of the gut microbiota in E. akaara, taking into account survival strategies and temporal dimensions, which yields valuable insights into the gut microbiota of E. akaara and provides a valuable reference to its aquaculture.

Lipogenesis is often highly upregulated in breast cancer brain metastases to adapt to intracranial low lipid microenvironments. Lipase inhibitors hold therapeutic potential but their intra-tumoral distribution is often blocked by the blood‒tumor barrier (BTB). BTB activates its Wnt signaling to maintain barrier properties, e.g., Mfsd2a-mediated BTB low transcytosis. Here, we reported VCAM-1-targeting nano-wogonin (W@V-NPs) as an adjuvant of nano-orlistat (O@V-NPs) to intensify drug delivery and inhibit lipogenesis of brain metastases. W@V-NPs were proven to be able to inactivate BTB Wnt signaling, downregulate BTB Mfsd2a, accelerate BTB vesicular transport, and enhance tumor accumulation of O@V-NPs. With the ability to specifically kill cancer cells in a lipid-deprived environment with IC50 at 48 ng/mL, W@V-NPs plus O@V-NPs inhibited the progression of brain metastases with prolonged survival of model mice. The combination did not induce brain edema, cognitive impairment, and systemic toxicity in healthy mice. Targeting Wnt signaling could safely modulate the BTB to improve drug delivery and metabolic therapy against brain metastases.

Abstract Purpose The study was designed to investigate the occurrence and risk factors of malnutrition in diabetic foot ulcers (DFU) patients and examine the association between malnutrition and length of stay (LOS). Methods This observational study included DFU hospitalized patients in two campuses of a hospital from January 2021 to June 2023. The diagnosis standard of malnutrition was established by using the Global Leadership Initiative on Malnutrition (GLIM) criteria. Patients were followed up to ascertain the length of hospitalization, and hospital stays longer than 17 days were considered as prolonged LOS. To explore the risk factors of malnutrition and the association between malnutrition and LOS, univariate and multivariate logistic regression analyses were performed. Results Overall 219 DFU patients were enrolled, malnutrition was identified in 38.36% of patients according to GLIM criteria, and 92 patients (42%) were recognized as prolonged LOS. Eremurus was described at the beginning of the 19th century. However, due to limited sampling and the small number of gene markers to date, its phylogeny and evolution are largely unknown. In this study, we analyzed plastomes from 27 species belonging to 2 subgenera and 3 sections of Eremurus, which are found in Central Asia (its center of diversity) and China. We also analyzed nuclear DNA ITS of 33 species, encompassing all subgenera and sections of the genus in Central Asia, southwest Asia and China. Our findings revealed that the genus was monophyletic, although both subgenera Eremurus and Henningia were found to be paraphyletic. Both plastome and nrDNA-based phylogenetic trees had three clades that did not reflect the current taxonomy of the genus. Our biogeographical and time-calibrated trees suggest that Eremurus originated in the ancient Tethyan area in the second half of the Eocene. Diversification of Eremurus occurred from the early Oligocene to the late Miocene. Paratethys Sea retreat and several orogenetic events, such as the progressive uplift of the Qinghai-Tibet Plateau and surrounding mountain belts (Altai, Pamir, Tian Shan), caused serious topographic and climate (aridification) changes in Central Asia that may have triggered a split of clades and speciation. In this transformed Central Asia, speciation proceeded rapidly driven mainly by vicariance caused by numerous mountain chains and specialization to a variety of climatic, topographic and soil conditions that exist in this region.

Abstract Background Accurate species delimitation is fundamental for testing evolutionary theory and provides essential implications for conservation management. The arctic-alpine genus Saxifraga L. (Saxifragaceae) is taxonomically complex and many species have not been critically assessed. The taxonomic and phylogenetic status of Saxifraga lancangensis Y.Y.Qian, considered as a synonym of Saxifraga mengtzeana Engl. & Irmsch. in previous studies, is re-evaluated in light of new evidence presented here. Results Evidence from morphological comparison and sequencing of plastid genome indicate that S. lancangensis belongs to Saxifraga sect. Irregulares Haw., and is closely related to Saxifraga geifolia Balf.f., and S. mengtzeana. However, S. lancangensis can be diagnosed by its petals with red and clawless base, leaf blade orbicular and leaf margin shallowly dentate. Conclusions The morphological and molecular evidence support the resurrection of S. lancangensis as a distinct species. An updated morphological description based on protologue and fresh material, diagnostic characters, and original photographs of the resurrected species are presented.

Abstract Background Dactylicapnos is a climbing herbaceous vine, distributed from the Himalayas to southwestern China, and some of the species have important medicinal values. However, the chloroplast genomes of Dactylicapnos have never been investigated. In this study, chloroplast genomes of seven Dactylicapnos species covering all three sections and one informal group of Dactylicapnos were sequenced and assembled, and the detailed comparative analyses of the chloroplast genome structure were provided for the first time. Results The results showed that the chloroplast genomes of Dactylicapnos have a typical quadripartite structure with lengths from 172,344 bp to 176,370 bp, encoding a total of 133–140 genes, containing 88–94 protein-coding genes, 8 rRNAs and 37–39 tRNAs. 31 codons were identified as relative synonymous codon usage values greater than one in the chloroplast genome of Dactylicapnos genus based on 80 protein-coding genes. The results of the phylogenetic analysis showed that seven Dactylicapnos species can be divided into three main categories. Phylogenetic analysis revealed that seven species form three major clades which should be treated as three sections. Conclusions This study provides the initial report of the chloroplast genomes of Dactylicapnos, their structural variation, comparative genomic and phylogenetic analysis for the first time. The results provide important genetic information for development of medical resources, species identification, infrageneric classification and diversification of Dactylicapnos.

Flower constancy describes the phenomenon that pollinators tend to successively visit flowers of a single species during foraging, reducing reproductive interference in natural communities. The extent of flower constancy is largely determined by the floral traits of co-flowering species. Both higher inter-specific and lower intraspecific differences of floral traits should contribute to a higher level of flower constancy. However, previous studies mainly focused on interspecific difference, and the intraspecific variation (consistency) of floral traits received much less attention. We hypothesise that selection may favour lower intraspecific floral trait variation in communities composed of multiple co-flowering congeners. We investigated the floral colour variation of three focal Pedicularis species that share pollinators in 19 communities composed of either single or multiple Pedicularis species. Colour was quantified using image-based colour analysis as perceived by pollinators. We found that most of the intrapopulation floral colour variation was below the colour discrimination threshold of bumblebees, implying strongly constrained by the visual selection by pollinators. Contrary to the hypothesis, there is no significant difference in intraspecific floral colour variation between different community contexts. It may be due to the relatively large interspecific floral colour differences of most co-flowering species. The influence of community context on intraspecific variation may be reflected in floral traits other than colours.

Sexual systems play important roles in angiosperm evolution and exhibit substantial variations among different floras. Thus, studying their evolution in a whole flora is crucial for understanding the formation and maintenance of plant biodiversity and predicting its responses to environmental change. In this study, we determined the patterns of plant sexual systems and their associations with geographic elements and various life-history traits in dry-hot valley region of southwestern China, an extremely vulnerable ecosystem. Of the 3166 angiosperm species recorded in this area, 74.5% were hermaphroditic, 13.5% were monoecious and 12% were dioecious, showing a high incidence of diclinous species. Diclinous species were strongly associated with tropical elements, whereas hermaphroditic species were strongly associated with temperate and cosmopolitan elements. We also found that hermaphroditism was strongly associated with showy floral displays, specialist entomophily, dry fruits and herbaceous plants. Dioecy was strongly associated with inconspicuous, pale-colored flowers, generalist entomophily, fleshy fruits, and woody plants, whereas monoecy was strongly associated with inconspicuous, pale-colored flowers, anemophily, dry fruits, and herbaceous plants. In addition, hermaphroditic species with generalist entomophily tended to flower in the dry season, whereas diclinous species with specialist entomophily tended to flower in the rainy season. However, independent of sexual systems, plants that produce dry fruits tended to flower in the rainy season and set fruits in the dry season, but the opposite pattern was found for fleshy fruit-producing plants. Our results suggest that in the dry-hot valleys, plant sexual systems are associated with geographic elements as well as various life-history traits that are sensitive to environmental change.

When benefiting other beneficiaries, cushion plants may reciprocally receive feedback effects. The feedback effects on different sex morphs, however, remains unclear. In this study, taking the gynodioecious Arenaria polytrichiodes as a model species, we aimed to assess the sex-specific facilitation intensity of cushion plant by measuring the beneficiary cover ratio, and to assess the potential costs in cushion reproductive functions by measuring the flower and fruit cover ratios. The total beneficiary cover ratio was similar between females and hermaphrodites. Females produced much less flowers but more fruits than hermaphrodites. These results suggested that females and hermaphrodites possess similar facilitation intensity, and female cushion A. polytrichoides may allocate more resources saved from pollen production to seed production, while hermaphrodites possibly allocate more resources to pollen production hence reducing seed production. The surface areas covered by beneficiaries produced less flowers and fruits than areas without beneficiaries. In addition, strong negative correlations between beneficiary cover and flower cover were detected for both females and hermaphrodites, but the correlation strength were similar for these two sex morphs. However, the correlation between beneficiary cover and fruit cover was only significantly negative for females, suggesting that beneficiary plants negatively affect fruit reproduction of females while have neutral effects on hermaphrodites. All the results suggest that to facilitate other beneficiaries can induce reproductive costs on cushion A. polytrichoides, with females possibly suffering greater cost than hermaphrodites. Such differentiation in reproductive costs between sex morphs, in long-term perspective, may imply sex imbalance in population dynamics.

Abstract Background Obesity is associated with elevated serum uric acid (SUA) levels and frequent gout flares. Losing weight can reduce the SUA level and gout flares. The effect of orlistat on SUA levels and gout flares in patients with overweight/obesity and hyperuricemia (HUA) has not been extensively studied. This study investigated the effects of orlistat on SUA levels and gout flares compared to placebo in overweight and obese patients with HUA. Methods A total of 72 Chinese patients with overweight/obesity and HUA were randomly divided into a placebo group (35, 48.6%) and an orlistat group (37, 51.4%); the trial lasted 12 weeks. The primary endpoints were the relative changes in body weight, the SUA level, and gout flares in the per-protocol population. Results Orlistat reduced the proportion of patients with gout flares (log-rank P = 0.023, hazard ratio = 0.31, 95% confidence interval 0.11–0.85). There was no significant difference in SUA level between the two groups. The average weight loss of the orlistat group was 2.85 kg, and the average weight loss of the placebo group was 0.76 kg. Balancing the biomass requirements of different functions for the purpose of population reproduction and persistence can be challenging for alpine plants due to extreme environmental stresses from both above- and below-ground sources. The presence of ecosystem engineers in alpine ecosystems effectively alleviates microenvironmental stresses, hence promoting the survival and growth of other less stress-tolerant species. However, the influence of ecosystem engineers on plant resource allocation strategies remains highly unexplored. In this study, we compared resource allocation strategies, including biomass accumulation, reproductive effort (RE), root fraction (RF), as well as relationships between different functions, among four alpine plant species belonging to Gentianaceae across bare ground, tussock grass-, cushion-, and shrub-engineered microhabitats. Shrub-engineered microhabitats exerted the strongest effects on regulating plant resource allocation patterns, followed by tussock grass- and cushion-engineered microhabitats. Additionally, apart from microhabitats, population background and plant life history also significantly influenced resource allocation strategies. Generally, plants established within engineered microhabitats exhibited higher biomass accumulation, as well as increased flower, leaf and stem production. Furthermore, individuals within engineered microhabitats commonly displayed lower RF, indicating a greater allocation of resources to above-ground functions while reducing allocation to root development. RE of annual plants was significantly higher than that of perennial plants. However, individuals of annual plants within engineered microhabitats showed lower RE compared to their counterparts in bare ground habitats; whereas perennial species demonstrated similar RE between microhabitat types. Moreover, RE was generally independent of plant size in bare-ground habitats but exhibited size-dependency in certain populations for some species within specific engineered microhabitat types. However, size-dependency did exist for absolute reproductive and root biomass allocation in most of the cases examined here. No trade-offs were observed between flower mass and flower number, nor between leaf mass and leaf number. The capacity of ecosystem engineers to regulate resource allocation strategies in associated plants was confirmed. However, the resource allocation patterns resulted synergistically from the ecosystem engineering effects, population environmental backgrounds, and plant life history strategies. In general, such regulations can improve individual survival and reproductive potential, potentially promoting population persistence in challenging alpine environments.

In the western South China Sea, the diversity and complex genesis of sand waves lead to difficulties in measuring the actual scour depth of offshore wind turbine foundations. The present study employs methods such as in-situ multibeam echo sounding along with numerical simulation techniques, combined with regional hydrological and geological data, to reveal the scour depth of pile foundations under seabed sand wave migration conditions (SPF-SWM). Unlike previous studies, this study examines pile foundation scour depth from both macroscopic and microscopic perspectives, and proposes a method for risk assessment and safety zoning during the service life of pile foundations (RASZ). Our findings demonstrate that the dominant flow directions in the modern hydrodynamic environment play a crucial role in shaping and migrating sand waves, providing guidance for macro-level offshore wind project siting. Moreover, the study quantifies the changes in pile foundation elevations during sand wave migration and assesses their impact on safety throughout the service life of various pile foundations. Additionally, by utilizing Flow3D’s numerical model and combining it with sampling data, the maximum scour depth likely to be experienced by pile foundations was calculated, establishing a micro-perspective on the range of pile foundation scour and analytically examining the coupling mechanism between sand wave migration and pile foundation scour in stages. By considering SPF-SWM, we estimated the actual depth variations of pile foundations in the study area, enabling effective assessment of scour conditions during their service life. This also provides ideas for delineating safety and risk zones, thereby offering precise locational analysis for the construction planning of offshore wind farm pile foundations.

Experimental studies were conducted on four extended end-plate joints subjected to cyclic loading at the column top, investigating the evolving patterns of the joints' mechanical performance. The paper provides a detailed analysis and discussion of the test joints' failure modes, ductility, stiffness degradation, and energy dissipation capacity. The Mann-Kendall (M − K) trend analysis tool was applied to the mechanical response curves, identifying key performance evolution points (evolution initiation point P and overall yield point Q). The trends in bolt forces, deformations, and strains at critical joints were effectively validated, revealing the transition of the energy system from quantitative to qualitative changes and the component's failure process from stability to instability. Additionally, based on the experimental joints' hysteresis curves and energy dissipation capacity, a theoretical hysteresis model was established to predict the joint's hysteresis curve and cumulative dissipated energy accurately. According to EC3 requirements, joints were classified as partially rigid connections. The experimental results of the initial rotational stiffness and plastic moment were further used to evaluate the calculated values in existing standards EN 1993-1-8, ANSI/AISC 358-16, and GB 51017-2017. The results indicate that extended end-plate connections possess sufficient strength, joint rotational stiffness, ductility, and energy dissipation capacity, making them suitable for seismic moment frames.

The alteration of consciousness during shamanic rituals is both a physical and mystical phenomenon. It involves psychological and spiritual experiences. Through ritual practices, shamans can connect with archetype within the collective unconscious, utilizing trance-inducing techniques for “hallucinatory exploration”. This study surveyed 75 participants to investigate the impact of prototype symbols in Shamanistic rituals on participants’ consciousness states focusing on Jungian psychology’s concept of archetype. The results indicate that archetype symbols in shamanic rituals can significantly influence participants’ conscious state, leading them to experience a conscious dissolution of the self. Furthermore, archetype symbols have different effects at the stages of consciousness change. In particular, during the “Visionary Restructuralization” stage, archetype symbols, such as patterns, masks, totems and music, brought participants’ consciousness to a peak and caused significant changes to it. These findings suggest that the metaphoric function of archetype symbols plays a crucial role in rituals. Archetype symbols connect the individual to the collective unconscious through visual images and symbolic imagery. They prompt the participants to experience emotional resonances that transcend individual experiences and affect their state of consciousness.

The color and texture of camouflaged objects closely resemble their backgrounds, making it difficult to distinguish between the target and the background. Additionally, camouflaged environments are complex, with some camouflaged objects overlapping significantly with the background, further complicating the identification of camouflaged target areas. To address these challenges, we present a novel double-branch camouflaged object detection algorithm, termed TDNet, inspired by biological vision and based on the classical convolutional neural network ResNet50. TDNet employs a two-stage target detection structure that utilizes location segmentation to accurately identify detection targets. Specifically, we introduce an internal feature interaction mechanism (IFE) that enhances camouflaged target feature representations through multi-dimensional spatial feature enhancement and multi-channel feature interaction, enabling the rough segmentation of camouflaged regions. To mitigate the issue of blurred boundaries of camouflaged targets, we devise a cross-layer feature fusion mechanism (CFU) to refine the borders of camouflaged regions. Moreover, we employ a multi-supervised approach to constrain the model’s degradation process, ensuring the accuracy of camouflaged object detection. Experimental results demonstrate that our proposed method outperforms nine classical camouflaged object detection algorithms, achieving state-of-the-art performance. Recently, the use of unmanned aerial vehicle (UAV) imagery for object detection in forest fire detection has gained significant attention and has shown remarkable performance. However, most existing object detection models have neglected the exploration of relationships between positive sample features, which is crucial for learning more representative and color-robust features. In addition, small objects in UAV images poses challenges in capturing sufficient object information and hinders accurate object detection. To address these issues, we propose FCLGYOLO that aims to constrain positive sample features and enrich the object information in the feature maps. Specifically, a feature invariance and covariance constraint structure proposed to maintain feature invariance among positive samples and remove internal correlations. Furthermore, a local guided global module proposed to enrich object positioning and semantic information in the feature map, which leverages local features that focus on spatial information to facilitate the learning of global features that focus on frequency information. It is interesting to show that FCLGYOLO performs well even in the presence of heavy smoke or tree occlusions. Compared with multiple state-of-the-art object detection models on a forest fire dataset, experimental results demonstrate the superiority of FCLGYOLO.

Abstract Multiferroic materials offer a promising avenue for manipulating digital information by leveraging the cross-coupling between ferroelectric and ferromagnetic orders. Despite the ferroelectricity has been uncovered by ion displacement or interlayer-sliding, one-unit-cell of multiferroic materials design and wafer-scale synthesis have yet to be realized. Here we develope an interface modulated strategy to grow 1-inch one-unit-cell of non-layered chromium sulfide with unidirectional orientation on industry-compatible c-plane sapphire. The interfacial interaction between chromium sulfide and substrate induces the intralayer-sliding of self-intercalated chromium atoms and breaks the space reversal symmetry. As a result, robust room-temperature ferroelectricity (retaining more than one month) emerges in one-unit-cell of chromium sulfide with ultrahigh remanent polarization. Besides, long-range ferromagnetic order is discovered with the Curie temperature approaching 200 K, almost two times higher than that of bulk counterpart. In parallel, the magnetoelectric coupling is certified and which makes 1-inch one-unit-cell of chromium sulfide the largest and thinnest multiferroics.

Abstract Background Kidney cancer is an immunogenic solid tumor, characterized by high tumor burden and infiltration of CD8+ T cells. Although immunotherapy targeting the PD1/CTLA-4 axis has demonstrated excellent clinical efficacy, clinical outcomes in most patients are poor. Methods We used the RNA sequencing data from the GEO database for KIRC GSE121636 and normal kidney tissue GSE131685, and performed single-cell analysis for cluster identification, pathway enrichment, and CD8+ T cell-associated gene identification. Subsequently, the significance of different CD8+ T-cell associated gene subtypes was elucidated by consensus clustering, pathway analysis, mutated gene analysis, and KIRC immune microenvironment analysis in the TCGA–KIRC disease cohort. Single gene analysis identified LAG3 as the most critical CD8+ T-cell-associated gene and its function was verified by cell phenotype and immunohistochemistry in KIRC. Results In the present study, CD8+ T-cell associated genes in KIRC were screened, including GZMK, CD27, CCL4L2, FXYD2, LAG3, RGS1, CST7, DUSP4, CD8A, and TRBV20-1 and an immunological risk prognostic model was constructed (risk score = − 0.291858656434841*GZMK − 0.192758342489394*FXYD2 + 0.625023643446193*LAG3 + 0.161324477181591*RGS1 − 0.380169045328895*DUSP4 − 0.107221347575037*TRBV20-1). LAG3 was identified and proved as the most critical CD8+ T cell-associated gene in KIRC. Conclusion We proposed and constructed an immunological risk prognostic model for CD8+ T cell-associated genes and identified LAG3 as a pivotal gene for KIRC progression and CD8+ T-cell infiltration. The model comprehensively explained the immune microenvironment and provided novel immune-related therapeutic targets and biomarkers in KIRC.

Accurate identification of electroencephalogram (EEG) signals forms the basis for the development and application of brain–computer interface (BCI) devices. Signal preprocessing is an essential part of most EEG classification and recognition systems.In this study, we proposed a method called Super-resolution level separation (SRLS) to add dimensions of information to the classification model. First, we calculated the correlation between the EEG signals acquired by each channel and divided these channels into multiple levels. Next, we used a super-resolution method to calculate common EEGs (C-EEGs) acquired by the channels. In addition, we designed an EEG–split-informer (ES-informer) model based on an informer model to enable small-sample users to obtain highly fitting C-EEGs. We then calculated the difference between the C-EEG and true EEG (T-EEG) to obtain the unique EEGs (U-EEGs) acquired by each channel, thus, adding dimensions to the data inputted to the classification model. Utilizing the 2008 2a motor imagery (MI) EEG dataset from the BCI Competition and the P300 paradigm data collected, EEGNet was employed as the classification model to validate the efficacy of the proposed SRLS method. The results of the experiments indicated that the SRLS method augmented the input dimension of the model and amplified the classification accuracy by over 7% for MI and 3.6% for P300. These findings demonstrate that SRLS is capable of enhancing the recognition accuracy of EEG.

Abstract Background How geographical isolation and ecological divergence act together to promote plant diversity in mountainous regions remains largely unknown. In this study, we chose two genera comprising a small number of species distributed in the Sino-Himalayan region, Megacodon (Gentianaceae) and Beesia (Ranunculaceae), which both exhibit a fragmented distribution pattern and are found across a wide range of elevations. By summarizing their common patterns of speciation and/or divergence processes, we aim to understand how environmental changes accelerated lineage diversification in the Sino-Himalayan region through ancient allopatry and ecological divergence. Results Using ddRAD-seq, chloroplast genome sequences, and specific molecular markers, we studied the phylogenetic relationships, population structure, and historical biogeography of Beesia and Megacodon. Both genera began to diverge from the late Miocene onwards, with ancient allopatry at lower elevations formed narrow-range species or relict populations. Mantel tests between genetic distance and climatic, elevational, or geographic distance revealed an isolation-by-distance pattern in Beesia and Megacodon stylophorus. Megacodon showed two clades occupying entirely different altitudinal ranges, whereas Beesia calthifolia exhibited a genetic divergence pattern along an elevation gradient. Furthermore, we conducted morphological measurements on Beesia calthifolia and found that different elevational groups had distinct leaf shapes. Conclusions The regional disjunctions of plant groups in the Sino-Himalayan region are drastic and closely related to several biogeographic boundaries. As a consequence of major geological and climate change, ecological divergence when different elevations are colonized often happens simultaneously within plant groups. Although habitat fragmentation and parapatric ecological divergence each spur speciation to different extents, a combined effect of these two factors is a common phenomenon in the Sino-Himalayan region.

Background: Ultrasound (US) has a high sensitivity in detecting monosodium urate (MSU) deposition in gout patients. However, the value of US in predicting gout flares has been reported only in a few monocentric studies. Objective: To investigate the association between gout flares in the previous year and US-detected MSU burden using two different US scores. Design: A retrospective study. Methods: Patients with gout were consecutively recruited to undergo musculoskeletal US examinations of their knees, ankles, and feet. The score derived from Outcome Measure in Rheumatology (hereinafter referred to as MSU score) and musculoskeletal US features-based (hereinafter referred to as MSKF score) were used to quantify the MSU burden of gout. Odds ratios for frequent gout flares were calculated. Results: We enrolled 1894 patients with gout (mean age: 45 years; gout duration: 5 years; males: 96.1%), experiencing a median of three flares over the past year. Of these, 428 (22.6%) patients reported frequent (⩾7) gout flares. The MSU and MSKF median scores were 6 and 9, respectively. For each five-point increase in MSU and MSKF score, the odds ratio of frequent gout flares increased 1.13-fold and 1.24-fold, respectively. The area under the curve (AUC) for the MSU and MSKF score was 0.635 [95% confidence interval (CI): 0.604–0.665] and 0.688 (95% CI: 0.659–0.718), respectively, (AUC difference 0.054, p value for AUC difference

Abstract Background The Sino-Himalayan flora harbors highly diverse high-elevation biotas, but our understanding of its evolutionary history in temporal and spatial dimensions is limited. In this study, we integrated a dated phylogenetic tree with comprehensive species distribution data to investigate changes over time and space in floristic elements, including the tropical, Tethys, northern temperate, and East Asian floristic elements, across the entire Sino-Himalaya and its three floristic regions: the Yunnan Plateau, Hengduan Mountains, and East Himalaya regions. Results Our results revealed that the Sino-Himalayan flora developed from lowland biomes and was predominantly characterized by tropical floristic elements before the collision between the Indian subcontinent and Eurasia during the Early Cenozoic. Subsequently, from the Late Eocene onwards, the uplifts of the Himalaya and Hengduan Mountains transformed the Sino-Himalayan region into a wet and cold plateau, on which harsh and diverse ecological conditions forced the rapid evolution of local angiosperms, giving birth to characteristic taxa adapted to the high altitudes and cold habitat. The percentage of temperate floristic elements increased and exceeded that of tropical floristic elements by the Late Miocene. Conclusions The Sino-Himalayan flora underwent four significant formation periods and experienced a considerable increase in endemic genera and species in the Miocene, which remain crucial to the present-day patterns of plant diversity. Our findings support the view that the Sino-Himalayan flora is relatively young but has ancient origins. The three major shifts in the divergence of genera and species during the four formation periods were primarily influenced by the uplifts of the Himalaya and Hengduan Mountains and the onset and intensification of the Asian monsoon system. Additionally, the temporal patterns of floristic elements differed among the three floristic regions of the Sino-Himalaya, indicating that the uplift of the Himalaya and surrounding areas was asynchronous. Compared to the Yunnan Plateau region, the East Himalaya and Hengduan Mountains experienced more recent and drastic uplifts, resulting in highly intricate topography with diverse habitats that promoted the rapid radiation of endemic genera and species in these regions.

Brandisia is a shrubby genus of about eight species distributed basically in East Asian evergreen broadleaved forests (EBLFs), with distribution centers in the karst regions of Yunnan, Guizhou, and Guangxi in southwestern China. Based on the hemiparasitic and more or less liana habits of this genus, we hypothesized that its evolution and distribution were shaped by the development of EBLFs there. To test our hypothesis, the most comprehensive phylogenies of Brandisia hitherto were constructed based on plastome and nuclear loci (nrDNA, PHYA and PHYB); then divergence time and ancestral areas were inferred using the combined nuclear loci dataset. Phylogenetic analyses reconfirmed that Brandisia is a member of Orobanchaceae, with unstable placements caused by nuclear-plastid incongruences. Within Brandisia, three major clades were well supported, corresponding to the three subgenera based on morphology. Brandisia was inferred to have originated in the early Oligocene (32.69 Mya) in the Eastern Himalayas–SW China, followed by diversification in the early Miocene (19.45 Mya) in karst EBLFs. The differentiation dates of Brandisia were consistent with the origin of keystone species of EBLFs in this region (e.g., Fagaceae, Lauraceae, Theaceae, and Magnoliaceae) and the colonization of other characteristic groups (e.g., Gesneriaceae and Mahonia). These findings indicate that the distribution and evolution of Brandisia were facilitated by the rise of the karst EBLFs in East Asia. In addition, the woody and parasitic habits, and pollination characteristics of Brandisia may also be the important factors affecting its speciation and dispersal.

In this work, two Cu–Al–Mn shape memory alloys (SMAs), Cu-12.5Al–3Mn and Cu-12.5Al–3Mn–1Ni, were prepared by wire arc melting deposition, and their shape recovery rates, ultimate recoverable strains, additive manufacturing microstructures, memory recovery behaviors, and phase transition properties were comparatively investigated. Meanwhile, the mechanism of Ni in these two alloys was discussed by combining the differences in phase composition of the two alloys. Results showed that the shape recovery rate of Cu-12.5Al–3Mn alloy decreased linearly with increasing pre-strain, from 91.23% at 4% pre-strain to 62.41% at 10% pre-strain, and its ultimate recoverable strain was approximately 7%; while the shape recovery rate of Cu-12.5Al–3Mn–1Ni SMA was 100% at 4%–8% pre-strain, and remained at 96.57% until the pre-strain was increased to 10%, i.e., the recoverable strain was more than 9%. In addition, the phase change temperature of the alloy with Ni addition decreased compared to that of the alloy without Ni addition, especially the temperature hysteresis was narrower. The results indicate the formation of a new κ phase after adding Ni to the alloy. The pinning effect of this new phase significantly improves the order of martensite within the alloy. Moreover, Ni also induces the transformation of the alloy, inhibiting the precipitation of the brittle phase γ2 and leading to a marked increase in the shape memory recovery rate and the recoverable strain of the additively fabricated Cu–Al–Mn alloy. Note that Ni transforms the alloy from hyper-eutectoid components to eutectoid components. Moreover, after adding Ni, the thermoelastic martensite phase transition temperature and temperature hysteresis decreased together with its eutectoid temperature, which made the additive manufacturing Cu–Al–Mn SMA exhibit an explosive response behavior.

Background: Changes to work-life balance has increased the incidence of cervical cancer among younger people. A minor ginseng saponin known as ginsenoside Rk1 can inhibit the growth and survival of human cancer cells; however, whether ginsenoside Rk1 inhibits HeLa cell proliferation is unknown. Methods and results: Ginsenoside Rk1 blocked HeLa cells in the G0/G1 phase in a dose-dependent manner and inhibited cell division and proliferation. Ginsenoside Rk1 markedly also activated the apoptotic signaling pathway via caspase 3, PARP, and caspase 6. In addition, ginsenoside Rk1 increased LC3B protein expression, indicating the promotion of the autophagy signaling pathway. Protein processing in the endoplasmic reticulum signaling pathway was downregulated in Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses, consistent with teal-time quantitative PCR and western blotting that showed YOD1, HSPA4L, DNAJC3, and HSP90AA1 expression levels were dramatically decreased in HeLa cells treated with ginsenoside Rk1, with YOD1 was the most significantly inhibited by ginsenoside Rk1 treatment. Conclusion: These findings indicate that the toxicity of ginsenoside Rk1 in HeLa cells can be explained by the inhibition of protein synthesis in the endoplasmic reticulum and enhanced apoptosis, with YOD1 acting as a potential target for cervical cancer treatment.

Our first record of the rare and scatteredly distributed Ixeridium sagittarioides for Guizhou, China, triggered a study to assess its systematic position. The species was placed in four different genera in the course of its taxonomic history and was recently treated with doubts as a member of Ixeridium in the Flora of China. Comparative morphological investigation and phylogenetic analyses based on the nuclear ribosomal DNA internal transcribed spacer (nrITS) and five non-coding plastid DNA regions (petD region, psbA-trnH, trnL-trnF, rpl32-trnL (UAG) and 5´rps16-trnQ (UUG) spacers) provided evidence that the species is not a member of Ixeridium and the Crepidinae but has evolved by ancient hybridisation of members of the Lactuca alliance (Lactucinae). It is reinstated as Lactuca sagittarioides and a comprehensive morphological description is provided, based on material from its entire range of distribution.

Fibrotic liver features excessive deposition of extracellular matrix (ECM), primarily produced from “activated” hepatic stellate cells (HSCs). While targeting human HSCs (hHSCs) in fibrosis therapeutics shows promise, the overall understanding of hHSC activation remains limited, in part because it is very challenging to define the role of human long non-coding RNAs (lncRNAs) in hHSC activation. To address this challenge, we identified another cell type that acts via a diverse gene network to promote fibrogenesis. Then, we identified the lncRNAs that were differentially regulated in activated hHSCs and the other profibrotic cell. Next, we conducted concurrent analysis to identify those lncRNAs that were specifically involved in fibrogenesis. We tested and confirmed that transdifferentiation of vascular smooth muscle cells (VSMCs) represents such a process. By overlapping TGFβ-regulated lncRNAs in multiple sets of hHSCs and VSMCs, we identified a highly selected list of lncRNA candidates that could specifically play a role in hHSC activation. We experimentally characterized one human lncRNA, named CARMN, which was significantly regulated by TGFβ in all conditions above. CARMN knockdown significantly reduced the expression levels of a panel of marker genes for hHSC activation, as well as the levels of ECM deposition and hHSC migration. Conversely, gain of function of CARMN using CRISPR activation (CRISPR-a) yielded the completely opposite effects. Taken together, our work addresses a bottleneck in identifying human lncRNAs that specifically play a role in hHSC activation and provides a framework to effectively select human lncRNAs with significant pathophysiological role.

Introduction: Inactivated parapoxvirus ovis (iPPVO) exerts strong immunomodulatory effects on innate immune cells, making it an attractive therapeutic candidate. However, little is known about the signaling pathways that are involved in iPPVO-induced immune responses. Methods: In this study, we systematically analyzed how different types of dendritic cells (DCs) react to iPPVO (Zylexis, strain D1701) in both BALB/c and C57BL/6 mice by flow cytometry and ELISAs, and investigated which signaling pathway is related to DC activation by Western blotting and protein profiling. Results: We demonstrated that bone marrow-derived conventional DCs (BM-cDCs) and bone marrow-derived plasmacytoid DCs (BM-pDCs) matured and secreted type I interferons in response to Zylexis stimulation in both mouse strains. Similarly, Zylexis promoted the secretion of IL-12/23p40 and TNF by pDCs. However, IL-12/23p40 and TNF secretion by cDCs were induced in BALB/c mice but not in C57BL/6 mice. Analyzing the underlying signaling pathways revealed that iPPVO-induced maturation of cDCs was Toll-like receptor 9 (TLR9) independent, while the maturation of pDCs partially depended on the TLR9 pathway. Moreover, the production of proinflammatory cytokines by cDCs and the secretion of IFN-α/β by pDCs partially depended on the TLR9 pathway in both mouse strains. Therefore, other signaling pathways seem to participate in the response of DCs to iPPVO, supported by protein profiling. Conclusion: Our data provide useful insights into the diversity of iPPVO sensors and their varying effects across different strains and species.

BackgroundDisulfidptosis, an emerging type of programmed cell death, plays a pivotal role in various cancer types, notably impacting the progression of kidney renal clear cell carcinoma (KIRC) through the tumor microenvironment (TME). However, the specific involvement of disulfidptosis within the TME remains elusive.MethodsAnalyzing 41,784 single cells obtained from seven samples of KIRC through single-cell RNA sequencing (scRNA-seq), this study employed nonnegative matrix factorization (NMF) to assess 24 disulfidptosis regulators. Pseudotime analysis, intercellular communication mapping, determination of transcription factor activities (TFs), and metabolic profiling of the TME subgroup in KIRC were conducted using Monocle, CellChat, SCENIC, and scMetabolism. Additionally, public cohorts were utilized to predict prognosis and immune responses within the TME subgroup of KIRC.ResultsThrough NMF clustering and differential expression marker genes, fibroblasts, macrophages, monocytes, T cells, and B cells were categorized into four to six distinct subgroups. Furthermore, this investigation revealed the correlation between disulfidptosis regulatory factors and the biological traits, as well as the pseudotime trajectories of TME subgroups. Notably, disulfidptosis-mediated TME subgroups (DSTN+CD4T-C1 and FLNA+CD4T-C2) demonstrated significant prognostic value and immune responses in patients with KIRC. Multiple immunohistochemistry (mIHC) assays identified marker expression within both cell clusters. Moreover, CellChat analysis unveiled diverse and extensive interactions between disulfidptosis-mediated TME subgroups and tumor epithelial cells, highlighting the TNFSF12-TNFRSF12A ligand-receptor pair as mediators between DSTN+CD4T-C1, FLNA+CD4T-C2, and epithelial cells.ConclusionOur study sheds light on the role of disulfidptosis-mediated intercellular communication in regulating the biological characteristics of the TME. These findings offer valuable insights for patients with KIRC, potentially guiding personalized immunotherapy approaches.

Abstract Methods accurately predicting the responses of colorectal cancer (CRC) and colorectal cancer liver metastasis (CRLM) to personalized chemotherapy remain limited due to tumor heterogeneity. This study introduces an innovative patient‐derived CRC and CRLM tumor model for preclinical investigation, utilizing 3d‐bioprinting (3DP) technology. Efficient construction of homogeneous in vitro 3D models of CRC/CRLM is achieved through the application of patient‐derived primary tumor cells and 3D bioprinting with bioink. Genomic and histological analyses affirm that the CRC/CRLM 3DP tumor models effectively retain parental tumor biomarkers and mutation profiles. In vitro tests evaluating chemotherapeutic drug sensitivities reveal substantial tumor heterogeneity in chemotherapy responses within the 3DP CRC/CRLM models. Furthermore, a robust correlation is evident between the drug response in the CRLM 3DP model and the clinical outcomes of neoadjuvant chemotherapy. These findings imply a significant potential for the application of patient‐derived 3DP cancer models in precision chemotherapy prediction and preclinical research for CRC/CRLM.

Polyploidization is a process that typically leads to instantaneous reproductive isolation and has, therefore, been considered as one of the major evolutionary forces in the species-rich Hengduan Mountains (HM), yet this topic remains poorly studied in the region. Allium sikkimense and its relatives (about eight species) compose a natural diploid–polyploid complex with the highest diversity in the HM and adjacent areas. A combination of nuclear ribosomal DNA (nrDNA), plastome, transcriptome, and ploidy identification through chromosome counting and flow cytometry is employed to reconstruct the phylogenetic relationships in this complex and to investigate the frequency and the evolutionary significance of polyploidy in the complex. The plastome failed to resolve the phylogenetic relationships of the different species in the A. sikkimense complex, and the phylogenetic tree based on nrDNA also has limited resolution. However, our study reveals a well-resolved phylogenetic framework for species in the A. sikkimense complex using more than 1,000 orthologous genes from the transcriptome data. Previously recognized morphospecies A. sikkimense are non-monophyletic and comprise at least two independently evolved lineages (i.e., cryptic species), each forming a clade with different diploid species in this complex. The embedded pattern of octoploid A. jichouense and tetraploid A. sp. nov. within different polyploid samples of A. sikkimense supports a possible scenario of budding speciation (via niche divergence). Furthermore, our results reveal that co-occurring species in the A. sikkimense complex usually have different ploidy levels, suggesting that polyploidy is an important process for reproductive isolation of sympatric Allium species. Phylogenetic network analyses suggested that the phylogenetic relationships of the A. sikkimense complex, allowing for reticulation events, always fit the dataset better than a simple bifurcating tree. In addition, the included or exserted filaments, which have long been used to delimit species, are highly unreliable taxonomically due to their extensive parallel and convergent evolution.

Abstract Glasshouse plants are species that trap warmth via specialized morphology and physiology, mimicking a human glasshouse. In the Himalayan alpine region, the highly specialized glasshouse morphology has independently evolved in distinct lineages to adapt to intensive UV radiation and low temperature. Here we demonstrate that the glasshouse structure – specialized cauline leaves – is highly effective in absorbing UV light but transmitting visible and infrared light, creating an optimal microclimate for the development of reproductive organs. We reveal that this glasshouse syndrome has evolved at least three times independently in the rhubarb genus Rheum. We report the genome sequence of the flagship glasshouse plant Rheum nobile and identify key genetic network modules in association with the morphological transition to specialized glasshouse leaves, including active secondary cell wall biogenesis, upregulated cuticular cutin biosynthesis, and suppression of photosynthesis and terpenoid biosynthesis. The distinct cell wall organization and cuticle development might be important for the specialized optical property of glasshouse leaves. We also find that the expansion of LTRs has likely played an important role in noble rhubarb adaptation to high elevation environments. Our study will enable additional comparative analyses to identify the genetic basis underlying the convergent occurrence of glasshouse syndrome.

Abstract Background The Sinodielsia clade of the subfamily Apioideae (Apiacieae) was established in 2008, and it is composed of 37 species from 17 genera. Its circumscription is still poorly delimited and unstable, and interspecific relationships in the clade lack comprehensive analysis. Chloroplast (cp.) genomes provide valuable and informative data sources for evolutionary biology and have been widely used in studies on plant phylogeny. To infer the phylogenetic history of the Sinodielsia clade, we assembled complete cp. genomes of 39 species and then performed phylogenetic analysis based on these cp. genome sequence data combined with 66 published cp. genomes from 16 genera relative to the Sinodielsia clade. Results These 39 newly assembled genomes had a typical quadripartite structure with two inverted repeat regions (IRs: 17,599–31,486 bp) separated by a large single-copy region (LSC: 82,048–94,046 bp) and a small single-copy region (SSC: 16,343–17,917 bp). The phylogenetic analysis showed that 19 species were clustered into the Sinodielsia clade, and they were divided into two subclades. Six mutation hotspot regions were detected from the whole cp. genomes among the Sinodielsia clade, namely, rbcL–accD, ycf4–cemA, petA–psbJ, ycf1–ndhF, ndhF–rpl32 and ycf1, and it was found that ndhF–rpl32 and ycf1 were highly variable in the 105 sampled cp. genomes. Conclusion The Sinodielsia clade was subdivided into two subclades relevant to geographical distributions, except for cultivated and introduced species. Six mutation hotspot regions, especially ndhF–rpl32 and ycf1, could be used as potential DNA markers in the identification and phylogenetic analyses of the Sinodielsia clade and Apioideae. Our study provided new insights into the phylogeny of the Sinodielsia clade and valuable information on cp. genome evolution in Apioideae.

Toxoplasma gondii (T. gondii) is an opportunistic parasite that can infect the central nervous system (CNS), causing severe toxoplasmosis and behavioral cognitive impairment. Mortality is high in immunocompromised individuals with toxoplasmosis, most commonly due to reactivation of infection in the CNS. There are still no effective vaccines and drugs for the prevention and treatment of toxoplasmosis. There are five developmental stages for T. gondii to complete life cycle, of which the tachyzoite and bradyzoite stages are the key to the acute and chronic infection. In this study, to better understanding of how T. gondii interacts with the host CNS at different stages of infection, we constructed acute and chronic infection models of T. gondii in astrocytes, and used label-free proteomics to detect the proteome changes before and after infection, respectively. A total of 4676 proteins were identified, among which 163 differentially expressed proteins (fold change ≥ 1.5 or ≤ 0.67 and p-value ≤ 0.05) including 109 up-regulated proteins and 54 down-regulated proteins in C8-TA vs C8 group, and 719 differentially expressed proteins including 495 up-regulated proteins and 224 down-regulated proteins in C8-BR vs C8-TA group. After T. gondii tachyzoites infected astrocytes, differentially expressed proteins were enriched in immune-related biological processes to promote the formation of bradyzoites and maintain the balance of T. gondii, CNS and brain. After T. gondii bradyzoites infected astrocytes, the differentially expressed proteins up-regulated the host's glucose metabolism, and some up-regulated proteins were strongly associated with neurodegenerative diseases. These findings not only provide new insights into the psychiatric pathogenesis of T. gondii, but also provide potential targets for the treatment of acute and chronic Toxoplasmosis.

In order to study the effect of typical pioneer herb plants in the metal smelting slag dump on the nutrients and heavy metal forms of the slag, the pioneer herb (Trifolium repens, Lolium perenne) rhizosphere microdomains on the lead-zinc smelting slag dump that has carried out vegetation reconstruction for 5 years were used as the research object, analyzing the differentiation characteristics of heavy metal content, occurrence forms and nutrients in two kinds of herbaceous plant rhizosphere micro-domain waste residues with different particle sizes (5 mm) during the ecological restoration process. The results showed that the plant nutrients in the waste residue were mainly distributed in the small particle size waste residue ryegrass> control, and showed obvious rhizosphere effects (rhizosphere> non rhizosphere). Two phytoremediation effects significantly affect the heavy metal content of waste residues and the distribution of their occurrence forms in different particle sizes: (1) The content of Pb and Cu in clover rhizosphere waste residue was significantly lower than that of the control, and promoted the conversion of Cd to residue; (2) The content of Cu and Cd in ryegrass rhizosphere waste residues with different particle sizes is redistributed, and the content in the fine particle size (

Abstract Objective Acute respiratory distress syndrome (ARDS) leads to life-threatening acute hypoxemic respiratory failure and requires mechanical ventilation. However, there is a lack of certainty regarding recruitment maneuvers (RMs) and positive end-expiratory pressure (PEEP) levels. Therefore, we performed a network meta-analysis to identify the optimal RM and PEEP levels of patients with ARDS. Methods We searched the PubMed, OVID and Cochrane Central Register of Controlled Trials (Central) databases for randomized controlled trials (RCTs). The primary outcome was death on the 28th day, and the secondary outcomes included in-hospital death, ventilator-free days, and barotrauma. The relative effect sizes were estimated by risk ratios (RRs) for binary variables and standard mean difference (SMD) for continuous variables. The efficacy of the interventions was ranked using surface under the cumulative ranking. Multiple intervention comparisons based on the Bayesian framework were performed to integrate the efficacy of all included strategies. Results Thirteen RCTs comprising 4410 patients were included in the network meta-analysis. In terms of death at 28 days, inconsistencies were found globally and locally in the tests. None of the ventilation strategies was significantly superior to the others on any outcomes. According to the relative rank probabilities, RM + lower PEEP levels showed the highest probability of reducing the risk of in-hospital death and reducing ventilator-free days. Lower PEEP levels showed the highest probability of benefitting barotrauma. The overall quality of the evidence per grade was moderate to low. Conclusions The no ventilation strategy is significantly superior to the other strategies. RM + lower PEEP levels has the highest probability of benefitting survival. The evidence has low overall quality and should be further studied.

Lignin (SCEL) was separated from steam-exploded eucalyptus wood fibers using a supercritical carbon dioxide (scCO2)–ethanol/water composite medium, which was compared with traditional industrial alkaline extraction of lignin (AL) and high-temperature ethanol extraction of lignin (EL). All three kinds of lignin were spray-dried to form microparticles. The obtained SCEL, AL, and EL particles were mixed with poly(vinyl alcohol) (PVOH) to prepare a composite film. Compared with the pure PVOH film, the transmittance of the composite film to ultraviolet light was reduced from more than 70% to less than 20%, and the tensile strengths of the SCEL/PVOH, AL/PVOH, and EL/PVOH films were increased by 79.2%, 39.1%, and 66.0%, respectively. The water contact angle was increased from 35.5° to 95.3°, 59.5°, and 66.7°, respectively. In terms of performance improvement, SCEL had a better effect. The SCEL particle size was small and uniform, and SCEL contained more S-type structural units and β-O-4 bonds, which is conducive to its uniform dispersion in the PVOH matrix and increases the contact area between lignin and ultraviolet light, thereby improving its anti-ultraviolet performance. The hydrophilic groups in lignin form a large number of hydrogen bonds with PVOH, which promotes the combination of lignin and PVOH, and at the same time exposes a large number of hydrophobic groups of lignin, which improves the mechanical properties and the hydrophobicity of the film.

Bridges’ expansion joints are prone to damage during operation, and repairing them often requires interruption of traffic, the impact of which can be minimized by using fast-hardening and early-strength expansion joint materials. In this study, a fast-hardening polymer cement composite (PCC) was developed using sulfate aluminate cement and ordinary silicate cement as binding agents and polymer powder as admixture. To improve the crack resistance of the material, several types of fibers were added and the effects of different fiber types and admixtures on the crack resistance of the material were compared using SCB tests. The results showed that the best effect of improving the crack resistance of concrete was achieved with a volume fraction of 0.5% of basalt fibers. Then, a test method for the interfacial shear properties of PCC materials and ordinary concrete was established, and the cohesive force model was selected as the interface simulation parameter for finite element analysis and compared with experimental data to verify its feasibility. Finally, based on the previously obtained PCC material parameters, a solid model of the expansion joint anchorage zone was established to study the mechanical properties of the expansion joint anchorage zone with the application of fast-hardening PCC material. This research provides a new way to develop fast-hardening and early-strength expansion joint materials with high crack resistance.

To improve the performance and efficiency of ordinary galloping energy harvesters (GEHs), this paper proposes a GEH with a striped bluff body. The fluid-structure coupling simulation of the bluff body and the oscillator of the energy harvester was carried out using COMSOL Multiphysics. The cloud diagram in the flow field, tip displacement at the tip of the beam, lifting and dragging force, and trace and frequency response of the harvester are analyzed. Simulation results show that the GEH with a striped bluff body has the characteristics of rising frequency and falling peak compared with the ordinary GEH, which can be locked at a fixed peak to form a stable periodic response. Experimental results show that the striped bluff body energy harvester displays better energy harvest performance. The output root mean square voltage can be increased by 119.2% at 2 m/s wind speed, which means that the galloping harvester with a striped bluff body has more stable dynamic characteristics and shows better energy harvest performance.

Abstract Background The dismal prognosis of hepatocellular carcinoma (HCC) is closely associated with characteristics of the tumour microenvironment (TME). Recent studies have confirmed the presence and potential influence of the microbiome in TME on cancer progression. Elucidating the relationship between microbes in the TME and cancer could provide valuable insights into novel diagnostic markers and therapeutic strategies for HCC and thus warrants a closer investigation of the role of intratumoural microbiome in the HCC TME. Methods We determined the presence of intratumoural microbiome using fluorescence in situ hybridisation, and explored the microbial community profiles in the HCC TME in paired tumour and adjacent normal tissues using 16S rDNA sequencing. Microbial signatures were characterised in the paired group, and their correlation with clinical characteristics was further investigated. We clustered the microbial signatures of tumour tissues by hepatotypes, and further analysis was performed to elucidate the independent prognostic value of the hepatotypes. Results This study revealed that microbial profiles and community networks differed notably between tumours and adjacent normal tissues. Proteobacteria and Actinobacteria were the most abundant phyla in the HCC TME. The TME microbial profiles also revealed heterogeneities between individuals and between multiple tumour lesions. Clustering of the microbial profiles into two hepatotypes revealed different microbial network patterns. Additionally, the hepatotypes were revealed to be independent prognostic factors in patients with resected HCC. Conclusions Our study illuminates the microbial profiles in the TME of HCC and presents the hepatotype as a potential independent biomarker for the prognostic prediction of HCC after surgery.

PurposeThis study aimed to explore the clinical value of non-invasive preoperative Edmondson-Steiner grade of hepatocellular carcinoma (HCC) using contrast-enhanced ultrasound (CEUS).Methods212 cases of HCCs were retrospectively included, including 83 cases of high-grade HCCs and 129 cases of low-grade HCCs. Three representative CEUS images were selected from the arterial phase, portal vein phase, and delayed phase and stored in a 3-dimensional array. ITK-SNAP was used to segment the tumor lesions manually. The Radiomics method was conducted to extract high-dimensional features on these contrast-enhanced ultrasound images. Then the independent sample T-test and the Least Absolute Shrinkage and Selection Operator (LASSO) were employed to reduce the feature dimensions. The optimized features were modeled by a classifier based on ensemble learning, and the Edmondson Steiner grading was predicted in an independent testing set using this model.ResultsA total of 1338 features were extracted from the 3D images. After the dimension reduction, 10 features were finally selected to establish the model. In the independent testing set, the integrated model performed best, with an AUC of 0.931.ConclusionThis study proposed an Edmondson-Steiner grading method for HCC with CEUS. The method has good classification performance on independent testing sets, which can provide quantitative analysis support for clinical decision-making.

Lesion-mimic mutants (LMM) spontaneously produce necrotic spots, a process not affected by environmental stress or pathogen infection. In this study, we identified a LMM, lesion mimic mutant 8 (lmm8) in rice (Oryza sativa). The lmm8 mutant produces brown and off-white lesions on its leaves during the second- and third-leaf stages. The lesion mimic phenotype of the lmm8 mutant was enhanced by light. At the mature stage, lmm8 mutant are shorter and exhibit inferior agronomic traits than the wild type. Contents of photosynthetic pigments and chloroplast fluorescence were significantly reduced in lmm8 leaves, along with increased production of reactive oxygen species and programmed cell death compared to the wild type. The mutated gene was identified as LMM8 (LOC_Os01g18320) by map-based cloning. A point mutation occurred in LMM8, causing a Leu to Arg mutation of the 146th amino acid of LMM8. It is an allele of SPRL1, encoding a protoporphyrinogen IX oxidase (PPOX) located in chloroplasts and involved in the biosynthesis of tetrapyrrole in chloroplasts. The lmm8 mutant showed enhanced resistance and broad-spectrum resistance. Together, our results demonstrate the importance of rice LMM8 protein in defense responses and plant growth in rice, and provides theoretical support for resistance breeding to improve rice yield.

Hepatocellular carcinoma (HCC) is a major global health burden, causing approximately 8.3 million deaths each year, and it is the third leading cause of cancer-related death worldwide, with a relative 5-year survival rate of around 18%. Due to the advanced stage of diagnosis in most patients, systemic treatment based on targeted therapy has become the only feasible option. Genomic studies have established a profile of molecular alterations in hepatocellular carcinoma with potentially actionable mutations, but these mutations have yet to be translated into clinical practice. The first targeted drug approved for systemic treatment of patients with advanced hepatocellular carcinoma was Sorafenib, which was a milestone. Subsequent clinical trials have identified multiple tyrosine kinase inhibitors, such as Lenvatinib, Cabozantinib, and Regorafenib, for the treatment of hepatocellular carcinoma, with survival benefits for the patient. Ongoing systemic therapy studies and trials include various immune-based combination therapies, with some early results showing promise and potential for new therapy plans. Systemic therapy for hepatocellular carcinoma is complicated by the significant heterogeneity of the disease and its propensity for developing drug resistance. Therefore, it is essential to choose a better, individualized treatment plan to benefit patients. Preclinical models capable of preserving in vivo tumor characteristics are urgently needed to circumvent heterogeneity and overcome drug resistance. In this review, we summarize current approaches to targeted therapy for HCC patients and the establishment of several patient-derived preclinical models of hepatocellular carcinoma. We also discuss the challenges and opportunities of targeted therapy for hepatocellular carcinoma and how to achieve personalized treatment with the continuous development of targeted therapies and bioengineering technologies.

The Qinghai-Tibet Plateau (QTP) is an important cushion plant hotspot. However, the distribution of cushion plants on the QTP is unknown, as are the factors that drive cushion plant distribution, limiting our understanding of the evolution of cushion species in the region. In this study, we assessed spatial patterns of total cushion plant diversity (including taxonomic and phylogenetic) over the entire QTP and compared patterns of diversity of cushion plants with different typologies (i.e., compact vs. loose). We also examined how these patterns were related to climatic features. Our results indicate that the southern QTP hosts the highest total cushion plant richness, especially in the south-central Hengduan Mountains subregion. The total number of cushion species declines from south to north and from southeast to northwest. Compact cushion plants exhibit similar patterns as the total cushion plant richness, whereas loose cushion plants show random distribution. Cushion plant phylogenetic diversity showed a similar pattern as that of the total cushion plant richness. In addition, cushion plant phylogenetic community structure was clustered in the eastern and southwestern QTP, whereas random or overdispersed in other areas. Climatic features represented by annual energy and water trends, seasonality and extreme environmental factors, had significant effects on cushion plant diversity patterns but limited effects on the phylogenetic community structure, suggesting that climatic features indeed promote the formation of cushion plants. Because cushion plants play vital roles in alpine ecosystems, our findings not only promote our understanding of the evolution and formation of alpine cushion plant diversity but also provide an indispensable foundation for future studies on cushion plant functions and thus alpine ecosystem sustainability in the entire QTP region.

The precision and reliability of electroencephalogram (EEG) data are essential for the effective functioning of a brain-computer interface (BCI). As the number of BCI acquisition channels increases, more EEG information can be gathered. However, having too many channels will reduce the practicability of the BCI system, raise the likelihood of poor-quality channels, and lead to information misinterpretation. These issues pose challenges to the advancement of BCI systems. Determining the optimal configuration of BCI acquisition channels can minimize the number of channels utilized, but it is challenging to maintain the original operating system and accommodate individual variations in channel layout. To address these concerns, this study introduces the EEG-completion-informer (EC-informer), which is based on the Informer architecture known for its effectiveness in time-series problems. By providing input from four BCI acquisition channels, the EC-informer can generate several virtual acquisition channels to extract additional EEG information for analysis. This approach allows for the direct inheritance of the original model, significantly reducing researchers’ workload. Moreover, EC-informers demonstrate strong performance in damaged channel repair and poor channel identification. Using the Informer as a foundation, the study proposes the EC-informer, tailored to BCI requirements and demanding only a small number of training samples. This approach eliminates the need for extensive computing units to train an efficient, lightweight model while preserving comprehensive information about target channels. The study also confirms that the proposed model can be transferred to other operators with minimal loss, exhibiting robust applicability. The EC-informer’s features enable original BCI devices to adapt to a broader range of classification algorithms and relax the operational requirements of BCI devices, which could facilitate the promotion of the use of BCI devices in daily life.

In this paper, a piezoelectric energy harvester based on periodic one-dimensional acoustic black holes (ABH) is proposed to improve efficiency. The harvesting performance of the energy harvester with different ABH-structures was numerically simulated through the commercial software, COMSOL Multiphysics. Finally, an experimental platform was set up to test several energy harvester samples. The results show that the energy aggregation effect of the bilateral ABH beam is better than that of the traditional ABH beam. In the optimal impedance matching, the maximum output power of the energy harvester with bilateral ABH type 3 is 112 mW, which is 2.8 times that of the energy harvester with traditional ABH. The simulation and experimental results show that the energy harvesting efficiency of the proposed ABH is much higher than that of the energy harvester with traditional ABH. It is expected to make some contributions to the further development of lightweight wireless sensors, equipment service life, bearing fault diagnosis, and so on.

Abstract In the Flora of China account of Saxifraga mengtzeana Engl. & Irmsch., eight synonyms were attributed to it and one variant, recognized as Saxifraga epiphylla Gornall & Ohba, was split from it. This study reevaluates the taxonomic status of some of the synonyms and of the segregated species in light of new evidence presented here. Morphological and molecular evidence demonstrate that collections from northwestern Yunnan and Sichuan are genetically differentiated from those in southeastern Yunnan and neighboring Guangxi. Observations in the field and in cultivation show that the peltate petiole attachment diagnostic of S. mengtzeana var. peltifolia Engl. & Irmsch. is developmentally labile. Similar observations combined with molecular data show that viviparous phenotypes, formerly treated as S. epiphylla, although largely under genetic control, occur sporadically throughout the ranges of both northern and southern taxa. Collections from northwestern Yunnan and Sichuan are best recognized as Saxifraga geifolia Balf.f., whereas those from southeastern Yunnan and neighboring Guangxi are S. mengtzeana. Peltate‐leaved variants of the latter are given no status and are relegated to complete synonymy. Viviparous phenotypes of S. mengtzeana and S. geifolia are recognized at the rank of forma.
