TFF3 wird bei mucosalen Verletzungen, unterschiedlichen ulcerativen Prozessen und Krebs vermehrt exprimiert und scheint vor allem bei der Abwehr, Instandhaltung und Reparatur des intestinalen Epithels eine Relevanz zu besitzen. Die Überexpression von TFF3 in HT-29/B6 führte in dieser Arbeit zu einem erhöhten protektiven Effekt gegenüber einer IFN-γ- und TNF‑α- induzierten Apoptose. Um diesen Effekt zu verstehen, haben wir unterschiedliche miRNAs und lncRNAs als potente Regulatoren untersucht, die laut Literaturrecherchen und in silico-Analysen eng im Zusammenhang mit TFF3-relevanten Signalwegen wie Wnt, PI3K und der Regulation der Apoptose stehen. Dabei stießen wir auf die antikorreliert exprimierte miR-491-5p und lncRNA PRINS. Gain- und loss of function-Experimente wiesen auf eine gegenseitige Regulation hin, wobei die direkte Interaktion von uns bewiesen werden konnte. Es drängte sich die Vermutung auf, dass der erhöhte antiapoptotische Effekt von TFF3 auf die Dysregulation und das Zusammenspiel der miR-491-5p und PRINS zurückzuführen sein könnte. Ein Ausgleich dieser veränderten Expression auf das Niveau von Kontrollzellen sollte unserer Hypothese nach auch den Schutz gegenüber der Induktion der Apoptose aufheben. Tatsächlich konnte der Anteil apoptotischer HT-29/B67/htff3 dadurch erhöht werden. Um den Mechanismus hinter der Wirkung beider nicht kodierenden RNAs zu untersuchen, wurden mittels kleiner inhibitorischer Moleküle verschiedene relevante Signalwege stillgelegt, wodurch die Bedeutung des PI3K-Signalweges erkennbar wurde. Die Inhibierung des PI3K-Signalweges führte zur Aufhebung der TFF3-vermittelten Dysregulation beider nicht kodierenden RNAs. Dabei scheinen TFF3 und miR-491-5p unabhängig von AKT in der Lage zu sein, PDK1 zu phosphorylieren und p70-S6K zu aktivieren. Die erhöhte Phosphorylierung der p70-S6K unabhängig von PDK1 in HT-29/B67/mock und die Untersuchung pro- und anti-apoptotisch wirkender Proteine führten uns zu PMAIP1, einem Regulator des pro-apoptotischen Bcl-2-Familienmitgliedes Mcl-1, der bekanntermaßen sogar in der Lage ist, Apoptose in Krebszellen auszulösen. Co-Lokalisierungen von PRINS und PMAIP1 bekräftigten die Hypothese einer potentiell gegenseitigen Regulation. Unseren Ergebnissen nach scheint TFF3 in der Lage zu sein, die Translokation von PMAIP1 vom Zellkern in das Zytoplasma PRINS-abhängig verhindern zu können. Final durchgeführte Co-Immunpräzipitationen deuteten auf eine direkte Bindung von PMAIP1 und PRINS hin. Weitere mechanistische Untersuchungen wären in Zukunft notwendig, um den genauen Einfluss von PRINS auf PMAIP1 entschlüsseln zu können. Besonders interessant wäre zusätzlich die Überprüfung des protektiven PRINS-Effektes in vivo als potentielles Therapeutikum bei der IBD, als Marker bei metastasierenden Krebserkrankungen oder zusätzliches Ziel im Rahmen einer TRAIL-basierten Chemotherapie., TFF3 is upregulated in mucosal injury, different ulcerative processes and cancer. It appears to have a relevance especially in the defense, maintenance and repair of the intestinal epithelium and is also closely related to tumor invasion, resistance to apoptosis and metastasis. The over-expression of TFF3 in HT-29/B6 resulted in an increased protective effect against IFN-γ and TNF-α induced apoptosis and caused dysregulation of various non-coding RNAs. Anti- correlated expression pattern after transfection with miR-491-5p-mimic as well as reporter gene assays confirmed the direct interaction of miR-491-5p with the lncRNA PRINS in HT-29/B6/htff3. miR-491-5p was shown to inhibit PRINS- expression while PRINS had no discernible effect on miR-491-5p. Moreover, PRINS expression seemed to be controlled independently of TFF3 and miR-491-5p by IFN-γ- and TNF-α. Both non-coding RNAs hold an impact on the previously observed antiapoptotic phenotype. Compensation of the dysregulated noncoding RNAs increased the number of apoptotic cells after TRAIL-induced apoptosis. Mechanistically, Western blot analyzes confirmed the involvement of PI3K signaling pathway in this context. Regulation of miR-491-5p and PRINS by IFN-γ/TNF-α seemed to activate the PI3K signaling pathway independent of AKT through regulation of PDK1 and p70-S6K. In addition, the TFF3-mediated inhibitory effect on miR-491-5p was reversible by inhibition of the PI3K signaling pathway with small molecular inhibitors. PRINS and miR-491-5p had an impact on expression of a variety of mRNA that encode for pro-or anti- apoptotic proteins and the regulation of PMAIP1, FOXK1 and FOXK2 was confirmed at protein level, too. In further analyzes PRINS and PMAIP1 showed nuclear colocalization, indicating a direct interaction. PRINS revealed its influence on PMAIP1’s cellular localization in the nucleus especially in combination with TRAIL-induced apoptosis. PRINS seemed to be able to prevent the translocation of PMAIP1 from the nucleus to the cytoplasm. Finally, co- immunoprecipitation suggested PMAIP1’s direct binding on PRINS. In future, further mechanistic studies will be necessary in order to determine the impact of PRINS on regulation of PMAIP1. Analyzes under in vivo conditions will be indicated to prove the potential of PRINS as a therapeutic target in IBD, as a marker in metastatic cancers or as part of a TRAIL-based chemotherapy.

Tumour necrosis factor-α (TNF-α) is a double-edged cytokine associated with pathogenesis of inflammatory-related cancers being also able to induce cancer cell death. In the process of tumour development or metastasis, cancer cells can become resistant to TNF-α. In trefoil factor 3 (TFF3) overexpressing colorectal adenocarcinoma cells (HT-29/B6), we observed enhanced resistance against TNF-α/interferon gamma-induced apoptosis. TFF3 is a secreted small peptide that supports intestinal tissue repair but is also involved in intestinal tumour progression and scattering. We hypothesised that TFF3 rescues intestinal epithelial cancer cells from TNF-α-induced apoptosis by involving regulatory RNA networks. In silico-based expression analysis revealed TFF3-mediated regulation of selected microRNAs as well as long non- coding RNAs (lncRNAs), whereas miR-491-5p was identified to target the lncRNA ‘psoriasis susceptibility-related RNA gene induced by stress’ (PRINS). RNA interference-based gain- and loss-of-function experiments examined miR-491-PRINS axis to exert the TFF3-mediated phenotype. Chemical inhibition of selected pathways showed that phosphatidylinositol 3-kinase/AKT accounts for TFF3-mediated downregulation of miR-491-5p and accumulation of PRINS. Moreover, we showed that PRINS colocalises with PMAIP1 (NOXA) in nuclei of HT-29/B6 possessing inhibitory effects. Immunoprecipitation experiments proved molecular interaction of PMAIP1 with PRINS. Our study provides an insight into RNA regulatory networks that determine resistance of colorectal cancer cells to apoptosis.

The GMO Risk Assessment and Communication of Evidence (GRACE; www.grace-fp7.eu) project is funded by the European Commission within the 7th Framework Programme. A key objective of GRACE is to conduct 90-day animal feeding trials, animal studies with an extended time frame as well as analytical, in vitro and in silico studies on genetically modified (GM) maize in order to comparatively evaluate their use in GM plant risk assessment. In the present study, the results of two 90-day feeding trials with two different GM maize MON810 varieties, their near-isogenic non-GM varieties and four additional conventional maize varieties are presented. The feeding trials were performed by taking into account the guidance for such studies published by the EFSA Scientific Committee in 2011 and the OECD Test Guideline 408. The results obtained show that the MON810 maize at a level of up to 33 % in the diet did not induce adverse effects in male and female Wistar Han RCC rats after subchronic exposure, independently of the two different genetic backgrounds of the event.
