Abstract

A new synthesis of (+)-cyclophellitol a potent beta-glucosidase inhibitor that was completed in nine steps from D-xylose is described. This represents the shortest method for preparing enantiopure cyclophellitol.

Abstract

Synthetic routes towards the three protected 3'-C-branched LNA-type phosphoramidite building blocks containing furanose rings locked in an N-type conformation starting from a known 3-C-allyl allofuranose derivative is reported. At low salt conditions a duplex stabilizing effect of introducing 3'-C-aminopropyl-LNA monomer relative to 3'-C-hydroxypropyl-LNA monomer was observed.

6. Synthesis of Gabosine A and N from Ribose by the Use of RingClosing Metathesis

Abstract

A concise synthetic route is described for the synthesis of gabosine A and N. The key step uses a zincmediated tandem reaction where methyl 5deoxy5iodo2,3OisopropylideneβDribofuranoside is fragmented to give an unsaturated aldehyde which is allylated in the same pot with 3benzoyloxy2methylallyl bromide. The functionalized octa1,7diene, thus obtained, is converted into the sixmembered gabosine skeleton by ringclosing olefin metathesis. Subsequent protective group manipulations and oxidation gives rise to gabosine N in a total of 8 steps from ribose while the synthesis of gabosine A employs an additional step for inverting a secondary hydroxy group. © WileyVCH Verlag GmbH & Co.
