Abstract

Whilst holding great promise for low noise, ease of operation and networking, useful photonic quantum computing has been precluded by the need for beyond-state-of-the-art components, manufactured by the millions. Here we introduce a manufacturable platform for quantum computing with photons. We benchmark a set of monolithically-integrated silicon photonics-based modules to generate, manipulate, network, and detect photonic qubits, demonstrating dual-rail photonic qubits with $99.98\% \pm 0.01\%$ state preparation and measurement fidelity, Hong-Ou-Mandel quantum interference between independent photon sources with $99.50\%\pm0.25\%$ visibility, two-qubit fusion with $99.22\%\pm0.12\%$ fidelity, and a chip-to-chip qubit interconnect with $99.72\%\pm0.04\%$ fidelity, not accounting for loss. Abstract

This article focuses on Japanese travelers between the ages of twenty and forty years old who come to Jamaica driven by their passion for its dancehall music scene. They are working-class sojourners alongside a smattering of expatriates looking for an experience that they view as affirmative, authentic, and, at times, escapist. Slackness is a Jamaican vernacular concept underscoring a resistance to hegemonic cultural and social norms, especially in relation to sexuality, and is central to dancehall. It plays a key role in drawing these Japanese visitors to the scene and ultimately Jamaica. While most men tend to view their stay in terms of acquiring subcultural capital, the desires of women are more diverse. Their varied expectations of and experiences in Kingston's dancehall scene, and engaging with its many sites--the city, the hostels, and the clubs--are windows to questioning what Japaneseness and sociocultural belonging means to many of these individuals. Engagement with slackness ranges from a brief respite to a complete break from convention and national expectations in Japan. In the end, for the dedicated follower, dancehall acts as a resistance to or even insulates individuals from dominant cultural expectations. Abstract

BackgroundPreclinical studies have shown synergy between radiation therapy and immunotherapy. However, in almost all preclinical models, radiation is delivered in single doses or short courses of high doses (hypofractionated radiation). By contrast in most clinical settings, radiation is delivered as standard small daily fractions of 1.8-2 Gy to achieve total doses of 50-54 Gy (fractionated radiation). We do not yet know the optimal dose and scheduling of radiation for combination with chemotherapy and immunotherapy.MethodsTo address this, we analyzed the effect of neoadjuvant standard fractionated and hypofractionated chemoradiation on immune cells in patients with locally advanced and borderline resectable pancreatic adenocarcinoma.ResultsWe found that standard fractionated chemoradiation resulted in a significant and extended loss of lymphocytes that was not explained by a lack of homeostatic cytokines or response to cytokines. By contrast, treatment with hypofractionated radiation therapy avoided the loss of lymphocytes associated with conventional fractionation.ConclusionHypofractionated neoadjuvant chemoradiation is associated with reduced systemic loss of T cells.Trial registrationClinicalTrials.gov NCT01342224, April 21, 2011; NCT01903083, July 2, 2013.

Abstract

Ocrelizumab (OCR) is a disease-modifying drug used for treatment of the neurologic disease relapsing-remitting multiple sclerosis (RRMS). Epstein-Barr virus (EBV) express different immune evasion mechanisms in the host, and a correlation between various virus and, in particular EBV, and MS development has been shown. Some studies have even assigned the effect of OCR on patients with MS to be caused by the removal of EBV-infected B cells. Based on this, the aim of the study was to investigate the effect of OCR on virus antibody (Ab) levels in patient with MS and to evaluate the relation between EBV and MS compared to other viruses. Ab levels to EBV nuclear antigen (EBNA)1, cytomegalovirus (CMV) pp52 antigens, human herpes virus (HHV) 6A polymerase processivity factor p41, and severe acute respiratory syndrome coronavirus (SARS- CoV)-2 spike (S) protein were investigated by enzyme-linked immunosorbent assays (ELISA). Samples from patients with MS (n = 42) were divided into baseline (-OCR) (n = 13) and patients with MS treated with OCR (+OCR) (n = 29) and compared to healthy controls (HCs) (n = 15). Furthermore, intrathecal synthesis of EBV EBNA1 and SARS-CoV-2 S protein Abs was investigated using ELISA in samples from patients with RRMS (n = 11) and patient with the onset symptom optic neuritis (ON) (n = 8). Finally, plasma samples from three HCs and patients with a clinically isolated symptom were used to validate the conjugation of Fluorescein isothiocyanate (FITC) to a programmed cell death protein 1 (PD-1) monoclonal Ab (mAb) in a validated Fluorescence-activated Cell Sorting (FACS) panel. At baseline SARS-CoV-2 S protein IgG and EBV EBNA1 IgG were elevated in patients with RRMS compared to HCs, whereas no significant difference was observed for CMV pp52 IgG and HHV-6 p41 IgG between the two groups. Abstract

Self-assembled hyaluronic acid-based nanogels are versatile drug carriers due to their biodegradable nature and gentle preparation conditions, making them particularly interesting for delivery of peptide therapeutics. This study aims to elucidate the relation between peptide structure and encapsulation in a nanogel. Key peptide properties that affect encapsulation in octenyl succinic anhydride-modified hyaluronic acid nanogels were identified as we explored the effect on nanogel characteristics using 12 peptides with varying charge and hydrophobicity. The size and surface properties of the microfluidics-assembled peptide-loaded nanogels were evaluated using dynamic light scattering, laser Doppler electrophoresis, and small angle neutron scattering. Additionally, the change in peptide secondary structure upon encapsulation in nanogels, their release from the nanogels, and the in vitro antimicrobial activity were assessed. In conclusion, the more hydrophobic peptides showed stronger binding to the nanogel carrier and localized internally rather than on the surface of the nanogel, resulting in more spherical nanogels with smoother surfaces and slower release profiles. In contrast, cationic and hydrophilic peptides localized at the nanogel surface resulting in fluffier nanogel structures and quick and more complete release in biorelevant medium. These findings emphasize that the advantages of nanogel delivery systems for different applications depend on the therapeutic peptide properties.

Abstract

IntroductionRoutine lymphadenectomy in the surgical treatment of intrahepatic cholangiocarcinoma (ICC) is not routinely performed. We aim to define predictive indicators of survival in patients with positive lymph nodes.MethodsThe National Cancer Data Base (NCDB) was queried for patients who underwent major hepatectomy for ICC between 1998 and 2011. Clinical and pathologic data were assessed using uni- and multi-variate analyses. A sub-analysis was performed on the 160 patients with positive lymph nodes.ResultsOf 849 patients with lymph node data, 57% had at least one lymph node examined. Median survival for lymph node negative patients was 37 months versus 15 months for lymph node positive patients. In lymph node positive patients, poorer survival was associated with not receiving chemotherapy (HR 1.83, p = 0.003), tumor size > 5 cm (p = 0.029), and older age (p < 0.0001). Lymph node positive patients age less than 45 had a median survival of 27 months.ConclusionsOverall survival in patients with lymph node metastases from ICC is poor. Adjuvant therapy was associated with a longer survival in lymph node positive patients, although prospective data are needed. Routine lymphadenectomy should be strongly considered to provide prognostic information and guidance for adjuvant therapy.
