Abstract Background: Microvascular invasion (MVI) is an independent risk factor of poor prognosis in hepatocellular carcinoma (HCC) and can be used to guide the diagnosis and treatment of HCC. The immune system serves as an integral role in the incidence and progression of HCC. However, the molecular biology correlation between MVI and tumor immunity and the value of combining the two parameters to predict patient prognosis and HCC response to treatment remain to be evaluated. Results: In this study, we used univariate Cox regression analysis and least absolute shrinkage and selection operator (LASSO) Cox analysis to establish the MVI and immune-related gene index (MIRGPI) including eight genes. We demonstrated that the MIRGPI was an independent risk factor in predicting the prognosis of HCC. Subsequently, our research established a nomogram model combining pathologic characteristics and verified its good clinical application value. In addition, our study found that the TP53 gene had a higher mutation frequency and a lower degree of immune infiltration in the high-risk group. The low-risk group had higher sensitivity to immunotherapy, sorafenib, and TACE treatment, and the high-risk group had higher sensitivity to common chemotherapeutic agents. Finally, SEMA3C was found to facilitate the proliferation, migration and invasive ability of HCC by in vitro and in vivo experiments, and its mechanism may be associated with the activation of the NF-Κb/EMT signaling pathway. Conclusions: In summary, the MIRGPI signature we developed is a reliable marker for the prediction of prognosis and treatment response, and is important for the prognostic assessment and individualized treatment of HCC.

Abstract Hepatocellular carcinoma (HCC) is the most common malignancy in the liver, with a poor prognosis. Transmembrane protein 209 (TMEM209) involves multiple biological processes, such as substance transportation and signal transduction, and is abundantly expressed in tumor tissues. However, the relationship between TMEM209 and HCC has not been comprehensively elucidated. In this study, we aimed to illustrate this issue by in vitro and in vivo experiments. Bioinformatic analysis and clinical sample validation revealed that TMEM209 was upregulated in HCC and correlated with reduced survival duration. Functionally, TMEM209 promoted the proliferation, migration, invasion, and EMT of HCC cells in vitro and facilitated tumor growth and metastasis in xenograft models. Mechanistically, TMEM209 promoted the proliferation and metastasis of HCC in a KPNB1-dependent manner. Specifically, TMEM209 could bind to KPNB1, thereby competitively blocking the interaction between KPNB1 and the E3 ubiquitin ligase RING finger and CHY zinc finger domain-containing protein 1 (RCHY1) and preventing K48-associated ubiquitination degradation of KPNB1. Ultimately, the Wnt/β-catenin signaling pathway was activated, contributing to the progression of the malignant phenotype of HCC. In conclusion, the molecular mechanism underlying the TMEM209/KPNB1/Wnt/β-catenin axis in HCC progression was elucidated. TMEM209 is a potential biomarker and therapeutic target for HCC.

Abstract Dysnatremia is common in donors and recipients of liver transplantation (LT). However, the influence of dysnatremia on LT prognosis remains controversial. This study aimed to investigate effects of donors’ and recipients’ serum sodium on LT prognosis. We retrospectively reviewed 248 recipients who underwent orthotopic LT at our center between January 2016 and December 2018. Abstract Objective To evaluate and compare the measurement properties and consistency between the Chinese versions of EQ-5D-3L and EQ-5D-Y-3L among Chinese adolescent populations aged 15–17 years. Methods Chinese adolescents aged 15–17 studying in high school were recruited through online survey. Social-demographic characteristics and self-reported EQ-5D-3L and EQ-5D-Y-3L responses were collected in the survey. The consistency of responses between the two measures was assessed using redistribution property, and the consistency of utility values was assessed by intraclass correlation coefficients (ICC). Convergent validity and known-group validity were examined using Spearman’s rank correlation, F-test and effect sizes, respectively. Sensitivity was compared using relative efficiency (RE). Results 762 respondents (48.8% male; age 15–17 years;) were recruited. The EQ-5D-3L showed a more severe ceiling effect than EQ-5D-Y-3L (78.2% vs. 66.0%). Respondents reported higher proportions of having problems in four dimensions using the EQ-5D-Y-3L than using the EQ-5D-3L. The consistency of corresponding dimensions between the two measures was relatively good, while non-negligible proportions of inconsistency were observed in “pain/discomfort” (11.4%) and “anxiety/depression” (15.7%) dimensions. Landslide susceptibility maps (LSMs) play an important role in landslide hazard risk assessments, urban planning, and land resource management. While states of motion and dynamic factors are critical in the landslide formation process, these factors have not received due attention in existing LSM-generation research. In this study, we proposed a valuable method for dynamically updating and refining LSMs by combining soil moisture products with Multi-Temporal Interferometric Synthetic Aperture Radar (MT-InSAR) data. Based on a landslide inventory, we used time-series soil moisture data to construct an index system for evaluating landslide susceptibility. MT-InSAR technology was applied to invert the displacement time series. Furthermore, the surface deformation rate was projected in the direction of the steepest slope, and the data was resampled to a spatial resolution consistent with that of the LSM to update the generated LSM. The results showed that varying soil moisture conditions were accompanied by dynamic landslide susceptibility. A total of 22% of the analyzed pixels underwent significant susceptibility changes (either increases or decreases) following the updating and refining processes incorporating soil moisture and MT-InSAR compared to the LSMs derived based only on static factors. The relative landslide density index obtained based on actual landslides and the analyses of Dongfeng, Haila town, and Dajie township confirmed the improved slow landslide prediction reliability resulting from the reduction of the false alarm and omission rates.

Earthquakes cause a huge number of landslides and alter the regional landslide risk distribution. As a result, after a significant earthquake, the landslide susceptibility maps (LSMs) must be updated. The study goal was to create seismic landslide susceptibility maps containing landslide causative variables which are adaptable to great changes in susceptibility after the Jiuzhaigou earthquake (MS 7.0) and to perform a rapid update of the LSM after the earthquake by means of the distributed scatterer interferometric synthetic aperture radar (DS-InSAR) technique. We selected the territory of Jiuzhaigou County (southwestern China) as the study region. Jiuzhaigou is a world-renowned natural heritage and tourist area of great human and ecological value. For landslide susceptibility mapping, we examined the applicability of three models (logistic regression, support vector machine, and random forest) for landslide susceptibility mapping and offered a strategy for updating seismic landslide susceptibility maps using DS-InSAR. First, using logistic regression, support vector machine, and random forest techniques, susceptibility models of seismic landslides were built for Jiuzhaigou based on twelve contributing variables. Second, we obtained the best model parameters by means of a Bayesian network and network search, while using five-fold cross-validation to validate the optimized model. According to the receiver operating characteristic curve (ROC), the SVM model and RF model had excellent prediction capability and strong robustness over large areas compared with the LR models. Third, the surface deformation in Jiuzhaigou was calculated using DS-InSAR technology, and the deformation data were adopted to update the landslide susceptibility model using the correction matrix. The correction of deformation data resulted in a susceptibility class transition in 4.87 percent of the research region. According to practical examples, this method of correcting LSMs for the continuous monitoring of surface deformation (DS-InSAR) was effective. Finally, we analyze the reasons for the change in the revised LSM and point out the help of ecological restoration in reducing landslide susceptibility. The results show that the integration of InSAR continuous monitoring not only improved the performance of the LSM model but also adapted it to track the evolution of future landslide susceptibility, including seismic and human activities.

The transmission distance of transmission lines is long, the line affected by the diversity of climate and topography of the corridors of transmission lines, differences in regional geological structure conditions, variability of rock and soil types, and the complexity of groundwater. Under the influence of extreme weather conditions (ice-covered, strong wind, etc.) and sudden geological disasters (such as mudslides, flash floods, earthquakes, etc.), catastrophic damage and basic deformation problems of the tower foundations are prone, and even tower collapse accidents occur in severe cases, which affect the safe operation of transmission lines. Monitoring the deformation of power transmission towers and surrounding grounds, it is critical to ensuring the normal operation of transmission lines by assessing and controlling potential risks in advance. In this paper, using ALOS-2 PALSAR radar satellite data, differential interferometry was used to monitor surface deformation near the Sichuan Jinsu line transmission channel. The analysis found that a significant landslide hazard was found near the transmission channel tower in Yibin-Zhaotong section of Jinsu, Sichuan Province, the cumulative deformation reaches 9cm.
