In this study, a novel nanocomposite of graphene oxide-zinc oxide-manganese (GO-ZnO-M) was successfully synthesized using a modified Hummers method and hydrothermal technique. Enhanced electrochemical performance was observed in the GO-ZnO-M nanocomposite when utilized as an electrode material for the detection of rutin, a flavonoid known for its significant health benefits. The GO-ZnO-M modified electrode demonstrated excellent sensitivity, selectivity, reproducibility, and stability during rutin detection. Furthermore, the sensor exhibited a wide linear detection range and a low detection limit. The practical applicability of the GO-ZnO-M/GCE sensor was assessed by detecting rutin in pharmaceutical tablet samples, yielding satisfactory results. This study highlights the potential of GO-ZnO-M nanocomposites as promising materials for the electrochemical detection of rutin and other biologically significant compounds.

Peripheral tumor-specific CD8+ T cells often fail to infiltrate into tumor parenchyma due to the immunosuppression of tumor microenvironment (TME). Meanwhile, a significant portion of tumor-specific CD8+ T cells infiltrated into TME are functionally exhausted. Despite the enormous success of anti-PD-1/PD-L1 immune-checkpoint blockade (ICB) treatment in a wide variety of cancer types, the majority of patients do not respond to this treatment largely due to the failure to efficiently drive tumor-specific CD8+ T cell infiltration and reverse their exhaustion states. Nowadays, tumor cell pyroptosis, a unique cell death executed by pore-forming gasdermin (GSDM) family proteins dependent or independent on inflammatory caspase activation, has been shown to robustly promote immune-killing of tumor cells by enhancing tumor immunogenicity and altering the inflammatory state in the TME, which would be beneficial in overcoming the shortages of anti-PD-1/PD-L1 ICB therapy. Therefore, in this review we summarize the current progresses of tumor cell pyroptosis in enhancing immune function and modulating TME, which synergizes anti-PD-1/PD-L1 ICB treatment to achieve better anti-tumor effect. We also enumerate several strategies to better amply the efficiency of anti-PD-1/PD-L1 ICB therapy by inducing tumor cell pyroptosis.

BackgroundSurgery is the main treatment for recurrent patellar dislocation (PD). However, due to the complexity of anatomical factors, there is still a lack of consensus on the choice of combined surgical methods. This study aimed to compare the clinical and radiological outcomes of medial patellofemoral ligament reconstruction combined with derotational distal femur osteotomies (MPFLR + DDFO) and combined with tibial tubercle osteotomies (MPFLR + TTO) for recurrent PD with increased femoral anteversion angles (FAA) and excessive tibial tubercle-trochlear groove (TT-TG) distance.MethodsIn this retrospective analysis, MPFLR + DDFO and MPFLR + TTO patients from 2015 to 2020 were included. Group A (MPFLR + DDFO, n = 42) and B (MPFLR + TTO, n = 46) were formed. Clinical outcomes included physical examinations, functional outcomes (Kujala, Lysholm, International Knee Documentation Committee (IKDC), visual analog scale (VAS) and intermittent and persistent osteoarthritis pain scale (ICOAP), Tegner scores), and complications. The Caton-Deschamps index (CD-I), patellar title angle, patellar congruence angle, patella-trochlear groove distance, TT-TG distance, and FAA were used to assess radiological outcomes.ResultsAll clinical outcomes improved significantly in both groups, but Group A had significantly better postoperative scores than Group B (Kujala: 89.8 ± 6.4 vs. 82.9 ± 7.4, P

A Cable-Driven Continuum Robot (CDCR) that consists of a set of identical Cable-Driven Continuum Joint Modules (CDCJMs) is proposed in this paper. The CDCJMs merely produce 2-DOF bending motions by controlling driving cable lengths. In each CDCJM, a pattern-based flexible backbone is employed as a passive compliant joint to generate 2-DOF bending deflections, which can be characterized by two joint variables, i.e., the bending direction angle and the bending angle. However, as the bending deflection is determined by not only the lengths of the driving cables but also the gravity and payload, it will be inaccurate to compute the two joint variables with its kinematic model. In this work, two stretchable capacitive sensors are employed to measure the bending shape of the flexible backbone so as to accurately determine the two joint variables. Compared with FBG-based and vision-based shape-sensing methods, the proposed method with stretchable capacitive sensors has the advantages of high sensitivity to the bending deflection of the backbone, ease of implementation, and cost effectiveness. The initial location of a stretchable sensor is generally defined by its two endpoint positions on the surface of the backbone without bending. A generic shape-sensing model, i.e., the relationship between the sensor reading and the two joint variables, is formulated based on the 2-DOF bending deflection of the backbone. To further improve the accuracy of the shape-sensing model, a calibration method is proposed to compensate for the location errors of stretchable sensors. Based on the calibrated shape-sensing model, a sliding-mode-based closed-loop control method is implemented for the CDCR. In order to verify the effectiveness of the proposed closed-loop control method, the trajectory tracking accuracy experiments of the CDCR are conducted based on a circle trajectory, in which the radius of the circle is 55mm. The average tracking errors of the CDCR measured by the Qualisys motion capture system under the open-loop and the closed-loop control are 49.23 and 8.40mm, respectively, which is reduced by 82.94%.

The absence of a conducive bone formation microenvironment between fractured ends poses a significant challenge in repairing large bone defects. A promising solution is to construct a bone formation microenvironment that mimics natural bone tissue. Biomimetic mineralized collagen possesses a chemical composition and microstructure highly similar to the natural bone matrix, making it an ideal biomimetic bone substitute material. The microstructure of biomimetic mineralized collagen is influenced by various factors, and its biomineralization and microstructure, in turn, affect its physicochemical properties and biological activity. We aimed to utilize mineralization time and solution concentration as variables and employed the polymer-induced liquid precursor strategy to fabricate mineralized collagen with diverse microstructures, to shed light on how mineralization parameters impact the material microstructure and physicochemical properties. We also investigated the influence of microstructure and physicochemical properties on cell biocompatibility and the bone-forming microenvironment. Through comprehensive characterization, we examined the physical and chemical properties of I-EMC under various mineralization conditions and assessed the in vitro and in vivo biocompatibility and osteogenic performance. By investigating the relationship between mineralization parameters, material physicochemical properties, and osteogenic performance, we revealed how microstructures influence cellular behaviors like biocompatibility and osteogenic microenvironment. Encouragingly, mineralization solutions with varying concentrations, stabilized by polyacrylic acid, successfully produced intrafibrillar and extrafibrillar mineralized collagen. Compared to non-mineralized collagen, all mineralized samples demonstrated improved bone-forming performance. Notably, samples prepared with a 1× mineralization solution exhibited relatively smooth surfaces with even mineralization. Extending the mineralization time enhanced the degree of mineralization and osteogenic performance. Conversely, samples prepared with a 2× mineralization solution had rough surfaces with large calcium phosphate particles, indicating non-uniform mineralization. Overall, our research advances the potential for commercial production of mineralized collagen protein products, characterized by dual biomimetic properties, and their application in treating various types of bone defects.

The growth plate is a cartilaginous tissue with three distinct zones. Resident chondrocytes are highly organized in a columnar structure, which is critical for the longitudinal growth of immature long bones. Once injured, the growth plate may potentially be replaced by bony bar formation and, consequently, cause limb abnormalities in children. It is well-known that the essential step in growth plate repair is the remolding of the organized structure of chondrocytes. To achieve this, we prepared an anatomy-inspired bionic Poly(ε-caprolactone) (PCL) scaffold with a stratified structure using three-dimensional (3D) printing technology. The bionic scaffold is engineered by surface modification of NaOH and collagen Ⅰ (COL Ⅰ) to promote cell adhesion. Moreover, chondrocytes and bone marrow mesenchymal stem cells (BMSCs) are loaded in the most suitable ratio of 1:3 for growth plate reconstruction. Based on the anatomical structure of the growth plate, the bionic scaffold is designed to have three regions, which are the small-, medium-, and large-pore-size regions. These pore sizes are used to induce BMSCs to differentiate into similar structures such as the growth plate. Remarkably, the X-ray and histological results also demonstrate that the cell-loaded stratified scaffold can successfully rebuild the structure of the growth plate and reduce limb abnormalities, including limb length discrepancies and angular deformities in vivo. This study provides a potential method of preparing a bioinspired stratified scaffold for the treatment of growth plate injuries.

Abstract To efficiently combat viral infectious diseases, it is important to develop broadly applicable countermeasures, and efficient antiviral systems can be developed by elaborating the relationship of antiviral efficiency with the interactions between antiviral agents and viruses. In the present study, conjugated polymer (CP)-based photodynamic therapy was used to inhibit RNA virus infections. A severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pseudotyped virus composed of an SARS-CoV-2 envelope coated with the S protein and luciferase RNA genome was employed to assess antiviral efficiency. Three cationic CPs with different backbone structures, fluorene-co-phenylene (PFP), thiophene (PMNT), and phenylene vinylene (PPV), exhibit different photoinactivation effects. The highly efficient photoinactivation of PPV and PMNT is derived from the complete photodegradation of spike proteins, nucleocapsid proteins and nucleic acids of SARS-CoV-2 after binding to the viral spike proteins. Although PFP showed the highest efficiency in the photodegradation of spike proteins due to its strong binding affinity, ineffective viral inhibition was observed, which occurred because the viral gene was partially damaged under light irradiation and the process of delivering the viral gene to cells received assistance. This work preliminarily reveals the effect of CP-virus interactions on their photoinactivation activity and should be beneficial for further research on the development of highly efficient antiviral PDT agents.

Combustion characteristics in a scramjet combustor equipped with a thin strut were observed and discussed in this paper. A series of numerical simulations were carried out under different flight dynamic pressure conditions. The parameters of cold flow field and combustion field were used to analyze the combustion characteristics. Based on the basic data, the mixing efficiency, characteristics of flame establishment and propagation as well as combustion field characteristics were discussed in this paper. The influence laws of lower dynamic pressure conditions were further revealed to optimize combustor performance. Results indicated that properly reducing the flight dynamic pressure can enhance the mixing of kerosene. The diffusion of kerosene determined the distribution of combustion zone and heat release. Then, the influencing factor that affected the chemical reaction rate was revealed to shorten chemical reaction time. And the higher flight Mach number made the flame propagation velocity faster and the combustion stability stronger. The fuel mixing became the main factor and low dynamic pressure had little effect on laminar flame propagation velocity under high Mach number conditions. The investigations in this paper are helpful for understanding the combustion characteristics under low dynamic pressure conditions.

Human heel pads commonly undergo cyclic loading during daily activities. Low cyclic loadings such as daily human walking tend to have less effect on the mechanical properties of heel pads. However, the impact of cyclic loading on cushion performance, a vital biomechanical property of heel pads, under engineering test condition remains unexplored. Herein, dynamic mechanical measurements and finite element (FE) simulations were employed to explore this phenomenon. It was found that the wavy collagen fibers in the heel pad will be straightened under cycle compression loading, which resulted in increased stiffness of the heel pad. The stiffness of the heel pads demonstrated an inclination to escalate over a span of 50,000 loading cycles, consequently resulting in a corresponding increase in peak impact force over the same loading cycles. Sustained cyclic loading has the potential to result in the fracturing of the straightened collagen fibers, this collagen breakage may diminish the stiffness of the heel pad, leading to a reduction in peak impact force. This work enhances understanding of the biomechanical functions of human heel pad and may provide potential inspirations for the innovative development of healthcare devices for foot complex.

Bisphosphonates (BPs), the stable analogs of pyrophosphate, are well-known inhibitors of osteoclastogenesis to prevent osteoporotic bone loss and improve implant osseointegration in patients suffering from osteoporosis. Compared to systemic administration, BPs-incorporated coatings enable the direct delivery of BPs to the local area, which will precisely enhance osseointegration and bone repair without the systemic side effects. However, an elaborate and comprehensive review of BP coatings of implants is lacking. Herein, the cellular level (e.g., osteoclasts, osteocytes, osteoblasts, osteoclast precursors, and bone mesenchymal stem cells) and molecular biological regulatory mechanism of BPs in regulating bone homeostasis are overviewed systematically. Moreover, the currently available methods (e.g., chemical reaction, porous carriers, and organic material films) of BP coatings construction are outlined and summarized in detail. As one of the key directions, the latest advances of BP-coated implants to enhance bone repair and osseointegration in basic experiments and clinical trials are presented and critically evaluated. Finally, the challenges and prospects of BP coatings are also purposed, and it will open a new chapter in clinical translation for BP-coated implants.

Mineralized collagen (MC) is the basic unit of bone structure and function and is the main component of the extracellular matrix (ECM) in bone tissue. In the biomimetic method, MC with different nanostructures of neo-bone have been constructed. Among these, extra-fibrous MC has been approved by regulatory agencies and applied in clinical practice to play an active role in bone defect repair. However, in the complex microenvironment of bone defects, such as in blood supply disorders and infections, MC is unable to effectively perform its pro-osteogenic activities and needs to be functionalized to include osteogenesis and the enhancement of angiogenesis, anti-infection, and immunomodulation. This article aimed to discuss the preparation and biological performance of MC with different nanostructures in detail, and summarize its functionalization strategy. Then we describe the recent advances in the osteo-inductive properties and multifunctional coordination of MC. Finally, the latest research progress of functionalized biomimetic MC, along with the development challenges and future trends, are discussed. This paper provides a theoretical basis and advanced design philosophy for bone tissue engineering in different bone microenvironments.

Rigid robots have found wide-ranging applications in manufacturing automation, owing to their high loading capacity, high speed, and high precision. Nevertheless, these robots typically feature joint-based drive mechanisms, possessing limited degrees of freedom (DOF), bulky structures, and low manipulability in confined spaces. In contrast, continuum robots, drawing inspiration from biological structures, exhibit characteristics such as high compliance, lightweight designs, and high adaptability to various environments. Among them, cable-driven continuum robots (CDCRs) driven by multiple cables offer advantages like higher dynamic response compared to pneumatic systems and increased working space and higher loading capacity compared to shape memory alloy (SMA) drives. However, CDCRs also exhibit some shortcomings, including complex motion, drive redundancy, challenging modeling, and control difficulties. This study presents a comprehensive analysis and summary of CDCR research progress across four key dimensions: configuration design, kinematics and dynamics modeling, motion planning, and motion control. The objective of this study is to identify common challenges, propose solutions, and unlock the full potential of CDCRs for a broader range of applications.

The influence of landscape structure on species communities is intimately connected to the spatial scales at which measurements are made. If we do not measure landscape structure at its most impactful scale, otherwise known as the “scale of effect”, we might fail to detect crucial community–landscape relationships. In our study, we focused on the “scale of effect” as it pertains to the relationship between urban bird diversity and landscape structure. We investigated eight types of landscape structure variables alongside elevation attributes across 16 spatial scales around 28 sampling sites in Kunming, Yunnan Province, China. Our objective was to identify the most influential scale at which the environmental variables under investigation affect the taxonomic and functional diversity of bird species. We utilized PLSR and VIP scores to overcome challenges posed by multicollinearity among predictors. We found that the influence of landscape characteristics on bird species richness and functional dispersion index (FDis) was scale-dependent. Notably, the influence of the area of open water on FDis is most pronounced at smaller scales, while at larger scales, open water dispersion becomes more essential. Additionally, in a highly fragmented urban matrix, the importance of forest connectivity may exceed that of patch size at medium scales. However, the significance of forest coverage increases with scale, which underscores the importance of preservation efforts at larger scales to prevent forest fragmentation. We also noted a considerable impact from landscape-level subdivision across almost all scales. These findings highlight the importance of shifting urban conservation planning towards a multi-scale approach, which would allow for the identification of priority intervention scales.

Background: Dissociation of the polyethylene liner after reverse shoulder arthroplasty could cause shoulder dislocation that could not achieve closed reduction. The cause of liner dissociation is currently unclear.Method: Non-homogeneous model of the bone was constructed and dynamic finite element analysis was utilized to simulate the impingement of the polyethylene liner and scapula during humeral adduction. The stress distribution of the fixation claws, their degree of deformation (DOD), and the stress of the impingement sites in three initial humeral postures (neutral, 30° flexion, and 30° extension) were measured and analyzed. The influence of the liner material stiffness was also investigated.Result: The impingement stress on the liner and scapula was 100–200 MPa, and different humeral postures caused different locations of impingement points. The fixation claws’ maximum principal stress (MPS) results were below 5 MPa. In the connection area between some fixation claws and the liner, compressive stresses on the inside and tensile stresses on the outside were observed, which showed that the fixation claws were prone to deform toward the center direction. The maximum DOD results of three initial humeral postures (neutral, 30° flexion, and 30° extension) were 3.6%, 2.8%, and 3.5%, respectively. The maximum DOD results of neutral initial humeral posture were 0.51% and 11.4% when the elastic modulus of the liner was increased and decreased by a factor of 10, respectively.Conclusion: The humeral adduction impingement could lead to the deformation of the claw-shaped liner fixation structure, which might be one of the reasons for the liner dissociation. The increased stiffness of the liner material helped to reduce the deformation of the fixation structure.

Abstract Background Gas-producing perianal abscess raises the possibility of clostridial infection, with Clostridium perfringens being the most common causative agent, which is highly lethal if untreated timely. As the treatment of clostridial infections often differs from that of non-clostridial infections, which they may closely resemble, the importance of accurate pathogenic organism identification cannot be overemphasized. The 16S rDNA of bacteria is highly conserved within a species and among species of the same genus but demonstrates substantial variation between different species, thus making it a suitable genomic candidate for bacterial detection and identification. Case presentation Here, we report the case of a 53-year-old patient who was admitted to the hospital for a gas-producing perianal abscess. The patient was managed with ceftizoxime and ornidazole and then received debridement and drainage at the lesion on the second day after admission. The bacterial cultures of the patient isolates from the debridement showed a coinfection of Escherichia coli and Enterococcus faecium. Although perianal redness and swelling subsided obviously after the surgery, the patient was febrile to 38.3℃ with his left upper thigh red and swollen, aggravated with tenderness and crepitus. Considering insufficient debridement and the risk of incorrect identification of pathogens, a second debridement and drainage were performed 4 days after the primary operation, and 16S rDNA sequencing of the isolates implicated Clostridium perfringens infection. Given the discrepancies in diagnostic results and the treatment outcomes, Enterococcus faecium was identified as sample contamination, and a diagnosis of coinfection of Clostridium perfringens and Escherichia coli in gas-producing perianal abscess was confirmed. The patient was then successfully treated with meropenem and vancomycin and was discharged at 27 days of admission. Conclusions This case represents the first report of coinfection of both clostridial and non-clostridial organisms in gas-producing perianal abscess and the first case reporting the use of 16S rDNA sequencing in the diagnosis of perianal abscess. Timely pathogen identification is critical for treating gas-producing perianal abscess and an antibiotic regimen covering both aerobic and anaerobic organisms is recommended before true pathogens are identified.

Abstract Heavy metal Cadmium (Cd2+) pollution has become a severe environmental problem for aquatic organisms. In crustaceans, gills (Gi) and hepatopancreas (Hp) play a vital role in the toxicology. However, in Macrobrachium rosenbergill, there are few researches about gill and hepatopancreases responding to Cd2+ stress at a molecular level. In this study, transcriptomic analysis was applied to characterize gene expression profiles of gills and hepatopancreas of M. rosenbergill after Cd2+ exposure for 0 h, 3 h and 3 d. Six cDNA libraries (Gi 0 h, Gi 3 h, Gi 3 d, Hp 0 h, Hp 3 h, and Hp 3 d) were constructed and a total of 66,676 transcripts and 48,991 unigenes were annotated. Furthermore, differentially expressed genes (DEGs) were isolated by comparing the Cd2+ treated time-point libraries (3 h and 3 d group) with the control library (0 h group). The results showed that most of the DEGs were down-regulated after Cd2+ exposure and the number of DEGs among gill groups were significantly higher than those among hepatopancreas groups. GO functional and KEGG pathway analysis suggested many key DEGs in response to the Cd2+ stress, such as metallothionein and Hemocyanin. Additionally, a total of six DEGs were randomly selected to further identify their expressional profile by qPCR. The results indicated that these DEGs were involved in the response to Cd2+. This comparative transcriptome provides valuable molecular information on the mechanisms of responding to Cd2+ stress in M. rosenbergii, which lays the foundation for further understanding of heavy metal stress.

The loosening of traditional prosthetics is among the leading causes of surgical failure of proximal femoral bone defects. A novel compound sleeve and stem prosthesis was designed using an optimization methodology that combined an octet-truss porous structure with density-based topology optimization to improve stability, promote bone ingrowth, and enhance biomechanical properties. Biomechanical changes were assessed using finite element analysis. The distribution of stress, the strain energy density, and the relative micromotion in the optimized group were considered. The optimized sleeve prosthesis achieved a 31.5% weight reduction. The maximum stresses in the optimized group were observed to decrease by 30.33 and 4.74% at the back sleeve and neck part of stem prosthesis, with a 29.52% increase in the femur, respectively. The average stress in most selected regions in the optimized group was significantly greater than that in the original group (p < 0.05). The maximum relative micromotion decreased by 15.18% (from 63.9 to 54.2 μm) in the optimized group. The novel designed compound sleeve and stem prosthesis could effectively improve the biomechanical performance of next-generation prosthetics and provide a microenvironment for bone ingrowth. The presented method could serve as a model for clinical practice and a platform for future orthopedic surgery applications.

Abstract Photosynthesis is promising in sequestrating carbon dioxide and providing food and biofuel. Recent findings have shown that luminescent materials could shift the wavelength of light to a more usable range for augmented photosynthesis. Among them, aggregation‐induced emission luminogens (AIEgens) have advantages of efficient light conversion, high biocompatibility, large Stokes’ shift, and so on. In this perspective, emerging reports of augmented photosynthesis with luminescent materials, especially the AIEgens are included. We emphasized the spectra shift characteristics, material formation, and sustainable development based on augmented photosynthesis.

Several studies have revealed that urban rivers with relatively complete habitat structures can support a high level of biodiversity in urban areas. However, few studies have analyzed the impact of the plan, design, and management of multi-scale urban riverside green space on bird communities by assessing several aspects of diversity (e.g., taxonomic and functional). In this study, we explored bird community variation (taxonomic and functional) among three rivers (28 sampling sites) with different levels of human disturbance along the urban development gradient in Kunming, Yunnan Province, China, during an annual cycle. This study related bird species richness and functional diversity to environmental variables at three spatial scales: landscape metrics at two different landscape scales (500 and 100 m radius) and vegetation structure at patch scales (20 m radius). Using partial least squares regression (PLSR) and multiple linear regression, we found that bird species richness and functional diversity were strongly related to various landscape- and patch-level variables. Multiple linear regression models revealed that, compared with the richness influenced by explanatory variables on three spatial scales (500, 100, and 20 m), the functional diversity was significantly influenced by explanatory variables on the 100 and 20 m scales. Tree cover at the 20 m scale had significantly positive impacts on bird species richness and functional diversity. Forest land in the 100 m scale had a significantly positive effect on bird species richness and functional diversity, while construction land in the two landscape scales (the 500- and 100 m scales) had a significantly negative effect on species richness. Accordingly, we suggest that urban riverside green space should be planned and managed at multiple scales.

Abstract Appropriate drainage duration is vital for the postoperative rehabilitation of patients with breast cancer (BC) undergoing modified radical mastectomy (MRM). To provide better and individualized postoperative management for these patients, this study explored independent predictors of postoperative drainage duration in patients with BC. This was a single-center retrospective cohort study. Patients diagnosed with BC and treated with MRM from May 2016 to April 2020 were randomly divided into training (n = 729) and validation (n = 243) cohorts. Univariate and multivariate Cox analyses revealed that the body mass index, serum albumin level, hypertension, number of total dissected axillary lymph nodes, and ratio of positive axillary lymph nodes were independent predictors of postoperative drainage duration in the training cohort. Based on independent predictors, a nomogram was constructed to predict the median postoperative drainage duration and the probability of retaining the suction drain during this period. This nomogram had good concordance and discrimination both in the training and validation cohorts and could effectively predict the probability of retaining the suction drain during drainage, thus assisting clinicians in predicting postoperative drainage duration and providing individualized postoperative management for patients with BC.

Osteoporosis-derived bone marrow mesenchymal stem cells (OP-BMSCs) have impaired cell function, low osteogenic activity, and potential osteoclast inducibility, resulting in insufficient osseointegration and increased risk of catastrophic postoperative complications after arthroplasty. The low autophagy activity of OP-BMSCs contributed to poor osseointegration, which could be regulated by the classical autophagic activator, rapamycin. Hence, it is of great significance to regulate autophagy by rapamycin to promote osseointegration at the three-dimensional (3D) printed prosthetic interfaces under osteoporotic environment. In this study, 3D printed porous titanium alloy prosthetic interfaces with bionic pore size and porosity were prepared and implanted into the distal femur of osteoporosis rabbits. Subsequently, a transdermal drug delivery device was used to deliver rapamycin to the interfaces implantation sites. Ultrasound-mediated rapamycin delivery could restore the declined cellular activities (including cell viability, proliferation, migration, and osteogenesis), as well as inhibit potential osteoclast-inducing capacity and adipogenic differentiation by upregulating the autophagy level of OP-BMSCs. Moreover, percutaneous ultrasound-mediated rapamycin delivery significantly improved bone ingrowth and osseointegration of prosthetic interfaces in osteoporotic environment. To conclude, this novel therapy combining 3D printed bionic prosthesis and percutaneous ultrasound-mediated rapamycin delivery can effectively promote prosthetic interfaces osseointegration by regulating autophagy after arthroplasty for patients with osteoporosis.

Photosynthetic biohybrid systems exhibit promising performance in biosynthesis; however, these systems can only produce a single metabolite and cannot further transform carbon sources into highly valuable chemical production. Herein, a photosynthetic biohybrid system integrating biological and chemical cascade synthesis was developed for solar-driven conversion of glucose to value-added chemicals. A new ternary cooperative biohybrid system, namely bacterial factory, was constructed by self-assembling of enzyme-modified light-harvesting donor-acceptor conjugated polymer nanoparticles (D-A CPNs) and genetically engineered Escherichia coli (E. coli). The D-A CPNs coating on E. coli could effectively generate electrons under light irradiation, which were transferred into E. coli to promote the 37% increment of threonine production by increasing the ratio of nicotinamide adenine dinucleotide phosphate (NADPH). Subsequently, the metabolized threonine was catalyzed by threonine deaminase covalently linking with D-A CPNs to obtain 2-oxobutyrate, which is an important precursor of drugs and chemicals. The 2-oxobutyrate yield under light irradiation is increased by 58% in comparison to that in dark. This work provides a new organic semiconductor-microorganism photosynthetic biohybrid system for biological and chemical cascade synthesis of highly valuable chemicals by taking advantage of renewable carbon sources and solar energy.

In recent years, there has been increasing interest in using biomimetic materials for bone tissue engineering. However, the high immunogenicity and low mechanical properties of biomimetic materials limit their applications in vivo. As an essential component of the extracellular matrix in bone tissue, collagen is ideal because it easily degrades and has strong plasticity and low immunogenicity. However, pure collagen alone does not yield satisfactory experimental results. Therefore, biomimetic bone tissue engineering scaffolds are often prepared together with other bioactive materials to better replicate the bone tissue’s biological microenvironment. To better understand the engineered scaffolds, we have summarized and analyzed current research in collagen-based biomimetic bone tissue engineering. This review focuses on the acquisition and processing of collagen, the mechanisms involved in collagen-induced osteogenesis, the biomimetic materials composed of collagen and different materials, and the application of collagen-based bioactive materials in bone tissue engineering.

Tumor resection and treatment of trauma-related regional large bone defects have major challenges in the field of orthopedics. Scaffolds that treat bone defects are the focus of bone tissue engineering. 3D printing porous titanium alloy scaffolds, prepared via electron beam melting technology, possess customized structure and strength. The addition of a growth factor coating to the scaffold introduces a specific form of biological activation. Vascular endothelial growth factor (VEGF) is key to angiogenesis and osteogenesis in vivo. We designed a porous titanium alloy scaffold/thermosensitive collagen hydrogel system, equipped with VEGF, to promote local osseointegration and angiogenesis. We also verified the VEGF release via thermosensitive collagen and proliferation and induction of the human umbilical vein endothelial cells (HUVECs) via the composite system in vitro. In vivo, using microscopic computed tomography (Micro-CT), histology, and immunohistochemistry analysis, we confirmed that the composite scaffold aids in angiogenesis-mediated bone regeneration, and promotes significantly more bone integration. We also discovered that the composite scaffold has excellent biocompatibility, provides bioactive VEGF for angiogenesis and osteointegration, and provides an important theoretical basis for the restoration of local blood supply and strengthening of bone integration.

The growth plate (GP) is a cartilaginous region situated between the epiphysis and metaphysis at the end of the immature long bone, which is susceptible to mechanical damage because of its vulnerable structure. Due to the limited regeneration ability of the GP, current clinical treatment strategies (e.g., bone bridge resection and fat engraftment) always result in bone bridge formation, which will cause length discrepancy and angular deformity, thus making satisfactory outcomes difficult to achieve. The introduction of cartilage repair theory and cartilage tissue engineering technology may encourage novel therapeutic approaches for GP repair using tissue engineered GPs, including biocompatible scaffolds incorporated with appropriate seed cells and growth factors. In this review, we summarize the physiological structure of GPs, the pathological process, and repair phases of GP injuries, placing greater emphasis on advanced tissue engineering strategies for GP repair. Furthermore, we also propose that three-dimensional printing technology will play a significant role in this field in the future given its advantage of bionic replication of complex structures. We predict that tissue engineering strategies will offer a significant alternative to the management of GP injuries.

Abstract Bacteria infected cells acting as “Trojan horses” not only protect bacteria from antibiotic therapies and immune clearance, but also increase the dissemination of pathogens from the initial sites of infection. Antibiotics are hard and insufficient to treat such hidden internalized bacteria, especially multidrug‐resistant (MDR) bacteria. Herein, aggregation‐induced emission luminogens (AIEgens) such as N,N‐diphenyl‐4‐(7‐(pyridin‐4‐yl) benzo [c] [1,2,5] thiadiazol‐4‐yl) aniline functionalized with 1‐bromoethane (TBP‐1) and (3‐bromopropyl) trimethylammonium bromide (TBP‐2) (TBPs) show potent broad‐spectrum bactericidal activity against both extracellular and internalized Gram‐positive pathogens. TBPs trigger reactive oxygen species (ROS)‐mediated membrane damage to kill bacteria, regardless of light irradiation. TBPs effectively kill bacteria without the development of resistance. Additionally, such AIEgens activate mitochondria dependent autophagy to eliminate internalized bacteria in host cells. Compared to the routinely used vancomycin in clinic, TBPs demonstrate comparable efficacy against methicillin‐resistant Staphylococcus aureus (MRSA) in vivo. The studies suggest that AIEgens are promising new agents for the treatment of MDR bacteria associated infections.

Abstract Background Restoration projects have been implemented worldwide to mitigate the adverse effects of the loss and degradation of wetland habitats. Much research has been carried out on the impacts on birds of wetland restoration and management projects in China. Studies have mainly investigated central or coastal wetlands, while inland wetlands in remote areas have been much less studied. We focused on examining the response of wild birds to wetland restoration in Dianchi Lake, south-west China. Methods The line transect method was performed at 26 sampling plots. Three of these were in the city, and to acquire all wild bird data 23 plots were located every 2–8 km along the shore of Dianchi Lake, between December 2011 and November 2013. We collected all related bird records by searching the available literature, articles, newspapers and records of birdwatchers to compare species variation before and after implementation of wetland restoration. To measure the relationships between waterbird assemblages and habitat structures, we used canonical correspondence analysis (CCA) to pair the main matrix of bird assemblages with a second matrix of habitat variables. Results We recorded 182 bird species belonging to 51 families and 17 orders. Of the species, 42 were new records for Kunming City and 20 were new records for Yunnan Province. Ten waterbird species were found to have disappeared from the shore of Dianchi Lake. CCA results indicated that waterbirds could be divided into four categories based on their habitat preference: synanthropic (wintering gulls), special habitat (shorebirds), semi-natural (wintering coots and ducks) and disturbance-tolerant (resident) species. Conclusions Our study is the first to consider the entire wild bird community throughout the year and discuss the species variation before and after wetland restoration projects launched for Dianchi Lake. Distinct habitat requirements of different waterbird groups were detected in our study, suggesting different types of restoration and management should be implemented.

A diabetic foot ulcer (DFUs) is a state of prolonged chronic inflammation, which can result in amputation. Different from normal skin wounds, various commercially available dressings have not sufficiently improved the healing of DFUs. In this study, a novel self-healing hydrogel was prepared by in situ crosslinking of N -carboxyethyl chitosan ( N -chitosan) and adipic acid dihydrazide (ADH) with hyaluronic acid-aldehyde (HA-ALD), to provide a moist and inflammatory relief environment to promote stem cell proliferation or secretion of growth factors, thus accelerating wound healing. The results demonstrated that this injectable and self-healing hydrogel has excellent swelling properties, stability, and mechanical properties. This biocompatible hydrogel stimulated secretion of growth factors from bone marrow mesenchymal stem cells (BM-MSCs) and regulated the inflammatory environment by inhibiting the expression of M1 macrophages and promoting the expression of M2 macrophages, resulting in granulation tissue formation, collagen deposition, nucleated cell proliferation, neovascularization, and enhanced diabetic wound healing. This study showed that N -chitosan/HA-ALD hydrogel could be used as a multifunctional injectable wound dressing to regulate chronic inflammation and provide an optimal environment for BM-MSCs to promote diabetic wound healing.

Patients with osteoporosis are more likely to suffer prosthesis displacement, subsidence or periprosthesis fracture after prosthesis replacement. This is due to excessive osteoclast activity and a lack of bone marrow mesenchymal stem cell (BMSC) integration at the implant-bone interface locally in the setting of osteoporosis. In order to inhibit local excessive bone resorption and promote bone integration, we have constructed a porous titanium/poloxamer 407 hydrogel system loaded with zoledronate (ZOL), a bisphosphonate, to promote osteointegration in a rabbit model of osteoporosis. This composite scaffold (ZOL/gTi) showed good biocompatibility and sustained ZOL release along with the degradation of hydrogel. Not only does this system provide a three-dimensional structural network for BMSC proliferation but also locally released ZOL promotes proliferation and osteogenic differentiation while inhibiting osteoclast differentiation of BMSCs in vitro. After implantation of ZOL/gTi into a distal femoral defect in the rabbits, micro-CT, histological, push-out, as well as genetic and protein analyses revealed significant promotion of osteointegration with the aid of this composite scaffold. Here, we provide an important theoretical basis for the research and development of bioactive implant interfaces for patients with osteogenic defects, such as osteoporosis. Background: Bionic surface nanopatterns of titanium (Ti) materials have excellent antibacterial effects in vitro for infection prevention. To date, there is a lack of knowledge about the in vivo bactericidal outcomes of the nanostructures on the Ti implant surfaces. Methods: A systematic review was performed using the PubMed, Embase, and Cochrane databases to better understand surface nanoscale patterns’ in vivo antibacterial efficacy. The inclusion criteria were preclinical studies (in vivo) reporting the antibacterial activity of nanopatterns on Ti implant surface. Ex vivo studies, studies not evaluating the antibacterial activity of nanopatterns or surfaces not modified with nanopatterns were excluded. Results: A total of five peer-reviewed articles met the inclusion criteria. The included studies suggest that the in vivo antibacterial efficacy of the nanopatterns on Ti implants’ surfaces seems poor. Conclusions: Given the small number of literature results, the variability in experimental designs, and the lack of reporting across studies, concluding the in vivo antibacterial effectiveness of nanopatterns on Ti substrates’ surfaces remains a big challenge. Surface coatings using metallic or antibiotic elements are still practical approaches for this purpose. High-quality preclinical data are still needed to investigate the in vivo antibacterial effects of the nanopatterns on the implant surface.

One of the goals of bone tissue engineering is to mimic native ECM in architecture and function, creating scaffolds with excellent biocompatibility, osteoinductive ability and mechanical properties. The aim of this study was to fabricate nanofibrous matrices by electrospinning a blend of poly (L-lactic-co-glycolic acid) (PLGA), hydroxyapatite (HA), and grapheme oxide (GO) as a favourable platform for bone tissue engineering. The morphology, biocompatibility, mechanical properties, and biological activity of all nanofibrous matrices were compared. The data indicate that the hydrophilicity and protein adsorption rate of the fabricated matrices were significantly increased by blending with a small amount of HA and GO. Furthermore, GO significantly boosted the tensile strength of the nanofibrous matrices, and the PLGA/GO/HA nanofibrous matrices can serve as mechanically stable scaffolds for cell growth. For further test in vitro, MC3T3-E1 cells were cultured on the PLGA/HA/GO nanofbrous matrices to observe various cellular activities and cell mineralization. The results indicated that the PLGA/GO/HA nanofibrous matrices significantly enhanced adhesion, and proliferation in MCET3-E1 cells and functionally promoted alkaline phosphatase (ALP) activity, the osteogenesis-related gene expression and mineral deposition. Therefore, the PLGA/HA/GO composite nanofibres are excellent and versatile scaffolds for applications in bone tissue regeneration.
