For ensuring the effectiveness of sterile insect technique ( SIT) programmes, maintaining the reproductive competitiveness and dispersal ability of mass-reared sterile males is essential. Inadvertent selection is an important genetic process that frequently occurs during mass rearing to produce sterile males. We investigated the effect of mass-rearing conditions on the responsiveness to sex pheromones and spontaneous flight activity of males of the sweetpotato weevil Cylas formicarius ( Coleoptera: Brentidae). There were no significant differences in the responsiveness to sex pheromones and spontaneous flight activity between wild and mass-reared strains. These results indicate that mass-reared strains of C. formicarius might not cause serious problems for implementing SIT programmes. [ABSTRACT FROM AUTHOR]

The sweetpotato weevil Cylas formicarius ( Fabricius) ( Coleoptera: Brentidae) and West Indian sweetpotato weevil Euscepes postfasciatus ( Fairmaire) ( Coleoptera: Curculionidae) are major pests of sweet potato Ipomoea batatas ( L.) Lam., in tropical and subtropical regions. Effects of intra- and inter-specific competition on the number of progeny (fecundity), body weight and developmental time of C. formicarius and E. postfasciatus were examined in single- and mixed-species treatments under laboratory conditions. Cylas formicarius tended to outcompete E. postfasciatus, whereas E. postfasciatus rather than C. formicarius tolerated higher conspecific densities. Background: Staging laparoscopy(SL)for patients with pancreatic cancer is used to identify occult abdominal metastasis (OAM). The aim of this study is to analyze the efficacy and safety of SL., Methods: Patients with pancreatic cancer who underwent SL before initial treatment at our hospital were included in this study. The short-term outcomes of the patients and the frequency of OAM detection were retrospectively analyzed., Results: Ten patients with pancreatic cancer were enrolled. The tumors were located in the head of the pancreas in 5 cases, and in the body and/or tail in 5 cases. The tumor diameter was 3 cm or more in 2 cases, and the serum CA19-9 level was 150 U/mL or more in 7 cases. No postoperative complications were observed in any of the patients. SL showed OAM in 2 patients., Conclusions: SL can be performed safely and may be useful in detecting OAM and optimizing treatment strategy for pancreatic cancer patients.

Quantity discrimination, is thought to be highly adaptive as it allows an organism to select greater amounts of food or larger social groups. In contrast to mammals, the processes underlying this ability are not as well understood in reptiles. This study examined the effects of ratio and number size on relative quantity discrimination in African spurred tortoises (Centrochelys sulcata). To assess these effects, tortoises were presented with trays containing favored food pieces in all possible number combinations between 1 and 7. The tortoises had to approach the tray they perceived as having the larger quantity. If correct, they received one piece of food as reinforcement. The results revealed that relative quantity discrimination was influenced by the ratio between the numbers of pieces, with performance improving as the ratio between the numbers increased. This finding suggests that the approximate number system or analogue magnitude estimation may control their behavior. However, as the number size increased, their performance declined, also suggesting that the approximate number system alone could not explain the present results., (© 2023. The Author(s).)

This paper presents the first case of the successful eradication of a Coleoptera pest species over a wide area using a combination of male annihilation technique (MAT) and sterile insect technique (SIT) application. The sweet potato weevil, Cylas formicarius, is one of the most destructive and widely distributed pests of sweet potato, Ipomoea batatas. A project to eradicate it was launched in 1994 on Kume Island, Okinawa Prefecture, Japan. The MAT application was first used from November 1994 to January 1999 to reduce the density of wild populations. The distribution and densities of weevils were assessed by trapping them and surveying infestation rates in wild hosts and sweet potatoes in the field. The C. formicarius populations were suppressed by approximately 90% and plant infestations were reduced from 9.5% to less than 0.1% by using the MAT. Then, hundreds of thousands to millions of sterile weevils were released each week (ca. 460 million in total from 1999 to 2012). As a result, based on an analysis of 12748 stems and 48749 tubers, no weevil infections were detected in the stems or tubers of sweet potato since 1997. Since 2009, almost no wild weevils were captured in traps, and in wild host and sweet potato surveys no weevils have been found in any of the 580 locations and 8833 samples since October 2011. As of 28 December, 2012, C. formicarius is considered to have been eradicated from Kume Island. Somatic cells can be reprogrammed into induced pluripotent stem (iPS) cells by forced expression of the transcription factors Oct3/4, Klf4, Sox2, and c-Myc (OKSM). Somatic cell nuclear transfer can also be utilized to reprogram somatic cells into totipotent embryos, suggesting that factors present in oocytes potentially enhance the efficiency of iPS cell generation. Here, we showed that preferentially expressed antigen of melanoma family member 12 (Pramef12), which is highly expressed in oocytes, enhances the generation of iPS cells from mouse fibroblasts. Overexpression of Pramef12 during the early phase of OKSM-induced reprogramming enhanced the efficiency of iPS cell derivation. In addition, overexpression of Pramef12 also enhanced expression of naïve pluripotency-associated genes, Gtl2 located within the Dlk1-Dio3 imprinted region essential for full pluripotency, glycolysis-associated genes, and oxidative phosphorylation-associated genes, and it promoted mesenchymal-to-epithelial transition during iPS cell generation. Furthermore, Pramef12 greatly activated β-catenin during iPS cell generation. The epithelial cell sheet functions as a barrier to prevent invasion of pathogens. It is necessary to eliminate intercellular gaps not only at bicellular junctions, but also at tricellular contacts, where three cells meet, to maintain epithelial barrier function. To that end, tight junctions between adjacent cells must associate as closely as possible, particularly at tricellular contacts. Tricellulin is an integral component of tricellular tight junctions (tTJs), but the molecular mechanism of its contribution to the epithelial barrier function remains unclear. In this study, we revealed that tricellulin contributes to barrier formation by regulating actomyosin organization at tricellular junctions. Furthermore, we identified α-catenin, which is thought to function only at adherens junctions, as a novel binding partner of tricellulin. α-catenin bridges tricellulin attachment to the bicellular actin cables that are anchored end-on at tricellular junctions. Thus, tricellulin mobilizes actomyosin contractility to close the lateral gap between the TJ strands of the three proximate cells that converge on tricellular junctions., (© 2022 Cho et al.)

A series of 3-oxygenated α-ionone analogs have been developed as highly specific male lures for the solanaceous fruit fly Bactrocera latifrons, a pest of solanaceous fruits. We compared the attractant and phagostimulant activities of analogs with or without (i) unsaturations at the 4,5- and/or 7,8-positions and (ii) oxygen moieties at the 3- and/or 9-positions of the ionone molecule. Since naturally occurring vomifoliol (V2) was found to induce a highly potent phagostimulant activity in B. latifrons males, related analogs including dehydrovomifoliol (V1), 6-hydroxy-α-ionone (U1), and 6-hydroxy-α-ionol (U2) were synthesized to evaluate their attractant and phagostimulant activities. Synthetic V1, V2, U1, and U2 exhibited low attractant activity, but their phagostimulant activity was relatively high. Optical isomers of 3-oxo-7,8-dihydro-α-ionone (P3) and V1 were prepared to examine the stereochemical specificity of attractants. (+)-(6R)-P3 and (+)-(6S)-V1 exhibited the corresponding activities, while their respective antipodal enantiomers were found entirely inactive., (© The Author(s) 2021. Background: Holding endoscopes by hand when performing eye surgery reduces the dexterity of the surgeon., Methods: A robotic endoscope holder called "Eye Explorer" is proposed to hold the endoscope and free the surgeon's hand., Results: This device satisfies the engineering and clinical requirements of eye surgery. The force for manual operation is less than 0.5 N. The observable ranges inside the patient's eye considering horizontal and vertical perspectives are 118° and 97°, and the motion of the holder does not interfere with the surgeon's hand and other surgical devices. The self-weight compensation can prevent the endoscope from falling when extra supporting force is released. When comparing the external force exerted on the eye by the Eye Explorer with that in case of manual operation, a decrease of more than 15% can be observed. Moreover, the consumption time of endoscope view adjustment using the Eye Explorer and manual operation does not significantly differ., Conclusion: The Eye Explorer allows dual-hand operation, facilitating a successful endoscopic eye surgery., (© 2020 The Authors. Background: Sensing the grasping force and displaying the force for the operator are important for safe operation in robot-assisted surgery. Although robotic forceps that senses the force by force sensors or driving torque of electric motors is proposed, the force sensors and the motors have some problems such as increase in weight and difficulty of the sterilization., Method: We developed a pneumatically driven robotic forceps that estimates the grasping torque and display the magnified torque for the operator. The robotic forceps has a master device and a slave robot, and they are integrated. In the slave side, the grasping torque is estimated by the pressure change in the pneumatic cylinder. A pneumatic bellows display the torque through a linkage., Results: We confirmed that the slave robot follows the motion of the master, and the grasping torque is estimated in the accuracy of 7 mNm and is magnified and displayed for the operator., Conclusions: The pneumatically driven robotic forceps has the capability in the estimation of the grasping torque and display of the torque. This work investigated the removal of selenite and selenate from water by green rust (GR) sulfate. Selenite was immobilized by simple adsorption onto GR at pH 8, and by adsorption-reduction at pH 9. Selenate was immobilized by adsorption-reduction to selenite and zero valent selenium (Se 0 ) at both pH 8 and 9. In the process, GR oxidized to a mixture of goethite (FeOOH) and magnetite (Fe 3 O 4 ). The kinetics of selenite and selenate sorption at the GR-water interface was described through a pseudo-second-order model. X-ray absorption spectroscopy data enabled to elucidate the concentration profiles of Se and Fe species in the solid phase and allowed to distinguish two removal mechanisms, namely adsorption and reduction. Selenite and selenate were reduced by GR through homogeneous solid-phase reaction upon adsorption and by heterogeneous reaction at the solid-liquid interface. The selenite reduced through heterogeneous reduction with GR was adsorbed onto GR but not reduced further. The redox reaction between GR and selenite/selenate was kinetically described through an irreversible second-order bimolecular reaction model based on XAFS concentration profiles. A nanopipette containing a solution of bis(benzo-15-crown-5) dissolved in 1,6-dichlorohexane was used as an ion-selective electrode (ISE) to probe K(+) for shear force-based constant-distance scanning electrochemical microscopy (SECM). In a previous study, the ISE responded only at low K(+) concentrations ([K(+)] < 1 mM), due to the depletion of the bis(benzo-15-crown-5) at the oil/water interface at high K(+) concentrations and the unstable response of the tip at the oil/water interface for shear force and current detection. In the present study, a nanopipette reshaped by heating and with the hydrophobic layer removed was used as the ISE. This modified ISE enabled a rapid response to changes in K(+) flux at a physiological concentration of K(+) and allowed SECM imaging on a nanometer scale. The fabricated nano-ISE was used as a probe for shear force-based SECM. Topography and K(+) flux images were obtained simultaneously at a polycarbonate membrane filter with 5 μm pores and human embryonic kidney 293 cells (HEK293). Several areas containing a K(+) flux larger than the surrounding areas were found in the SECM images of the HEK293 cells, which indicated the existence of K(+) channels.

The removal mechanisms of silicate using an aluminum hydroxide co-precipitation process was investigated and compared with an adsorption process, in order to establish an effective and validated method for silicate removal from wastewater. Adsorption isotherms, XRD and FT-IR analyses showed that silicate uptake occurred by adsorption to boehmite for initial Si/Al molar ratios smaller than two, but by precipitation of poorly crystalline kaolinite for the ratios larger than two, in both co-precipitation and adsorption processes. Silicate was removed by two steps: (i) an initial rapid uptake in a few seconds; and (ii) a slow uptake over several hours in both processes. The uptake rate in the first step was higher in the co-precipitation process than in adsorption process, presumably due to increased silicate adsorption to boehmite and rapid precipitation of kaolinite. These results suggest that silicate removal using aluminum salts could be effectively achieved if the pH adjustment and aluminum concentration are strictly controlled.

Over the past decade, rapid development of imaging technologies has resulted in the introduction of improved imaging devices, such as multi-modality scanners that produce combined positron emission tomography-computed tomography (PET-CT) images. The adoption of picture archiving and communication systems (PACS) in hospitals have dramatically improved the ability to digitally share medical image studies via portable storage, mobile devices and the Internet. This has in turn led to increased productivity, greater flexibility, and improved communication between hospital staff, referring physicians, and outpatients. However, many of these sharing and viewing capabilities are limited to proprietary vendor-specific applications. Furthermore, there are still interoperability and deployment issues which reduce the rate of adoption of such technologies, thus leaving many stakeholders, particularly outpatients and referring physicians, with access to only traditional still images with no ability to view or interpret the data in full. In this paper, we present a distribution architecture for medical image display across numerous devices and media, which uses a preprocessor and an in-built networking framework to improve compatibility and promote greater accessibility of medical data. Our INVOLVE2 system consists of three main software modules: 1) a preprocessor, which collates and converts imaging studies into a compressed and distributable format; 2) a PACS-compatible workflow for self-managing distribution of medical data, e.g. via CD USB, network etc; 3) support for potential mobile and web-based data access. The focus of this study was on cultivating patient-centric care, by allowing outpatient users to comfortably access and interpret their own data. As such, the image viewing software included on our cross-platform CDs was designed with a simple and intuitive user-interface (UI) for use by outpatients and referring physicians. Furthermore, digital image access via mobile devices or web-based access enables users to engage with their data in a convenient and user-friendly way. We evaluated the INVOLVE2 system using a pilot deployment in a hospital environment.

The sterile insect technique (SIT) is widely used for suppressing or eradicating target pest insect populations. In the eradication programs using the SIT, a large number of sterile insects are marked and released in the field. In Japan, Cylas fonnrmicarius (F.) group (Coleoptera: Brentidae) weevils are marked with a fluorescent powder dye to monitor the progress of such programs. However, this monitoring technique is not fully effective because of the disappearance or contamination of the dye. Therefore, an alternative marking method is required. Currently, a rare color morph such as piceous elytra (PE) is used for visible marking of C. formicarius group weevils. A PE-monomorphic strain has previously been established by artificial selection from a small locally distributed population; this can lead to reduced survival and genetic changes in behavioral traits due to inbreeding depression. In this study, we evaluated the survival rate and mating behavior of PE males of C. formicarius group. The characteristics of the PE males were similar to those of the wild strain (WS) males. Thus, we considered that PE males were suitable for visible marking in the eradication programs using the SIT.

There exist many viewers for single-modal medical images that are efficient and are equipped with powerful analysis tools. However, there is a distinct lack of efficient image viewers for multi-modality images, particularly for displaying multiple follow-up studies that depict a patient's response to treatment over time. Such viewers would be required to display large amounts of image data. In this study, we present the TAGIGEN viewer--a web-based image viewer designed specifically for the visualisation of multi-modality follow-up studies. We innovate by defining a series of dynamically generated image grid layouts that display sets of related images together in order to improve the ability to compare and assimilate the myriad images. We adopted a web-based client-server image streaming technology, thus enabling interactive navigation of the images in a computationally efficient manner. Furthermore, our web-based approach is interoperable and requires no software installation. We evaluated the ability of our viewer in displaying and understanding a patient's follow-up images in a case study with combined positron emission tomography and computed tomography (PET-CT) follow-up scans. We conducted a usability survey on 10 participants to measure the usefulness of our viewer, used as an outpatient viewer e.g. viewer designed for use by the patients, in tracking a patient's disease state across four PET-CT studies. Our initial results suggest that our viewer was able to efficiently visualise the patient data over time, and that the web-based implementation was fast (loading on average within 5.6 seconds with real-time navigation) and easy to use (overall survey score higher than 4 / 5).

The number of West Indian sweet potato weevils, Euscepes postfasciatus, being mass-reared in a facility for use in sterile insect technique (SIT) eradication programs has undergone a drastic reduction. A neogregarine protozoan pathogen Farinocystis sp. (an undescribed species) was detected in vivo in the mass-reared E. postfasciatus. We investigated the effects of this disease on the longevity and fecundity of host weevils and the incubation time of the disease in the host body under mass-rearing conditions. Our results demonstrated that infection by this Farinocystis sp. decreased both longevity and fecundity in E. postfasciatus. In particular, the pathogen severely limited the production of progeny by infected females compared to healthy females. Therefore, we consider this protozoan infection to be the major cause of the decreased E. postfasciatus production in the mass-rearing facility., (Copyright © 2010 Elsevier Inc. Weevils constitute the most species-rich animal group with over 60,000 described species, many of which possess specialized symbiotic organs and harbor bacterial endosymbionts. Among the diverse microbial associates of weevils, Nardonella spp. represent the most ancient and widespread endosymbiont lineage, having co-speciated with the host weevils for over 125 million years. Thus far, however, no empirical work on the role of Nardonella for weevil biology has been reported. Here we investigated the biological role of the Nardonella endosymbiont for the West Indian sweet potato weevil, Euscepes postfasciatus. This insect is an experimentally tractable pest insect that can easily be reared on a natural diet of sweet potato root as well as on an agar-based artificial diet. By larval feeding on an antibiotic-containing artificial diet, Nardonella infection was effectively eliminated from the treated insects. The antibiotic-treated insects exhibited significantly lighter body weight and lower growth rate than the control insects. Then, the antibiotic-treated insects and the control insects were respectively allowed to mate and oviposit on fresh sweet potatoes without the antibiotic. The offspring of the antibiotic-treated insects, which were all Nardonella-negative, exhibited significantly lighter body weight, smaller body size, lower growth rate and paler body color in comparison with the offspring of the control insects, which were all Nardonella-positive. In conclusion, the Nardonella endosymbiont is involved in normal growth and development of the host weevil. The biological role of the endosymbiont probably underlies the long-lasting host-symbiont co-speciation in the evolutionary course of weevils.

Irradiation has been widely used in suppression or eradication programs that use the sterile insect technique (SIT) or in studying sperm competition. Although it is well known that irradiation has negative impacts on reproductive (sperm) cells, previous studies have assumed that sperm from irradiated males behave identically to normal sperm in the female reproductive tract after mating. In this study, we used the West Indian sweetpotato weevil Euscepes postfasciatus to investigate the effect of irradiation on the abundance and viability of sperm in female spermatheca for 14 days after copulation. The abundance of sperm in females did not change throughout the experimental period, and sperm viability gradually decreased regardless of irradiation. In this weevil, irradiated sperm appear to behave identically to normal sperm in females for 14 days following irradiation/copulation. Therefore, the effect of irradiation on sperm viability within the female spermatheca is considered to be insignificant.

Purpose: This study was designed to evaluate the effect of anterior stromal puncture to treat patients with symptomatic bullous keratopathy., Methods: Fourteen patients with bullous keratopathy who had decreased vision and were or were not awaiting penetrating keratoplasty were evaluated before and 1, 4, 12, and 24 weeks after anterior stromal punctures with a 25-gauge needle. The patients were questioned about pain intensity, foreign-body sensation, and photophobia. Biomicroscopy, esthesiometry, and pachymetry results were evaluated. The results before and after the punctures were compared using the McNemar test, Friedman test, and multiple comparison tests., Results: The comparisons among before and 1, 4, and 12 weeks after the anterior stromal puncture of pain (p = 0.00000556), photophobia (p = 0.0252), foreign-body sensation (p = 0.0000743), and esthesiometry (p = 0.0243) showed significant statistical differences in terms of decreasing symptoms and corneal sensitivity. The average pachymetry did not show statistical difference among before and 1, 4, 12, and 24 weeks after the procedure (p = 0.956). We did not find important differences in terms of corneal neovascularization., Conclusion: Anterior stromal puncture is an effective, simple, and low-cost alternative for treating patients with symptomatic bullous keratopathy.
