Abstract

Abstract: Erythroascorbic acid (eAsA) is a five-carbon analog of ascorbic acid, and it is synthesized from d-arabinose by d-arabinose dehydrogenase (ARA) and d-arabinono-γ-lactone oxidase. We found an NAD+-specific ARA activity which is operative under submillimolar level of d-arabinose in the extracts of Saccharomyces cerevisiae. The hypothetical protein encoded by YMR041c showed a significant homology to a l-galactose dehydrogenase which plays in plant ascorbic acid biosynthesis, and we named it as Ara2p. Recombinant Ara2p showed NAD+-specific ARA activity with K m =0.78mM to d-arabinose, which is 200-fold lower than that for the conventional NADP+-specific ARA, Ara1p. Gene disruptant of ARA2 lost entire NAD+-specific ARA activity and the conspicuous increase in intracellular eAsA by exogenous d-arabinose feeding, while the double knockout mutant of ARA1 and ARA2 still retained measurable amount of eAsA. It demonstrates that Ara2p, not Ara1p, mainly contributes to the production of eAsA from d-arabinose in S. cerevisiae. [Copyright &y& Elsevier]

Abstract

The authors conducted a survey on the conception and attitude to leprosy using questionnaire in September of 1982. The social groups examined were university students and one of their parents, and the persons staying in a home for the aged. The source of the information about leprosy was the massmedia for 40.2% of the persons. Many persons had obtained knowledge on the severe and miserable social situation of the patients before and during the World War II. Eighty-six percent of the persons had had no contacts with such patients and they seemed to build up the body picture of the patient only with the visualization of the image and an emotional shade. Half of those surveyed thought that the disease was infectious. Concerning the social contact in society, the aged persons seemed to show flexibility in the opinion while those in the middle-aged group showed a negative attitude. Though the present results can not be generalized to the whole society, they may partially reflect the present general attitude toward leprosy.

Abstract

YBa2Cu3O7−δpowders were prepared by the thermal decomposition of a heteronuclear complex, CuY1/3Ba2/3(dhbaen)(NO3)1/3(H2O)3. The products of the complex thermal decomposition were analyzed by TG-DTA, XRD, SEM-Auger and XPS. The decomposition of the CuY1/3Ba2/3-complex was obtained at about 500°C and the product was a mixture of oxides and carbonates. The formation of YBa2Cu3O7−δproceeded at 800°C, with a gradual decomposition of the carbonates. A homogeneous distribution of each element, Y, Ba, and Cu, was observed for the decomposed CuY1/3Ba2/3-complex by SEM-Auger analysis. The binding energy values of Ba3d5/2and O1s photolines from Ba and O in the superconductive lattice did not shift during the sputtering period. Furthermore, the formation of Ba rich regions on the surface was depressed by using the complex as a starting material for homogeneous 123-oxide, YBa2Cu3O7−δ.

24. Fine structure of rat liver, adrenal, testis and seminal vesicle in experimental emaciation

Abstract

Experimentally emaciated male rats were produced by a bilateral electrical destruction of a part of hypothalamus. In a typical case, when the animals were fixed by perfusion, dissected, and organs weighed, the body weight became 1/2 of the control in 10 weeks. The weight of the viscera (including the subserous fat) was more decreased in comparison with the controls than the weight of the body wall (including extremities and the subcutaneous fat). The weight of the liver became 1/3, the adrenal 1/4, the testis 1/6 and the seminal vesicle 1/19 of the control. Light and electron microscopic examinations showed atrophy and fatty degeneration in the liver, atrophy of the zona reticularis in the adrenal, failure of spermatogenesis, especially at its spermiogenetic stage, in the testis, and an apoptosis in glandular epithelial cells of the seminal vesicle. Two weeks after partial hypothalamus destruction, the weight of the body wall was more decreased in comparison with the controls than the weight of the viscera. Possible pathophysiological mechanisms are discussed. An experimental model of electron microscopical research of apoptosis are presented.
