Objectives: We evaluated the noise reduction effects of deep learning reconstruction (DLR) and hybrid iterative reconstruction (HIR) in brain computed tomography (CT). Methods: CT images of a 16 cm dosimetry phantom, a head phantom, and the brains of 11 patients were reconstructed using filtered backprojection (FBP) and various levels of DLR and HIR. The slice thickness was 5, 2.5, 1.25, and 0.625 mm. Phantom imaging was also conducted at various tube currents. The noise reduction ratio was calculated using FBP as the reference. For patient imaging, overall image quality was visually compared between DLR and HIR images that exhibited similar noise reduction ratios. Results: The noise reduction ratio increased with increasing levels of DLR and HIR in phantom and patient imaging. For DLR, noise reduction was more pronounced with decreasing slice thickness, while such thickness dependence was less evident for HIR. Although the noise reduction effects of DLR were similar between the head phantom and patients, they differed for the dosimetry phantom. Variations between imaging objects were small for HIR. The noise reduction ratio was low at low tube currents for the dosimetry phantom using DLR; otherwise, the influence of the tube current was small. In terms of visual image quality, DLR outperformed HIR in 1.25 mm thick images but not in thicker images. Conclusions: The degree of noise reduction using DLR depends on the slice thickness, tube current, and imaging object in addition to the level of DLR, which should be considered in the clinical use of DLR. The effective dose (ED) in computed tomography (CT) may be calculated by multiplying the dose–length product (DLP) by a conversion factor. As children grow, automatic exposure control increases the DLP, while the conversion factor decreases; these two changes affect the ED in opposite ways. The aim of this study was to investigate the methods of ED estimation according to age in pediatric brain CT. We retrospectively analyzed 980 brain CT scans performed for various clinical indications in children. The conversion factor at each age, in integer years, was determined based on the values at 0, 1, 5, and 10 years provided by the International Commission on Radiological Protection (ICRP), using a curve (curve method) or lines (linear method). In the simple method, the ED was estimated using the ICRP conversion factor for the closest age. We also analyzed the ED estimated by a radiation dose management system. Although the median DLP at each age increased with age, the median ED estimated by the curve method was highest at 0 years, decreased with age, and then plateaued at 9 years. The linear method yielded mildly different results, especially at 2 and 3 years. The ED estimated by the simple method or the radiation dose management system showed inconsistent, up-and-down changes with age. In conclusion, the ED in pediatric brain CT decreases with age despite increased DLP. Determination of the conversion factor at each age using a curve is expected to contribute to estimating the ED in pediatric CT according to age. [ABSTRACT FROM AUTHOR]

Chinese hamster ovary (CHO) cells are major host cells for biopharmaceuticals. During culture, the chromosome number of CHO cells alters spontaneously. Here, we investigated the effects of artificial changes in the chromosome number on productivity. When cell fusion between antibody‐producing CHO‐K1‐derived cells was induced, we observed a wide range of aneuploidy that was not detected in controls. In particular, antibody productivities were high in clone‐derived cell populations that retained a diverse chromosome number distribution. We also induced aneuploid cells using 3‐aminobenzamide that causes chromosome non‐disjunction. After induction of aneuploidy by 3‐aminobenzamide, cells with an increased chromosome number were isolated, but cells with a decreased chromosome number could not be isolated. When antibody expression vectors were introduced into these isolated clones, productivity tended to increase in cells with an increased chromosome number. Further analysis was carried out by focusing on clone 5E8 with an average chromosome number of 37. When 5E8 cells were used as host, the productivity of multiple antibodies, including difficult‐to‐express antibodies, was improved compared with CHO‐K1 cells. The copies of exogenous genes integrated into the genome were significantly increased in 5E8 cells. These findings expand the possibilities for host cell selection and contribute to the efficient construction of cell lines for recombinant protein production. [ABSTRACT FROM AUTHOR]

In computed tomography (CT), organ dose modulation (ODM) reduces radiation exposure from the anterior side to reduce radiation dose received by the radiosensitive organs located anteriorly. We investigated the effects of ODM applied to a part of the scan range on radiation dose in body CT. The thorax and thoraco-abdominopelvic region of an anthropomorphic whole-body phantom were imaged with and without ODM. ODM was applied to various regions, and the tube current modulation curves were compared. Additionally, the dose indices were compared with and without ODM in thoracic and thoraco-abdominopelvic CTs in 800 patients. ODM was applied to the thyroid in male patients and to the thyroid and breast in female patients. In phantom imaging of the thorax, the application of ODM below the scan range decreased the tube current, and that to the breast showed a further decrease. Decreased tube current was also observed in phantom imaging of the thoraco-abdominopelvic regions with ODM below the scan range, and the application of ODM to the whole scan range, thyroid, breast, and both thyroid and breast further reduced the tube current in the region to which ODM was applied. In patient imaging, the dose indices were significantly lower with ODM than without ODM, regardless of the scan range or sex. The absolute reduction in dose-length product was larger for thoraco-abdominopelvic CT (male, 43.2 mGy cm; female, 59.7 mGy cm) than for thoracic CT (male, 30.8 mGy cm; female, 37.6 mGy cm) in both sexes, indicating dose reduction in the abdominopelvic region to which ODM was not applied. In conclusion, The application of ODM in body CT reduces radiation dose not only in the region to which ODM is applied but also outside the region. Dose-length product (DLP) is widely used as an indicator of the radiation dose in computed tomography. The aim of this study was to investigate the significance of sex and body weight in DLP-based monitoring of the radiation dose. Eight hundred computed tomographies of the chest performed using four different scanners were analysed. The DLP was compared with body weight by linear regression in men and women separately. The DLP was positively correlated with body weight, and dependence on sex and weight differed among scanners. Standard DLP values adjusted for sex and weight facilitated interscanner comparison of the radiation dose and its dependence on sex and weight. Adjusting the DLP for sex and weight allowed one to identify examinations with possibly excessive doses independently of weight. Monitoring the DLP in relation to sex and body weight appears to aid detailed comparison of the radiation dose among imaging protocols and scanners and daily observations to find unexpected variance., (© The Author 2015. Unlabelled: We evaluated the effective dose (ED) of the CT component of whole-body PET/CT using software dedicated to CT dose estimation and from dose-length product (DLP) values to establish practical methods of ED estimation., Methods: Eighty adult patients who underwent (18)F-FDG whole-body PET/CT were divided into groups A and B, each consisting of 20 men and 20 women. In group A, ED of the CT component was calculated using CT-Expo for 6 anatomic regions separately, and whole-body ED was obtained by summing the regional EDs (CT-Expo method). DLP was calculated for each of the 6 regions and multiplied by a corresponding conversion factor described in International Commission on Radiological Protection publication 102 to obtain the ED for each region (regional DLP method). Whole-body ED was also calculated as the product of a whole-body DLP value provided by the scanner automatically and a conversion factor (simple DLP method). Moreover, the ED/DLP values were calculated using whole-body ED estimated by the CT-Expo method and the scanner-derived DLP, to optimize the conversion factor. In group B, the optimized conversion factor was applied for the estimation of ED by the simple DLP method., Results: In group A, the regional DLP method allowed an accurate estimation of mean whole-body ED as a result of counterbalance of mild overestimation in men and mild underestimation in women, regarding the CT-Expo method as a standard. The simple DLP method using a conversion factor for the trunk (0.015 mSv/mGy/cm) caused overestimation. On the basis of the ED/DLP values in group A, a modified conversion factor of 0.013 mSv/mGy/cm and sex-specific conversion factors of 0.012 and 0.014 mSv/mGy/cm for men and women, respectively, were determined. In group B, the use of the modified conversion factor improved accuracy, and the use of sex-specific conversion factors eliminated sex-dependent residual errors., Conclusion: ED of the CT component of whole-body PET/CT can be assessed by multiplying the scanner-derived DLP by a conversion factor optimized for whole-body PET/CT., (© 2015 by the Society of Nuclear Medicine and Molecular Imaging, Inc.)
