Abstract

Biodiversity loss and climate change have severely impacted ecosystems and livelihoods worldwide, compromising access to food and water, increasing disaster risk, and affecting human health globally. Nature-based Solutions (NbS) have gained interest in addressing these global societal challenges. Although much effort has been directed to NbS in urban and terrestrial environments, the implementation of NbS in marine and coastal environments (blue NbS) lags. The lack of a framework to guide decision-makers and practitioners through the initial planning stages appears to be one of the main obstacles to the slow implementation of blue NbS. To address this, we propose an integrated conceptual framework, built from expert knowledge, to inform the selection of the most appropriate blue NbS based on desired intervention objectives and social-ecological context. Our conceptual framework follows a four incremental steps structure: Step 1 aims to identify the societal challenge(s) to address; Step 2 highlights ecosystem services and the underlying biodiversity and ecological functions that could contribute to confronting the societal challenge(s); Step 3 identify the specific environmental context the intervention needs to be set within (e.g. the spatial scale the intervention will operate within, the ecosystem's vulnerability to stressors, and its ecological condition); and Step 4 provides a selection of potential blue NbS interventions that would help address the targeted societal challenge(s) considering the context defined through Step 3. 17. Phylogeny and bioprospecting: The diversity of medicinal plants used in cancer management.

Abstract

Societal Impact Statement Summary As the second‐leading cause of mortality worldwide, cancer is a major focus of drug discovery research. Traditional plant knowledge can guide the search for undiscovered compounds, but the efficacy of this approach for cancer, a highly complex disease affecting diverse tissues, is unknown. We investigated the patterns underlying plant selection for cancer treatment globally, finding certain lineages are repeatedly targeted. While this indicates therapeutic value, their relatedness with plants used for unrelated ethnobotanical uses suggests that plants are probably selected to treat cancer‐associated symptoms, rather than addressing tumour growth. Careful re‐examination and scoring of ethnobotanical reports may make the prediction of lineages for drug discovery more informative. Cancer is a highly diverse disease and as the second‐leading cause of death worldwide is a focus of drug discovery research. Natural products have been shown to be a useful source of novel molecules for treating cancer. It is likely there are many plants with undiscovered molecules of therapeutic value, however identifying new leads from the vast diversity of plants is very challenging. Traditional knowledge might inform bioprospecting by predicting lineages of plants rich in therapeutically useful molecules. We characterise the phylogenetic diversity of plants used in traditional cancer management using a comprehensive genus‐level phylogeny of angiosperms, and a list of 597 genera used globally to treat different cancers. We phylogenetically predict which lineages may have elevated potential for drug discovery and assess the quality of the prediction. We demonstrate the independent and repeated targeting of specific lineages of plants by different peoples in different parts of the world. However, the lineages we report here as rich in plants used in traditional cancer management coincide with those for other ethnobotanical applications and contain few plants with proven anti‐cancer activity. That the same lineages are used to treat different cancers is suggestive of independent discovery of therapeutic value. However, it is likely that the traditional knowledge explored here is shaped by the selection of plants conferring milder effects for treating wider symptoms, such as tiredness or nausea, rather than for halting tumour growth. Accurate prediction of useful plant lineages for cancer management requires more nuanced information than is commonly provided in ethnobotanical records. [ABSTRACT FROM AUTHOR]

29. Supermatrix analyses and molecular clock rooting of Fabales: Exploring the effects of outgroup choice and long branch attraction on topology

Abstract

Fabales is a cosmopolitan angiosperm order that consists of four families: Leguminosae (Fabaceae), Polygalaceae, Surianaceae, and Quillajaceae. Despite the great interest in this group, a convincing phylogeny of the order is still not available. Therefore, the aim of this study was to explicitly test for possible long branch attraction (LBA) problems within Fabales for the first time, and determine whether low tree stemminess and unequal branch lengths could worsen this problem. Supermatrix analysis of Fabales was carried out using previously. published plastid matK, trnL, rbcL, and newly sequenced nuclear sqd1 regions for 678 taxa in total, including 43 outgroup taxa from families of Fabidae. We employed additional analyses, such as simulations, network analyses, sampling different outgroup taxa (random or real), removing fast evolving sites and fast evolving taxa, and molecular clock rooting, to identify both LBA and (or) rooting problems. These analyses clearly show that the Fabales phylogeny has been influenced by the sampling of outgroup taxa, but not LBA. However, network analyses show that even though it is weak, there is a consistent phylogenetic signal among the rapidly radiated Fabales families, which can be traced by further analyses. While, molecular clock rooting analysis yielded a (Leguminosae(Polygalaceae(Surianaceae+Quillajaceae))) topology with strong support for the first time here, supermatrix analyses yielded a ((Leguminosae+Polygalaceae)(Surianaceae+Quillajaceae)) with low-moderate support. Les Fabales forment un ordre d'angiospermes cosmopolite constitué dequatre familles, les Leguminosae (Fabaceae), les Polygalaceae, les Surinaceae et les Quillajaceae. Malgré le grand intérêt suscité par ce groupe, une phylogénie convaincante n'est pas encore établie pour cet ordre. Par conséquent, l'objectif de cette étude consiste à vérifier explicitement et pour la première fois l'existence de problèmes éventuels d'attraction des longues branches (LBA, long branch attraction) au sein des Fabales et à déterminer si un faible facteur de << stemminess >> (c.-à-d. courtes branches internes et longues branches terminales) et des longueurs de branches inégales pourraient aggraver ce problème. L'analyse de la supermatrice des Fabales a été réalisée à l'aide de régions précédemment publiées des plastides matK, trnL, rbcL et la région récemment séquencée de sqd1 nucléaire de 678 taxons au total, dont 43 taxons hors groupe des familles des Fabidae. Les auteurs ont utilisé des analyses supplémentaires telles que les simulations, les analyses de réseau, l'échantillonnage de taxons hors groupe différents (aléatoires ou réel), l'élimination des sites à évolution rapide et des taxons à évolution rapide et l'enracinement de l'horloge moléculaire, afin de déterminer les problèmes d'attraction des longues branches ou d'enracinement. Ces analyses montrent clairement que la phylogénie des Fabales a été influencée par l'échantillonnage de taxons hors groupe, mais pas par l'attraction des longues branches. Toutefois, les analyses de réseau montrent que même s'il est faible, il existe un signal phylogénétique constant parmi les familles de Fabales à radiation rapide, dont on peut trouver la trace par des analyses plus approfondies. Alors que l'analyse d'enracinement de l'horloge moléculaire produisait une topologie (Leguminosae(Polygalaceae(Surinaceae+Quillajaceae))) fortement soutenue ici pour la première fois, les analyses de la supermatrice produisaient une topologie ((Leguminosae+Polygalaceae)(Surinaceae+ Quillajaceae)) avec un soutien faible/modéré.
