10. Characterization of Volatile Organic Compounds of Giant Salamander (Andrias davidianus) Oil Adulterated with Different Amounts of Peanut Oil by Gas Chromatography-Ion Mobility Spectrometry Combined with Chemometrics

Abstract

The volatile components in giant salamander oil adulterated with different amounts of peanut oil (0%, 5%, 10%, 20%, 30%, and 100%) were studied by gas chromatography-ion mobility spectroscopy (GC-IMS) combined with chemometrics. The results showed that a total of 41 volatile compounds were identified in all samples, including 21 aldehydes, 6 ketones, 4 alcohols, 4 heterocyclic compounds, 3 esters, 2 sulfur-containing compounds and 1 acid. With increasing adulteration level, the contents of aldehydes, heterocycles, acids and esters increased, while the contents of ketones, alcohols and sulfur compounds decreased. A partial least squares regression (PLSR) model between volatile components and adulteration level was established. The correlation coefficient (R2) values for the calibration and verification sets were 0.992 4 and 0.988 2, respectively, indicating that the reliability of the model. Principal component analysis (PCA) showed that the cumulative contribution rate of the first two principal components (PC) was 94.3%, indicating that the different adulteration levels could be well distinguished by volatile components. Thirteen differential volatile compounds with variable importance for the projection (VIP) scores greater than one, including seven aldehydes, three ketones, one alcohol, one sulfur compound and one ester, were selected by partial least squares-discriminant analysis (PLS-DA). PCA and cluster analysis showed that these differential volatile components could also be used to distinguish the different adulterated salamander oil samples. This study can provide technical support for the nondestructive rapid identification of adulterated giant salamander oil.

22. Research on Optimal Design of Spudcan Structures to Ease Spudcan-Footprint Interactions in Clay and Comparative Analyses with Different Measures

Abstract

In order to ease consequences of spudcan-footprint interactions during jack-up rigs reinstalling in the vicinity of an existing seabed footprint, three new types of spudcan shapes, that is, a lotus-shaped spudcan with six circular holes, a flat-bottomed spudcan, and a concave-shaped spudcan, were proposed to perform an optimizing study of the spudcan structures, and the effectiveness of them were analyzed comparatively with other different measures. Firstly, 3D Large Deformation Finite Element (LDFE) analyses were carried out using the Coupled Eulerian-Lagrangian (CEL) method in the commercial finite element package ABAQUS. After calibrating the validity of the numerical calculation model against existing centrifuge test data and LDFE results, the differences in interaction mechanism between the novel spudcans and the generic spindle-shaped spudcan were studied when penetrating near an existing footprint with an eccentric distance of 0.5D, and the horizontal range of plastic deformation of the disturbed soils, the inclination angle of the spudcan and the offset distance of the pile legs were analyzed comparatively as well. The results show that the proposed novel spudcans can mitigate the maximum horizontal sliding force and the maximum bending moment at the top of the pile leg obviously, compared with those of the generic one, which were reduced by 32.59%, 22.47%, 28.18%, and 26.32%, 12.88%, 18.02%, respectively. It also can be seen that all novel structures can ease the adverse consequences of spudcan-footprint interactions effectively, and can improve the in -place stability of the spudcan as well. Finally, three possible measures in mitigating interactions of the spudcan-footprint were contrasted, that is, the novel spudcan (represented by the lotus-shaped spudcan with six holes), stomping method, and perforation drilling method. The results show that all of them can reduce adverse impacts induced by interactions of the spudcan-footprint, and also can improve the in-place stability of the spudcan during reinstallation. In addition, among them, according to the effect of reducing the additional stress of the spudcan, the effectiveness of them can be listed as follows: perforation drilling near an existing footprint > the lotus-shaped spudcan with six holes > stomping method. In terms of the vertical bearing capacity of the spudcan, the lotus-shaped spudcan with six holes can improve it as much as 16.33% compared with the spindle-shaped structure due to the particularity of the structure. While reducing the continuity and strength of soil foundations, the perforation drilling measure leads to the decrease of the vertical bearing capacity of the spudcan by 13.07%. It can be concluded that all the three measures have merits and demerits, so the relevant construction environment conditions and engineering practice should be fully considered when selecting measures to deal with interactions of the spudcan-footprint.

24. Use of GC-IMS and Stoichiometry to Characterize Flavor Volatiles in Different Parts of Lueyang Black Chicken during Slaughtering and Cutting.

Abstract

Chilled and cut chicken is preferred by consumers for its safeness and readiness to cook. To evaluate the quality characteristics of various chilled chicken products, differences in volatile organic components (VOCs) of six different cut parts (breast, back, leg, heart, liver, and gizzard) of Lueyang black chicken were characterized through gas chromatography–ion mobility spectroscopy (GC-IMS) combined with stoichiometry. A total of 54 peaks in the signal of VOCs were detected by GC-IMS, and 43 VOCs were identified by qualitative analysis. There were 22 aldehydes (20.66–54.07%), 8 ketones (25.74–62.87%), 9 alcohols (4.17–14.69%), 1 ether (0.18–2.22%), 2 esters (0.43–1.54%), and 1 furan (0.13–0.52%), in which aldehydes, ketones, and alcohols were the main categories. Among the six cut parts, the relative content of aldehydes (54.07%) was the highest in the gizzard, and the relative content of ketones (62.87%) was the highest in the heart. Meanwhile, the relative content of alcohols (14.69%) was the highest in the liver. Based on a stable and reliable predictive model established by orthogonal partial least squares–discriminant analysis (OPLS-DA), 3-hydroxy-2-butanone (monomer and dimer), acetone, 2-butanone monomer, hexanal (monomer and dimer), isopentyl alcohol monomer, and n-hexanol monomer were picked out as characteristic VOCs based on variable importance in projection (VIP value > 1.0, p < 0.05). Principal component analysis (PCA) and the clustering heatmap indicated that the characteristic VOCs could effectively distinguish the six cut parts of Lueyang black chicken. The specific VOCs responsible for flavor differences among six different cut parts of Lueyang black chicken were hexanal (monomer and dimer) for the gizzard, 2-butanone monomer and hexanal dimer for the breast, hexanal monomer for the back, 3-hydroxy-2-butanone monomer for the leg, 3-hydroxy-2-butanone (monomer and dimer) for the heart, and acetone and isopentyl alcohol monomer for the liver. Abstract

By using the 3-point QCD sum rules, we calculate the transition form factors of $D$ decays into the spin triplet axial vector mesons $a_1(1260)$, $f_1(1285) $, $f_1(1420)$. In the calculations, we consider the quark contents of each meson in detail. In view of the fact that the isospin of $a_1(1260)$ is one, we calculate the $D^+ \rightarrow a_1^0 (1260)$ and $D^0 \rightarrow a_1^- (1260)$ transition form factors separately. In the case of $ f_1(1285), f_1(1420)$, the mixing between light flavor $SU(3)$ singlet and octet is taken into account. Based on the form factors obtained here, we give predictions for the branching ratios of relevant semileptonic decays, which can be tested in the future experiments., Comment: 14 pages, 13 figures

In this study, the sensory evaluation of Lueyang black chicken breast meat during steaming was first performed, and their volatile organic compounds (VOCs) were further analyzed by gas chromatography-ion mobility spectroscopy (GC-IMS) combined with stoichiometry. The sensory results demonstrated that the Lueyang black chicken breast meat steamed for 15–30 min was more acceptable in taste, flavor, and chewiness. A total of 60 volatile flavor signal peaks were obtained, and 46 VOCs were recognized from qualitative analysis, containing 24 aldehydes (51.19–72.57%), 8 ketones (10.15–16.97%), 9 alcohols (7.98-13.16%), 2 furans (2.24–10.85%), 2 esters (0.54–1.56%), and 1 ether (0.05–2.47%). A stable and reliable prediction model was set up by orthogonal partial least squares-discriminant analysis (OPLS-DA), and 18 characteristic VOCs (including 10 aldehydes, 3 alcohols, 3 ketones, 1 furan, and 1 ether) were picked out through variable importance in the projection (VIP>1.0 and p < 0.05). Principal component analysis (PCA) results indicated that the cumulative contribution ratio was 92% with PC1 68.7% and PC2 23.3%, respectively, indicating that these characteristic VOCs could well discriminate the steaming time of Lueyang black chicken breast meat. Heatmap clustering analysis also demonstrated a similar distinguishing effect. Abstract

In order to study the buckling failure of lining delamination of thin-walled lined composite pipe, a numerical analysis model was established by using bilinear cohesion relationship, the buckling mode and critical load of the composite pipe obtained by linear buckling are taken as reference values, the interlayer initial defects were introduced to carry out the nonlinear buckling analysis on the composite pipe lining structure with thin wall lining. The relative displacement curve and deformation morphology of the relative displacement of the lining with the change of external pressure were obtained, and the results are consistent with the existing test results. Based on this model, the buckling sensitivity of lining pipe was analyzed. The results show that the size of interlayer initial defect is the main factor that affects the critical buckling pressure of liner, but it has little influence on the propagation pressure after buckling; However, the increase of interlayer bonding effect significantly improves the buckling resistance and propagation pressure of lining pipe; The ratio of outer tube wall diameter to thickness and liner thickness have significant effects on the critical buckling pressure of liner. 44. The Mechanism of Plantaginis Semen in the Treatment of Diabetic Nephropathy based on Network Pharmacology and Molecular Docking Technology

Abstract

Background: Diabetic nephropathy (DN) is one of the common complications of diabetes. Plantaginis Semen (PS) has a variety of therapeutic effects, however its mechanism on DN is unclear.Objective: This paper aims to find the ingredients, the key targets, and the action pathways of PS on DN from the perspective of network pharmacology.Methods: The databases of network pharmacology, such as Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP), Pharmmapper, OMIM, DrugBank, Gene- Cards, TTD, Disgenet, STRING, and Cytoscape software, were used to find the main ingredients and targets. Gene Ontology (GO) function and Kyoto Encyclopedia of Genome and Genomes (KEGG) pathway enrichment analysis were used to reveal the potential pathways of the PS on DN. The GEO database was used to find the targets of DN based on valid experimental research. The molecular docking technology was used to evaluate the combination between ingredients of PS and the targets.Results: A total of 9 active ingredients and 216 potential therapeutic targets were obtained for PS on DN. Hub targets were discovered by the Cytoscape software analysis. CASP3 was screened by Venn diagram by making intersection between GSE30529 and hub genes. Moreover, CASP3 was combined with one of the nine active ingredients, quercetin, by molecular docking analysis. The KEGG pathways were mainly involved in diabetic nephropathy, and were simultaneously associated with CASP3 as followed: AGE-RAGE signaling pathway in diabetic complications, apoptosis, lipid and atherosclerosis, MAPK signaling pathway, TNF signaling pathway, IL-17 signaling pathway, and p53 signaling pathway.Conclusion: PS can have the treatment on DN through CASP3. Quercetin, as one of the nine active ingredients, can be bounded to CASP3 to inhibit apoptosis in DN. PS can also take action on DN probably through many pathways. The role of PS on DN through other pathways still needs to be further elaborated.

Abstract

Background and Objective: Diabetes is an important risk factor for periodontitis, and circular RNA (circRNA) may play an important role in aggravating inflammation and accelerating disease progression by regulating miRNA/mRNA. This study aimed to investigate the role and mechanism of the hsa_circ_0084054/miR‐508‐3p/PTEN axis in the progression of periodontitis with diabetes. Methods: First, circRNA sequencing was used to screen the differentially expressed circRNAs of periodontal ligament cells (PDLCs) treated with high glucose and/or Porphyromonas gingivalis lipopolysaccharide (LPS) in vitro, and the overtly differentially expressed hsa_circ_0084054 was selected and was also verified in periodontal ligament (PDL) tissue from periodontitis patients with diabetes. Then, its ring structure was tested by Sanger sequencing, RNase R, and actinomycin D assays. The bioinformatics analysis, dual luciferase reporter assay, and RIP assay were used to explore the interaction of hsa_circ_0084054/miR‐508‐3p/PTEN axis, whose effects on inflammation, oxidative stress, and apoptosis of PDLCs were evaluated through the measurement of inflammatory factors, reactive oxygen species (ROS), total superoxide dismutase (SOD), malondialdehyde (MDA), and Annexin V/PI assay. Results: By high‐throughput sequencing, it was found that hsa_circ_0084054 was significantly increased in HG + LPS group compared with control group and LPS group, which was also verified in periodontal ligament (PDL) tissue from periodontitis patients with diabetes. Silencing hsa_circ_0084054 in PDLCs decreased the expression of inflammatory factors (IL‐1β, IL‐6, TNF‐α), the levels of ROS and MDA, and the proportion of apoptotic cells; conversely, SOD activity was enhanced. In addition, we found that hsa_circ_0084054 could up‐regulate the expression of PTEN through sponge miR‐508‐3p to inhibit AKT phosphorylation, finally trigger the aggravation of oxidative stress and inflammation in periodontitis patients with diabetes. Conclusion: hsa_circ_0084054 can aggravate inflammation and promote the progression of periodontitis with diabetes by regulating miR‐508‐3p/PTEN signaling axis, which may serve as a new target for the intervention of periodontitis with diabetes. Abstract

Forgiveness, an emotion regulation strategy for overcoming personal distress and healing injuries following interpersonal conflicts, is greatly modulated by parental bonding (e.g. parental care and overprotection) and genetic predispositions. Inspired by the roles of dopamine β-hydroxylase (DBH) gene in empathy and emotion regulation, we examined how parental bonding interacted with the DBH-1021C/T (i.e. a polymorphism accounts for 35–52% of variance in DBH activity) in forgiveness. Results indicated that (1) the score on forgiveness was positively related to parental care, but negatively related to parental overprotection; (2) parental care significantly interacted with the DBH-1021C/T in forgiveness with differential susceptibility model. The TT/CT genotypes were related to lower forgiveness than the CC genotype among individuals with lower parental care, butwere related to higher forgiveness among individuals with optimal parental care. Abstract

Precise control of the morphology of nanocrystals is vital in various fields, especially for the conductive materials and catalysts. In this work, ethanolamine (ETA)-induced synthesis of sesame cake-like Cu/Ag core–shell nanosheets (Cu/AgNSs) has been reported for the first time. It is found that the ETA molecules may play a "clip-on" role in the formation of sesame cake-like Cu/AgNSs. In the system, the lone-pair electrons of oxygen and nitrogen in ETA can not only bind to the vacant orbital of Cu element, thus promoting ETA to preferentially adsorb on the surface of copper nanosheets (CuNSs), but also make Ag+ ions tend to form silver–amine complexes, eventually resulting in the gentle reduction of Ag+ ions on the surface of CuNSs and the final formation of Ag shell layer. Moreover, the prepared sesame cake-like Cu/AgNSs exhibited excellent catalytic activity (turnover frequency, TOF = 12.05 min–1) in the reduction of p-nitrophenol to p-aminophenol when compared with the most reported silver-based catalysts.
