13. Studying the Internal Wave Generation Mechanism in the Northern South China Sea Using Numerical Simulation, Synthetic Aperture Radar, and In Situ Measurements.

Abstract

The internal waves in the South China Sea are highly correlated with the tidal currents in the Luzon Strait, which makes it possible to establish an internal wave prediction model based on internal wave kinematics. However, the kinematic model requires the input of the exact location and time of the initial internal wave for which the generation mechanism of internal waves in the northern South China Sea must be well understood. By analyzing the internal wave field in the northern South China Sea (SCS) simulated using the MIT General Circulation Model (MITgcm) and observations from satellite synthetic aperture radar (SAR) and mooring temperature–salinity–depth (TSD) chains, the source regions and propagation initiation times of internal waves are identified for three typical tidal phases, i.e., the diurnal-tide-dominated phase (DTP), transition tide phase (TTP), and semidiurnal-tide-dominated phase (STP). The generation procedures of Type A and Type B internal waves are discussed in detail with those data. The present study reveals that Type A and Type B waves are generated at the eastern and western ridges, respectively, and both commence their westward propagation at the peak of the eastward tidal flow. The dynamics of lee waves and the resonance effect with double ridges constitute the generation mechanisms of internal waves in the northern SCS. Combined with varying configurations of tidal conditions, topography, and stratification, the generation procedures of Type A and Type B waves in the DTP, TTP, and STP are elucidated with the generation mechanism in a unified and self-consistent way. In short, during DTP, weaker A waves alternate with weaker B waves each day; during TTP, strong A waves and strong B waves appear alternately every day; and there are two weak A waves per day during the STP. The generation mechanism can help in developing future empirical models for generating internal waves using tidal currents, topography, and stratification without requiring complex fluid dynamics calculations. Abstract

Transition metal doping is an ideal strategy to construct multifunctional and efficient nanozymes for biosensing. In this work, a metal-doped CoMnOx nanozyme was designed and synthesized by hydrothermal reaction and high-temperature calcination. Based on its oxidase activity, an "on-off-on" smartphone sensing platform was established to detect ziram and Cu2+. The obtained flower-shaped CoMnOx could exhibit oxidase-, catalase-, and laccase-like activities. The oxidase activity mechanism of CoMnOx was deeply explored. O2 molecules adsorbed on the surface of CoMnOx were activated to produce a large amount of O2·-, and then, O2·- could extract acidic hydrogen from TMB to produce blue oxTMB. Meanwhile, TMB was oxidized directly to the blue product oxTMB via the high redox ability of Co species. According to the excellent oxidase-like activity of CoMnOx, a versatile colorimetric detection platform for ziram and Cu2+ was successfully constructed. The linear detection ranges for ziram and Cu2+ were 5~280 μM and 80~360 μM, and the detection limits were 1.475 μM and 3.906 μM, respectively. In addition, a portable smartphone platform for ziram and Cu2+ sensing was established for instant analysis, showing great application promise in the detection of real samples including environmental soil and water. 27. Construction of CuO/Fe2O3Nanozymes for Intelligent Detection of Glufosinate and Chlortetracycline Hydrochloride

Abstract

In this work, CuO/Fe2O3nanozymes with high peroxidase-like activity were synthesized by using hydrothermal and calcination methods. The high-resolution transmission electron microscopy (HRTEM) proved that the heterogeneous interface of CuO/Fe2O3was the main reason for the high enzyme-like activity. Strong interactions of CuO and Fe2O3were successfully verified by X-ray absorption near-edge structure (XANES) characterization. Experiments and density functional theory (DFT) calculations were also used to explain the increased enzyme activity. The heterogeneous interface acted as the main active center, facilitating the electron transfer from CuO to Fe2O3. A colorimetric and intelligent sensing system was constructed based on deep learning. Using the peroxidase-like activity of CuO/Fe2O3, a platform for glufosinate pesticides and chlortetracycline hydrochloride (CTC) with the signal “on–off–on” changes were established. The limit of detection (LOD) of glufosinate and CTC was 28 and 0.69 μM, respectively. It was successfully applied in the detection of environmental water and soil. This study can provide an intelligent detection method for environmental monitoring.

Abstract

Tomato is a major source of dietary carotenoids that play an important role in human health. Improving the carotenoid profiles of tomato fruit has become a primary aspect in tomato breeding. The tomato ‘Huang Song’ landrace grows vigorously but with low contents of carotenoids in this yellowish fruit. To overcome this problem and extend lycopene to high-value astaxanthin, we co-expressed four genes encoding for phytoene synthase (crtB), β-carotene hydroxylase (BHY), β-carotene ketolase (BKT), and lycopene β-cyclase (LCYB), respectively, in the fruit of the landrace via linking the genes with 2A-sequences and driving their expression with the E8 promoter. The transgenic lines showed similar phenotypes to WT except for the fruit colors. The transgenic fruit exhibited deep red color due to the accumulation of novel ketocarotenoids including astaxanthin, canthaxanthin, and ketolutein and enhanced production of native carotenoids which were 20 ~ 44-fold of the control. The T0, T1, and T2 plants exhibited similar growing status and carotenoid profiles, indicating that the transformants were genetically stable. The transgenes resulted in upregulation of most of the endogenous carotenogenic genes in fruit at mature green stage and more plastoglobules in chromoplast. This study provides insights into metabolic engineering of tomato for enhanced production of value-added carotenoids.

Abstract

BackgroundPhlebopus portentosus and mealy bugs form a fungus-insect gall on the roots of host plants. The fungus and mealy bugs benefit mutually through the gall, which is the key link in the nutritional mechanism of P. portentosus. The cavity of the fungus-insect gall provides an ideal shelter for mealy bugs survival and reproduction, but how does P. portentosus benefit from this symbiotic relationship?Methodology and resultsAnatomical examination of fungus-insect galls revealed that one or more mealy bugs of different generations were living inside the galls. The mealy bug's mouthpart could penetrate through the mycelium layer of the inside of the gall and suck plant juice from the host plant root. Mealy bugs excreted honeydew inside or outside the galls. The results of both honeydew agar medium and quartz tests showed that the honeydew can attract and promote the mycelial growth of P. portentosus. A test of the relationship between the honeydew and the formation of the fungus-insect gall showed that honeydew promoted gall formation.ConclusionsAll experimental results in this study show that the honeydew secreted by mealy bugs can attract and promote the mycelial growth of P. portentosus, forming a fungus-insect gall, because mealy bugs' honeydew is rich in amino acids and sugars.
