The pile-fermentation conditions and raw materials used play a vital role in determining the stability and quality of dark tea. In this study, sensory quality evaluation, metabolomics, and microbiomics techniques were used to investigate the effect of pile-fermentation duration on the taste quality of single-cultivar large-leaf dark tea (SLDT) and its underlying metabolite and microbial mechanisms. The study revealed that a 60-day duration resulted in a better SLDT sensory quality, with astringency and bitterness significantly reduced and sweetness increased. Catechins and theaflavins with ester structures, L-epicatechin, methyl gallate, protocatechuic acid, gallic acid, salicin, chlorogenic acid, and neochlorogenic acid were key taste metabolites contributing to the reduction of astringency and bitterness. Salicylic acid and D-sorbitol helped form the sweetness. Correlation analysis found out Aspergillus, Thermomyces, Bacillus, Staphylococcus, and Micrococcaceae were core functional microorganisms linked to these metabolites, helping to foster the higher quality of SLDT. Microorganisms shaped the taste quality of SLDT through metabolic processes and enzyme secretion during pile-fermentation. This study provided insights into the metabolite basis and microbiological mechanisms of SLDT taste formation and offered guidance for optimizing production processes to improve the stability and quality of dark tea.

Rosemary extract (RE) had strong antioxidant activity which was used as an ideal substitute for antibiotic. However, few studies were explored its effects on fish species. Therefore, present study was conducted to determine the effects of RE on disease-resistance and nutrient metabolism in largemouth bass. Fish with initial body weight 9.07 g ± 0.01 g were fed with diets supplemented with gradient RE levels (0 %, 0.3 %, 0.6 % and 0.9 %) for 8 weeks. Following 8 weeks trial, Edwardsiella tarda challenge test were conducted to explore the ability of disease-resistance. Data on Edwardsiella tarda challenge test showed that all experimental groups exhibited higher survival rate than control group. RE supplementation significantly decreased the Ca2+ concentration and apoptosis rate and increased the mitochondrial membrane potential in blood cells. Meanwhile, the plasma biochemical indexes of antioxidant, immune response and key regulators in antioxidant, immune response presented an increasing pattern after RE supplementation. The growth trial results showed that visceral adipose index (VAI) and abdominal fat weight (AFW) showed a decreasing trend with the increase of RE addition levels, which further decreased growth performance. Further analyzed the lipid metabolism, we found that dietary RE addition markedly inhibited growth hormone-insulin like growth factor (GH-IGF), target of rapamycin (TOR) signalings and activated amino acid response (AAR) signaling to enhance lipid catabolism. Beyond that, the increased antioxidant capacity in turn reduces muscle cooking loss, hardness and increased muscle cohesiveness, springiness, chewiness. In conclusion, based on immunity, antioxidant capacity, disease-resistance and muscle quality, dietary 0.3–0.6 % RE addition was suggested in aquafeeds.

Butene-based elastomers were prepared by bulk polymerization of butene-1 and he-xene-1 using self-made supported titanium catalyst, and the optimization of polymerization process was studied. The structure of the copolymers was characterized by nuclear magnetic resonance spectrometer and differential scanning calorimetry, and the physical and mechanical properties of the copolymers were investigated. The results showed that the optimum polymerization conditions were as follow：n(monomers)/n(catalyst) was in range of 20 000 to 25 000, n(triethylaluminum)/n(supported titanium catalyst) was in range of 250 to 300, reaction tem-perature was 45 ℃, reaction time was 4 h，hydrogen pressure was in range of 0.10 to 0.15 MPa. The copolymer contained a random copolymer structure of butene-1 and hexene-1. The copolymer had no fixed melting point. The tensile strength and elastic modulus of the copolymer decreased with the increase of the amount of hexene-1, and the nominal fracture strain of the copolymer reached 534% when the mass fraction of hexene-1 was 20%.

Abstract It is important to explore the effective approaches to prevent dry age-related macular degeneration (AMD). In this study, significantly decreased full-field electroretinograms wave amplitudes and disordered retina structures were detected in rat retinas of sodium iodate induced dry AMD model. Six a- and b-wave amplitudes and the antioxidant activities were significantly increased, and the outer nuclear layer thickness was significantly improved in the rat retinas treated with the combination of Lactobacillus fermentum NS9 (LF) and aronia anthocyanidin extract (AAE) compared with the model. The effects were much better than the treatment with AAE alone. The proteomics analysis showed the expressions of α-, β- and γ-crystallins were increased by 3–8 folds in AAE treated alone and by 6–11 folds in AAE + LF treatment compared with the model, which was further confirmed by immuno-blotting analysis. Analysis of gut microbial composition indicated that higher abundance of the genus Parasutterella and species P. excrementihominis was found in the AAE + LF treatment compared with the other groups. The results indicated that the combined treatment of AAE + LF is a potential way to prevent the retina degeneration which is significantly better than the AAE treated alone.

Abstract Crocodiles are cultured in large numbers in Asia and other places in order to protect wild resources and meet the needs of human life. In this study, crocodile (Crocodylus siamensis) meat proteins were extracted and hydrolyzed into peptides, their antioxidant peptides were isolated and purified by silica gel chromatography and identified by LC/MS. Crocodile meat proteins were optimally extracted with water and hydrolyzed by papain based on the degree of hydrolysis and antioxidant activity. The hydrolysates were fractionated by ultrafiltration into 3 kDa, 3–30 kDa, and ≥ 30 kDa fractions. The 3 kDa fraction showed most antioxidant activity of the hydrolysates. Its active peptides were separated by silica gel column chromatography and purified by silica gel TLC, based on TLC bio-autographic assays of the activity. Four highly active peptides were identified by LC/MS as SSLTIQFVEGQFVDSYDPTIENTFTK, VPPHIY, VAPEEHPVLLTEAPLNPK, and RNGLPGPIGPAG. The identified peptides were synthesized and showed 50% free radical scavenging activities at 1.0 mg/mL, equal or higher to ascorbic acid at 0.5 mg/mL, in both DPPH and ABTS assays. The results indicated that the 3 kDa hydrolyzed peptides of crocodile meat had high antioxidant activity and the active peptides can be effectively separated and purified by silica gel column chromatography and TLC.

Atherosclerosis, a chronic inflammatory condition characterized by plaque formation, often leads to instability, particularly under Type 2 diabetes mellitus (T2DM) conditions, which exacerbate cardiovascular risks. However, the molecular mechanisms underlying this process remain incompletely understood. In this study, we investigated the correlation between acute coronary syndrome (ACS) and serum levels of Nε-carboxyethyl-lysin (CEL), a prominent advanced glycation end product (AGE) elevated in T2DM, in a cohort of 225 patients with coronary artery disease. Using a murine model of atherosclerosis complicated by T2DM, we examined the effects of CEL on plaque stability and macrophage autophagy. Our findings revealed that elevated serum CEL levels are independently associated with increased ACS incidence. Metabolomic profiling identified CEL as a key AGE contributing to plaque instability in diabetic conditions. Mechanistically, CEL disrupted macrophage autophagy and plaque stability by perturbing the Receptor for Advanced Glycation End products (RAGE)/Liver Kinase B1 (LKB1)/AMP-activated Protein Kinase 1 (AMPK1)/Sirtuin 1 (SIRT1) signaling cascade. This pathway further regulated autophagic activity through SIRT1-mediated acetylation of Zinc Finger with KRAB and SCAN Domains 3 (ZKSCAN3). These findings highlight CEL's critical role in promoting plaque instability in T2DM by impairing key molecular pathways that regulate autophagy, offering potential therapeutic targets for managing atherosclerosis in diabetic patients. One of the long-standing challenges in the field of titanium matrix composites is achieving the synergistic optimization of high strength and excellent ductility. When pursuing high strength characteristics in materials, it is often difficult to consider their ductility. Therefore, this study prepared a Ti1400 alloy and in situ synthesized TiC-reinforced (TiC + Ti1400)/TC4 composites using low-energy ball milling and spark plasma sintering technology, followed by hot rolling, to obtain titanium matrix composites with excellent mechanical properties. The Ti1400 alloy bonded well with the matrix, forming uniformly distributed Ti1400 regions within the matrix, and TiC particles were discontinuously distributed around the TiC-lean regions, forming a three-dimensional network structure. The (TiC + Ti1400)/TC4 composites effectively enhanced their yield strength to 1524 MPa by using 3 wt.% of Ti1400 alloy while preserving an impressive elongation of 9%. When the Ti1400 alloy content reaches 20 wt.%, the overall mechanical properties of the composites decrease. A small amount of Ti1400 does not reduce the strength of the composite. On the contrary, it can undergo stress-induced phase transformation during plastic deformation, thereby coordinating deformation, which not only provides higher strain hardening and increases tensile strength but also benefits ductility. Background: A meta‐analysis study was conducted to determine how to predict the effect of postoperative atrial fibrillation after cardiac surgery. Hypothesis: Long‐term mortality and cardiovascular morbidity are linked to postoperative atrial fibrillation. Method: Until August 2024, a comprehensive literature study was completed, and 3486 connected studies were revised. The 38 selected studies included 241 299 cardiac surgery participants at the beginning of the study. The odds ratio (OR) and 95% confidence intervals (CIs) were used to look at the effect of atrial fibrillation after heart surgery using two‐sided methods and either a fixed or random model. Results: Individuals with cardiac surgery with postoperative atrial fibrillation had significantly higher mortality at 1 year (OR, 1.39; 95% CI, 1.12–1.72, p < 0.001), mortality at 5 years (OR, 1.61; 95% CI, 1.33–1.94, p < 0.001), mortality at 10 years (OR, 1.61; 95% CI, 1.39–1.87, p < 0.001), and overall stroke (OR, 1.61; 95% CI, 1.34–1.94, p < 0.001) compared to without postoperative atrial fibrillation. Conclusions: Individuals with cardiac surgery with postoperative atrial fibrillation had significantly higher mortality at 1 year, mortality at 5 years, mortality at 10 years, and overall stroke compared to those without postoperative atrial fibrillation. To validate this discovery, more research and caution must be implemented when interacting with its values. Summary: Postoperative atrial fibrillation is the most frequent side effect following heart surgery. Rare earth elements (REEs) are indispensable in modern industry and technology, driving an urgent demand for innovative, eco-friendly recovery technologies. As space exploration advances, the impact of microgravity on microorganisms has become a focal point, yet the effects on microbial growth and REEss recovery remain uncharted. This study investigates the biosorption of REEs by Burkholderia cepacia (B. cepacia) and Aspergillus niger (A. niger) from a mixed solution containing La, Ce, Pr, Nd, Sm, Er, and Y under varying initial concentrations, pH levels, and microgravity conditions. We observed that the medium's pH rose with B. cepacia and fell with A. niger when cultured in normal gravity conditions, suggesting distinct metabolic responses. Notably, microgravity significantly altered microbial morphology and metabolite profiles, significantly enhancing REEs recovery efficiency. Specifically, the recovery of B. cepacia of Ce and Pr peaked at 100%, and A. niger achieved full recovery of all tested REEs at pH 1.5 (suboptimal growth conditions). This study pioneers the application of biosorption for the recovery of REEs in microgravity conditions, presenting a promising strategy for future resource exploitation by space biomining. Abstract Background Methyltransferase-like 3 (METTL3) is a member of the m6A methyltransferase family and acts as an oncogene in cancers. Recent studies suggest that host innate immunity is regulated by the enzymes controlling m6A epitranscriptomic changes. Here, we aim to explore the associations between the levels of METTL3 and CD33+ myeloid-derived suppressor cells (MDSCs) in tumour tissues and the survival of patients with cervical cancer (CC). Methods Specimens of paraffin embedded tumour from 197 CC patients were collected. The expression levels of METTL3 and CD33 were measured by immunohistochemical (IHC) staining. The clinical associations of the IHC variants were analysed by Pearson’s or Spearman’s chi-square tests. Overall survival (OS) and disease-free survival (DFS) were estimated by the Kaplan–Meier method and log-rank test. Hazard ratios (HRs) and independent significance were obtained via Cox proportional hazards models for multivariate analyses. METTL3 in CD33+ cells or CC-derived cells was knocked down by METTL3-specific siRNA, and MDSC induction in vitro was performed in a co-culture system in the presence of METTL3-siRNA and METTL3-knockdown-CC-derived cells compared with that of the corresponding controls. Results We found that tumour tissues displayed increased levels of METTL3 and CD33+ MDSCs compared with tumour-adjacent tissues from the same CC patients. Importantly, METTL3 expression was positively related to the density of CD33+ cells in tumour tissues (P = 0.011). We further found that the direct CD33+CD11b+HLA-DR− MDSC induction and tumour-derived MDSC induction in vitro were decreased in the absence of METTL3. The level of METTL3 in tumour microenvironments was significantly related to advanced tumour stage. The levels of METTL3 and CD33+ MDSCs in tumour tissues were notably associated with reduced DFS or OS. Cox model analysis revealed that the level of METTL3 in tumour cells was an independent factor for patient survival, specifically for DFS (HR = 3.157, P = 0.022) and OS (HR = 3.271, P = 0.012), while the CD33+ MDSC number was an independent predictor for DFS (HR: 3.958, P = 0.031). Interestingly, in patients with advanced-disease stages (II–IV), METTL3 in tumour cells was an independent factor for DFS (HR = 6.725, P = 0.010) and OS (HR = 5.140, P = 0.021), while CD33+ MDSC density was an independent factor for OS (HR = 8.802, P = 0.037). Conclusion Our findings suggest that CD33+ MDSC expansion is linked to high levels of METTL3 and that METTL3 and CD33+ MDSCs are independent prognostic factors in CC.
