Background: Hepatozoonosis has been reported in many species around the world. Few incidences have been reported in various species of wild felids. Tigers are endangered large cats and are protected under the Wildlife Protection Act, 1972 under Schedule I. The study was carried out to estimate the positivity rate of hepatozoonosis in tigers of the Vidarbha region of Maharashtra, India. Methods: Blood (n = 21) or tissue samples (n = 5) were collected from 26 wild captured / zoo-born or dead tigers during the quarantine period/post-mortem examination. Blood smear examination along with Polymerase Chain Reaction (PCR) studies were conducted for the detection of hepatozoonosis. All the amplicons from the positive samples were purified and sequenced, and the sequences were subjected to nBLAST analysis to detect the species of Hepatozoon. The sequences were deposited into public domain database of National Center for Biotechnology Information (NCBI) and accession numbers were allotted. A phylogenetic study was undertaken to understand the evolutionary lineage of the pathogen. Tissue distribution studies were carried out on tissue samples received during post mortem. A clinical case in a tiger cub was managed and sub-clinical cases were monitored for relapse. Age-wise, sex-wise, region-wise and captive time-wise positivity rate was estimated. The data was analyzed using statistical tools. Results: A total of 12 tigers were found positive for H. felis during the screening. A clinical case was diagnosed and successfully treated. The age group of 0–3 years reported a positivity rate of 66.66%, and all the cases found positive were reported between the age group of 0–7 years. Males reported a positivity rate of 58.33 per cent, while females reported 35.71%. Taboba and Andhari Tiger Reserve of the state had a positivity rate of 52.94 per cent. However, the statistical analysis for blood parameters and positivity rate by 't' test and Chi-squared test were found to be non-significant. Conclusions: An overall positivity rate of 46.15% indicates the wide distribution of hepatozoonosis among wild tigers of the Vidarbha region of Maharashtra, India, which is strategically important considering the gene flow and migration of tigers. Hepatozoonosis can progress to clinical outcomes in young animals and require veterinary intervention. Molecular tools and phylogenetic studies can supplement important data on circulating species of Hepatozoon in the field. In vertebrate hosts, the disease primarily occurs through ingestion of the hemoparasite or its components. Hepatozoon spp. infection typically remains subclinical, only becoming diagnosed upon microscopic observation of the hemoparasite during blood smear analysis. However, in certain instances, moderate or severe clinical manifestations may occur. Due to limited research focusing on Hepatozoonosis, diagnosis can present challenges, as well as selecting the appropriate test to confirm it, with PCR being considered the preferred method for detection. A two-and-a-half-month-old Golden Retriever puppy was examined, exhibiting symptoms of vomiting, lethargy, bloody diarrhea, anorexia, and hyperthermia (39°C - 102,2°F). Diagnosis was confirmed by observation of Hepatozoon spp. parasites in the blood smear, alongside severe anemia, presence of microagglutination, and elevated total protein levels. The treatment was initiated with Doxycycline, and following the patient´s improvement, the owner opted to proceed with castration and subsequently administer two doses of Imidocarb Dipropionate at 5mg/kg with a 14-day interval.

Babesia is a diverse genus of piroplasms that parasitize the red blood cells of a wide variety of mammals and avian species, including humans. There is a lack of knowledge on the Babesia species of carnivores and mesomammals in the eastern United States and the potential impacts of these species on the health of humans and domestic animals. We surveyed 786 wild mammals in the eastern United States by testing blood, spleen, and heart samples with PCR targeting the 18S rRNA region of apicomplexan parasites. We also performed PCR targeting the cytochrome c oxidase subunit 1 (cox1) region of each unique clade identified with 18S rRNA sequencing. We found a high positivity of Babesia spp. infection in raccoons (Procyon lotor), foxes (Vulpes vulpes and Urocyon cinereoargenteus), and striped skunks (Mephitis mephitis), and low positivity in Virginia opossums (Didelphis virginiana). No Babesia infections were detected in coyotes (Canis latrans), black bears (Ursus americanus), groundhogs (Marmota monax), muskrats (Ondatra zibethicus), or mink (Neovison vison). Skunks carried a diverse number of strains including a potential novel species of Babesia related to B. gibsoni, a strain closely related to a B. microti-like species known to cause disease in river otters, as well as a separate B. microti-like strain. Raccoons primarily carried B. microti-like strains, though there was a high diversity of sequences including Babesia lotori, Babesia sensu stricto MA230, and Babesia sp. ‘Coco.’ Foxes exclusively carried B. vulpes. In addition to Babesia spp., a high positivity of Hepatozoon spp. infection was found in mink, while low positivity was found in raccoons and muskrats. Wildlife in the eastern United States carry a diverse range of Babesia species including several novel strains of unknown clinical significance.

Abstract Background Canine hepatozoonosis caused by Hepatozoon canis is a common infection in dogs, with frequent case reports from the Mediterranean region and more recently from several Central European countries, such as Hungary and Germany. Despite the high prevalence of H. canis in red foxes, no infections have been reported to date in dogs in Poland. We describe here the first autochthonous cases of H. canis infection in dogs, including their clinical features, and report the prevalence of H. canis in grey wolves from different regions of Poland. Methods Thin smears prepared from blood samples collected from dogs were evaluated by microscopic examination. A total of 60 wolves and 47 dogs were tested. Infections were confirmed by PCR and sequencing. Results Gamonts of H. canis were found in > 50% of the neutrophils of two dogs and in < 10% of the neutrophils in another five dogs. Molecular typing by PCR sequencing of the 18S ribosomal RNA gene fragment confirmed infections in 11 dogs from different regions of Poland, in 2.7% of dogs attending veterinary practices in central Poland and in 35% of wolves from various geographical regions of Poland. Clinical features manifested mostly in older dogs, and the most common signs were anaemia and apathy. Young dogs usually remained asymptomatic. Conclusions This is the first report of H. canis infection in dogs and wolves in Poland. Although the exact vector of the parasite is not known, veterinary practitioners should be aware of this new parasitosis and should consider appropriate diagnostics to confirm/exclude this infection. Feline hepatozoonosis is a vector-borne disease caused by different species of the genus Hepatozoon, i.e. Hepatozoon felis, Hepatozoon silvestris and Hepatozoon canis. Knowledge on the biology, epidemiology and taxonomy of Hepatozoon spp. is still limited, despite the fact that the number of documented Hepatozoon spp. infections in domestic cats increased in recent years in different countries. This study was carried out to evaluate the prevalence and the genetic profile of Hepatozoon spp. in cats living on the island of Skopelos, Greece. Individual blood samples were collected from 54 owned cats and were subjected to Giemsa-stained blood smear examination to investigate the presence of Hepatozoon spp. gamonts and to a specific PCR protocol targeting the 18S rRNA gene of Hepatozoon. A total of 45 cats (83.3%) were found infected by Hepatozoon spp. by at least one of the methods applied. In particular, 43 (79.6%) of the cats were PCR-positive, and in 6 (11.1%) cats gamonts of Hepatozoon spp. were found in the blood smears. A total of 26 H. felis sequences were obtained and the presence of three undescribed single nucleotide polymorphisms were detected. The present results indicate that H. felis species complex may be hyperendemic in isolated/confined areas. In such contexts, geographical isolation may favor the origin of new genotypes or haplotypes or even new species.

Incidence of hepatozoonosis has been described in a variety of animal species but information on cats is scarce. To investigate the occurrence of this pathogen in domestic cats, blood samples were collected from domestic cats from three districts of Kerala (Thrissur, Kannur and Wayanad). Field-stained blood smears were examined to detect the presence of gamonts of Hepatozoon spp. and the samples were subjected to molecular analysis by PCR amplification. Out of 122 blood samples screened, none of the sample revealed the presence of gamonts of Hepatozoon on microscopic examination. Polymerase chain reaction targeting 18S rRNA gene of Hepatozoon spp. demonstrated seven positive cases with a prevalence of 5.74 per cent. Results of this study indicate that hepatozoonosis is established within the domestic cats in Kerala and warrant the adoption of control measures.

Simple Summary: There is currently no available data on the prevalence of vector-borne pathogens (VBPs) in stray cats in Germany. In this study, clinically healthy stray cats were investigated for selected VBPs. Of the 50 stray cats included, 22% tested positive for at least one VBP by direct and 64% by indirect detection methods. Stray cats may therefore represent important pathogen reservoirs and contribute to the spread of VBPs, many of which have zoonotic potential, for example, Bartonella spp., Rickettsia spp., and Anaplasma phagocytophilum. Bacterial, protozoal, and viral vector-borne pathogens (VBPs) can cause infections in cats. There is little information on feline VBP prevalence in Germany. Stray cats are frequently exposed to vectors but receive no veterinary care. The aim of this study was to determine the prevalence of selected VBPs in stray cats. EDTA blood and serum samples were taken from apparently healthy stray cats during a spay/neuter campaign in the federal state of Thuringia. Overall, 11/50 (22%) and 32/50 (64%) cats tested positive for at least one VBP by direct and indirect detection methods, respectively. PCR testing of EDTA blood detected hemotropic Mycoplasma spp. in 12% of cats, Hepatozoon spp. in 10%, and Anaplasma phagocytophilum in 4%. PCR testing for Rickettsia spp. and piroplasms was negative. IFAT on serum samples showed 46% of cats had detectable antibodies for Bartonella spp., 30% for Rickettsia spp., and 16% for A. phagocytophilum. The cats were additionally tested for feline coronavirus, FIV, and FeLV to identify potential risk factors for pathogen contact and/or infections. No correlation between FIV and FeLV status and VBP positivity was detected. Haemoparasitic infections persist as one of the major challenges in companion animal health. Despite the increased incidence, studies on tick-borne apicomplexan haemoparasites infecting domestic cats are comparatively fewer. These vector-borne diseases are emerging problems in cats across the globe. Feline haemoparasites include Hepatozoon spp. such as H. felis, H. canis and H. silvestris, Piroplasm-causing organisms such as Cytauxzoon felis, Babesia felis, B. cati, B. herpailuri and B. pantherae and haemoflagellates such as Trypanosoma evansi. Ticks have the major role in the transmission of hepatozoonosis, cytauxzoonosis and babesiosis. Haematophagous insects including Tabanus, Stomoxys and Lyperosia act as vectors for trypanosomosis. Routine diagnosis of infection is based on microscopic identification of parasites in blood smears or tissues. Application of molecular diagnostic techniques like polymerase chain reaction (PCR) for rapid diagnosis and epidemiological studies will enhance the potential of veterinary services. Knowledge on occurrence and clinical manifestations of haemoparasitic infections will aid in improving the health of cats and facilitate the development of an efficient disease control strategy. In the last few years, the number of studies on feline hepatozoonosis has increased, but our knowledge on the actual species of Hepatozoon and/or different genotypes affecting felines is still incipient. At least three species, namely Hepatozoon felis, H. canis, and H. silvestris, have been isolated from domestic cats in various countries. Additionally, there are indications that other species and genotypes may affect felines in given geographic areas. This study was carried out to investigate the occurrence of Hepatozoon spp. in cats from Niterói, a municipality within the metropolitan area of Rio de Janeiro, Brazil. Individual blood samples were collected from 28 cats enrolled in a spaying/castration program. DNA was extracted from all samples and subjected to sequencing specific for Hepatozoon spp. DNA of H. felis was found in 21/28 cats (75%), and four genetic polymorphisms never described thus far were detected. This is the first report of H. felis in cats living in the State of Rio de Janeiro, and the present data confirm that H. felis is a species complex encompassing different genotypes circulating within cat populations. Further studies are warranted to investigate whether different genotypes have different biology or pathogenicity for felids.

Abstract Background Hepatozoon canis is a protozoal agent that is known to be transmitted by oral uptake of H. canis-infected Rhipicephalus sanguineus sensu lato ticks in dogs. Vertical transmission of H. canis has only been described once in a study evaluating dogs from Japan. The aim of this study was to investigate the parasitological status of puppies from a bitch that had tested positive for Hepatozoon spp. prior to giving birth. Findings A 4-year-old, female, pregnant dog imported from Italy (Sardinia) to Germany showed clinical signs of lethargy and tachypnoea and tested positive for H. canis by PCR. The dog gave birth to eight puppies, one of which was stillborn and another that had to be reanimated. Haematology, buffy coat analysis and a biochemistry profile were performed for each dog. EDTA-blood of the surviving seven puppies and bone marrow, liver, spleen, amniotic fluid, and umbilical cord of the stillborn puppy was tested for the presence of Hepatozoon spp. by PCR. The mother and the seven surviving puppies tested positive for H. canis by PCR at day 62 post-partum. Gamonts were detected in all dogs by buffy coat evaluation. Haematological and biochemistry results revealed mild abnormalities. In the stillborn puppy, spleen, umbilical cord, and amniotic fluid were positive for H. canis. Conclusion The results confirm that vertical transmission is a possible route of H. canis infection in dogs, demonstrated by molecular detection of the pathogen in the stillborn puppy. In the seven surviving puppies, vertical transmission was the most likely transmission route. A potential impact of the level of parasitaemia on the health of puppies, as well as its pathogenesis, should be investigated further. Graphical Abstract

Objectives: Three species of protozoal Hepatozoon species (H felis, H canis and H silvestris) are known to infect cats in Europe. The objective of this study was to determine the prevalence of Hepatozoon species in samples from cats living in Germany that were submitted to a veterinary laboratory. Methods: The study included cats tested for Hepatozoon species by PCR between 2007 and 2020 by the Laboklin laboratory. Travel history and haematological results were documented for cats with positive test results. From 2018 onwards, a partial 18S rRNA Hepatozoon gene fragment was sequenced from cats with positive PCR results. Results: Sixty-four of 931 cats (7%) tested positive for Hepatozoon species. Sex and age did not have a statistically significant impact. Sequencing was carried out for 16 samples and revealed H felis in all cases. All cats with positive test results and a relevant travel history had been imported from the Mediterranean or south-eastern Europe. There were no autochthonous infections with Hepatozoon species. Leukocytosis, haemoconcentration and anaemia were the most common haematological abnormalities. Conclusions and relevance: Although infections with Hepatozoon species in cats are usually subclinical, it may be useful to screen cats imported from the Mediterranean and south-eastern Europe for these pathogens to prevent local transmission cycles. There was no evidence of autochthonous infections in Germany; however, further investigations regarding a possible transmission of Hepatozoon species from infected cats to blood-feeding arthropods in Germany may be of interest. EDTA-blood of the surviving seven puppies and bone marrow, liver, spleen, amniotic fluid, and umbilical cord of the stillborn puppy was tested for the presence of Hepatozoon spp. by PCR. The mother and the seven surviving puppies tested positive for H. canis by PCR at day 62 post-partum. Gamonts were detected in all dogs by buffy coat evaluation. Haematological and biochemistry results revealed mild abnormalities. In the stillborn puppy, spleen, umbilical cord, and amniotic fluid were positive for H. canis. Conclusion: The results confirm that vertical transmission is a possible route of H. canis infection in dogs, demonstrated by molecular detection of the pathogen in the stillborn puppy. In the seven surviving puppies, vertical transmission was the most likely transmission route. Feline hepatozoonosis is a vector-borne disease caused by different species of the genus Hepatozoon , i.e. Hepatozoon felis , Hepatozoon silvestris and Hepatozoon canis . Knowledge on the biology, epidemiology and taxonomy of Hepatozoon spp. is still limited, despite the fact that the number of documented Hepatozoon spp. infections in domestic cats increased in recent years in different countries. This study was carried out to evaluate the prevalence and the genetic profile of Hepatozoon spp. in cats living on the island of Skopelos, Greece. Individual blood samples were collected from 54 owned cats and were subjected to Giemsa-stained blood smear examination to investigate the presence of Hepatozoon spp. gamonts and to a specific PCR protocol targeting the 18S rRNA gene of Hepatozoon . A total of 45 cats (83.3%) were found infected by Hepatozoon spp. by at least one of the methods applied. In particular, 43 (79.6%) of the cats were PCR-positive, and in 6 (11.1%) cats gamonts of Hepatozoon spp. were found in the blood smears. A total of 26  H. felis sequences were obtained and the presence of three undescribed single nucleotide polymorphisms were detected. The present results indicate that H. felis species complex may be hyperendemic in isolated/confined areas. Abstract This study aimed to investigate the genetic diversity of Hepatozoon spp. in rodents from Valdivia, Chile. A total of 74 rodents (synanthropic n=38; wild n=36) were trapped in Valdivia. We performed conventional PCR assays for Apicomplexa organisms targeting two overlapping 18S rDNA gene fragments (600 bp and 900 bp) followed by sequencing of selected amplicons. Hepatozoon spp. occurrence was 82.43% (61/74). Twelve sequences obtained from the 600 bp and ten from the 900 bp 18S rDNA fragments were identified as Hepatozoon sp. Six sequences obtained from 18S rDNA-based overlapping PCR protocols were used for concatenated (1,400 bp) phylogenetic, haplotype and distance analyses. Hepatozoon spp. 18S rDNA concatenated sequences from the present study were detected in Oligoryzomys longicaudatus, Rattus norvegicus, Mus musculus, and Abrothrix longipilis grouped with Hepatozoon species earlier described in rodents and reptiles from Chile and Brazil. Nucleotide polymorphism of the six 18S rDNA sequences (1,400 bp) from this study, and other Chilean sequences from rodents and rodent’s ticks, showed high diversity with a total of nine Chilean haplotypes. Three haplotypes from Valdivia were identified for the first time in this study, suggesting the circulation of novel haplotypes in rodents from southern Chile.

In this study, a case of hepatozoonosis with concurrent TVT unresponsive to chemotherapy in a 10-year old female German shepherd dog treated with Vincristine for 10 weeks at the State Veterinary Clinic, Abeokuta, was referred to the Veterinary Teaching Hospital, Federal University of Agriculture, Abeokuta, Nigeria. A diagnosis of hepatozoonosis and TVT were made based on clinical presentations including protruding mass from the vulva, parasitological technique and cytological evaluation of the tumour mass and bone marrow using optical fine needle aspirate. Pathological changes observed in various organs at post-mortem, following euthanasia were cachexia, pale and icteric ocular and oral mucous membranes, enlarged and oedematous lymph nodes. Histopatholgical lesions were round to oval neoplastic cells from the growth observed in the vagina. There were different developmental stages of the merogony of Hepatozoon canis in the bone marrow. We concluded that concurrent hepatozoonosis and TVT and, possibly, the geriatric state of the patient contributed largely to the problem of unresponsiveness to treatment. Hepatozoonosis is caused by protozoa of the genus Hepatozoon. In dogs, the infection is caused mainly by Hepatozoon canis, and there are a few descriptions of the prevalence of this infection in the Northeast region of Brazil, especially in the semi-arid region. Therefore, we aimed to determine the prevalence of Hepatozoon canis infection in dogs in the rural area of Sousa, Paraíba, Brazil, as well as to determine the possible clinical and epidemiological aspects of this infection. Ninety-eight dogs in the rural zone of the municipality of Sousa that were at least 4 months old were evaluated, regardless of their breed or gender. Clinical examinations were carried out, and samples of systemic and peripheral blood were collected to determine the presence of the parasite in blood smears and carry out hemograms. In addition, epidemiological questionnaires about animal health and food management were completed. The prevalence of H. canis infections in dogs was 8.1% (8/98). There were three main changes in the hematological status: thrombocytopenia, anemia and hyperproteinemia, mainly related to percentage of leukocyte infection ≥5%, and also to the presence of clinical signs such as mucopurulent secretion, lymphadenomegaly, dry skin, pale mucous membranes, and lean or cachectic body score. There is not any available data about the presence of Hepatozoon infections in dogs in Kyrgyzstan. In the study we aimed that investigate the presence of Hepatozoon canis and the prevalance of Hepatozoon infections in dogs from Kyrgyzstan using polymerase chain reaction (PCR) and sequence analysis. To determine the prevelance of hepatozoonosis in dogs, a total of 170 blood samples were applied to PCR to amplify a fragment of 666 bp found in 18S SSU rRNA gene of Hepatozoon spp. The PCR results shown that Hepatozoon infection rate was 28.8% (49/170) in dogs. Eleven representative positive samples were sequenced to classification of the species. The nucleotide sequences were compared to the H. canis sequences which registered in GenBank using the basic local alignment search tool. Results of sequence analyse of 11 amplicons indicated that 8 were 100% identical and the other 3 sequences shared 99% similarity with H. canis. The sequences were deposited in Genbank with accession numbers from MG917709 to MG917719. It was the first record of H. canis in dogs in Kyrgyzstan.

Genetic diversity within partial 18S rRNA sequences from Hepatozoon protozoan parasites from wild felines in South Africa was assessed and compared with data from domestic cats to assess patterns of host specificity. Lions, leopards, servals, a caracal and an African wildcat were all positive for parasites of the Hepatozoon felis -complex. However, haplotypes were not species-specific, and potential mixed infections were widespread. Genetic diversity within partial 18S rRNA sequences from Hepatozoon protozoan parasites from domestic cats in South Africa was assessed and compared against published data to assess global biogeographic patterns. Multiple distinct haplotypes of Hepatozoon felis were identified, as well as an unrelated Hepatozoon lineage. Hepatozoon felis genetic diversity globally is very high, indicating a likely complex of species. The recently described Hepatozoon apri from wild boars is closely related to some lineages of H. felis. Sarcocystis and Babesia parasites were also detected. Since Hepatozoon felis is apparently a species complex, potential differences between genetically distinct forms need to be assessed. The finding of an unrelated Hepatozoon indicates that felids can be infected by more species of Hepatozoon than currently known, and that trophic interactions may increase the number of Hepatozoon species found in carnivores. A dog was presented with epistaxis, melena, exercise intolerance, tick infestation and decreased feed intake. Clinical examination revealed pale mucous membranes, pyrexia, lymphadenopathy, depression and respiratory distress. Hematobiochemical examination revealed anemia, leukocytosis and thrombocytopenia, decreased levels of total serum protein, albumin and elevated creatinine. Upon microscopic examination of peripheral blood and buffy coat smear, H. canis morulae were identified in neutrophils and E. canis was also detected by nested PCR. The case was diagnosed as mixed Ehrlichia canis and Hepatozoon canis infection. Dog was treated with Doxycycline @10 mg/ kg b. wt. once in a day orally for twenty eight days. Imidocarb dipropionate 12% w/v injection @ 6.6 mg/ kg b. wt. subcutaneously twice fourteen days apart and other supportive treatment resulted in both clinical and parasitological cure. Hepatozoon apri n. sp. is described from Japanese wild boars Sus scrofa leucomystax in Japan. The gamonts in the peripheral blood leukocytes were 11.6 ± 1.4 × 6.7 ± 1.3 μm in size. The meronts in the muscle tissues were 35.0–47.5 μm in length and 26.5–30 μm in width. A high rate (53.0%) of infection was found by nested PCR using muscle specimens from 181 wild boars captured in Tokushima, Japan. A phylogenetic analysis based on 18S rRNA gene sequences revealed that H. apri n. sp. detected in wild boars is closely related to Hepatozoon spp. isolated from carnivores. This report describes a dog infected with Hepatozoon canis, the first canine infection in the Republic of Korea. A 2-year-old intact male Maltese dog presented with anorexia and depression. Physical examinations revealed mild dehydration and hyperthermia (39.8°C), and blood analysis showed pancytopenia. Diff-Quik staining of blood smear specimens showed the presence of ellipsoidal shaped structures (gamonts of H. canis) within a small number of neutrophils. Real-time PCR analysis using whole blood confirmed infection by H. canis. The clinical condition of the dog improved after symptomatic treatment and administration of doxycycline. Although a molecular epidemiologic survey in Korea showed H. canis infection of dogs, to our knowledge this is the first report of a dog infection in Korea molecularly shown to be H. canis. Changing scenario of climate resulting from global warming and adversity of nature has also resulted in emergence and re-emergence of diseases transmitted by arthropods. Increasing trends of population growth of dogs has increased the chance of disease transmission due to readily available susceptible host. Babesiosis and Hepatozoonosis and Ehrlichiosis are the main arthropod borne diseases of dogs prevalent in India. The present article explains the importance of these arthropod transmitted infections in kennel dogs, research progress and reason for their emergence in the present scenario. [Vet. Tick-borne diseases are emerging worldwide and have an important zoonotic relevance. Dogs play an important role in the epidemiology of several zoonotic tick-borne pathogens acting as sentinels and/or reservoirs. This study focused on the molecular identification of tick-borne pathogens in blood samples of 153 autochthonous asymptomatic dogs in Maio Island, Cape Verde archipelago. Eighty-four (54.9%) dogs were positive for one or more pathogens. Fifty-five (35.9%) dogs were infected with Hepatozoon canis , 53 (34.6%) with Anaplasma platys, five (3.3%) with Ehrlichia canis and Rickettsia monacensis , an emerging human pathogen, was also identified in a single dog (0.7%). The former three pathogens cause important canine tick-borne diseases that are transmitted or potentially transmitted by Rhipicephalus sanguineus s.l., the only hard tick identified in Cape Verde. Furthermore, Wolbachia spp. was amplified from the blood of one dog. None of the dogs were positive for Anaplasma phagocytophilum , Borrelia burgdorferi sensu lato, Midichloria mitochondrii , Bartonella spp., Babesia spp. or Theileria spp. Fifty-four (35.3%) animals showed single infections and 30 (19.6%) co-infections, with A. platys and H. canis co-infection being the most frequent (28 dogs, 18.3%). The frequency of E. canis infection was statistically different among age groups ( P = 0.017), being higher among dogs older than 4 years compared to younger dogs. Infection by A. platys was also statistically different among age groups ( P = 0.031), being higher in dogs younger than 2 years compared to older dogs. The statistical analyses showed no significant association of PCR positivity with gender or location. The frequency of tick-borne pathogens detected in dogs in Maio Island, including R. monacensis, highlights the need to improve diagnosis and control in order to prevent the risk of transmission of these pathogens among dogs and humans living in or travelling to this touristic island. • Out of 104 coatis sampled in central-western Brazil, 80 (76.9%) were positive for Hepatozoon procyonis in at least one capture. • Hepatozoon procyonis gametocytes were detected in coatis during 4 annual recaptures, showing fluctuation of the parasitemia. • Taking into account gender, age, season and sampling location, no statistical differences were observed regarding H. procyonis infection. • Hepatozoon procyonis seems not to affect clinical and hematological parameters of infected coatis. This study aimed to morphologically and molecularly detect Hepatozoon procyonis in ring-tailed coatis' (Nasua nasua) blood and associated ticks from central-western Brazil, Campo Grande, Mato Grosso do Sul state and also evaluate the impact of the protozoa in blood parameters and coati´s health. Samplings were performed in a conservation area Parque Estadual do Prosa (PEP) and in a Brazilian Air Force Private Area namely Vila da Base Aérea (VBA), between March 2018 and April 2019. We collected 165 blood samples, 61 from recaptured coatis. Peripheral blood smears were stained with Romanovsky-type stain for H. procyonis parasitemia assessment. DNA extracted from blood samples and ticks (Amblyomma spp.) were submitted to a nested PCR (nPCR) assay based on the 18S rRNA gene for Hepatozoon spp. Out of 104 individuals sampled, 80 (77%) were positive for H. procyonis in at least one capture. Overall, 67/165 (40.6%) blood smears showed H. procyonis gametocytes (PEP: 41/63 - 65%; VBA: 26/102 - 25.5%). Parasitemia based on 500 assessed leucocytes ranged from 1 (0.2%) to 50 (10%) and 1 (0.2%) to 25 (5%), from animals sampled in PEP and VBA, respectively. Fluctuation on the parasitemia was observed during recaptures. nPCR results showed higher positivity when compared to blood smears, i.e. 112/165 (68%) positive blood samples [PEP: 41/63 (65%), VBA: 26/102 (25.5%)]. In total, 63/248 (25.4%) tick DNA samples were positive at nPCR for Hepatozoon sp., including 32/87 (37%) pools (1 to 10 larvae) of Amblyomma larvae, 21/105 (20%) pools (1 to 5 nymphs) of Amblyomma sculptum nymphs, 9/43 (21%) pools (1 to 5 nymphs) of Amblyomma dubitatum nymphs, and 1/12 (8%) A. sculptum adult female. The partial 18S rRNA sequence from one coati's blood sample and one representative of each positive tick species randomly selected from each area for sequencing (1,000 bp) showed 100% identity with sequences of H. procyonis from GenBank previously detected in coatis. Regarding H. procyonis infection, no statistical differences were obtained when comparing males vs. females (p-value 0.67), immature animals vs. adults (p-value 0.31), rainy vs. dry season (p-value 0.51) and sampling location (p-value 0.42). No noticeable alteration in blood parameters or heath status was observed in parasite animals. H. procyonis circulates in a high prevalence in coatis from central-western Brazil. Many tick-borne infections are increasing in geographic range as a result of activities such as reforestation and climate change. A history of outdoor activity in tick-endemic regions, together with consistent clinical signs (such as fever, splenomegaly, polyarthritis, thrombocytopenia), should raise suspicion for tick-borne infectious disease. Diseases with short incubation periods are best diagnosed with organism-detection assays. When the incubation period is long and organism numbers are low, diagnosis often relies on antibody testing, but it may be difficult to associate positive tests with infection. Canine hepatozoonosis is a disease caused by protozoan Hepatozoon sp., recently discovered in Brazil, with few cases. It is transmitted mainly through ingestion of Rhipicephalus sanguineus with oocysts of H. canis. Pathogenicity and symptoms are variable depending on the age of the animal, the degree of infection and the association with other diseases. The symptoms can present in lethal asymptomatic. The diagnosis of hepatozoonosis is based on blood smears, biopsy, serology and PCR. The objective is to describe the clinical and laboratory findings in two dogs with hepatozoonosis met in Fortaleza, Ceará. The first case, male, 3 months old, showed aggression, circling, lethargy hind limbs, anorexia, seizures, pale mucous membranes and eye discharge. The second case, bitch, 1 year old, had anorexia. On physical examination, we observed the presence of ticks, pale mucous membranes, and temperature 38.6 ° C. Blood samples were collected for hemogram, biochemical testing and research hematozoa. The diagnosis was characterized by the display of gametocytes of Hepatozoon sp in neutrophils in blood smears. Regenerative anemia, mild neutrophilia, reactive lymphocytes and monocytes vacuoles were observed. One had thrombocytopenia. Hyperbilirubinemia, hypoalbuminemia are present. ALT levels were within normal limits, however, AST, CPK, BUN and creatinine were found altered. One animal died. We conclude that this is a hepatozoonosis disease in Fortaleza, Ceará. Two male dogs aged between 2 and 3 months belonging to German Shepherd and non descript breed were brought to Outpatient unit of Department of Veterinary Medicine, Veterinary College, Bidar with history of anorexia, weakness and going down in body condition since 15 days. Clinical examination revealed fever, enlargement of popliteal and submandibular lymph nodes, pale mucous membranes and cachectic body condition in both the cases. Haemato-biochemical examination revealed anemia, leukocytosis, thrombocytopenia, hypoalbuminemia and elevated levels of blood urea nitrogen, creatinine and alanine amino transferase. On blood smear examination, gamonts of Hepatozoon canis organisms were noticed in neutrophils. Based on these observations a diagnosis of hepatozoonosis was made. This study was conducted to investigate Babesia canis vogeli, B. canis canis, B. canis rossi, B. gibsoni, Hepatozoon canis, Ehrlichia canis and Anaplasma phagocytophilum species, and to determine the molecular characterizations of the isolates in totally 400 whole blood samples of dogs in Kayseri region. According to the Real Time PCR results, the prevalence of E.canis, B.canis canis, B. gibsoni, A. phagocytophilum, H. canis, and B. canis vogeli was detected as 14.5%, 12.0%, 9.0%, 7.8%, 5.3%, and 2.3%, respectively while B.canis rossi was not detected in the examined samples. 182 (89.7%) out of the 400 samples were found to be infected with a single parasite species, and 21 (10.3%) were found to be infected with two species. According to the pairwise comparisons of 18S rRNA gene region of the isolates under B. canis canis, B.canis vogeli and B. gibsoni, 1.4±0.2%, 0.3±0.2%, and 0.9±0.3% genetic distance were detected with the other similar isolates from the world, respectively. Based on the 16S rRNA gene sequence alignments, 100% identity was found among the 3 isolates of E. canis while 0.1% genetic difference was determined with the isolates from the world. With respect to ankA gene region of A. phagocytophilum, 99.8±0.2% identity and 0.9±0.3% genetic difference were found among the 3 isolates obtained from Kayseri region and the isolates from the world, respectively. The 2 H. canis isolates were showed 100% identity to each other and 0.2±0.1% genetic difference were determined with the other isolates from the world with respect to 18S rRNA gene region. In conclusion, the molecular prevalence of tick-borne protozoon and rickettsial infections in dogs in Kayseri region was determined and the molecular characterizations of the obtained isolates were performed by analyzing the various gene regions in this study. The objective of this survey was to investigate the prevalence of Hepatozoon infection in dogs in the rural and urban areas of Uberlândia, Brazil by PCR and molecular characterization. DNA was obtained from blood samples collected from 346 local dogs from both genders and various ages. Seventeen PCR products from positive blood samples of urban dogs and 13 from the rural dogs were sequenced. Partial sequences of the 18S rRNA gene indicated that all 30 dogs were infected with Hepatozoon canis similar in sequence to H.?canis from southern Europe. Four local dog sequences were submitted to GenBank (accessions JN835188; KF692038; KF692039; KF692040).
