1. Structural Investigations of Phthalazinone Derivatives as Allosteric Inhibitors of Human DNA Methyltransferase 3A.

Abstract

The development of new therapeutics targeting enzymes involved in epigenetic pathways such as histone modification and DNA methylation has received a lot of attention, particularly for targeting diverse cancers. Unfortunately, irreversible nucleoside inhibitors (azacytidine and decitabine) have proven highly cytotoxic, and competitive inhibitors are also problematic. This work describes synthetic and structural investigations of a new class of allosteric DNA methyltransferase 3A (DNMT3A) inhibitors, leading to the identification of several critical pharmacophores in the lead structure. Specifically, we find that the tetrazole and phthalazinone moieties are indispensable for the inhibitory activity of DNMT3A and elucidate other modifiable regions in the lead compound.

3. EELS Studies of Cerium Electrolyte Reveal Substantial Solute Concentration Effects in Graphene Liquid Cells

Abstract

Graphene liquid cell transmission electron microscopy is a powerful technique to visualize nanoscale dynamics and transformations at atomic resolution. However, the solution in liquid cells is known to be affected by radiolysis, and the stochastic formation of graphene liquid cells raises questions about the solution chemistry in individual pockets. In this study, electron energy loss spectroscopy (EELS) was used to evaluate a model encapsulated solution, aqueous CeCl3. First, the ratio between the O K-edge and Ce M-edge was used to approximate the concentration of cerium salt in the graphene liquid cell. It was determined that the ratio between oxygen and cerium was orders of magnitude lower than what is expected for a dilute solution, indicating that the encapsulated solution is highly concentrated. To probe how this affects the chemistry within graphene liquid cells, the oxidation of Ce3+ was measured using time-resolved parallel EELS. It was determined that Ce3+ oxidizes faster under high electron fluxes, but reaches the same steady-state Ce4+ concentration regardless of flux. The time-resolved concentration profiles enabled direct comparison to radiolysis models, which indicate rate constants and g-values of certain molecular species are substantially different in the highly concentrated environment. Finally, electron flux-dependent gold nanocrystal etching trajectories showed that gold nanocrystals etch faster at higher electron fluxes, correlating well with the Ce3+ oxidation kinetics. Understanding the effects of the highly concentrated solution in graphene liquid cells will provide new insight on previous studies and may open up opportunities to systematically study systems in highly concentrated solutions at high resolution.

Abstract

A novel method for joining four components together in a single pot leading to an assortment of N-amino-benzylated phenols is described. The method involves the addition of different Grignard reagents to various o-OBoc salicylaldehydes in the presence of assorted 4,5-dihydrooxazoles, followed by aqueous workup. Seventeen examples are presented with varied (-R, -R -R″, -R‴, -R⁗, and Cn) substituents.

6. Concern about Experiencing Downward Socioeconomic Mobility Generates Precarious Types of Motivation among Students of Color

Abstract

Students' beliefs about whether they will experience changes in their socioeconomic status influence their academic motivation. We propose that students who are concerned about downward socioeconomic mobility will focus their attention on negative academic outcomes and exhibit motivational goals oriented towards preventing negative possibilities and that this relationship will be particularly pronounced among students of color. Two studies investigated the relationship between college students' concerns about downward socioeconomic mobility and their adoption of academic achievement goals. The more that students of color expressed concerns about experiencing downward socioeconomic mobility, the more they adopted academic mastery-avoidance goals ([beta] = 0.76), whereas there was no significant relationship between concerns about downward socioeconomic mobility and mastery-avoidance goals among White students ([beta] = -0.24; Study 1). Experimentally induced concerns about downward socioeconomic mobility increased academic mastery-avoidance goals among students of color ([beta] = -0.58) but decreased mastery-avoidance goals among White students ([beta] = 0.46; Study 2). Together, results indicate that there is a strong relationship between concerns about downward socioeconomic mobility and mastery-avoidance goals among students of color, highlighting the importance of understating how students of color make sense of their future socioeconomic prospects in order to most effectively support their academic trajectories positively.

Abstract

Spatial resolution of stimulated emission depletion (STED) microscopy varies with sample labeling techniques and microscope components, e.g., lasers, lenses, and photo-detectors. Fluctuations in the intensity of the depletion laser decrease achievable resolution in STED microscopy; the stronger the fluctuations, the higher the average intensity needed to achieve a given resolution. This phenomenon is encountered in every STED measurement. However, a theoretical framework that evaluates the effect of intensity fluctuations on spatial resolution is lacking. This article presents an analytical formulation based on a stochastic model that characterizes the impact of the laser fluctuations and correlation time on the depletion efficiency in the continuous wave (CW) STED microscopy. We compared analytical results with simulations using a wide range of intensity noise conditions and found a high degree of agreement. The stochastic model used considers a colored noise distribution for the laser intensity fluctuations. Simple analytical expressions were obtained in the limit of small and large fluctuations correlation time. These expressions fitted very well the available experimental data. Abstract

The size and shape of semiconductor nanocrystals govern their optical and electronic properties. Liquid cell transmission electron microscopy (LCTEM) is an emerging tool that can directly visualize nanoscale chemical transformations and therefore inform the precise synthesis of nanostructures with desired functions. However, it remains difficult to controllably investigate the reactions of semiconductor nanocrystals with LCTEM, because of the highly reactive environment formed by radiolysis of liquid. Here, we harness the radiolysis processes and report the single-particle etching trajectories of prototypical semiconductor nanomaterials with well-defined crystalline facets. Lead selenide nanocubes represent an isotropic structure that retains the cubic shape during etching via a layer-by-layer mechanism. The anisotropic arrow-shaped cadmium selenide nanorods have polar facets terminated by either cadmium or selenium atoms, and the transformation trajectory is driven by etching the selenium-terminated facets. LCTEM trajectories reveal how nanoscale shape transformations of semiconductors are governed by the reactivity of specific facets in liquid environments.

Abstract

Test and evaluation is a necessary process for ensuring that engineered systems perform as intended under a variety of conditions, both expected and unexpected. In this work, we consider the unique challenges of developing a unifying test and evaluation framework for complex ensembles of cyber-physical systems with embedded artificial intelligence. We propose a framework that incorporates test and evaluation throughout not only the development life cycle, but continues into operation as the system learns and adapts in a noisy, changing, and contended environment. The framework accounts for the challenges of testing the integration of diverse systems at various hierarchical scales of composition while respecting that testing time and resources are limited. A generic use case is provided for illustrative purposes and research directions emerging as a result of exploring the use case via the framework are suggested.

Abstract

Liquid cell electron microscopy enables the study of nanoscale transformations in solvents with high spatial and temporal resolution, but for the technique to achieve its potential requires a new level of control over the reactivity caused by radical generation under electron beam irradiation. An understanding of how to control electron-solvent interactions is needed to further advance the study of structural dynamics for complex materials at the nanoscale. We developed an approach that scavenges radicals with redox species that form well-defined redox couples and control the electrochemical potential in situ. This approach enables the observation of electrochemical structural dynamics at near-atomic resolution with precise control of the liquid environment. Analysis of nanocrystal etching trajectories indicates that this approach can be generalized to several chemical systems. The ability to simultaneously observe heterogeneous reactions at near-atomic resolution and precisely control the electrochemical potential enables the fundamental study of complex nanoscale dynamics with unprecedented detail.

Abstract

Graphene liquid cell transmission electron microscopy (TEM) has enabled the observation of a variety of nanoscale transformations. Yet understanding the chemistry of the liquid cell solution and its impact on the observed transformations remains an important step toward translating insights from liquid cell TEM to benchtop chemistry. Gold nanocrystal etching can be used as a model system to probe the reactivity of the solution. FeCl3 has been widely used to promote gold oxidation in bulk and liquid cell TEM studies, but the roles of the halide and iron species have not been fully elucidated. In this work, we observed the etching trajectories of gold nanocrystals in different iron halide solutions. We observed an increase in gold nanocrystal etch rate going from Cl-- to Br-- to I--containing solutions. This is consistent with a mechanism in which the dominant role of halides is as complexation agents for oxidized gold species. Additionally, the mechanism through which FeCl3 induces etching in liquid cell TEM remains unclear. Ground-state bleaching of the Fe(III) absorption band observed through pulse radiolysis indicates that iron may react with Cl2·- radicals to form an oxidized transient species under irradiation. Complete active space self-consistent field (CASSCF) calculations indicate that the FeCl3 complex is oxidized to an Fe species with an OH radical ligand. Together our data indicate that an oxidized Fe species may be the active oxidant, while halides modulate the etch rate by tuning the reduction potential of gold nanocrystals.

16. The Influence of Socioeconomic Status and Concerns about Experiencing Downward Socioeconomic Mobility on Education-Related Resource Hoarding

Abstract

While the proportion of students from lower socioeconomic status (SES) backgrounds enrolled in higher education continues to increase, rates of upward SES mobility among people from the lower end of the socioeconomic distribution remain extremely low and socioeconomic inequality in the United States continues to rise. The current manuscript examines how SES-based intergroup processes within the context of higher education may contribute to both stagnant socioeconomic mobility and ever-expanding socioeconomic inequality despite ostensive growing demographic diversity within higher education. Specifically, the present dissertation investigates the extent to which college students engage in education-related resource hoarding, particularly when they are concerned about experiencing downward socioeconomic mobility. Four studies examined the relationship between SES, concerns about downward socioeconomic mobility, and education-related resource hoarding among college students. I hypothesized that college students with higher SES would be more likely to engage in education-related resource hoarding than their lower SES counterparts, and that students with higher SES would be even more likely to engage in education-related resource hoarding when they were concerned about experiencing downward socioeconomic mobility than students with higher SES who were not concerned about downward mobility. Collectively, these four studies provide some evidence of education-related resource hoarding among students from both higher and lower SES groups. Education-related resource hoarding practiced by students within educational contexts can significantly limit the opportunities of other students, create additional obstacles for students to overcome within educational spaces, and ultimately contribute to extant societal inequality. 17. AutoDetect-mNP: An Unsupervised Machine Learning Algorithm for Automated Analysis of Transmission Electron Microscope Images of Metal Nanoparticles

Abstract

The synthesis quality of artificial inorganic nanocrystals is most often assessed by transmission electron microscopy (TEM) for which high-throughput advances have dramatically increased both the quantity and information richness of metal nanoparticle (mNP) characterization. Existing automated data analysis algorithms of TEM mNP images generally adopt a supervised approach, requiring a significant effort in human preparation of labeled data that reduces objectivity, efficiency, and generalizability. We have developed an unsupervised algorithm AutoDetect-mNP for automated analysis of TEM images that objectively extracts morphological information on convex mNPs from TEM images based on their shape attributes, requiring little to no human input in the process. The performance of AutoDetect-mNP is tested on two data sets of bright field TEM images of Au nanoparticles with different shapes and further extended to palladium nanocubes and cadmium selenide quantum dots, demonstrating that the algorithm is quantitatively reliable and can thus serve as a generalizable measure of the morphology distributions of any mNP synthesis. The AutoDetect-mNP algorithm will aid in future developments of high-throughput characterization of mNPs and the future advent of time-resolved TEM studies that can investigate reaction mechanisms of mNP synthesis and reactivity.

Abstract

Natural photosynthesis relies on a sophisticated charge transfer pathway among multiple components with precise spatial, energetic, and temporal organizations in the aqueous environment. It continues to inspire and challenge the design and fabrication of artificial multicomponent colloidal nanostructures for solar-to-fuel conversion. Herein, we introduce a plasmonic photocatalyst synthesized with colloidal methods with five integrated components including cocatalysts installed in orthogonal locations. The precise deposition of individual inorganic components on an Au/TiO2 nanodumbell nanostructure is enabled by photoreduction and photo-oxidation, which selectively occurs at the TiO2 tip sites and Au lateral sites, respectively. Under visible-light irradiation, the photocatalyst exhibited activity of oxygen evolution from water without scavengers. We demonstrate that each component is essential for improving the photocatalytic performance. In addition, mechanistic studies suggest that the photocatalytic reaction requires combining the hot charge carriers derived from exciting both the d-sp interband transition and the localized surface plasmon resonance of Au.

20. Exploring log-normal distributions in nascent entrepreneurship outcomes

Abstract

Recently, Joo, Aguinis and Bradley (2017), using a novel distribution pitting technique, have found that the exponential tail distributions-- exponential and power law with an exponential cut-off -- and their generative mechanism - namely, incremental differentiation -, are the most frequent distribution in many individual outputs across different organizations, sectors, jobs and activities. However, this may not be totally accurate in nascent entrepreneurship processes: the first section of this research shows that the lognormal distribution in entrepreneurial outcomes seems predominant throughout the different panels - i.e., longitudinal studies - in different countries. We have studied those in which the datasets are in the public domain: Australia, Sweden, US PSED I & II (Reynolds, 2017b). The power law distribution with an exponential cut-off may also be a plausible fit in some particular panel outcomes variables. A definitive conclusion regarding which of these two distributions may be the better fit will require the analysis of the rest of 14 still ongoing longitudinal projects around the world. The pervasiveness of lognormality offers relevant clues to understand nascent entrepreneurial processes, their generative mechanism, and it will offer strategies to allocate resources to foster and promote new entrepreneurial ventures. The second section of this research is the design and implementation of a baseline agent-based model as a research tool, "A nascent entrepreneurial agent-based model". Inspired by previous simpler entrepreneurial models, our model introduces new layers of complexity, making possible parametrization and calibration. This baseline model, initially with parameters similar to the public available panel datasets --Australia, Sweden, US PSED --, is able to generate the patterns that were found in the empirical results: the heavy-tailed distributions. Although PSED-type of longitudinal panels have been performed in more than a dozen countries, their results and datasets are not publicly available yet. This base model is, therefore, flexible in order to be easily adapted to each of the empirical dataset under study. The model, at this initial stage, has not been fully parametrized and calibrated for any specific country. The baseline model takes the main parameters from the datasets available as examples, in order to show that multiplicative processes --as main generative mechanism-- are able to simulate the empirical patterns. The baseline model is designed as a research tool to experiment and to help entrepreneurship researchers to test their theories, and for exploring in more detail the mechanisms involved in the emergence of new ventures. The baseline model and its background documentation will be openly available to the research community in two major agent-based repositories. Taking this baseline model as a "backbone", researchers can change parameters, agents, behaviours, schedules or global variables for their own theory building or calibration of their specific country's simulation.

28. Minds of Monsters: Scary Imbalances Between Cognition and Emotion.

Abstract

Four studies investigate a fear of imbalanced minds hypothesis that threatening agents perceived to be relatively mismatched in capacities for cognition (e.g., self-control and reasoning) and emotion (e.g., sensations and emotions) will be rated as scarier and more dangerous by observers. In ratings of fictional monsters (e.g., zombies and vampires), agents seen as more imbalanced between capacities for cognition and emotion (high cognition–low emotion or low cognition–high emotion) were rated as scarier compared to those with equally matched levels of cognition and emotion (Studies 1 and 2). Similar effects were observed using ratings of scary animals (e.g., tigers, sharks; Studies 2 and 3), and infected humans (Study 4). Moreover, these effects are explained through diminished perceived control/predictability over the target agent. These findings highlight the role of balance between cognition and emotion in appraisal of threatening agents, in part because those agents are seen as more chaotic and uncontrollable. Abstract

The ultimate goal of this work is to create an engineered aerosol that acts as one‐way smoke, i.e. it creates an asymmetric vision environment in which the ability to image objects depends on the viewing direction. To this end, a rapid, one‐pot synthesis of copper‐based microclubs is developed that consists of a Cu2O octahedron attached to a Cu2O@Cu shaft. Millions of synthesized particles are analyzed in minutes with a FlowCam to provide a robust statistical analysis of their geometry, and rapidly elucidate the roles of the reaction constituents on the particle shape and yield. The combination of asymmetry in both shape and composition introduces a 30% difference in scattering of light propagating parallel to the microclub axis from opposing directions. This work represents a first step toward the creation of an asymmetric imaging environment with an aerosol consisting of acoustically aligned microclubs. [ABSTRACT FROM AUTHOR]

Abstract

It is unknown if the current size and shape of protected areas (PA) in Costa Rica favors retention of viable populations of wildlife. The western border of Carara National Park (CNP) and its surroundings were characterized at a landscape level in order to consider their implications for biodiversity conservation. 54 400 hectares of Costa Rican Central Pacific tree cover were analyzed (1997, 2000, 2005) to calculate the size of the PAs, mean shape index, mean patch fractal dimension and edge density. Aside from that, we performed eight habitat evaluations on the western border of the CNP (costanera-sur highway) to assess tree composition, regeneration, litter, horizontal obstruction, and canopy cover. It was determined that the PAs were between 36 and 5 242 hectares long. We observed that the tree cover increased from 21 231,8 hectares in 1997 to 29 006,9 hectares in 2000, and decreased to 26 933,4 hectares in 2005. We found out that most of the tree cover (2005) belongs to CNP and ZP Cerros de Turrubares, but both of them present high values of edge density and mean shape index, suggesting that they are susceptible to alteration and fragmentation. The four evaluated points have a similar successional stage. In order to maintain the potential of CNP as an area for biodiversity conservation, we recommend reducing the maximum speed limit in the region adjacent to the park. It is also important to establish frequent monitoring of the tree cover and promote reforestation programs to create corridors that stop the area's fragmentation and facilitate biodiversity conservation., Comment: 6 pages, 1 figure, 4 tables

37. Design and control of active lower limb prosthesis with sensorial frame.

Abstract

In this article, we have designed an active prosthesis for the lower limb of a human. The unique design incorporates an exoskeletal extension that serves as a low-cost sensory framework, providing feedback on residual signals, namely the states of the hip and thigh. The controller takes advantage of system modeling and employs a simple proportional–derivative control. A stability analysis has been conducted. Abstract

Gender and ethnicity are increasingly studied topics within I-O psychology, helpful for understanding the composition of collectives, experiences of marginalized group members, and differences in outcomes between demographics and capturing diversity at higher levels. However, the absence of explicit, structured, demographic information online makes applying these research questions to Big Data sources challenging. We highlight how deep neural networks can be used to infer demographics based on people's names, which are commonly found online (e.g., social media profiles, employee pages, and membership rosters), using broad international data to train and evaluate the effectiveness of these models and find that validity coefficients meet minimum reliability thresholds at the individual level (r gender =.91, r ethnicity =.80) highlighting their ability to contextualize and facilitate Big Data research. Using empirical data extracted from databases, websites, and mobile apps, we highlight how these models can be applied to large organizational data sets by presenting illustrative demonstrations of research questions that incorporate the information provided by the model. 50. Analysis and interpretation of ambient data for CO2, CH4 and CO recorded in Southwest London during 2000 to 2012

Abstract

CO2 and CH4 current levels are the highest recorded during the Anthropocene epoch due, mostly, to emissions from anthropogenic activities. These emissions have modified the biochemical cycle of C and earth's thermal equilibrium. In contrast, CO emissions have decreased since 2000 following the strict controls on vehicle emissions. Atmospheric observations can be used to detect trends and changes in CO2, CH4 and CO concentrations, and also, to locate and characterize natural and anthropogenic sources. In situ measurements of atmospheric CO2, CH4 and CO have been performed at Royal Holloway University of London since 2000 to 2012 and standardised using NOAA calibration gases. CO2, CH4 and CO varied on time scales ranging from minutes to inter-annual and annual cycles. Diurnal cycles were observed and varied with the length daylight which influences the net soil uptake of CO2, the concentration of OH radicals and, the degree of vertical mixing. There is a greater influence of CO2 and CO anthropogenic emissions during weekdays when fossil fuel use and combustion processes are higher than at weekends not observed for CH4. CO2 showed an increasing trend of 2.45 ppm yr-1 data for the whole dataset; a higher rate of increase than the observed global trend due to higher regional combustion emissions such as airplanes, houses and cars on highly loaded motorways. By contrast, CO showed a progressive decline not linear, but if linearized the decline rate was 15.3 ppb CO yr-1, while CH4 concentration remained steady. The data set was split into 8 categories (45°) using wind sector analysis. This shows that the greatest concentrations for the three carbon gas are recorded from the NE, E and SE sectors. The lowest concentrations were observed for air from the S and SW sectors. Back trajectory and meteorological analysis of the data confirmed that the dominant sources of CO2, CH4 and CO are anthropogenic emissions from Greater London area and the dense local road network to the East of the measurement site. Nowadays, compared with Mace Head (West Ireland) data, the CO2 measured at Egham is slightly higher than at Mace Head while CO and CH4 is not far above Atlantic background levels during large periods of the year when prevailing S-SW winds reach the site. The Egham record implies that controls on CO emissions subsequent to legislation have been extremely successful the UK.
