Yüksek Yağlı Diyetle İndüklenmiş Obez Sıçanlarda Aposininin Testis Üzerine Etkisi: Histolojik ve Biyokimyasal Değerlendirmeİrem Hersek, Prof. Dr. Feriha ErcanHistoloji ve Embriyoloji Anabilim DalıAmaç: Bu çalışmada yüksek yağlı diyetle (YYD) indüklenmiş obezitenin sıçan testisi üzerine olumsuz etkilerini ve aposininin (APO) etkilerini histolojik ve biyokimyasal yöntemlerle göstermek amaçlandı. Gereç ve Yöntem: Çalışmada YYD Grubu (n=10) ve YYD+APO Grubuna (n=10) 4 ay boyunca yüksek yağlı yem verildi. YYD+APO Grubuna 3. aydan itibaren 4 hafta boyunca APO (25mg/kg) orogastrik gavaj ile haftada 5 gün uygulandı. STD grubundaki sıçanlar (n=8) standart yemle beslendi. Deney sonunda anestezi altında kan, testis ve epididimis örnekleri alındı ve histolojik ve biyokimyasal değerlendirmeler için hazırlandı. Elde edilen veriler istatistiksel yöntemler ile değerlendirildi. Bulgular ve Sonuç: YYD grubunda seminifer tübüllerde hasar, apoptik hücrelerde, NOX-2 pozitif hücrelerde ve hasarlı sperm sayısında artış, proliferatif hücre sayısında ve progresif sperm motilitesinde düşüş olduğu gözlendi. YYD grubunda, lipid ve hormon profillerinin değiştiği, MDA, MPO ve 8-OHDg aktivitesinin arttığı, GSH ve SOD düzeyinin ise düştüğü gözlendi. YYD+APO grubunda ise tüm bu histolojik ve biyokimyasal parametrelerin iyileştiği gözlendi. Sonuç: Yüksek yağlı diyet ile oluşturulan obezitenin sıçanlarda, testis morfolojisinde hasara, apoptotik ve NOX-2 pozitif hücre sayısında artışa ve sperm parametrelerinin bozulmasına sebep olduğu, NOX-2 inhibitörü olan APO’nun ise oksidatif mekanizmaları kısmen engelleyerek koruyucu etki yaptığı düşünülmektedir. --------------------Histological Evaluation of Effects of Apocynin on Testis in High Fat Diet-Induced Obese Rats Irem Hersek, Prof. Dr. Feriha ErcanDepartment of Histology and Embriology Aim: In this study, it was aimed to show the negative effects of high fat diet (HFD) induced obesity on rat testis and the effects of apocyninio (APO) by histological and biochemical methods. Materials and Methods: In the study, high fat feed was given to HFD Group (n = 10) and HFD + APO Group (n = 10) for 4 months. YYD + APO Group was administered with APO (25mg / kg) orogastirik gavage 5 days a week for 4 weeks starting from the 3rd month. STD group rats (n = 8) were fed with standard feed. At the end of the experiment, blood, testis and epididymis samples were taken under anesthesia and prepared for histological and biochemical evaluations. The data obtained were evaluated by statistical methods. Results and Conclusion: Damage in seminiferous tubules, increased number of apoptic cells, NOX-2 positive cells and damaged sperm, decreased number of proliferative cells and progressive sperm motility were observed in the HFD group. Lipid and hormone profiles were changed, MDA, MPO and 8-OHDg activity increased, GSH and SOD levels were decreased in the HFD group. In the YYD + APO group, all these histological and biochemical parameters improved. Conclusion: It is thought that caused by high fat diet obesity causes damage in testicular morphology, increase in apoptotic and NOX-2 positive cell numbers and deterioration of sperm parameters, and APO, which is a NOX-2 inhibitor, has a protective effect by partially inhibiting oxidative mechanisms.
