The objectives of this mixed-methods study were to gather survey and interview data about the barriers and facilitators from grantees funded by the National Institute on Disability, Independent Living, and Rehabilitation Research (NIDILRR) and to extract themes that could inform program changes that would increase technology translation (TT) success in assistive technology (AT). We developed a TT Barriers and Facilitators survey consisting of Likert scale and multiple-choice questions about barriers and facilitators to TT. With survey respondents who were willing, we conducting a semi-structured interview and asked pointed questions to expand upon survey response rankings and perceived barriers and facilitators. The questions were framed to explore the grantee's personal experience with ATTT and what helped and hindered their individualised processes. Across survey and interview respondents, the three most common themes when exploring the barriers and facilitators of TT were funding, incentives, and collaboration. Results indicate that there is a need for increased collaboration and access to additional resources such as funding for pilot grants, support to assess technology marketability, help to navigate regulatory and legal aspects, and assistance in establishing goals to help grantees successfully transfer assistive technologies to consumers. A large amount of research and development into assistive technology does not lead to tech transfer which means that these technologies are not getting to the people that need them. Educating tech transfer offices at universities about how to transfer AT would improve outcomes greatly. Creating a community of practice where grantees can find academic or industry partners would also increase the likelihood of tech transfer. The most massive stars provide an essential source of recycled material for young clusters and galaxies. While very massive stars (VMSs, M>100  |$\rm {\rm M}_{\odot }$|⁠) are relatively rare compared to O stars, they lose disproportionately large amounts of mass already from the onset of core H-burning. VMS have optically thick winds with elevated mass-loss rates in comparison to optically thin standard O-star winds. We compute wind yields and ejected masses on the main sequence, and we compare enhanced mass-loss rates to standard ones. We calculate solar metallicity wind yields from MESA stellar evolution models in the range 50–500  |$\rm {\rm M}_{\odot }$|⁠ , including a large nuclear network of 92 isotopes, investigating not only the CNO-cycle, but also the Ne–Na and Mg–Al cycles. VMS with enhanced winds eject 5–10 times more H-processed elements (N, Ne, Na, Al) on the main sequence in comparison to standard winds, with possible consequences for observed anticorrelations, such as C–N and Na–O, in globular clusters. We find that for VMS 95 per cent of the total wind yields is produced on the main sequence, while only ∼ 5 per cent is supplied by the post-main sequence. This implies that VMS with enhanced winds are the primary source of 26Al, contrasting previous works where classical Wolf–Rayet winds had been suggested to be responsible for galactic 26Al enrichment. Finally, 200  |$\rm {\rm M}_{\odot }$| stars eject 100 times more of each heavy element in their winds than 50  |$\rm {\rm M}_{\odot }$| stars, and even when weighted by an IMF their wind contribution is still an order of magnitude higher than that of 50  |$\rm {\rm M}_{\odot }$| stars. The ages of stars have historically relied on isochrone fitting of standardized grids of models. While these stellar models have provided key constraints on observational samples of massive stars, they inherit many systematic uncertainties, mainly in the internal mixing mechanisms applied throughout the grid, fundamentally undermining the isochrone method. In this work, we utilize the mass–lumiosity (M–L) plane of Higgins & Vink as a method of determining stellar age, with mixing-corrected models applying a calibrated core overshooting αov and rotation rate to fit the observational data. We provide multiple test-beds to showcase our new method, while also providing comparisons to the commonly used isochrone method, highlighting the dominant systematic errors. We reproduce the evolution of individual O stars, and analyse the wider sample of O and B supergiants from the VLT-FLAMES Tarantula Survey, providing dedicated models with estimates for αov, Ω/Ωcrit, and ultimately stellar ages. The M–L plane highlights a large discrepancy in the spectroscopic masses of the O supergiant sample. Furthermore the M–L plane also demonstrates that the evolutionary masses of the B supergiant sample are inappropriate. Finally, we utilize detached eclipsing binaries, VFTS 642 and VFTS 500, and present their ages resulting from their precise dynamical masses, offering an opportunity to constrain their interior mixing. For the near-TAMS system, VFTS 500, we find that both components require a large amount of core overshooting (αov ≃ 0.5), implying an extended main-sequence width. The most massive stars dominate the chemical enrichment, mechanical and radiative feedback, and energy budget of their host environments. Yet how massive stars initially form and how they evolve throughout their lives is ambiguous. The mass loss of the most massive stars remains a key unknown in stellar physics, with consequences for stellar feedback and populations. In this work, we compare grids of very massive star (VMS) models with masses ranging from 80 to 1000 M⊙, for a range of input physics. We include enhanced winds close to the Eddington limit as a comparison to standard O-star winds, with consequences for present-day observations of ∼50–100 M⊙ stars. We probe the relevant surface H abundances (X s) to determine the key traits of VMS evolution compared to O stars. We find fundamental differences in the behaviour of our models with the enhanced-wind prescription, with a convergence on the stellar mass at 1.6 Myr, regardless of the initial mass. It turns out that X s is an important tool in deciphering the initial mass due to the chemically homogeneous nature of VMS above a mass threshold. We use X s to break the degeneracy of the initial masses of both components of a detached binary, and a sample of WNh stars in the Tarantula Nebula. We find that for some objects, the initial masses are unrestricted and, as such, even initial masses of the order 1000 M⊙ are not excluded. At the end of its life, a very massive star is expected to collapse into a black hole (BH). The recent detection of an 85  M⊙ BH from the gravitational wave event GW 190521 appears to present a fundamental problem as to how such heavy BHs exist above the approximately 50  M⊙ pair-instability (PI) limit where stars are expected to be blown to pieces with no remnant left. Using mesa , we show that for stellar models with non-extreme assumptions, 90–100  M⊙ stars at reduced metallicity (⁠|$Z/\mbox{ $\mathrm{Z}_{\odot }$}\le 0.1$|⁠) can produce blue supergiant progenitors with core masses sufficiently small to remain below the fundamental PI limit, yet at the same time lose an amount of mass via stellar winds that is small enough to end up in the range of an 'impossible' 85  M⊙ BH. The two key points are the proper consideration of core overshooting and stellar wind physics with an improved scaling of mass-loss with iron (Fe) contents characteristic for the host galaxy metallicity. Our modelling provides a robust scenario that not only doubles the maximum BH mass set by PI, but also allows us to probe the maximum stellar BH mass as a function of metallicity and cosmic time in a physically sound framework. Summary: Ensuring faithful homologous recombination in allopolyploids is essential to maintain optimal fertility of the species. Variation in the ability to control aberrant pairing between homoeologous chromosomes in Brassica napus has been identified. The current study exploited the extremes of such variation to identify genetic factors that differentiate newly resynthesised B. napus, which is inherently unstable, and established B. napus, which has adapted to largely control homoeologous recombination.A segregating B. napus mapping population was analysed utilising both cytogenetic observations and high‐throughput genotyping to quantify the levels of homoeologous recombination.Three quantitative trait loci (QTL) were identified that contributed to the control of homoeologous recombination in the important oilseed crop B. napus. One major QTL on BnaA9 contributed between 32 and 58% of the observed variation. This study is the first to assess homoeologous recombination and map associated QTLs resulting from deviations in normal pairing in allotetraploid B. napus.The identified QTL regions suggest candidate meiotic genes that could be manipulated in order to control this important trait and further allow the development of molecular markers to utilise this trait to exploit homoeologous recombination in a crop. Context. Current massive star evolution grids are not able to simultaneously reproduce the empirical upper luminosity limit of red supergiants, the Humphrey-Davidson (HD) limit, nor the blue-to-red (B/R) supergiant ratio at high and low metallicity. Although previous studies have shown that the treatment of convection and semi-convection plays a role in the post-main-sequence (MS) evolution to blue or red supergiants (RSGs), a unified treatment for all metallicities has not been achieved so far. Aims. We focus on developing a better understanding of what drives massive star evolution to blue and red supergiant phases, with the ultimate aim of reproducing the HD limit at varied metallicities. We discuss the consequences of classifying B and R in the B/R ratio and clarify what is required to quantify a relatable theoretical B/R ratio for comparison with observations. Methods. For solar, Large Magellanic Cloud (50% solar), and Small Magellanic Cloud (20% solar) metallicities, we develop eight grids of MESA models for the mass range 20-60 M⊙ to probe the effect of semi-convection and overshooting on the core heliumburning phase. We compare rotating and non-rotating models with efficient (ffsemi = 100) and inefficient semi-convection (ffsemi = 0:1), with high and low amounts of core overshooting (ffov of 0.1 or 0.5). The red and blue supergiant evolutionary phases are investigated by comparing the fraction of core He-burning lifetimes spent in each phase for a range of masses and metallicities. Results. We find that the extension of the convective core by overshooting ffov = 0:5 has an effect on the post-MS evolution that can disable semi-convection, leading to more RSGs, but a lack of BSGs. We therefore implement ffov = 0:1, which switches on semi-convective mixing, but for standard ffsemi = 1 would result in an HD limit that is higher than observed at low Z (Large and Small Magellanic Clouds). Therefore, we need to implement very efficient semi-convection of ffsemi = 100, which reproduces the HD limit at log L=Lβ ~ 5:5 for the Magellanic Clouds while simultaneously reproducing the Galactic HD limit of log L=Lβ ~ 5:8 naturally. The effect of semi-convection is not active at high metallicities because the envelope structure is depleted by strong mass loss such that semi-convective regions could not form. Conclusions. Metallicity-dependent mass loss plays an indirect, yet decisive role in setting the HD limit as a function of Z. Traditionally, the pair instability (PI) mass gap is located between 50 and 130 M⊙, with stellar mass black holes (BHs) expected to 'pile up' towards the lower PI edge. However, this lower PI boundary is based on the assumption that the star has already lost its hydrogen (H) envelope. With the announcement of an 'impossibly' heavy BH of 85 M⊙ as part of GW 190521 located inside the traditional PI gap, we realized that blue supergiant (BSG) progenitors with small cores but large hydrogen envelopes at low metallicity (Z) could directly collapse to heavier BHs than had hitherto been assumed. The question of whether a single star can produce such a heavy BH is important, independent of gravitational wave events. Here, we systematically investigate the masses of stars inside the traditional PI gap by way of a grid of 336 detailed mesa stellar evolution models calculated across a wide parameter space, varying stellar mass, overshooting, rotation, semiconvection, and Z. We evolve low Z stars in the range 10−3 < Z /Z⊙ < Z SMC, making no prior assumption regarding the mass of an envelope, but instead employing a wind mass-loss recipe to calculate it. We compute critical carbon–oxygen and helium core masses to determine our lower limit to PI physics, and we provide two equations for M core and M final that can also be of use for binary population synthesis. Assuming the H envelope falls into the BH, we confirm the maximum BH mass below PI is M BH ≃ 93.3 M⊙. Our grid allows us to populate the traditional PI gap, and we conclude that the distribution of BHs above the traditional boundary is not solely due to the shape of the initial mass function, but also to the same stellar interior physics (i.e. mixing) that which sets the BH maximum. Research at the intersection of cognitive science and education has major benefits for both researchers and education practitioners. Yet, a number of logistical and communication challenges must be carefully negotiated to maximize the benefits of these collaborations. In this article, we trace the investments and major publications that have sought to bridge cognitive science and education over the past 40 years to provide context for the papers in a special issue of the Journal of Cognition and Development "Found in Translation: Cognitive Development Meets Education. We use the Institute of Education Sciences' investments in research at the intersection of cognitive science and education as a case study for what the field has primarily focused on over the past two decades and, finally, we offer a broad overview of the themes that this special issue touches on and a commentary on how we believe the field can improve. Context. Massive star evolution is dominated by various physical effects, including mass loss, overshooting, and rotation, but the prescriptions of their effects are poorly constrained and even affect our understanding of the main sequence. Aims. We aim to constrain massive star evolution models using the unique test-bed eclipsing binary HD 166734 with new grids of MESA stellar evolution models, adopting calibrated prescriptions of overshooting, mass loss, and rotation. Methods. We introduce a novel tool, called the mass-luminosity plane or M−L plane, as an equivalent to the traditional HR diagram, utilising it to reproduce the test-bed binary HD 166734 with newly calibrated MESA stellar evolution models for single stars. Results. We can only reproduce the Galactic binary system with an enhanced amount of core overshooting (αov = 0.5), mass loss, and rotational mixing. We can utilise the gradient in the M−L plane to constrain the amount of mass loss to 0.5–1.5 times the standard prescription test-bed, and we can exclude extreme reduction or multiplication factors. The extent of the vectors in the M−L plane leads us to conclude that the amount of core overshooting is larger than is normally adopted in contemporary massive star evolution models. We furthermore conclude that rotational mixing is mandatory to obtain the correct nitrogen abundance ratios between the primary and secondary components (3:1) in our test-bed binary system. Conclusions. Our calibrated grid of models, alongside our new M−L plane approach, present the possibility of a widened main sequence due to an increased demand for core overshooting. The increased amount of core overshooting is not only needed to explain the extended main sequence, but the enhanced overshooting is also needed to explain the location of the upper-luminosity limit of the red supergiants. The ketone body ester (R)-3-hydroxybutyryl-(R)-3-hydroxybutyrate and its (S,S) enantiomer were prepared in a short, operationally simple synthetic sequence from racemic β-butyrolactone. Enantioselective hydrolysis of β-butyrolactone with immobilized Candida antarctica lipase-B (CAL-B) results in (R)-β-butyrolactone and (S)-β-hydroxybutyric acid, which are easily converted to (R) or (S)-ethyl-3-hydroxybutyrate and reduced to (R) or (S)-1,3 butanediol. Either enantiomer of ethyl-3-hydroxybutyrate and 1,3 butanediol are then coupled, again using CAL-B, to produce the ketone body ester product. The heavy selection pressure due to intensive breeding of Brassica napus has created a narrow gene pool, limiting the ability to produce improved varieties through crosses between B. napus cultivars. One mechanism that has contributed to the adaptation of important agronomic traits in the allotetraploid B. napus has been chromosomal rearrangements resulting from homoeologous recombination between the constituent A and C diploid genomes. Determining the rate and distribution of such events in natural B. napus will assist efforts to understand and potentially manipulate this phenomenon. The Brassica high-density 60K SNP array, which provides genome-wide coverage for assessment of recombination events, was used to assay 254 individuals derived from 11 diverse cultivated spring type B. napus. These analyses identified reciprocal allele gain and loss between the A and C genomes and allowed visualization of de novo homoeologous recombination events across the B. napus genome. The events ranged from loss/gain of 0.09 Mb to entire chromosomes, with almost 5% aneuploidy observed across all gametes. There was a bias toward sub-telomeric exchanges leading to genome homogenization at chromosome termini. The A genome replaced the C genome in 66% of events, and also featured more dominantly in gain of whole chromosomes. These analyses indicate de novo homoeologous recombination is a continuous source of variation in established Brassica napus and the rate of observed events appears to vary with genetic background. The Brassica napus 60K Illumina Infinium™ SNP array has had huge international uptake in the rapeseed community due to the revolutionary speed of acquisition and ease of analysis of this high-throughput genotyping data, particularly when coupled with the newly available reference genome sequence. However, further utilization of this valuable resource can be optimized by better understanding the promises and pitfalls of SNP arrays. We outline how best to analyze Brassica SNP marker array data for diverse applications, including linkage and association mapping, genetic diversity and genomic introgression studies. We present data on which SNPs are locus-specific in winter, semi-winter and spring B. napus germplasm pools, rather than amplifying both an A-genome and a C-genome locus or multiple loci. Common issues that arise when analyzing array data will be discussed, particularly those unique to SNP markers and how to deal with these for practical applications in Brassica breeding applications. Key message : The Brassica napus Illumina array provides genome-wide markers linked to the available genome sequence, a significant tool for genetic analyses of the allotetraploid B. napus and its progenitor diploid genomes. Abstract: A high-density single nucleotide polymorphism (SNP) Illumina Infinium array, containing 52,157 markers, was developed for the allotetraploid Brassica napus. A stringent selection process employing the short probe sequence for each SNP assay was used to limit the majority of the selected markers to those represented a minimum number of times across the highly replicated genome. As a result approximately 60 % of the SNP assays display genome-specificity, resolving as three clearly separated clusters (AA, AB, and BB) when tested with a diverse range of B. napus material. This genome specificity was supported by the analysis of the diploid ancestors of B. napus, whereby 26,504 and 29,720 markers were scorable in B. oleracea and B. rapa, respectively. Forty-four percent of the assayed loci on the array were genetically mapped in a single doubled-haploid B. napus population allowing alignment of their physical and genetic coordinates. Although strong conservation of the two positions was shown, at least 3 % of the loci were genetically mapped to a homoeologous position compared to their presumed physical position in the respective genome, underlying the importance of genetic corroboration of locus identity. In addition, the alignments identified multiple rearrangements between the diploid and tetraploid Brassica genomes. Although mostly attributed to genome assembly errors, some are likely evidence of rearrangements that occurred since the hybridisation of the progenitor genomes in the B. napus nucleus. Based on estimates for linkage disequilibrium decay, the array is a valuable tool for genetic fine mapping and genome-wide association studies in B. napus and its progenitor genomes. Turnip mosaic virus (TuMV) is the major virus infecting crops of the genus Brassica worldwide. A dominant resistance gene, TuRB01b, that confers immunity to the virus isolate UK 1 (a representative pathotype 1 isolate of TuMV) on Brassica rapa was identified in the Chinese cabbage cultivar Tropical Delight. The TuRB01b locus was mapped to a 2.9-cM interval on B. rapa chromosome 6 (A6) that was flanked by RFLP markers pN101e1 and pW137e1. This mapping used a first backcross (B1) population segregating for the resistance gene at TuRB01b and sets of RFLP markers employed in previous mapping experiments in Brassica. Virus-plant interaction phenotypes were assayed in inbred progeny derived from B1 individuals to allow different virus isolates to be tested. Comparative mapping confirmed that A6 of B. rapa was equivalent to chromosome 6 of Brassica napus (A6) and that the map position of TuRB01b in B. rapa could be identical to that of TuRB01 in B. napus. Detailed evaluation of plant-virus interactions showed that TuRB01 and TuRB01b had indistinguishable specificities to a range of TuMV isolates. The possibility that TuRB01 and TuRB01b represent similar or identical alleles at the same A genome resistance locus suggests that B. napus acquired TuRB01 from the B. rapa gene pool. Classical Wolf-Rayet (WR) stars mark an important stage in the late evolution of massive stars. As hydrogen-poor massive stars, these objects have lost their outer layers, while still losing further mass through strong winds indicated by their prominent emission line spectra. Wolf-Rayet stars have been detected in a variety of different galaxies. Their strong winds are a major ingredient of stellar evolution and population synthesis models. Yet, a coherent theoretical picture of their strong mass-loss is only starting to emerge. In particular, the occurrence of WR stars as a function of metallicity (Z) is still far from being understood. To uncover the nature of the complex and dense winds of Wolf-Rayet stars, we employ a new generation of model atmospheres including a consistent solution of the wind hydrodynamics in an expanding non-LTE situation. With this technique, we can dissect the ingredients driving the wind and predict the resulting mass-loss for hydrogen-depleted massive stars. Our modelling efforts reveal a complex picture with strong, non-linear dependencies on the luminosity-to-mass ratio and Z with a steep, but not totally abrupt onset for WR-type winds in helium stars. With our findings, we provide a theoretical motivation for a population of helium stars at low Z, which cannot be detected via WR-type spectral features. Our study of massive He-star atmosphere models yields the very first mass-loss recipe derived from first principles in this regime. Implementing our first findings in stellar evolution models, we demonstrate how traditional approaches tend to overpredict WR-type mass loss in the young Universe. [ABSTRACT FROM AUTHOR]

Although the biomechanical properties of the various types of running foot strike (rearfoot, midfoot, and forefoot) have been studied extensively in the laboratory, only a few studies have attempted to quantify the frequency of running foot strike variants among runners in competitive road races. We classified the left and right foot strike patterns of 936 distance runners, most of whom would be considered of recreational or sub-elite ability, at the 10 km point of a half-marathon/marathon road race. We classified 88.9% of runners at the 10 km point as rearfoot strikers, 3.4% as midfoot strikers, 1.8% as forefoot strikers, and 5.9% of runners exhibited discrete foot strike asymmetry. Rearfoot striking was more common among our sample of mostly recreational distance runners than has been previously reported for samples of faster runners. We also compared foot strike patterns of 286 individual marathon runners between the 10 km and 32 km race locations and observed increased frequency of rearfoot striking at 32 km. A large percentage of runners switched from midfoot and forefoot foot strikes at 10 km to rearfoot strikes at 32 km. The frequency of discrete foot strike asymmetry declined from the 10 km to the 32 km location. Abstract: Inositol 1,4,5-trisphosphate receptor channels control release of from the endoplasmic reticulum into the cytosol of a cell. The binding of both 1,4,5-trisphosphate and activating is required for the channel to open. At high concentrations, s are inhibited. s are composed of four identical subunits and form in clusters. Many models have been proposed to describe how the binding of and to subunits results in the opening and closing of s. Here we compare the opening and closing probability distributions for clusters of s, resulting from three different models. The distributions are calculated both analytically, using a method we have developed, and with simulations. Abstract: Microdomains of calcium (i.e., areas on the nanometer scale that have qualitatively different calcium concentrations from that in the bulk cytosol) are known to be important in many situations. In cardiac cells, for instance, a calcium microdomain between the L-type channels and the ryanodine receptors, the so-called diadic cleft, is where the majority of the control of calcium release occurs. In other cell types that exhibit calcium oscillations and waves, the importance of microdomains in the vicinity of clusters of inositol trisphosphate receptors, or between the endoplasmic reticulum (ER) and other internal organelles or the plasma membrane, is clear. Given the limits of computational power, it is not currently realistic to model an entire cellular cytoplasm by incorporating detailed structural information about the ER throughout the entire cytoplasm. Hence, most models use a homogenised approach, assuming that both cytoplasm and ER coexist at each point of the domain. Conversely, microdomain models can be constructed, in which detailed structural information can be incorporated, but, until now, methods have not been developed for linking such a microdomain model to a model at the level of the entire cell. Using the homogenisation approach we developed in an earlier paper [Goel, P., Friedman, A., Sneyd, J., 2006. Homogenization of the cell cytoplasm: the calcium bidomain equations. SIAM J. Multiscale Modeling Simulation, in press] we show how a multiscale model of a calcium microdomain can be constructed. In this model a detailed model of the microdomain (in which the ER and the cytoplasm are separate compartments) is coupled to a homogenised model of the entire cell in a rigorous way. [Display omitted] Quorum sensing is a bacterial signaling system that involves the synthesis, secretion and detection of signal molecules called autoinducers. The main autoinducer in Gram-negative bacteria are acylated homoserine lactones, produced by the LuxI family of autoinducer synthases and detected by the LuxR family of autoinducer receptors. Quorum sensing allows for changes in gene expression and bacterial behaviors in a coordinated, cell density dependent manner. Quorum sensing controls the expression of virulence factors in some human pathogens, making quorum sensing an antibacterial drug target. Very massive stars (VMS) dominate the physics of young clusters due to their ionizing radiation and extreme stellar winds. It is these winds that determine their lifepaths until expiration. Observations in the Arches Cluster show that VMS all have similar temperatures. The VLT-FLAMES Tarantula Survey analysed VMS in the 30 Doradus (30 Dor) region of the Large Magellanic Cloud (LMC) also finding a narrow range of temperatures, albeit at higher values – likely a metallicity effect. Using mesa , we study the main-sequence evolution of VMS with a new mass-loss recipe that switches from optically thin O-star winds to optically thick Wolf–Rayet-type winds through the model-independent transition mass-loss rate of Vink & Gräfener. We examine the temperature evolution of VMS with mass loss that scales with the luminosity-over-mass (L / M) ratio and the Eddington parameter (Γe), assessing the relevance of the surface hydrogen (H) abundance that sets the number of free electrons. We present grids of VMS models at Galactic and LMC metallicity and compare our temperature predictions with empirical results. Models with a steep Γe dependence evolve horizontally in the Hertzsprung–Russel (HR) diagram at nearly constant luminosities, requiring a delicate and unlikely balance between envelope inflation and enhanced mass loss over the entire VMS mass range. By contrast, models with a steep L / M -dependent mass loss are shown to evolve vertically in the HR diagram at nearly constant T eff, naturally reproducing the narrow range of observed temperatures, as well as the correct trend with metallicity. Atherosclerosis is a chronic cardiovascular disease which increases risk of major cardiovascular events including myocardial infarction and stroke. Elevated plasma concentrations of asymmetric dimethylarginine (ADMA) have long been recognised as a hallmark of cardiovascular disease and are associated with cardiovascular risk factors including hypertension, obesity and hypertriglyceridemia. In this review, we discuss the clinical literature that link ADMA concentrations to increased risk of the development of atherosclerosis. The formation of atherosclerotic lesions relies on the interplay between vascular dysfunction, leading to endothelial activation and the accumulation of inflammatory cells, particularly macrophages, within the vessel wall. Here, we review the mechanisms through which elevated ADMA contributes to endothelial dysfunction, activation and reactive oxygen species (ROS) production; how ADMA may affect vascular smooth muscle phenotype; and finally whether ADMA plays a regulatory role in the inflammatory processes occurring within the vessel wall. The Humphreys–Davidson (HD) limit sets the boundary between evolutionary channels of massive stars that end their lives either as the red supergiants (RSGs) or as the hotter blue supergiants (BSGs) and Wolf–Rayet stars. Mixing in the envelopes of massive stars close to their Eddington limit is crucial for investigating the upper luminosity limit of the coolest supergiants. We study the effects of excess mixing in superadiabatic layers that are dominated by radiation pressure, and we critically investigate the effects of mixing and mass-loss on the evolution of RSGs with log (T eff/K) < 3.68 – as a function of metallicity. Using MESA, we produce grids of massive star models at three metallicities: Galactic (Z⊙), LMC |$(\frac{1}{2}{\rm Z}_\odot)$|⁠ , and SMC |$(\frac{1}{5}{\rm Z}_\odot)$|⁠ , with both high and low amounts of overshooting to study the upper luminosity limit of RSGs. We systematically study the effects of excess mixing in the superadiabatic layers of post-main-sequence massive stars, overshooting above the hydrogen core and yellow supergiant (YSG) mass-loss rates on the fraction of core helium burning time spent as a RSG. We find that the excess mixing in the superadiabatic layers is stronger at lower metallicities, as it depends on the opacities in the hydrogen bump at log (T eff/K) ≈ 4, which become more pronounced at lower metallicity. This shifts the cut-off luminosities to lower values at lower metallicities, thus balancing the first-order effect of mass-loss. The opposing effects of mass-loss and excess envelope mixing during post-main-sequence evolution of stars with higher overshooting potentially results in a metallicity-independent upper luminosity limit. Very massive stars (VMS) up to 200–300 M⊙ have been found in the Local Universe. If they would lose little mass, they produce intermediate-mass black holes or pair-instability supernovae (PISNe). Until now, VMS modellers have extrapolated mass-loss versus metallicity (Z) exponents from optically thin winds, resulting in a range of PISN thresholds that might be unrealistically high in Z , as VMS develop optically thick winds. We utilize the transition mass-loss rate of Vink and Gräfener (2012) that accurately predicts mass-loss rates of Of/WNh ('slash') stars that characterize the morphological transition from absorption-dominated O-type spectra to emission-dominated WNh spectra. We develop a wind efficiency framework, where optically thin winds transition to enhanced winds, enabling us to study VMS evolution at high redshift where individual stars cannot be resolved. We present a MESA grid covering Z ⊙/2 to Z ⊙/100. VMS above the transition evolve towards lower luminosity, skipping the cool supergiant phase but directly forming pure He stars at the end of hydrogen burning. Below the transition, VMS evolve as cooler luminous blue variables (LBVs) or yellow hypergiants (YHGs), naturally approaching the Eddington limit. Strong winds in this YHG/LBV regime – combined with a degeneracy in luminosity – result in a mass-loss runaway, where a decrease in mass increases wind mass loss. Our models indicate an order-of-magnitude lower metallicity threshold for PISN than usually assumed, at Z ⊙/20 due to our mass-loss runaway. While future work on LBV mass loss could affect the PISN threshold, our framework will be critical for establishing definitive answers on the PISN threshold and galactic chemical evolution modelling. Asymmetric dimethylarginine (ADMA) is generated through the irreversible methylation of arginine residues. It is an independent risk factor for cardiovascular disease, currently thought to be due to its ability to act as a competitive inhibitor of the nitric oxide (NO) synthase enzymes. Plasma ADMA concentrations increase with obesity and fall following weight loss; however, it is unknown whether they play an active role in adipose pathology. Here, we demonstrate that ADMA drives lipid accumulation through a newly identified NO-independent pathway via the amino-acid sensitive calcium-sensing receptor (CaSR). ADMA treatment of 3T3-L1 and HepG2 cells upregulates a suite of lipogenic genes with an associated increase in triglyceride content. Pharmacological activation of CaSR mimics ADMA while negative modulation of CaSR inhibits ADMA driven lipid accumulation. Further investigation using CaSR overexpressing HEK293 cells demonstrated that ADMA potentiates CaSR signalling via G q intracellular Ca2+ mobilisation. Summary: Camelina neglecta is a diploid species from the genus Camelina, which includes the versatile oilseed Camelina sativa. These species are closely related to Arabidopsis thaliana and the economically important Brassica crop species, making this genus a useful platform to dissect traits of agronomic importance while providing a tool to study the evolution of polyploids. A highly contiguous chromosome‐level genome sequence of C. neglecta with an N50 size of 29.1 Mb was generated utilizing Pacific Biosciences (PacBio, Menlo Park, CA) long‐read sequencing followed by chromosome conformation phasing. Comparison of the genome with that of C. sativa shows remarkable coincidence with subgenome 1 of the hexaploid, with only one major chromosomal rearrangement separating the two. Synonymous substitution rate analysis of the predicted 34 061 genes suggested subgenome 1 of C. sativa directly descended from C. neglecta around 1.2 mya. Higher functional divergence of genes in the hexaploid as evidenced by the greater number of unique orthogroups, and differential composition of resistant gene analogs, might suggest an immediate adaptation strategy after genome merger. The absence of genome bias in gene fractionation among the subgenomes of C. sativa in comparison with C. neglecta, and the complete lack of fractionation of meiosis‐specific genes attests to the neopolyploid status of C. sativa. The assembled genome will provide a tool to further study genome evolution processes in the Camelina genus and potentially allow for the identification and exploitation of novel variation for Camelina crop improvement. Camelina sativa, a largely relict crop, has recently returned to interest due to its potential as an industrial oilseed. Molecular markers are key tools that will allow C. sativa to benefit from modern breeding approaches. Two complementary methodologies, capture of 3′ cDNA tags and genomic reduced-representation libraries, both of which exploited second generation sequencing platforms, were used to develop a low density (768) Illumina GoldenGate single nucleotide polymorphism (SNP) array. The array allowed 533 SNP loci to be genetically mapped in a recombinant inbred population of C. sativa. Alignment of the SNP loci to the C. sativa genome identified the underlying sequenced regions that would delimit potential candidate genes in any mapping project. In addition, the SNP array was used to assess genetic variation among a collection of 175 accessions of C. sativa, identifying two sub-populations, yet low overall gene diversity. Purpose: This study assesses the rate of enhancement of breast fibroglandular tissue after administration of a magnetic resonance imaging (MRI) gadolinium-based contrast agent and determines its relationship with response to neoadjuvant therapy (NAT) in women with breast cancer.Method: Women with locally advanced breast cancer (N = 19) were imaged four times over the course of NAT. Dynamic contrast-enhanced (DCE) MRI was acquired after administration of a gadolinium-based contrast agent with a temporal resolution of 7.27 s. The tumor, fibroglandular tissue, and adipose tissue were semi-automatically segmented using a manually drawn region of interest encompassing the tumor followed by fuzzy c-means clustering. The rate and relative intensity of signal enhancement were calculated for each voxel within the tumor and fibroglandular tissue.Results: The rate of fibroglandular tissue enhancement after contrast agent injection declined by an average of 29 % over the course of NAT. This decline was present in 16 of the 19 patients in the study. The rate of enhancement is significantly higher in women who achieve pathological complete response (pCR) after both 1 cycle (68 % higher, p < 0.05) and after 3-5 cycles of NAT (58 % higher; p < 0.05). The relative intensity of fibroglandular enhancement correlates with the rate of enhancement (R2 = 0.64, p < 0.001) and is higher in women who achieve pCR after both 1 cycle and after 3-5 cycles of NAT (p < 0.05, both timepoints).Conclusion: The rate of fibroglandular tissue enhancement declines over the course of therapy, provides novel information not reflected by tumoral measures, and may predict pathological response early in the course of therapy, with smaller declines in enhancement in women who achieve favorable response. Abstract: Background: Merkel cell polyomavirus (MCV or MCPyV) is a recently discovered human polyomavirus that is implicated in the pathogenesis of Merkel cell carcinoma (MCC). Although the transmission route for MCV is not yet known, other polyomaviruses, such as BKV, cause non-malignant pathology in the urinary tract. Like MCC, prostate cancer predominantly affects the elderly. Furthermore, prostate cancers and premalignant precursors exhibit chronic inflammation, which suggests a possible infectious involvement. We therefore examined whether MCV might participate in the pathogenesis of prostate cancer. Objective: To determine the presence of MCV RNA in prostate cancer and surrounding stroma or normal prostate tissue. Study design: RNA was extracted from 28 patient-matched cancerous and 28 benign prostate epithelial samples, and six additional cancer-adjacent stromal samples. All tissues were laser-capture micro-dissected. DNA and RNA from a sequence-verified MCV-containing MCC tumor served as a positive control. Quantitative reverse-transcription PCR was used to assess the presence or absence of MCV T antigen transcript. Results: No MCV T antigen was detected in prostate carcinomas, patient-matched benign samples, or tumor-adjacent stroma, with appropriate sensitivity of the assay demonstrated by an MCC tumor. Conclusions: MCV infection appears unlikely to be a significant factor in prostate carcinogenesis and there is no evidence of the prostate serving as a reservoir for MCV. [Copyright &y& Elsevier]

Ethiopian mustard (Brassica carinata A. Braun) is an emerging sustainable source of vegetable oil, in particular for the biofuel industry. The present study exploited genome assemblies of the Brassica diploids, Brassica nigra and Brassica oleracea, to discover over 10,000 genome-wide SNPs using genotype by sequencing of 620 B. carinata lines. The analyses revealed a SNP frequency of one every 91.7 kb, a heterozygosity level of 0.30, nucleotide diversity levels of 1.31 × 10−05, and the first five principal components captured only 13% molecular variation, indicating low levels of genetic diversity among the B. carinata collection. Genome bias was observed, with greater SNP density found on the B subgenome. The 620 lines clustered into two distinct sub-populations (SP1 and SP2) with the majority of accessions (88%) clustered in SP1 with those from Ethiopia, the presumed centre of origin. SP2 was distinguished by a collection of breeding lines, implicating targeted selection in creating population structure. Two selective sweep regions on B3 and B8 were detected, which harbour genes involved in fatty acid and aliphatic glucosinolate biosynthesis, respectively. Brassica napus seed composition traits (fibre, protein, oil and fatty acid profiles), seed colour and yield-associated traits are regulated by a complex network of genetic factors. Although previous studies have attempted to dissect the underlying genetic basis for these traits, a more complete picture of the available quantitative trait loci (QTL) variation and any interaction between the different traits is required. In this study, QTL mapping for eleven seed composition traits, seed colour and a yield-related trait (TSW) was conducted in a spring-type canola-quality B. napus doubled haploid (DH) population from a cross between black-seeded (DH12075) and yellow-seeded (YN01-429) lines across five environments. A major QTL associated with fibre traits (acid detergent fibre, acid detergent lignin and neutral detergent fibre) and seed colour (whiteness index) was mapped on chromosome N9 across the five environments. Multi-trait analysis identified QTL which had pleiotropic effect for seed colour and other composition traits. Multi-environment analysis revealed genetic (QTL) × environment effects on most QTL. Single nucleotide polymorphisms (SNPs) have rapidly become the molecular marker of choice in plant and animal association mapping (AM) studies. In this work, a genome-wide association study (GWAS) and candidate quantitative trait loci (cQTL) approaches were used to identify SNP markers associated with seed quality traits, in a Brassica napus L. association panel composed of 89 adapted winter oilseed rape accessions. Six seed quality traits (oil and protein content, linolenic acid, total glucosinolates, hemicellulose and cellulose content) were evaluated in two different locations for two seasons. For GWAS, 4025 SNP markers evenly distributed along the B. napus genome were genotyped using a 6K Illumina array platform. For cQTL, 100 SNP markers previously discovered in genomic regions underlying seed quality QTL were genotyped using a competitive allele-specific PCR (KASPar). Analysis of the population structure revealed the presence of two weakly differentiated subpopulations (FST = 0.037), with 82 % of the pairwise kinship comparisons ranging from 0 to 0.1. The GWAS approach resulted in the identification of 17 and 5 significant associations for seed glucosinolate content and seed hemicellulose content, respectively. The cQTL approach identified 4 significant associations for seed glucosinolate content and 6 significant associations for seed hemicellulose content. The associated SNPs were consistently identified across environments and were mapped to previously reported QTL. The objectives of our study were to (i) examine the expression levels of candidate genes in seedling leaves of B. napus and correlate these with seedling development as well as (ii) detect genome regions associated with gene expression levels and seedling development traits in B. napus by genome-wide association mapping. Results: The expression levels of the 15 candidate genes examined in the 509 B. napus inbreds showed an averaged standard deviation of 5.6 across all inbreds and ranged from 3.2 to 8.8. The gene expression differences between the 509 B. napus inbreds were more than adequate for the correlation with phenotypic variation of seedling development. The average of the absolute value correlations of the correlation coefficients of 0.11 were observed with a range from 0.00 to 0.39. The candidate genes GER1, AILP1, PECT, and FBP were strongly correlated with the seedling development traits. In a genome-wide association study, we detected a total of 63 associations between single nucleotide polymorphisms (SNPs) and the seedling development traits and 31 SNP-gene associations for the candidate genes with a P-value < 0.0001. For the projected leaf area traits we identified five different association hot spots on the chromosomes A2, A7, C3, C6, and C7. Conclusion: A total of 99.4% of the adjacent SNPs on the A genome and 93.0% of the adjacent SNPs on the C genome had a distance smaller than the average range of linkage disequilibrium. Therefore, this genome-wide association study is expected to result on average in 14.7% of the possible power. Compared to previous studies in B. napus, the SNP marker density of our study is expected to provide a higher power to detect SNP-trait/-gene associations in the B. napus diversity set. Information on genetic diversity and population structure is needed for continued improvement in Brassica napus L. A total of 169 B. napus lines, collected between 1970 and 2000 in Australia, Canada, China, Europe, Japan, Pakistan, and South Korea, were genotyped with 84 simple sequence repeat markers. Nei's unbiased genetic diversity (H) and Shannon's information index (SI) showed that genetic diversity was highest among lines from Europe followed by South Korea, Japan, China, and Pakistan while lines from Australia and Canada had the lowest diversity. Pairwise comparison of the populations using Nei's genetic identity (I) and genetic distance (D) showed greater differentiation between Australia and Canada versus China and Pakistan. Accessions from South Korea and Japan were most similar, confirming an historical exchange of germplasm. The analysis of unique alleles and allele richness revealed that the Pakistani population was the richest followed by Europe and South Korea. Analysis of molecular variance revealed that 9% of the variation was accounted for by geographical region and 91% by lines. Both Structure and principal coordinate analyses indicated two subpopulations in the collection confirming separation of genotypes due to growth habit adaptation. Information on B. napus genotypes containing unique alleles is valuable for breeding programs in different parts of the world seeking to improve agronomic traits and adaptability. Multiple human epidemiologic studies link caffeinated (but not decaffeinated) beverage intake with significant decreases in several types of cancer, including highly prevalent UV-associated skin carcinomas. The mechanism by which caffeine protects against skin cancer is unknown. Ataxia telangiectasia and Rad3-related (ATR) is a replication checkpoint kinase activated by DNA stresses and is one of several targets of caffeine. Suppression of ATR, or its downstream target checkpoint kinase 1 (Chkl1, selectively sensitizes DNA-damaged and malignant cells to apoptosis. Agents that target this pathway are currently in clinical trials. Conversely, inhibition of other DNA damage response pathways, such as ataxia telangiectasia mutated (ATM) and BRCA1, promotes cancer. To determine the effect of replication checkpoint inhibition on carcinogenesis, we generated transgenic mice with diminished ATR function in skin and crossed them into a UV-sensitive background, Xpc-/-. Unlike caffeine, this genetic approach was selective and had no effect on ATM activation. These transgenic mice were viable and showed no histological abnormalities in skin. Primary keratinocytes from these mice had diminished UV-induced Chkl phosphorylation and twofold augmentation of apoptosis after UV exposure (P = 0.006). With chronic UV treatment, transgenic mice remained tumor-free for significantly longer (P = 0.003) and had 69% fewer tumors at the end of observation of the full cohort (P = 0.019), compared with littermate controls with the same genetic background. This study suggests that inhibition of replication checkpoint function can suppress skin carcinogenesis and supports ATR inhibition as the relevant mechanism for the protective effect of caffeinated beverage intake in human epidemiologic studies. The architecture of the Brassica napus genome is marked by its evolutionary origins. The genome of B. napus was formed from the hybridization of two closely related diploid Brassica species, both of which evolved from an hexaploid ancestor. The extensive whole genome duplication events in its near and distant past result in the allotetraploid genome of B. napus maintaining multiple copies of most genes, which predicts a highly complex and redundant transcriptome that can confound any expression analyses. A stringent assembly of 142 399 B. napus expressed sequence tags allowed the development of a well-differentiated set of reference transcripts, which were used as a foundation to assess the efficacy of available tools for identifying and distinguishing transcripts in B. napus; including microarray hybridization and 3′ anchored sequence tag capture. Microarray platforms cannot distinguish transcripts derived from the two progenitors or close homologues, although observed differential expression appeared to be biased towards unique transcripts. The use of 3′ capture enhanced the ability to unambiguously identify homologues within the B. napus transcriptome but was limited by tag length. The ability to comprehensively catalogue gene expression in polyploid species could be transformed by the application of cost-efficient next generation sequencing technologies that will capture millions of long sequence tags. Targeted genomic selection methodologies, or sequence capture, allow for DNA enrichment and large-scale resequencing and characterization of natural genetic variation in species with complex genomes, such as rapeseed canola (Brassica napus L., AACC, 2n=38). The main goal of this project was to combine sequence capture with next generation sequencing (NGS) to discover single nucleotide polymorphisms (SNPs) in specific areas of the B. napus genome historically associated (via quantitative trait loci –QTL– analysis) to traits of agronomical and nutritional importance. A 2.1 million feature sequence capture platform was designed to interrogate DNA sequence variation across 47 specific genomic regions, representing 51.2 Mb of the Brassica A and C genomes, in ten diverse rapeseed genotypes. All ten genotypes were sequenced using the 454 Life Sciences chemistry and to assess the effect of increased sequence depth, two genotypes were also sequenced using Illumina HiSeq chemistry. As a result, 589,367 potentially useful SNPs were identified. Analysis of sequence coverage indicated a four-fold increased representation of target regions, with 57% of the filtered SNPs falling within these regions. Sixty percent of discovered SNPs corresponded to transitions while 40% were transversions. Interestingly, fifty eight percent of the SNPs were found in genic regions while 42% were found in intergenic regions. Further, a high percentage of genic SNPs was found in exons (65% and 64% for the A and C genomes, respectively). Two different genotyping assays were used to validate the discovered SNPs. Validation rates ranged from 61.5% to 84% of tested SNPs, underpinning the effectiveness of this SNP discovery approach. Most importantly, the discovered SNPs were associated with agronomically important regions of the B. napus genome generating a novel data resource for research and breeding this crop species.
