Legionella pneumophila (Lp) is an opportunistic drinking water pathogen that can cause infections through the inhalation of Lp-containing aerosols and can occur in premise plumbing systems. In this work, the use of copper (Cu) and silver (Ag) ions was evaluated at the bench and pilot scale to determine (i) the effective independent concentrations of copper and silver that are efficacious in inactivating Lp, (ii) the impact of various water quality parameters on the efficaciousness of copper and silver ions, and (iii) the effectiveness and practicality of using dissociation to produce ions at the pilot scale. At the bench scale, it was determined that 0.3 ppm and 0.03 ppm of Cu and Ag, respectively, achieved 6-log inactivation of Lp in 5 h in experimental buffer. But, in dechlorinated filter-sterilized tap water, the same concentrations of Cu were not effective, and the effectiveness of Ag was slower. pH and dissolved inorganic carbon content were found to be important parameters in determining if the use of Cu and Ag ions is appropriate. At the pilot scale, dissociation was successfully used to produce Cu and Ag ions. Target levels of ions were met at the pilot scale but were difficult to achieve, and no impact was observed on Lp concentrations. Results from this study suggest that there are important caveats in the application of this technology when applied in a drinking water matrix and prior understanding of a system's water chemistry may be important to determine the effectiveness of Lp disinfection using Cu and Ag.IMPORTANCEThis work sheds light on the effectiveness of using Cu and Ag ions to inactivate (or kill) Legionella pneumophila. This study examined the washoff of Bacillus globigii (Bg) spores from concrete, asphalt, and grass surfaces by stormwater. Bg is a nonpathogenic surrogate for Bacillus anthracis, which is a biological select agent. Areas (2.74 m × 7.62 m) of concrete, grass, and asphalt were inoculated twice at the field site during the study. Spore concentrations were measured in runoff water after seven rainfall events (1.2-65.4 mm) and complimentary watershed data were collected for soil moisture, depth of water in collection troughs, and rainfall using custom-built telemetry units. An average surface loading of 10 7.79 Bg spores/m 2 resulted in peak spore concentrations in runoff water of 102, 260, and 4.1 CFU/mL from asphalt, concrete, and grass surfaces, respectively. Spore concentrations in the stormwater runoff were greatly reduced by the third rain event after both inoculations, but still detectable in some samples. When initial rainfall events occurred longer after the initial inoculation, the spore concentrations (both peak and average) in the runoff were diminished. The study also compared rainfall data from 4 tipping bucket rain gauges and a laser disdrometer and found they performed similarly for values of total rainfall accumulation while the laser disdrometer provided additional information (total storm kinetic energy) useful in comparing the seven different rain events. The soil moisture probes are recommended for assistance in predicting when to sample sites with intermittent runoff. Sampling trough level readings were critical to understanding the dilution factor of the storm event and the age of the sample collected. Collectively the spore and watershed data are useful for emergency responders faced with remediation decisions after a biological agent incident as the results provide insight into what equipment to deploy and that spores may persist in runoff water at quantifiable levels for months. Stream burial, the rerouting of streams into underground culverts, is common in industrialized and densely populated urban areas. While stream burial is common in urban environments, direct characterization of the within-culvert environment is rare and it is unclear if buried reaches reflect neighboring open reaches regarding habitat, biota, and water chemistry. Additionally, for a buried stream, the entrance and exit of the culvert are abrupt habitat transitions within the stream channel, and it is unknown if these transitions lead to similarly abrupt responses in biotic and abiotic characteristics or if responses are gradual. Quantifying the within-culvert environment and transitions upon entering/exiting the culvert has rarely been done but can help inform management practices regarding how these systems are impacted and establish a baseline for evaluating daylighting or stream restoration projects. To understand how culverts affect longitudinal biotic and abiotic characteristics of urban streams, we evaluated longitudinal patterns of physical habitat characteristics, stream water physiochemistry, periphyton biomass, and macroinvertebrate density and diversity in two urban streams that included long (>100 m) culvert reaches. Abrupt transitions in a suite of abiotic and biotic variables were observed at the entrances and exits of the culverts whereas some variables showed no response to the culvert presence. Periphyton biomass and macroinvertebrate density were reduced by 98% and 92%, respectively, by culverts in the two streams. Within the culverts, we observed greater water depths (average of 10 cm outside vs 26 cm within the culvert), finer benthic substrate, and diversity of macroinvertebrates was reduced by 50%. Nutrient concentrations, in contrast, showed no response to the presence of a culvert.
