This study introduces a novel approach for the sensitive and accurate detection of small molecule metabolites, employing metal-phenolic network (MPN) functionalized AuNPs as both adsorbent and matrix to enhance laser desorption/ionization mass spectrometry (LDI-MS) performance. The MPN comprising tannic acid (TA) and transition metal ions (Fe 3+ , Co 2+ , Ni 2+ , Cu 2+ , or Zn 2+ ) was coated on the surface of AuNPs, forming metal-TA network-coated AuNPs (M-TA@AuNPs). The M-TA@AuNPs provided a tunable surface for specific interactions with analytes, displaying distinct enrichment efficacies for different amino acids, especially for Cu-TA@AuNPs exhibiting the highest affinity for histidine (His). Under the optimized condition, the proposed method enabled ultrasensitive detection of His, with good linearity (R 2  = 0.9627) in the low-concentration range (50 nM-1 μM) and a limit of detection (LOD) as low as 0.9 nM. A highly sensitive, selective and recyclable histidine detection method based on magnetic Fe 3 O 4 @mTiO 2 (M-TiO 2 ) nanocomposites with SERRS was developed. Mesoporous M-TiO 2 nanoparticles were functionalized with 4-aminothiophenol and then coupled with histidine through an azo coupling reaction in 5 min, producing the corresponding azo compound. The strong and specific SERRS response of the azo product allowed for ultrasensitive and selective detection for histidine with an M-TiO 2 device loaded with Ag NPs due to the molecular resonance effect and plasmonic effect of Ag NPs under a 532 nm excitation laser. The sensitivity was further enhanced with the magnetic enrichment of M-TiO 2 . The limit of detection (LOD) was as low as 8.00 × 10 -12 mol/L. The M-TiO 2 demonstrated applicability towards histidine determination in human urine without any sample pretreatment. Additionally, the M-TiO 2 device can be recycled for 3 cycles with the photodegradation of the azo product under UV irradiation due to TiO 2 -assisted and plasmon-enhanced photocatalysis. In summary, a multifunctional and recyclable M-TiO 2 device was synthesized based on azo coupling and SERRS spectroscopy for ultra-sensitive and specific histidine sensing. In addition, the proposed system demonstrated the potential for the multiplex determination of toxic compounds in the fields of food safety, industrial production and environmental protection, which benefit from the fingerprint property and universality of SERRS.

In this study, Cu 2+ modulated silver nanoclusters were constructed for the turn-on, label-free detection of L-histidine. Six Ag NCs protected by oligonucleotides (DNA-Ag NCs) were tested in a series of experiments. Finally, A-DAN-Ag NCs were chosen as the best candidate due to their excellent fluorescent properties. The fluorescence of A-DAN-Ag NCs was quenched using Cu 2+ through energy or electron transfer. However, quenched fluorescence could be restored dramatically in the presence of L-histidine due to Cu 2+ liberation from A-DAN-Ag NCs and because of the chelation between the imidazole group of L-histidine and Cu 2+ . The proposed sensor exhibited high selectivity towards L-histidine over other amino acids, with a limit of detection (LOD) of 0.096 μM ranging from 0 to 8 μM. The proposed sensor succeeded in detecting L-histidine in diluted human urine. Therefore, the sensor has promising practical applications in biological systems.

Histidine plays an essential role in most biological systems. Changes in the homeostasis of histidine and histidine-rich proteins are connected to several diseases. Herein, we report a water-soluble Cu(II) coordination polymer, labeled CuCP, for the fluorimetric detection of histidine and histidine-rich proteins and peptides. Single-crystal structure determination of CuCP revealed a two-dimensional wavy network structure in which a carboxylate group connects the individual Cu(II) dimer unit in a syn-anti conformation. The weakly luminescent and water-soluble CuCP shows turn-on blue emission in the presence of histidine and histidine-rich peptides and proteins. The polymer can also stain histidine-rich proteins via gel electrophoresis. The limits of quantifications for histidine, glycine-histidine, serine-histidine, human serum albumin (HSA), bovine serum albumin, pepsin, trypsin, and lysozyme were found to be 300, 160, 600, 300, 600, 800, 120, and 290 nM, respectively. Utilizing the fluorescence turn-on property of CuCP, we measured HSA quantitatively in the urine samples. We also validated the present urinary HSA measurement assay with existing analytical techniques. Job's plot, 1 H NMR, high-resolution mass spectrometry (HRMS), electron paramagnetic resonance (EPR), fluorescence, and UV-vis studies confirmed the ligand displacement from CuCP in the presence of histidine.

A high-throughput fluorimetric assay for histidine was developed, using a 96-well plates platform. The analyte reacts selectively with o -phthalaldehyde under mild alkaline conditions to form a stable derivative. Instrumental-free detection was carried out using a smartphone after illumination under UV light (365 nm). The method was proved to be linear up to 100 μM histidine, with an LLOQ (lower limit of quantification) of 10 μM. The assay was only prone to interference from glutathione and histamine that exist in the urine samples at levels that are orders of magnitude lower compared to histidine. Human urine samples were analyzed following minimum treatment and were found to contain histidine in the range of 280 to 1540 μM. The results were in good agreement with an HPLC corroborative method.

Somapacitan is a reversible albumin-binding growth hormone (GH) derivative in clinical development for once-weekly administration in patients with adult GH deficiency (AGHD) and children with GH deficiency (GHD). To date, the use of somapacitan in AGHD or severe AGHD has been approved in the USA and Japan, respectively. This study (ClinicalTrials.gov, NCT02962440) investigated the absorption, metabolism and excretion, as well as the pharmacokinetics (PK), of tritium-labelled somapacitan ([ 3 H]-somapacitan). Seven healthy males received a single subcutaneous dose of 6 mg somapacitan containing [ 3 H]-somapacitan 20 MBq. Blood, serum, plasma, urine, faeces, and expired air were collected for radioactivity assessment. Metabolites were identified and quantified in plasma and urine collected. The PK of plasma components were determined, and the radioactive peaks of the most abundant plasma metabolites and urine metabolites were selected for analysis. Twenty-eight days after dosing, 94.0% of the administered dose was recovered as [ 3 H]-somapacitan-related material, most of which was excreted in urine (80.9%); 12.9% was excreted in faeces, and an insignificant amount (0.2%) was exhaled in expired air. PK properties of [ 3 H]-somapacitan-related material appeared to be consistent across plasma, serum and blood. Three abundant plasma metabolites (P1, M1 and M1B) and two abundant urine metabolites (M4 and M5) were identified. The total exposure of intact somapacitan accounted for 59% of the total exposure of all somapacitan-related material, P1 accounted for 21% and M1 plus M1B accounted for 12%. M4 and M5 were the most abundant urine metabolites and accounted for 37% and 8% of the dosed [ 3 H]-somapacitan radioactivity, respectively. No intact somapacitan was found in excreta. Two subjects had six adverse events (AEs); all were mild in severity and unlikely to be related to trial product. The majority of dosed [ 3 H]-somapacitan (94%) was recovered as excreted metabolites. Urine was the major route for excretion of somapacitan metabolites, followed by faeces, and exhalation in expired air was negligible. The low molecular weights of identified urine metabolites demonstrate that somapacitan was extensively degraded to small residual fragments that were excreted (fully biodegradable). The extensive metabolic degradation and full elimination of metabolites in excreta were the major clearance pathways of somapacitan and the key elements in its biological fate. A single dose of 6 mg somapacitan (containing [ 3 H]-somapacitan) in healthy male subjects was well tolerated with no unexpected safety issues identified., (Copyright © 2021. Bladder cancer (BC) is a common malignancy of the urinary system and a leading cause of death worldwide. In this work, untargeted metabolomic profiling of biological fluids is presented as a non-invasive tool for bladder cancer biomarker discovery as a first step towards developing superior methods for detection, treatment, and prevention well as to further our current understanding of this disease. In this study, urine samples from 24 healthy volunteers and 24 BC patients were subjected to metabolomic profiling using high throughput solid-phase microextraction (SPME) in thin-film format and reversed-phase high-performance liquid chromatography coupled with a Q Exactive Focus Orbitrap mass spectrometer. The chemometric analysis enabled the selection of metabolites contributing to the observed separation of BC patients from the control group. Relevant differences were demonstrated for phenylalanine metabolism compounds, i.e., benzoic acid, hippuric acid, and 4-hydroxycinnamic acid. Furthermore, compounds involved in the metabolism of histidine, beta-alanine, and glycerophospholipids were also identified. Thin-film SPME can be efficiently used as an alternative approach to other traditional urine sample preparation methods, demonstrating the SPME technique as a simple and efficient tool for urinary metabolomics research. Moreover, this study's results may support a better understanding of bladder cancer development and progression mechanisms.

Background and Objectives: Genetic variants in NAT8 , a liver- and kidney-specific acetyltransferase encoding gene, have been associated with eGFR and CKD in European populations. Higher circulating levels of two NAT8 -associated metabolites, N- δ -acetylornithine and N-acetyl-1-methylhistidine, have been linked to lower eGFR and higher risk of incident CKD in the Black population. We aimed to expand upon prior studies to investigate associations between rs13538, a missense variant in NAT8 , N-acetylated amino acids, and kidney failure in multiple, well-characterized cohorts., Design, Setting, Participants, & Measurements: We conducted analyses among participants with genetic and/or serum metabolomic data in the African American Study of Kidney Disease and Hypertension (AASK; n =962), the Atherosclerosis Risk in Communities (ARIC) study ( n =1050), and Bio Me , an electronic health record-linked biorepository ( n =680). Separately, we evaluated associations between rs13538, urinary N-acetylated amino acids, and kidney failure in participants in the German CKD (GCKD) study ( n =1624)., Results: Of 31 N-acetylated amino acids evaluated, the circulating and urinary levels of 14 were associated with rs13538 ( P <0.05/31). Higher circulating levels of five of these N-acetylated amino acids, namely, N- δ -acetylornithine, N-acetyl-1-methylhistidine, N-acetyl-3-methylhistidine, N-acetylhistidine, and N2,N5-diacetylornithine, were associated with kidney failure, after adjustment for confounders and combining results in meta-analysis (combined hazard ratios per two-fold higher amino acid levels: 1.48, 1.44, 1.21, 1.65, and 1.41, respectively; 95% confidence intervals: 1.21 to 1.81, 1.22 to 1.70, 1.08 to 1.37, 1.29 to 2.10, and 1.17 to 1.71, respectively; all P values <0.05/14). None of the urinary levels of these N-acetylated amino acids were associated with kidney failure in the GCKD study., Conclusions: We demonstrate significant associations between an NAT8 gene variant and 14 N-acetylated amino acids, five of which had circulation levels that were associated with kidney failure., (Copyright © 2021 by the American Society of Nephrology.)

In this study, a novel simple type of label-free, ultra-sensitive, and highly selective UV-Vis absorption and naked-eye detection of histidine (His) and lysine (Lys) using a dye/metal ion ensemble is developed. The outcoming high sensitivity and selectivity for histidine and lysine were attained by changing the metal ions. The indicator is released due to its displacement from the murexide (Mure)/Cu 2+ complex by histidine and the change in absorbance may be due to the further complexation of lysine with the additional coordination sites present in the zinc atom of Mure/Zn 2+ complex. The label-free chemosensor provided sensitive and selective detection of l-histidine and l-lysine with detection limits of 9.1 and 9.4 nmol L -1 , respectively. The protocol especially offers high selectivity for the determination of His and Lys among amino acids found in human urine samples. In the present study, the determination of histidine (HIS) by an on-line flow method based on the concept of zone fluidics is reported. HIS reacts fast with o -phthalaldehyde at a mildly basic medium (pH 7.5) and in the absence of additional nucleophilic compounds to yield a highly fluorescent derivative ( λ ex / λ em = 360/440 nm). The flow procedure was optimized and validated, paying special attention to its selectivity and sensitivity. The LOD was 31 nmol·L -1 , while the within-day and day-to-day precisions were better than 1.0% and 5.0%, respectively ( n = 6). Random urine samples from adult volunteers ( n = 7) were successfully analyzed without matrix effect (<1%). Endogenous HIS content ranged between 116 and 1527 μmol·L -1 with percentage recoveries in the range of 87.6%-95.4%.

Objective: Excess catecholamine release by pheochromocytomas and paragangliomas (PPGL) leads to characteristic clinical features and increased morbidity and mortality. The influence of PPGLs on metabolism is ill described but may impact diagnosis and management. The objective of this study was to systematically and quantitatively study PPGL-induced metabolic changes at a systems level., Design: Targeted metabolomics by liquid chromatography-tandem mass spectrometry of plasma specimens in a clinically well-characterized prospective cohort study., Methods: Analyses of metabolic profiles of plasma specimens from 56 prospectively enrolled and clinically well-characterized patients (23 males, 33 females) with catecholamine-producing PPGL before and after surgery, as well as measurement of 24-h urinary catecholamine using LC-MS/MS., Results: From 127 analyzed metabolites, 15 were identified with significant changes before and after surgery: five amino acids/biogenic amines (creatinine, histidine, ornithine, sarcosine, tyrosine) and one glycerophospholipid (PCaeC34:2) with increased concentrations and six glycerophospholipids (PCaaC38:1, PCaaC42:0, PCaeC40:2, PCaeC42:5, PCaeC44:5, PCaeC44:6), two sphingomyelins (SMC24:1, SMC26:1) and hexose with decreased levels after surgery. Patients with a noradrenergic tumor phenotype had more pronounced alterations compared to those with an adrenergic tumor phenotype. Weak, but significant correlations for 8 of these 15 metabolites with total urine catecholamine levels were identified., Conclusions: This first large prospective metabolomics analysis of PPGL patients demonstrates broad metabolic consequences of catecholamine excess. Robust impact on lipid and amino acid metabolism may contribute to increased morbidity of PPGL patients.

A highly selective and sensitive electroanalysis strategy has been developed for sensing histamine (HTA) and histidine (His) with "turn-on" signal outputs using copper nanocomposites (Cu NCs) of reduced copper metal-organic frameworks (Cu MOFs). It was discovered that the Cu NC-modified electrodes could display the sharp and stable oxidation peaks of solid-state CuCl electrochemistry at a low potential (about -0.10 V). More interestingly, once HTA or His was introduced, the peaking currents of the electrodes would increase due to the specific interaction between Cu 2+ and imidazole groups of HTA or His. A highly selective electroanalysis method was thereby developed for the detection of both HTA and His in the concentration range of 0.010-100 μM. Besides, the application feasibility of the developed electroanalysis strategy was demonstrated for the evaluation of HTA and His separately in red wine and urine samples. Such an electroanalysis candidate for HTA and His holds great potential for wide applications in the fields of food analysis and clinical disease diagnosis.

A 25 μL analytical glass syringe has been used for isoelectric focusing (IEF) utilizing the stainless-steel needle and plunger as electrodes. The generation of protons and hydroxyl ions at the electrodes facilitated a neutralization reaction boundary (NRB) mechanism to focus different amphoteric compounds, such as hemoglobin, bovine serum albumin, R-phycoerythrin, and histidine, within minutes. After optimization of different experimental parameters affecting the IEF process and the coupling of the IEF syringe with electrospray ionization mass spectrometry (ESI-MS), a BGE composed of NH 4 Ac, 1.0 mM, pH 4.0, in 70.0% (v/v) acetonitrile was used for the IEF of histidine. A voltage of -200 V was applied for 5.0 min to accomplish the IEF and increased to -400 V during the infusion to ESI-MS at a flow rate of 4.0 μL/min. The coaxial sheath liquid consisting of 0.2% (v/v) formic acid was added at 4.0 μL/min. The detection limit was found to be 2.2 μg/mL and a nonlinear quadratic fit calibration curve was constructed for histidine over the range of 4.0-64.0 μg/mL with a correlation coefficient ( r) = 0.9998. The determination of histidine in spiked urine samples as relevant for the diagnosis of histidinemia was demonstrated by the IEF syringe-ESI-MS system with accuracy from 88.25% to 102.16% and a relative standard deviation less than 11%.

A simple coumarin-based Schiff base (probe L) was successfully developed. It showed strong green fluorescence emission at 527 nm with a 70.3% of fluorescence quantum efficiency (Φ F ). However, after the addition of common metal ions, probe L can only combine with Cu 2+ ions and displayed significant fluorescence quenching of > 96.2% (Φ F  = 2.7%) due to the paramagnetic quenching action from Cu 2+ . Conversely, by the coordination action of cysteine (Cys) and histidine (His), the quenching fluorescence of the complex (L-Cu 2+ ) between probe L and Cu 2+ ions was recovered mostly because the Cys and His can usurp Cu 2+ of L-Cu 2+ and led to the liberation of probe L. Based on the fluorescence changes of probe L with the actions of Cu 2+ , Cys, and His, an on-off-on reversible fluorescence probe for sensitive and specific monitoring Cu 2+ , Cys, and His has been prepared. More importantly, the probe L and L-Cu 2+ ensemble can be used, respectively, to test Cu 2+ and Cys/His in live cells and human urine samples with great reliability.

A novel label-free fluorescent sensor for histidine was developed based upon Cu 2+ -specific DNAzyme, hybridization chain reaction(HCR) and triplex DNA. Cu 2+ can bind to the histidine, in the presence of histidine, leading to the inhibition of the cleavage of substrate strand of Cu 2+ -dependent DNAzyme, then the intact substrate strand trigger the HCR between H1 and H2. The HCR product can be recognized by triplex-forming oligonucleotide (TFO) through triplex formation and reported by the fluorescence of berberine, the fluorescence intensity of the sensing system was proportional to the concentration of histidine during the range of 5.7-455 nmol L -1 , with a detection limit of 2.0 nmol L -1 ., (Copyright © 2019 Elsevier B.V. Urine adulteration to circumvent positive drug testing is a fundamental challenge for toxicological laboratories all over the world. Untargeted mass spectrometry (MS) methods used in metabolomics had previously revealed uric acid (UA), histidine, methylhistidine, and their oxidation products, for example 5-hydroxyisourate (HIU) as potential biomarkers for urine adulteration using potassium nitrite (KNO 2 ). These markers should be further evaluated for their reliability, stability, and routine applicability. Influence of KNO 2 concentration, urinary pH, reaction time, and stability at room temperature, 4°C, and - 20°C was determined in urine under varying conditions. Analysis was performed after protein precipitation with acetonitrile by liquid chromatography-high resolution mass spectrometry (LC-HRMS). Receiver operating characteristics (ROC) analysis was applied for cut-off evaluation after biomarker quantification (n = 100 per group). Blinded measurements (n = 50) were performed to check the general applicability to identify adulterated samples under routine conditions. The higher the adulterant concentration, the lower the concentrations of histidine, methylhistidine, and UA. In return, amounts of their oxidation products increased. Highest changes were observed under weak acid conditions (pH 4-5). Storage at -20°C ensured sufficient stability for all oxidative markers over one month. ROC evaluated biomarker performance and application to unknown samples revealed satisfying results, with HIU as the most suitable biomarker (positive predictive value (PPV) 100%), followed by UA (PPV 93%). We proposed a colorimetric method for l-histidine detection based on Cu 2+ -mediated DNAzyme and G-quadruplex-hemin complex catalyzed oxidation of 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS). In this system, after the addition of l-histidine, the formation of G-quadruplex-hemin complex will be disturbed, thus the colorimetric signal intensity conversely corresponds to the concentration of histidine. In this assay, a lower detection limit of l-histidine (50 nM) is addressed comparing to previously reported colorimetric methods. The cost is extremely low as the proposed design is both label-free and enzyme-free. All the more vitally, the colorimetric detection procedure is substantially straightforward with no further modification processes. Purpose: Asthma-COPD overlap (ACO) is heterogeneous in nature and requires a unified diagnostic approach. We investigated the urinary levels of l-histidine, a precursor of histamine related to inflammatory responses, as a new candidate biomarker for diagnosing this condition., Patients and Methods: We performed a prospective multicenter cohort study with retrospective analysis of 107 patients, who were divided into three groups: asthma, COPD, and ACO, according to the Spanish guidelines algorithm. Urinary l-histidine levels were measured using liquid chromatography-mass spectrometry. High-resolution metabolomic analysis, coupled with liquid chromatography-mass spectrometry and followed by multivariate statistical analysis, was performed on urine samples to discriminate between the metabolic profiles of the groups., Results: Urinary l-histidine levels were significantly higher in patients with ACO than in those with asthma or COPD, but the subgroups of ACO, classified according to disease origin, did not differ significantly. High urinary l-histidine level was a significant factor for the diagnosis of ACO even after adjusting for age, sex, and smoking amount. Among patients with airflow obstruction, the urinary l-histidine levels were elevated in patients with a documented history of asthma before the age of 40 years or bronchodilator responsiveness ≥400 mL; bronchodilator responsiveness ≥200 mL of forced expiratory volume in 1 second and exceeding baseline values by 12% on two or more visits; blood eosinophil count ≥300 cells·mm -3 ; and frequent exacerbations ( P < 0.05)., Conclusion: Urinary l-histidine could be a potential biomarker for ACO, regardless of the diversity of diagnostic definitions used., Competing Interests: Disclosure The authors report no conflicts of interest in this work.

A highly sensitive sensor for detection of histidine (His) based on the nitrogen-doped graphene quantum dots (N-GQDs)-Cu 2+ system has been designed. The N-GQDs were synthesized by one-step hydrothermal approach according to previous report. The fluorescence of N-GQDs can be effectively quenched by Cu 2+ due to the binding between Cu 2+ and functional groups on the surface of N-GQDs. The high affinity of His to Cu 2+ enables Cu 2+ to be dissociated from the surface of N-GQDs and recovering the fluorescence. The sensor displayed a sensitive response to His in the concentration range of 0-35μmolL -1 , with a detection limit of 72.2nmolL -1 . A new, precise, and very selective method for increasing the impact and assessment of histidine as a biomarker for early diagnosis of histidinemia disease in new born children was developed. The method depends on the formation of the ion pair associate between histidine and the nano optical samarium tetracycline [Sm-(TC) 2 ] + complex doped in sol-gel matrix in a borate buffer of pH 9.2. The [Sm-(TC) 2 ] + complex has +I net charge which is very selective and sensitive for [histidine] - at pH 9.2 in serum and urine samples of histidinemia disease. Histidine enhances the luminescence intensity of the nano optical [Sm-(TC) 2 ] + complex at 645nm after excitation at 400nm, in borate buffer, pH 9.2. The remarkable enhancement of the luminescence intensity at 645nm of nano [Sm-(TC) 2 ] + complex doped in sol-gel matrix by various concentrations of the histidine was successfully used as an optical probe for the assessment of histidine in different serum and urine samples of new born children infected by histidinemia. The calibration plot was achieved over the concentration range 1.4×10 -5 - 6.5×10 -10 molL -1 histidine with a correlation coefficient of (0.998) and a detection limit of (3.2×10 -10 molL -1 ). We assessed the association between urinary metabolites, genetic variants, and incident chronic kidney disease (CKD) in the Framingham Offspring cohort. Among the participants, 193 individuals developed CKD (estimated glomerular filtration rate under 60 ml/min/1.73m 2 ) between cohort examinations 6 (1995-1998) and 8 (2005-2008, mean follow-up 9.7 years). They were age- and sex-matched to 193 control individuals free of CKD. A total of 154 urinary metabolites were measured using mass spectrometry, and the association between metabolites and CKD was examined using logistic regression. Next, we tested the genetic associations of each metabolite with an Illumina exome chip. Urinary glycine and histidine were associated with a lower risk of incident CKD with an odds ratio of 0.59 (95% confidence interval [CI] 0.43-0.80) and 0.65 (0.50-0.85) respectively, per one standard deviation increase in metabolite concentration. Follow-up in the Atherosclerosis Risk in Communities cohort confirmed the association of urinary glycine with CKD. In exome chip analyses, 36 single nucleotide polymorphisms at 30 loci were significantly associated with 31 metabolites. We surveyed exome chip findings for associations with known renal function loci such as rs8101881 in SLC7A9 coding for an amino acid transporter, which has been associated with a lower risk of CKD. We found this polymorphism was significantly associated with higher levels of lysine and NG-monomethyl-L-arginine (NMMA). Increased urinary lysine and NMMA were associated with a lower risk of CKD (0.73 [0.50-0.90] and 0.66 [0.53-0.83], respectively) in the univariate model. Thus, low urinary glycine and histidine are associated with incident CKD. To date urinary metabolic profiling has been applied to define a specific metabolic fingerprint of hepatocellular carcinoma on a background of cirrhosis. Its utility for the stratification of other complications of cirrhosis, such as hepatic encephalopathy (HE), remains to be established. Urinary proton nuclear magnetic resonance ( 1 H-NMR) spectra were acquired and NMR data from 52 patients with cirrhosis (35 male; 17 female, median (range) age [60 (18-81) years]) and 17 controls were compared. A sub-set of 45 patients (33 male; 12 female, [60 (18-90) years, median model for end stage liver disease (MELD) score 11 (7-27)]) were fully characterised by West-Haven criteria, Psychometric Hepatic Encephalopathy Score (PHES) and electroencephalogram (EEG), and defined as overt HE (OHE, n = 21), covert HE (cHE, n = 7) or no HE (n = 17). Urinary proton nuclear magnetic resonance ( 1 H-NMR) spectra were analysed by partial-least-squares discriminant analysis (PLS-DA). The results showed good discrimination between patients with cirrhosis (n = 52) and healthy controls (n = 17) (R2X = 0.66, R2Y = 0.47, Q2Y = 0.31, sensitivity-60 %, specificity-100 %) as the cirrhosis group had higher 1-methylnicotinamide with lower hippurate, acetate, phenylacetylglycine and N-methyl nicotinic acid levels. While patients with OHE could be discriminated from those with no HE, with higher histidine, citrate and creatinine levels, the best models lack robust validity (R2X = 0.65, R2Y = 0.48, Q2Y = 0.12, sensitivity-100 %, specificity-64 %) with the sample size used. Urinary 1 H-NMR metabolic profiling did not discriminate patients with cHE from those without HE, nor discriminate subjects on the basis of PHES/EEG result or MELD score. In conclusion, patients with cirrhosis showed different urinary 1 H-NMR metabolic profiles compared to healthy controls and those with OHE may be distinguished from those with no HE although larger studies are required. However, urinary 1 H-NMR metabolic profiling did not discriminate patients with differing grades of HE or according to severity of underlying liver disease.

This study on interstitial cystitis (IC) aims to identify a unique urine metabolomic profile associated with IC, which can be defined as an unpleasant sensation including pain and discomfort related to the urinary bladder, without infection or other identifiable causes. Although the burden of IC on the American public is immense in both human and financial terms, there is no clear diagnostic test for IC, but rather it is a disease of exclusion. Very little is known about the clinically useful urinary biomarkers of IC, which are desperately needed. Untargeted comprehensive metabolomic profiling was performed using gas-chromatography/mass-spectrometry to compare urine specimens of IC patients or health donors. The study profiled 200 known and 290 unknown metabolites. The majority of the thirty significantly changed metabolites before false discovery rate correction were unknown compounds. Partial least square discriminant analysis clearly separated IC patients from controls. The high number of unknown compounds hinders useful biological interpretation of such predictive models. Given that urine analyses have great potential to be adapted in clinical practice, research has to be focused on the identification of unknown compounds to uncover important clues about underlying disease mechanisms.

Bretschneider (histidine-tryptophan-ketoglutarate, HTK) solution employed for induction of cardioplegic arrest possesses a high histidine concentration (198 mM). Due to the large volume administered, massive amounts of histidine are incorporated. The aim of the study was to evaluate alterations in amino acid and nitrogen metabolism originating from histidine degradation. Between 07/2014 and 10/2014, a total of 29 consecutive patients scheduled for elective isolated coronary artery bypass grafting with cardiopulmonary bypass (CPB) were enrolled in this prospective observational study. The patients received 1.6 L cardioplegic Bretschneider solution on average. Blood gas and urine samples obtained were analyzed for amino acid as well as urea and ammonium concentrations. After CPB initiation, plasma histidine concentration greatly increased to 21,000 µM to reach 8000 µM at the end. Within the operative period, plasma concentrations of aspartate, glutamate, asparagine, alanine, and glutamine increased variable in magnitude. During the same time, urinary analysis revealed histidine excretion of 19,500 µmol in total and marked elevations in glutamate and glutamine excretion. The absolute amounts of urea and ammonium excreted additionally were 3 mmol and 8 mmol, respectively. Already during CPB, distinct amounts of the histidine administered are metabolized, mainly to other amino acids, but only small amounts to urea and ammonia. Thus, the impact of the histidine incorporated on acid-base status in the intraoperative phase is minor. On the other hand, intraoperative provision of several amino acids arising from histidine metabolism might mitigate postaggression syndrome.

A selenosugar (selenosugar 1, methyl-2-acetamido-2-deoxy-1-seleno-β-D-galactopyranoside) was identified in aqueous extracts of muscle tissue of three marine fish species, mackerel (Scomber scombrus), sardine (Sardina pilchardus), and tuna (Thunnus albacares), by high-performance liquid chromatography coupled to elemental and high-resolution molecular mass spectrometry. Selenoneine (2-selenyl-Nα, Nα, Nα-trimethyl-L-histidine), a known selenium compound in fish, was the major form of selenium in the aqueous extracts, and the methylated derivative of selenoneine, namely Se-methylselenoneine, was also identified as a minor natural constituent in the fish. Selenosugar 1, a major urinary excretion product of selenium often found in organs and body fluids related to selenium excretion, has so far not been reported in muscle tissue. Se-methylselenoneine has been proposed as the main urinary metabolite from selenoneine. This first report of selenosugar 1 and Se-methylselenoneine as natural constituents of fish muscle tissue opens up a new perspective on the role of these compounds in selenium metabolism and is relevant to selenium supplementation studies.

In the present study, a new strategy based on Cu(2+) mediated DNA-templated silver nanoclusters (DNA-Ag NCs) was developed, as a label-free, on-off-on fluorescent probe for the detection of l-histidine. Eight synthesized DNA oligonucleotides (D1-D8) were experimentally tested, and D5-Ag NCs was finally selected for l-histidine detection due to its best fluorescent properties. The fluorescence emission of D5-Ag NCs could be quenched by Cu(2+) via electron or energy transfer. Upon addition of l-histidine, the chelation between Cu(2+) and the imidazole group of l-histidine leads to Cu(2+) liberation from D5-Ag NCs, and subsequently results in a dramatic fluorescence enhancement of the probe. The method displayed a good selectivity toward l-histidine over all the other amino acids, with a linear relationship in the range of 0.20-80μM, and a limit of detection (LOD) of 4.3nM. By means of employing 11-mercaptoundecanoic acid (11-MUA) as a reducing agent and protecting ligand, we present straightforward one-pot preparation of fluorescent Ag/Au bimetallic nanoclusters (namely AgAuNCs@11-MUA) from AgNO3 and HAuCl4 in alkaline aqueous solution at room temperature. It is found that the fluorescence of AgAuNCs@11-MUA has been selectively quenched by Cu(2+) ions, and the nonfluorescence off-state of the as-prepared AgAuNCs@11-MUA-Cu(2+) ensemble can be effectively switched on upon the addition of histidine and cysteine. By incorporating Ni(2+) ions and N-ethylmaleimide, this phenomenon is further exploited as an integrated logic gate and a specific fluorescence turn-on assay for selectively and sensitively sensing histidine and cysteine has been designed and established based on the original noncovalent AgAuNCs@11-MUA-Cu(2+) ensemble. Under the optimal conditions, histidine and cysteine can be detected in the concentration ranges of 0.25-9 and 0.25-7 μM; besides, the detection limits are found to be 87 and 111 nM (S/N = 3), respectively. Furthermore, we demonstrate that the proposed AgAuNCs@11-MUA-based fluorescent assay can be successfully utilized for biological fluids sample analysis.

We report a turn-on phosphorescence probe for detection of histidine based on Co(2+)-adsorbed N-acetyl-L-cysteine (NAC) capped Mn: ZnS quantum dots (QDs) which is directly synthesized by the hydrothermal method. The phosphorescence of NAC-Mn: ZnS QDs is effectively quenched by Co(2+) attributing to the adsorption of Co(2+) onto the surface of QDs with a concomitant in suppressing the recombination process of hole and electron of QDs. The phosphorescence of Co(2+)-adsorbed NAC-Mn: ZnS QDs can be recovered by binding of Co(2+) with histidine. The quenching and regeneration of the phosphorescence of NAC-Mn: ZnS QDs have been studied in detail. The as-prepared QDs-based probe is applied to determine histidine with a linear range of 1.25-30 μM and a detection limit of 0.74 μM. The relative standard deviation for eleven repeat detections of 20 μM histidine is 0.65%. Co(2+)-adsorbed NAC-Mn: ZnS QDs show high sensitivity and good selectivity to histidine over other amino acids, metal ions and co-existing substances. We elucidated theoretically and experimentally that counter-ions in background electrolyte (BGE) play a role of booster for electrokinetic injection (EKI) for the determination of cationgenic weak electrolytes and amino acids in neutral aqueous solutions using capillary electrophoresis (CE). The pH change in the sample solution caused by the migration of counter-ions resulted in the increase of analyte mobility and hence the increase of the amount of analyte injected into the capillary. This type of EKI was named as counter-ion boosted EKI. Using the counter-ion boosted EKI-capillary zone electrophoresis (CZE), the limit of detections (LODs, S/N=3) for creatinine (4.8nM) and l-histidine (9.0nM) were lowest ever achieved by CE with UV detection. The RSDs (n=3) of the migration time for creatinine and l-histidine were obtained as 0.35% and 0.34%, for peak areas of 13% and 12%, and for peak heights of 12% and 8.5%, respectively. A new compound L soluble in water derived from vitamin B6 was successfully synthesized and characterized. The complex LAl(3+) is a turn-on selective fluorescent probe for histidine in HEPES buffer, with detection and quantification limits of 0.3 μM and 0.6 μM, respectively. The application of the complex LAl(3+) in real urine samples is reported.

An ensemble-based fluorescent sensor HS-1 + Cu(2+) for detection of histidine is reported. Complex HS-1 + Cu(2+) sensitively senses histidine at pH 7.4 in aqueous media. The quantitative determination of histidine in urine and fetal calf serum is also conducted.

In this work, Ni(2+)-modified gold nanoclusters were fabricated for fluorescence turn-on detection of histidine. The fluorescence of Au NCs was first quenched by Ni(2+). Then, the addition of histidine can restore the fluorescence of Au NCs by binding with Ni(2+) and removing it from the surface of the Au NCs. This architecture ensured non-toxic, cost-effective, label-free and sensitive detection of histidine. The developed Au NCs-based fluorescent sensor offered high selectivity for histidine over other amino acids. The relative standard deviation (RSD) for eleven replicate detections was 2.7%. The detection limit for histidine is 30 nM. The recovery of spiked histidine in human urine samples ranges from 95 to 104%.

A simple and efficient colorimetric method for the naked-eye detection and quantification of histidine in biological fluids was developed based on an indicator-displacement assay (IDA) and the Ni(2+)-histidine affinity pair. In this IDA approach, a commercially available dye, murexide, was used as the indicator and the selective detection of histidine was achieved based on the competition between indicator and histidine for the binding with Ni(2+). The competition of histidine with murexide for Ni(2+) resulted in an obvious color change of the solution from yellow to purple, and the permitted naked-eye detection of trace histidine. The developed bioassay allows the rapid, sensitive and selective detection of histidine in urine samples, and does not need complicated sample pretreatment. The detection limit was 0.4 μM with a linear range from 2 to 30 μM. The relative standard deviation for 11 replicate detections of 8 μM histidine was 2.0%. The developed sensor was successfully applied to the determination of histidine in human urine samples with recoveries from 97 to 105%.

Speciation analysis of selenium in human urine allowed for the first time the identification of a novel selenium metabolite, Se-methylselenoneine. Despite a concentration at low ppb level, its characterization was achieved after sample purification by solid phase extraction (SPE) followed by the parallel coupling of the bidimensional RP/HILIC chromatography with ICP-MS and ESI-LTQ Orbitrap MS detection. To confirm its biological significance with regards to selenoneine, the recently discovered analog of ergothioneine, and to discard the possibility of sample preparation artifacts, a new method was developed to monitor its actual presence, as well as the occurrence of its sulfur and/or non-methylated analogs, in non-preconcentrated urine and blood samples of non-supplemented humans. It consisted in a HILIC ESI-MS(3) method in high resolution mode (resolution 30 000 at m/z 400) with large isolation width windows for precursor ions. These two particular settings allowed respectively to keep observing the specific mass defect of selenium- and sulfur-containing molecules and to maintain the characteristic selenium pattern in product ions created through MS(n) fragmentations. As a result, all four metabolites were detected in blood and three of them in urine. Moreover, different ratios "methylated/non-methylated" were observed between urine and blood samples, which seemed to indicate their active metabolization. The analytical tool developed here will be of a great importance to further study the occurrence and the potential metabolic role in mammalian organelles, cells and fluids of these very particular and promising redox metabolites.

Physical exercise modifies animal metabolism profoundly. Until recently, biochemical investigations related to exercise focused on a small number of biomolecules. In the present study, we used a holistic analytical approach to investigate changes in the human urine metabolome elicited by two exercise sessions differing in the duration of the rest interval between repeated efforts. Twelve men performed three sets of two 80 m maximal runs separated by either 10 s or 1 min of rest. Analysis of pre- and postexercise urine samples by (1)H NMR spectroscopy and subsequent multivariate statistical analysis revealed alterations in the levels of 22 metabolites. Urine samples were safely classified according to exercise protocol even when applying unsupervised methods of statistical analysis. Separation of pre- from postexercise samples was mainly due to lactate, pyruvate, hypoxanthine, compounds of the Krebs cycle, amino acids, and products of branched-chain amino acid (BCAA) catabolism. Separation of the two rest intervals was mainly due to lactate, pyruvate, alanine, compounds of the Krebs cycle, and 2-oxoacids of BCAA, all of which increased more with the shorter interval. Metabonomics provides a powerful methodology to gain insight in metabolic changes induced by specific training protocols and may thus advance our knowledge of exercise biochemistry.

We describe a new CE method with UV-detection for the quantification of histidine (His) and its methylated forms 1-methylhistidine and 3-methylhistidine, both in plasma and urine. Analytes were basically resolved using a 60 mmol/L Tris-phosphate run buffer pH 2.2 in less than 12 min. The use of a mixture of ACN/ammonia (80:20) for protein precipitation allows the quantitative recovery of all His from plasma. The optimization of the sample volume injection permits to reach an LOD of 20 nmol/L, thus improving the sensitivity of about hundred times in comparison to the previous described assays. Moreover, the opportunity to also measure creatinine in the same run makes it possible to evaluate the renal function contemporarily, thus avoiding further dosages with significant time saving. The application method has been proved by measuring His, 1-methylhistidine and 3-methylhistidine in 44 healthy subjects. The high affinity of histidine to Ni2+ has long been recognized in metal ion affinity chromatography for the separation and purification of histidine-tagged proteins. Besides, such affinity pair has been explored in modern nanotechnology for constructing functional nanoparticle-histidine-tagged protein conjugates. However, the use of Ni2+-histidine affinity pair in conjunction with optically-active nanomaterials for sensor design, to our knowledge, has not been reported yet. Here we report a turn-on photoluminescent sensor for histidine based on Ni2+-modulated homocysteine (Hcy)-capped CdTe quantum dots (QDs) by taking the advantages of this well-known Ni2+-histidine affinity pair and photoluminescent QDs. The photoluminescence of Hcy-capped CdTe QDs can be effectively quenched by Ni2+ due to the binding of Ni2+ to the Hcy on the surface of the QDs and the electron transfer from the photoexcited QDs to Ni2+. The high affinity of histidine to Ni2+ enables Ni2+ to be dissociated from the surface of Hcy-capped CdTe QDs to form stable complex with histidine in solution, thereby recovering the photoluminescence of Hcy-capped CdTe QDs. The Ni2+ induced photoluminescence quenching and subsequent histidine-induced photoluminescence recovery for Hcy-capped CdTe QDs build a solid base for the present QD-based turn-on photoluminescent sensor for detecting histidine. The developed QD-based sensor gives excellent selectivity for histidine over other amino acids with the limit of detection (3 s) of 0.3 μM. The relative standard deviation for 11 replicate detections of 15 μM histidine was 2.7%. Affinity analysis is a key biotechnique used in the fields of biology and biomedicine. Herein, we advanced the concept of moving affinity boundary (MAB) using metal ion Ni(II) and histidine (His) as the model inorganic ion and ligand, respectively, developed the simple method of MAB affinity capillary electrophoresis (MAB-ACE), and carried out the relative experiments. The experiments manifested that (a) an MAB could be created with the model metal ion and ligand; (b) the MAB-ACE could specifically capture His rather than other amino acids, or numerous metabolites in human urine; and (c) the capture had the merits of simultaneous focusing and separation to the target metabolite of His. It was further revealed that the specificity of MAB-ACE was originated from the selective affinity interaction and the effective control of affinity conditions. The analyses of His in raw urine by the MAB-ACE are in agreement with those via the standard amino acid analyzer, indicating the reliability of the developed method. Additionally, the MAB-ACE with UV detector had good sensitivity (LOD = 43 ng mL(-1), S/N = 3), 1.0-150 microM linearity and <5% intra-/inter-day variations. The novel method has an evident potential application for capture of a target metabolite in complex biological sample.

The feasibility of microwave-accelerated derivatization for capillary electrophoresis (CE) with laser-induced fluorescence (LIF) detection was evaluated. The derivatization reaction was performed in a domestic microwave oven. Histidine (His), 1-methylhistidine (1-MH) and 3-methylhistidine (3-MH) were selected as test analytes and fluorescein isothiocyanate (FITC) was chosen as a fluorescent derivatizing reagent. Parameters that may affect the derivatization reaction and/or subsequent CE separation were systematically investigated. Under optimized conditions, the microwave-accelerated derivatization reaction was successfully completed within 150 s, compared to 4-24 h in a conventional water-bath derivatization process. This will remarkably reduce the overall analysis time and increase sample throughput of CE-LIF. The detection limits of this method were found to be 0.023 ng/mL for His, 0.023 ng/mL for 1-MH, and 0.034 ng/mL for 3-MH, respectively, comparable to those obtained using traditional derivatization protocols. Background: Urinary enzymes, including the brush border enzyme alanine aminopeptidase (AAP), are reliable biomarkers of nephrotoxicity and represent early indicators of tubular damage. The aim of the present study was to develop and validate an automated urinary AAP activity assay., Methods and Results: For this purpose, we adapted a photometric test using the substrate L-alanine-4-nitroanilide to a dedicated clinical chemistry analyser. Due to the unavailability of calibrators with established AAP activity, we assessed the AAP activity of a commercially available control material using the photometric test. This material was subsequently employed as a calibrator for the automated assay. Quality assurance was accomplished with independent control materials. A fast gel filtration step of urine samples using Sephadex G50 was demonstrated to be necessary before AAP analysis to avoid underestimation of enzyme activity due to ammonia and amino acids. We compared the automated assay with a manual photometric assay and obtained excellent correlation between both methods (r2=0.946, n=50)., Conclusions: We conclude that the newly developed automated assay represents a user-friendly and analytical tool to determine urinary AAP activity in a medium- to high-throughput laboratory. Potential applications include clinical diagnostic work and toxicological studies.

Urinary mutagenicity was evaluated in relation to environmental mutagen exposure (i.e., diet, indoor/outdoor activities, residential area etc.) on the day prior to sample collection, and also considering factors that contribute to the variability of Salmonella mutagenicity assay results. Overnight urine samples from 283 healthy non-smoking residents of northeast Italy (46% males, 20-62 years) were analyzed for mutagenicity on sensitive Salmonella typhimurium strain YG1024 with S9 mix employing the preincubation version of the plate incorporation assay (i.e., the Salmonella reverse mutation test). Urinary mutagenicity varied between 0.02 and 9.84 rev/ equiv. ml, and 7% of samples were positive (i.e., sample elicited a two-fold increase in revertants). There was an evident increase in mutagenicity in subjects with increased intake of mutagen-rich meals (n = 80) (P < 0.01 and positive urine 13% vs. 5%, P = 0.025). Indoor-exposed subjects (n = 65) also showed a higher percentage of positive urine (14% vs. 5%, P = 0.015). In particular, those subjects exposed to cooking fumes the previous evening (n = 28) revealed higher urinary mutagenicity (P = 0.035, positive urine 25% vs. 5%, P < 0.001) than non-indoor exposed. The sources of variability of the mutagenicity assay, mainly the histidine content of the urine concentrate (z = 4.06, P < 0.0001), and to a lesser extent bacterial inoculum size (z = 2.33, P = 0.019), also significantly influenced urinary mutagenicity values. In a linear multiple regression analysis, their effects were still significant (i.e., histidine content P = 0.026 and inoculum size P = 0.021), but the effects of diet, indoor exposure, and other environmental exposures (i.e., traffic and heating system exhausts, residential area) were not. It is concluded that the previous day's exposure to mutagen-rich meals and cooking fumes may influence the presence of mutagenic activity in the overnight urine of non-smoking subjects. This mutagenic activity, which remains in contact with bladder mucosa for several hours, could be considered risk factors for colorectal adenoma and possibly other cancers (i.e., bladder) in non-smokers. A 12-year-old child presented with asymptomatic, noninflammatory, generalized peeling of the skin since early childhood. He was diagnosed as having type A continual peeling skin syndrome. Associated increased excretion of cystine and histidine in the urine has hitherto not been reported.

Increased intake of chromium (Cr) often leads to improvements in glucose, insulin, lipids, and related variables in studies involving humans and experimental and farm animals. However, the results are often variable, depending not only on the selection of subjects but also dietary conditions and the form of supplemental Cr used. Our objective was to find a Cr supplement suitable for humans that was absorbed better than any of those available. Chromium absorption by six adult subjects, three males and three females, was determined based on the amount of Cr excreted in the urine in the initial 2 d following intake of 200 microg of Cr of the various forms of chromium tested. The absorption of the newly synthesized complexes was greatest for those containing histidine. Urinary Cr losses for six control subjects consuming 200 microg of Cr as Cr histidinate increased from basal levels of 256+/-48 to 3670+/-338 ng/d compared with 2082+/-201 ng for Cr picolinate, the currently most popular nutrient supplement, in the 48 h following Cr consumption. Chromium histidinate complexes were stable and absorption was similar to the initial values after more than 2 yr. Mixing of some of the complexes with starch, which was postulated to improve Cr absorption, was shown to essentially block Cr absorption within 1 mo. These data demonstrate that urinary Cr losses need to be determined because stability and absorption of the Cr complexes varies widely and could be responsible for the variability in some of the Cr supplementation studies. Chromium histidinate complexes are absorbed better than any of the Cr complexes currently available and need to be evaluated as Cr nutritional supplements.

1. WHO-coordinated Cardiovascular Diseases and Alimentary Comparison Study (CARDIAC) and its follow-up, MONALISA Study covering 60 populations in 25 countries indicated an increasing prevalence of obesity, even in developing countries. 2. The index of obesity, BMI was confirmed to be related positively with both systolic and diastolic blood pressure (BP) in 60 CARDIAC populations and with the age-adjusted mortality rates of coronary heart diseases (CHD) in 25 populations where reliable demographic data were available. 3. CHD mortality rates were inversely related with biomarkers for soy bean and fish intakes, isoflavones and taurine in 24-h urine samples in 25 CARDIAC study populations. 4. Population samples were divided into two groups: soy eaters and non-eaters as well as fish eaters and non-eaters, respectively, based on WHO-CARDIAC study data. Soy eaters as well as fish eaters had significantly lower BMI, BP and serum cholesterol levels, which are three major CHD risk factors. 5. BMI was inversely related with 24-h urinary isoflavones excretion, the biomarker of soy intake and also with 24-h urinary excretion of histidine, relatively rich in fish. 6. Since both nutrients, isoflavones as phytoestrogen and histidine as the precursor of histamine may affect central appetite mechanisms, soy and fish intake may be recommended to control obesity, the major common risk of lifestyle-related diseases.

Background: Histidinaemia is an autosomal recessive disorder affecting the hepatic enzyme histidine ammonia lyase (histidase) resulting in elevated plasma and urinary histidine and is prototypic of a series of hepatic cytosolic enzyme defects., Aims: To characterise the physiology of murine histidinaemia with respect to histidine excretion and catabolism, and explore the potential for manipulating cellular and whole body histidase metabolism by gene transfer., Materials and Methods: We studied his/his mice which have a G to A substitution in the gene encoding histidase, using both in vitro transduction of isolated hepatocytes by lipofection with wild-type histidase cDNA, and in vivo transduction of whole liver using a retroviral construct., Results and Conclusion: Histidase cDNA expression restored histidase activity in vivo and in vitro towards normal levels, demonstrated both at the cellular level and by whole body metabolic studies, establishing the potential of this model for the development of new gene therapeutic approaches., (Copyright Blackwell Munksgaard 2004)

The amino acid histidine is metabolized to glutamic acid in mammalian tissue. Formiminoglutamic acid (FIGLU) is an intermediary in this reaction, and tetrahydrofolic acid is the coenzyme that converts it to glutamic acid. A test for folate deficiency concerns the measurement of urinary FIGLU excretion after a histidine load. It was observed that folate-deficient individuals receiving the histidine for the FIGLU test made hematological response that alleviated the anemia associated with this deficiency. This was unusual in that a biochemical test to determine the deficiency results in a beneficial effect for one aspect of the deficiency. The studies reported in this paper give a metabolic explanation for this phenomenon. Urine was collected for 24 hr from 25 folate-deficient subjects, 10 vitamin B(12)-deficient subjects, and 15 normal controls. Urinary excretion of histidine was a mean of 203 mg with a range of 130-360 mg for the folate-deficient subjects; 51.5 mg with a range of 30-76.6 mg for normal subjects; and 60.0 mg with a range of 32.3-93.0 mg for the vitamin B(12)-deficient subjects. All the folate-deficient subjects subsequently made a hematological response to the histidine administered for the FIGLU test. No hematological response was observed in the vitamin B(12)-deficient individuals. When folic acid was given to folate-deficient subjects who received no histidine, urinary histidine levels returned to normal levels rapidly and this was followed by a hematological response. Others have shown that volunteers fed a histidine-free diet developed anemia. In normal subjects, histidine is excreted much more in the urine than other essential amino acids are. Hemoglobin protein contains 10% histidine. Under normal conditions, dietary histidine can supply sufficient histidine to prevent anemia. When the dietary intake is diminished or the urinary excretion is greatly increased, anemia results. It is concluded that folate deficiency causes histidine depletion through increased urinary excretion of this amino acid. Feeding histidine replenishes tissue levels of histidine, resulting in hemoglobin regeneration. Folic acid administration results in return of histidine to normal urinary levels. Thus, a combination of folic acid histidine would be beneficial for folate deficient individuals.

The essentiality of histidine in healthy adults is a controversial topic. To study the potential metabolic effects of a lack of exogenous histidine, four healthy adults consumed a histidine-free diet, with adequate energy and 1.0 g/(kg. d) of an L-amino acid mixture for 48 d. Protein metabolism was monitored every 4 d by using indicator amino acid (L-[1-(13)C]phenylalanine) oxidation (in four subjects) and [(15)N]glycine (in one subject). Urine samples (24-h) were collected for measurement of urea, total nitrogen, creatinine, 3-methylhistidine (3-MH), histidine and beta-alanine. Albumin, transferrin and hematologic concentrations were measured on d 0, 24 and 48. Urinary excretion of nitrogen, urea, creatinine and 3-MH were not affected by the histidine-free diet. However, there was a significant (P < 0.001) linear decline (24-28%) in whole-body protein turnover. Significant (P < 0.05) decreases in albumin (12%), transferrin (17%) and hemoglobin (Hb) (11%) concentrations occurred slowly over the histidine depletion period. The urinary excretion of beta-alanine (an index of carnosine catabolism) generally increased in the smallest subject during the consumption of histidine-free diet. This study demonstrates that a lack of histidine in the diet for a prolonged period resulted in an accommodation of protein turnover and phenylalanine oxidation, measured by the (13)C-phenylalanine indicator amino acid. The extensive metabolic accommodation, together with decreases in Hb, albumin and transferrin during histidine depletion, leaves unresolved the issue of whether histidine is a dietary essential amino acid in healthy adults.

To explore the nutritional significance of urea hydrolysis for human subjects, male infants being treated for severe undernutrition were given oral doses of 10 mg [15N15N]urea every 3 h for 36 h, on admission, during rapid growth and after repletion with either moderate or generous intakes of protein. Urea hydrolysis was calculated from the 15N enrichment of urinary urea, and where possible, lysine, alanine, glycine and histidine were isolated from urine by preparative ion-exchange chromatography for measurement of 15N enrichment. Sufficient N was obtained for 15N enrichment of lysine to be measured on fifteen occasions from six children. Urea hydrolysis accounted for half of all urea production with 130 (SD 85) mg N/kg hydrolysed per d, most of which appeared to be utilized in synthetic pathways. Of the samples analysed successfully, nine samples of lysine were enriched with 15N (mean atom percent excess 0.0102, range 0.0017-0.0208) with relative enrichment ratios with respect to lysine of 1.63 (range 0.18-3.15), 1.96 (range 0.7-3.73) and 0.9 (range 0.4-1.8) for glycine, alanine and histidine respectively. Enriched samples were identified at each treatment phase and 68% of the variation in lysine enrichment was explained by the variation in urea enrichment with 54% explained by the overall rate of delivery of 15N to the lower gastrointestinal tract. The results indicate a minimum of 4.7 mg lysine per kg body weight made available by de novo synthesis with the more likely value an order of magnitude higher. Thus, urea hydrolysis can improve the quality of the dietary protein supply by enabling an increased supply of lysine and other indispensable amino acids.
