Abstract

Antibiotic resistance in pathogens is a growing threat to human health. Of particular concern is resistance to carbapenem, which is an antimicrobial agent listed as critically important by the World Health Organization. With the global spread of carbapenem-resistant organisms, there is an urgent need for new treatment options. Shewanella algae is an emerging pathogen found in marine environments throughout the world that has increasing resistance to carbapenem. The organism is also a possible antibiotic resistance reservoir in humans and in its natural habitat. The development of CRISPR/Cas9-based methods has enabled precise genetic manipulation. A number of attempts have been made to knock out resistance genes in various organisms. The study used a single plasmid containing CRISPR/Cas9 and recE/recT recombinase to reverse an antibiotic-resistant phenotype in S. algae and showed bla OXA-55 -like gene is essential for the carbapenem resistance. This result demonstrates a potential validation strategy for functional genome annotation in S. algae.

Abstract

Since 2011, the Global Energy and Water cycle Exchanges (GEWEX) Water Vapor Assessment (G-VAP) has provided performance analyses for state-of-the-art reanalysis and satellite water vapour products to the GEWEX Data and Analysis Panel (GDAP) and the user community in general. A significant component of the work undertaken by G-VAP is to characterise the quality and uncertainty of these water vapour records to (i) ensure full exploitation and (ii) avoid incorrect use or interpretation of results. This study presents results from the second phase of G-VAP, where we have extended and expanded our analysis of total column water vapour (TCWV) from phase 1, in conjunction with updating the G-VAP archive. For version 2 of the archive, we consider 28 freely available and mature satellite and reanalysis data products, remapped to a regular longitude–latitude grid of 2° × 2° and on monthly time steps between January 1979 and December 2019. We first analysed all records for a "common" short period of 5 years (2005–2009), focusing on variability (spatial and seasonal) and deviation from the ensemble mean. We observed that clear-sky daytime-only satellite products were generally drier than the ensemble mean, and seasonal variability/disparity in several regions up to 12 kg m -2 related to original spatial resolution and temporal sampling. For 11 of the 28 data records, further analysis was undertaken between 1988–2014. Within this "long period", key results show (i) trends between - 1.18 ± 0.68 to 3.82 ± 3.94 kg m -2 per decade and - 0.39 ± 0.27 to 1.24 ± 0.85 kg m -2 per decade were found over ice-free global oceans and land surfaces, respectively, and (ii) regression coefficients of TCWV against surface temperatures of 6.17 ± 0.24 to 27.02 ± 0.51 % K -1 over oceans (using sea surface temperature) and 3.00 ± 0.17 to 7.77 ± 0.16 % K -1 over land (using surface air temperature). It is important to note that trends estimated within G-VAP are used to identify issues in the data records rather than analyse climate change. Additionally, breakpoints have been identified and characterised for both land and ocean surfaces within this period. Finally, we present a spatial analysis of correlations to six climate indices within the long period, highlighting regional areas of significant positive and negative correlation and the level of agreement among records. Abstract —J. BLUNDEN, T. BOYER, AND E. BARTOW-GILLIES Earth’s global climate system is vast, complex, and intricately interrelated. Many areas are influenced by global-scale phenomena, including the “triple dip” La Niña conditions that prevailed in the eastern Pacific Ocean nearly continuously from mid-2020 through all of 2022; by regional phenomena such as the positive winter and summer North Atlantic Oscillation that impacted weather in parts the Northern Hemisphere and the negative Indian Ocean dipole that impacted weather in parts of the Southern Hemisphere; and by more localized systems such as high-pressure heat domes that caused extreme heat in different areas of the world. Underlying all these natural short-term variabilities are long-term climate trends due to continuous increases since the beginning of the Industrial Revolution in the atmospheric concentrations of Earth’s major greenhouse gases. In 2022, the annual global average carbon dioxide concentration in the atmosphere rose to 417.1±0.1 ppm, which is 50% greater than the pre-industrial level. Global mean tropospheric methane abundance was 165% higher than its pre-industrial level, and nitrous oxide was 24% higher. All three gases set new record-high atmospheric concentration levels in 2022. Sea-surface temperature patterns in the tropical Pacific characteristic of La Niña and attendant atmospheric patterns tend to mitigate atmospheric heat gain at the global scale, but the annual global surface temperature across land and oceans was still among the six highest in records dating as far back as the mid-1800s. It was the warmest La Niña year on record. Many areas observed record or near-record heat. Europe as a whole observed its second-warmest year on record, with sixteen individual countries observing record warmth at the national scale. Abstract

Since 2011 the Global Energy and Water cycle Exchanges (GEWEX) Water Vapor Assessment (G-VAP) has provided performance analyses for state-of-the-art reanalysis and satellite water vapour products to the GEWEX Data and Analysis Panel (GDAP) and the user community in general. A significant component of the work undertaken by G-VAP is to characterise the quality and uncertainty of these water vapour records to; i) ensure full exploitation and ii) avoid incorrect use or interpretation of results. This study presents results from the second phase of G-VAP, where we have extended and expanded our analysis of Total Column Water Vapour (TCWV) from phase 1, in conjunction with updating the G-VAP archive. For version 2 of the archive, we consider 28 freely available and mature satellite and reanalysis data products, remapped to a regular longitude-latitude grid of 2°× 2°, and on monthly time steps between January 1979 and December 2019. We first analysed all records for a 'common' short period of five years (2005–2009), focusing on variability (spatial & seasonal) and deviation from the ensemble mean. We observed that clear-sky daytime-only satellite products were generally drier than the ensemble mean, and seasonal variability/disparity in several regions up to 12 kg/m2 related to original spatial resolution and temporal sampling. For 11 of the 28 data records, further analysis was undertaken between 1988–2014. Within this 'long period', key results show i) trends between -1.18±0.68 to 3.82±3.94 kg/m2/decade and -0.39±0.27 to 1.24±0.85 kg/m2/decade were found over ice-free global oceans and land surfaces respectively, and ii) regression coefficients of TWCV against surface temperatures of 6.17±0.24 to 27.02±0.51 %/K over oceans (using sea surface temperature) and 3.00±0.17 to 7.77±0.16 %/K over land (using surface air temperature). It is important to note that trends estimated within G-VAP are used to identify issues in the data records rather than analyse climate change. Additionally, breakpoints have been identified and characterised for both land and ocean surfaces within this period. Finally, we present a spatial analysis of correlations to six climate indices within the "long period", highlighting regional areas of significant positive and negative correlation and the level of agreement among records. Abstract

Global Navigation Satellite System (GNSS) Radio Occultation (RO) data play an essential role in improving numerical weather prediction (NWP) and monitoring climate change. The NOAA Commercial RO Purchase Program (CDP) purchased RO data provided by Spire Global Inc. To ensure the data quality from Spire Global Inc. is consistent with other RO missions, we need to quantify their accuracy and retrieval uncertainty carefully. In this work, Spire Wet Profile (wet temperature profile) data from 7 September 2021 to 31 October 2022, processed by the University Corporation for Atmospheric Research (UCAR), and COSMIC-2 (Constellation Observing System for Meteorology, Ionosphere, and Climate-2/Formosa Satellite Mission 7) data are evaluated through comparison with NOAA-20 Advanced Technology Microwave Sounder (ATMS) microwave sounder measurements and collocated RS41 radiosonde measurements. Through the Community Radiative Transfer Model (CRTM) simulation, we convert the Spire and COSMIC-2 RO retrievals to ATMS brightness temperature (BT) at sounding channels CH07 to CH14 (temperature channels), with weighting function peak heights from 8 km to 35 km, and CH19 to CH22 (water vapor channels), with weighting function peak heights ranging from 3.2 km to 6.7 km, and compare the simulations with the collocated NOAA-20 ATMS measurements over ocean. Using ATMS observations as references, Spire and COSMIC-2 BTs agree well with ATMS within 0.07 K for CH07-14 and 0.20 K for CH19-22. The trends between Spire and COSMIC-2 are consistent within 0.07 K/year over the oceans for ATMS CH07-CH13 and CH19-22, indicating that Spire/COSMIC-2 wet profiles are, in general, compatible with each other over oceans. The RO retrievals and RS41 radiosonde observation (RAOB) comparison shows that above 0.2 km altitude, RS41 RAOB matches Spire/COSMIC-2 temperature profiles well with a temperature difference of <0.13 K, and the trends between Spire and COSMIC-2 are consistent within 0.08 K/year over land, indicating that Spire/COSMIC-2 wet profiles are overall compatible with each other through RS41 RAOB measurements over land. In addition, the consistency of Spire and COSMIC-2 based on different latitude intervals, local times, and signal-to-noise ratios (SNRs) through ATMS was evaluated. The results show that the performance of Spire is comparable to COSMIC-2, even though COSMIC-2 has a higher SNR. The high quality of RO profiles from Spire is expected to improve short- and medium-range global numerical weather predictions and help construct consistent climate temperature records. Abstract

In recent years, Global Navigation Satellite System (GNSS) radio occultation (RO) has become a critical observation system for global operational numerical weather prediction. Constellation Observing System for Meteorology, Ionosphere, Climate (COSMIC) 2 (COSMIC-2) has been a backbone RO mission for NOAA. NOAA also began to purchase RO data from commercial sources in 2020. To ensure the consistent quality of RO data from different sources, NOAA Center for Satellite Applications and Research (STAR) has developed capabilities to process all available RO data from different missions. This paper describes the STAR RO processing systems which convert the pseudo-range and carrier phase observations to excess phases and bending angles (BAs). We compared our COSMIC-2 data products with those processed by the University Corporation for Atmospheric Research (UCAR) COSMIC Data Analysis and Archive Center (CDAAC). We processed more than twelve thousand COSMIC-2 occultation profiles. Our results show that the excess phase difference between UCAR and STAR is within a few centimeters at high altitudes, although the difference increases towards the lower atmosphere. The BA profiles derived from the excess phase are consistent with UCAR. The mean relative BA differences at impact height from 10 to 30 km are less than 0.1% for GLObal NAvigation Satellite System (GLONASS) L2C signals and Global Positioning System (GPS) L2C and L2P signals. The standard deviations are 1.15%, 1.15%, and 1.32% for GLONASS L2C signal and for GPS L2C and L2P signals, respectively. The BA profiles agree with those derived from European Center for Medium-range Weather Forecast (ECMWF) reanalysis version 5 (ERA5). The Signal-to-Noise-Ratio (SNR) plays an essential role in the processing. 43. Inverting COSMIC-2 Phase Data to Bending Angle and Refractivity Profiles Using the Full Spectrum Inversion Method

Abstract

The radio occultation technique provides stable atmospheric measurements that can work as a benchmark for calibrating and validating satellite-sounding data. Launched on 25 June 2019, the Constellation Observing System for Meteorology, Ionosphere, and Climate 2 and Formosa Satellite Mission 7 (COSMIC-2/FORMOSAT-7) are expected to produce about 5000 high-quality RO observations daily over the tropics and subtropics. COSMIC-2 constellation consists of 6 Low Earth Orbit (LEO) satellites in 24° inclination orbits at 720 km altitude and distributed mainly between 45°N to 45°S. The COSMIC-2 observations have uniform temporal coverage between 30°N to 30°S. This paper presents an independent inversion algorithm to invert COSMIC-2 geometry and phase data to bending angle and refractivity. We also investigate the quality of Global Navigation Satellite System (GNSS) and LEO position vectors derived from the UCAR COSMIC Data Analysis and Archive Center (CDAAC). The GNSS and LEO position vectors are stable with LEO position variations < 1.4 mm/s. The signal-to-noise ratio (SNR) on the L1 band ranges from 300–2600 v/v with a mean of 1600 v/v. The inversion algorithm developed at NOAA Center for Satellite Applications and Research (STAR) uses the Full Spectrum Inversion (FSI) method to invert COSMIC-2 geometry and phase data to bending angle and refractivity profiles. The STAR COSMIC-2 bending angle and refractivity profiles are compared with in situ radiosonde, the current COSMIC-2 products derived from CDAAC, and the collocated European Center for Medium-Range Weather Forecasts (ECMWF) climate reanalysis data ERA5. The mean bias at 8–40 km altitude among the UCAR, ERA5, and STAR is <0.1% for both bending and refractivity, with a standard deviation in the range of 1.4–2.3 and 0.9–1.1% for bending angles refractivity, respectively. In the lowest 2 km, the RO bias relative to ERA-5 shows a strong latitudinal and SNR dependence.

44. Consistency and Stability of SNPP ATMS Microwave Observations and COSMIC-2 Radio Occultation over Oceans

Abstract

Radio occultation (RO) sensor measurements have critical roles in numerical weather prediction (NWP) by complementing microwave and infrared sounder measurements with information of the atmospheric profiles at high accuracy, precision, and vertical resolution. This study evaluates Constellation Observing System for Meteorology, Ionosphere, and Climate 2 (COSMIC-2) wet temperature and humidity data products’ consistency and stability through inter-comparison with SNPP advanced technology microwave sounder (ATMS) measurements. Through the community radiative transfer model (CRTM), brightness temperature (BT) at SNPP ATMS channels are simulated with COSMIC-2 retrieved atmospheric profiles from two versions of the University Corporation for Atmospheric Research (UCAR) wet profiles (WETprf and WETpf2) as inputs to the CRTM simulation. The analysis was focused on ATMS sounding channels CH07–14 and CH19–22 with sounding weighting function peak heights from 3.2 to 35 km. The COSMIC-2 vs. ATMS inter-comparison indicates that their BT biases are consistent, and the latitudinal difference is <0.3 K over three latitudinal regions. The differences between the two versions of UCAR COSMIC-2 wet profiles are identified and attributed to the differences in the implementation of 1DVAR retrieval algorithms. The stability between UCAR near real-time COSMIC-2 wet profile data and ATMS measurements is also well-maintained. It is demonstrated that the well-sustained quality of COSMIC-2 RO data makes itself a well-suited reference sensor to capture the calibration update of SNPP ATMS. Furthermore, the impacts of the assimilation of COSMIC-2 data into the European Centre for Medium-Range Weather Forecasts (ECMWF) model after 25 March 2020, are evaluated by trending observation-minus-background (O-B) biases, which confirms the statistically significant positive impacts of COSMIC-2 on the ECMWF reanalysis. Abstract

Water vapor plays an important role in both climate change processes and atmospheric chemistry and photochemistry. Global water vapor vertical profile can be derived from satellite infrared and microwave sounders. However, no single remote sensing technique is capable of completely fulfilling the needs for numerical weather prediction, chemistry, and climate studies in terms of vertical resolution, spatial and temporal coverage, and accuracy. In addition to the passive infrared and microwave sounder observations, the active global positioning system (GPS) radio occultation (RO) technique can also provide all-weather temperature and moisture profiles. In this chapter, we describe the current developments of global water vapor vertical profile and total precipitable water derived from active GPS RO measurements. In addition, we also demonstrate the potential improvement of global water vapor estimates using combined active GPS RO and passive IR/MW particularly from Atmospheric InfraRed Sounder (AIRS) and Advanced Technology Microwave Sounder (ATMS) measurements. Results show that because RO data are very sensitive to water vapor variation in the moisture rich troposphere, the RO data are able to provide extra water vapor information for the combined AIRS/ATMS and RO retrievals in the lower troposphere.
