Polyethylene glycol (PEG) is one of the environmentally benign solvent options for green chemistry. It readily absorbs water when exposed to the atmosphere. The Molecular Dynamics (MD) simulations of PEG200, a commercial mixture of low molecular weight polyethyelene glycol oligomers, as well as di-, tetra-, and hexaethylene glycol are presented to study the effect of added water impurities up to a weight fraction of 0.020, which covers the typical range of water impurities due to water absorption from the atmosphere. Each system was simulated a total of four times using different combinations of two force fields for the water (SPC/E and TIP4P/2005) and two force fields for the PEG and oligomer (OPLS-AA and modified OPLS-AA). The observed trends in the effects of water addition were qualitatively quite robust with respect to these force field combinations and showed that the water does not aggregate but forms hydrogen bonds at most between two water molecules. In general, the added water causes overall either no or very small and nuanced effects in the simulation results. Specifically, the obtained water RDFs are mostly identical regardless of the water content. The added water reduces oligomer hydrogen bonding interactions overall as it competes and forms hydrogen bonds with the oligomers. The loss of intramolecular oligomer hydrogen bonding is in part compensated by oligomers switching from inter- to intramolecular hydrogen bonding. The interplay of the competing hydrogen bonding interactions leads to the presence of shallow extrema with respect to the water weight fraction dependencies for densities, viscosities, and self-diffusion coefficients, in contrast to experimental measurements, which show monotonous dependencies. Submerged freshwater macrophytes are crucial for the functioning of lakes. Their growth and survival follow environmental conditions like light, temperature, and nutrient availability. Hence, the impending increase in water temperature as well as changes of nutrients and turbidity will lead to changes in macrophyte geographic and depth distribution: Herein, we assess these potential changes. We apply an eco‐physiological macrophyte growth model to simulate biomass growth and survival of virtual species defined by random trait combinations within expert‐derived trait ranges for oligotraphentic, mesotraphentic, and eutraphentic species groups in deep lakes in Bavaria, Germany, which cover clear, moderate, and turbid lake conditions. The emergent potential species richness is compared with empirically observed species richness to evaluate general predictions for current conditions. Thereafter, we apply the model to scenarios of temperature increase and of turbidity and nutrient change to assess potential changes in species richness and the influence of species' traits on being an environmental change 'winner' or 'loser'. We find a cross‐lake, hump‐shaped pattern of potential species richness along depth. This largely reflects observed patterns, although mismatches were also detected and might be explained by missing processes and environmental heterogeneity within the lake. Rising temperature leads to increased richness of potential species in all lake types, species groups, and depths. Turbidity and nutrient change effects depend on depth and lake type. 'Loser species' under increased turbidity and nutrient level are light consumptive and sensible to disturbances, while 'winner species' have a high biomass production. These findings show that the hump‐shaped depth distributions of submerged macrophyte diversity can emerge solely considering eco‐physiology. The differential responses to environmental changes imply that management measures must account for lake type because those responses can have opposite trends depending on lake depth and type. This study is seeking a better understanding of polyethylene glycol (PEG) as a solvent to promote its use in chemical synthesis. The effect of adding two solutes of interest, 2,2,6,6-tetramethylpiperidinyloxyl (TEMPO) and 5-tert-butylisophthalic acid (5-TBIPA) to PEG200 (average molar weight of 200 g·mol−1) on the solution density, viscosity, and self-diffusion coefficients is monitored in a temperature range of 298.15–358.15 K to deduce how these solutes interact with the PEG200 solvent. The effect of water, the most common impurity in PEGs, is also monitored and found to be nearly negligibly small. Addition of (5-TBIPA) increases solution density and viscosity. Combined with the observation that 5-TBIPA consistently self-diffuses at about half the rate as PEG200 at all investigated experimental conditions, this suggests strong attractive solute–solvent interactions likely through hydrogen bonding interactions. In contrast, addition of TEMPO causes lower solution densities and viscosities suggesting that the solute–solvent interactions of TEMPO lead to an overall weakening of the intermolecular interactions present compared to neat PEG200. Inspection of the viscosity and self-diffusion temperature dependence reveals slight deviations from the Arrhenius equation. Interestingly, the activation energies obtained from the viscosity and the self-diffusion data are essentially identical in values suggesting that the same dynamic processes and thus the same activation barriers govern translational motion and momentum transfer in these PEG200 solutions. Motivation Cancer is one of the leading causes of death worldwide. Despite significant improvements in prevention and treatment, mortality remains high for many cancer types. Hence, innovative methods that use molecular data to stratify patients and identify biomarkers are needed. Promising biomarkers can also be inferred from competing endogenous RNA (ceRNA) networks that capture the gene–miRNA gene regulatory landscape. Thus far, the role of these biomarkers could only be studied globally but not in a sample-specific manner. To mitigate this, we introduce spongEffects, a novel method that infers subnetworks (or modules) from ceRNA networks and calculates patient- or sample-specific scores related to their regulatory activity. Results We show how spongEffects can be used for downstream interpretation and machine learning tasks such as tumor classification and for identifying subtype-specific regulatory interactions. In a concrete example of breast cancer subtype classification, we prioritize modules impacting the biology of the different subtypes. In summary, spongEffects prioritizes ceRNA modules as biomarkers and offers insights into the miRNA regulatory landscape. Notably, these module scores can be inferred from gene expression data alone and can thus be applied to cohorts where miRNA expression information is lacking. Background Eukaryotic gene expression is controlled by cis -regulatory elements (CREs), including promoters and enhancers, which are bound by transcription factors (TFs). Differential expression of TFs and their binding affinity at putative CREs determine tissue- and developmental-specific transcriptional activity. Consolidating genomic datasets can offer further insights into the accessibility of CREs, TF activity, and, thus, gene regulation. However, the integration and analysis of multimodal datasets are hampered by considerable technical challenges. While methods for highlighting differential TF activity from combined chromatin state data (e.g. chromatin immunoprecipitation [ChIP], ATAC, or DNase sequencing) and RNA sequencing data exist, they do not offer convenient usability, have limited support for large-scale data processing, and provide only minimal functionality for visually interpreting results. Results We developed TF-Prioritizer, an automated pipeline that prioritizes condition-specific TFs from multimodal data and generates an interactive web report. We demonstrated its potential by identifying known TFs along with their target genes, as well as previously unreported TFs active in lactating mouse mammary glands. Additionally, we studied a variety of ENCODE datasets for cell lines K562 and MCF-7, including 12 histone modification ChIP sequencing as well as ATAC and DNase sequencing datasets, where we observe and discuss assay-specific differences. Conclusion TF-Prioritizer accepts ATAC, DNase, or ChIP sequencing and RNA sequencing data as input and identifies TFs with differential activity, thus offering an understanding of genome-wide gene regulation, potential pathogenesis, and therapeutic targets in biomedical research. Alzheimer's disease (AD) is a neurodegenerative disease whose molecular mechanisms are activated several years before cognitive symptoms appear. Genotype-based prediction of the phenotype is thus a key challenge for the early diagnosis of AD. Machine learning techniques that have been proposed to address this challenge do not consider known biological interactions between the genes used as input features, thus neglecting important information about the disease mechanisms at play. To mitigate this, we first extracted AD subnetworks from several protein–protein interaction (PPI) databases and labeled these with genotype information (number of missense variants) to make them patient-specific. Next, we trained Graph Neural Networks (GNNs) on the patient-specific networks for phenotype prediction. We tested different PPI databases and compared the performance of the GNN models to baseline models using classical machine learning techniques, as well as randomized networks and input datasets. The overall results showed that GNNs could not outperform a baseline predictor only using the APOE gene, suggesting that missense variants are not sufficient to explain disease risk beyond the APOE status. Nevertheless, our results show that GNNs outperformed other machine learning techniques and that protein–protein interactions lead to superior results compared to randomized networks. Nevertheless, although Nafamostat-mediated inhibition of -1PRF did not translate into reduced SARS-CoV-2 replication in our study, interference with SARS-CoV-2 -1PRF by more potent compounds could still represent a promising antiviral strategy. • Stromal cells are of mesenchymal origin and provide the structure and support the function of organs. • Multiomics has unveiled the origins and previously unknown functional diversity of stromal cells. • Stromal cells such as fibroblasts are now considered members of the innate immune system. • Specific therapeutic targeting of pathogenic fibroblast subsets shows promise for curing inflammatory diseases. In the last fifteen years it has become apparent that tissue-resident mesenchymal cells such as fibroblasts, which are the structural elements of all organs, play a cardinal role in the pathology of immune-mediated inflammatory diseases. We now know that all fibroblasts originate from universal pan-organ cellular ancestors and that they are diversified into more specific subsets according to the functional needs of their home tissue-and its activation state. In arthritis, a plethora of activated joint-resident and migrating fibroblast types have been recently described that are central for pathogenesis and persistence of inflammatory joint-disease. Here we provide a current overview on the multiple inflammatory and immune-related functions of fibroblasts and how they could be curbed to induce long-lasting abatement of disease. Road agencies employ different criteria and methods to compare treatment alternatives and develop maintenance and rehabilitation (M&R) programmes for their networks. The maximisation of benefits has been integrated in several leading pavement management systems and used for network-level programming since the 1980s. The approach defines benefit as the area between the performance curves with and without M&R actions based on an aggregated index, representing the overall pavement condition. Further simplifications like the screening of solutions using an efficiency frontier and the incremental benefit–cost technique have made it possible to apply the method with the available computing power at the time. Based on a case study of 1000 road sections, this paper analyses the effect of benefit maximisation on annual budget, network condition, treatment selection, trigger values and remaining life. M&R programmes minimising costs and maximising benefits are compared at the project and network level for various budget scenarios. The results show that the maximisation of benefits based on an aggregated condition index leads to substantially higher agency costs and favours the selection of expensive treatments with earlier timing, irrespective of actual failure causes. Investigating diversity gradients helps to understand biodiversity drivers and threats. However, one diversity gradient is rarely assessed, namely how plant species distribute along the depth gradient of lakes. Here, we provide the first comprehensive characterization of depth diversity gradient (DDG) of alpha, beta, and gamma species richness of submerged macrophytes across multiple lakes. We characterize the DDG for additive richness components (alpha, beta, gamma), assess environmental drivers, and address temporal change over recent years. We take advantage of yet the largest dataset of macrophyte occurrence along lake depth (274 depth transects across 28 deep lakes) as well as of physiochemical measurements (12 deep lakes from 2006 to 2017 across Bavaria), provided publicly online by the Bavarian State Office for the Environment. We found a high variability in DDG shapes across the study lakes. The DDGs for alpha and gamma richness are predominantly hump‐shaped, while beta richness shows a decreasing DDG. Generalized additive mixed‐effect models indicate that the depth of the maximum richness (Dmax) is influenced by light quality, light quantity, and layering depth, whereas the respective maximum alpha richness within the depth gradient (Rmax) is significantly influenced by lake area only. Most observed DDGs seem generally stable over recent years. However, for single lakes we found significant linear trends for Rmax and Dmax going into different directions. The observed hump‐shaped DDGs agree with three competing hypotheses: the mid‐domain effect, the mean–disturbance hypothesis, and the mean–productivity hypothesis. The DDG amplitude seems driven by lake area (thus following known species–area relationships), whereas skewness depends on physiochemical factors, mainly water transparency and layering depth. The spin of the electron is nowadays replacing the charge as basic carrier of information not only in spintronics applications, but also in the emerging field of quantum information. Topological quantum materials, where spin-momentum locking is believed to lead to particularly long spin lifetimes, are regarded as a promising platform for such applications. However, spin-orbit coupling, that is essential to all topological matter, at the same time gives rise to spin mixing and decoherence as a major obstacle for quantum computing. Here, we give experimental evidence that hot-spots of spin-mixing and spin-conserving contributions of the spin-orbit operator coexist in an archetypal topological Dirac metal, and that these hot spots can have a strongly anisotropic distribution of their respective wave vectors with respect to the spin quantization direction. Our results can be understood within a theory that takes into account the decomposition of the spin-orbit Hamiltonian into spin-conserving and spin-flip terms, contributing to a better understanding of quantum decoherence in topological materials, in general. A common feature of most pavement management systems is the aggregation of condition data collected from periodic surveys to appropriate long homogeneous sections, aiming to reduce the amount of data to be analysed. It can be shown that regardless of the methods used for sectioning and prediction, homogeneous sections cannot describe the true variation in condition and failure time, leading to either over- or underestimation of service life and hence suboptimal selection of timing and type of maintenance and rehabilitation (M&R) treatments. Furthermore, the generation of homogeneous sections hinders the derivation of project-level performance models from historical condition surveys due to shifting of sections and greatly reduces the number of possible work-zone solutions in the optimisation. This paper illustrates the deviations resulting from different levels of data aggregation based on a case study of 1000 road sections. The results of common sectioning algorithms are then compared to those derived from a LCC-optimisation approach based on accurate condition predictions on short survey sections and work-zones based on economies of scale. The conclusions of this work may be useful to road agencies in avoiding substantial drawbacks of data aggregation and improving project-level M&R treatment recommendations based on the same input condition data. mumps virus (MuV) fusion protein (F) plays a crucial role for the entry process and spread of infection by mediating fusion between viral and cellular membranes as well as between infected and neighboring cells, respectively. The fusogenicity of MuV differs depending on the strain and might correlate with the virulence; however, it is unclear which mechanisms contribute to the differentiated fusogenicity. The cleavage motif of MuV F is highly conserved among all strains, except the amino acid residue at position 8 (P8) that shows a certain variability with a total of four amino acid variants (leucine [L], proline [P], serine [S], and threonine [T]). We demonstrate that P8 affects the proteolytic processing and the fusogenicity of MuV F. The presence of L or S at P8 resulted in a slower proteolysis of MuV F by furin and a reduced ability to mediate cell-cell fusion. However, virus-cell fusion was more efficient for F proteins harboring L or S at P8, suggesting that P8 contributes to the mechanism of viral spread: P and T enable a rapid spread of infection by cell-to-cell fusion, whereas viruses harboring L or S at P8 spread preferentially by the release of infectious viral particles. Our study provides novel insights into the fusogenicity of MuV and its influence on the mechanisms of virus spread within infected tissues. Assuming a correlation between MuV fusogenicity and virulence, sequence information on the amino acid residue at P8 might be helpful to estimate the virulence of circulating and emerging strains. Existing pavement management systems (PMSs) aggregate short survey sections into long homogeneous sections (typically 1–5 km), aiming to reduce the amount of data and formulate a tractable optimisation problem. However, assuming that sections in a similar condition will undergo a similar deterioration of this condition is incorrect, resulting in imprecise predictions and violations of thresholds. Thus, any maintenance and rehabilitation (M&R) optimisation approach based on long road sections and a single condition indicator (aggregated or not) cannot provide applicable results at the project level. This paper provides a holistic model framework and consistent methodology for a new PMS, abolishing the need for composite condition indices, data aggregation and homogeneous sections in the M&R optimisation process. The proposed bottom-up optimisation is based on short survey sections (25–50m) and solves for optimal treatment type, timing, work-zone length and layout, minimising agency, road user and environmental life cycle costs. Instead of using just one condition indicator, the method models the effects of each treatment on multiple distress types and their combinations. Moreover, the short survey sections are grouped into larger work zones based on purely economic criteria (i.e. economies-of-scale costs). Treatment cost, temporary traffic control cost, and work-zone user cost models are derived as a function of the project size and calibrated for actual prices in Austria. Genetic algorithms are employed as a part of the solution algorithm, being capable of handling an unlimited number of road sections. While there are numerous studies showing that neutrophil extracellular traps (NETs) contribute to autoimmune inflammation and cause bystander tissue injury, human individuals with genetic impairments in NET formation curiously often suffer from exacerbated autoimmune diseases and/or chronic inflammatory conditions. These findings are confirmed in some mouse models of systemic lupus erythematosus (SLE) and gouty arthritis, where an absence of neutrophils or impairment of NET formation leads to exacerbation of autoimmunity and chronic inflammation. Thus, aside from their role as archetypical proinflammatory cells, neutrophils in general, and NETs in particular, can also interrupt the self-amplifying loop of cell activation and cell recruitment that characterizes neutrophilic inflammation. Here, we review the current state-of-the-science regarding anti-inflammatory and immune-regulatory action of NETs. We give an overview about the mechanistic involvement of NET-associated neutrophil serine proteases and suggest how tailored induction of NET formation could be exploited for the treatment of chronic autoinflammatory disorders. An accurate estimation of service life is of primary interest in pavement management systems limiting the time frame for maintenance and rehabilitation (M&R) treatments. Common condition prediction models are derived by regression analysis at the road network level based on empirical data from periodic condition surveys. If a particular section has not failed prior to the last survey or the condition has improved (e.g. due to treatment), it is considered as censored. If censoring is neglected the performance functions, service lives and estimated costs may show substantial bias. The authors who acknowledge this problem have used standard statistical (survival analysis) techniques accounting for censoring. However, any road section may fail due to different but dependent competing failure causes (risks), each leading to treatments. This constitutes a special type of censoring that cannot be addressed with traditional survival analysis methods relying on the assumption of independent censoring. As the number of failure causes usually exceeds one (e.g. fatigue, permanent deformation, thermal cracking), this case is quite common. Moreover, the time until a first failure depends on the sign and degree of correlation between present failure types being modelled by the overall survival function. This paper presents a critical review and comparison of common regression, Markov chain and survival analysis models with and without correlated competing risks based on computer-generated data. Using performance history and distress progression models at the section level in combination with survival analysis improves the accuracy of predictions in comparison. Furthermore, the paper proposes a simultaneous modelling of joint and marginal service life distributions based on copula functions as generalised solution accounting for dependence between competing risks. Ebola virus (EBOV) and Nipah virus (NiV) infection of humans can cause fatal disease and constitutes a public health threat. In contrast, EBOV and NiV infection of fruit bats, the putative (EBOV) or proven (NiV) natural reservoir, is not associated with disease and it is currently unknown how these animals control the virus. The human interferon (IFN)-stimulated antiviral effector protein tetherin (CD317, BST-2) blocks release of EBOV- and NiV-like particles from cells and is counteracted by the EBOV glycoprotein (GP). In contrast, it is unknown whether fruit bat tetherin restricts virus infection and is susceptible to GP-driven antagonism. Here, we report the sequence of fruit bat tetherin and show that its expression is IFN-stimulated and associated with strong antiviral activity. Moreover, we demonstrate that EBOV-GP antagonizes tetherin orthologues of diverse species but fails to efficiently counteract fruit bat tetherin in virus-like particle (VLP) release assays. However, unexpectedly, tetherin was dispensable for robust IFN mediated inhibition of EBOV spread in fruit bat cells. Thus, the VLP-based model system mimicking tetherin-mediated inhibition of EBOV release and its counteraction by GP seems not to adequately reflect all aspects of EBOV release from IFN-stimulated fruit bat cells, potentially due to differences in tetherin expression levels that could not be resolved by the present study. In contrast, tetherin expression was essential for IFN-dependent inhibition of NiV infection, demonstrating that IFN-induced fruit bat tetherin exerts antiviral activity and may critically contribute to control of NiV and potentially other highly virulent viruses in infected animals. The influenza virus hemagglutinin (HA) facilitates viral entry into target cells. Cleavage of HA by host cell proteases is essential for viral infectivity, and the responsible enzymes are potential targets for antiviral intervention. The type II transmembrane serine protease (TTSP) TMPRSS2 has been identified as an HA activator in cell culture and in the infected host. However, it is less clear whether TMPRSS2-related enzymes can also activate HA for spread in target cells. Moreover, the activity of cellular serine protease inhibitors against HA-activating TTSPs is poorly understood. Here, we show that TMPRSS11A, another member of the TTSP family, cleaves and activates the influenza A virus (FLUAV) HA and the Middle East respiratory syndrome coronavirus spike protein (MERS-S). Moreover, we demonstrate that TMPRSS11A is expressed in murine tracheal epithelium, which is a target of FLUAV infection, and in human trachea, suggesting that the protease could support FLUAV spread in patients. Finally, we show that HA activation by the TMPRSS11A-related enzymes human airway tryptase and DESC1, but not TMPRSS11A itself, is blocked by the cellular serine protease inhibitor hepatocyte growth factor activator inhibitor type-1 (HAI-1). Our results suggest that TMPRSS11A could promote FLUAV spread in target cells and that HA-activating TTSPs exhibit differential sensitivity to blockade by cellular serine protease inhibitors. Abstract Reactive oxygen species (ROS) are created in cells during oxidative phosphorylation by the respiratory chain in the mitochondria or by the family of NADPH oxidase (NOX) complexes. The first discovered and most studied of these complexes, NOX2, mediates the oxidative burst in phagocytes. ROS generated by NOX2 are dreadful weapons: while being essential to kill ingested pathogens they can also cause degenerative changes on tissue if production and release are not balanced by sufficient detoxification. In the last fifteen years evidence has been accumulating that ROS are also integral signaling molecules and are important for regulating autoimmunity and immune-mediated inflammatory diseases. It seems that an accurate redox balance is necessary to sustain an immune state that both prevents the development of overt autoimmunity (the bright side of ROS) and minimizes collateral tissue damage (the dark side of ROS). Herein, we review studies from rodent models of arthritis, lupus, and neurodegenerative diseases that show that low NOX2-derived ROS production is linked to disease and elaborate on the underlying cellular and molecular mechanisms and the translation of these results to disease in humans. The interferon-induced antiviral host cell protein tetherin can inhibit the release of several enveloped viruses from infected cells. The Ebola virus (EBOV) glycoprotein (GP) antagonizes tetherin, but the domains and amino acids in GP that are required for tetherin antagonism have not been fully defined. A GXXXA motif within the transmembrane domain (TMD) of EBOV-GP was previously shown to be important for GP-mediated cellular detachment. Here, we investigated whether this motif also contributes to tetherin antagonism. Mutation of the GXXXA motif did not impact GP expression or particle incorporation and only modestly reduced EBOV-GPdriven entry. In contrast, the GXXXA motif was required for tetherin antagonism in transfected cells. Moreover, alteration of the GXXXA motif increased tetherin sensitivity of a replication-competent vesicular stomatitis virus (VSV) chimera encoding EBOV-GP. Although these results await confirmation with authentic EBOV, they indicate that a GXXXA motif in the TMD of EBOV-GP is important for tetherin antagonism. Moreover, they provide the first evidence that GP can antagonize tetherin in the context of an infectious EBOV surrogate. IMPORTANCE The glycoprotein (GP) of Ebola virus (EBOV) inhibits the antiviral host cell protein tetherin and may promote viral spread in tetherin-positive cells. However, tetherin antagonism by GP has so far been demonstrated only with virus-like particles, and it is unknown whether GP can block tetherin in infected cells. Moreover, a mutation in GP that selectively abrogates tetherin antagonism is unknown. Here, we show that a GXXXA motif in the transmembrane domain of EBOV-GP, which was previously reported to be required for GP-mediated cell rounding, is also important for tetherin counteraction. Moreover, analysis of this mutation in the context of vesicular stomatitis virus chimeras encoding EBOV-GP revealed that GPmediated tetherin counteraction is operative in infected cells. To our knowledge, these findings demonstrate for the first time that GP can antagonize tetherin in infected cells and provide a tool to study the impact of GP-dependent tetherin counteraction on EBOV spread. The colligative property freezing point depression is evaluated as a means for estimating the extent of aggregation for solutions of poly(ethylene oxide) alcohol (C10E6) nonionic surfactant in cyclohexane. Combined with additional measurements of self-diffusion coefficients, it is shown that both unaggregated C10E6 as well as reverse micelles are significantly present for the entire range of measured C10E6 concentration (0.048-2.35 mol kg-1). A change in speciation near 0.2 mol kg-1 is indicated by the results from both freezing point depression and self-diffusion coefficient measurements. It is shown that average reverse micelle radii and aggregation numbers obtained from the ratio of solvent and C10E6 self-diffusion coefficients are consistent with prior reported results. However, unreasonably small radii for the reverse micelles as well as for the cyclohexane were obtained from analysis of the results by the Stokes-Einstein equation using additional measured solution viscosities. The concentration of reverse micelles and unaggregated C10E6 was calculated from the freezing point depression results using the aggregation numbers obtained from ratio of self-diffusion coefficients. These concentrations indicate that the reverse micelles become smaller in average size and increase in number with increasing temperature without an increase in unaggregated C10E6. The ambient solubility of the mineral salts NaCl, KCl, CsCl, KBr, K2SO4 and CuSO4 is reported as a function of composition of mixed binary solvents consisting of water with polyethylene glycol (PEG 200), ethoxylated C9-11 alcohols (C10E6) and ethoxylated, or propoxylated C10-12 alcohols (C11E6P1). Solubility gradually decreases with decreasing water content and follows the order PEG 200 > C10E6 > C11E6P1. Solubility of CsCl and KBr was found to be surprisingly high in neat PEG 200, on the order of 1 mol kg-1, and in neat C10E6, on the order of 0.1 mol · kg-1. Periodic condition assessments of pavements together with condition predictions are the basis for investment decisions in every pavement management system (PMS). Typical approaches include surveys of distress types every 3–6 years with analysis rating and calculation of condition indices for road safety and/or structural health. Furthermore, advanced PMS prediction models allow a comparison of maintenance alternatives and an optimisation of investment strategies. This paper presents an overview of current survey and rating approaches in Germany, Switzerland and Austria, together with an impact analysis of different methods, utilised deterministic performance functions and condition threshold (trigger) values for all major distress types. The core of this paper is a comparison of common deterministic condition prediction models with discrete stochastic approaches and prediction models based on advanced regression techniques mainly from scientific literature and an innovative stochastic continuous time and continuous state space process (HOFFMANN – Process). All prediction models are applied to real-world data from condition surveys in Austria and the long-term pavement performance Database (USA) at single-section and network level. The paper provides evidence why deterministic prediction approaches are leading to substantial bias in condition distribution and remaining service life as they do not account for the stochastic nature of pavements. Classic Markov-chain approaches do not account for censoring of survey data and neglect changes in transition probabilities with increasing age. Applying common bivariate and multiple regression techniques may also lead to certain bias due to collinearity effects and specification bias. The paper provides mathematical evidence on ways to avoid these shortcomings based on the presented innovative stochastic process leading to a higher reliability in condition assessment, rating and accuracy of condition predictions. The positive prognostic value of HPV-infections in oropharyngeal squamous cell cancer (OSCC) patients has led to the initiation of prospective clinical trials testing the value of treatment de-escalation. It is unclear how to define patients potentially benefiting from de-escalated treatment, whether a positive smoking history impacts survival data and what kind of de-escalation might be best. Here, we investigate the effect of HPV-status, smoking habit and treatment design on overall survival (OS) and progression free survival (PFS) of 126 patients with tonsillar SCC (TSCC) who underwent CO 2 -laser-surgery and risk adapted adjuvant treatment. HPV-DNA-, HPV-mRNA-, and p16 INK4A -expression were analysed and results were correlated to OS and PFS. Factors tested for prognostic value included HPV-status, p16 INK4A -protein expression, therapy and smoking habit. Log rank test and p-values ≤0.05 defined significant differences between groups. The highest accuracy of data with highest significance in this study is given when the HPV-RNA-status is considered. Using p16 INK4A -expression alone or in combination with HPV-DNA-status, would have misclassified 23 and 7 patients, respectively. Smoking fully abrogates the positive impact of HPV-infection in TSCC on survival. Non-smoking HPV-positive TSCC patients show 10-year OS of 100% and 90.9% PFS when treated with adjuvant RCT. The presented data show that high-precision HPV-detection methods are needed, specifically when treatment decisions are based on the results. Furthermore, smoking habit should be included in all studies and clinical trials testing HPV-associated survival. Vesicular stomatitis virus (VSV) release from infected cells is inhibited by the interferon (IFN)-inducible antiviral host cell factor tetherin (BST-2, CD317). However, several viruses encode tetherin antagonists and it is at present unknown whether residual VSV spread in tetherin-positive cells is also promoted by a virus-encoded tetherin antagonist. Here, we show that the viral glycoprotein (VSV-G) antagonizes tetherin in transfected cells, although with reduced efficiency as compared to the HIV-1 Vpu protein. Tetherin antagonism did not involve alteration of tetherin expression and was partially dependent on a GXXXG motif in the transmembrane domain of VSV-G. However, mutation of the GXXXG motif did not modulate tetherin sensitivity of infectious VSV. Deicing agents in winter maintenance are critical in providing safe roads at all times. Sodium chloride (SC) is the most common, efficient and favourable agent but has a drawback of high corrosiveness, thus substantially shortening the service life of metal-based transport infrastructures. This work focuses on corrosion and addresses approaches for the following corrosion test methods that quantify the mass loss of metals for different deicing agents: standardised neutral salt spray test (ISO 9227), salt solution immersion test (ASTM G31-72) and alternate immersion test (ISO 11130). A wide range of different deicing agents with and without corrosion inhibitors are tested on unalloyed steel, and an analytical and visual comparison is made. Results reveal a substantially reduced corrosion effect at reasonable costs by selecting the appropriate deicing agent with or without corrosion inhibitors. The large scale of the Ebola virus disease (EVD) outbreak in West Africa in 2013-2016 raised the question whether the host cell interactions of the responsible Ebola virus (EBOV) strain differed from those of other ebolaviruses. We previously reported that the glycoprotein (GP) of the virus circulating in West Africa in 2014 (EBOV2014) exhibited reduced ability to mediate entry into two nonhuman primate (NHP)-derived cell lines relative to the GP of EBOV1976. Here, we investigated the molecular determinants underlying the differential entry efficiency. We found that EBOV2014-GP-driven entry into diverse NHP-derived cell lines, as well as human monocyte-derived macrophages and dendritic cells, was reduced compared to EBOV1976-GP, although entry into most human- and all bat-derived cell lines tested was comparable. Moreover, EBOV2014 replication in NHP but not human cells was diminished relative to EBOV1976, suggesting that reduced cell entry translated into reduced viral spread. Mutagenic analysis of EBOV2014-GP and EBOV1976-GP revealed that an amino acid polymorphism in the receptor-binding domain, A82V, modulated entry efficiency in a cell line-independent manner and did not account for the reduced EBOV2014-GP-driven entry into NHP cells. In contrast, polymorphism T544I, located in the internal fusion loop in the GP2 subunit, was found to be responsible for the entry phenotype. Interferon-induced transmembrane proteins (IFITMs) can inhibit the cellular entry of several enveloped viruses, including simian immunodeficiency virus (SIV). The blockade of SIV by IFITMs is isolate specific, raising the question of which parameters impact sensitivity to IFITM. We show that the virion context in which SIV-Env is presented and the efficiency of virion incorporation determine Env susceptibility to inhibition by IFITMs. Thus, determinants other than the nature of the envelope protein can impact the IFITM sensitivity of viral entry. New World bats have recently been discovered to harbor influenza A virus (FLUAV)-related viruses, termed bat-associated influenza A-like viruses (batFLUAV). The internal proteins of batFLUAV are functional in mammalian cells. In contrast, no biological functionality could be demonstrated for the surface proteins, hemagglutinin (HA)-like (HAL) and neuraminidase (NA)-like (NAL), and these proteins need to be replaced by their human counterparts to allow spread of batFLUAV in human cells. Here, we employed rhabdoviral vectors to study the role of HAL and NAL in viral entry. Vectors pseudotyped with batFLUAV-HAL and -NAL were able to enter bat cells but not cells from other mammalian species. Host cell entry was mediated by HAL and was dependent on prior proteolytic activation of HAL and endosomal low pH. In contrast, sialic acids were dispensable for HAL-driven entry. Finally, the type II transmembrane serine protease TMPRSS2 was able to activate HAL for cell entry indicating that batFLUAV can utilize human proteases for HAL activation. Collectively, these results identify viral and cellular factors governing host cell entry driven by batFLUAV surface proteins. They suggest that the absence of a functional receptor precludes entry of batFLUAV into human cells while other prerequisites for entry, HAL activation and protonation, are met in target cells of human origin. Ebola and marburgviruses, members of the family Filoviridae, can cause severe hemorrhagic fever in humans. The ongoing Ebola virus (EBOV) disease epidemic in Western Africa claimed more than 11,300 lives and was associated with secondary cases outside Africa, demonstrating that filoviruses pose a global health threat. Bats constitute an important natural reservoir of filoviruses, including viruses of the recently identified Cuevavirus genus within the Filoviridae family. However, the interactions of filoviruses with bat cells are incompletely understood. Here, we investigated whether filoviruses employ different strategies to enter human and bat cells. For this, we examined host cell entry driven by glycoproteins (GP) from all filovirus species into cell lines of human and fruit bat origin. We show that all GPs were able to mediate entry into human and most fruit bat cell lines with roughly comparable efficiency. In contrast, the efficiency of entry into the cell line EidNi/41 derived from a straw-colored fruit bat varied markedly between the GPs of different filovirus species. Furthermore, inhibition studies demonstrated that filoviruses employ the same host cell factors for entry into human, non-human primate and fruit bat cell lines, including cysteine proteases, two pore channels and NPC1 (Niemann-Pick C1 molecule). Finally, processing of GP by furin and the presence of the mucin-like domain in GP were dispensable for entry into both human and bat cell lines. Collectively, these results show that filoviruses rely on the same host cell factors for entry into human and fruit bat cells, although the efficiency of the usage of these factors might differ between filovirus species. A bat virus with high phylogenetic relatedness to human mumps virus (MuV) was identified recently at the nucleic acid level. We analyzed the functional activities of the hemagglutinin-neuraminidase (HN) and the fusion (F) proteins of the bat virus (batMuV) and compared them to the respective proteins of a human isolate. Transfected cells expressing the F and HN proteins of batMuV were recognized by antibodies directed against these proteins of human MuV, indicating that both viruses are serologically related. Fusion, hemadsorption, and neuraminidase activities were demonstrated for batMuV, and either bat-derived protein could substitute for its human MuV counterpart in inducing syncytium formation when coexpressed in different mammalian cell lines, including chiropteran cells. Cells expressing batMuV glycoproteins were shown to have lower neuraminidase activity. The syncytia were smaller, and they were present in lower numbers than those observed after coexpression of the corresponding glycoproteins of a clinical isolate of MuV (hMuV). The phenotypic differences in the neuraminidase and fusion activity between the glycoproteins of batMuV and hMuV are explained by differences in the expression level of the HN and F proteins of the two viruses. In the case of the F protein, analysis of chimeric proteins revealed that the signal peptide of the bat MuV fusion protein is responsible for the lower surface expression. These results indicate that the surface glycoproteins of batMuV are serologically and functionally related to those of hMuV, raising the possibility of bats as a reservoir for interspecies transmission. The viscosity, conductivity, proton T 1 relaxation, and the self-diffusion coefficients of cation, anion and water are presented for the binary system water–1-butyl-3-methylimidazolium methanesulfonate ([C 4 mim][MeSO 3 ]) for the temperature and composition ranges of (298.15–343.14) K and ionic liquid mole fractions 0 < x IL < 0.5. The temperature dependence of these properties at fixed compositions was analyzed to obtain Arrhenius activation energies, which were found to be linearly dependent to x IL for x IL greater than about 0.15 for the self-diffusion coefficients, viscosity and proton T 1 relaxation times (except for the water proton T 1 relaxation times). The specific x IL dependencies of the various Arrhenius activation energies are qualitatively different from what was observed in a prior study of the water–1-ethyl-3-methylimidazolium methanesulfonate ([C 2 mim][MeSO 3 ]) binary system [J. Mol. Liq. 160 (2011) 166–179]. In particular, the T 1 relaxation behavior is markedly different und is discussed in detail along with other comparisons between these two water–IL binary systems. Henipaviruses are associated with pteropodid reservoir hosts. The glycoproteins G and F of an African henipavirus (strain M74) have been reported to induce syncytium formation in kidney cells derived from a Hypsignathus monstrosus bat (HypNi/1.1) but not in nonchiropteran BHK-21 and Vero76 cells. Here, we show that syncytia are also induced in two other pteropodid cell lines from Hypsignathus monstrosus and Eidolon helvum bats upon coexpression of the M74 glycoproteins. The G protein was transported to the surface of transfected chiropteran cells, whereas surface expression in the nonchiropteran cells was detectable only in a fraction of cells. In contrast, the G protein of Nipah virus is transported efficiently to the surface of both chiropteran and nonchiropteran cells. Even in chiropteran cells, M74-G was predominantly expressed in the endoplasmic reticulum (ER), as indicated by colocalization with marker proteins. This result is consistent with the finding that all N-glycans of the M74-G proteins are of the mannose-rich type, as indicated by sensitivity to endo H treatment. These data indicate that the surface transport of M74-G is impaired in available cell culture systems, with larger amounts of viral glycoprotein present on chiropteran cells than on nonchiropteran cells. The restricted surface expression of M74-G explains the reduced fusion activity of the glycoproteins of the African henipavirus. The rapid spread of the SARS-CoV-2 Omicron variant suggests that the virus might become globally dominant. Further, the high number of mutations in the viral spike protein raised concerns that the virus might evade antibodies induced by infection or vaccination. Here, we report that the Omicron spike was resistant against most therapeutic antibodies but remained susceptible to inhibition by sotrovimab. Similarly, the Omicron spike evaded neutralization by antibodies from convalescent patients or individuals vaccinated with the BioNTech-Pfizer vaccine (BNT162b2) with 12- to 44-fold higher efficiency than the spike of the Delta variant. Neutralization of the Omicron spike by antibodies induced upon heterologous ChAdOx1 (Astra Zeneca-Oxford)/BNT162b2 vaccination or vaccination with three doses of BNT162b2 was more efficient, but the Omicron spike still evaded neutralization more efficiently than the Delta spike. These findings indicate that most therapeutic antibodies will be ineffective against the Omicron variant and that double immunization with BNT162b2 might not adequately protect against severe disease induced by this variant. Polyethylene glycol (PEG) is an industrial commodity produced for applications foremost in the medial and personal care business. This review focuses on the much less explored application of using PEG as a chemical solvent. This review highlights some of the successful chemical synthesis strategies to illustrate the advantages of using PEG as an environmentally friendly reaction medium. These advantages include its ability to (a) dissolve a wide range of chemicals including mineral salts, (b) serve as a catalyst because of its acid/base functionalities, (c) complex metal cations, and (d) engage in redox chemistry. New developments of combining PEG with other green solvents and/or functionalizing PEGs are covered as well. The present state of physicochemical studies of PEG as a solvent is also provided and clearly shows the need for future research in this area to further promote the effective use of PEG as a medium for chemistry. Growth experiments with macrophytes revealed anomalies in the growth and development of Najas intermedia. Further analyses showed that these anomalies were caused by differences between the male and female plants. To examine the gender-related differences, the results of two field experiments and a phenological monitoring programme were analysed. The field experiments were conducted in four different lakes in Southern Germany. Over the course of 2 years, eight different sediments were used to research the interaction between the gender and the growth of N. intermedia plants. The phenological monitoring programme, on the other hand, was focused on the development of three different populations of N. intermedia in three Southern German lakes. The results of the experiments revealed significant differences in the gender-related development: Depending on the environmental conditions, male plants grew between 20 and 40% faster than female plants. Additionally, the male plants developed flowers earlier in the vegetation period and died before the female plants. As a result, male and female plants coexisted in the same lake only for a short period of time. This might be a strategy to reduce the competitive pressure between the genders and to gain an advantage over rival species. [ABSTRACT FROM AUTHOR]

Bats (Chiroptera) host major human pathogenic viruses including corona-, paramyxo, rhabdo- and filoviruses. We analyzed six different cell lines from either Yinpterochiroptera (including African flying foxes and a rhinolophid bat) or Yangochiroptera (genera Carollia and Tadarida) for susceptibility to infection by different enveloped RNA viruses. None of the cells were sensitive to infection by transmissible gastroenteritis virus (TGEV), a porcine coronavirus, or to infection mediated by the Spike (S) protein of SARS-coronavirus (SARS-CoV) incorporated into pseudotypes based on vesicular stomatitis virus (VSV). The resistance to infection was overcome if cells were transfected to express the respective cellular receptor, porcine aminopeptidase N for TGEV or angiotensin-converting enzyme 2 for SARS-CoV. VSV pseudotypes containing the S proteins of two bat SARS-related CoV (Bg08 and Rp3) were unable to infect any of the six tested bat cell lines. By contrast, viral pseudotypes containing the surface protein GP of Marburg virus from the family Filoviridae infected all six cell lines though at different efficiency. Notably, all cells were sensitive to infection by two paramyxoviruses (Sendai virus and bovine respiratory syncytial virus) and three influenza viruses from different subtypes. These results indicate that bat cells are more resistant to infection by coronaviruses than to infection by paramyxoviruses, filoviruses and influenza viruses. Furthermore, these results show a receptor-dependent restriction of the infection of bat cells by CoV. The implications for the isolation of coronaviruses from bats are discussed. Abstract: It has been proposed that p16INK4A qualifies as a surrogate marker for viral oncogene activity in head and neck cancer (HNSCC). By analyzing 78 HNSCC we sought to validate the accuracy of p16INK4A as a reliable marker of active HPV infections in HNSCC. To this end we determined HPV DNA (HPVD) and E6*I mRNA (HPVR) expression status and correlated these results with p16INK4A staining. In tonsillar SCC 12/20 were HPVD+ and 12/12 of these showed active HPV infections whereas in non-tonsillar SCC 10/58 were HPVD+ and 5/10 showed active HPV infections. Thus, we prove about 8% of non-tonsillar SCC to be also correlated with HPV-associated carcinogenesis. Strikingly, 3/14 (21.4%) of tonsillar and non-tonsillar HPVD+/HPVR+ cases did not show p16INK4A overexpression and these cases would have been missed when applying initial p16INK4A staining only. However, in 13 cases negative for HPV, DNA p16INK4A was overexpressed. In conclusion, our data confirm tonsillar SCC to be predominantly but not only associated with active HPV infections. Furthermore, our data show that p16INK4A overexpression is not evident in a subgroup of HNSCC with active HPV infection. Definitive HPV data should therefore be utilized in diagnostics and treatment modalities of HPV positive and HPV negative HNSCC patients, resulting in a paradigm shift regarding these obviously different tumor entities. The ferritic steel 16Mo3 is commonly used for heat exchangers and steam generators. The temperature loading conditions for these applications range typically from 200 to 500°C. For a basic characterization of the steel 16Mo3, the properties were determined by means of tensile tests with different strain rates. At 200 and 300°C, a negative strain-rate sensitivity was observed which is due to dynamic strain ageing (Portevin-LeChâtelier effect). To characterize the low cycle fatigue (LCF) behavior of this steel, isothermal strain-controlled LCF tests were carried out at 200 and 500°C at different strain amplitudes. The cyclic stress response curves are showing principally different courses at the investigated temperatures. The stress relaxation behavior was obtained by cyclic load enhancement tests at various strain amplitudes with dwell times of 30 and 120 s. [ABSTRACT FROM AUTHOR]

Infection with Ebola virus (EBOV) causes hemorrhagic fever in humans with high case-fatality rates. The EBOV-glycoprotein (EBOV-GP) facilitates viral entry and promotes viral release from human cells. African fruit bats are believed not to develop disease upon EBOV infection and have been proposed as a natural reservoir of EBOV. We compared EBOV-GP interactions with human cells and cells from African fruit bats. We found that susceptibility to EBOV-GP-dependent infection was not limited to bat cells from potential reservoir species, and we observed that GP displayed similar biological properties in human and bat cells. The only exception was GP localization, which was to a greater extent intracellular in bat cells as compared to human cells. Collectively, our results suggest that GP interactions with fruit bat and human cells are similar and do not limit EBOV tropism for certain bat species. [ABSTRACT FROM AUTHOR]

Abstract: Autoimmune responses to heterogeneous nuclear ribonucleproteins (hnRNP) occur in many systemic autoimmune diseases, particularly in patients with rheumatoid arthritis (RA) and systemic lupus erythematosus. In RA, humoral and/or cellular autoimmunity to hnRNP-A2/B1 is the most prominent anti-nuclear reactivity, being detectable in more than 50% of patients. However, its pathogenic role has not been fully elucidated yet. Here, we report that splenocytes from rats with pristane-induced arthritis transfer disease after in vitro restimulation with hnRNP-A/B antigens. Remarkably, disease transfer can be blocked by nuclease treatment of hnRNPs and is also achieved with splenocytes stimulated with hnRNP-A/B associated DNA or RNA oligonucleotides (ON) alone. Induction of proinflammatory cytokines in splenocytes stimulated with hnRNP-A/Bs or ONs involves Toll-like receptors (TLR) 7 and 9 but not TLR3. Furthermore, although T cells are the main mediators of disease transfer they require restimulation with TLR-activated antigen-presenting cells such as macrophages in order to become arthritogenic. Thus, the autoantigenic properties of hnRNPs appear to be mediated by their associated nucleic acids binding to TLR7 and 9. Our data explain the specific selection of hnRNP-A2/B1 as autoantigen in RA and reveal the requirement of interaction between innate and adaptive immunity to initiate and drive inflammation in autoimmune arthritis. A series of butadiyne-linked zinc porphyrin oligomers, with one, two, three, and four porphyrin units and lengths of up to 75 A, have been spin-labeled at both ends with stable nitroxide TEMPO radicals. The pulsed EPA technique of double electron electron resonance (DEER) was used to probe the distribution of intramolecular end-to-end distances, under a range of conditions. DEER measurements were carried out at 50 K in two types of dilute solution glasses: deutero-toluene (with 10% deutero-pyridine) and deutero- o-terphenyl (with 5% 4-benzyl pyndine). The complexes of the porphyrin oligomers with monodentate ligands (pyridine or 4-benzyl pyridine) principally adopt linear conformations. Nonlinear conformations are less populated in the lower glass-transition temperature solvent. When the oligomers bind star-shaped multidentate ligands, they are forced to bend into nonlinear geometries, and the experimental end-to-end distances for these complexes match those from molecular mechanics calculations. Our results show that porphyrin-based molecular wires are shape-persistent, and yet that their shapes can deformed by binding to multivalent ligands. Self-assembled ladder-shaped 2:2 complexes were also investigated to illustrate the scope of DEER measurements for providing structural information on synthetic noncovalent nanostructures. Rheumatoid arthritis (RA) is a severe autoimmune disorder of unknown etiology. Major autoantigens include immunoglobulin G (which is targeted by rheumatoid factor), citrullinated proteins, and the heterogeneous nuclear ribonucleoprotein (hnRNP) A2. To obtain more insight into the pathogenic role of arthritogenic autoantigens, we studied autoimmune responses in two animal models of RA, which are independent of immunization with exogenous antigens, namely human tumor necrosis factor-α (TNF-α)-transgenic (hTNFtg) mice and rats with pristane-induced arthritis (PIA). Serologic and cellular studies revealed autoantibodies to hnRNP-A2 in these animals and pronounced T-cell reactivity to hnRNP-A2 and the presence of rheumatoid factor in PIA, while citrullinated antigens were not targeted. Furthermore, hnRNP-A2 was found to be highly overexpressed in the joints of mice and rats affected by arthritis. Thus, unspecific proinflammatory stimuli, such as TNF-α or pristane, may induce pathogenic autoimmune reactions that may drive an inflammatory process, leading to the development of erosive arthritis.
