Engineering nanocarriers with low cellular interaction during systemic circulation, but with high uptake in targeted cells, is a great challenge and is further complicated by the BBB. As a first step in obtaining such a delivery system, this study aims at designing a lipid nanoparticle (LNP) able to efficiently encapsulate siRNA by a combination of titratable cationic lipids. The targeted delivery is obtained through the design of a two-stage system where the first step is conjugation of angiopep to the surface of the LNP for targeting the low-density lipoprotein receptor-related protein-1 expressed on the BBB. Second, the positively charged LNPs are masked with a negatively charged PEGylated (poly(ethylene glycol)) cleavable lipopeptide, which contains a recognition sequence for matrix metalloproteinases (MMPs), a class of enzymes often expressed in the tumor microenvironment and inflammatory BBB conditions. Proteolytic cleavage induces PEG release, including the release of four glutamic acid residues, providing a charge switch that triggers a shift of the LNP charge from weakly negative to positive, thus favoring cellular endocytosis and release of siRNA for high silencing efficiency. This work describes the development of this two-stage nanocarrier-system and evaluates the performance in brain endothelial and glioblastoma cells with respect to uptake and gene silencing efficiency. The ability of activation by MMP-triggered dePEGylation and charge shift is demonstrated to substantially increase the uptake and the silencing efficiency of the LNPs.Keywords: matrix metalloproteinase, cleavable PEG-lipid, gene therapy, BBB, angiopep, nanocarrier

The strain relief and dislocation motion in III-nitride films that are grown on step-free 4H-SiC mesas are studied. The results show that the nucleation of threading dislocations in the absence of interfacial steps is an extremely difficult process, which proves that the strain relief for such III-nitride films is not dominated by the greater excess stress driving dislocation tilt.

Research has been conducted on soluble reduced iron-sulfur clusters with nuclearites. The synthesis of these clusters which possess tertiary phosphine ligation and are based on Fe (sub)4 S (sub)4 cubane-type structural motif is described.

Research has been conducted on the site-differentiated cyanide-substituted hexanuclear rhenium(III) selenide clusters. The preparation of these complexes has been carried out via the heterogeneous reactions of iodo clusters with AgCN in chloroform and the details are reported.

The collective XAS results for HC4H2-[Fe4S4]-Ni and HC5H-[Fe4S4]-M demonstrate the presence of an Fe4S4 cluster and support the existence of the distorted square-planar coordination units [Ni (super II) (S.Cys)(N.His)(Sub 3)] and [Ni(super II)(S.Cys)(Sub 2)(N.His)(sub 2)] in the HC4H2 and HC5H metallopeptides, respectively. The physical processes and the chemical reactions involved in magnetization transfer between water and large proteins, such as collagen, in bovine Achilles tendon are studied. The acquisition of water signal, which originates from the protein magnetization in the DQF-magnetization transfer (MT) technique, give rise to the possibility of creating images that are related directly to the amount of protein and its intramolecular dipolar interactions.

Research has been conducted on the nitrogenase clusters. The preparation of these clusters in different oxidation states, the study of the ground-state electronic properties of these states and the hypothesis that the hyigher-nuclearity clusters could be produced from the single-cubane species are discussed.

Research has been conducted on the structurally characterized tungstoenzymes containing mononuclear active sites. The stable coordination units of bis(dithiolene)tungsten (IV,V,VI) complexes related to the enzyme sites have been examines via the exploratory synthesis and the results indicate that tungsten(IV) complexes which have been obtained via substitution are carbonyl displacement products.

Analysis of electron delocalization and spin ordering in the mixed-valence binuclear iron complexes shows a correlation between these two electronic properties and their dependence on the metal-metal separation. Data also point out fast electron hopping on the Mossbauer time scale.
