Describes the essential features and results of an Extended General Physics course offered as an alternative to the general physics sequence at Rutgers University. Finds that by integrating many proven-successful instructional techniques and structuring the time that students spend in the course, the program has fostered the success of at-risk as well as average students. The chorio-amniotic membrane, in avian embryos, begins as a fold that rises like a parabolic wave, anterior to the head, then expands and moves posteriorly, to envelop the embryo. The mechanism by which this process occurs is not well understood. Nile Blue sulfate labeling of cells on the membrane allowed general membrane movements to be followed. Results suggested that the membrane expands both from tractoring of the fold and also from fusion of the folds that arise on either side of the embryo (the sides of the parabola). We found, by immunochemistry, that the leading edge of the chorio-amniotic fold and the region of fusion stained intensely for Phospho Inositide 3 kinase (PI3 kinase), an enzyme important in [Ca.sup.2+] signaling pathways. PI3 kinase is known to affect cell division, cytoskeletal rearrangements and cell-cell adhesion. We inhibited PI3 kinase by applying the toxin, wortmannin, to developing embryos. Regardless of the application procedure (bath or wortmannin-soaked beads) the posterior progression of the fold stopped and the membrane ballooned away from the embryo. We are currently investigating the possibility that PI3 kinase regulates cell shape and/or adhesive properties of cells along the leading edge of the chorio-amniotic fold.

At the beginning of the COVID-19 pandemic, the Georgia Institute of Technology made the decision to keep the university doors open for on-campus attendance. To manage COVID-19 infection rates, internal resources were applied to develop and implement a mass asymptomatic surveillance program. The objective was to identify infections early for proper follow-on verification testing, contact tracing, and quarantine/isolation as needed. Program success depended on frequent and voluntary sample collection from over 40,000 students, faculty, and staff personnel. At that time, the nasopharyngeal (NP) swab, not saliva, was the main accepted sample type for COVID-19 testing. However, due to collection discomfort and the inability to be self-collected, the NP swab was not feasible for voluntary and frequent self-collection. Therefore, saliva was selected as the clinical sample type and validated. A saliva collection kit and a sample processing and analysis workflow were developed. The results of a clinical sample-type comparison study between co-collected and matched NP swabs and saliva samples showed 96.7% positive agreement and 100% negative agreement. During the Fall 2020 and Spring 2021 semesters, 319,988 samples were collected and tested. The program resulted in maintaining a low overall mean positivity rate of 0.78% and 0.54% for the Fall 2020 and Spring 2021 semesters, respectively. For this high-throughput asymptomatic COVID-19 screening application, saliva was an exceptionally good sample type.

Background: Six months into the COVID-19 pandemic, college campuses faced uncertainty regarding the likely prevalence and spread of disease, necessitating large-scale testing to help guide policy following re-entry., Methods: A SARS-CoV-2 testing program combining pooled saliva sample surveillance leading to diagnosis and intervention surveyed over 112,000 samples from 18,029 students, staff and faculty, as part of integrative efforts to mitigate transmission at the Georgia Institute of Technology in Fall 2020., Results: Cumulatively, we confirmed 1,508 individuals diagnostically, 62% of these through the surveillance program and the remainder through diagnostic tests of symptomatic individuals administered on or off campus. The total strategy, including intensification of testing given case clusters early in the semester, was associated with reduced transmission following rapid case increases upon entry in Fall semester in August 2020, again in early November 2020, and upon re-entry for Spring semester in January 2021. During the Fall semester daily asymptomatic test positivity initially peaked at 4.1% but fell below 0.5% by mid-semester, averaging 0.84% across the Fall semester, with similar levels of control in Spring 2021., Conclusions: Owing to broad adoption by the campus community, we estimate that the program protected higher risk staff and faculty while allowing some normalization of education and research activities., Competing Interests: The authors report no conflicts of interest., (Copyright © 2021 The Author(s). Background: Many studies of animal movement have focused on directed versus area-restricted movement, which rely on correlations between step-length and turn-angles and on stationarity through time to define behavioral states. Although these approaches might apply well to grazing in patchy landscapes, species that either feed for short periods on large, concentrated food sources or cache food exhibit movements that are difficult to model using the traditional metrics of turn-angle and step-length alone., Results: We used GPS telemetry collected from a prey-caching predator, the cougar ( Puma concolor, Linnaeus ), to test whether combining metrics of site recursion, spatiotemporal clustering, speed, and turning into an index of movement using partial sums, improves the ability to identify caching behavior. The index was used to identify changes in movement characteristics over time and segment paths into behavioral classes. The identification of behaviors from the Path Identification Index (PII) was evaluated using field investigations of cougar activities at GPS locations. We tested for statistical stationarity across behaviors for use of topographic view-sheds. Changes in the frequency and duration of PII were useful for identifying seasonal activities such as migration, gestation, and denning. The comparison of field investigations of cougar activities to behavioral PII classes resulted in an overall classification accuracy of 81%., Conclusions: Changes in behaviors were reflected in cougars' use of topographic view-sheds, resulting in statistical nonstationarity over time, and revealed important aspects of hunting behavior. Incorporating metrics of site recursion and spatiotemporal clustering revealed the temporal structure in movements of a caching forager. The movement index PII, shows promise for identifying behaviors in species that frequently return to specific locations such as food caches, watering holes, or dens, and highlights the potential role memory and cognitive abilities play in determining animal movements.

Landscape genetic studies based on neutral genetic markers have contributed to our understanding of the influence of landscape composition and configuration on gene flow and genetic variation. However, the potential for species to adapt to changing landscapes will depend on how natural selection influences adaptive genetic variation. We demonstrate how landscape resistance models can be combined with genetic simulations incorporating natural selection to explore how the spread of adaptive variation is affected by landscape characteristics, using desert bighorn sheep (Ovis canadensis nelsoni) in three differing regions of the southwestern United States as an example. We conducted genetic sampling and least-cost path modeling to optimize landscape resistance models independently for each region, and then simulated the spread of an adaptive allele favored by selection across each region. Optimized landscape resistance models differed between regions with respect to landscape variables included and their relationships to resistance, but the slope of terrain and the presence of water barriers and major roads had the greatest impacts on gene flow. Genetic simulations showed that differences among landscapes strongly influenced spread of adaptive genetic variation, with faster spread (1) in landscapes with more continuously distributed habitat and (2) when a pre-existing allele (i.e., standing genetic variation) rather than a novel allele (i.e., mutation) served as the source of adaptive genetic variation. The combination of landscape resistance models and genetic simulations has broad conservation applications and can facilitate comparisons of adaptive potential within and between landscapes.

Background: Approximately 0.05% of Emergency Department visits in the United States are related to supraventricular tachycardia (SVT). The majority of patients convert with an atrioventricular nodal blocking medication., Case Report: We report a case of SVT that converted after administration of calcium gluconate after failing conversion with adenosine., Conclusion: Conversion to normal sinus rhythm resulted after administration of i.v. calcium in our patient. The present study explored the delay discounting of future and past monetary rewards by pathological gamblers. Using a multiple baseline design, following repeated exposure to choices between smaller immediate and larger delayed consequences, participants completed a relational responding task that attempted to alter the psychological functions of irrelevant stimuli and to affect subsequent delay discounting. Results support previous literature on the discounting of delayed consequences by pathological gamblers, illustrate that the discounting of past rewards occurs in a similar fashion to the well-documented literature on the discounting of future rewards, and that magnitude of discounting can be altered.

Trunk neural crest cells primarily form neurons, nerve supportive cells of the peripheral nervous system and melanocytes. We are interested in signal transduction pathways that affect the production of peripheral neurons or melanocytes. Quail neural crest cell cultures were treated with a variety of drugs that affect components of protein kinase A- (PKA-), protein kinase C- (PKC-) and inositol-3-phosphate- (I3P-) dependent pathways. Forskolin, a drug that increases cAMP levels, augmented melanocyte populations and reduced neuronal populations in our cultures. H8 and H89, two drugs that inhibit PKA, reduced melanocyte populations well below control levels. Down regulation of PKC with a phorbol ester, PMA, or with calphostin C inhibited neurogenesis. PMA also enhanced melanogenesis. Increasing intracellular calcium levels (with A23187 or thapsigargin) resulted in cultures with few melanocytes but many neurons, compared to untreated controls. An antagonist of the I3P pathway, wortmannin, prevented the appearance of neurons but did not affect melanocyte populations. In summary, molecules that altered the PKA-dependent pathway affected melanogenesis. Manipulations of the I3P and/or PKC-dependent pathways influenced neurogenesis. The relationship between health care providers and pharmaceutical companies and other commercial interests is ethically complex. The common practice of gift giving takes many forms including free samples, sponsorship of medical education, loan of equipment, and gifts ranging from those of nominal value such as pens to more valuable gifts such as golf outings or dinners. Gift giving is a practice that serves both the recipient and the giver, but, in the medical setting, it raises the question of whether this is to the detriment of patient care. Although health care professionals may believe they are able to ignore influence from commercial interests, human judgment research indicates that decision-makers are generally unaware of biases affecting their decisions. This is an issue of organizational ethics as well. Institutions that allow commercial interests to give some form of gift are allowing the appearance of bias as well as placing the burden of avoiding bias on the individual rather than on the institution. Conflict-of-interest analysis indicates that best practice is to limit or eliminate the influence of commercial interests, ensuring that professionals are better able to exercise their independent objective judgment.

The Rieske 2Fe-2S protein is a distinguishing subunit of the photosynthetic electron transport cytochrome b6f complex in chloroplast and cyanobacterial thylakoid membranes. We have constructed plasmids for overproduction in Escherichia coli of fusion, full-length, and truncated forms of the Rieske (PetC) protein from the cyanobacterium Nostoc sp. PCC 7906. A glutathione S-transferase/Rieske fusion protein was used to prepare specific chicken egg-yolk antibodies against the Rieske protein. Expression of the nonfusion petC gene in a T7 RNA polymerase promoter vector produced copious quantities of the full-length Rieske protein predominantly as inclusion bodies. The highly enriched, Rieske protein from inclusion bodies has been denatured in guanidine hydrochloride and refolded and the characteristic 2Fe-2S cluster reconstituted in vitro by incubation with iron and sulfide under reducing conditions. Purification by chromatography on Whatman DE52 cellulose and ultrafiltration through a 30000 molecular weight cutoff membrane yielded pure and predominantly monomeric Rieske protein. Reconstituted Rieske preparations showed intense and highly characteristic gx = 1.74, gy = 1.89, and gz = 2.03 "Rieske-type" electron paramagnetic resonance signals at 15 K. Two methods of reconstitution yielded Rieske preparations in which 20-60% of the protein contained 2Fe-2S clusters as determined by EPR spin quantitation. The reconstituted Rieske protein was soluble and stable at 4 degrees C in buffers containing nonionic detergents and showed a redox midpoint potential of +321 mV at pH 7.0 as determined by optical circular dichroism (CD) spectroscopy. These data demonstrate the in vitro restoration of a Cys and His liganded 2Fe-2S cluster and provide the basis for mutational and structural analysis of a PetC Rieske protein of oxygenic photosynthesis.

In the embryos of glossiphoniid leeches, as in many annelids, cytoplasmic reorganization prior to first cleavage generates domains of yolk-deficient cytoplasm (called teloplasm) that are sequestered during the first three cell divisions to the D' macromere. Subsequently, the D' macromere generates a set of embryonic stem cells (teloblasts) that are the progenitors of the definitive segmental tissues. The hypothesis that fate-determining substances are localized within the teloplasm and segregated to the D' macromere during cleavage is supported by experiments in which a redistribution of yolk-defcient cytoplasm changes the fate of blastomeres that inherit it (Astrow et al. 1987; Devries 1973; Nelson and Weisblat 1992). As a step toward identifying fate-determining factors in teloplasm, we describe the distribution of polyadenylated RNAs (polyA+ RNA) in the early embryo of the leech, Helobdella triserialis, as inferred from in situ hybridization using tritiated polyuridylic acid (3H-polyU). Our results indicate that polyA+ RNA colocalizes with teloplasm during cytoplasmic rearrangements resulting in teloplasm formation, and that it remains concentrated in the teloplasm during the cell divisions and a second cytoplasmic rearrangement during early embryogenesis. Lesser amounts of polyA+ RNA appear to be localized in cortical cytoplasm at most stages.

In embryos of the glossiphoniid leech, Helobdella triserialis, as in many annelids, cytoplasmic reorganization prior to first cleavage generates distinct animal and vegetal domains of yolk-deficient cytoplasm, called teloplasm. Both domains are sequestered to the D' macromere, progenitor of the definitive segmental tissues, during the first three rounds of cell division. And it has been believed that during the fourth round of cell division, the obliquely equatorial cleavage of macromere D' cleanly segregates animal teloplasm into an ectodermal precursor, cell DNOPQ, and vegetal teloplasm into a mesodermal precursor, cell DM. But here we report a hitherto unobserved cytoplasmic rearrangement between the second and the fourth divisions that seems to mix the animal and vegetal domains of teloplasm. The newly observed rearrangement consists of the movement of vegetal teloplasm toward the animal pole of cell D' between the second and the fourth cell divisions. Animal and vegetal teloplasms form a single pool of teloplasm in cell D' which is then divided between DM and DNOPQ at the fourth division. The movement of teloplasm was inferred by examination of embryos fixed and sectioned between the second and the fourth rounds of cleavage and was confirmed in living embryos microinjected with rhodamine 123, a fluorescent mitochondrial stain.

Fertilized eggs of the leech Helobdella triserialis undergo a cytoplasmic reorganization which generates domains of nonyolky cytoplasm, called teloplasm, at the animal and vegetal poles. The segregation of teloplasm to one cell of the eight-cell embryo is responsible for a unique developmental fate of that cell, i.e., to give rise to segmental ectoderm and mesoderm. We have studied the cytoplasmic movements that generate teloplasm using time-lapse video microscopy; the formation and migration of rings of nonyolky cytoplasm were visualized using transmitted light, while the movements of mitochondria into these rings were monitored with epifluorescence after labeling embryos with rhodamine 123, a fluorescent mitochondrial dye. To examine the likelihood that cytoskeletal elements play a role in the mechanism of teloplasm formation in Helobdella, we examined the distribution of microtubules and microfilaments during the first cell cycle by indirect immunofluorescence and rhodamine-phalloidin labeling, respectively. The cortex of the early embryo contained a network of microtubules many of which were oriented parallel to the cell surface. As teloplasm formation ensued, microtubule networks became concentrated in the animal and the vegetal cortex relative to the equatorial cortex. More extensive microtubule arrays were found within the rings of teloplasm. Actin filaments appeared in the form of narrow rings in the cortex, but these varied apparently randomly from embryo to embryo in terms of number, size, and position. The role of microtubules and microfilaments in teloplasm formation was tested using depolymerizing agents. Teloplasm formation was blocked by microtubule inhibitors, but not by microfilament inhibitors. These results differ significantly from those obtained in embryos of the oligochaete Tubifex hattai, suggesting that the presumably homologous cytoplasmic reorganizations seen in these two annelids have different cytoskeletal dependencies.

In the normal development of glossiphoniid leech embryos, cytoplasmic reorganization prior to the first cleavage generates visibly distinct domains of yolk-deficient cytoplasm, called teloplasm. During an ensuing series of stereotyped and unequal cell divisions, teloplasm is segregated primarily into cell CD of the two-cell stage and then into cell D of the four-cell and eight-cell stages. The subsequent fate of cell D is also unique in that it alone undergoes further cleavages which generate five bilateral pairs of embryonic stem cells, the mesodermal (M) and ectodermal (N, O/P, O/P, and Q) teloblasts. Here we report studies on the effects of centrifugation on cleavage pattern and protein composition of individual blastomeres of the leech Helobdella triserialis. Centrifugation partially stratifies the cytoplasm of each cell, generating a layer of clear cytoplasm in cell CD derived largely from teloplasm. After centrifuging embryos at the two-cell stage, clear cytoplasm present in cell CD and normally inherited by cell D is redistributed and can be inherited by both cells C and D at the second cleavage. The developmental fates of cells C and D in centrifuged embryos correlate with the amount of clear cytoplasm they receive. In particular, when clear cytoplasm has been distributed roughly equally between the two cells, both cell C and cell D undergo further cleavages resembling the pattern of divisions normally associated with cell D. Likewise, non-yolk-associated proteins, normally found in higher quantities in cell D than in cell C, appear evenly disbursed between the two cells under conditions which induce this fate change. These results are consistent with the idea that the fates of cells C and D are influenced by the distribution or cellular localization of cytoplasmic components.

The acetylcholine receptor (AChR) from Torpedo marmorata electric organ exists in a light form (α2βγδ) of apparent molecular weight 250,000. The association of two light forms via an intermolecular δ-δ disulfide bridge results in the AChR heavy form. In adult Torpedo electric organ extracts the heavy form constitutes about 70% of the AChR. We report that, in contrast, embryonic electric organ extracts contain only about 30% of the heavy form, the rest being light form. In addition, amongst a library of 38 monoclonal antibodies (mAbs), all of those that distinguished between embryonic and adult AChR did so only because they precipitated the heavy form of the AChR better than the light form., (Copyright © 1984.
