The 25th Annual Western Canadian Gastrointestinal Cancer Consensus Conference (WCGCCC) was held in Winnipeg, Manitoba, on 26–27 October 2023. The WCGCCC is an interactive multidisciplinary conference that was attended by healthcare professionals from across Western Canada (British Columbia, Alberta, and Manitoba) who are involved in the care of patients with gastrointestinal cancer. Surgical, medical, and radiation oncologists; pathologists; oncology nurses; pharmacists; and a family physician in oncology (FPO) participated in presentation and discussion sessions for the purpose of developing the recommendations presented here. This consensus statement addresses current issues in the management of gastroesophageal cancers.

Abstract The use of microbial inoculant is a promising strategy to improve plant health, but their efficiency often faces challenges due to difficulties in successful microbial colonization in soil environments. To this end, the application of biostimulation products derived from microbes is expected to resolve these barriers via direct interactions with plants or soil pathogens. However, their effectiveness and mechanisms for promoting plant growth and disease resistance remain elusive. In this study, we showed that root irrigation with the extracts of Streptomyces ahygroscopicus strain 769 (S769) solid fermentation products significantly reduced watermelon Fusarium wilt disease incidence by 30% and increased the plant biomass by 150% at a fruiting stage in a continuous cropping field. S769 treatment led to substantial changes in both bacterial and fungal community compositions, and induced a highly interconnected microbial association network in the rhizosphere. The root transcriptome analysis further suggested that S769 treatment significantly improved the expression of the MAPK signalling pathway, plant hormone signal transduction and plant–pathogen interactions, particular those genes related to PR‐1 and ethylene, as well as genes associated with auxin production and reception. Together, our study provides mechanistic and empirical evidences for the biostimulation products benefiting plant health through coordinating plant and rhizosphere microbiome interaction.

Natural and geological environmental conditions have an important impact on the planning and construction of sponge cities. This paper analyzes geological factors that influence the usage of natural sponge bodies, taking the Shuanghe lake district of Zhengzhou airport zone as an example. An evaluation system with seven factors has been established and the weights of these factors are determined using the analytic hierarchy process (AHP) method. Overlay analysis is then carried out on all factors using GIS to evaluate the geological suitability of the construction of the sponge city. The results show that geologically suitable area for city construction in Shuanghe lake district accounts for 12.3%, relatively suitable area accounts for 76.1%, and relatively unsuitable area accounts for 11.6%. For suitable and relatively suitable areas, we should make full use of the advantages of surface infiltration, vadose zone transportation and aquifer storage to build a sponge city infrastructure with geological engineering as the main component, supplemented by engineering measures such as surface water storage and drainage, and jointly establish a sustainable urban hydrological cycle. For less suitable areas, artificial rain and flood control works, such as roof garden, should be considered. The findings of this paper can serve as an important reference for sponge city planning and construction not only in the research area but also in other regions with similar geological conditions.

Objectives: Long-term use of evidence-based antiplatelet therapy is recommended for management of stable coronary artery disease (SCAD). However, non-adherence to antiplatelet drugs is common in older patients. This study aimed to evaluate the incidence and impact of antiplatelet therapy cessation on clinical outcomes of older patients with SCAD.Methods: A total of 351 consecutive eligible very older patients (≥80 years) with SCAD from the PLA General Hospital were included. Baseline demographics, clinical characteristics, and clinical outcomes were collected during follow-up. Patients were divided into cessation group and standard group based on whether discontinuing of antiplatelet drugs. The primary outcome was major adverse cardiovascular events (MACE) and secondary outcomes were minor bleeding and all-cause mortality.Results: A total of 351 participants, with a mean age of 91.76 ± 5.01 years old (range 80–106 years) were included in statistical analysis. The antiplatelet drug cessation rate was 60.1%. There were 211 patients in cessation group and 140 patients in standard group. During a median follow-up of 98.6 months, the primary outcome of MACE occurred in 155 patients (73.5%) in the cessation group and 84 patients (60.0%) in the standard group (HR = 1.476, 95% CI:1.124-1.938, p = 0.005). Cessation of antiplatelet drugs increased the rates of angina (HR = 1.724, 95% CI:1.211-2.453, p = 0.002) and non-fatal MI (HR = 1.569, 95% CI:1.093-2.251, p = 0.014). The secondary outcomes of minor bleeding and all-cause mortality were similar between the two groups.Conclusion: Among very older patients with SCAD, antiplatelet therapy cessation significantly increased the risk of MACE, and continuous antiplatelet drug therapy didn’t increase the risk of minor bleeding.

Simplicillium species are widely distributed with a broad spectrum of hosts and substrates. Generally, these species are entomopathogenic or mycoparasitic. Notably, some isolates of Simplicillium lanosoniveum and Simplicillium obclavatum were obtained from human tissues. In this study, two fungi were isolated from the annular itchy patch of infected skin of a 46-year-old man with diabetes mellitus. Based on a combination of morphological characteristics and phylogenetic analysis, a novel species, Simplicillium sinense, was introduced herein. It morphologically differs from the remaining Simplicillium in the size of phialides and conidia. Additionally, it grows slowly on YPD at 37°C. Antimicrobial susceptibility testing presented that this fungus is resistant to most azole antifungals. Therefore, the diagnosis of tinea faciei was made, and after 2 weeks of being treated with oral terbinafine (250 mg, once a day) and topical terbinafine cream for 1 month, the rash was mainly resolved and no recurrence happened after 6 months of follow-up. Herein, Simplicillium sinense was introduced as a new fungal taxon. Meanwhile, a case of superficial infection caused by S. sinense was reported. So far, it is the third Simplicillium species obtained from human tissue. Meanwhile, terbinafine is recommended as the first-line antifungal treatment against Simplicillium infection.

In previous studies, researchers have used singing to treat respiratory function in patients with spinal cord injury. However, few studies have examined the way in which vocal training affects respiratory neural plasticity in patients with spinal cord injury. Vocal respiratory training (VRT) is a type of vocal muscle-related treatment that is often a component of music therapy (MT) and focuses on strengthening respiratory muscles and improving lung function. In this randomized controlled study, we analyzed the therapeutic effects of VRT on respiratory dysfunction at 3 months after cervical spinal cord injury. Of an initial group of 37 patients, 26 completed the music therapy intervention, which comprised five 30-minute sessions per week for 12 weeks. The intervention group (n = 13) received VRT training delivered by professional certified music therapists. The control group (n = 13) received respiratory physical therapy delivered by professional physical therapists. Compared with the control group, we observed a substantial increase in respiratory function in the intervention group after the 12-week intervention. Further, the nerve fiber bundles in the respiratory center in the medulla exhibited a trend towards increased diversification, with an increased number, path length, thickness, and density of nerve fiber bundles. These findings provide strong evidence for the effect of music therapeutic VRT on neural plasticity. This study was approved by the Ethics Committee of China Rehabilitation Research Center (approval No. 2020-013-1) on April 1, 2020, and was registered with the Chinese Clinical Trial Registry (registration No. ChiCTR2000037871) on September 2, 2020.

Compared with the traditional BB84 protocol, the counterfactual quantum key distribution (QKD) does not rely on any signal travelling in the quantum channel, and therefore can present a security advantage where Eve cannot fully access signal. However, the practical system may be damaged in a scenario where the devices are untrusted. In this paper, we analyze the security of counterfactual QKD in untrusted detectors scenario. We show that the requirement to disclose “which detector clicked” has become the main loophole in all counterfactual QKD versions. An eavesdropping scheme which is similar to the memory attack on device-independent QKD could break its security by exploiting detectors' imperfections. We consider two different counterfactual QKD protocols and analyze their security against this major loophole. One is a modified Noh09 protocol, which would be secure in untrusted detectors context. Another is a variant of counterfactual QKD with high efficiency (Phys. Rev. A 104 (2021) 022424) against a series of detectors side-channel attacks as well as against other attacks that exploit detectors imperfections.

BackgroundDiabetic foot disease is a serious complication of diabetes mellitus. Patients with diabetes mellitus have a 25% lifetime risk for developing a foot ulcer, and between 14% and 24% of patients require a major or minor lower limb amputation due to severe gangrene. However, decisions concerning whether to amputate or whether to perform a major or minor lower limb amputation, and how best to determine the amputation plane remain unclear.MethodsTo consolidate the current literature with expert opinion to make recommendations that will guide surgical amputation for patients with diabetic foot ulcers. A total of 23 experts experienced in surgical treatment of patients with diabetic foot ulcers formed an expert consensus panel, and presented the relevant evidence, discussed clinical experiences, and derived consensus statements on surgical amputation for patients with diabetic foot ulcers. Each statement was discussed and revised until a unanimous consensus was achieved.ResultsA total of 16 recommendations for surgical amputation for patients with diabetic foot ulcers were formulated. The experts believe that determination of the amputation plane should be comprehensively evaluated according to a patient's general health status, the degree of injury, and the severity of lower limb vasculopathy. The Wagner grading system and the severity of diabetic lower extremity artery disease are important criteria when determining the degree of amputation. The severity of both diabetic foot infection and systemic underlying diseases are important factors when considering appropriate treatment. Moreover, consideration should also be given to a patient's socioeconomic status. Given the complexities of treating the diabetic foot, relevant issues in which consensus could not be reached will be discussed and revised in future.ConclusionThis expert consensus could be used to guide doctors in clinical practice, and help patients with diabetic foot ulcers gain access to appropriate amputation treatment.

To investigate the potential relationship between Ikaros family genes and skin cutaneous melanoma (SKCM), we undertook a pan-cancer analysis of the transcriptional signature and clinical data of melanoma through multiple databases. First, 10,327 transcriptomic samples from different cancers were included to determine the overall characteristics and clinical prognoses associated with Ikaros gene expression across cancer types. Second, differentially expressed genes analysis, prognostic evaluation, and gene set enrichment analysis were employed to investigate the role of Ikaros (IKZF) genes in SKCM. Third, we evaluated the relationship between Ikaros family genes and SKCM immune infiltrates and verified the findings using the GEO single-cell sequencing dataset. The results show that Ikaros genes were widely expressed among different cancer types with independently similar patterns as follows: 1. IKZF1 and IKZF3, and 2. IKZF2 and IKZF4–5. IKZF2 and IKZF5 were downregulated in the primary tumor, and IKZF1–3 expression decreased significantly as the T-stage or metastasis increased in SKCM. Moreover, high IKZF1–3 expression was associated with better overall survival, disease-specific survival, and progression-free interval. IKZF3 is an independent prognostic factor of SKCM. Among Ikaros genes, the expression of IKZF1 and IKZF3 positively correlated with the infiltration level of CD4+ T cells and CD8+ T cells, B cells, and Tregs in SKCM and negatively correlated with the infiltration level of M0 and M1 macrophages. Moreover, single-cell sequencing data analysis revealed that IKZF1 and IKZF3 were mainly expressed by immune cells. Correlation analysis shows the immune factors and drug responses associated with IKZF3 expression. In conclusion, the present study is the first, to our knowledge, to identify a pan-cancer genomic signature of the Ikaros gene family among different cancers. Expression of these family members, particularly high levels of IKZF3, indicate positive immunological status and beneficial clinical outcomes of SKCM. IKZF3 may therefore serve as potential targets for immunotherapy of melanoma.

Purpose: The present study was carried out to investigate the global m6A-modified RNA pattern and possible mechanisms underlying the pathogenesis of keloid.Method: In total, 14 normal skin and 14 keloid tissue samples were first collected on clinics. Then, three samples from each group were randomly selected to be verified with the Western blotting to determine the level of methyltransferase and demethylase. The total RNA of all samples in each group was isolated and subjected to the analysis of MeRIP sequencing and RNA sequencing. Using software of MeTDiff and htseq-count, the m6A peaks and differentially expressed genes (DEGs) were determined within the fold change >2 and p-value < 0.05. The top 10 pathways of m6A-modified genes in each group and the differentially expressed genes were enriched by the Kyoto Encyclopedia of Genes and Genomes signaling pathways. Finally, the closely associated pathway was determined using the Western blotting and immunofluorescence staining.Results: There was a higher protein level of WTAP and Mettl3 in the keloid than in the normal tissue. In the keloid samples, 21,020 unique m6A peaks with 6,573 unique m6A-associated genetic transcripts appeared. In the normal tissue, 4,028 unique m6A peaks with 779 m6A-associated modified genes appeared. In the RNA sequencing, there were 847 genes significantly changed between these groups, transcriptionally. The genes with m6A-methylated modification and the upregulated differentially expressed genes between two tissues were both mainly related to the Wnt signaling pathway. Moreover, the hyper-m6A-modified Wnt/β-catenin pathway in keloid was verified with Western blotting. From the immunofluorescence staining results, we found that the accumulated fibroblasts were under a hyper-m6A condition in the keloid, and the Wnt/β-Catenin signaling pathway was mainly activated in the fibroblasts.Conclusion: The fibroblasts in the keloid were under a cellular hyper-m6A-methylated condition, and the hyper-m6A-modified highly expressed Wnt/β-catenin pathway in the dermal fibroblasts might promote the pathogenesis of keloid.

The development of Xiongan New Area in Hebei Province, China, as a significant national choice, has considerable strategic significance for the integrated growth of Beijing, Tianjin, and Hebei. This paper proposes a cloud model for the suitability evaluation of the construction of Xiongan New Area based on entropy weight analysis, taking into account the geological conditions, groundwater environment, environmental geological problems, and other factors of the suitability of image city development. According to the research, the suitability evaluation findings for the project building employing the cloud model are in strong accord with those of the traditional model and have some application potential. The evaluation’s findings indicate that the project construction in Xiongan New Area is acceptable, with suitable and relatively suitable sites making up 81.4% of the total area and excellent circumstances for project development, construction, and usage. This study offers helpful direction for Xiongan New Area’s urban land-space design and serves as a useful point of comparison for studies looking at the viability of other deep Quaternary Plain region engineering buildings.

Abstract Background In gut microbiome studies, the cultured gut microbial resource plays essential roles, such as helping to unravel gut microbial functions and host-microbe interactions. Although several major studies have been performed to elucidate the cultured human gut microbiota, up to 70% of the Unified Human Gastrointestinal Genome species have not been cultured to date. Large-scale gut microbial isolation and identification as well as availability to the public are imperative for gut microbial studies and further characterizing human gut microbial functions. Results In this study, we constructed a human Gut Microbial Biobank (hGMB; homepage: hgmb.nmdc.cn ) through the cultivation of 10,558 isolates from 31 sample mixtures of 239 fresh fecal samples from healthy Chinese volunteers, and deposited 1170 strains representing 400 different species in culture collections of the International Depository Authority for long-term preservation and public access worldwide. Following the rules of the International Code of Nomenclature of Prokaryotes, 102 new species were characterized and denominated, while 28 new genera and 3 new families were proposed. hGMB represented over 80% of the common and dominant human gut microbial genera and species characterized from global human gut 16S rRNA gene amplicon data (n = 11,647) and cultured 24 “most-wanted” and “medium priority” taxa proposed by the Human Microbiome Project. We in total sequenced 115 genomes representing 102 novel taxa and 13 previously known species. Further in silico analysis revealed that the newly sequenced hGMB genomes represented 22 previously uncultured species in the Unified Human Gastrointestinal Genome (UHGG) and contributed 24 representatives of potentially “dark taxa” that had not been discovered by UHGG. The nonredundant gene catalogs generated from the hGMB genomes covered over 50% of the functionally known genes (KEGG orthologs) in the largest global human gut gene catalogs and approximately 10% of the “most wanted” functionally unknown proteins in the FUnkFams database. Conclusions A publicly accessible human Gut Microbial Biobank (hGMB) was established that contained 1170 strains and represents 400 human gut microbial species. hGMB expands the gut microbial resources and genomic repository by adding 102 novel species, 28 new genera, 3 new families, and 115 new genomes of human gut microbes. In this study, the vocal intonation therapy (VIT) was compared with the standard respiratory therapy for people suffering from respiratory dysfunction as a result of cervical spinal cord injury (CSCI) to observe its effect on vocal quality. Thirty patients with vocal dysfunction after CSCI with the injury time of more than 3 months were screened for inclusion in the trial, and 18 patients completed the 12-weeks, each participant had 60 sessions in total in the clinical trial. All patients were allocated to the intervention group or the control group. The intervention group received VIT training and the control group received respiratory phonation therapy. Both groups were trained by professional therapists, and the training time was 30 min/day, 5 days/week, for 60 sessions for each group in a total of 12 weeks. In the Baseline (T0), mid-intervention period (after 6 weeks, T1), and after intervention (after 12 weeks, T2), the vocal quality of the two groups of patients was tested with a computer-aided real-time audio analyzer 2.1.6 (Adobe Systems, United States) for Sing-SPL (p < 0.0001), Speech-SPL (p < 0.0001), SNL (p < 0.0001), and F0 (p < 0.0001) of the intervention group were significantly improved compared with the control group. In comparing the spectrometry analysis of vocal quality for the 2 groups of participants, there was a significant difference in the results of Sing-SPL and Speech-SPL acoustic analysis in the intervention group of patients at T2 (after 12 weeks) compared to the control group. Vocal intonation therapy—music therapy can improve the speech sound quality of cervical CSCI patients and provide CSCI patients with a practical, highly operable treatment that has both functional training effects and can bring a pleasant experience that can be promoted in the medical field. Abstract. Background:. There are an increasing number of patients with oral sensory complaints (OSCs) presenting to our dental clinic. For most dentists, it is difficult to distinguish burning mouth syndrome (BMS) from other oral mucosal diseases that may cause symptoms such as burning mouth. It is beneficial to effectively distinguish OSC patients to reduce misdiagnosis and eliminate burning symptoms as much as possible. Methods:. Patients with oral burning sensations in the oral mucosal disease clinic were collected from the Peking University Hospital of Stomatology between September 1, 2014 and December 31, 2018. After excluding oral candidiasis, anemic stomatitis, dental material allergy, and other diseases from patients with oral sensory complaints, basic conditions such as gender, age, education level, job status, hyperglycemia, hypertension, hyperlipidemia, history of brain abnormalities, history of cervical spondylitis, history of thyroid disease, history of thyroid disease and insomnia were obtained. The BMS patients were compared with the control group. The t test and Chi-square test were used for statistical analysis to compare the clinical symptoms of these diseases and explore the risk factors for BMS. Results:. In this case-control study, 395 patients (321 females and 74 males, mean age 55.26 ± 10.51 years) with oral sensory complaints and 391 healthy controls (281 females and 110 males, mean age 47.11 ± 13.10 years) were enrolled, among which, 8.4% (33/395) had oral candidiasis, 1.3% (5/395) had dental material allergy, 0.8% (3/395) had anemic stomatitis and 0.5% (2/395) had lichen planus. A total of 352 patients were eventually diagnosed with BMS. Anxiety and depression were more severe in BMS patients, as were the incidences of sleep disorders and brain abnormalities. Abstract Background Flammulina filiformis (previously known as Asian F. velutipes) is a popular commercial edible mushroom. Many bioactive compounds with medicinal effects, such as polysaccharides and sesquiterpenoids, have been isolated and identified from F. filiformis, but their biosynthesis and regulation at the molecular level remains unclear. In this study, we sequenced the genome of the wild strain F. filiformis Liu355, predicted its biosynthetic gene clusters (BGCs) and profiled the expression of these genes in wild and cultivar strains and in different developmental stages of the wild F. filiformis strain by a comparative transcriptomic analysis. Results We found that the genome of the F. filiformis was 35.01 Mb in length and harbored 10,396 gene models. Thirteen putative terpenoid gene clusters were predicted and 12 sesquiterpene synthase genes belonging to four different groups and two type I polyketide synthase gene clusters were identified in the F. filiformis genome. The number of genes related to terpenoid biosynthesis was higher in the wild strain (119 genes) than in the cultivar strain (81 genes). Most terpenoid biosynthesis genes were upregulated in the primordium and fruiting body of the wild strain, while the polyketide synthase genes were generally upregulated in the mycelium of the wild strain. Moreover, genes encoding UDP-glucose pyrophosphorylase and UDP-glucose dehydrogenase, which are involved in polysaccharide biosynthesis, had relatively high transcript levels both in the mycelium and fruiting body of the wild F. filiformis strain. Conclusions F. filiformis is enriched in a number of gene clusters involved in the biosynthesis of polysaccharides and terpenoid bioactive compounds and these genes usually display differential expression between wild and cultivar strains, even in different developmental stages. This study expands our knowledge of the biology of F. filiformis and provides valuable data for elucidating the regulation of secondary metabolites in this unique F. filiformis strain.

Different austenitizing temperatures were used to obtain medium-carbon low-alloy (MCLA) steels with different initial states of tempering, including lath martensite microstructures and matrix carbon contents. The effects of these factors on the tempered carbides and impact toughness of the MCLA steels were investigated via optical microscopy (OM), scanning electron microscopy (SEM), electron backscatter diffraction (EBSD), X-ray diffraction (XRD) and transmission electron microscopy (TEM). The results showed that the matrix carbon content and lath martensite substructure sizes could be effectively controlled by the austenitizing temperature. The morphologies of the tempered carbides precipitated at the boundaries were controlled by both the matrix carbon content and the lath martensite substructure sizes. Higher matrix carbon content and boundary densities increased the nucleation rate and promoted the formation of spherical carbides. The spheroidization of carbides was conducive to improving the impact toughness. However, the decrease in the density of high-angle grain boundaries and the appearance of long strip-shaped carbides drastically reduced the impact toughness.

Addition of Sc is capable of greatly improving the mechanical properties of aluminum alloy welded joints, reducing the hot crack sensitivity coefficient; thus, it could solve the welding problem of ultra-high strength aluminum alloys. Friction stir welding has the advantages of small heat-affected zone, low residual stress, and small deformation of welding work piece, making it a good choice for welding materials with high heat crack sensitivity. In this article, the microstructure and properties of friction stir welding (FSW) joints of Al–Zn–Mg–Cu–Zr–(Sc) alloys were characterized via optical microscopy (OM), transmission electron microscopy (TEM), micro-hardness testing, and universal tensile testing. The mechanism of the effect of adding Sc element on improving the welding properties of the ultra-high strength Al–Zn–Mg–Cu–Zr alloy was explored. The results show that the welding joints of Al–Zn–Mg–Cu–Zr–(Sc) alloy exhibit similar microstructure characteristics. The welding nugget zone (WNZ) displays a dynamic recrystallization feature comprising fine and uniform equiaxed grains with high density dislocations. Most of the aged precipitates dissolve into the matrix in the WNZ. The grains in the thermal-mechanical affected zone (TMAZ) are elongated with higher dislocation density, and residual aged precipitates coarsened remarkably. The heat-affected zone (HAZ) retains the same grain morphology as the base metal. Most of the aged η' precipitates grow, and a few coarsen to be the η phase in this zone. However, 0.17% (mass fraction) Sc addition increases the ultimate tensile strength of FSW joint by 43 MPa, yield strength by 23 MPa, elongation by 2.3%, and the welding coefficient up to 74.1%. Al3(Sc, Zr) dispersoids are found to achieve the following: 1) strongly inhibit the movement of dislocations, sub-grain boundaries, and grain boundaries; 2) significantly refine grains while retaining several sub-structures; and 3) factor in Orowan precipitation strengthening. Therefore, the mechanical properties of FSW joints can be improved using the refined grain, sub-structure, and precipitation strengthening mechanisms.

Resistance to powdery mildew is an important trait of interest in many wheat breeding programs. The information on genes conferring resistance to powdery mildew in wheat cultivars is useful in parental selection. Winter wheat breeding line DH51302 derived from Liangxing 99 and cultivar Shimai 26 derived from Jimai 22 showed identical infection patterns against 13 isolates of Blumeria graminis f. sp. tritici (Bgt) that causes wheat powdery mildew. DH51302 and Shimai 26 were crossed to a powdery mildew susceptible cultivar Zhongzuo 9504 and the F2:3 families were used in molecular localization of the resistance genes. Fourteen polymorphic markers, which were linked to Pm52 from Liangxing 99, were used to establish the genetic linkage maps for the resistance genes PmDH51302 and PmSM26 in DH51302 and Shimai 26, respectively. These genes were placed in the same genetic interval where Pm52 resides. Analysis of gene-linked molecular markers indicated that PmDH51302 and PmSM26 differed from other powdery mildew resistance genes on chromosome arm 2BL, such as Pm6, Pm33, Pm51, MlZec1, MlAB10, and Pm64. Based on the results of reaction patterns to different Bgt isolates and molecular marker localization, together with the pedigree information, DH51302 and Shimai 26 carried the same gene, Pm52, which confers their resistance to powdery mildew.

Analysis of genetic progress for lodging-related traits provides important information for further improvement of lodging resistance. Forty winter wheat cultivars widely grown in the Yellow-Huai River Valleys Winter Wheat Zone (YHWZ) of China during the period of 1964–2015 were evaluated for several lodging-related traits in three cropping seasons. Plant height, height at center of gravity, length of the basal second internode, and lodging index decreased significantly in this period, and the average annual genetic gains for these traits were −0.50 cm or −0.62%, −0.27 cm or −0.60%, −0.06 cm or −0.63%, and −0.01 or −0.94%, respectively. Different from other traits, stem strength showed a significant increasing trend with the breeding period, and the annual genetic gains were 0.03 N or 0.05%. Correlation analysis showed that lodging index was positively correlated with plant height, height at center of gravity, and length of the basal second internode, but negatively correlated with stem strength. Meanwhile, significantly positive correlations were observed between plant height, height at center of gravity, and length of the basal first and second internodes. By comparison with the wild types, dwarfing genes had significant effects on all lodging-related traits studied except for length of the basal first internode and stem strength. Principle component analysis demonstrated that plant height and stem strength were the most important factors influencing lodging resistance. Clustering analysis based on the first two principle components further indicated the targets of wheat lodging-resistant breeding have changed from reducing plant height to strengthening stem strength over the breeding periods. This study indicates that the increase of stem strength is vital to improve lodging resistance in this region under the high-yielding condition when plant height is in an optimal range.

Abstract. Background:. Blood glucose control is closely related to type 2 diabetes mellitus (T2DM) prognosis. This multicenter study aimed to investigate blood glucose control among patients with insulin-treated T2DM in North China and explore the application value of combining an elastic network (EN) with a machine-learning algorithm to predict glycemic control. Methods:. Basic information, biochemical indices, and diabetes-related data were collected via questionnaire from 2787 consecutive participants recruited from 27 centers in six cities between January 2016 and December 2017. An EN regression was used to address variable collinearity. Then, three common machine learning algorithms (random forest [RF], support vector machine [SVM], and back propagation artificial neural network [BP-ANN]) were used to simulate and predict blood glucose status. Additionally, a stepwise logistic regression was performed to compare the machine learning models. Results:. The well-controlled blood glucose rate was 45.82% in North China. The multivariable analysis found that hypertension history, atherosclerotic cardiovascular disease history, exercise, and total cholesterol were protective factors in glycosylated hemoglobin (HbA1c) control, while central adiposity, family history, T2DM duration, complications, insulin dose, blood pressure, and hypertension were risk factors for elevated HbA1c. Before the dimensional reduction in the EN, the areas under the curve of RF, SVM, and BP were 0.73, 0.61, and 0.70, respectively, while these figures increased to 0.75, 0.72, and 0.72, respectively, after dimensional reduction. Moreover, the EN and machine learning models had higher sensitivity and accuracy than the logistic regression models (the sensitivity and accuracy of logistic were 0.52 and 0.56; RF: 0.79, 0.70; SVM: 0.84, 0.73; BP-ANN: 0.78, 0.73, respectively). Conclusions:. More than half of T2DM patients in North China had poor glycemic control and were at a higher risk of developing diabetic complications. The EN and machine learning algorithms are alternative choices, in addition to the traditional logistic model, for building predictive models of blood glucose control in patients with T2DM.

Ergosterol derivatives exhibited copious promising biological activities. The fungus Gyromitra esculenta is widely distributed in Europe and North America. In order to examine the chemical properties of Gyromitra esculenta, a phytochemical study has been preceded and resulted in the isolation of the steroid, ergosta-5, 22-dien-3β-ol (brassicasterol), from its methanol extract. The complete identification and absolute configuration of the isolated compound have been established by X-ray structural analysis to be (22E, 24R)-24-methylcholesta-5, 22-dien-3beta-ol. The reported cytotoxicity and the great structural similarity of the isolated compound with the cocrystallized ligand of the aromatase enzyme inspired us to run molecular docking studies against that protein. Ergosta-5, 22-dien-3β-ol occupied the target protein with a binding mode almost the same as the cocrystallized ligand and a binding affinity of −33.55 kcal/mol, which was better than that of the cocrystallized ligand (−22.61 kcal/mol). This promising result encouraged us to conduct in silico ADMET and toxicity studies of ergosta-5, 22-dien-3β-ol against 6 models, and the results expected the likeness of the isolated compound to be a drug. In conclusion, ergosta-5, 22-dien-3β-ol has been isolated from Gyromitra esculenta, identified by X-ray structural analysis, and exhibited promising in silico activities against aromatase enzyme.

In this paper, a simple sub-loading yield surface model for both normally consolidated and over-consolidated clay is proposed with emphasis on the effect of the yield surface shape. Compared with the modified Cam-clay model, only one additional material parameter is introduced to reflect geometry features of the yield surface. A higher-order stress–dilatancy relation is given in the current study, leading to a new yield function capable of offering an adequate description of the yield surface of soil samples in the p–q plane. By introducing the concept of the sub-loading yield surface and the unified hardening parameter, the proposed model can capture the main features of the over-consolidated clay with dilatancy and strain-softening behavior and the main features of the normally consolidated clay with contraction and strain-hardening behavior. The results show that adjusting the yield surface leads to more accurate predictions than the modified Cam-clay model. The proposed model can also reasonably describe its mechanical behavior for clay samples.

Abstract Three new heptelidic acid derivatives (1–3) including two new dimeric esters and two known heptelidic acid analogues (4 and 5) were isolated from the solid culture of mushroom Lentinellus ursinus. The structures of new compounds were confirmed by the analysis of NMR and HRESIMS spectroscopic data. The biosynthetic origin of compounds 1–5 was postulated. Compounds 1–5 exhibited no antibacterial activity against Staphylococcus aureus and Escherichia coli at the dose of 100 μM. Graphical Abstract

Two new secondary metabolites, kongiilines A and B (1, 7), and two asperphenamate derivatives, asperphenamates B and C (5–6), together with 16 known compounds (2–4, 8–20), were isolated from Tibetan Plateau fungi Penicillium kongii and Penicillium brasilianum. This is the first report on asperphenamates B and C as naturally occurring compounds, and that aspterric acid is isolated from P. brasilianum for the first time. Their structures were elucidated by different spectroscopic techniques including high-resolution electrospray ionisation mass spectrum, 1D nuclear magnetic resonance (NMR), and 2D NMR as well as electronic circular dichroism. Compounds 4, 5, and 10 exhibited cytotoxicity activities against human colon carcinoma HCT116 cell line with IC50 values of 88.16, 77.68, and 36.92 μM, respectively. Fungi from Tibetan Plateau represent important and rich resources for the investigation of new chemicals.

Purpose: Patients with left-sided breast cancer (LSBC) are at increased risk of cardiac morbidity from adjuvant breast radiation therapy (ABRT). Breath-hold (BH) techniques substantially reduce the radiation received by heart during radiation therapy for LSBC. However, a subset of patients with LSBC are ineligible for BH techniques due to an inability to breath-hold or because of other comorbidities. To reduce radiation to the heart, we routinely use a custom-made breast shell for the treatment of patients with LSBC who are ineligible for BH techniques. This study evaluates the dosimetric impact of using a breast shell for patients with LSBC undergoing ABRT. Methods and materials: Sixteen consecutive patients with LSBC who failed BH and underwent ABRT using a breast shell during the period of 2014 to 2016 were identified. Treatment was planned using field-in-field tangents with a prescribed dose of 42.5 Gy in 16 fractions. Comparisons between plans with and without a shell were made for each patient using a paired t test to quantify the sparing of organs at risk (OARs) and target coverage. Results: There was no statistically significant difference in the planning target volume of breast coverage. Autologous fat transplantation has been applied widely in clinic. However, the low survival rate is still a problem to be solved. Studies shows that the human adipose-derived stem cells (ADSCs) transfected by vascular endothelial growth factor (VEGF) can improve the survival rate of autologous fat transplantation. Our study is to evaluate the effects of the conditioned medium of VEGF-transfected human adipose-derived stem cells (VEGF-ADSCs-CM) on fat transplantation. ADSCs were isolated and transfected with MOI = 40. The study was divided into three groups, VEGF-ADSCs-CM group, normal-ADSCs-CM group and control group. The conditioned media for VEGF-ADSCs-CM group and normal-ADSCs-CM group were collected, and then mixed with fat, with the mixtures being injected into the back of nude mice. On 4, 7, 15, 30, 60 days after transplantation, the grafts were evaluated on the wet weight, histology, ELISA and western blot. As the results revealed, the survival rate of VEGF-ADSCs-CM group was highest with the best fat cell morphology, and the VEGF secretion of VEGF-ADSCs-CM group was also highest. Therefore, our study demonstrates that VEGF-ADSCs-CM can improve the survival rate of fat transplantation effectively, and VEGF-ADSCs-CM can be regarded as an effective assisted method for fat transplantation.

Local sympathetic denervation by surgical sympathectomy is used in the treatment of lower limb ulcers and ischemia, but the restoration of cutaneous sympathetic nerve functions is less clear. This study aims to explore the recovery of cutaneous sympathetic functions after bilateral L2–4 sympathectomy. The skin temperature of the left feet, using a point monitoring thermometer, increased intraoperatively after sympathectomy. The cytoplasm of sympathetic neurons contained tyrosine hydroxylase and dopamine β-hydroxylase, visualized by immunofluorescence, indicated the accuracy of sympathectomy. Iodine starch test results suggested that the sweating function of the hind feet plantar skin decreased 2 and 7 weeks after lumbar sympathectomy but had recovered by 3 months. Immunofluorescence and western blot assay results revealed that norepinephrine and dopamine β-hydroxylase expression in the skin from the sacrococcygeal region and hind feet decreased in the sympathectomized group at 2 weeks. Transmission electron microscopy results showed that perinuclear space and axon demyelination in sympathetic cells in the L5 sympathetic trunks were found in the sympathectomized group 3 months after sympathectomy. Although sympathetic denervation occurred in the sacrococcygeal region and hind feet skin 2 weeks after lumbar sympathectomy, the skin functions recovered gradually over 7 weeks to 3 months. In conclusion, sympathetic functional recovery may account for the recurrence of hyperhidrosis after sympathectomy and the normalization of sympathetic nerve trunks after incomplete injury. The recovery of sympathetic nerve function was slower in the limbs than in the torso after bilateral L2–4 sympathectomy.

Hydrology is an old discipline due to its early origination, as well as a young discipline due to its insufficient scientific foundation as a natural science. Thus hydrology has long been haunted by a debate between natural functionalities found in observations vs. model results built on many simplified assumptions. We define the hydrological maze as puzzles, paradoxes, or complexity involved in hydrologic measurements and interpretations. The objective of this study was to reveal the hydrological maze through a comprehensive review of decades of work since the 1980s on observations and experiments using a combination of natural and artificial catchments at the Chuzhou Hydrology Laboratory in China, highlighting the role of hydropedology in hillslope and catchment hydrology based on long-term monitoring of surface and subsurface flows at various soil depths and at different spatial scales. A conception has emerged that indicates the fundamental control of hydropedological factors (such as soil types, soil properties, and their spatial variations) as the trigger for the hydrological maze, including runoff generation, runoff composition, flow heterogeneity, and various hydrological puzzles. It is clear that the vadose zone is the key source for nonlinear and dissipative complexity in the hydrological maze that is intertwined with hydrochemical and hydroecological dynamics. Therein lies the hope for new hydrological insights and possible solutions to the hydrological maze.

In allusion to the flaws in software cost model and optimal release policy, inadequate consideration for real debugging, a cost model and optimal release policy for SRGM (Software Reliability Growth Model) incorporating imperfect debugging is proposed. A SRGM is presented, based on incomplete debugging, introduction of new faults and TE (Testing Effort). It is verified to describe real testing process well by actual failure data set and has better performance as compared to other models. Based on the proposed SRGM, a formulation of cost function is also established especially considering the impact of imperfect debugging on cost. Furthermore, the optimal release policies given limited reliability objective and the uncertainty in actual total cost exceeding expected one are developed and elaborated. Finally, a numerical example and related data analyses are illustrated and parameter sensitivity analysis is performed.

Tea is one of the most popular beverages across the world and is made exclusively from cultivars of Camellia sinensis. Many wild relatives of the genus Camellia that are closely related to C. sinensis are native to Southwest China. In this study, we first identified the distinct genetic divergence between C. sinensis and its wild relatives and provided a glimpse into the artificial selection of tea plants at a genome-wide level by analyzing 15,444 genomic SNPs that were identified from 18 cultivated and wild tea accessions using a high-throughput genome-wide restriction site-associated DNA sequencing (RAD-Seq) approach. Six distinct clusters were detected by phylogeny inferrence and principal component and genetic structural analyses, and these clusters corresponded to six Camellia species/varieties. Genetic divergence apparently indicated that C. taliensis var. bangwei is a semi-wild or transient landrace occupying a phylogenetic position between those wild and cultivated tea plants. Cultivated accessions exhibited greater heterozygosity than wild accessions, with the exception of C. taliensis var. bangwei. Thirteen genes with non-synonymous SNPs exhibited strong selective signals that were suggestive of putative artificial selective footprints for tea plants during domestication. The genome-wide SNPs provide a fundamental data resource for assessing genetic relationships, characterizing complex traits, comparing heterozygosity and analyzing putatitve artificial selection in tea plants.

Effects of tempering temperature on the microstructures and low temperature impact toughness of 42CrMo4-V steel were investigated. Microstructures of 42CrMo4-V steel after tempering at 570–720 °C for 3 h were experimentally investigated using scanning electron microscopy (SEM), electron back-scattered diffraction (EBSD), transmission electron microscopy (TEM) and X-ray diffraction (XRD). The results showed that the carbide precipitation sequence of 42CrMo4-V steel is M8C7 → M3C and the absorbed energies of 42CrMo4-V steel increase greatly from 21.7 J to 132.3 J with increasing tempering temperature from 570 °C to 720 °C. The changes of impact toughness with increasing tempering temperature were attributed to the softening of matrix, the evolution of carbide precipitates and grain structures.

Seismic data is commonly acquired sparsely and irregularly, which necessitates the regularization of seismic data with anti-aliasing and anti-leakage methods during seismic data processing. We propose a novel method of 4D anti-aliasing and anti-leakage Fourier transform using a cube-removal strategy to address the combination of irregular sampling and aliasing in high-dimensional seismic data. We compute a weighting function by stacking the spectrum along the radial lines, apply this function to suppress the aliasing energy, and then iteratively pick the dominant amplitude cube to construct the Fourier spectrum. The proposed method is very efficient due to a cube removal strategy for accelerating the convergence of Fourier reconstruction and a well-designed parallel architecture using CPU/GPU collaborative computing. To better fill the acquisition holes from 5D seismic data and meanwhile considering the GPU memory limitation, we developed the anti-aliasing and anti-leakage Fourier transform method in 4D with the remaining spatial dimension looped. The entire workflow is composed of three steps: data splitting, 4D regularization, and data merging. Numerical tests on both synthetic and field data examples demonstrate the high efficiency and effectiveness of our approach. [ABSTRACT FROM AUTHOR]
