In 2020 and 2021, peach fruit with natural disease and decay were collected from orchards in Zhejiang Province. Then the fungal pathogens were isolated from the diseased spots and identified by morphological characteristics, molecular biological identification (rDNA-ITS sequence analysis) and pathogenicity detection. The antifungal effect of cinnamon essential oil on the fungal pathogens in vitro and in vivo were also investigated. The results showed that two fungal pathogens with strong pathogenicity were isolated, and identified as Monilinia fructicola and Rhizopus stolonifer, which caused brown rot and soft rot, respectively. Cinnamon essential oil inhibited the growth of M. fructicola and R. stolonifer, and the minimum inhibitory concentrations were 0.4 and 0.8 mL/L, respectively. M. fructicola was more sensitive to cinnamon essential oil. Cinnamon essential oil fumigation could reduce the disease incidence and lesion diameter of brown rot and soft rot of peach fruit caused by these two fungal pathogens. Therefore, cinnamon essential oil could be used as a natural fungicide to control fungal diseases of peach fruit.

Online monitoring of tool wear is an indispensable part of industrial production. It allows for real-time assessment of tool condition, enabling tool replacement at the optimal time. This not only reduces the time costs associated with downtime for inspections but also effectively prevents the deterioration of workpiece quality due to excessive wear. Therefore, online monitoring of tool wear is essential for ensuring machining quality and efficiency. This research presents a monitoring technique for tool wear while performing micro-milling by extracting features from machining signals. Initially, the wear mechanism, which includes initial, regular, and severe wear, is analyzed. Tool wear labels are then constructed. Subsequently, the recursive feature elimination (RFE) algorithm optimizes the original feature set from the processed signal, resulting in a subset of relevant features that accurately describe the tool wear. The optimal subset is utilized as an input vector for extreme gradient boosting (XGBoost) training. Next, the best hyperparameter combination for the monitoring model is determined using Bayesian optimization (BO), ensuring the generalization performance of the monitoring model. Finally, the two-edge wear monitoring model is established based on the RFE-BO-XGBoost. To validate the superiority of the RFE-BO-XGBoost model, the performance of the XGBoost model with default parameters in tool wear monitoring is also investigated. The results demonstrate that the constructed monitoring model can effectively identify the tool wear stage. More notably, it precisely predicts the wear status of two cutting edges simultaneously, enabling qualitative and quantitative monitoring of tool wear under normal manufacturing settings.

Abstract Background Tumor-associated macrophages (TAMs) significantly influence the progression, metastasis, and recurrence of esophageal squamous cell carcinoma (ESCC). The aberrant expression of long noncoding RNAs (lncRNAs) in ESCC has been established, yet the role of lncRNAs in TAM reprogramming during ESCC progression remains largely unexplored. Methods ESCC TAM-related lncRNAs were identified by intersecting differentially expressed lncRNAs with immune-related lncRNAs and performing immune cell infiltration analysis. The expression profile and clinical relevance of LINC00330 were examined using the TCGA database and clinical samples. The LINC00330 overexpression and interference sequences were constructed to evaluate the effect of LINC00330 on ESCC progression. Single-cell sequencing data, CIBERSORTx, and GEPIA were utilized to analyze immune cell infiltration within the ESCC tumor microenvironment and to assess the correlation between LINC00330 and TAM infiltration. ESCC-macrophage coculture experiments were conducted to investigate the influence of LINC00330 on TAM reprogramming and its subsequent effect on ESCC progression. The interaction between LINC00330 and C–C motif ligand 2 (CCL2) was confirmed through transcriptomic sequencing, subcellular localization analysis, RNA pulldown, silver staining, RNA immunoprecipitation, and other experiments. Results LINC00330 is significantly downregulated in ESCC tissues and strongly associated with poor patient outcomes. Overexpression of LINC00330 inhibits ESCC progression, including proliferation, invasion, epithelial–mesenchymal transition, and tumorigenicity in vivo. LINC00330 promotes TAM reprogramming, and LINC00330-mediated TAM reprogramming inhibits ESCC progression. LINC00330 binds to the CCL2 protein and inhibits the expression of CCL2 and downstream signaling pathways. CCL2 is critical for LINC00330-mediated TAM reprogramming and ESCC progression. Conclusions LINC00330 inhibited ESCC progression by disrupting the CCL2/CCR2 axis and its downstream signaling pathways in an autocrine fashion; and by impeding CCL2-mediated TAM reprogramming in a paracrine manner. The new mechanism of TAM reprogramming mediated by the LINC00330/CCL2 axis may provide potential strategies for targeted and immunocombination therapies for patients with ESCC. Graphical Abstract

Abstract Ferroptosis has important value in cancer treatment. It is significant to explore the new ferroptosis-related lncRNAs prediction model in Hepatocellular carcinoma (HCC) and the potential molecular mechanism of ferroptosis-related lncRNAs. We constructed a prognostic multi-lncRNA signature based on ferroptosis-related differentially expressed lncRNAs in HCC. qRT-PCR was applied to determine the expression of lncRNA in HCC cells. The biological roles of NRAV in vitro and in vivo were determined by performing a series of functional experiments. Furthermore, dual-luciferase reporter and RNA immunoprecipitation (RIP) assays were used to confirm the interaction of NRAV with miR-375-3P. We identified 6 differently expressed lncRNAs associated with the prognosis of HCC. Kaplan–Meier analyses revealed the high-risk lncRNAs signature associated with poor prognosis of HCC. Moreover, the AUC of the lncRNAs signature showed utility in predicting HCC prognosis. Further functional experiments show that the high expression of NRAV can strengthen the viciousness of HCC. Interestingly, we found that NRAV can enhance iron export and ferroptosis resistance. Further study showed that NRAV competitively binds to miR-375-3P and attenuates the inhibitory effect of miR-375-3P on SLC7A11, affecting the prognosis of patients with HCC. In conclusion, We developed a novel ferroptosis-related lncRNAs prognostic model with important predictive value for the prognosis of HCC. NRAV is important in ferroptosis induction through the miR-375-3P/SLC7A11 axis.

Abstract Background Targeting the tumor microenvironment (TME) has emerged as a promising strategy in cancer treatment, particularly through the utilization of immune checkpoint blockade (ICB) agents such as PD-1/PD-L1 inhibitors. Despite partial success, the presence of tumor-associated macrophages (TAMs) contributes to an immunosuppressive TME that fosters tumor progression, and diminishes the therapeutic efficacy of ICB. Blockade of the CD47/SIRPα pathway has proven to be an effective intervention, that restores macrophage phagocytosis and yields substantial antitumor effects, especially when combined with PD-1/PD-L1 blockade. Therefore, the identification of small molecules capable of simultaneously blocking CD47/SIRPα and PD-1/PD-L1 interactions has remained imperative. Methods SMC18, a small molecule with the capacity of targeting both SIRPα and PD-L1 was obtained using MST. The efficiency of SMC18 in interrupting CD47/SIRPα and PD-1/PD-L1 interactions was tested by the blocking assay. The function of SMC18 in enhancing the activity of macrophages and T cells was tested using phagocytosis assay and co-culture assay. The antitumor effects and mechanisms of SMC18 were investigated in the MC38-bearing mouse model. Results SMC18, a small molecule that dual-targets both SIRPα and PD-L1 protein, was identified. SMC18 effectively blocked CD47/SIRPα interaction, thereby restoring macrophage phagocytosis, and disrupted PD-1/PD-L1 interactions, thus activating Jurkat cells, as evidenced by increased secretion of IL-2. SMC18 demonstrated substantial inhibition of MC38 tumor growths through promoting the infiltration of CD8+ T and M1-type macrophages into tumor sites, while also priming the function of CD8+ T cells and macrophages. Moreover, SMC18 in combination with radiotherapy (RT) further improved the therapeutic efficacy. Conclusion Our findings suggested that the small molecule compound SMC18, which dual-targets the CD47/SIRPα and PD-1/PD-L1 pathways, could be a candidate for promoting macrophage- and T-cell-mediated phagocytosis and immune responses in cancer immunotherapy.

Abstract Studies on the associations of blood pressure (BP) and the risk of venous thromboembolism (VTE) had been performed neither among pregnant women nor in Chinese population. This study included participants of pregnant women from a retrospective multicenter cohort, between May 2020 and April 2023. Systolic BP (SBP) and diastolic BP (DBP) of the participants were measured in the third trimester. The incidences of VTE (including deep venous thrombosis and/or pulmonary embolism) at 42 days postpartum were followed. With regards to SBP, pregnant women in the Q1 (≤114 mmHg), Q2 (115–122 mmHg), and Q4 group (≥131 mmHg) had increased risk of VTE than those in Q3 group (123–130 mmHg), with ORs 4.48 [1.69, 11.85], 3.52 [1.30, 9.59], and 3.17 [1.12, 8.99], respectively. Compared with pregnant women with the Q4 of DBP (≥85 mmHg), women of Q1 (≤71 mmHg) were found to have elevated risk of VTE (OR 2.73 [1.25, 5.96]). A one standard deviation decrease of DBP (9 mmHg) was related with 37% elevated risk of VTE (OR 1.37 [1.05, 1.79]). This study demonstrated a U‐shaped association of SBP in the third trimester and VTE postpartum and inverse association of DBP in the third trimester and VTE postpartum.

Suaeda glauca Bunge produces dimorphic seeds on the same plant, with brown seeds displaying non-dormant characteristics and black seeds exhibiting intermediate physiological dormancy traits. Previous studies have shown that black seeds have a very low germination rate under natural conditions, but exogenous GA3 effectively enhanced the germination rate of black seeds. However, the physiological and molecular mechanisms underlying the effects of GA3 on S. glauca black seeds are still unclear. In this study, transcriptomic profiles of seeds at different germination stages with and without GA3 treatment were analyzed and compared, and the TTF, H2O2, O2–, starch, and soluble sugar contents of the corresponding seed samples were determined. The results indicated that exogenous GA3 treatment significantly increased seed vigor, H2O2, and O2– contents but decreased starch and soluble sugar contents of S. glauca black seeds during seed dormancy release. RNA-seq results showed that a total of 1136 DEGs were identified in three comparison groups and were involved mainly in plant hormone signal transduction, diterpenoid biosynthesis, flavonoid biosynthesis, phenylpropanoid biosynthesis, and carbohydrate metabolism pathway. Among them, the DEGs related to diterpenoid biosynthesis (SgGA3ox1, SgKAO and SgGA2ox8) and ABA signal transduction (SgPP2Cs) could play important roles during seed dormancy release. Most genes involved in phenylpropanoid biosynthesis were activated under GA3 treatment conditions, especially many SgPER genes encoding peroxidase. In addition, exogenous GA3 treatment also significantly enhanced the expression of genes involved in flavonoid synthesis, which might be beneficial to seed dormancy release. In accordance with the decline in starch and soluble sugar contents, 15 genes involved in carbohydrate metabolism were significantly up-regulated during GA3-induced dormancy release, such as SgBAM, SgHXK2, and SgAGLU, etc. In a word, exogenous GA3 effectively increased the germination rate and seed vigor of S. glauca black seeds by mediating the metabolic process or signal transduction of plant hormones, phenylpropanoid and flavonoid biosynthesis, and carbohydrate metabolism processes. Our results provide novel insights into the transcriptional regulation mechanism of exogenous GA3 on the dormancy release of S. glauca black seeds. The candidate genes identified in this study may be further studied and used to enrich our knowledge of seed dormancy and germination.

Abstract Brain tumour diagnosis is significant for both physicians and patients, but the low contrast and the artefacts of MRI glioma images always affect the diagnostic accuracy. The existing mainstream image enhancement methods are insufficient in improving contrast and suppressing artefacts simultaneously. To enrich the field of glioma image enhancement, this research proposed a glioma image enhancement method based on histogram modification and total variational using stochastic parallel gradient descent (SPGD) algorithm. Firstly, this method modifies the cumulative distribution function on the image histogram and performs gamma correction on the image according to the modified histogram to obtain a contrast‐enhanced image. Then, the method suppresses the artefacts of glioma images by total variational and wavelet denoising algorithm. To get better enhancement images, the optimal parameters in the proposed method are searched by the SPGD algorithm. The statistical studies performed on 580 real glioma images demonstrate that the authors’ approach can outperform the existing mainstream image enhancement methods. The results show that the proposed method increases the discrete entropy of the image by 8.9% and the contrast by 2.8% compared to original images. The enhanced images are produced by the proposed method with a natural appearance, appealing contrast, less degradation, and reasonable detail preservation.

Abstract Higher-order topological phases featured by hierarchical topological states (HTSs) have spawned a paradigm for developing robust multidimensional wave manipulation. While non-Hermitian skin effects (NHSEs) entail that bulk states collapse to open boundaries as local skin modes, the topological transport properties at the interplay between HTS and NHSE are still at early stage of exploration. Here, we report the non-Hermitian reconstruction of HTSs by incorporating the interplay of NHSEs and HTSs, which manifests robust and controllable topological transport properties. By a feasible design in coupled resonant optical waveguides, we demonstrate that zero-dimensional topological states of HTSs only undergo non-Hermitian reconstruction at finitely small system sizes, while nonzero-dimensional topological states of HTSs undergo non-Hermitian reconstruction independent of bulk states. We link the behaviour of zero-dimensional topological states to the restriction of their spatially non-negligible couplings under a macroscopic non-reciprocal framework. Our study unveils the interplay mechanism between NHSEs and HTSs, and underpins topological applications in various wave systems.

Objective: To reveal the bacteriostatic activity of hybrid antimicrobial peptide NK-LPd, and discuss its further exploitation potential. Methods: Two glycines were utilized as the linker to connect the active fragments of NK-lysin and Piscidin, and the physicochemical properties and functional structures of the hybrid antimicrobial peptides were predicted by bioinformatics. The hybrid antimicrobial peptide NK-LPd gene was optimized according to Pichia pastoris codon preference principle. The optimized gene was amplified by gene splicing via overlap extension polymerase chain reaction (gene splicing by overlap extension polymerase chain reaction, SOE-PCR) and inserted into the secretory expression vector pPIC9K. The recombinant plasmid was chemically transformed into P. pastoris KM71 competent cells. The recombinant expression was induced by 0.5% methanol as the inducer, which products were purified by affinity purification and its activity was evaluated. Results: KM71/pPIC9K-NK-LPd was successfully constructed. After expression for 5 days, an expression product with a relative molecular mass of about 6 kDa was obtained in the supernatant, which was in line with the expected size. Bacteriostatic activity experiments showed that NK-LPd had strong bacteriostatic activity against Gram-negative and Gram-positive bacteria. Compared with the parent peptides, the bacteriostatic activity of NK-LPd was significantly enhanced. Conclusion: The hybrid antimicrobial peptide NK-LPd was designed and expressed in P. pastoris KM71, and its bacteriostatic activity was superior to that of the parent peptides. This study could provide technical reference for the design and production of novel hybrid antimicrobial peptides.

To keep pace with the demands of modern photonic integration technology, the electro-optic modulator should feature multi-parameter tunable components and a compact size. Here, we propose a hybrid structure that can modulate the multi-parameters of surface plasmon polaritons (SPPs) simultaneously with a compact size by controlling the electron concentration of indium tin oxide (ITO) in the near-infrared spectrum. The length, width and height of the device are only 15 μm, 5 μm and 9 μm, respectively. The numerical results show that when the electron concentration in ITO changes from 7.5 × 1026 m−3 to 9.5 × 1026 m−3, the variation in amplitude, wavelength and phase are 49%, 300 nm and 347°, respectively. The demonstrated structure paves a new way for multi-parameter modulation and the realization of ultracompact modulators.

Abstract An adaptive neural network dynamic surface control method for strict feedback non‐linear systems with model uncertainty is proposed in this paper. The uncertain parts are approached by RBF neural network; meanwhile, the virtual controller is designed to make system stable according to dynamic surface control. The method of Lyapunov is used to prove the stability and convergence of the system. The simulation results prove the feasibility of this controller and prove that the controller has an advantage to approach the uncertain non‐linear system and make system have well convergence and traceability.

Abstract Background Pulmonary hypertension (PH) is a lethal vascular disease with limited therapeutic options. The mechanistic connections between alveolar hypoxia and PH are not well understood. The aim of this study was to investigate the role of mitotic regulator Polo-like kinase 1 (PLK1) in PH development. Methods Mouse lungs along with human pulmonary arterial smooth muscle cells and endothelial cells were used to investigate the effects of hypoxia on PLK1. Hypoxia- or Sugen5416/hypoxia was applied to induce PH in mice. Plk1 heterozygous knockout mice and PLK1 inhibitors (BI 2536 and BI 6727)-treated mice were checked for the significance of PLK1 in the development of PH. Results Hypoxia stimulated PLK1 expression through induction of HIF1α and RELA. Mice with heterozygous deletion of Plk1 were partially resistant to hypoxia-induced PH. PLK1 inhibitors ameliorated PH in mice. Conclusions Augmented PLK1 is essential for the development of PH and is a druggable target for PH.

Abstract Demand responsive transit (DRT) with app‐based reservation platforms is experiencing a renaissance to bring the tremendous potential for mobility in the urban universe. Nevertheless, how to attract and retain passengers for long‐term use has become one of the most significant problems. The decision‐making psychology of passengers is often overlooked but incredibly critical in the practical applicability of DRT services. This paper proposes a more flexible DRT service with soft time windows considering boundedly rational passengers. A compensation mechanism is developed to alleviate the dissatisfaction of passengers while considerably promoting the system efficiency. A two‐stage model is designed to incorporate bounded rationality into the optimization process of mixed demand, including the static phase for reservation passengers and the dynamic phase for real‐time passengers. To enhance the computational efficiency, a hybrid heuristic algorithm combining spatiotemporal clustering and non‐dominated sorting genetic algorithm (NSGA)‐II is constructed to obtain the Pareto solutions set. An illustrative example of the Nguyen–Dupuis network is presented to demonstrate the validity of the algorithm. Subsequently, a large‐scale case study in Beijing evaluated the applicability of DRT in the real‐world network. The results reveal that dynamic DRT with compensation mechanism can substantially improve the system performance while ensuring the service quality. The response rate of passengers has been dramatically promoted to 80%. The operating profit has been enormously improved by up to 73%. Therefore, this study is radically conducive to understanding the passenger's decision‐making psychology while constructing a more cost‐efficient flexible strategy for the service provider.

Park and Ride (P&R) as a demand management tool has the effect of reducing traffic congestion in urban centers, saving energy and reducing pollutant emissions. Since 2000, many cities in China have been constructing P&R facilities, which have partially alleviated urban traffic congestion and provided a time-reliable mode of travel for commuters heading to urban centers. However, in recent years, due to the pricing policy of the P&R facility, there has been an insufficient supply of P&R facilities in many places. In fact, the P&R system prefers to welcome travelers who make long-distance subway rides and does not want those who make short-distance subway rides to occupy more parking spaces. To address this, this paper proposes a tiered pricing strategy that considers charging parking fees based on the distance traveled by commuters after switching to public transportation, to improve the utilization of P&R. That is, charge less for parking for long-distance subway riders and more for short-distance subway riders. Firstly, based on questionnaire data from SP surveys, a fixed pricing mixed logit model (FP model) and a tiered pricing mixed logit model (TP model) for P&R facilities are constructed. Utilizing two models, we explored the mechanisms underpinning traveler’s mode choice influenced by daily habits and travel considerations through the comparison of the two models to validate the effectiveness of the tiered pricing for P&R facilities. The study found that the implementation of a tiered pricing method for P&R facilities increases its attractiveness to long-distance subway ride travelers, resulting in a higher proportion of long-distance subway riders among P&R commuters. In the study’s last section, a marginal effect analysis was conducted on the per-kilometer cost (Pkm) within the P&R model. This analysis determined the optimal Pkm for three subway travel distances within the P&R model. Consequently, it calculated the corresponding P&R parking fees for these three subway travel distances. Additionally, we have predicted the implementation effects of the tiered pricing scheme.

BackgroundSeasons were found to be related to the occurrences of venous thromboembolism (VTE) in hospitalized patients. No previous study has explored whether seasons were associated with VTE risk in pregnant women. This study aimed to investigate the relationships between the season of delivery and VTE risk during hospitalization among pregnant women.MethodsThis is a multi-center retrospective cohort study of pregnant women. Participants were those who delivered at seven designated sites in Hubei Province, China, during the period from January 2017 to December 2022. They were categorized according to their season/month of delivery. Information on new-onset VTE during hospitalization was followed.ResultsApproximately 0.28% (104/37,778) of the pregnant women developed new-onset VTE during hospitalization for delivery. After adjustment, compared with participants in the spring group, participants in the summer, autumn, and winter groups had an increased risk of VTE during hospitalization. The ORs were 2.59 [1.30, 5.15], 2.83 [1.43, 5.60], and 2.35 [1.17, 4.75] for the summer, autumn, and winter groups, respectively. Pregnant women in the combined group (summer + autumn + winter) had an increased risk of VTE during hospitalization than those in the spring group (OR, 2.59 [1.39, 4.85]). By restricting the analyses among pregnant women without in vitro fertilization, gestational diabetes mellitus, and preterm, the results still remained robust. Compared with participants who delivered in March, April, and May, participants who delivered in June, July, September, November, December, and February had a higher risk of VTE during hospitalization.ConclusionThis study demonstrated that pregnant women who delivered in summer, autumn, and winter had an increased VTE risk during hospitalization compared with those who delivered in spring.

Summary: Background: Adolescent idiopathic scoliosis (AIS) affects up to 5% of the population. The efficacy of school-aged screening remains controversial since it is uncertain which curvatures will progress following diagnosis and require treatment. Patient demographics, vertebral morphology, skeletal maturity, and bone quality represent individual risk factors for progression but have yet to be integrated towards accurate prognostication. The objective of this work was to develop composite machine learning-based prediction model to accurately predict AIS curves at-risk of progression. Methods: 1870 AIS patients with remaining growth potential were identified. Curve progression was defined by a Cobb angle increase in the major curve of ≥6° between first visit and skeletal maturity in curves that exceeded 25°. Separate prediction modules were developed for i) clinical data, ii) global/regional spine X-rays, and iii) hand X-rays. The hand X-ray module performed automated image classification and segmentation tasks towards estimation of skeletal maturity and bone mineral density. A late fusion strategy integrated these domains towards the prediction of progressive curves at first clinic visit. Findings: Composite model performance was assessed on a validation cohort and achieved an accuracy of 83.2% (79.3–83.6%, 95% confidence interval), sensitivity of 80.9% (78.2–81.9%), specificity of 83.6% (78.8–84.1%) and an AUC of 0.84 (0.81–0.85), outperforming single modality prediction models (AUC 0.65–0.78). Interpretation: The composite prediction model achieved a high degree of accuracy. Upon incorporation into school-aged screening programs, patients at-risk of progression may be prioritized to receive urgent specialist attention, more frequent follow-up, and pre-emptive treatment. Summary: Background: Two previous studies found alkaline phosphatase (ALP) levels were related with the development of venous thromboembolism (VTE) in hospitalised patients. VTE is a leading cause of death during pregnancy and postpartum. No prior study has investigated the associations of ALP levels and VTE postpartum, and the related mechanisms remain unclear. This study aimed to investigate the associations between ALP levels and VTE postpartum, and to reveal the potential mechanisms. Methods: In this retrospective cohort study, we included pregnant women who planned to deliver at the Department of Obstetrics and Gynecology in the three designated hospitals in a multicentre cohort of pregnant women in Wuhan, China, during two recruitment periods of January 1, 2018 to December 31, 2019, and May 14, 2020 to March 25, 2022. A total of 10,044 participants with serum ALP and whole blood hemoglobin measurements in late pregnancy (median, 37 (35, 39) weeks) were enrolled. The participants’ incidences of VTE (deep venous thrombosis and/or pulmonary embolism) postpartum were confirmed from the medical records. Pregnant women with new-onset VTE postpartum (within 6 weeks after delivery) were confirmed as VTE cases. Findings: Approximately 0.8% (79/10,044) of the pregnant women were diagnosed with VTE postpartum. In the unadjusted model, pregnant women with the lowest quintile of serum ALP levels (≤116 U/L) in late pregnancy had higher risk of VTE postpartum compared with those with the highest quintile (≥199 U/L) (OR, 2.83 [1.32, 6.05]). After adjusting for covariates of demographic, life style, birth outcomes, and other liver enzymes, pregnant women with the lowest quintile of serum ALP levels (≤116 U/L) in late pregnancy had increased risk of VTE postpartum compared with those with the highest quintile (≥199 U/L) (OR, 2.48 [1.14, 5.40]). A one standard deviation decrease of ln-transformed ALP levels were associated with elevated risk of VTE postpartum (OR, 1.29 [1.02, 1.62]). Significant negative associations of ALP with VTE were found in the unadjusted and adjusted models. The effect of exogenous γ-aminobutyric acid (GABA) and GABA synthesis inhibitor 3-mercaptopropionic acid (3-MP) on GABA metabolism and quality in fresh-cut pumpkin was investigated, and the possible mechanism was studied. The results indicated that exogenous GABA induced the accumulation of endogenous GABA and promoted the conversion of glutamic acid to GABA in fresh-cut pumpkin through the catalytic conversion of highly active glutamate decarboxylase (GAD). The 3-MP-treated pumpkin retained lower GABA content than the control, and the activities of GAD and GABA transaminase (GABA-T) were inhibited. The GABA-treated pumpkins possessed relatively lower polyamines (putrescine, spermine and spermidine) contents in comparison with the control during storage. 3-MP treatment restrained the activities of polyamine oxidase (PAO), diamine oxidase (DAO) and amino aldehyde decarboxylase (AMADH). These results demonstrated that GABA treatment could activate the polyamine degradation pathway and promote GABA enrichment in pumpkin. Meanwhile, no significant differences in maximum colony count, color, β-carotene content and soluble solids content were observed among treatments during storage, which suggested that exogenous GABA had little effect on the quality of fresh-cut pumpkin.

With growing demands for portable electronics, flexible aqueous energy storage devices such as supercapacitors and Zn‐based batteries have drawn tremendous interest from both academic and industrial fields. Organo‐hydrogels, crosslinked amphiphilic polymers filled with organic solvents and water, are regarded as an ideal electrolyte candidate for portable electronic applications, especially due to their superior environmental adaptation. Organo‐hydrogels can reserve the advantages of the corresponding hydrogels and achieve some unique features such as broad temperature tolerance, high mechanical stability, strong interfacial interaction, and decent ionic conductivity. This review outlines the composition fundamentals, preparation methods, and comprehensive properties of reported organo‐hydrogel electrolytes. In particular, supercapacitors and Zn‐based batteries that operate under harsh temperature conditions and unusual mechanical deformations endowed by organo‐hydrogel electrolytes are further highlighted. Moreover, a unique perspective on the current challenges and future development directions of the organo‐hydrogel electrolytes for flexible energy storage is also provided.

With the deepening and expansion of semiconductor technology and research, in order to continuously optimize the structure and performance of semiconductor detectors, a high-precision hexagonal spiral silicon drift detector (SDD) is proposed in this paper. In order to obtain a more accurate spiral ring structure, this paper goes beyond the first-order formula in the Taylor expansion for calculating the radius of the spiral ring. Based on the first-order formula, the second-order formula for calculating the radius of the spiral ring is further developed and derived. The point coordinates are obtained by combining the radius, angle, and ring spacing change formula to obtain a more accurate spiral ring structure. The actual number of turns is more accurate than that obtained from first-order approximation, which better solves the problem of accurate calculation of the number of spiral rings and the structure of the spiral SDD in the existing technology, that is, the accurate calculation of the radius of the spiral ring. In order to verify the abovementioned theory, we model this new structure and use Technology Computer-Aided Design to system simulate and study its electrical properties, including potential distribution, electric field distribution, and electron concentration distribution. According to the simulation results, compared with the first-order formula, the second-order formula has better electrical properties; more uniform distribution of potential, electric field, and electron concentration; and a clearer electron drift channel.

Among the 3D electrode Si detectors for high energy particle and X-ray detection, the traditional 3D-Trench electrode Si detector is a semiconductor detector that is widely used and discussed. Aiming at removing the shortcomings of the traditional 3D-Trench electrode Si detectors such as uneven electric field distribution, asymmetric electric potential, and the existence of some dead zone, we propose a new type 3D-Spherical Electrode Detectors and carry out extensive and systematic studies of their physical properties. We simulated detector electric field, electric potential, electron concentration distribution, full depletion voltage, leakage current, capacitance, the incident particle induced transient current and the weighting field. We systematically studied and analyzed detector’s electrical characteristics. By comparing with the traditional 3D-Trench electrode Si detectors, the new detector structure has more uniform electric field and potential, and less depletion voltage, leakage current and capacitance.

The accurate acquisition of the residual stress field is the key to clarifying the cold expansion strengthening mechanism of compression bushings, optimizing the extrusion process parameters, and improving the structural fatigue life. In the actual cold expansion strengthening process, the resultant distribution of residual stresses is influenced by the relative extrusion amount, mandrel structure, material properties, and extrusion speed. In this article, the distribution patterns of residual stress after cold extrusion are investigated through a combination of finite element simulation and experimental measurements using a micro-region stress tester. To examine the redistribution law of the stress field of cold expansion reinforcement under external load, the compression bushing-lug-plate-reinforced structure is loaded and unloaded. The results show that large circumferential residual compressive stresses are distributed in the hole wall of the compression bushing after cold expansion. Radial residual stresses are also compressive stresses, although the values are small. In addition, the reinforced structure after cold extrusion presents a large difference in the stress redistribution rules compared with cold extrusion after the load is applied and removed.

In our previous studies, the silicon drift detector (SDD) structure with a constant spiral ring cathode gap (g) and a given surface electric field has been partially investigated based on the physical model that gives an analytical solution to the integrals in the calculations. Those results show that the detector has excellent electrical characteristics with a very homogeneous carrier drift electric field. In order to cope with the implementation of the theoretical approach with a complete set of technical parameters, this paper performs different theoretical algorithms for the technical implementation of the detector performance using the Taylor expansion method to construct a model for cases where the parameter “j” is a non-integer, approximating the solution with finite terms. To verify the accuracy of this situation, we performed a simulation of the relevant electrical properties using the Sentaurus TCAD tool 2018. The electrical properties of the single and double-sided detectors are first compared, and then the effects of different equal gaps g (g = 10 μm, 20 μm, and 25 μm, respectively) on the electrical properties of the double-sided detectors are analyzed and demonstrated. By analyzing and comparing the electrical characteristics data from the simulation results, we can show that the double-sided structure has a larger transverse drift electric field, which improves the spatial position resolution as well as the response speed. The effect of the gap size on the electrical characteristics of the detector is also analyzed by analyzing three different gap bifacial detectors, and the results show that a 10 μm equal gap is the optimal design. Such results can be used in applications requiring large-area SDD, such as the pulsar X-ray autonomous navigation. in the future to provide navigation and positioning space services for spacecraft deep-space exploration.

Rice blast is arguably the most devastating fungal disease of rice. Utilization of resistance genes to breed resistant cultivars is one of the most economical and environmentally friendly approaches to combat the disease. Pi25, a major resistance gene conferring broad-spectrum resistance to both leaf and neck blast, is an ideal gene resource to improve the resistance of rice varieties to blast. Recently, several allele-specific markers were developed. However, they were deficiently efficient due to either an additional process of restriction enzyme digestion for cleaved amplified polymorphic sequence (CAPS) markers or the risk of false-positive error in identifying susceptible Tetep allele (Pi25TTP) for PCR-based markers. In this study, based on a conserved single nucleotide polymorphism (SNP) between resistant and susceptible alleles, a tetra-primer amplification refractory mutation system (ARMS)-PCR marker was developed. The new marker, namely Pi25-2687R3, could effectively distinguish the resistant Gumei 2 (GM2) allele (Pi25GM2) and the susceptible allele Pi25TTP. Moreover, a perfect consistency of genotyping was exhibited between Pi25-2687R3 and published CAPS marker CAP3/Hpy 99I. A more accurate genotyping was also displayed compared to the previous PCR-based SNP marker Pi25-2566. Our finding proved that Pi25-2687R3 could achieve the same result as CAP3/Hpy 99I with less workload and cost and could promote the accuracy in the identification of genotypes superior to Pi25-2566. This study provided a quick and reliable functional marker for discriminating Pi25 alleles, which would be a valuable tool for genotypic assay and rice molecular breeding of blast resistance.

Abstract Twisted bilayer systems with discrete magic angles, such as twisted bilayer graphene featuring moiré superlattices, provide a versatile platform for exploring novel physical properties. Here, we discover a class of superflat bands in general twisted bilayer systems beyond the low-energy physics of magic-angle twisted counterparts. By considering continuous lattice dislocation, we obtain intrinsic localized states, which are spectrally isolated at lowest and highest energies and spatially centered around the AA stacked region, governed by the macroscopic effective energy potential well. Such localized states exhibit negligible inter-cell coupling and support the formation of superflat bands in a wide and continuous parameter space, which can be mimicked using a twisted bilayer nanophotonic system. Our finding suggests that general twisted bilayer systems can realize continuously tunable superflat bands and the corresponding localized states for various photonic, phononic, and mechanical waves.

Abstract Background Metastasis is the leading cause of mortality in human cancers, including esophageal squamous cell carcinoma (ESCC). As a pro-inflammatory cytokine, IL-32 was reported to be a poor prognostic factor in many cancers. However, the role of IL-32 in ESCC metastasis remains unknown. Methods ESCC cells with ectopic expression or knockdown of IL-32 were established and their effects on cell motility were detected. Ultracentrifugation, Transmission electron microscopy and Western blot were used to verify the existence of extracellular vesicle IL-32 (EV-IL-32). Coculture assay, immunofluorescence, flow cytometry, and in vivo lung metastasis model were performed to identify how EV-IL-32 regulated the crosstalk between ESCC cells and macrophages. Results Here, we found that IL-32 was overexpressed and positively correlated to lymph node metastasis of ESCC. IL-32 was significantly higher in the tumor nest compared with the non-cancerous tissue. We found that IL-32β was the main isoform and loaded in EV derived from ESCC cells. The shuttling of EV-IL-32 derived from ESCC cells into macrophages could promote the polarization of M2 macrophages via FAK-STAT3 pathway. IL-32 overexpression facilitated lung metastasis and was positively correlated with the proportion of M2 macrophages in tumor microenvironment. Conclusions Taken together, our results indicated that EV-IL-32 derived from ESCC cell line could be internalized by macrophages and lead to M2 macrophage polarization via FAK-STAT3 pathway, thus promoting the metastasis of ESCC. These findings indicated that IL-32 could serve as a potential therapeutic target in patients with ESCC.

Photonic topological insulators with robust boundary states can enable great applications for optical communication and quantum emission, such as unidirectional waveguide and single-mode laser. However, because of the diffraction limit of light, the physical insight of topological resonance remains unexplored in detail, like the dark line that exists with the crystalline symmetry-protected topological edge state. Here, we experimentally observe the dark line of the Z2 photonic topological insulator in the visible range by photoluminescence and specify its location by cathodoluminescence characterization, and elucidate its mechanism with the p-d orbital electromagnetic field distribution which calculated by numerical simulation. Our investigation provides a deeper understanding of Z2 topological edge states and may have great significance to the design of future on-chip topological devices.

Abstract Background Despite surgical advances, acute type A aortic dissection remains a life-threatening disease with high mortality and morbidity. Tracheostomy is usually used for patients who need prolonged mechanical ventilation in the intensive care unit (ICU). However, data on the risk factors for requiring tracheostomy and the impact of tracheostomy on outcomes in patients after Stanford type A acute aortic dissection surgery (AADS) are limited. Methods A retrospective single-institutional study including consecutive patients who underwent AADS between January 2016 and December 2019 was conducted. Patients who died intraoperatively were excluded. Univariate analysis and multivariate logistic regression analysis were used to identify independent risk factors for postoperative tracheostomy (POT). A nomogram to predict the probability of POT was constructed based on independent predictors and their beta-coefficients. The area under the receiver operating characteristic curve (AUC) was performed to assess the discrimination of the model. Calibration plots and the Hosmer–Lemeshow test were used to evaluate calibration. Clinical usefulness of the nomogram was assessed by decision curve analysis. Propensity score matching analysis was used to analyze the correlation between requiring tracheostomy and clinical prognosis. Results There were 492 patients included in this study for analysis, including 55 patients (11.2%) requiring tracheostomy after AADS. Compared with patients without POT, patients with POT experienced longer ICU and hospital stay and higher mortality. Age, cerebrovascular disease history, preoperative white blood cell (WBC) count and renal insufficiency, intraoperative amount of red blood cell (RBC) transfusion and platelet transfusion were identified as independent risk factors for POT. Our constructed nomogram had good discrimination with an AUC = 0.793 (0.729–0.856). Good calibration and clinical utility were observed through the calibration and decision curves, respectively. For better clinical application, we defined four intervals that stratified patients from very low to high risk for occurrence of POT. Conclusions Our study identified preoperative and intraoperative risk factors for POT and found that requiring tracheostomy was related to the poor outcomes in patients undergoing AADS. The established prediction model was validated with well predictive performance and clinical utility, and it may be useful for individual risk assessment and early clinical decision-making to reduce the incidence of tracheostomy.

Diabetic ulcer (DU) has been recognized as one of the most prevalent and serious complications of diabetes. However, the clinical efficacy of standard treatments for DU remains poor. Traditional Chinese medicine (TCM) shows a positive therapeutic effect on DU. Specifically, Zizhu ointment (ZZO) has been widely used to treat DU in long-term clinical practice, but the exact mechanism by which it promotes DU wound healing remains unknown. In this study, network analysis and high-performance liquid chromatography–high resolution mass spectrometry (UPLC-HRMS) were conducted to identify the active compounds of ZZO. We detected isovalerylshikonin (ISO), mandenol, daidzein, kaempferol, and formononetin in both network analysis and UPLC-HRMS. Moreover, ZZO could ameliorate DU by regulating the phosphatidylinositol-3-kinase (PI3K)/protein kinase B (AKT) and inflammation signaling pathways, according to the results of KEGG analysis. We established a DU mouse model with a high-fat diet and streptozotocin injection in vivo to evaluate the network analysis result. The experimental results showed that ZZO could inhibit inflammation, remodel fibrous tissue, and promote angiogenesis in the DU area, facilitating wound healing in DU mice. Moreover, the PI3K/AKT signaling pathway was indeed activated by ZZO treatment, promoting macrophage M2 polarization. In addition, we used molecular docking technology to evaluate the binding sites between ZZO and the PI3K/AKT pathway. The results showed that ISO has a good binding interaction with AKT. Moreover, ISO promoted M2 polarization in macrophages in a dose-dependent manner in vitro. Our study found that ZZO could promote DU wound healing by inhibiting inflammation, which was achieved by macrophage M2 polarization through activating the PI3K/AKT pathway. Further studies have demonstrated that ISO plays major role in the above process. These findings provide a theoretical basis for further preclinical evaluation and lay a foundation for nano-gel compound treatment with ZZO.

Abstract Background Pneumonia is a common complication after Stanford type A acute aortic dissection surgery (AADS) and contributes significantly to morbidity, mortality, and length of stay. The purpose of this study was to identify independent risk factors associated with pneumonia after AADS and to develop and validate a risk prediction model. Methods Adults undergoing AADS between 2016 and 2019 were identified in a single-institution database. Patients were randomly divided into training and validation sets at a ratio of 2:1. Preoperative and intraoperative variables were included for analysis. A multivariate logistic regression model was constructed using significant variables from univariate analysis in the training set. A nomogram was constructed for clinical utility and the model was validated in an independent dataset. Results Postoperative pneumonia developed in 170 of 492 patients (34.6%). In the training set, multivariate analysis identified seven independent predictors for pneumonia after AADS including age, smoking history, chronic obstructive pulmonary disease, renal insufficiency, leucocytosis, low platelet count, and intraoperative transfusion of red blood cells. The model demonstrated good calibration (Hosmer–Lemeshow χ2 = 3.31, P = 0.91) and discrimination (C-index = 0.77) in the training set. The model was also well calibrated (Hosmer–Lemeshow χ2 = 5.73, P = 0.68) and showed reliable discriminatory ability (C-index = 0.78) in the validation set. By visual inspection, the calibrations were good in both the training and validation sets. Conclusion We developed and validated a risk prediction model for pneumonia after AADS. The model may have clinical utility in individualized risk evaluation and perioperative management.

The NAC (NAM, ATAF1/2 and CUC2) transcription factors are ubiquitously distributed in plants and play critical roles in the construction of plant organs and abiotic stress response. In this study, we described the cloning of a Suaeda liaotungensis K. NAC transcription factor gene SlNAC4, which contained 1450 bp, coding a 331 amino acid. We found that SlNAC4 was highly expressed in stems of S. liaotungensis, and the expression of SlNAC4 was considerably up-regulated after salt, drought, and ABA treatments. Transcription analysis and subcellular localization demonstrated that the SlNAC4 protein was located both in the nucleus and cytoplasm, and contained a C-terminal transcriptional activator. The SlNAC4 overexpression Arabidopsis lines significantly enhanced the tolerance to salt and drought treatment and displayed obviously increased activity of antioxidant enzymes under salt and drought stress. Additionally, transgenic plants overexpressing SlNAC4 had a significantly higher level of physiological indices. Interestingly, SlNAC4 promoted the expression of ABA metabolism-related genes including AtABA1, AtABA3, AtNCED3, AtAAO3, but inhibited the expression of AtCYP707A3 in overexpression lines. Using a yeast one-hybrid (Y1H) assay, we identified that the SlNAC4 transcription factor could bind to the promoters of those ABA metabolism-related genes. These results indicate that overexpression of SlNAC4 in plants enhances the tolerance to salt and drought stress by regulating ABA metabolism.

Diamond tools are extensively used in ultra-precision machining due to their exceptional performance. However, when machining challenging materials like Ti6Al4V, diamond tools experience significant wear due to poor machining properties and catalytic effects. Tool wear not only impacts machining quality but also escalates machining costs and energy consumption. Cutting fluids are commonly employed to mitigate interfacial reactions and suppress tool wear. However, traditional cutting fluids are difficult to penetrate the cutting area and have limited lubrication and cooling capabilities. Therefore, in this paper, a technique combining graphene nanofluid and minimum-quantity lubrication (MQL) is used to suppress diamond tool wear. Firstly, micro-milling experiments for Ti6Al4V alloy are conducted using diamond tools in the graphene nanofluid MQL and under a dry environment. The experimental results show that tool wear is effectively suppressed by graphene nanofluids. Subsequently, the cutting process in both environments (graphene nanofluid MQL, dry) is simulated. The suppression mechanism of graphene nanofluid MQL for diamond tool wear is evaluated from phase transition, atomic transfer process, and amorphous behavior of diamond structure. The simulation results show that the contact characteristics, cutting force, and cutting temperature are improved by graphene nanofluids. Tool wear is effectively reduced. In addition, the removal efficiency of workpiece materials has also been improved. This work provides a technical basis for exploring the application of graphene nanofluids in diamond tool damage suppression and micro-milling.

Measuring the correlation between belief functions is an important issue in Dempster–Shafer theory. From the perspective of uncertainty, analyzing the correlation may provide a more comprehensive reference for uncertain information processing. However, existing studies about correlation have not combined it with uncertainty. In order to address the problem, this paper proposes a new correlation measure based on belief entropy and relative entropy, named a belief correlation measure. This measure takes into account the influence of information uncertainty on their relevance, which can provide a more comprehensive measure for quantifying the correlation between belief functions. Meanwhile, the belief correlation measure has the mathematical properties of probabilistic consistency, non-negativity, non-degeneracy, boundedness, orthogonality, and symmetry. Furthermore, based on the belief correlation measure, an information fusion method is proposed. It introduces the objective weight and subjective weight to assess the credibility and usability of belief functions, thus providing a more comprehensive measurement for each piece of evidence. Numerical examples and application cases in multi-source data fusion demonstrate that the proposed method is effective.

Abstract Background Hyperlactatemia may be caused by increased production due to tissue hypoxia or non-hypoxia. The aim of this study was first to identify risk factors for postoperative hyperlactatemia (POHL) after Stanford type A acute aortic dissection surgery (AADS) and construct a predictive model, and second to evaluate the impact of POHL on prognosis. Methods This retrospective study involved patients undergoing AADS from January 2016 to December 2019 in Wuhan Union Hospital. Multivariate logistic regression analysis was performed to identify independent risk factors for POHL. A nomogram predicting POHL was established based on these factors and was validated in the original dataset. The receiver operating characteristic curve was drawn to assess the ability of postoperative lactate levels to predict the in-hospital mortality. Results A total of 188 patients developed POHL after AADS (38.6%). Male gender, surgery history, red blood cell transfusion and cardiopulmonary bypass time were identified as independent predictors. The C-index of the prediction model for POHL was 0.72, indicating reasonable discrimination. The model was well calibrated by visual inspection and goodness-of-fit test (Hosmer–Lemeshow χ2 = 10.25, P = 0.25). Decision and clinical impact curves of the model showed good clinical utility. The overall in-hospital mortality rate was 10.1%. Postoperative lactate levels showed a moderate predictive power for postoperative in-hospital mortality (C-index: 0.72). Conclusion We developed and validated a prediction model for POHL in patients undergoing AADS, which may have clinical utility in personal risk evaluation and preventive interventions. The POHL could be a good predictor for in-hospital mortality.

Samples of pure titanium with a purity of 99.99 wt.% were successfully processed by equal channel angular pressing (ECAP) at cryogenic temperature. Deformed microstructures of ECAPed samples were characterized by electron backscattering diffraction (EBSD), and its homogeneity was studied by color-coded contour mapping of microhardness distribution. EBSD results indicate the original coarse grains with an average grain size of 196 μm were significantly refined to 0.56 μm after four passes, and that the prism slip systems were activated in the whole deformation process and with increasing passes number more slip systems were activated. The hardness mapping reveals that the microstructure of samples prior to ECAP is basically homogeneous but becomes inhomogeneous after one pass and then gradually homogeneous with additional passes. In addition, the grain refinement of ECAPed titanium at cryogenic temperature was discussed and confirmed by CDRX. Also, the factors of influence on the homogeneity of microstructure were analyzed.

With advancements in artificial intelligence, wearable motion recognition systems based on flexible nanomaterial sensors exhibit excellent potential for harmonious human–machine interaction. However, the sensing stability and demand of large‐scale arrays limit the application of flexible nanomaterial sensors. Herein, a data glove system based on simple multiwalled carbon nanotube (MWCNT) sensors and a lightweight deep‐learning algorithm to achieve accurate gesture recognition is proposed. A regional‐crack mechanism is introduced through the microspine structure to enhance the strain sensitivity. Moreover, an efficient signal processing strategy based on an adaptive wavelet threshold function to improve the robustness and anti‐interference of signals obtained from MWCNT sensors, which exhibit strong generalization and can be used in other nanomaterial strain sensor. Based on the depth‐wise separable convolution, a novel hybrid convolutional neural network (CNN) long short‐term memory (LSTM) model for gesture recognition is constructed. The proposed model achieves an average accuracy of 97.5% and recognition accuracy of 30 gestures with an average recognition time of 2.173 ms based on only five sensors. The fabricated data glove is a promising platform for low‐cost and wearable human–machine interaction that can be directly interfaced in applications such as robotic hands, smart cars, and first‐person shooting games.

Background The development of cancer is largely dependent on the accumulation of somatic mutations, indicating the potential to develop cancer chemoprevention agents targeting mutation drivers. However, ideal cancer chemoprevention agents that can effectively inhibit the mutation drivers have not been identified yet.Methods The somatic mutation signatures and expression analyses of APOBEC3B were performed in patient with pan-cancer. The computer-aided screening and skeleton-based searching were performed to identify natural products that can inhibit the activity of APOBEC3B. 4-nitroquinoline-1-oxide (4-NQO)-induced spontaneous esophageal squamous cell carcinoma (ESCC) and azoxymethane/dextran sulfate sodium (AOM/DSS)-induced spontaneous colon cancer mouse models were conducted to investigate the influences of APOBEC3B inhibitor on the prevention of somatic mutation accumulation and cancer progression.Results Here, we discovered that the cytidine deaminase APOBEC3B correlated somatic mutations were widely observed in a variety of cancers, and its overexpression indicated poor survival. SMC247 (3, 5-diiodotyrosine), as a source of kelp iodine without side effects, could strongly bind APOBEC3B (KD=65 nM) and effectively inhibit its deaminase activity (IC50=1.69 µM). Interestingly, 3, 5-diiodotyrosine could significantly reduce the clusters of mutations, prevent the precancerous lesion progression, and prolong the survival in 4-NQO-induced spontaneous ESCC and AOM/DSS-induced spontaneous colon cancer mouse models. Furthermore, 3, 5-diiodotyrosine could reduce colitis, increase the proportion and function of T lymphocytes via IL-15 in tumor microenvironment. The synergistic cancer prevention effects were observed when 3, 5-diiodotyrosine combined with PD-1/PD-L1 blockade.Conclusions This is the first prove-of-concept study to elucidate that the natural product 3, 5-diiodotyrosine could prevent somatic mutation accumulation and cancer progression through inhibiting the enzymatic activity of APOBEC3B. In addition, 3, 5-diiodotyrosine could reduce the colitis and increase the infiltration and function of T lymphocytes via IL-15 in tumor microenvironment. 3, 5-diiodotyrosine combined with PD-1/PD-L1 blockade could elicit synergistic cancer prevention effects, indicating a novel strategy for both prevent the somatic mutation accumulation and the immune-suppressive microenvironment exacerbation.

Various strategies to assemble protein building blocks into one-, two- and three-dimensional hierarchical nanostructures were described, but controlling the transformation between those different assemblies is largely uninvestigated. Here, the authors describe a protein interface redesign strategy and use it for the self-assembly transformation of dimeric building blocks from hollow protein nanocage to filament, nanorod and nanoribbon.

In this paper, two types of adsorption materials SA-C-Fe and SA-C-Fe(C) were prepared using bagasse biochar produced by one-step microwave pyrolysis and activation for Cr(VI) removal of wastewater. The adsorption materials were characterized, and Cr(VI) adsorption performance, kinetics and thermodynamics on adsorption materials were studied. Results show that microwave pyrolysis/activation contributes to developed pore structure and abundant active functional groups, resulting in high Cr(VI) adsorption capacities. The optimal preparation conditions for biochar is: microwave power 500 W, ZnCl2/bagasse ratio 2.5:1 and pyrolysis/activation time 15 min, and the specific surface area of biochar is 1,787.64 m2/g. The Cr(VI) adsorption of the two materials is more in line with the pseudo-second-order kinetic model, and the adsorption process is dominated by chemical adsorption. The static removal experiment of Cr(VI) using SA-C-Fe and SA-C-Fe(C) has the best removal effect at pH = 2, and the whole adsorption process is more in line with the Langmuir-Freundlich isotherm model. Calculated by the pseudo-two-order kinetic model and the Langmuir-Freundlich isothermal model, the maximum adsorption rate for Cr(VI) of SA-C-Fe and SA-C-Fe(C) are 211.87 mg/g and 388.92 mg/g, respectively. The removal process is mainly dominated by three mechanisms: electrostatic adsorption, ion exchange and redox reactions. The improvement of Cr(VI) adsorption capacity is attributed to more developed pore structure. The results offer beneficial reference for the application of low-cost carbon-based adsorption materials for pollutants separation, and effectively realize the utilization of bagasse pyrolysis by-products.

ObjectiveTo compare the performance of clinical factors, FS-T2WI, DWI, T1WI+C based radiomics and a combined clinic-radiomics model in predicting the type of serous ovarian carcinomas (SOCs).MethodsIn this retrospective analysis, 138 SOC patients were confirmed by histology. Significant clinical factors (P < 0.05, and with the area under the curve (AUC) > 0.7) was retained to establish a clinical model. The radiomics model included FS-T2WI, DWI, and T1WI+C, and also, a multisequence model was established. A total of 1,316 radiomics features of each sequence were extracted; the univariate and multivariate logistic regressions, cross-validations were performed to reduce valueless features and then radiomics signatures were developed. Nomogram models using clinical factors, combined with radiomics features, were developed in the training cohort. The predictive performance was validated by receiver operating characteristic curve (ROC) analysis and decision curve analysis (DCA). A stratified analysis was conducted to compare the differences between the combined radiomics model and the clinical model in identifying low- and high-grade SOC.ResultsThe AUC of the clinical model and multisequence radiomics model in the training and validation cohorts was 0.90 and 0.89, 0.91 and 0.86, respectively. By incorporating clinical factors and multi-radiomics signature, the AUC of the radiomic-clinical nomogram in the training and validation cohorts was 0.98 and 0.95. The model comparison results show that the AUC of the combined model is higher than that of the uncombined models (P= 0.05, 0.002).ConclusionThe nomogram models of clinical factors combined with MRI multisequence radiomics signatures can help identifying low- and high-grade SOCs and a provide a more comprehensive, effective method to evaluate preoperative risk stratification for SOCs.

The appointed-time leader-following consensus problem for second-order multi-agent systems with external disturbance on directed graphs is addressed. A distributed controller based on the cumulative position difference and cumulative velocity difference is proposed, which does not require prior knowledge of external disturbances. It is shown that the proposed controller guarantees the prescribed performance of the controlled systems, namely, to keep the cumulative position difference within a predefined boundary envelope. Furthermore, by employing a novel performance function, it is ensured that the position tracking error converges to an arbitrarily small expected region within the appointed time. Different from most existing finite/fixed-time control methods, here the settling time and the convergence region can both be predefined, which are also independent of initial conditions and the system parameters. Finally, the theoretical results are verified by simulations.

Soil salinization is an increasing agricultural problem around the world, affecting crop productivity and quality. Seed germination and seedling establishment are susceptible to salt stress. Suaeda liaotungensis is a halophyte with strong salt tolerance that produces dimorphic seeds to adapt to the saline environment. Differences in physiological characteristics, seed germination, and seedling establishment in response to salt stress between dimorphic seeds in S. liaotungensis have not been reported. The results showed that brown seeds had significantly higher H2O2 and O2−. levels and betaine content, as well as POD and CAT activities, while they had significantly lower MDA and proline contents and SOD activity than black seeds. Light promoted the germination of brown seeds in a certain temperature range, and brown seeds could reach a higher germination percentage in a wide temperature range. However, light and temperature had no effect on the germination percentage of black seeds. Brown seeds had higher germination than black seeds under the same NaCl concentration. The final germination of brown seeds was significantly decreased as salt concentration increased, whereas this had no effect on the final germination of black seeds. POD and CAT activities, as well as MDA content, in brown seeds were significantly higher than those in black seeds during germination under salt stress. Additionally, the seedlings from brown seeds were more tolerant to salinity than those from black seeds. Therefore, these results will give an in-depth understanding of the adaptation strategies of dimorphic seeds to a salinization environment, and better exploitation and utilization of S. liaotungensis.

Abstract Background Seed dimorphism has been thought to be a bet-hedging strategy that helps plants survive in the disturbed environment and has been widely studied for its ecological adaptation mechanism. Many studies showed that seed-associated microorganisms play an important role in enhancing plant fitness, but information regarding endophytic bacteria associated with dimorphic seeds is limited. This study explores the influence of seed coat structure and seed phytochemical properties on the community composition and diversity of endophytic bacteria of dimorphic seeds of Suaeda glauca. In this study, we used 16S rRNA high-throughput gene sequencing method to compare the community composition and bacterial diversity between brown and black seeds of Suaeda glauca. Results A significant difference was observed in seed coat structure and phytochemical properties between brown and black seeds of S. glauca. Total 9 phyla, 13 classes, 31 orders, 53 families, 102 genera were identified in the dimorphic seeds. The dominant phyla were Proteobacteria, Firmicutes, and Actinobacteria. The results showed that seed dimorphism had little impact on the diversity and richness of endophytic bacterial communities but significantly differs in the relative abundance of the bacterial community between brown and black seeds. At the phylum level, Actinobacteria tend to be enriched significantly in brown seeds. At the genus level, Rhodococcus, Ralstonia, Pelomonas and Bradyrhizobium tend to be enriched significantly in brown seeds, while Marinilactibacillus was mainly found in black seeds. Besides, brown seeds harbored a large number of bacteria with plant-growth-promoting traits, whereas black seeds presented bacteria with enzyme activities (i.e., pectinase, cellulolytic and xylanolytic activities). Conclusion The endophytic bacterial community compositions were significantly different between dimorphic seeds of Suaeda glauca, and play an important role in the ecological adaptation of dimorphic seeds by performing different biological function roles. The endophytic bacterial communities of the dimorphic seeds may be influenced mainly by the seed coat structureand partly by the seed phytochemical characteristics. These findings provide valuable information for better understanding of the ecological adaptation strategy of dimorphic seeds in the disturbed environment.

An Fe(II) complex with DPA-Bpy (DPA-Bpy = N,N-bis(2-pyridinylmethyl)-2,20-bipyridine-6 -methanamine) as the ligand was synthesized and characterized to mimic bleomycin. The binding constants (Kb) of the complex with calf thymus DNA and human serum albumin (HSA) were quantitatively evaluated using fluorescence spectroscopy, with Kb as 5.53×105 and 2.40×104 M−1, respectively; the number of the average binding site (n) is close to 1. The thermodynamic analyses suggested that the electrostatic interactions exist between the complex and DNA, and the hydrogen bonding and Van der Waals force exist for the complex and HSA. The Fe complex exhibits cleavage ability toward pBR322 DNA, and the crystal structure of the HSA Fe complex adduct at 2.4 Å resolution clearly shows that His288 serves as the axial ligand of the Fe center complexed with a pentadentate DPA-Bpy ligand. Furthermore, the cytotoxicity of the complex was evaluated against HeLa cells. Both the Fe complex and HSA Fe complex adduct show obvious effect on cell proliferation with an IC50 of 1.18 and 0.82 μM, respectively; they induced cell apoptosis and arrested cell cycles at S phase. This study provides insight into the plausible mechanism underlying their metabolism and pharmacological activity.

BackgroundHypoxemia is a common complication after Stanford type A acute aortic dissection surgery (AADS), however, few studies about hypoxemia after AADS exist. The aims of this study were to identify independent risk factors for hypoxemia after AADS and to clarify its association with clinical outcomes.MethodsPatients undergoing AADS from 2016 to 2019 in our hospital were identified and used as a training set. Preoperative variables were first screened by univariate analysis and then entered into a multivariate logistic regression analysis to identify independent risk factors. A nomogram and an online risk calculator were constructed based on the logistic model to facilitate clinical practice and was externally validated in an independent dataset.ResultsSevere hypoxemia developed in 119 of the 492 included patients (24.2%) and poorer clinical outcomes were observed in these patients. Five independent risk factors for severe hypoxemia after AADS were identified by multivariate analysis, including older age, smoking history, renal insufficiency, higher body mass index, and white blood cell count. The model showed good calibration, discrimination, and clinical utility in the training set, and was well validated in the validation set. Risk stratification was performed and three risk groups were defined as low, medium, and high risk groups. Hypertension was identified as an independent risk factor for moderate hypoxemia besides the five predictors mentioned above, and renal insufficiency was not significant for mild hypoxemia by multivariate analysis. In addition, although frozen elephant trunk was associated with increased risk of postoperative hypoxemia in the univariate analysis, frozen elephant trunk was also not identified as an independent risk factor for postoperative hypoxemia in the multivariate analysis.ConclusionHypoxemia was frequent following AADS, related to poorer clinical outcomes. Predictors were identified and a nomogram as well as an online risk calculator predicting severe hypoxemia after AADS was developed and validated, which may be helpful for risk estimation and perioperative management.

Background Postoperative headache (POH) is frequent after cardiac surgery; however, few studies on risk factors for POH exist. The aims of the current study were to explore risk factors related to POH after elective cardiac surgery and to establish a predictive system. Methods and Results Adult patients undergoing elective open‐heart surgery under cardiopulmonary bypass from 2016 to 2020 in 4 cardiac centers were retrospectively included. Two thirds of the patients were randomly allocated to a training set and one third to a validation set. Predictors for POH were selected by univariate and multivariate analysis. POH developed in 3154 of the 13 440 included patients (23.5%) and the overall mortality rate was 2.3%. Eight independent risk factors for POH after elective cardiac surgery were identified, including female sex, younger age, smoking history, chronic headache history, hypertension, lower left ventricular ejection fraction, longer cardiopulmonary bypass time, and more intraoperative transfusion of red blood cells. A nomogram based on the multivariate model was constructed, with reasonable calibration and discrimination, and was well validated. Decision curve analysis revealed good clinical utility. Finally, 3 risk intervals were divided to better facilitate clinical application. Conclusions A nomogram model for POH after elective cardiac surgery was developed and validated using 8 predictors, which may have potential application value in clinical risk assessment, decision‐making, and individualized treatment associated with POH.

The current study examined a multiple mediator model based on the control-value theory. A sample of 433 Chinese junior middle school students participated in questionnaires assessing teacher-student relationships (TSRs), academic emotions, academic value (AV), and academic self-efficacy (ASE). English test scores were collected as an indicator of students' English academic performance. The results indicated the following: (a) for academic enjoyment, the TSR has an effect on academic achievement through the chain intermediary of AV and ASE; (b) for academic relaxation, the TSR has an effect on English academic achievement through the single intermediary of academic relaxation and the chain intermediary of AV, ASE, and academic relaxation; (c) for academic anxiety, the TSR has an effect on English academic achievement through the single intermediary of AV, the single intermediary of academic anxiety, and the chain intermediary of AV, ASE, and academic anxiety; (d) for academic boredom, the TSR has an effect on English academic achievement through the single intermediary of academic boredom. These findings enrich the relevant research on the control-value theory and provide important insights for improving students' English academic achievement.

A new type of 3D electrode detector, named here as the Implanted-Epi Silicon 3D-Spherical Electrode Detector, is proposed in this work. Epitaxial and ion implantation processes can be used in this new detector, allowing bowl-shaped electrodes to penetrate the silicon completely. The distance between the bowl cathode and the central collection electrode is basically the same, thus the total depletion voltage of Implanted-Epi Silicon 3D-Spherical Electrode Detectors is no longer directively correlated with the thickness of the silicon wafer, but only related to the electrode spacing. In this work, we model the device physics of this new structure and use a simulation program to conduct a systematic 3D simulation of its electrical characteristics, including electric potential and electric field distributions, electron concentration profile, leakage current, and capacitance, and compare it to the traditional 3D detectors. The theoretical and simulation study found that the internal electric potential of the new detector was smooth and no potential saddle point was found. The electric field is also uniform, and there is no zero field and a low electric field area. Compared with the traditional silicon 3D electrode detectors, the full depletion voltage is greatly reduced and the charge collection efficiency is improved. As a large electrode spacing (up to 500 μm) can be realized in the Implanted-Epi Silicon 3D-Spherical Electrode Detector thanks to their advantage of a greatly reduced full depletion voltage, detectors with large pixel cells (and thus small dead volume) can be developed for applications in photon science (X-ray, among others).
