Osteosarcoma is the most common malignant bone tumor without efficient management for improving 5-year event-free survival. Immunotherapy is also limited due to its highly immunosuppressive tumor microenvironment (TME). Pore-forming gasdermins (GSDMs)-mediated pyroptosis has gained increasing concern in reshaping TME, however, the expressions and relationships of GSDMs with osteosarcoma remain unclear. Herein, gasdermin E (GSDME) expression is found to be positively correlated with the prognosis and immune infiltration of osteosarcoma patients, and low GSDME expression was observed. A vector termed as LPAD contains abundant hydroxyl groups for hydrating layer formation was then prepared to deliver the GSDME gene to upregulate protein expression in osteosarcoma for efficient TME reshaping via enhanced pyroptosis induction. Atomistic molecular dynamics simulations analysis proved that the hydroxyl groups increased LPAD hydration abilities by enhancing coulombic interaction. The upregulated GSDME expression together with cleaved caspase-3 provided impressive pyroptosis induction. The pyroptosis further initiated proinflammatory cytokines release, increased immune cell infiltration, activated adaptive immune responses and create a favorable immunogenic hot TME. The study not only confirms the role of GSDME in the immune infiltration and prognosis of osteosarcoma, but also provides a promising strategy for the inhibition of osteosarcoma by pore-forming GSDME gene delivery induced enhanced pyroptosis to reshape the TME of osteosarcoma.

Abstract Glyphosate, widely used to manage weeds in cassava crops, simultaneously inhibits cassava growth, necessitating the development of herbicide-tolerant cassava varieties. In this study, screened 262 cassava varieties, identifying the glyphosate-resistant (GR) variety ZM8701 and the glyphosate-sensitive (GS) variety SC9. Transcriptomic analysis via Illumina sequencing revealed differentially expressed genes associated with resistance, including Cytochrome P450, GST, GT, ABC transporters, and others such as MIOX1, LHCA1, PPH, HSP26, HSP83A, and UGT73C5. Notably, the EMB3004 gene, involved in the biosynthesis of aromatic amino acids, was significantly upregulated in resistant varieties, suggesting a key role in countering glyphosate’s inhibition of the shikimic acid pathway. These genes are pivotal in enhancing cell wall biosynthesis, optimizing photosynthesis, and improving detoxification processes. This research elucidated the molecular mechanisms underlying cassava’s resistance to glyphosate, thereby laying the groundwork for breeding programs aimed at developing herbicide-resistant varieties. Abstract Background Developing and enriching genetic resources plays important role in the crop improvement. The flag leaf affects plant architecture and contributes to the grain yield of wheat (Triticum aestivum L.). The genetic improvement of flag leaf traits faces problems such as a limited genetic basis. Among the various genetic resources of wheat, Thinopyrum intermedium has been utilized as a valuable resource in genetic improvement due to its disease resistance, large spikes, large leaves, and multiple flowers. In this study, a recombinant inbred line (RIL) population was derived from common wheat Yannong15 and wheat-Th. intermedium introgression line SN304 was used to identify the quantitative trait loci (QTL) for flag leaf-related traits. Results QTL mapping was performed for flag leaf length (FLL), flag leaf width (FLW) and flag leaf area (FLA). A total of 77 QTLs were detected, and among these, 51 QTLs with positive alleles were contributed by SN304. Fourteen major QTLs for flag leaf traits were detected on chromosomes 2B, 3B, 4B, and 2D. Additionally, 28 QTLs and 8 QTLs for flag leaf-related traits were detected in low-phosphorus and drought environments, respectively. Based on major QTLs of positive alleles from SN304, we identified a pair of double-ended anchor primers mapped on chromosome 2B and amplified a specific band of Th. intermedium in SN304. Moreover, there was a major colocated QTL on chromosome 2B, called QFll/Flw/Fla-2B, which was delimited to a physical interval of approximately 2.9 Mb and contained 20 candidate genes. Through gene sequence and expression analysis, four candidate genes associated with flag leaf formation and growth in the QTL interval were identified. Conclusion These results promote the fine mapping of QFll/Flw/Fla-2B, which have pleiotropic effects, and will facilitate the identification of candidate genes for flag leaf-related traits. Additionally, this work provides a theoretical basis for the application of Th. intermedium in wheat breeding.

Preventing local tumor recurrence while promoting bone tissue regeneration is an urgent need for osteosarcoma treatment. However, the therapeutic efficacy of traditional photosensitizers is limited, and they lack the ability to regenerate bone. Here, a piezo-photo nanoheterostructure is developed based on ultrasmall bismuth/strontium titanate nanocubes (denoted as Bi/SrTiO3), which achieve piezoelectric field-driven fast charge separation coupling with surface plasmon resonance to efficiently generate reactive oxygen species. These hybrid nanotherapeutics are integrated into injectable biopolymer hydrogels, which exhibit outstanding anticancer effects under the combined irradiation of NIR and ultrasound. In vivo studies using patient-derived xenograft models and tibial osteosarcoma models demonstrate that the hydrogels achieve tumor suppression with efficacy rates of 98.6 % and 67.6 % in the respective models. Furthermore, the hydrogel had good filling and retention capabilities in the bone defect region, which exerted bone repair therapeutic efficacy by polarizing and conveying electrical stimuli to the cells under mild ultrasound radiation. This study provides a comprehensive and clinically feasible strategy for the overall treatment and tissue regeneration of osteosarcoma.

IntroductionAccumulating evidence has supported that gut microbiota and metabolite profiles play indispensable roles in the pathogenesis of colorectal cancer (CRC), which ranks as the third most common cancer and the second leading cause of cancer-related deaths worldwide. However, alterations in tumoral or circulating microbiomes in CRC remain incompletely understood. It has been well-documented that tissue or serum microbiomes with low microbial biomass could be screened by use of 2bRAD sequencing for microbiome (2bRAD-M) at the species resolution.MethodsIn order to validate the microbial biomarkers distinguishing CRC and the variations in microorganisms present in serum and tumors, we performed 2bRAD-M to characterize the microbiomes in serum and cancer tissues of CRC patients with and without lymph node or liver metastasis.ResultsThe composition of dominated microbiota in serum was different from that of tissue samples, while the microbial community composition of tumors was similar to that of the tumor-adjacent tissues. The analysis of α-diversity and β-diversity has revealed notable variations in serum microbiota diversities in CRC patients, particularly those with liver metastasis. Multiple CRC-specific microbial species, such as Moraxella A cinereus, Flavobacterium sp001800905, and Acinetobacter albensis, were identified in serum. Complicated functions and KEGG pathways were also confirmed in CRC according to the metastasis status.DiscussionThis study has found significant alterations in the microbial compositions and diversities in CRC and CRC-specific microbial species in both circulation and cancer tissues, which may serve as promising biomarkers for the screening, diagnosis and prognosis prediction of CRC. In particular, CRC-specific bacterial taxa are promising markers, holding transformative potentials in establishing personalized screening and risk stratification, refining much earlier non-invasive diagnostic approaches, and enhancing diagnostic sensitivity.

With the continuous development of IoT technology, the application of Long Rang wireless link transmission in urban environments has gradually increased, making reliable wireless link quality transmission particularly important. Traditional wireless link quality assessments in urban environments have not comprehensively considered the various factors affecting LoRa link quality, nor have they assessed link quality by integrating fundamental environmental factors with digital models. This paper proposes a LoRa link quality assessment model that combines static and dynamic environmental factors. First, this paper analyzes the factors influencing LoRa link quality transmission through extensive real-environment experiments. Then, the First Fresnel Zone concept is considered to simulate LoRa link transmission. This is done by constructing a three-dimensional model of the urban environment and a three-dimensional model of the First Fresnel Zone to measure the proportions of various factors affecting link transmission. This paper employs a Bidirectional Long Short-Term Memory method to establish a static environment evaluation model. The reliability of this model is demonstrated by comparing the Mean Squared Error between the scores of the natural environment and the predicted scores. By comparing with three other methods, it was found that the static parameter evaluation model used in this paper is optimal. Subsequently, the static parameter evaluation model scores were combined with dynamic parameters to establish a dynamic environment evaluation model through a CNN-GRU-Attention mechanism. This model considers the influencing factors of time series. The MSE comparison with natural environmental conditions revealed that the dynamic parameter evaluation model scores are sufficient to indicate the link quality in the current urban environment.

Abstract Background German chamomile (Matricaria chamomilla L.) is an important medicinal plant, and the essential oils in the flowers have various biological activities. Genetic transformation systems are important for plant quality improvement and molecular research. To the best of our knowledge, a genetic transformation system has not yet been reported for German chamomile. Results In this study, we developed Agrobacterium-mediated transformation protocols for German chamomile callus tissues. This involved optimizing key parameters, such as hygromycin and cefotaxime concentrations, bacterial density, and infection and co-culture durations. We also performed gas chromatography–mass spectrometry analysis to identify volatile compounds in non-transgenic and transgenic callus and hairy root tissues. Furthermore, to compare and verify the callus transformation system of German chamomile, we transferred McFPS to the hairy roots of German chamomile. The results showed that the optimal conditions for Agrobacterium-mediated callus tissue transformation were as follows: explant, petiole; cefotaxime concentration, 300 mg/L; hygromycin concentration, 10 mg/L; and bacterial solution concentration, OD600 = 0.6; callus transformation efficiency was the highest when the co-culture time was 3 days. Conclusions Establishment of a high-efficiency callus transformation system will lay the foundation for gene function identification in German chamomile.

LoRa (long range) is a communication technology that employs chirp spread spectrum modulation. Among various low-power wide area network (LPWAN) technologies, LoRa offers unique advantages, including low power consumption, long transmission distance, strong anti-interference capability, and high network capacity. Addressing the issue of power-constrained end devices in IoT application scenarios, this paper proposes an adaptive data rate (ADR) algorithm for LoRa networks designed for power-constrained end devices (EDs). The algorithm evaluates the uplink communication link state between the EDs and the gateway (GW) by using a combined weighting method to comprehensively assess the signal-to-noise ratio (SNR), received signal strength indication (RSSI), and packet reception rate (PRR), and calculates a list of transmission power and data rates that ensure stable and reliable communication between the EDs and the GW. By using ED power consumption models, network throughput models, and ED latency models to evaluate network performance, the Zebra optimization algorithm is employed to find the optimal data rate for each ED under power-constrained conditions while maximizing network performance. Test results show that, in a single ED scenario, the average PRR achieved by the proposed ADR algorithm for power-constrained EDs in LoRa networks is 14% higher than that of the standard LoRaWAN ADR algorithm. In a multi-ED link scenario (50 end devices), the proposed method reduces the average power consumption of EDs by 10% compared to LoRaWAN ADR, achieves a network throughput of 6683 bps, and an average latency of 2.10 s, demonstrating superior performance overall. The proposed method shows unique advantages in LoRa networks with power-constrained EDs and a large number of EDs, as it not only reduces the average power consumption of the EDs but also optimizes network throughput and average latency.

Attitude information is as important as position information in describing and localizing objects. Based on this, this paper proposes a method for object attitude sensing utilizing ultra-high frequency passive RFID technology. This method adopts a double tag array strategy, which effectively enhances the spatial freedom and eliminates phase ambiguity by leveraging the phase difference information between the two tags. Additionally, we delve into the issue of the phase shift caused by coupling interference between the two tags. To effectively compensate for this coupling effect, a series of experiments were conducted to thoroughly examine the specific impact of coupling effects between tags, and based on these findings, a coupling model between tags was established. This model was then integrated into the original phase model to correct for the effects of phase shift, significantly improving the sensing accuracy. Furthermore, we considered the influence of the object rotation angle on phase changes to construct an accurate object attitude recognition and tracking model. To reduce random errors during phase measurement, we employed a polynomial regression method to fit the measured tag phase information, further enhancing the precision of the sensing model. Compared to traditional positioning modes, the dual-tag array strategy essentially increases the number of virtual antennas available for positioning, providing the system with more refined directional discrimination capabilities. The experimental results demonstrated that incorporating the effects of inter-tag coupling interference and rotation angle into the phase model significantly improved the recognition accuracy for both object localization and attitude angle determination. Specifically, the average error of object positioning was reduced to 12.3 cm, while the average error of attitude angle recognition was reduced to 8.28°, making the method suitable for various practical application scenarios requiring attitude recognition.

Utilizing reliable and accurate positioning and navigation systems is crucial for saving the lives of rescue personnel and accelerating rescue operations. However, Global Navigation Satellite Systems (GNSSs), such as GPS, may not provide stable signals in dense forests. Therefore, integrating multiple sensors like GPS and Inertial Measurement Units (IMUs) becomes essential to enhance the availability and accuracy of positioning systems. To accurately estimate rescuers’ positions, this paper employs the Adaptive Unscented Kalman Filter (AUKF) algorithm with measurement noise variance matrix adaptation, integrating IMU and GPS data alongside barometric altitude measurements for precise three-dimensional positioning in complex environments. The AUKF enhances estimation robustness through the adaptive adjustment of the measurement noise variance matrix, particularly excelling when GPS signals are interrupted. This study conducted tests on two-dimensional and three-dimensional road scenarios in forest environments, confirming that the AUKF-algorithm-based integrated navigation system outperforms the traditional Extended Kalman Filter (EKF), Unscented Kalman Filter (UKF), and Adaptive Extended Kalman Filter (AEKF) in emergency rescue applications. The tests further evaluated the system’s navigation performance on rugged roads and during GPS signal interruptions. The results demonstrate that the system achieves higher positioning accuracy on rugged forest roads, notably reducing errors by 18.32% in the north direction, 8.51% in the up direction, and 3.85% in the east direction compared to the EKF. Furthermore, the system exhibits good adaptability during GPS signal interruptions, ensuring continuous and accurate personnel positioning during rescue operations.

The Basic Leucine Zipper (bZIP) transcription factors (TFs) family is among of the largest and most diverse gene families found in plant species, and members of the bZIP TFs family perform important functions in plant developmental processes and stress response. To date, bZIP genes in Platycodon grandiflorus have not been characterized. In this work, a number of 47 PgbZIP genes were identified from the genome of P. grandiflorus, divided into 11 subfamilies. The distribution of these PgbZIP genes on the chromosome and gene replication events were analyzed. The motif, gene structure, cis-elements, and collinearity relationships of the PgbZIP genes were simultaneously analyzed. In addition, gene expression pattern analysis identified ten candidate genes involved in the developmental process of different tissue parts of P. grandiflorus. Among them, Four genes (PgbZIP5, PgbZIP21, PgbZIP25 and PgbZIP28) responded to drought and salt stress, which may have potential biological roles in P. grandiflorus development under salt and drought stress. Four hub genes (PgbZIP13, PgbZIP30, PgbZIP32 and PgbZIP45) mined in correlation network analysis, suggesting that these PgbZIP genes may form a regulatory network with other transcription factors to participate in regulating the growth and development of P. grandiflorus. This study provides new insights regarding the understanding of the comprehensive characterization of the PgbZIP TFs for further exploration of the functions of growth and developmental regulation in P. grandiflorus and the mechanisms for coping with abiotic stress response.

Diabetes mellitus (DM) refers to a group of chronic diseases with global prevalence, characterized by persistent hyperglycemia resulting from various etiologies. DM can harm various organ systems and lead to acute or chronic complications, which severely endanger human well-being. Traditional treatment mainly involves controlling blood sugar levels through replacement therapy with drugs and insulin; however, some patients still find a satisfactory curative effect difficult to achieve. Extensive research has demonstrated a close correlation between enteric dysbacteriosis and the pathogenesis of various types of DM, paving the way for novel therapeutic approaches targeting the gut microbiota to manage DM. Fecal microbiota transplantation (FMT), a method for re-establishing the intestinal microbiome balance, offers new possibilities for treating diabetes. This article provides a comprehensive review of the correlation between DM and the gut microbiota, as well as the current advancements in FMT treatment for DM, using FMT as an illustrative example. This study aims to offer novel perspectives and establish a theoretical foundation for the clinical diagnosis and management of DM.

Abstract Modeling and motion extraction of human upper limbs are essential for interpreting the natural behavior of upper limb. Owing to the high degrees of freedom (DOF) and highly dynamic nature, existing upper limb modeling methods have limited applications. This study proposes a generic modeling and motion extraction method, named Primitive-Based triangular body segment method (P-BTBS), which follows the physiology of upper limbs, allows high accuracy of motion angles, and describes upper-limb motions with high accuracy. For utilizing the upper-limb modular motion model, the motion angles and bones can be selected as per the research topics (The generic nature of the study targets). Additionally, P-BTBS is suitable in most scenarios for estimating spatial coordinates (The generic nature of equipment and technology). Experiments in continuous motions with seven DOFs and upper-limb motion description validated the excellent performance and robustness of P-BTBS in extracting motion information and describing upper-limb motions, respectively. P-BTBS provides a new perspective and mathematical tool for human understanding and exploration of upper-limb motions, which theoretically supports upper-limb research.

Objective. This study aims to evaluate the impact and predictive value of the preoperative NPRI on short-term complications and long-term prognosis in patients undergoing laparoscopic radical surgery for colorectal cCancer (CRC). Methods. A total of 302 eligible CRC patients were included, assessing five inflammation—and nutrition-related markers and various clinical features for their predictive impact on postoperative outcomes. Emphasis was on the novel indicator NPRI to elucidate its prognostic and predictive value for perioperative risks. Results. Multivariate logistic regression analysis identified a history of abdominal surgery, prolonged surgical duration, CEA levels ≥5 ng/mL, and NPRI ≥ 3.94 × 10−2 as independent risk factors for postoperative complications in CRC patients. The Clavien–-Dindo complication grading system highlighted the close association between preoperative NPRI and both common and severe complications. Multivariate analysis also identified a history of abdominal surgery, tumor diameter ≥5 cm, poorly differentiated or undifferentiated tumors, and NPRI ≥ 2.87 × 10−2 as independent risk factors for shortened overall survival (OS). Additionally, a history of abdominal surgery, tumor maximum diameter ≥5 cm, tumor differentiation as poor/undifferentiated, NPRI ≥ 2.87 × 10−2, and TNM Stage III were determined as independent risk factors for shortened disease-free survival (DFS). Survival curve results showed significantly higher 5-year OS and DFS in the low NPRI group compared to the high NPRI group. The incorporation of NPRI into nomograms for OS and DFS, validated through calibration and decision curve analyses, attested to the excellent accuracy and practicality of these models. Conclusion. Preoperative NPRI independently predicts short-term complications and long-term prognosis in patients undergoing laparoscopic colorectal cancer surgery, enhancing predictive accuracy when incorporated into nomograms for patient survival.

Abstract Background Esophageal cancer (EC) is a global health problem. Asia represents a huge burden of EC globally, and incidence and mortality vary considerably across different Asian regions. Methods Data on incidence, mortality, and preference were extracted from GLOBOCAN 2020. Age‐standardized incidence and mortality rates were calculated overall by sex, age, country, region, and continent. The predicted burden of incidence and mortality in 2040 was calculated based on global demographic projections. Results It was estimated there were 481 552 new cases of and 434 363 deaths from EC in Asia in 2020, accounting for 79.7% and 79.8% of world EC cases and deaths, respectively. EC incidence and mortality in Asia ranked the highest among all continents. Eastern Asia represents the highest age‐standardized world incidence rate (ASWIR) of 12.3 per 100 000 for all Asian regions. Western Asia represents the lowest ASWIR of 1.7 per 100 000, accounting for 0.7% of the globe. There exist obvious differences in epidemiological features in Asian countries, including incidence, mortality, prevalence, and mortality incidence ratio. There is forecast to be up to 781 000 new cases of EC in Asia by 2040, with increasing rates of 63% for incidence and 72% for mortality from 2020. Conclusions Asia has an increasing number of EC cases and deaths. Strategies for targeting in high‐incidence areas, the elderly, and survival should be prioritized to reduce the global EC burden, especially in low‐ and middle‐income countries in Asia.

With an increasing worldwide prevalence, hepatocellular carcinoma (HCC) is the most common primary malignant tumor of the liver in the world. It is also the primary reason for cancer-related death in the world. The pathogenesis of HCC is complex, such as DNA methylation changes, immune regulatory disorders, cell cycle disorders, chromosomal instability, and so on. Although many studies have been conducted on HCC, the molecular mechanisms of HCC are not completely understood. At present, there is no effective treatment for HCC. Hydrogen sulfide (H2S) has long been regarded as a toxic gas with the smell of rotten eggs, but recent studies have shown that it is an important gasotransmitter along with carbon monoxide (CO) and nitric oxide (NO). Increasing evidence indicates that H2S has multiple biological functions, such as anti-inflammation, anti-apoptosis, anti-oxidative stress, and so on. Recently, a lot of evidence has shown that H2S has a “double-edged sword” effect in HCC, but the mechanism is not fully understood. Here, we reviewed the progress on the role and mechanism of H2S in HCC in recent years, hoping to provide a theoretical reference for future related research.

Abstract Background The roles of circRNA and N6‐methyladenosine (m6A) methylation in Crohn's disease (CD) have drawn much attention. Therefore, this investigation aimed to discover how the m6A modification of circRNAs contributes to CD progression. Methods The study performed circRNA sequencing on colon samples from four CD patients and four normal controls (NCs) to screen for dysregulated circRNAs. Quantitative real‐time polymerase chain reaction (qRT‐PCR) was performed to validate the candidate circRNA expression and determine its correlation to CD‐associated inflammatory indicators. In vivo and in vitro investigations were conducted to examine the functions and pathways of circPRKAR1B in CD, besides investigating the m6A modification role in circRNA expression modulation. Results The RNA‐seq revealed that hsa_circ_0008039 (circPRKAR1B) was the most significant upregulated circRNA and was identified as the candidate circRNA for further examinations. Relative circPRKAR1B expression was significantly upregulated in CD colon tissues and closely related to CD‐associated inflammatory indices. The circPRKAR1B expression and function were regulated by methyltransferase‐like 3 (METTL3)‐mediated m6A methylation. In vitro studies indicated that circPRKAR1B promoted pyroptosis mediated by NLRP3 inflammasome (NLRP3; nucleotide‐binding oligomerization domain, leucine‐rich repeat and pyrin domain‐containing 3) and impaired autophagy by interacting with the RNA‐binding protein (RBP) SPTBN1, (SPTBN1; spectrin beta, non‐erythrocytic 1). The in vivo investigations revealed the treatment effects of si‐circPRKAR1B and si‐METTL3 in colitis models of IL‐10‐deficient mice. Conclusion Our study reveals that METTL3‐mediated m6A modification of circPRKAR1B promotes Crohn's colitis by aggravating NLRP3 inflammasome‐mediated pyroptosis via autophagy impairment in colonic epithelial cells.

Storage area networks (SANs) are one of the prevalent reliable data storage solutions. However, cascading failures triggered by data overloading have become a major threat to SANs, preventing the desired quality of service from being delivered to users. Based on our preliminary works on studying the impacts of data loading on the reliability performance of SANs, this paper advances the state of the art by implementing node degree-based load redistribution strategies to enhance the SAN reliability, thus mitigating or even preventing the occurrence of cascading failures during the mission time. Load-based and reliability-based node selection rules are considered, which choose nodes with the highest load level and the lowest reliability for load redistribution, respectively. The relationship between data loading and reliability of an individual SAN component is modeled using the accelerated failure-time model with the power law. The SAN reliability is assessed using a combinatorial decision diagram-based approach. The application and effectiveness of the proposed load redistribution strategies are demonstrated and compared through a case study of an SAN with the mesh topology.

Multienergy system (MES) operation has proven beneficial to the flexibility and efficiency of energy usage. Although efforts have been made toward the simulation and optimization of MESs, successful MES deployment is a grand challenge for market operations. The previous literature focuses on the modeling of the MES and the formulation of corresponding locational marginal prices (LMPs). A detailed analysis of LMP properties in an MES market-clearing model has not been fully developed. Although coupling analysis between different energy systems has been conducted from a system and power flow perspective, a coupling analysis with a specific indicator from a LMP perspective could provide operators with new insight into MES integration. Therefore, this paper first identifies six properties of the LMP in a unified MES market-clearing model. Then, based on the identified properties, two new types of critical load levels (CLL), Self-CLL and Cross-CLL, are defined and discussed to investigate the MES price pattern. Next, the Self-CLLs and Cross-CLLs for an MES are quantified by grey relation analysis (GRA) to deliver an MES coupling analysis from the perspective of LMP patterns. The identified properties and proposed analysis are demonstrated with a small MES case and a large one.

Abstract Background Bidens pilosa L., an annual herb, has recently been shown to be a potential Cd-hyperaccumulating plant. The germination characteristics of B. pilosa have been documented, while the difference among populations remains unclear. Understanding variability in seed germination among populations is crucial for determining which populations to use for soil remediation programs. Results Present study was conducted to compare the requirements of temperature and water potential for germination of B. pilosa cypselae (the central type, hereafter seeds) from three populations using the thermal time, hydrotime, and hydrothermal time models. Seeds of three populations were incubated at seven constant temperatures (8, 12, 15, 20, 25, 30, and 35 °C) and at each of four water potentials (0, -0.3, -0.6, and -0.9 MPa). The results showed that germination percentage and rate of B. pilosa seeds were significantly by population, temperature, water potential and their interaction except for the interaction of population and water potential. Seeds from Danzhou population displayed a higher base temperature (T b) for germination than those from Guilin and Baoshan population, however the ceiling temperature (T c) had no consistent level among the populations but varied according to the water potential. Abstract Background Range of motion (ROM) measurements are essential for diagnosing and evaluating upper extremity conditions. Clinical goniometry is the most commonly used methods but it is time-consuming and skill-demanding. Recent advances in human tracking algorithm suggest potential for automatic angle measuring from RGB images. It provides an attractive alternative for at-distance measuring. However, the reliability of this method has not been fully established. The purpose of this study is to evaluate if the results of algorithm are as reliable as human raters in upper limb movements. Methods Thirty healthy young adults (20 males, 10 females) participated in this study. Participants were asked to performed a 6-motion task including movement of shoulder, elbow and wrist. Images of movements were captured by commercial digital cameras. Each movement was measured by a pose tracking algorithm (OpenPose) and compared with the surgeon-measurement results. The mean differences between the two measurements were compared. Pearson correlation coefficients were used to determine the relationship. Reliability was investigated by the intra-class correlation coefficients. Results Comparing this algorithm-based method with manual measurement, the mean differences were less than 3 degrees in 5 motions (shoulder abduction: 0.51; shoulder elevation: 2.87; elbow flexion:0.38; elbow extension:0.65; wrist extension: 0.78) except wrist flexion. Abstract This study aimed to at explore exploring the biological functions of dysregulated circRNA in Crohn’s disease (CD) pathogenesis, with the overarching goal of and providing potential novel therapeutic targets. CircRNA microarray and quantitative real time-polymerase chain reaction (qRT-PCR) analyses were performed to investigate and verify the candidate dysregulated circRNA. The Next, clinical, in vivo, and in vitro studies were performed to investigate explore the biological function and mechanisms of the candidate circRNA in CD. The therapeutic effect of poly (lactic-co-glycolic acid)-microspheres (PLGA MSs)-carried oe-circGMCL1 in experimental colitis models of IL-10 knock-out mice was assessed. CircGMCL1 was identified as the candidate circRNA by microarray and qRT-PCR analyses. Results showed that circGMCL1 expression was negatively correlated with CD-associated inflammatory indices, suggesting that it is a CD-associated circRNA. Microarray and bioinformatics analyses identified miR-124-3p and Annexin 7 (ANXA7) as its downstream mechanisms. The in vitro studies revealed that circGMCL1 mediates its effects on autophagy and NLRP3 inflammasome-mediated pyroptosis in epithelial cells through the ceRNA network. Moreover, the in vivo studies identified the therapeutic effect of PLGA MSs-carried oe-circGMCL1 in experimental colitis models. This study suggests that circGMCL1 protects intestinal barrier function against Crohn’s colitis through alleviating NLRP3 inflammasome-mediated epithelial pyroptosis by promoting autophagy through regulating ANXA7 via sponging miR-124-3p. Therefore, circGMCL1 can serve as a potential biological therapeutic target for Crohn’s colitis.

BackgroundThe role of the microbiota-gut-brain axis in Parkinson’s disease (PD) has received increasing attention. Although gender differences are known to an essential role in the epidemiology and clinical course of PD, there are no studies on the sex specificity of the microbiota-gut-brain axis in the development and progression of PD.MethodsFresh fecal samples from 24 PD patients (13 males, 11 females) were collected for metagenomic sequencing. The composition and function of the gut microbiota were analyzed by resting-state functional magnetic resonance imaging (fMRI). Gender-dependent differences in brain ALFF values and their correlation with microbiota were further analyzed.ResultsThe relative abundance of Propionivibrio, Thermosediminibacter, and Flavobacteriaceae_noname was increased in male PD patients. LEfse analysis showed that Verrucomicrobial, Akkermansiaceae, and Akkermansia were dominant in the males. In female patients, the relative abundance of Propionicicella was decreased and Escherichia, Escherichia_coli, and Lachnospiraceae were predominant. The expression of the sesquiterpenoid and triterpenoid biosynthesis pathways was increased in male PD patients and was statistically different from females. Compared to the Male PD patients, female patients showed decreased ALFF values in the left inferior parietal regions, and the relative abundance of Propionivibrio was positively correlated with the regional ALFF values.ConclusionOur study provides novel clinical evidence of the gender-specific relationship between gut microbiota alterations and brain function in PD patients, highlighting the critical role of the microbiota-gut-brain axis in gender differences in PD.

IntroductionGut microbiota reportedly play a critical role in some autoimmune diseases by maintaining immune homeostasis. Only a few studies have examined the correlation between gut microbiota and the onset of primary immune thrombocytopenia (ITP), especially in children. The purpose of this study was to investigate changes in the composition and diversity of the fecal microbiota of children with ITP, as well as the correlation between such microbiota and the onset of ITP.MethodsTwenty-five children newly diagnosed with ITP and 16 healthy volunteers (controls) were selected for the study. Fresh stool samples were collected to identify changes in the composition and diversity of gut microbiota as well as for potential correlation analysis.ResultsIn ITP patients, the phyla that were most frequently encountered were Firmicutes (54.3%), followed by Actinobacteria (19.79%), Bacteriodetes (16.06%), and Proteobacteria (8.75%). The phyla that were predominantly found in the controls were, Firmicutes (45.84%), Actinobacteria (40.15%), Bacteriodetes (3.42%), and Proteobacteria (10.23%). Compared with those of the controls, the proportions of Firmicutes and Bacteriodetes in the gut microbiota of ITP patients were increased while the proportions of Actinobacteria and Proteobacteria were decreased. Furthermore, gut microbiota in ITP patients varied by age group, showed specific changes in diversity, and were correlated with antiplatelet antibodies. The fiber-filled polymer composite is one of the best materials which provides a symmetrical superior strength and stiffness to structures. With the strengthening of people’s environmental protection and resource reuse consciousness, the development of renewable materials, especially natural fiber-filled polymer composites, is receiving great attention. This study investigated the mechanical properties of polymer composites incorporating waste materials from the food processing industry and agricultural sources. Waste vegetal fiber-filled polymer biocomposites (WVFFPBs) with varying fiber types and 3D printing orientations were systematically fabricated. Subsequently, the tensile tests were executed to comprehensively assess the anisotropic mechanical behaviors of the WVFFPBs. The results demonstrated that WVFFPBs performed excellent anisotropic mechanical properties compared to pristine matrix samples as print orientation changed. As the printing angle increased from 0° to 90°, the tensile mechanical properties of the WVFFPBs displayed a decreasing trend. Moreover, the print orientation–anisotropic mechanical behavior relationship of 3D-printed WVFFPBs was revealed through the analysis of the material manufacturing characteristics and damage features.

Background This study aimed to investigate outcomes after extremity arterial injury repair and examined the association between outcomes and the degree of soft tissue injury and vascular repair methods. Methods A retrospective study was conducted on 106 patients (108 cases) who underwent emergent microsurgical repair of extremity arterial injury due to trauma and non-perfusion of the affected extremity. The cases were divided into three groups by degree of associated soft tissue injuries: (A) adequate soft tissue coverage over the injured major vessels after radical debridement, (B) inadequate soft tissue coverage over the injured major vessels after radical debridement, and (C) radical debridement was not feasible due to unclear extent of injured soft tissue. Differences in vascular repair methods and outcomes among the three groups were analyzed. Results In Group A (n = 61), microvascular suture and vessel graft achieved 95.1% and 85.0% successful limb reperfusion, respectively. In Group B (n = 31), vessel reconstruction with flap coverage achieved 100% successful reperfusion. Vessel graft achieved 28.6% successful limb reperfusion, while there were no cases of successful reperfusion using microvascular sutures. Inflammasomes play an important role in innate immunity. As a signal platform, they deal with the excessive pathogenic products and cellular products related to stress and injury. So far, the best studied and most characteristic inflammasome is the NLR-family pyrin domain-containing protein 3(NLRP3) inflammasome, which is composed of NLRP3, apoptosis associated speck like protein (ASC) and pro-caspase-1. The formation of NLRP3 inflammasome complexes results in the activation of caspase-1, the maturation of interleukin (IL)-1β and IL-18, and pyroptosis. Many studies have demonstrated that NLRP3 inflammasome not only participates in tumorigenesis, but also plays a protective role in some cancers. Hepatocellular carcinoma (HCC) is a major cause of cancer-related mortality. Currently, due to the lack of effective treatment methods for HCC, the therapeutic effect of HCC has not been ideal. Therefore, it is particularly urgent to explore the pathogenesis of HCC and find its effective treatment methods. The increasing evidences indicate that NLRP3 inflammasome plays a vital role in HCC, however, the related mechanisms are not fully understood. Hence, we focused on the recent progress about the role of NLRP3 inflammasome in HCC, and analyzed the relevant mechanisms in detail to provide reference for the future in-depth researches.

Endoplasmic reticulum (ER) is an important organelle for protein translation, folding and translocation, as well as the post-translational modification and assembly of newly synthesized secreted proteins. When the excessive accumulation of misfolded and/or unfolded proteins exceeds the processing capacity of ER, ER stress is triggered. The integrated intracellular signal cascade, namely the unfolded protein response, is induced to avoid ER stress. ER stress is involved in many pathological and physiological processes including myocardial diseases. For a long time, hydrogen sulfide (H2S) has been considered as a toxic gas with the smell of rotten eggs. However, more and more evidences indicate that H2S is an important gas signal molecule after nitric oxide and carbon monoxide, and regulates a variety of physiological and pathological processes in mammals. In recent years, increasing studies have focused on the regulatory effects of H2S on ER stress in myocardial diseases, however, the mechanism is not very clear. Therefore, this review focuses on the role of H2S regulation of ER stress in myocardial diseases, and deeply analyzes the relevant mechanisms so as to lay the foundation for the future researches.

This study aimed to determine the effects of different dosages of an organic amendment, consisting of a Chinese herbal compound, on cassava growth and rhizosphere microbial diversity. In this study, cassava SC8 growth was determined under four treatments. The growth was promoted with the application of an organic herbal compound amendment, biochar powder, and an agent, in the following groups: 900 kg/ha of organic amendment powder (topdressing) + agent (T1); 1200 kg/ha of organic amendment powder (topdressing) + agent (T2); 1500 kg/ha of organic amendment powder (topdressing) + agent (T3); and an unfertilized control (T4). After 240 days of treatment, the height, the diameter of the stem, the maximum perimeter of the cassava, the tuber number, the weight of a single tuber, the yield per hectare, the mineral elements in the cassava plant, the ash content in the starch and the microbial diversity in the soil were measured. Compared with the control group, the application of 1200 kg of the Chinese herbal compound amendment per ha (T2) had the strongest positive effect on cassava growth, while the height, diameter of the stem and yield per hectare all showed increases of 24.22%, 18.82% and 35.71%, respectively. As for the dry matter content of the tubers, that of T2 decreased by 5.96% compared with T4, while no substantial difference existed among the other groups. The Fe content in T2 decreased by 29.46%, and the Zn in T3 increased by 39.48%, while the contents of other metal ions in the four treatment groups showed no substantial difference. At the same time, the experiment also revealed that, in terms of the bacterial abundance in the soil, Streptophyta dominated in T2, and Fimbriimonas was the unique genera of T2. In addition, the abundances of Nocardioides and unclassfied_Spartobacteria in the T2 group were greatly considerable compared to the other treatment groups. In terms of the fungal abundance in the soil, after being treated with different concentrations, the composition of the soil’s fungi differed greatly among the groups, and the samples at different concentrations contained more than 20 species of unique bacteria. At the genus level, compared with the control group, the relative abundance of unclassfied_Chaetomiaceae in the treatment groups was considerably reduced, and the relative abundance of Psathyrella in T1 increased substantially. The results indicated that the organic amendment could improve the growth of cassava, as well as help to enhance the diversity and optimize the composition of the rhizosphere microbiome.

BackgroundUlcerative colitis (UC) is a chronic autoimmune-related disease that causes inflammation of the intestine. Ankylosing spondylitis (AS) is a common extraintestinal complication of UC involving the sacroiliac joint. However, the pathogenesis of AS secondary to UC has not been studied. This study aimed to investigate the shared pathways and potential common biomarkers of UC and AS.MethodsMicroarray data downloaded from the Gene Expression Omnibus (GEO) database were used to screen differentially expressed genes (DEGs) in the UC and AS datasets. Weighted gene co-expression network analysis (WGCNA) was performed to identify co-expression modules related to UC and AS. Shared genes were then further analyzed for functional pathway enrichment. Next, the optimal common biomarker was selected using SVM-RFF and further validated using two independent GEO datasets. Finally, immune infiltration analysis was used to investigate the correlation of immune cell infiltration with common biomarkers in UC and AS.ResultsA total of 4428 and 2438 DEGs in UC and AS, respectively, were screened. Four modules were identified as significant for UC and AS using WGCNA. A total of 25 genes overlapped with the strongest positive and negative modules of UC and AS. KEGG analysis showed these genes may be involved in the mitogen-activated protein kinase (MAPK) signaling pathway. GO analysis indicated that these genes were significantly enriched for RNA localization. PAN3 was selected as the optimal common biomarker for UC and AS. Immune infiltration analysis showed that the expression of PAN3 was correlated with changes in immune cells.ConclusionThis study first explored the common pathways and genetic diagnostic markers involved in UC and AS using bioinformatic analysis. Results suggest that the MAPK signaling pathway may be associated with both pathogeneses and that PAN3 may be a potential diagnostic marker for patients with UC complicated by AS.

Plant-specific TIFY [TIF(F/Y)XG] proteins serve important roles in the regulation of plant stress responses. This family encodes four subfamilies of proteins, JAZ (JASMONATE ZIM-domain), PPD (PEAPOD), ZML (Zinc-finger Inflorescence-like), and TIFY. In this work, a total of 16 JAZ, 3 PPD, 7 ZML, and 2 TIFY genes were found in cassava (Manihot esculenta Crantz) at the genome-wide level. The phylogenetics, exon-intron structure, motif organization, and conserved domains of these genes were analyzed to characterize the members of the JAZ, PPD, and ZML subfamilies. Chromosome location and synteny analyses revealed that 26 JAZ, PPD, and ZML genes were irregularly distributed across 14 of the 18 chromosomes, and 18 gene pairs were implicated in large-scale interchromosomal segmental duplication events. In addition, JAZ, PPD, and ZML gene synteny comparisons between cassava and three other plant species (Arabidopsis, Populus trichocarpa, and rice) uncovered vital information about their likely evolution. The prediction of protein interaction network and cis-acting elements reveal the function of JAZ, PPD, and ZML genes. Subsequently, expression patterns of JAZ, PPD, and ZML genes were validated by qRT-PCR as being expressed in response to osmotic, salt, and cadmium stress. Moreover, almost all JAZ subfamily genes were responsive to jasmonic acid (JA) treatment. In particular, MeJAZ1, MeJAZ13, and MeJAZ14, were highly up-regulated by three treatments, and these genes may deserve further study. This comprehensive study lays the groundwork for future research into TIFY family genes in cassava and may be valuable for genetic improvement of cassava and other related species.

Intelligent transportation systems (ITS) urgently need to realize vehicle identification, dynamic monitoring, and traffic flow monitoring under high-speed motion conditions. Vehicle tracking based on radio frequency identification (RFID) and electronic vehicle identification (EVI) can obtain continuous observation data for a long period of time, and the acquisition accuracy is relatively high, which is conducive to the discovery of rules. The data can provide key information for urban traffic decision-making research. In this paper, an RFID tag motion trajectory tracking method based on RF multiple features for ITS is proposed to analyze the movement trajectory of vehicles at important checkpoints. The method analyzes the accurate relationship between the RSSI, phase differences, and driving distances of the tag. It utilizes the information weight method to obtain the weights of multiple RF characteristics at different distances. Then, it calculates the center point of the common area where the vehicle may move under multi-antenna conditions, confirming the actual position of the vehicle. The experimental results show that the average positioning error of moving RFID tags based on dual-frequency signal phase differences and RSSI is less than 17 cm. This method can provide real-time, high-precision vehicle positioning and trajectory tracking solutions for ITS application scenarios such as parking guidance, unmanned vehicle route monitoring, and vehicle lane change detection.

Thinopyrum intermedium has been used as a resource for improving resistance to biotic and abiotic stresses and yield potential in common wheat. Wheat line SN304 was derived from a cross between common wheat cultivar Yannong 15 and Th. intermedium. Genomic in situ hybridization (GISH) produced no hybridization signal in SN304 using Th. intermedium genomic DNA as a probe, but fluorescence in situ hybridization (FISH) using oligonucleotides AFA-3, AFA-4, pAs1-1, pAs1-3, pAs1-4, pAs1-6, pSc119.2-1, and (GAA)10 as probes detected hybridization signals on chromosomes 2A, 7A, 2B, 3B, 6B, and 7B in SN304 that differed from Yannong 15. Results of specific markers also indicated that there were Th. intermedium chromatin introgressions on different chromosomes in SN304. In a hydroponic culture experiment, SN304 not only produced more biomass and higher stem and leaf dry weight but also accumulated more phosphorus than Yannong 15 under phosphorus-deficiency stress. Moreover, SN304 produced a lower pH and released more organic acids, especially oxalic acid, than Yannong 15, which suggests that SN304 exudates enabled more absorbance of P than Yannong 15 under comparable conditions. The results indicate that SN304 is a wheat-Th. intermedium introgression line with tolerance to phosphorus-deficiency stress.

Ionogels is a kind of hybrid materials composed of ionic liquids (ILs) and solid polymer network matrix, has been extensively investigated in the most recent decade. Due to the excellent mechanical properties and ionic conductivity, their promising applications in flexible stress and strain sensors have been proposed and explosively developed. In this review, we briefly summarize research progresses on ionogel based flexible stress and strain sensors (IFSSs) from five aspects, including material synthesis, device fabrication, working principles, characteristics and performances, and potential applications. Some outlooks and perspectives are also proposed at the end of review. The review is expected to provide reference and new insights into the research of IFSS.

Abstract In eukaryotes, histones and their variants are essential for chromatin structure and function; both play important roles in the regulation of gene transcription, as well as the development of tumors. We aimed to explore the genomics data of hepatocellular carcinoma (HCC), combined with literature analysis, in terms of the histone variant H2A.Z. Cell phenotype assay confirmed the effect of H2A.Z on the proliferation, metastasis, apoptosis, and cell cycle of HCC cells. H2A.Z was shown to function via the tumor dysregulation signaling pathway, with BCL6 as its interacting protein. In addition, the acetylation level of H2A.Z was higher in HCC and was related to tumor formation. We found the acetylation of H2A.Z to be related to and regulated by lincZNF337-AS1. LincZNF337-AS1 was found to bind to H2A.Z and KAT5 at different sites, promoting the acetylation of H2A.Z through KAT5. We concluded that, in HCC, H2A.Z is an oncogene, whose acetylation promotes the transcription of downstream genes, and is regulated by lincZNF331-AS1.

Powdery mildew is one of the most devastating foliar diseases in wheat production. The wild relative Thinopyrum ponticum (2n = 10x = 70) has been widely used in wheat genetic improvement due to its superior resistance to both biotic and abiotic stresses. In the present study, two wheat-Th. ponticum introgression lines named SN0293-2 and SN0293-7 were developed from the progenies of a cross between the octoploid Trititrigia SNTE20 and common wheat, including the elite cultivar Jimai 22. They had a novel powdery mildew resistance gene (temporarily named PmSN0293) putatively from Th. ponticum pyramided with Pm2 and Pm52, exhibiting excellent Pm resistance at both the seedling and adult stages. Sequential GISH-FISH detected no signal of Th. ponticum in these two lines but a pair of T1BL·1RS in SN0293-2. Chromosomal structural variations were also observed obviously in SN0293-2 and SN0293-7. Through the Wheat 660K SNP array, 157 SNPs, 134 of which were on 6A, were found to be specific to Th. ponticum. Based on the data combined with DNA re-sequencing, seven specific markers, including one CAPS marker on 2B and six CAPS and Indel markers on 6A, were developed, confirming their wheat-Th. ponticum introgression nature. Furthermore, the two lines displayed positive plant height and produced more kernels and higher 1,000-grain weight. Excellent resistance with desirable agronomic traits makes them valuable in wheat breeding programs.

The block chain technology has immense potential in many different applications, including but not limited to cryptocurrencies, financial services, smart contracts, supply chains, healthcare services, and energy trading. Due to the critical nature of these applications, it is pivotal to model and evaluate dependability of the block chain-based systems, contributing to their reliable and robust operation. This paper models and analyzes the dependability of Bitcoin nodes subject to Eclipse attacks and state-dependent mitigation activities. Built upon the block chain technology, the Bitcoin is a peer-to-peer cryptocurrency system enabling an individual user to trade freely without the involvement of banks or any other types of intermediate agents. However, a node in the Bitcoin is vulnerable to the Eclipse attack, which aims to monopolize the information flow of the victim node. A semi-Markov process (SMP) based approach is proposed to model the Eclipse attack behavior and possible mitigation activities that may prevent the attack from being successful during the attack process. The SMP model is then evaluated to determine the steady-state dependability of the Bitcoin node. Numerical examples are provided to demonstrate the influence of the time to restart the Bitcoin software and time to detect and delete the malicious message on the Bitcoin node dependability.

Abstract Background Thinopyrum intermedium (2n = 6x = 42) is an important wild perennial Triticeae species exhibiting many potentially favorable traits for wheat improvement. Wheat-Th. intermedium partial amphiploids serve as a bridge to transfer desirable genes from Th. intermedium into common wheat. Results Three octoploid Trititrigia accessions (TE261–1, TE266–1, and TE346–1) with good resistances to stripe rust, powdery mildew and aphids were selected from hybrid progenies between Th. intermedium and the common wheat variety ‘Yannong 15’ (YN15). Genomic in situ hybridization (GISH), fluorescence in situ hybridization (FISH) and multicolor GISH (McGISH) analyses demonstrated that the three octoploid Trititrigia possess 42 wheat chromosomes and 14 Th. intermedium chromosomes. The 14 alien (Th. intermedium) chromosomes belong to a mixed genome consisting of J-, JS- and St-genome chromosomes rather than a single J, JS or St genome. Different types of chromosomal structural variation were also detected in the 1A, 6A, 6B, 2D and 7D chromosomes via FISH, McGISH and molecular marker analysis. The identity of the alien chromosomes and the variationes in the wheat chromosomes in the three Trititrigia octoploids were also different. Conclusions The wheat-Th. intermedium partial amphiploids possess 14 alien chromosomes which belong to a mixed genome consisting of J-, JS- and St- chromosomes, and 42 wheat chromosomes with different structural variations. These accessions could be used as genetic resources in wheat breeding for the transfer of disease and pest resistance genes from Th. intermedium to common wheat.

CdSSe NFs thin film is used as a SERS substrate with an ultralow limit of detection (LOD, 10−14 M), high apparent enhancement factor (EF, 3.62 × 109), and excellent SERS stability (relative standard deviation, RSD = 3.05%) for probe molecules of Rh6G. Further, CdSSe NFs substrate is successfully applied in the sensitive, quantitative, and label-free analysis of ciprofloxacin (CIP) and enrofloxacin (ENR) antibiotics, which exhibit LODs of below 0.5 ppb. This excellent SERS platform may be widely utilized for sensitive life science and environmental sensing.

V2P (vehicle-to-pedestrian) communication can improve road traffic efficiency, solve traffic congestion, and improve traffic safety. It is an important direction for the development of smart transportation in the future. Existing V2P communication systems are limited to the early warning of vehicles and pedestrians, and do not plan the trajectory of vehicles to achieve active collision avoidance. In order to reduce the adverse effects on vehicle comfort and economy caused by switching the “stop–go” state, this paper uses a PF (particle filter) to preprocess GPS (Global Positioning System) data to solve the problem of poor positioning accuracy. An obstacle avoidance trajectory-planning algorithm that meets the needs of vehicle path planning is proposed, which considers the constraints of the road environment and pedestrian travel. The algorithm improves the obstacle repulsion model of the artificial potential field method, and combines it with the A* algorithm and model predictive control. At the same time, it controls the input and output based on the artificial potential field method and vehicle motion constraints, so as to obtain the planned trajectory of the vehicle’s active obstacle avoidance. The test results show that the vehicle trajectory planned by the algorithm is relatively smooth, and the acceleration and steering angle change ranges are small. Based on ensuring safety, stability, and comfort in vehicle driving, this trajectory can effectively prevent collisions between vehicles and pedestrians and improve traffic efficiency.

Abstract Background Long intergenic non-protein coding RNA 00342 (LINC00342) has been identified as a novel oncogene. However, the functional role of LINC00342 in colorectal cancer (CRC) remains unclear. Methods The expression of LINC00342 is detected by real-time PCR (RT-PCR) analysis. Cell proliferation, migration and invasion and xenograft model are examined to analyze the biological functions of LINC00342 in vitro and in vivo using colony formation, would healing and transwell analyses. Dual-luciferase reporter and RNA immunoprecipitation (RIP) assays are used to identify the target interactions between LINC00342, miR-19a-3p and aminopeptidase like 1 (NPEPL1). Results LINC00342 was highly expressed in CRC. Down-regulation of LINC00342 inhibited cell proliferation and metastasis of CRC cells. Moreover, knocking down LINC00342 inhibited the tumor growth in vivo. Mechanistic investigation revealed that LINC00342 might sponge miR-19a-3p to regulate NPEPL1 expression. Further investigation indicated that the ontogenesis facilitated by LINC00342 was inhibited due to the depletion of NPEPL1. Conclusion LINC00342 promotes CRC progression by competitively binding miR-19a-3p with NPEPL1.

Abstract Background This study aimed to investigate the association between enhanced recovery after surgery (ERAS) nursing program and stoma-related complications (SRCs) and prognosis in patients with low rectal cancer (LRC) undergoing abdominoperineal resection with sigmoidostomy. Methods LRC patients who underwent elective abdominoperineal resection with sigmoidostomy between May 2016 and May 2019 were retrospectively enrolled. The occurrence of early major or minor SRCs (within postoperative 30 days) was set as the primary end-point. Clinicopathological variables and laboratory tests were compared between patients with or without SRCs. The univariate and multivariate logistic regression analyses were performed to investigate risk factors for SRCs. Hospitalization satisfaction-related and prognosis-related variables were compared between LRC patients with or without ERAS nursing program. Results A total of 288 patients were enrolled and the incidence of SRCs was 26.7% (77/288). ERAS nursing program was the only independent risk factor for SRCs in LRC patients (OR 2.04, 95%CI 1.31–3.12, P = 0.016) by the multivariate logistic regression analysis. Moreover, ERAS nursing program was associated with higher hospitalization satisfaction rate, faster bowel function recovery, better psychological status, and higher quality of life. Conclusions ERAS nursing program was a protective factor for SRCs and associated with improved prognosis in LRC patients undergoing elective abdominoperineal resection with sigmoidostomy.

Abstract Background Lesion-mimic and premature aging (lmpa) mutant lmpa1 was identified from the ethyl methane sulfonate (EMS) mutant library in the bread wheat variety Keda 527 (KD527) background. To reveal the genetic basis of lmpa1 mutant, phenotypic observations and analyses of chlorophyll content and photosynthesis were carried out in lmpa1, KD527 and their F1 and F2 derivatives. Further, bulked segregation analysis (BSA) in combination with a 660 K SNP array were conducted on the F2 segregation population of lmpa1/Chinese spring (CS) to locate the lmpa1 gene. Results Most agronomic traits of lmpa1 were similar to those of KD527 before lesion-like spots appeared. Genetic analysis indicated that the F1 plants from the crossing of lmpa1 and KD527 exhibited the lmpa phenotype and the F2 progenies showed a segregation of normal (wild type, WT) and lmpa, with the ratios of lmpa: WT = 124:36(χ2 = 1.008

Distributed energy resources are capable of enhancing grid resilience through island operations in contingency. This paper proposes a multi-year, multi-criteria generation expansion model to achieve distribution power resilience through renewable energy integration. In cases that distribution circuits or fuel lifeline are destroyed post natural disasters, wind- and solar-based generators form island microgrids to power the critical load. The goal is to determine the sizing, siting and maintenance of distributed energy resources such that system cost and power shortage in contingency are minimized. Moment methods and central limit theorem are used to characterize the spatial climate uncertainty, generation intermittency, and voltage variation. The research contributions are twofold. First, the expansion model achieves triple goals by meeting the annual load growth, reducing the carbon footprint, and enhancing the grid resilience manifested as prevention and survivability. Second, we present an improved direct zigzag algorithm to search for the Pareto solutions. Numerical examples show that the zigzag search outperforms evolutionary-based algorithms as it can identify higher-quality non-dominant solutions by exploring a wider Pareto frontier.

Fecal microbiota transplantation (FMT), which is thought to have the potential to correct dysbiosis of gut microbiota, has been used to treat inflammatory bowel disease (IBD) for almost a decade. Here, we report an interventional prospective cohort study performed to elucidate the extent of and processes underlying microbiota engraftment in IBD patients after FMT treatment. The cohort included two categories of patients: (a) patients with moderate to severe Crohn’s disease (CD) (Harvey–Bradshaw Index ≥ 7, n = 11) and (b) patients with ulcerative colitis (UC) (Montreal classification S2 and S3, n = 4). All patients were treated with a single FMT (via mid‐gut, from healthy donors), and follow‐up visits were performed at baseline, 3 days, 1 week, and 1 month after FMT (missing time points included). At each follow‐up time point, fecal samples and clinical metadata were collected. For comparative analysis, 10 fecal samples from 10 healthy donors were included to represent the diversity level of normal gut microbiota. Additionally, the metagenomic data of 25 fecal samples from five individuals with metabolic syndrome who underwent autologous FMT treatment were downloaded from a previous published paper to represent fluctuations in microbiota induced during FMT. All fecal samples underwent shotgun metagenomic sequencing. We found that 3 days after FMT, 11 out of 15 recipients were in remission (three out of four UC recipients; 8 out of 11 CD recipients). Generally, bacterial colonization was observed to be lower in CD recipients than in UC recipients at both species and strain levels. Furthermore, across species, different strains displayed disease‐specific displacement advantages under two‐disease status. Finally, most post‐FMT species (> 80%) could be properly predicted (area under the curve > 85%) using a random forest classification model, with the gut microbiota composition and clinical parameters of pre‐FMT recipients acting as factors that contribute to prediction accuracy.

In order to overcome the limitations of conventional therapeutic systems in the treatment of cancer, nanoparticles (NPs) have been rapidly produced and developed as a separate treatment method for control of cancer. Synthesis of nanoparticles using plant-based materials (green synthesis), due to the easy and cost-effective synthesis, production of non-toxic, sustainable and environmentally friendly products, can be considered the most appropriate method for preparation of NPs. In this study, after synthesis of Bi2O3 NPs using Ginger (Zingiber officinale) root (rhizome) extract, the synthesized NPs were characterized and their potential application as selective anticancer agents against HCT116 colorectal cancer cells was evaluated through regulation of PI3K/AKT/mTOR signaling pathway, whereas the human kidney (HK-2) cells were used as normal cells. FTIR analysis showed a band at 673 cm−1 attributed to Bi-O vibration with a fingerprint region at 1291 cm−1 demonstrating the attachment of the organic molecules to the synthesized Bi2O3 NPs. UV–visible study showed a λmax of around 268 nm, whereas XRD analysis showed eight clear peaks, demonetizing the crystalline phase of synthesized Bi2O3 NPs. TEM analysis showed that spherical-shaped Bi2O3 NPs have a size range of 20–50 nm with a man size of around 35 nm. Finally, DLS analysis determined that Bi2O3 NPs have a hydrodynamic size of about 71.19 nm (PDI of 0.179) and a zeta potential value of −44.39 mV, revealing the good colloidal stability of NPs. Cellular assays (MTT, LDH, flow cytometry, and RT-qPCR) showed that synthesized Bi2O3 NPs selectively induced anticancer effects against HCT116 colorectal cancer cells through membrane leakage, generation of ROS, induction of apoptosis via dysregulation of Bax, Bcl-2 and caspase-3 at mRNA level mediated via regulation of PI3K/AKT/mTOR signaling pathway. In conclusion, it may be suggested that the presence study could provide useful information for the potential anticancer effects mediated by synthesized Bi2O3 NPs in vitro, although further studies, including in vivo studies and clinical trials, are needed to support our findings.

Introduction and objectives: Long non-coding RNAs (lncRNAs) have great potential as therapeutic targets in hepatocellular carcinoma (HCC). In this study, we aimed to uncover the function and molecular mechanism of long intergenic non-protein coding RNA 1006 (LINC01006) in HCC. Materials and methods: Mice were injected with HCC cells in order to establish the HCC model. Quantitative reverse transcription polymerase chain reaction was used to determine the expression levels of LINC01006, cell adhesion molecule 1 (CADM1), and microRNA (miR)-194-5p in HCC tissues and cells. The cell proliferation, invasion, and migration abilities were assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide, transwell, and wound healing assays. The interrelation between LINC01006, miR-194-5p, and CADM1 was confirmed by a dual-luciferase reporter assay. Western blotting was employed to assess the relative protein expression level of CADM1. Results: LINC01006 and CADM1 displayed upregulation, but miR-194-5p exhibited downregulation in HCC cells and tissues. Short hairpin (sh)-LINC01006 and miR-194-5p mimics repressed the proliferative, migratory, and invasive capacities of HCC cells, and injection of sh-LINC01006 restrained the growth of HCC tumours in mice. LINC01006 served as a competing endogenous RNA of miR-194-5p and was inversely correlated with miR-194-5p. CADM1 was targeted by miR-194-5p, inversely correlated with miR-194-5p, and positively associated with LINC01006. Furthermore, transfection of pcDNA-CADM1 or the miR-194-5p inhibitor reversed the suppressive effects of sh-LINC01006 on the proliferation, invasion, and migration abilities of HCC cells. Conclusions: Downregulation of LINC01006 repressed the development of HCC by sponging miR-194-5p to modulate the expression of CADM1, implying its potential as a therapeutic target for HCC.

Glycerol-induced resistance to various pathogens has been reported in different plants. Glycerol kinase (GK), a vital rate-limiting enzyme that catalyzes glycerol conversion to glycerol-3-phosphate (G3P), participates in responses to both abiotic and biotic stresses. However, its physiological importance in rice defenses against pathogens remains unclear. In this research, quantification analysis revealed that GK levels were significantly induced in rice leaves infected by Xanthomonas oryzae pv. oryzae (Xoo) strain PXO99. A typical GK-encoding gene OsNHO1 was cloned in rice. The transcriptional levels of OsNHO1 were significantly induced by salicylic acid, jasmonic acid, and Xoo-PXO99. Ectopic expression of OsNHO1 partially rescued the resistance to P. s. pv. phaseolicola in the Arabidopsis nho1 mutant. In the overexpressing transgenic rice lines (OsNHO1-OE), the content of GK and the transcriptional level of OsNHO1 were increased and the resistance to bacterial blight and blast was improved, while reduced OsNHO1 expression impaired the resistance in OsNHO1-RNAi lines. The wax contents and expression of the wax synthesis regulatory genes were significantly increased in the overexpression lines but decreased in the OsNHO1-RNAi lines. We then confirmed the interaction partner of OsNHO1 using yeast two-hybrid and bimolecular fluorescence complementation assays. The transcription of the interaction partner-encoding genes OsSRC2 and OsPRs in OsNHO1-RNAi lines was downregulated but upregulated in OsNHO1-OE lines. Thus, we concluded that OsNHO1 provided disease resistance by affecting the wax content and modulating the transcription levels of PR genes.

Objective. This study aimed to evaluate whether the site of C7 neurotomy affects spinal cord glial cell activation and pain-related behavior on the paralyzed side in a rat stroke model. Methods. After middle cerebral artery occlusion (MCAO) was induced in male Sprague-Dawley rats, they underwent C7 neurotomy 0, 2, and 4 mm distal to the intervertebral foramen on the paralyzed side. Pain-related behavior and immunofluorescence examination of spinal cord glial cell activation in the ipsilateral C7 dorsal horn were evaluated. Results. Mechanical paw withdrawal threshold (MPWT) was lower, and the number of microglia and astrocytes (/mm2) was higher as the distance between the site of C7 neurotomy and the intervertebral foramen decreased from 4 mm to 0. Conclusion. The site of C7 neurotomy affects MPWT and spinal cord glial proliferation in rats with MCAO. Nerve division closer to intervertebral foramen resulted in lower MPWT and higher degree of glial proliferation in the spinal cord.
