Cells express a large array of membrane receptors on their surface that function as a communication channel between the extra- and intracellular environment of the cell. Ligands for these receptors span a wide range of biomolecules, from proteins to carbohydrates to small molecules. Some receptors are continuously recycling between the membrane and the inside of a cell, whereas others are in a steady-state at the membrane and need ligand binding for their activation and subsequent internalization. Synthetic molecules that bind to these membrane receptors can be used to either modulate their function, or to target a reporter group (i.e. a fluorescent dye) and/or a bio-active compound (drug, protein) to cells that express this receptor, ensuring delivery to a specific cell-type. The research described in this Thesis combines synthetic and biochemical methodologies to create ligands that interact selectively with membrane receptors of the GPCR and lectin-binding families. Attachment of synthetic probes, proteins or cytostatic molecules to these ligands by a variety of chemical and enzymatic methods ensured their uptake exclusively into cells that expressed the receptor of interest. Visualization of this process was enabled by the incorporation of a fluorescent dye into the final constructs.

In just a few years, traffic management has been transformed from an everyday, passive task for the highway authority to an important policy instrument. This transformation is not without reason. Traffic management has improved the safety and operations on our roadways, while it is relatively inexpensive, flexible and rapidly deployable. Transportation professionals are also convinced that the potential of traffic management is far greater than previously imagined.

In de afgelopen decennia heeft het mobiliteitsvraagstuk zich ontwikkeld tot een hardnekkig probleem. De sterke toename van de vraag naar mobiliteit (en capaciteit) van de laatste jaren komt voor een groot deel voor rekening van het personenauto- en vrachtwagenverkeer. De combinatie van autonome groei van het autogebruik en een sterke groei van de bedrijvigheid binnen, maar ook buiten de Randstad, heeft er toe geleid dat de kwaliteit van het mobiliteitsproduct steeds verder onder druk is komen te staan. Hoewel het aantal verplaatsingen per persoon per dag in de afgelopen decennia niet veranderd is, steeg het aantal afgelegde kilometers per persoon per dag van 28 km in 1985 naar 35 km in 2008. Ook de vervoersprestatie van vrachtverkeer groeide gestaag door. Het hoofdwegennet, dat in Nederland toch al een groot aandeel in het aantal afgelegde kilometers heeft, wordt voor een steeds groter deel voor regionale verkeersbewegingen (bijvoorbeeld woon-werkverkeer) gebruikt. De toename van het aantal inwoners, banen en autobezit zorgde in de periode 2000-2008 voor een toename van het aantal autokilometers met ongeveer 14%. In diezelfde periode nam mede daardoor het reistijdverlies toe met 55%1. De gevolgen van de toename van het aantal afgelegde kilometers, gekoppeld aan de zware belasting van het wegennet en de beperkte uitbreiding van de capaciteit in de afgelopen jaren, zijn dat de deur-tot-deur reistijden langer worden, terwijl tegelijkertijd de voorspelbaarheid steeds verder afneemt. Deze ontwikkeling blijft niet beperkt tot de Randstad. Wat nu nog een aantrekkelijke regio buiten de Randstad is met een (nog) redelijke bereikbaarheid met enkele taaie knelpunten, dreigt onder invloed van een vastlopende en overkokende Randstad een regio met Randstedelijke bereikbaarheidsproblemen te worden. De groei van het wegverkeer is vooral een logisch gevolg van sociaal-economische ontwikkeling, maar tegelijkertijd vormen de gevolgen van deze groei van het wegverkeer ook een bedreiging voor verdere economische ontwikkeling. De bereikbaarheidskaart van Nederland2 laat dit duidelijk zien.

Robustness of a network is a main objective for road network managers these days, and has therefore become an important study area for transportation scientists. This article discusses one specific aspect in assessing road network robustness: the consequences of the closure of a link. These spillback effects have been examined in a dedicated traffic simulator in which the representation of spillback can be switched on and off. The impacts are studied in a simulation study of a road network of a regional size in which sequentially links are blocked. Two scenarios for route choice are considered: a fixed route choice based on a daily congestion pattern and a route choice adapted to the actual congestion caused by the closure. The study has also shown the influence of information which makes travellers adapt their routes. Road network robustness and characteristics of vulnerable links are evaluated for both spillback and non-spillback cases. It is found that a valid spillback modelling is a prerequisite for correctly analysing the robustness of the network as a whole, as well as for correctly indicating the locations in the network where a closure causes the largest delays. Furthermore, without simulating spillback, it is not possible to identify correctly the most vulnerable links for the network performance., European Journal of Transport and Infrastructure Research, Vol 8 No 4 (2008)
